Biogeography and biosystematics of plum curculio, Conotrachelus nenuphar (Herbst)/Wolbachia interactions

This research focused on the reproductive incompatibility and genetic differences between the two strains of plum curculio, Conotrachelus nenuphar (Herbst). Two molecular markers served as the basis for the strain distribution analysis of plum curculio and Wolbachia symbiont. One marker is the partial mitochondrial cytochrome oxidase gene subunit I (mtCOI) of plum curculio. Another marker is the Wolbachia Surface Protein (wsp) gene of Wolbachia associated with plum curculio. First, the reproductive compatibility of cross-populations mating in plum curculio was studied during the summers of 2004 and 2006. The results confirmed the reproductive incompatibility among plum curculio geographic populations. A unidirectional incompatibility was revealed in an approximate north and south transect of the range of plum curculio (4 x 4 two factorial design: NY, VA, FL, and WV): there was a significant low fertility in WV males mated with NY (40%) and VA (29%) females. The Florida population showed a different pattern: FL males have a significantly lower fertility with VA (46%) and WV (37%) females while FL females were compatible with all males from the four populations. The results of experiment 2 indicated that within the northern geographic area populations (3 x 3 two factorial design: NY, MA, and NJ) were compatible with each other. An opposite unidirectional reproductive incompatibility was revealed in the combination of NJ males with FL females, which showed a significant low fertility (47%). A bi-directional incompatibility occurred between FL and WV reciprocal cross mating. FL males mated with WV females (26%) and WV males mated with FL females (21%) both have the significant low fertility compared to fertility of within their population matings. The genetic diversity among plum curculio populations from different geographic locations was investigated using the partial mtCOI gene. A total of 50 samples from 10 populations were sequenced. PCR products were 863 bp in length. A total of 23 unique sequence haplotypes were found in the 50 samples tested. Haplotype G (n = 5), L (n = 12) and T (n = 13) comprised 60% of 50 samples. The nucleotide distances between those haplotypes ranged from 0.12% to 4.87%. Genetic distances between northern and southern group plum curculios range from 4.17% to 4.87%. Two distinct major clades were found, using three different phylogenetic analyses: 1) neighbor joining (NJ), 2) maximum-parsimony (MP), and 3) maximum-likelihood (ML). 100% bootstraps support the northern clade and the southern clade was strongly supported (100/100/86, NJ/MP/ML) as well. The mid-southern subclade within the southern clade was also strongly supported (70/82/71, NJ/MP/ML) and the far-southern subclade was supported in NJ tree (81%) but was not resovled in MP and ML trees. The mid-southern subclade included haplotypes from two NJ, Washington, VA (Ra), Blacksburg, VA (BL) and 50% of WV populations and the far-southern subclade included haplotypes from FL, GA, Whitethorne, VA (Ke), Troutville, VA (Bo) and another 50% of WV populations. The results suggested that the northern and the southern clade could correspond with the northern and southern strains, respectively, of plum curculio. In this study, the mtCOI sequence was highly informative as a molecular marker in that it was useful to distinguish C. nenuphar from northern and from southern geographic locations in the eastern United States. However, the number of generations per year of several geographic populations within the southern clade still needs to be determined. The distribution of Wolbachia infection associated with plum curculio strains was investigated. 91 of 93 samples were infected by Wolbachia. Three unique Wolbachia strains were identified. The strains wCne1 and wCne2 (593 bp) were 97% identical, and their sequences were both 84% identical with wCne3 (590 bp). The wsp sequence of wCne1 was 99% identical to Wolbachia sequenced from the neotropical beetle, Chelymorpha alternans Boheman (Keller et al. 2004). The wCne2 sequence was 98.5% identical to the flower bug, Orius nagaii Yasunaga (Miura and Tagami, unpublished). The wCne3 sequence was 100% identical to Wolbachia sequenced from the tephritid fruit fly, Dacus destillatoria (Jamnongluk et al. 2000) and the ant, Formica exsecta (Reuter and Keller 2003). PCR - Restriction Fragment Length Polymorphism (RFLP) was used for superinfection detection. Of 93 samples, 15 (16.1%), 21 (22.6%), 19 (20.4%), 36 (38.7%) samples were infected by wCne1, wCne2, wCne1 plus wCne2, and wCne3, respectively. Only two (2.2%) samples had no infection. The wCne3 strain was always present as a single infection. Therefore, current results suggest that Wolbachia strains approximate the distribution of plum curculio strains: the northern strain is infected with wCne1 and wCne2 strains in supergroup B, the southern strain is infected with wCne3 strain in supergroup A and the mid-Atlantic region is the convergence area. Compared with the haplotype distribution of plum curculio mtCOI gene, there was a closer relation of the mid-southern PC clade to the far-southern clade than to the northern clade. However, Wolbachia symbionts in mid-southern PC are more closely related to those in northern PC than to those in far-southern PC. The relationship of Wolabchia infection with reproductive incompatibility between plum curculio populations is also discussed. / Ph. D.

