"Estudo in vitro do potencial cariostático dos selantes resinosos, dos cimentos ionoméricos, do diamino fluoreto de prata e do verniz fluoretado, aplicados em superfície oclusal de molares permanentes humanos".

O objetivo deste trabalho foi avaliar, in vitro, ao Microscópio de Luz Polarizada (MLP), o potencial cariostático dos selantes resinosos, dos cimentos ionoméricos, do diamino fluoreto de prata e do verniz fluoretado aplicados em superfície oclusal de molares permanentes humanos. Para tanto, 70 terceiros molares humanos hígidos, extraídos por razões clínicas, foram divididos aleatoriamente em sete grupos (N=10): GI – controle (sem selamento); GII – selante resinoso sem flúor Delton ®; G III – selante resinoso fluoretado Fluroshield ®; GIV – cimento de ionômero de vidro (CIV) de presa rápida Ketac Molar ®; GV – CIV modificado por resina Vitremer ®; GVI – solução de diamino fluoreto de prata (DFP) a 30% Safluoride ®; GVII – verniz fluoretado a 2,26% Duraflur ®. Após profilaxia com jato de bicarbonato, os materiais foram aplicados na superfície oclusal, de acordo com as orientações dos fabricantes, sendo os dentes impermeabilizados com verniz com exceção da área das fissuras e de 1 mm ao redor das mesmas, sendo submetidos a ciclagens de desremineralização durante 14 dias. Após a ciclagem, foram seccionados e as secções obtidas lixadas, polidas e embebidas em água durante 36 horas, para análise ao MLP, sendo medidas transversalmente nas fotografias, a profundidade (µm) e a área m2) da lesão através de grade quadriculada em seis pontos padronizados ao longo da lesão. As médias das 6 medidas foram calculadas nos 7 grupos e comparadas estatisticamente através do teste não-paramétrico de Kruskal-Wallis e de Comparações Múltiplas de Dunn, ao nível de 5% de probabilidade (p<0,05). Observou-se diferença estatisticamente significante (P= 0.000000372) a nível de profundidade da lesão (H= 50,5) com menores valores para o GVII (0,29 µm), GVI (0,61 µm) e GV (0,89 µm) em comparação aos materiais do GIV (3,69 µm), GII (3,67 µm) e GIII (2,10 µm) que não diferiram do GI (5,81 µm). Quanto à área da lesão, houve diferença estatisticamente significante (P= 0.0000256; H= 51,3), com os menores valores para os tratamentos do GV (0,81µm2), GVII (1,78µm2), GVI (2,86 µm2), GIV (3,69 µm2) e GIII (4,91 µm2), comparados ao GII (16,39 µm2) e GI (24,04µm2), que não diferiram entre si. Concluiu-se que, os materiais fluoretados são eficazes em controlar o início e/ou a progressão da lesão de cárie na superfície oclusal quando comparados com o selante resinoso sem flúor e o não selamento. / The aim of this research was to evaluate, in vitro, with polarized light microscopy (PLM), the cariostatic potential of resin sealants and glass-ionomer cements, diamine silver fluoride solution and fluoride varnish applied on occlusal surface of human permanent molars. In order to achieve this goal, 70 caries free third human molars, extracted for clinical reasons, were randomly divided into seven groups (N=10): GI – control (no sealing treatment); GII – resin sealant without fluoride Delton ®; GIII – fluoride-releasing sealant Fluroshield ®; GIV – glass-ionomer cement Ketac Molar ®; GV – resin-modified glass-ionomer cement Vitremer ®; GVI – 30% diamine silver fluoride solution (DSF) Safluoride ®; GVII – 2,26% fluoride varnish Duraflur ®. After dental prophylaxis with sodium bicarbonate, the materials were applied on the occusal surface, according to the manufacturer’s instructions, and all tooth surfaces were covered with acid-resistant varnish except for the fissures and 1mm rim of exposed enamel surrounding this area. The specimens were then exposed to 14-day de-remineralization cycles. After this period, they were sectioned, grounded, polished and imbibed in water for 36 hours, then examined with PLM and photographed. The depth (µm) of the artificial caries lesions was assessed from the photomicrographs by measuring traverses running perpendicular to the enamel surface at six standardized points, whereas the area (µm2) was measured using a reticulated device. Data were analyzed using Kruskal-Wallis and Dunn’s multiple comparison tests at 5% of significance (p<0,05). It was observed a statistically significant difference (P= 0.000000372) in lesion depht (H=50,5) with lower values for GVII (0,29 µm), GVI (0,61 µm) and GV (0,89 µm) when compared to GIV (3,69 µm), GII (3,67 µm) and GIII (2,10 µm), which differences were not statistically significant in relation to GI (5,81 µm). Considering the lesion area, there was a statistically significant difference (P=0.0000256; H= 51,3), with lower values for GV (0,81µm2), GVII (1,78µm2), GVI (2,86 µm2), GIV (3,69 µm2) and GIII (4,91 µm2), when compared to GII (16,39 µm2) and GI (24,04µm2), which differences were not statistically significant. It was concluded that, fluoridated materials are effective in controlling the initiation and/or progression of caries lesion on the occlusal surface when compared to resin sealant without fluoride and no treatment.

