Estas são utilizadas em formulações específicas para gatos para reduzir a densidade energética de alimentos, previner e/ou auxilar o tratamento de doenças gastrintestinais e metabólicas e também para promover saúde intestinal devido ao estímulo de microorganismos produtores de ácidos graxos voláteis. Contudo, pouca informação existe a respeito das consequências metabólicas do consumo de fibras por gatos, seu efeito na excreção de água e eletrólitos, produção de urina ou mesmo o efeito da absorção de ácidos orgânicos derivados da fermentação microbiana intestinal. Este estudo verificou a influencia da fibra dietetica no balanço hidro-eletrolitico e de macroelementos, produção de oxalato e equilibrio ácido-basico de gatos. Foram utilizados 18 gatos adultos e três dietas: BP0, um dieta controle sem adição de fibra; PB8, com adição de 8% de polpa de beterrada e PB16, com adição de 16% de polpa de beterraba. Após 15 dias de adaptação, o consumo de alimento e água foram monitorados e as fezes e urina foram coletadas durante 8 dias. Neste período foram determinados o balanço hídrico dos animais, densidade, pH e volume de urina e coletadas amostras para determinação da concentração urinária de cálcio, fósforo, cloreto, sódio, potássio, magnésio, enxofre, ácido úrico, citrato e oxalato, para calculo da supersaturação relativa para oxalato de cálcio (SSR OxCa). These are used in specific formulations for cats to reduce food energy density, avoid or assist the treatment of gastrointestinal and metabolic diseases and also to promote intestinal health due to stimulus of microorganisms to produce volatile fatty acids. However, there is little information about the metabolic consequences of fiber intake for cats, their effect on water and electrolytes excretion, urine production, or even the effect of absorption of organic acids derived from intestinal microbial fermentation. The present study verified the influence of dietary fiber in water and macroelements balance, oxalate production and acid-base balance of cats. It were used 18 adult cats and three diets: BP0, a control diet without adding fiber; BP8 with 8% beet pulp added and BP16, with 16% beet pulp added. After 15 days of adaptation, food and water consumption were monitored, and feces and urine were collected 8 consecutive days. Avaliação de biofilme de proteus mirabilis em modelo experimental de fluxo dinâmico /Camargo, Gabriela Maria Pavan de Arruda. January 2006 (has links)
Resumo: O objetivo do presente trabalho foi o de verificar a formação de incrustações e o bloqueio do cateter de Foley utilizando-se um modelo laboratorial de bexiga humana. Para tanto, foram utilizadas duas urinas artificiais de diferentes composições: a) urina AS composta por dez solutos em concentrações semelhantes as encontradas na urina humana de 24 horas, acrescida de gelatina; b) urina AT composta por 4 solutos também encontrados na urina humana, mas em concentrações maiores e suplementada com ovalbumina de galinha. Também foi utilizada a urina de 24 horas de três homens. As urinas contaminadas com o P. mirabilis foram bombeadas (0,5ml/min) para o frasco em que o cateter estava inserido até a oclusão do cateter. A Microscopia Eletrônica de Varredura (MEV) foi utilizada para verificar a presença de biofilme nos segmentos dos cateteres. Foi observado uma diferença significante no peso dos segmentos dos cateteres após a canalização das urinas AS, AT e UH contaminadas com o P. mirabilis vs a canalização das urinas sem o microrganismo (p<0,05). O tempo de bloqueio dos cateteres que canalizaram a urina AS vs urina AT e UH vs urina AT também foram diferentes (p<0,05). O tempo de bloqueio dos cateteres, o número de células viáveis presentes no inóculo inicial e no momento do bloqueio do cateter, e variação no peso dos segmentos dos cateteres após a canalização com as urinas sem a adição do P. mirabilis e contaminadas com o P. mirabilis não foram diferentes para as urinas AS, AT e UH. As três urinas examinadas mostraram a estabilização do P. mirabilis e a manutenção em 108UFC/ml bem como a formação de biofilme. Os cateteres que canalizaram a urina AS e UH apresentaram tempos semelhantes de bloqueio. Os cateteres que utilizaram a urina AT foram bloqueados mais rapidamente (p<0,05). Não houve alteração de peso dos segmentos dos cateteres quando testados com o P. mirabilis entre as urinas. / Abstract: The aim of the present work was to verify formation of encrustations and occlusion on Foley catheter using a laboratorial model of human bladder. Two artificial urines with different compositions were used: a) AS urine consisted by ten solutes in concentrations similar to those found in 24 hour human urine, added gelatin; b) AT urine consisted by four solutes, also found in human urine but in higher concentrations, and supplemented with chicken ovalbumin and UH 24 hour urine of three men. Urines contaminated with P. mirabilis were pumped (0,5ml/min) to flasks where the catheter was inserted reaching catheter occlusion. Scanning Electronic Microscopy (SEM) was used to check the presence of biofilms in catheter segments. The period of catheter occlusions after canalization was determined with the three urines, as well as the number of P. mirabilis viable cells present in the initial inoculum and in the end of the experiment. The period