The causes and consequences of variation in the cloacal microbiome of tree swallows (Tachycineta bicolor)

Animals are ecological landscapes that host communities of microbes often referred to as microbiomes. These microbes can be transferred between individuals when they come into contact, such as during mating. Microbes that reside in or on any aspect of a host that becomes exposed to the reproductive tract or gametes comprise the reproductive microbiome. These microbes within the reproductive microbiome are important to overall host biology because they can influence host reproductive function, and thus play a role in shaping host ecology, evolution, and fitness. Though previous work has revealed much about the impact of beneficial and pathogenic microbes within the reproductive tract, much is left to be learned from describing the dynamic nature of the reproductive microbiome, and ultimately, how it affects host fitness. For my dissertation, I asked questions regarding how and why reproductive microbiome diversity varies among individuals. For instance, does reproductive microbiome diversity vary with respect to the number of mates or mating activity? Does reproductive microbiome diversity vary with host age or breeding stage? Are there fitness consequences associated with differences in reproductive microbiome diversity? To explore these questions, I studied tree swallows (Tachycineta bicolor), a socially monogamous bird in which both females and males engage in extra-pair mating activity. I focused on the cloacal microbiome as it is the site of contact during mating, and thus where microbes can be sexually transferred between individuals. I found that social partners did not have more similar cloacal microbiome diversity compared to other individuals in the same population, and that cloacal microbiome diversity was similar between sexes (Chapter II). By combining an observational approach with a hormone implant manipulation, I found that neither the number of sires per brood nor the increased mating activity of females significantly influenced cloacal microbiome richness or community structure. However, female age and breeding stage did significantly correlate with cloacal microbiome richness and community structure (Chapter III). Based on these findings, I hypothesized that the effect of mating activity on variation in the cloacal microbiome may only be detectable over a female's lifetime, and not within a single breeding season. In addition, I found evidence for a relationship between lay date and cloacal microbiome structure, after controlling for age. And I found that older females lay earlier in the season compared to younger, first-time breeding females (Chapter IV). These results provide support for a relationship between lay date and the cloacal microbiome and highlight the importance of age to this relationship. I then performed observational and experimental studies where I assessed the number of mates per female and manipulated female sexual activity with hormone implants. Sporisorium scitamineum é um fungo basidiomiceto causador do carvão da cana-de-açúcar, uma doença com impacto negativo no cultivo da cana-de-açúcar, e com ocorrência em todos os países produtores. A manifestação da doença na cultura da cana depende da formação de uma hifa dicariótica a partir da anastomose de duas hifas haplóides compatíveis com relação ao tipo de reação sexual (mating-type). O controle do cruzamento sexuado (mating) é realizado pela expressão de um conjunto de genes presentes em dois loci, a e b. O locus a codifica um lipopeptídeo com função de feromônio e um receptor de feromônio, responsáveis pelo reconhecimento de células compatíveis e fusão de hifas, enquanto o locus b codifica fatores de transcrição que controlam a expressão de genes responsáveis pela manutenção das hifas dicarióticas durante o processo de infecção e crescimento do fungo dentro da planta. Apesar de desempenharem função essencial no processo de infecção e manutenção da doença em cana-de-açúcar, o conhecimento a respeito da organização genômica ou da função dos demais genes presentes nos loci a e b em S. scitamineum e em outros fungos causadores de carvão é ainda incipiente. Desta forma, o objetivo geral do presente trabalho foi isolar as regiões genômicas relacionadas aos genes de cruzamento em S. scitamineum e analisar comparativamente com regiões similares já descritas e depositadas em bancos de dados públicos. Para o isolamento destas regiões, foi construída uma biblioteca genômica em BAC de uma linhagem haplóide de S. scitamineum, a Ssc39 (+), isolada de uma variedade de cana-de-açúcar com sintomas de alta susceptibilidade. Foram selecionados 11 clones por PCR. Os insertos foram sequenciados e utilizados para confirmação da montagem dos loci no sequenciamento do genoma do fungo. Apesar do fungo S. scitamineum apresentar sistema bipolar de reação sexual assim como o fungo U. hordei, as análises comparativas de ambos os locus indicaram que S. scitamineum apresenta maior similaridade com o fungo S. reilianum principalmente com o alelo a1, no qual apresenta sistema tetrapolar de reação sexual. A anotação e caracterização dos genes do tipo de reação sexual (mating type) possibilitaram a comparação e melhor entendimento sobre esses genes de grande importância na patogenicidade e no ciclo de vida do fungo. / Sporisorium scitamineum is a basidiomycete fungus causing the smut disease in sugarcane, with a negative impact on the cultivation of sugarcane, and occurring in all producing countries. The manifestation of the disease in sugarcane crop depends on the formation of a dikaryotic hyphae originated of the anastomosis of two haploid mating type compatible cells. The control of the sexual crossing (mating) is performed by expression of a set of genes present in two loci, a and b. The locus a encodes a lipopeptide with the function of pheromone and pheromone membrane receptor responsible for cell recognition and compatible hyphal fusion, whereas the locus b encodes transcription factors that control the expression of genes responsible for the maintenance of the dikaryotic hyphal growth in plant. Although they play an essential role in the maintenance of infection and disease in sugarcane process, knowledge about the genomic organization and function of other genes in these two loci of S. scitamineum and other smut fungi is still incipient. Thus, the overall goal of this work was to isolate genomic regions related to the mating type in S. scitamineum and to perform a comparative analyze with similar regions described and deposited in public databases. For the isolation of these regions, we constructed a genomic BAC library of a haploid strain of S. scitamineum, the Ssc39 (+), isolated from a variety of sugarcane with symptoms of high susceptibility. Eleven clones were selected by PCR. The inserts were sequenced and used to confirm the assembly of both loci in the genome sequencing of the fungus. Although S. scitamineum belongs to the class of bipolar system of sexual response as well as the fungus U. hordei , the comparative analysis of both loci indicated that S. scitamineum shows greater similarity to the S. reilianum mainly with A1 allele, which has a tetrapolar system sexual response. The annotation of the genes and characterization mating type genes enabled the comparison and better understanding of the importance of these genes in the life cycle of the fungus.