Este estudo quantificou micrometricamente o índice de adaptação à dentina (presença ou ausência de fenda interfacial) de vários agentes protetores à base de hidróxido de cálcio de ativação química - Dycal e de luz visível - Prisma VLC Dycal e Ultra-Blend® plus e cimento de ionômero de vidro convencional - Ketac Bond e fotoativado - Vitrebond; associados a diferentes tratamentos de superfície dentinária, com e sem a presença de sistema restaurador adesivo (Primer & Bond 2.1 + TPH Spectrum). Os tratamentos dentinários foram jatos de ar/água, condicionamento com ácido poliacrílico a 25% por 10s e ácido fosfórico a 37% por 15s. Foram utilizados 200 dentes, os quais numa metade foi desgastado o esmalte e os preparos realizados em dentina e na outra metade repetiu-se o preparo cavitário envolvendo esmalte e dentina. Cavidades tipo classe V foram realizadas com 3mm de diâmetro e 0,8mm de profundidade em dentina e 0,8mm quando envolvido o esmalte. Após o preenchimento da cavidade, os corpos-de-prova foram incluídos em resina epóxica e seccionados longitudinalmente no sentido vestíbulo-lingual. Foram obtidas réplicas em negativo utilizando-se material de moldagem à base de polivinil siloxana (Aquasil), para a avaliação da desadaptação à dentina, por meio de microscópia de luz refletida em campo escuro. Os dados obtidos microscopicamente foram submetidos aos Testes de Kruskal-Wallis e as comparações múltiplas pelo Teste de Dunn. Os resultados demonstraram que quando comparados o comprimento e a largura da desadaptação à dentina dos agentes protetores, entre as condições experimentais com e sem a presença do sistema restaurador adesivo, o cimento de ionômero de vidro fotopolimerizável Vitrebond obteve os melhores resultados, sobrepujando, inclusive, o Ketac Bond ativado quimicamente. Numa posição intermediária vêm o Ultra-Blend® plus e o cimento de hidróxido de cálcio auto-ativado Dycal e, por último, o cimento de hidróxido de cálcio fotopolimerizável Prisma VLC Dycal, que apresentou a maior desadaptação. / The present study evaluated the length and width of interface gaps between dentin and the following lining materials: chemically cured Dycal Advanced Formula II and light cured Prisma VLC Dycal and Ultra-Blend® plus calcium hydroxide bases; chemically cured Ketac Bond and light cured Vitrebond glass ionomer cements. Dentin cavities prepared in human teeth were pre-treated with only an air/water Spray or with a 25% Polyacrylic acid solution for 10s, or a 37% Phosphoric acid solution for 15s. One half of cavities were prepared in dentin (3mm wide, 0.8mm deep) and have been filled with only the linning materials. The other half of the cavities were enamel dentin cavities had a 0.8mm with enamel layer, 3mm wide, 0.8mm deep below the dentinoenamel junction. Theses cavities received a layer of lining material have then been treated with an adhesive system an restored with a composite resin. In order to access the dentinal interfacial adaptation, the specimens were sectioned longitudinally in buccal to lingual direction thorough the center of the restorations. Low viscosity polyvinyl siloxane impression material (Aquasil) was used to obtain negative replicas. The replicas were examined with a Zeiss Photomicroscope and the length and width of the gaps were measured. The values were submitted to Kruskall Wallis Test and Multiple Comparation Dunn Test. Results demonstrated that, on the experimental conditions used, the glass ionomer cements obtained the best results, specially the Vitrebond. The Ultra-Blend plus and Dycal, showed an intermediary position. Prisma VLC Dycal showed the powerest adaptation.