CFU/ml as well as biofilm formation. Catheters that canalized AS and HU urines showed similar occlusion periods. Glicosaminoglicanos (GAG) no urotélio e na urina possuem um efeito protetor contra desordens tais com urolitíase e cistite intersticial. Adicionalmente, foi demonstrado que hormônios sexuais femininos podem modular a excreção urinária de GAG (Maroclo M.V. et al. J Urol. 2005;173:1789-92). O presente estudo investigou se os anticoncepcionais hormonais orais combinados (AHOC) influenciam na excreção urinária de GAG. Amostras de urina foram doadas por mulheres jovens que utilizavam anticoncepcionais orais regularmente: EE + D (0,030 mg etinilestradiol + 3,0 mg drosperinona), n=9; EE + C (0,035 mg etinilestradiol + 2,0 mg acetato de ciproterona), n=9; e EE + G (0,020 mg etinilestradiol + 0,075 mg gestodeno), n=7. Controles foram 10 mulheres da mesma faixa etária não usuárias de AHOC. Valores de GAG urinário total das primeira e segunda metades do ciclo menstrual foram expressos como μg ácido hexurônico por mg de creatinina urinária. A proporção de GAG sulfatado foi determinada pela eletroforese em gel de agarose. Em contraste com o grupo controle, a excreção de GAG total na primeira e segunda metades do ciclo menstrual foi bastante similar nos grupos usuários de AHOC. Valores de excreção em todo o ciclo foram maior no grupo usuário de AHOC (p < 0,01), particularmente para o grupo usuário de EE + C, no qual os valores foram o dobro dos valores do controle (p < 0,005). Comparados com o controle, os 3 grupos usuários de AHOC produziram diferenças marcantes na composição urinária de GAG sulfatado, com diminuição de aproximadamente 50% no HS (p < 0,02) e DS (p < 0,02), e um aumento próximo de 100% no CS (p < 0,004). O padrão de excreção urinário de GAG está claramente alterado em mulheres que ingerem os AHOC estudados. Sendo assim, esta mudança na excreção constitui um efeito benéfico adicional dos AHOC, pois ela aumenta o potencial efeito protetor dos GAG contra desordens do trato urinário. / Glycosaminoglycans (GAG) on the urothelium and in the urine have protective effects against disorders such as urolithiasis and interstitial cystitis. Additionally, we have shown that female sex hormones may modulate urinary GAG excretion (Maroclo MV et al. J Urol 2005;173:1789-92). Here we investigated whether oral hormonal contraceptives (OC) affect urinary GAG excretion. Urine specimens were obtained from young women regularly taking the following OC: EE + D (0.030 mg ethinyl estradiol + 3.0 mg drospirenone), n=9; EE + C (0.035 mg ethinyl estradiol + 2.0 mg cyproterone acetate), n=9; and EE + G (0.020 mg ethinyl estradiol + 0.075 mg gestodene), n=7. Controls were from ten age-matched women not taking OC. Total urinary GAG from the first and second halves of the menstrual cycle was assayed as g hexuronic acid per mg urinary creatinine. Content of sulfated GAG species was determined by agarose gel electrophoresis. In contrast to controls, excretion of total GAG in the first and second halves of the menstrual cycle was quite similar in the OC groups. Excretion values for the whole cycle were higher in the OC groups (p < 0.01), especially for EE + C, in which the values were twice those from controls (p < 0.005). Compared with controls, the three OC produced marked changes in the composition of urinary sulfated GAG, with decreases of ~50% in HS (p < 0.02) and DS (p < 0.02), and a ~100% increase in CS (p < 0.004). The pattern of urinary GAG excretion is clearly changed in women taking commonly used OC. However, this altered excretion should constitute an additional beneficial effect of OC, as it enhances the protective effects of GAG against urinary tract disorders.

Ecological Sanitation in Uganda: Promotion through Demonstration Facilities and Potential for <i>Ascaris</i> Reduction by Free Ammonia Inactivation Using Stored UrineTrimmer, John Thomas 01 January 2015 (has links)
As Uganda works to transform itself into an industrialized, middle-income country in the coming decades, the country is faced with a number of problematic trends that could hinder this transition. High population growth and urbanization are quickly forcing small towns to deal with issues of limited space and the aesthetic conditions within sanitation systems, while declining soil fertility in surrounding rural areas calls into question the future nutritional security of the growing population. Ecological Sanitation (Eco-San) systems, which are designed to recover nutrients from human excreta, may help to address these trends.
Improved sanitation coverage in Uganda is currently estimated to be 34%, with most people using either improved or unimproved pit latrines. Eco-San systems, especially Urine-Diverting Dry Toilets (UDDTs, also referred to as composting toilets), have been promoted in the country, but uptake has been slow. Additionally, while UDDTs generally treat human feces to a greater degree than pit latrines and composting toilets (another type of Eco-San system), concerns have been raised as to the inactivation of environmentally persistent pathogens, such as Ascaris lumbricoides eggs. This research focused on two potential solutions to the issues of effective promotion and Ascaris inactivation, evaluating them in the context of Kalisizo, a small town in southern Uganda.
