Windows Azure Platform is product of the Microsoft company aimed to host web applications. With this product, Microsoft is reacting on the new trend in IT -- cloud computing. This diploma thesis deals with all the aspects of developing and running PHP applications on Windows Azure Platform. Thesis is divided into two parts -- theoretical and practical. In the theoretical part, Windows Azure Platform is described in detail. The description consists of it's main parts, position in the world of cloud computing, price and main competitors. In the practical part, migration of school PHP application developed during 4IT445 course from LAMP platform to Windows Azure Platform is described. Except the migration itself, the benefits of new platform to application are described. In the thesis conclusion, development and running applications on both platforms is compared. The target of the thesis is to write first complete Czech text about running PHP applications on Windows Azure Platform and to help PHP developers to chose which way to go.

The theoretical part is focused on lights, notions and calculations in the area of light technology, basic lightning sources and lamps. This work is also about the principle of lightning measuring. The main aim of the work is design of a new artificial lightning system in a High school agriculture in Prerov. At first, the control measuring of existing lightning was done which is mentioned in this work. From the results of control measuring is clear, that existing lighting system in classroom does not suit requirements, which are mentioned in the valid technical norm. Design of a new lightning system meets the requirements in all of the rooms in the building of school. Design of lightning system was done in two variants. The first variant uses lamps with fluorescent tube light sources. The second variant uses lamps with LED light sources. Also is here evaluated financial difficulty of possible installation and comparation of each variants with respect energetic difficulty.

This master thesis deals with design of injection molding technology for plastic lamp shade made of polycarbonate labelled Lexan 925. The injection is performed into injection mold with hot runner system. Initially there is a general study related to plastic materials and injection molding technology. Further the study continues with technological solution of production of the lamp shade. The designed injection mold is two-plate, double cavity mold with two hot nozzles and is equipped with a thread-unscrewing machine. This thesis contains part of the documentation and item list of the injection mold, along with technical-economical evaluation of the developed solution.

Advanced Detection Methods of Genomic Barcodes for Genotyping Escherichia coli LibrariesEger, Nicole January 2021 (has links)
Pooled cell strain libraries are a powerful tool allowing to investigate the influence of genetic modifications on phenotypes in high throughput single-cell assays. To link the genotype to phenotype in each cell of the library, unique 20 base pairs (bp) long barcodes are used to allow in situ genotyping after phenotyping via fluorescence microscopy. In previous studies, these barcode sequences were expressed from high copy number plasmids resulting in a high number of targets for detection via fluorescence in situ hybridization (FISH) and thus, a strong readout signal. However, constant selection pressure must be applied on the cells to maintain the foreign plasmid DNA which may influence the phenotype. Inserting unique barcodes on the chromosome ensures stability of the construct which is required for some genomic library applications. However, the low copy number of the barcode sequence often requires an additional step of DNA amplification for efficient detection. In this study, two methods for barcode amplification were investigated. First, amplification from the double stranded DNA upon binding of peptide nucleic acids and subsequent amplification via rolling circle amplification (AmPPR). Second, amplification from genomic DNA or cDNA via loop-mediated isothermal amplification (LAMP). Whereas the AmPPR approach remained unsuccessful, chromosomal barcode sequences were successfully amplified in situ via LAMP and subsequently detected using FISH. I show that LAMP can potentially be a quick, specific, and elegant amplification technique for in situ genotyping in microfluidic devices. However, nonspecific amplification and partly nonspecific readout signals when using LAMP remain a problem and need to be further investigated before implementing this method on pooled libraries.