O objetivo deste estudo foi avaliar a influência do tratamento do plasma não térmico (PNT) na resistência adesiva de um sistema adesivo autocondicionante aplicado em diferentes espessuras de dentina humana, bem como observar mudanças na micromorfologia das interfaces dentina/resina em MEV. Molares humanos foram divididos em dois grupos (n = 30) de acordo com a espessura da camada de dentina: superficial (S) (2 mm de espessura) e profunda (P) (1 mm de espessura). Cada grupo foi dividido em três subgrupos (n = 10) de acordo com o tratamento de superfície da dentina: grupo SPA (2 mm dentina + plasma + adesivo); Grupo SAP (2 mm dentina + adesivo + plasma); Grupo SA (2mm de dentina + adesivo); PPA (1 mm dentina + plasma + adesivo); Grupo PAP (1 mm dentina + adesivo + plasma) e grupo PA (1 mm dentina + adesivo). Foi confeccionada uma restauração em resina composta e as amostras armazenadas (48 h). Palitos de dentina/resina com área adesiva de aproximadamente 1 mm² foram obtidos e submetidos ao teste de microtração. O padrão de fraturas foi analisado por microscopia óptica. Os dados de resistência foram analisados pelos testes estatísticos ANOVA dois fatores e Tukey (5%). As análises das interfaces adesivas em MEV foram realizadas em amostras selecionadas. Os resultados mostraram uma diferença significativa na resistência adesiva entre a dentina superficial e profunda; independentemente do tratamento de superfície, a dentina superficial apresentou maiores valores de resistência adesiva. Entre os grupos tratados com e sem plasma, a resistência adesiva foi maior quando o plasma foi aplicado na superfície da dentina; no entanto a aplicação de plasma antes ou depois do o adesivo não foi estatisticamente diferente. The objective of this study was to evaluate the influence of non-thermal plasma (NTP) treatment on dentin bond strength of a self-etching adhesive system to different thicknesses of human dentin as well as to observe SEM micromorphology changes of the resin/dentin interfaces. Human molars were divided into two groups (n = 30) according to dentin layer thickness: superficial (S) (2 mm thick) and deep (D) (1 mm thick). Each group was divided into three subgroups (n = 10) according to dentin surface treatment: SPA group (2 mm dentin + plasma + adhesive); SAP group (2 mm dentin + adhesive + plasma); Group SA (2 mm dentin + adhesive); PPA (1 mm dentin + plasma + adhesive); PAP group (1 mm dentin + adhesive + plasma) and PA group (1 mm dentin + adhesive). A composite resin restoration was built up and specimens stored (48 h). Teeth sticks with adhesive area of approximately 1 mm² were obtained and submitted to microtensile test. The pattern of fractures was analyzed with optical microscopy. Bond strength data was analyzed by two-way ANOVA and Tukey (5%) statistical tests. Resin/dentin interfaces SEM analysis was conducted on selected specimens. The results showed a significant difference in bond strength between superficial and deep dentin; regardless surface treatment superficial dentin showed higher bond strength. Among the groups treated with and without plasma, bond strength was higher when plasma was applied on dentin surface; however plasma application before or after the adhesive was not statistically different. Cem dentes posteriores humanos foram divididos em cinco grupos (n = 20) de acordo protocolo de envelhecimento que receberam e posteriormente subdivididos em dois subgrupos (n = 10) de acordo com o tempo de armazenamento, totalizando dez subgrupos: In vivo (permaneceu em atividade bucal por 24 horas ou 6 meses); Estufa (armazenado em água destilada em estufa a 37 °C por 24 horas ou 6 meses); Ciclagem (submetidos a 27/666 ou 5000/120000 ciclos termomecânicos); Estufa com Pressão Pulpar Simulada (PPS) (restauração realizada sob PPS e em seguida armazenado em água destilada em estufa a 37 °C por 24 horas ou 6 meses); Ciclagem com PPS (restauração realizada sob PPS e em seguida submetidos a 27/666 -24 horas- ou 5000/120000 -6 meses- ciclos termomecânicos). Após a abertura da cavidade, a restauração foi realizada com a aplicação de ácido fosfórico