Demonstration facilities have been reported to effectively convince local stakeholders of the benefits and advantages of UDDTs, thereby increasing long-term uptake of the technology in the surrounding community. However, an unresolved question concerns whether these facilities should be installed in household or institutional settings. The initial effects of demonstration facilities constructed at local primary schools in Kalisizo were evaluated by assessing local knowledge and attitudes regarding UDDTs, both before installation and after several months of operation, through focus group discussions and key informant interviews. In general, this promotion strategy proved to be successful. After installation, students exhibited a marked increase in knowledge regarding these facilities and their benefits, and opinions were strongly positive. These changes were seen in users of the facilities as well as non-users, and students expressed clear acceptance of using the products of the toilets to fertilize crops. The introduction of an improved sanitation system at the schools also appears to have sparked other improvements related to sanitation and hygiene. In the future, it is likely that students will be compelling representatives for UDDTs within their households and communities.
Regarding the treatment of persistent pathogens, previous work has demonstrated that the elevation of free ammonia levels to levels that can inactivate Ascaris eggs can be achieved through the urea addition. In this research, use of stored urine as an ammonia source for treatment of fecal products from UDDTs in Uganda was investigated. Mixtures of stored urine, fecal products from UDDTs, and wood ash were prepared, and treatment conditions (pH, temperature, ammonia concentration) were compared to the results of previous Ascaris inactivation studies to determine whether this strategy would be a feasible and effective treatment alternative. Results indicated that a volumetric mixture containing two parts stored urine and one part fecal products could provide 4-log1¬0 inactivation of Ascaris eggs after five months of indoor storage or after three months of outdoor storage. This strategy could improve the safety of recovered products while maintaining their agricultural value. Social acceptance of the treatment system appears to be possible with proper education efforts, and a cost comparison showed that this system may be more economically favorable than typical double-vault UDDTs.

Urine Diversion & Reuse in Australia : A homeless paradigm or sustainable solution for the future?Cordell, Dana January 2006 (has links)
Diverting urine from faeces or mixed wastewater and reusing it to fertilize crops, is a traditional method used in Asia. It is also a contemporary approach to sustainable nutrient and water management in Scandinavia and other parts of Europe. Urine diversion and reuse is a proven socio-technical system that has significant potential benefits on both a local and global scale, such as recirculating scarce plant nutrients like phosphorus back to agriculture, reducing eutrophication of waterways and improving water and sanitation systems. This thesis explores the nature of these benefits in Australia and the global context and what barriers would need to be overcome if a urine diversion and reuse system were implemented in Australia to achieve significant environmental benefits. These questions are investigated through stakeholder interviews in Sweden, to identify the ‘lessons learnt’ from the Swedish experience with urine diversion and reuse, and, through interviews with relevant stakeholders in Australia to identify possible barriers and opportunities, costs and benefits, and roles and responsibilities in the Australian context. Findings from both the stakeholder interviews are triangulated with other sources of knowledge, such as the literature, personal communications and a qualitative assessment of costs and benefits. This thesis found that while urine diversion is likely to benefit the Australia situation and warrants further research, these benefits are fragmented and spread across a range of discourses and separate institutions. Its acceptance and effective introduction into Australia might therefore be challenged by its lack of a single obvious organisational home. To overcome this and other identified challenges, several recommendations are made. For example, an Australian demonstration trial of urine diversion and reuse is recommended where clear drivers and opportunities exist, such as: in new developments adjacent to agricultural land; in regions where algal blooms are a critical problem and are predominantly caused by municipal sewage discharges; and where synergies with waterless urinals are being considered for water conservation value. This thesis does not promote urine diversion and reuse as the ‘silver bullet’ to Australia’s water and nutrient problems, however it does recommend that it be considered on an equal basis next to other possible options. For example, if reducing nutrient loads on receiving water bodies is a key objective, then a cost-effective analysis of urine diversion and reuse, compared to other options to reduce nutrient loads, could be undertaken, ensuring all relevant costs and benefits to the whole of society are included in the analysis.

Ecological Sanitation (Ecosan) and the Kimberley ExperienceJonah, Albert January 2007 (has links)
The Hull Street Integrated Housing Project, in Kimberley, is one of the projects supported by the Swedish International Development Cooperation Agency, Sida, in South Africa. The vision of the project is to provide low cost housing for the people of Kimberley. As a way of ensuring sustainability, the project adopts the Ecological sanitation (Ecosan) approach where urine and faeces are separated from the source. The concept of Ecosan is new to many people around the world. To make the concept workable and acceptable effective implementation strategies are required. At the Hull Street, after the first of the four phases 144 unit houses have been completed all fitted with the UDS. Urine from the UDS as well as the greywater from the kitchen and bathroom are connected to infiltrate into the ground. This arrangement is called the “quick-fix”. The faeces from the houses are sent to the compost yard for composting so that the residents could use the compost in their gardens. This study which involves interview with some selected workers and residents in Hull Street focuses on the modus operandi of the Ecosan unit of the Hull Street project with special emphasis on the methods of human excreta disposal and education strategies.