The cognitive biology of mate choice in túngara frogs (Physalaemus pustulosus)

Sexual selection is responsible for a great diversity of elaborate male traits. A general female preference for males that have exaggerated traits drives this process, but the reasons females exhibit this preference are often unclear. Recent advances in understanding signal evolution have emerged from studies of receiver psychology that focus on how receivers perceive and process communication signals. I apply the perspective of receiver psychology to understand female preference for elaborate signals in túngara frogs (Physalaemus pustulosus). Male túngara frogs produce advertisement calls of variable complexity. Females exhibit a strong preference for complex to simple calls, but previous studies have not found consistent patterns of preference between calls of variable complexity. In my doctoral research, I investigate the function of variable complexity in túngara frogs. Specifically, I address the following questions: 1) Are calls of variable complexity especially relevant to females in certain contexts? Do males respond to female behavior by increasing their production of complex calls? 2) Does male to female proximity influence female response to call complexity? 3) Are females constrained by their perceptual biology in discriminating differences in call complexity? 4) Can females remember attractive males over silences between bouts of advertising? Is working memory for attractive males dependent upon signal complexity? And 5) Does signal memorability increase with signal complexity in a linear relationship? These studies provide several new perspectives to an understanding of female preference for elaborate signals. Phonotaxis experiments demonstrate that females use elicitation behaviors to influence male production of complex calls, that proximity influences female response to signal elaboration, that females are constrained by their perceptual biology in discriminating between complex calls, that memory can influence the evolution of signal complexity, and that memorability and signal complexity share a non-linear relationship. / text