a 37% (Vigodent), sistema adesivo Adper Single Bond 2 (3M ESPE) e resina composta Grandio (Voco), seguindo a orientação dos fabricantes. Após os diferentes tratamentos os espécimes foram seccionados obtendo-se palitos que foram submetidos ao teste de microtração. Os valores obtidos foram submetidos ao teste ANOVA dois fatores (protocolo de envelhecimento e tempo de armazenamento) e em seguida ao Teste de Tukey (5%). As further subdivided into two subgroups (n = 10) according to the time of storage were performed: 24 hours (24 hours in vivo 24 hours in water, 27/666 thermal and mechanical cycles) 6 months (6 months in vivo, 6 months in water and 5000/120000 thermal and mechanical cycles). After the opening of the cavity, the restoration was carried out with the use of phosphoric acid 37% (Vigodent), Single Bond 2 adhesive system (3M ESPE) and composite resin Grandio (Voco), following the lead of the manufacturers. After being held aging protocols for the storage time subgroup belonging specimens were sectioned obtaining sticks were submitted to microtensile test. Efeito de diferentes concentrações de alfatocoferol em gel sobre a adesão em dentes tratados endodonticamente e clareados /

Orientador: Andréa Abi Rached Dantas / Resumo: Estudos têm encontrado resultados promissores para o uso de antioxidantes na reversão da redução da adesão, ocasionada pela impregnação de radicais livres em dentina clareada. O objetivo do presente estudo foi avaliar o efeito de diferentes concentrações (15, 20 e 25%) de um gel de alfatocoferol na resistência à fratura, formação de camada híbrida e resistência de união de um adesivo em substrato clareado com peróxido de hidrogênio 38% (PH 38%). Sessenta incisivos bovinos foram distribuídos em 6 grupos (n=10): G1 - sem tratamento clareador (controle negativo); G2 - PH 38% (controle positivo); G3 - PH 38% + alfatocoferol gel 15%; G4 - PH 38% + alfatocoferol gel 20%; G5 - PH 38% + alfatocoferol gel 25%; G6 - PH 38% + ascorbato de sódio gel 10%. Os dentes foram preparados, submetidos a 3 aplicações de 15min de PH 38%. Os respectivos antioxidantes foram aplicados por 10 min e, em seguida, os dentes foram restaurados. O teste de resistência à fratura foi realizado 24h após o procedimento clareador em máquina de ensaio mecânico (EMIC). A formação da camada híbrida foi avaliada por meio de microscopia eletrônica de varredura (MEV) e a resistência de união, por meio do teste de microtração. Os dados obtidos com cada um dos três experimentos independentes foram submetidos à análise estatística específica (Kruskal Wallis/Dunn - a=0,05). The use of antioxidants has emerged as an interesting alternative to solve this problem. The aim of this study was evaluating the effect of different concentrations (15, 20 e 25%) of an alpha-tocopherol gel on fracture resistance, hybrid layer formation and bond strength of an adhesive system used in endodontic treated and bleached teeth (hydrogen peroxide 38% - HP 38%). Sixty bovine incisors were divided into six groups (n=10): G1 - negative control; G2 - 38% HP (positive control); G3 - 38% HP + 15% alpha-tocoferol; G4 - 38% HP + 20% alpha-tocoferol; G5 - 38% HP + 25% alpha-tocoferol; G6 - 38% HP + 10% sodium ascorbate. The specimens were prepared and submitted a bleaching using a gel with 38% HP (3x15 minutes). The antioxidant gels were applied for 10 minutes and the teeth was restored. Orientador: Edson Alves de Campos / Resumo: Um dos principais responsáveis pelo sucesso nos tratamentos restauradores são os sistemas adesivos, uma técnica que tem ganhado grande atenção na atualidade é a que faz uso de sistemas autocondicionantes. O objetivo deste trabalho foi avaliar a resistência de união de diferentes sistemas adesivos autocondicionantes de 2 passos, com graus de acidez distintos, frente ao tratamento prévio com novas formulações de dentifrícios, em substrato dentinário normal e hipermineralizado artificialmente. Cento e vinte oito dentes incisivos inferiores bovinos hígidos foram selecionados e aleatoriamente divididos em 2 categorias de acordo com o tipo de dentina: normal (N=64) e