Procurou-se avaliar parâmetros fisiológicos de antas (Tapirus terrestris) de vida livre capturadas no Parque Estadual Morro do Diabo, estado de São Paulo, Brasil, entre 1996-2008 pelo Programa Anta Mata Atlântica. Paralelamente, avaliou-se a exposição desses animais a patógenos de interesse em saúde pública e saúde animal. Nesse período foram obtidas amostras de 35 animais. Os valores médios de hemograma e bioquímica sérica foram comparados com os valores de referência do ISIS, através do uso do teste t de Student. Os resultados médios de todos os parâmetros avaliados estavam dento do intervalo de variação descrito para a espécie em cativeiro. Títulos sorológicos incluíram Encefalite Equina Oeste (n=1 animal), Encefalite Equina Leste (n=6), Leptospira interrogans sorovar pomona (n=6), Leptospira interrogans sorovar autumnalis (n=1), Leptospira interrogans sorovar hebdomadi (n=1), Língua Azul (n=5), Rinotraqueíte Infecciosa Bovina (IBR) (n=1). Não foi detectada a presença de anticorpos para Febre Aftosa, Doença de Aujeszky, Parvovirose Suína, Diarréia Viral Bovina, Leucose Bovina, Estomatite Vesicular, Anemia Infecciosa Equina e Brucelose. Todas as capturadas apresentavam alta infestação de carrapatos, identificados como Amblyomma brasiliense, Amblyomma cajennense, Amblyomma coelebs, Amblyomma ovale, Amblyomma naponense, Haemaphysalis juxtakochi e Rhipicephalus (Boophilus) microplus. Alguns animais apresentaram Tunga penetrans e mosca-do-chifre (Haematobia irritans). Oito amostras de urinas de antas de vida livre foram analisadas e descritas pela primeira vez. Os swabs das cavidades naturais apresentaram Staphylococcus aureus em todos os orifícios. Escherichia coli em todos os orifícios, com exceção da orelha. Proteus sp. em todos os orifícios, com exceção do prepúcio. Ânus e vagina presentaram as mesmas bactérias, já prepúcio e ânus se distinguiram pelo Staphylococcus saprophyticus. / It was evaluated physiologic parameters of wild Lowland tapirs (Tapirus terrestris) captured in Morro do Diabo State Park, São Paulo state, Brazil, between 1996 2008 by Atlantic Rainforest Lowland Tapir Program. Together, it was evaluated the expose of these animals to pathogens of public health and animal health interest. During this time it was collected samples from 35 animals. The medium value of blood cells and serum biochemistry were compared with reference values of ISIS, with the t test of Student. All medium values of parameters evaluated were inside of regular range described for the specie in captivity. Serologic titles include West Equine Encephalitis (n= 1 animal), East Equine Encephalitis (n=6), Leptospira interrogans sorovar pomona (n=6), Leptospira interrogans sorovar autumnalis (n=1), Leptospira interrogans sorovar hebdomadi (n=1), Bluetongue (n=5), Infectious Bovine Rhinotraqueitis (IBR) (n=1). It was not detected antibodies to Foot and Mouth Disease, Aujeszky\'s disease, Porcine Parvovirosis, Bovine Viral Diarrhea, Bovine Leucosis, Vesicular Stomatitis, Equine Infectious Anemia and Brucelosis. All captured animals had high level of infestation of ticks, identified as Amblyomma brasiliense, Amblyomma cajennense, Amblyomma coelebs, Amblyomma ovale, Amblyomma naponense, Haemaphysalis juxtakochi and Rhipicephalus (Boophilus) microplus. Some animals had Tunga penetrans and mosca-do-chifre (Haematobia irritans). Eight samples of urine from wild tapirs were analyzed and described first time. The swabs from natural cavity showed Staphylococcus aureus in all of them. Escherichia coli in all cavities, except ears. Proteus sp. in all cavities, except prepucial. Anus and vagina showed the same kinds of bacteria, but prepucial and vagina show the same bacteria, and prepucial and anus to be different because Staphylococcus saprophyticus.