Thus, mating behavior of P. nigrispinus was investigated as function of male body size (i), previous mating status (ii), risk of predation (iii), interrupted mating (iv), and spermatogenesis (v). The female choice for mating with large or small body size males was investigated under three conditions: multiples choice within a population of large and small males; partial choice (ca. only one pair of small and large male) and no choice (only small or large male). Time to initiate a copula and its duration was observed after pairing males and females under the risk of predation imposed by the presence of the predatory wasp Polistes versicolor Oliver (Hym.: Vespidae). Furthermore, females had their reproductive success evaluated with interrupted mating at different intervals (30, 60, 120 and 240 minutes afterinitiating a mating) and when mating with males previously mated. The spermatogenesis of these mated and unmated males was investigated. The outcomes show that P. nigrispinus female mates with males disregarding their body size, previous mating status and, exhibits no mating partner preference in successive mates. In addition, duration of mating, partner choice and female fertility were not influenced by male body size. The risk of predation did not alter mating behavior and duration of mating. However, females with interrupted mating at 30, 60, 120 and 240 minutesexhibited low egg viability (0, 3.1, 7.7, and 34%, respectively) compared to females with uninterrupted mating (338 to 671 minutes) that showed egg viability of 74.2%. The dissection of males of different mating status shows that P. nigrispinus exhibits continuous spermatogenesis and absence of a storage seminal vesicle. Spermatozoa were present in the lumen of the vas deferents independent of mating status. The spermatheca does not change size according to the duration of mating, although its duct exhibited elasticity with significant deformation after mating for 30 minutes. Females under uninterrupted mating exhibited spermathecal duct fully expanded and spermatheca filled with spermatozoa. Based on the results, the reproductive success of P. nigrispinus male is related to the duration of copula and female requires multiple mates to fill spermatheca with spermatozoa. However, the reproductive success of the species does not depend on males’ body size and their mating status. / A realização de múltiplas e longas cópulas por Podisus nigrispinus (Dallas) (Hem.: Pentatomidae) tem sido uma incógnita. Assim, este trabalho teve como objetivo investigar o comportamento reprodutivo de macho de P. nigrispinus. A cópula foi estudada em função do tamanho do macho, status prévio de acasalamento do macho e risco de predação (i). Também, foram analisados a espermatogênese e o efeito do tempo de cópula na transferência de espermatozóides para fêmeas (ii), bem como foram descritas a espermateca e estruturas relacionadas quanto à morfologia interna e modificações ocorridas durante a cópula (iii). A escolha da fêmea por machos virgens ou previamente acasalados foi estudada sob chance múltipla, parcial ou sem chance de escolha por machos grandes ou pequenos. O comportamento para o acasalamento e duração da cópula sob risco de predação [presença de Polistes versicolor Oliver (Hym.: Vespidae)] foi comparado com casais sem risco de predação. Fêmeas tiveram osucesso de fertilização e inseminação avaliado através da interrupção da cópula após 30, 60, 120 e 240 minutos. Em função do status de acasalamento, machos foram dissecados e investigados quanto a produção e transferência de espermatozóides para as fêmeas. Os resultados mostram que o acasalamento em P. nigrispinus independe do tamanho e status prévio de acasalamento dosmachos, além de não apresentar preferência de parceiro. A duração da cópula, escolha do parceiro e a fertilidade das fêmeas não sofrem influência do tamanho do macho. Também, o risco de predação não alterou o comportamento de acasalamento e duração de cópula. Fêmeas com cópulas interrompidas após 30, 60, 120 e 240 minutos apresentaram baixa viabilidade de ovos (0; 3,1; 7,7; e 34%, respectivamente), enquanto aquelas sem interrupção de cópula (338 a 671 minutos) obtiveram 74,2% de viabilidade de ovos. Também, foi observado que machos possuem espermatogênese contínua e ausência de vesícula seminal. A análise do ducto deferente mostra espermatozóides presentes no seu lúmen independente do status de acasalamento (machos virgens, 0h, 12h ou 24h após término da cópula). Em fêmeas, o volume da espermateca não sofre alteração com a cópula. Contudo, ocorre mudança de formato do ducto elástico da espermateca a partir de 30 minutos do início da cópula. Fêmeas que tiveram cópula sem interrupção apresentaram espermatecas completamente preenchidas com espermatozóides. Para P. nigrispinus o sucesso de inseminação e fertilização de fêmeas é dependente da duração da cópula e de múltiplas cópulas. Por outro lado, o sucesso reprodutivo não depende do tamanho do macho e de seu status de acasalamento.