hipermineralizada (N=64). Os dentes tiveram sua superfície vestibular desgastada para expor superfície dentinária plana e lixados para a exposição de uma superfície dentinária uniforme e padronizada. Os dentes foram aleatoriamente divididos em dois diferentes grupos, de acordo com o tipo de adesivo autocondicionante utilizado: Clearfil SE Bond (grupo C) (n=32) e AdheSE (grupo A) (n=32). Foram então aleatoriamente divididos em quatro subgrupos, de acordo com o tipo de dentifrício aplicado ou a ausência deste. Entre as 152 lesões selecionadas, quatro grupos experimentais formaram-se a partir do hábito de fumar e da estratégia de utilização do adesivo Scotchbond Universal (3M/ESPE): G1)condicionamento total em não fumantes; G2)condicionamento seletivo em não fumantes; G3)condicionamento total em fumantes; G4)condicionamento seletivo em fumantes. As restaurações foram feitas de forma pareada, sendo que cada paciente recebeu, no mínimo, 2 restaurações. A resina composta nanoparticulada Filtek Z350XT (3M/ESPE) foi utilizada em todas as restaurações, as quais foram executadas por apenas um operador e avaliadas por dois examinadores calibrados, em uma semana (baseline), 6 e 12 meses após sua execução. Os critérios USPHS modificados foram utilizados. Os dados foram analisados estatisticamente pelo teste qui-quadrado (para associação entre os grupos) e McNemar, o qual associou as alterações dos resultados ao longo do tempo. Não houve diferença estatística entre os grupos para os critérios retenção, descoloração marginal, sensibilidade, cárie recorrente, cor, integridade marginal, contorno axial e textura superficial nos períodos de avaliação. Nas avaliações ao longo do tempo, houve diferença estatística apenas no critério descoloração marginal, aos 12 meses, nos grupos 1, 3 e 4 quando comparado ao período inicial. Não houve diferença estatística entre os pacientes fumantes e os não fumantes. Em relação à estratégia de união, o condicionamento total não foi diferente estatisticamente do condicionamento seletivo ao longo dos 12 meses de avaliação. Com base nos dados apresentados, pode-se concluir que o hábito de fumar e a estratégia de utilização do adesivo não influenciaram no comportamento clínico das restaurações no período de um ano.<br> / Abstract : The purpose of this randomized clinical trial was to evaluate the clinical performance of composite restorations in non-carious cervical lesions performed in smokers and non-smoking patients with different adhesive strategies. Among the 152 selected lesions, four experimental groups were formed according with the smoking habit and the variation on the bond strategy of the Scotchbond Universal Adhesive (3M/ESPE): G1)total-etching in non-smokers; G2)selective etching in non-smokers; G3)total-etching in smokers; G4)selective etching in smokers. The restorations were paired: each patient received at least two restorations. The nanofilled composite resin Filtek Z350XT (3M/ESPE) was used in all restorations, which were performed incrementally by one operator. Two calibrated examiners evaluated the restorations one week (baseline), 6 months and 12 months after implementation of the same. The modified USPHS criteria were used. Data were analyzed by chi-square test (for associations between groups) and McNemar, which combined the results of changes over time. The results showed no statistical difference between groups for the criteria retention, marginal discoloration, sensitivity, recurrent decay, color, marginal integrity, surface texture and axial contour in the evaluation periods. The assessments over time showed statistical difference only in marginal discoloration, at 12 months in the Groups 1, 3 and 4 when compared to the initial period. There were no statistical differences for all criteria evaluated among smokers and non-smokers. Regarding the bond strategy, the total-etching was not statistically different from selective etching over the 12-month evaluation. Based on the presented data it could be concluded that smoking and adhesive strategy did not influence the clinical performance of restorations over one year.