Avaliação de biomarcadores da exposição humana à fumonisina B1 nos alimentos em municípios dos estados de São Paulo e Santa Catarina, Brasil / Evaluation of biomarkers of human exposure to dietary fumonisin B1 in cities from São Paulo and Santa Catarina states, BrazilKeliani Bordin 25 February 2015 (has links)
A fumonisina B1 (FB1) e uma micotoxina produzida pelo metabolismo secundário de espécies de Fusarium, principalmente F. verticillioides e F. proliferatum, os quais contaminam diversos alimentos antes e apos o processamento, sobretudo o milho e derivados, gerando graves problemas para a Saúde Pública e a qualidade dos alimentos. O objetivo deste trabalho foi avaliar a exposição humana a FB1 presente nos alimentos através da estimativa de ingestão da toxina na dieta e da análise de diferentes biomarcadores presentes em amostras de sangue, urina e cabelo. Além disso, foram investigados os efeitos da toxina através da avaliação de ácido fólico presentes em alimentos e em soro, e os níveis de uréia e creatinina presentes em soro. O estudo foi realizado em dois municípios dos Estados de São Paulo e Santa Catarina, cujos respectivos voluntários foram categorizados como de baixo consumo de derivados de milho (Grupo A, voluntários de Pirassununga/SP) e de alto consumo de derivados de milho (Grupo B, voluntários de Erval Velho/SC). As amostras de alimentos do Grupo A (Pirassununga/SP) foram fornecidas pelos voluntários (n=100) nos meses de Junho/2011, Setembro/2011, Dezembro/2011 e Marco/2012. Os voluntários do Grupo B (Erval Velho/SC) (n=20) forneceram amostras de alimentos no mês de Abril/2012. Em cada grupo, uma lista com 20 alimentos a base de milho foi entregue aos voluntários, para fornecimento de amostras daqueles disponíveis em suas respectivas residências em cada mês de amostragem, totalizando 122 amostras de derivados de milho no Grupo A e 17 amostras no Grupo B coletadas durante o estudo. Adicionalmente, aplicou-se um Questionário de Frequência Alimentar (QFA) e um Inquérito Recordatório de 24 horas (QIR - 24 h) no momento das coletas de amostras. Em cada mês de amostragem de alimentos, foram coletadas amostras de sangue, urina (somente Grupo A) e cabelo dos voluntários, sendo as amostras armazenadas a -20ºC (urina e cabelo) ou -80ºC (sangue) até o momento das análises. As amostras de alimentos foram submetidas a análise de FB1, sendo que as de farinha de milho foram também analisadas quanto ao teor de ácido fólico. Ambas as análises foram feitas através de cromatografia líquida de alta eficiência (CLAE). Em soro, foram avaliadas a relação esfinganina/esfingosina (Sa/So), resíduos de FB1, ácido fólico, uréia e creatinina. Em urina, foram analisados os níveis de FB1, creatinina para correção do volume urinário e a relação Sa/So. Em cabelo, foram analisados os resíduos de FB1 através de CLAE acoplada a espectrometria de massas. Todos os métodos de análise foram submetidos a procedimento de otimização e validação intra--laboratorial. A incidência de FB1 nos alimentos foi, em média, 72% (n=122) nas amostras do Grupo A (Pirassununga/SP) e 35% (n=17) no Grupo B (Erval Velho/SC). Os maiores níveis foram encontrados em amostras de pipoca provenientes do Grupo B, com uma amostra excedendo o limite de tolerância estabelecido no Brasil (2,500 µg kg-1). A ingestão diária provável média (IDPM) de FB1 no Grupo A foi de 63,3 ng kg-1 peso corpóreo (p.c.) dia-1, que corresponde a 3,1% da ingestão provisória máxima tolerável (IPMT) recomendada para fumonisinas (2.000 ng kg-1 p.c. dia-1). A IDPM do Grupo B apresentou uma média de 190,1 ng kg-1 p.c. dia-1 o que corresponde a 9,5% da IDMT. As concentrações de ácido fólico nas amostras de farinha de milho variaram de < 0,3 µg kg-1 (limite de quantificação do método) a 1.705 µg kg-1, com média de 713 ± 435 µg kg-1. Somente uma amostra apresentou nível de ácido fólico acima do valor mínimo estabelecido pela ANVISA. Em urina, a incidência de FB1 foi de 33,4% (n=251), com níveis médios de 3,19 ± 3,15 ng mg-1 de creatinina. Não houve correlação (P>0,05) entre as concentrações de FB1 na urina e nos alimentos. Os níveis de esfinganina foram mais elevados em mulheres, com 25,0% (n=116) de amostras positivas, em comparação à urina de homens, 10,4% (n=96). A relação Sa/So apresentou em média 0,91, 0,77 e 0,89 para urina de mulheres, homens e em combinação, respectivamente. Em soro, os níveis de esfingosina foram em média 2,48 ng mL-1 para o Grupo A e 5,01 ng mL-1 para o Grupo B. A relação Sa/So variou de 0,06 a 3,19 com média de 0,79 para o Grupo A e 0,78 para o Grupo B. Embora tenha havido correlação positiva (r=0,574, P<0,05) entre a relação Sa/So no soro e os dados de consumo de milho e derivados obtidos no QIR-24 h, não foram observadas correlações (P>0,05) entre a ingestão de FB1 e a relação Sa/So na urina ou soro. A concentração de ácido fólico no soro variou de 6,7 a 24,0 ng mL-1 (média de 13,4 ± 5,4 ng mL-1), com ambos os grupos (A e B) apresentando resultados dentro dos valores de referências. Não foram observados níveis detectáveis de FB1 nas amostras de soro. No entanto, FB1 foi detectada em 4 amostras de cabelo humano (7,2%) dos Grupos A e B, cuja concentração média foi de 21,3 ± 12,1 ng g-1. Em síntese, os resultados obtidos nas análises de biomarcadores de FB1 no presente trabalho estão de acordo com os valores de IDPM encontrados, indicando que a exposição a FB1 nas