Male mating tactics in the rose bitterling (Rhodeus ocellatus) and European bitterling (Rhodeus amarus)

The aim of this study was to investigate the basis to male mating decisions in two related species of bitterling: Rhodeus ocellatus and R. amarus. Bitterling have a resource-based mating system; females lay eggs in the gills of live freshwater mussels and males fertilize the eggs by releasing sperm into the inhalant syphon of the mussel. Male bitterling perform courtship behaviour and aggressively defend mussels in a territory from which they exclude other males. Using laboratory and field experiments it was shown that male aggressive behaviour is inherited through additive maternal genes. Male aggression is also influenced by the number of conspecific males encountered in competition for a mussel, and by the degree of clustering of mussels. Limited availability of mussels results in stronger selection on traits males use in mating context: hence they are more aggressive, larger and more colourful. The differences in mating behaviours in different environments may indicate a conflict between male dominance and female choice, but have not led to reproductive isolation. Resource availability during ontogenesis and male density during embryogenesis, however, do not exert an effect on male aggressive behaviour. Red carotenoid-based nuptial coloration functions as an inter- and intra-sexual signal and undergoes rapid variation in response to changes in mating context. Male bitterling do not modulate their courtship and aggressive behaviour in response to variation in female size, and their choice of mussel species is influenced by, and consistent with, female oviposition choice.

Os tratamentos utilizados foram: Controle (TC) dieta controle; Flushing (TF), dieta controle mais 300 g de fubá de milho; Flushing com nível 1 de própolis (TFP1), TF mais 8 mL de solução alcoólica de própolis/animal/dia; Flushing com nível 2 de própolis (TFP2), TF mais 12 mL de solução alcoólica de própolis/animal/dia. Foram utilizadas 16 fêmeas nulíparas vazias, sendo quatro por tratamento, distribuídas em experimento inteiramente casualizado, submetidas a um protocolo de sincronização de cio e posteriormente cobertas. Os animais foram estabulados por um período de adaptação de 10 dias, seguido de 30 dias de período experimental e alimentados duas vezes ao dia, às sete e 16 horas. Para determinar o perfil hematobioquímico, amostras de sangue foram coletadas a cada cinco dias, sendo a primeira coleta realizada com os animais em jejum e a segunda, duas horas após a alimentação das sete horas da manhã, durante 15 dias na fase pré cobrição e 15 dias na pós cobrição, para estimar os teores de ureia, proteínas totais, albumina, globulinas e a enzima AST. Utilizou-se ultrassonografia para confirmação de prenhez trinta dias após cobrição. Verificou-se que o tratamento com menor concentração de extrato etanólico de própolis Verde Alecrim apresentou potencial para diminuir consumo de MS e que o extrato etanólico de própolis demonstrou ser eficiente na diminuição do consumo de matéria seca e na concentração de ureia sérica. Os tratamentos TC, TF e TFP1 apresentaram mesma taxa de prenhez, superior ao TFP2, contudo o TFP1 resultou em maior taxa de partos gemelares. O tratamento com menor concentração de própolis reduziu a concentração de ureia sérica nos momentos jejum e duas horas após alimentação e na fase pós cobrição. Os teores dos metabólitos, em sua maioria, encontram-se dentro dos limites de valores de referência / The aim of this research was to evaluate diets with and without inclusion of propolis ethanolic extract Verde Alecrim on dry matter intake, metabolic profile and its relationships of reproductive aspects and to compare metabolic profile values with reference values established on the scientific literature. Treatments were: Control (CT), control diet; Flushing (FT), control diet plus 300 g/animal/day of corn meal; Flushing with level 1 of propolis (FTP1), FT plus 8 mL/animal/day of propolis ethanolic extract; Flushing plus level 2 of propolis (FTP2), FT plus 12 mL/animal/day of propolis ethanolic extract. Sixteen Santa Inês nulliparous ewes, empty, four per treatment, were assigned in a complete randomized design, in a factorial scheme and subjected to an estrus synchronization protocol and covered. The females were housed and the experiment consisted of an adjustment period of ten days, followed by an experimental period of 30 days (15 days for pre-mating and 15 days for post mating). Animals were fed twice a day at 7 AM and 4 PM. To determine the hematobiochemical profile, blood samples were collected at each five days in fasting and two hours after during 15 days in the pre-mating phase and 15 days in the post mating to estimate urea, total protein, albumin, globulins and AST enzyme contents. Ultrasound to confirm pregnancy thirty days following mating was used. It was found that treatment with lower concentrations of propolis ethanol extract Verde Alecrim presented the potential to decrease DM intake intake and that the ethanol extract of propolis has proved effective in decreasing the dry matter intake and concentration of serum urea. The CT, FT and FP1T provided the same pregnancy rate, superior to TFP2, however FP1T resulted in higher rate of twin births. Treatments with a lower concentration of propolis reduced urea content in serum in fasting and two hours after feeding and in the post-mating phase. Most of metabolite contents were within reference values, according to the literature