The present study is divided in three chapters, whose aims: 1) To evaluate the effect of demineralization by different acid solutions in MMP-2 activation on human dentin (Chapter 1); 2) To evaluate the effect of a one-step adhesive system on dentinal MMP-2 and -9 activation using in situ zymography and an enzymatic activity assay (Chapter 2); and 3) To evaluate the ability of MMPs inhibition by 1-ethyl-3-(3- dimethylaminopropryl) carbodiimide (EDC) EDC cross-linker on dentin (Chapter 3). As for the methodology approaches, 3 in vitro studies were performed to evaluate enzymatic expression of human dentin. Dentin protein was extracted after demineralization by 1% phosphoric acid, 10% phosphoric acid and 10% citric acid for 1, 5 and 10 minutes, and subjected to gelatin zymographic and activity assay (Chapter 1). Dentin powder/slabs were treated with one-step adhesive Adper Easy Bond (3M ESPE) and MMP-2 and -9 activities were evaluated using in situ zymography and quantified by means of an specific enzymatic assay (Chapter 2). Dentin powder/slabs were treated with one of those adhesive systems: Optibond FL or Scotchbond 1XT with or without pre-treatment using EDC. The enzymatic activity was analyzed using gelatin zymography and in situ zymography (Chapter 3). MMP-2 activity was present in all tested groups and increased after demineralization by 10% phosphoric acid and 10% citric acid (Chapter 1). After treated with one-step adhesive, enzymatic activity increased. In situ evaluation showed that MMPs action is associated with the exposed and unprotected collagen promoted by adhesive monomers (Chapter 2). Zymograms revealed increased expression of dentin endogenous MMP-2 and -9 after adhesives systems application, while the use of 0.3M EDC as a primer, inactivated dentin gelatinizes (Chapter 3). It can be concluded that both acid solutions (Chapter 1) and self-etch adhesives (Chapter 2) are able to activate dentin MMPs and that solutions like EDC cross-linker can inhibit the activation of those enzymes (Chapter 3). / As metaloproteinases são enzimas capazes de degradar o colágeno exposto na interface dentina/resina provocando sérios danos à manutenção da integridade da interface adesiva. Este estudo está dividido em três capítulos, cujos objetivos foram: 1) Avaliar o efeito da desmineralização através de diferentes ácidos na ativação de MMP-2 em dentina humana (Capítulo 1); 2) Avaliar o efeito de um sistema adesivo de passo único na ativação de MMP-2 e -9 dentinária usando zimografia in situ e teste de atividade enzimática; 3) Avaliar a capacidade do agente de ligação cruzada 1-etil-3-(3-dimetilaminopropril) carbodiamida (EDC) na inibição da atividade das MMPs em dentina (Capítulo 3). Como abordagens metodológicas foram realizados 3 estudos in vitro com avaliação enzimática em dentina humana. Proteína dentinária foi extraída após desmineralização com os ácidos fosfórico a 1% e a 10% e cítrico a 10% por 1, 5 e 10 min, e submetida a zimografia gelatinosa e teste de análise enzimática especifica, para a avaliação da atividade de MMP-2 (Capítulo 1). Dentina em pó/blocos foi tratada com o adesivo de passo único Adper Easy Bond (3M ESPE) e a atividade de MMP-2 e -9 foi avaliada através de zimografia in situ e quantificada através do ELISA (Capítulo 2). Dentina em pó/blocos foi tratada com os sistemas adesivos Optibond FL ou Scotchbond 1XT com ou sem tratamento prévio com EDC. A atividade enzimática foi analisada através de zimografia gelatinosa e in situ (Capítulo 3). A atividade de MMP-2 esteve presente em todos os grupos testados e aumentou após desmineralização com ácido fosfórico 10% e ácido cítrico 10% (Capítulo 1). Após tratamento com o adesivo de passo único, houve aumento da atividade enzimática dentinária. A análise in situ mostrou que a ação das MMPs está associada ao colágeno exposto e não protegido pelos monômeros adesivos (Capítulo 2). Zimograma revelou um aumento na expressão de MMP-2 e -9 após a exposição aos sistemas adesivos, enquanto o uso de EDC 0.3M como prétratamento, inativou as gelatinases dentinárias (Capítulo 3). Pode-se concluir que tanto soluções ácidas (Capítulo 1), quanto os adesivos autocondicionantes são capazes de ativar as MMPs dentinárias (Capítulo 2) e que algumas substâncias, como o agente de ligação cruzada EDC, são capazes de inibir a ativação destas enzimas (Capítulo 3).