populações estudadas não representa um risco a saúde. / Fumonisin B1 (FB1) is a mycotoxin produced by the secondary metabolism of Fusarium species, mainly F. verticillioides and F. proliferatum, which contaminates foods before and after processing and causes serious problems to public health and food quality. The aim of this study was to evaluate the human exposure to FB1 in food by means of estimated intake of toxin in the diet, and analysis of different biomarkers in serum, urine and hair. In addition, folic acid in food and blood as well urea and creatinin in serum were investigated to evaluate the toxin effects. The study was conducted in two cities of Sao Paulo and Santa Catarina States, where the respective volunteers were categorized as low-consumers of corn products (Group A, volunteers from Pirassununga/SP) and high-consumers of corn products (Group B, volunteers from Erval Velho/SC). Food samples from Group A (Pirassununga/SP) were provided by volunteers (n=100) in June/2011, September/2011, December/2011 and March/2012. The volunteers from Group B (Erval Velho/SC) (n=20) provided food samples in April/2012. In each group, a list of 20 corn products was given to volunteers, to allow them to check and collect the food items available in their homes at each sampling time. The total number of samples of corn products provided by the volunteers were 122 and 17 in Group A and Group B, respectively. Addicionally, a Food Frequency Questionnaire (FFQ) and a 24-Hours Dietary Recall Questionnaire (24h-DRQ) were applied by the time of sample collections. In each month of food samples collection, samples of blood, urine (only Group A) and hair from the volunteers were collected and storage at -20ºC (urine and hair) or -80ºC (blood) until analysis. Food samples were submitted to determination of FB1, and corn meal samples were also evaluated for folic acid levels. Both analysis were performed by high performance liquid chromatography (HPLC). In serum, analyses included sphinganine/sphingosine ratio (Sa/So), FB1 residue, folic acid, urea and creatinine. In urine, the levels of FB1, creatinine to correct urinary volume and Sa/So ratio were evaluated. In hair, FB1 residues were analysed by HPLC coupled to mass spectrometry. All the analytical methods were submitted to optimization and intra-laboratorial validation procedures. The mean incidences of FB1 in corn products were 72% (n=122) in samples of Group A (Pirassununga/SP), and 35% (n=17) of Group B (Erval Velho/SC). The higher levels were found in popcorn from Group B, with one sample exceeding the tolerance limit established in Brazil (2,500 µg kg-1). The mean probable daily intake (PDIM) of FB1 in Group A was 63.3 ng kg-1 body weigh (b.w.) day-1, which corresponds to 3.1% of provisional maximum tolerable intake (PMTDI) recommended for fumonisins (2,000 ng kg-1 b.w. day-1). PDIM of Group B was 190.1 ng kg-1 b.w. day-1, which represents 9.5% of PMTDI. Folic acid levels in corn meal ranged from < 0,3 µg kg-1 (quantification limit) to 1.705 µg kg-1, with a mean of 713 ± 435 µg kg-1. Sa/So ratio ranged from 0.06 to 3.19 with a mean of 0.79 to Group A and 0.78 to Group B. Although a positive correlation (r=0.574, P<0.05) was found between Sa/So ratio in serum and corn consumption data obtained by 24h-DRQ, no correlation was observed (P>0,05) with FB1 intake and Sa/So ratio in urine or serum. Folic acid concentration in serum ranged from 6.7 to 24.0 ng mL-1 (mean of 13.4 ± 5.4 ng mL-1), with both groups (A and B) presenting levels within the reference valuies. There were no detectable levels of FB1 in serum samples. However, FB1 was detected in 4 human hair samples (7.2%) of Groups A and B, at a mean concentration was 21.3 ± 12.1 ng g-1. In summary, the results obtained in the analyses of FB1 biomarkers in the present study are in agreement with the PDIM values found, hence indicating that FB1 exposure in the populations studied do not represent a health concern.

Previous issue date: 2015-12-09 / A presença de diferentes substâncias genotóxicas, como os produtos do decaimento do 210Pb, um elemento amplamente encontrado no cigarro, pode causar danos genéticos e contribuir para evolução de neoplasias malignas, o que evidencia a necessidade de monitoração constante dos fumantes. Bioindicadores, a exemplo de micronúcleos (MN’s) são largamente empregados nos estudos das ações de agentes genotóxicos em seres humanos. Normalmente, em grupos formados por fumantes, são utilizadas amostras da mucosa bucal para avaliação da frequência de micronúcleos. No entanto, raros são os estudos que procuram utilizar como matriz amostras de urina para essas determinações. O presente trabalho avaliou a frequência de micronúcleo em células esfoliativas na urina, bem como os teores de Chumbo-210 em urina coletada durante 24 horas de indivíduos fumantes e não fumantes, com o objetivo de avaliar a utilização desta matriz como indicadora da suscetibilidade a danos genéticos em voluntários fumantes. Foram coletadas amostras de urina de 30 voluntários (15 não fumantes e 15 fumantes). Para as análises de micronúcleo em urina foram obtidas células esfoliativas através de uma série de centrifugações e lavagens com NaCl a 0,9%. A fixação e coloração empregadas foram metanol/ácido acético e Giemsa, respectivamente. Para determinar as concentrações de 210Pb, utilizou-se a técnica da resina trocadora de íons. Nesse