Ultraviolet (UV) light has been used for disinfection purposes for over 100 years. Irradiation by UV light is a method to disinfect surfaces in order to prevent microbiological growth. At Tetra Pak this is of great importance as they are manufacturer of filling machines. Those filling machines must ensure a certain level of sterility on all packages produced. The irradiation process can be simulated using Geant4 which is a software package that tracks particles through matter. The simulation model used today does not consider the absorption of photons inside of medium-pressure UV lamps. By understanding the absorption that takes place in the lamp, one can quantify how changes in the design would impact the emitter output. In this master's thesis, the aim is to develop a model that can describe the interaction of photons with a medium-pressure UV lamp. An absorption model was suggested and developed with the assumption of local thermodynamical equilibrium and existing Hg radiative data. A simulation including the collision process in Geant4 was used. In this collision process the non-radiative transition probabilities were assumed to be the same as that of the radiative, this was done in order to demonstrate how it can be done. It resulted in collisions populating other states allowing more transitions to be present in the final output spectrum. The collision process and a method for computing the Einstein's emission coefficient with the software package General Relativistic Atomic Structure Package is proposed as future work. / I över 100 år har ultraviolet (UV) ljus använts till desinficering. UV bestrålning är en metod för att desinficera ytor med målet att förhindra mikrobiologisk tillväxt. För Tetra Pak som är ledande inom tillverkning av fyllmaskiner är det extra viktigt. Förpackningarna inuti fyllningsmaskinerna måste garantera en viss nivå av sterilitet för alla förpackningar. Dagens simuleringar av medeltrycks UV lampa utförs i Geant4 som är ett mjukvarupaket som möjliggör följandet av partiklar genom olika medium. Detta görs utan att ta hänsyn till absorptionen av fotoner. Genom att förstå absorptionen som sker i lampans gas kan man kvantifiera hur förändringar i design skulle påverka emittorns utgående effekt. I detta examensarbete är målet att utveckla en modell som kan beskriva hur fotoner växelverkar med gasen i en medeltrycks UV lampa. En modell utvecklas och föreslås med antagandet att lokalt termodynamisk jämvikt råder och att enbart Hg strålnings data används. En simulering med en kollisionsprocess i Geant4 inkluderades. I denna kollisionsprocess antas den icke-optiska övergångssannolikheten vara densamma som för de optiska övergångarna. Detta inkluderades för att demonstrera hur en sådan process kan gå till. Detta resulterade i att kollisionerna populerade andra tillstånd vilket gjorde att dessa övergångar visade sig i utgående spektrum. Kollisionsprocessen och en metod för att beräkna Einsteins emissions koefficient med mjukvarupaketet General Relativistic Atomic Structure Package föreslås även som framtida arbete.

Development of a novel loop-mediated isothermal amplification (LAMP) assay for the rapid detection of Clostridium perfringensChaudhary, Deepa 09 December 2022 (has links) (PDF)
Clostridium perfringens (C. perfringens) causes diseases such as necrotic enteritis in poultry. Current methods for early detection of C. perfringens, including culture, and molecular techniques (PCR and qPCR), have logistical limitations in detecting pathogens on-site because they require specialized equipment and long sample preparation time. This study describes developing a highly sensitive on-site LAMP assay combined with a simple DNA extraction method to detect C. perfringens. Results showed that the LAMP assay was highly specific and sensitive with the optimal reaction conditions: 63°C for 30 mins with primer concentration of 1.2 mM FIP/BIP and 0.2 mM F3/B3. The HotSHOT method was the most convenient DNA extraction method for the on-site LAMP assay with respect to the processing time, cost, and minimal equipment requirement. Therefore, the HotSHOT DNA extraction method followed by the LAMP assay could serve as a rapid on-site molecular test for C. perfringens in the field.

Lumilos is a work investigating the possibilities to use light and color in an interactive design context.I have explored different light sources and compared them to each other and given a shortdescription of the history of light and how it has been developed during the industrialization in the western world. I have also investigated what is currently done in the field and summarized that with examples in the categories passive-, colored-, material- and interactive light. The conclusion is that working with interactive color and light is a hard task. There are many different angles to take into account such as personal likings and earlier experiences that is uncontrollable by the designer. Yet there are many possibilities and light is a material that is moldable in many forms.I have put the knowledge about light and color into practice and developed an interactive light wall that is visualizing energy consumption with the use of different colors of light and animations. The wall is built using modules to enable it to be scaled up and down to fit different contexts.