caso, o 210Pb foi precipitado na forma de PbCrO4. O detector Canberra Tennelec S5E foi utilizado para determinar o 210Pb. As concentrações de 210Pb na urina dos fumantes variaram de 49,7 a 158,65 mBq.L-1, enquanto os não fumantes apresentaram valores abaixo de 45 mBq (LD). Nesta pesquisa, os resultados relacionados ao 210Pb sugerem que fatores como a idade, tempo de tabagismo e quantidade de cigarros não exerceram influência estatisticamente significante sobre os valores de excreção do 210Pb, entretanto o hábito tabagista contribui para o aumento das concentrações encontradas na urina. Foi encontrada uma frequência média para micronúcleo em não fumantes de 0,0036 ± 0,001 e, em fumantes foi de 0,0068 ± 0,002. Os resultados indicam um aumento expressivo da frequência de micronúcleos em células esfoliativas de indivíduos fumantes em relação aos não fumantes, sendo este aumento estatisticamente significativo. A análise estatística dos componentes principais (ACP) sugere que a quantidade de cigarros está relacionada com os micronúcleos, no entanto, não houve correlação estatisticamente significante com as concentrações de Chumbo-210. Os resultados obtidos sugerem que a urina pode ser utilizada como matriz biológica para a avaliação da suscetibilidade a danos genéticos ocasionados pelos compostos presentes no cigarro, comportando-se como uma ferramenta eficiente para a monitoração humana. / The presence of different genotoxic substances in cigarette smoke can cause genetic damage and contribute to development of malignancies, which highlights the need for constant monitoring of smokers. Biological indicators, such as the frequency of micronuclei (MN) are widely used to study the action of genotoxic agents in human beings. Usually in groups formed by smokers, the buccal mucosa samples are used to assess the frequency of micronuclei. However, few studies that seeks to use as a template urine samples for these determinations. The aim of this study was to evaluate the frequency of micronuclei in exfoliated cells as well as Lead-210 levels in 24-hour urine from smokers and nonsmokers, in order to evaluate the use of this matrix as an indicator of susceptibility to genetic damage in smokers volunteers. Because it is an array easy to obtain and allow an analysis of various elements were collected urine samples from 30 volunteers (15 smokers and 15 non-smokers). For the analysis of micronucleus, exfoliative cells from urine were obtained by a series of centrifugations and washings with NaCl 0.9%. Carnoy I fixation and Giemsa staining were employed, respectively. To determine the 210Pb, we used the technique of ion exchange resin. In this case, the 210Pb was precipitated in the form of PbCrO4. 210Pb beta counts were conducted in a Canberra Tennelec S5E detector with a detection limit of 45 mBq. The results indicated that the urine is an excellent matrix for assessing the degree of exposure to lead-210, especially for smokers, the group of most interest to monitoring. The non-smokers present values above 45mBq and Smokers have shown higher values for 210Pb in urine that ranged from 49.7 to 158.65 mBq.L-1. In this research, the results related to this radionuclide suggest that factors such as age, smoking time and number of cigarettes have little influence on the 210Pb excretion values, though the tobacco habit contributes to the concentrations found in the urine. The average rate for micronucleus found in nonsmokers was 0.0036 ± 0.001 and 0.0068 ± 0.002 in the smokers. The results showed a significant increase in micronuclei frequency in exfoliated cells of smokers compared to nonsmokers, which is statistically significant increase. Statistical analysis also suggest that the number of cigarettes are related to the micronuclei, however there is no statistical correlation with the concentrations of Lead-210. The results suggest that urine can be used with biological matrix for assessing genetic susceptibility to damage caused by the present compounds on the cigarette, behaving as an efficient tool for human monitoring.

Previous issue date: 2010-02-19 / Conselho Nacional de Pesquisa e Desenvolvimento Científico e Tecnológico - CNPq / This study was carried out at the Experimental Station of the Agronomic Institute of Pernambuco (IPA) in Itambé, dry Forest Zone of Pernambuco. Two grazing trials were performed. The first one evaluated nutrient deposition via animal excreta on grazed Pennisetum purpureum Schum. pastures; the second one characterized the soil and theroot system of two tropical grasses under different grazing intensities. Pastures of Pennisetum purpureum Schum clone IRI 381 were submitted to different post-grazing stubble heights (40, 80, and 120 cm) and Brachiaria decumbens Stapf. pastures under different stocking rates (2, 4, and 6 AU/ ha). A rotational stocking was applied with grazing cycle of 35 days ( 32 days resting period and three days of grazing period) in the rainy season and 70 days (67 days resting period and three days of grazing period) in the dry season. Crossbred Holstein-Zebu cattle were used as experimental animals. A complete randomized blocks design was used, with three replications per treatment. The amount of excreta returned to the pasture and chemical composition of urine and feces were evaluated in seven different sampling periods. In addition, soil fertility, soil physics, and root system variables were also measured at different soil layers. The average concentration of minerals in feces, varied according to the period of assessment with average values of 16,0; 1,7; 0,20, 9,67, 5,77, and 5,24 g/kg for N, P, Na, K, Ca and Mg, respectively. Mineral concentrations in urine were 2,99; 1,11; 34,86; 0,29 and 0,23 g/L of N, Na, K, Ca and Mg, respectively. The post-grazing stubble heights affected only the concentration of N and Mg in the dung, where the treatment with 40 cm postgrazing stubble height presented higher concentration of these nutrients. Return of nitrogen through dung and urine contributed with 506 g N/ha.day, 47% of this via urine and 53% via dung. The N, K and Na (89%, 95% and 96%) returned mainly via urine; Ca and Mg (78 e 81%) returned mainly via dung. Phosphorus was found only in dung. The return of N was similar in feces and urine. Characteristics of the soil and root system were not affected by post-grazing stubble heights and stocking rates. The top soil layer (0-20 cm) showed greater root length (59 and 54cm/200 g soil), root length density (0,37 and 0,33 cm/cm3), and root biomass (2,04 and 2,84 T/ha), and carbon storage (61 and 91 Mg.ha-1), respectively to elephant grass and Brachiaria. The samelayer (0-20) presented higher soil P concentration regardless of the grass. Brachiaria accumulated more carbon in the soil due to its greater capacity of soil coverage, with greater C stock at the 0-20 cm soil layer. The root system of both species were mainly in the 0-20 cm soil layer, however, B. decumbens presented greater root biomass in the deeper soil layers (40-100 cm) compared to P. purpureum. Based on the observed results, nutrient return via animal excreta is of great importance for maintaining pasture ecosystem, mainly due to deposition of N, P, and K. Future researches aiming to improve excreta distribution on pasture surface must be performed, maximizing nutrient return and reducing application of chemical fertilizers. / Este estudo foi realizado na Estação Experimental do Instituto Agronômico de Pernambuco (IPA), no município de Itambé, Zona da Mata Seca de Pernambuco, e objetivou avaliar dois ensaios, sendo o primeiro realizado através da avaliação da deposição de nutrientes via excreta animal em pastagens de Pennisetum purpureum Schum. e o segundo, através da caracterização do solo e do sistema radicular em pastagens de gramíneas tropicais manejadas sob diferentes intensidades de pastejo. Para tal, foram avaliadas pastagens de capim elefante clone IRI 381, submetidas a diferentes alturas de resíduo pós pastejo (40, 80 e 120 cm) e Brachiaria decumbens sob diferentes lotações animais (2, 4 e 6 UA/ha), com lotação intermitente e ciclo de pastejo de 35 dias, na época chuvosa (32 dias de descanso e três dias de ocupação) e, 70 dias, na época seca (67 dias de descanso e três dias de ocupação). Foram utilizados bovinos mestiços holandês/zebu como animais pastejadores. O delineamento experimental utilizado foi de blocos casualizados, com três repetições. Foram realizadas avaliações referentes à quantidade excretada e composição química das fezes e urina, em sete períodos de avaliação, além de análise de fertilidade e física do solo, e características do sistema radicular de gramineas tropicais em diferentes profundidades. A concentração média de minerais nas fezes variaram de acordo com o período de avaliação, sendo observados valores médios de 16,0; 1,7; 0,20; 9,67; 5,77 e 5,24 g/kg para N, P, Na, K,Ca e Mg, respectivamente. A urina apresentou concentrações de 2,99; 1,11; 34,86; 0,29 e 0,23; g/L de N, Na, K, Ca e Mg, respectivamente. As alturas de resíduo pós pastejo influenciaram apenas a concentração de N e Mg nas fezes, onde o tratamento de 40 cm apresentou maior concentação desses nutrientes. O retorno de nitrogênio por meio das fezes e urina contribuiu, a cada dia de pastejo, com 506 g N/ha.dia, sendo 47% deste proveniente da urina e 53% via fezes. O K e Na (95% e 96%) retornaramprincipalmente via urina; Ca e Mg (78 e 81%) foram mais presentes nas fezes dos animais, já o P foi encontrado apenas nas fezes. O retorno de N foi semelhante nas fezes e urina (53 e 47%). As características do solo e sistema radicular não foram influenciadas pelas alturas de resíduo pós pastejo e lotações animais. A camada superficial do solo (0-20 cm) apresentou maior comprimento (59 e 54 cm/200g de solo), densidade (0,37 e 0,33 cm/cm3) e biomassa radicular (2,04 e 2,84 Mg/ha), bem como estoque de carbono (61 e 91 Mg.ha-1), respectivamente para o capim elefante e a braquiária. A braquiária acumulou maior quantidade de carbono no solo, em virtude de sua maior capacidade de cobertura do mesmo, sendo esse estoque maior nas camadas superficiais. O sistema radicular de ambas as espécies concentra-se principalmente na camada de 0-20 cm, todavia, B. decumbens apresentou maior biomassa radicular em camadas mais profundas do solo (40 – 100 cm) quando comparada ao P. purpureum. Diante dos resultados observados no presente trabalho, o retorno de nutrientes via excreta animal é de grande importância para a manutenção do ecossistema daspastagens, principalmente relacionado à deposição do N, P e K. Futuras pesquisas objetivando melhorar a distribuição dos excrementos no pasto devem ser realizadas, de forma a maximizar o retorno dos nutrientes depositados, gerando economia na aplicação de fertilizantes químicos.