Estudo da resistência de união ao cisalhamento de sistemas adesivos: influência de diferentes substratos

Os sistemas adesivos com etapa prévia de condicionamento ácido da estrutura dental utilizados foram: Scotchbond Multi Purpose Plus e Single Bond, e os sistemas adesivos autocondicionantes: Clearfill SE Bond e Adper Prompt L-Pop. Foram utilizados 80 dentes humanos, sendo 40 hígidos e 40 cariados (n=10), cortados com disco de diamante, expondo a superfície de dentina, e a área de adesão padronizada, em 4 mm de diâmetro. Para os dentes cariados, após corte inicial, estes eram corados com solução evidenciadora de cárie, e o tecido corado removido, até se obter dentina com características de viabilidade. Nos Grupos controle, os sistemas adesivos eram aplicados em dentina hígida, e nos Grupos experimentais, aplicados em dentina afetada por cárie. Confeccionaram-se cilindros de resina composta Z-100, com auxílio de matriz bipartida, os corpos-de-prova obtidos, armazenados em água destilada a 37°C por 24 horas, e submetidos ao ensaio de cisalhamento, a uma velocidade de 0,5 mm/min. Os resultados foram submetidos à Análise de Variância, e ao teste de Tukey, ao nível de 5% de significância. Observou-se influência do fator substrato para os valores de resistência de união, diminuídos significantemente quando sistemas adesivos de condicionamento ácido total eram aplicados em dentina afetada por cárie. O presente estudo indicou que a resistência de união ao cisalhamento é dependente do tipo de dentina e do tipo de sistema adesivo utilizado. / The objective of this study was to evaluate the shear bond strength of total-etch and self-etch adhesives to caries-affected versus normal dentine. The adhesive systems with previous stage of conditioning acid of the dental structure used were: Scotchbond Multi Purpose Plus and Single Bond, and the self-etching adhesives: Clearfill SE Bond and Adper Prompt L-Pop. Eighty (80) human teeth were used, being forty (40) normal dentine and forty (40) caries-affected, cut with diamond disk, exposing the dentine surface, and the area of standardized adhesion, in 4 diameter mm. For the caries dentine, after initial cut, these were staining with caries detector solution, and the infected dentine removed, until if to obtain dentine with viability characteristics. In the Groups control, the adhesive systems were applied in normal dentine, and in the experimental Groups, applied in caries-affected dentine. Cylinders of Z100 resin composite were condensed into a bipartite mold on the dentine and light activate. The samples were stored in 37oC water for 24 hours, then shear tested at a crosshead speed of 0,5 mm/min. The results were submitted to the ANOVA and to the Tukey test (p=0,05). Influence of the factor substratum was observed for the values the shear bond strength, significantly low when adhesive systems total-etch were applied in caries-affected dentine. The present study indicated that shear bond strength depended upon both the type of dentin and the adhesive system.

Materiais e Métodos. No primeiro estudo foi avaliada a influência da CLX em solução aquosa ou alcoólica na umectabilidade da dentina hígida e afetada por cárie por um sistema adesivo. Foram preparadas superfícies planas de dentina em 100 terceiros molares hígidos, das quais 50 foram submetidas ao processo microbiológico de indução de cárie. Dentro de cada condição de substrato, hígido e afetado por cárie, as superfícies foram divididas em 5 grupos (n=10): com smear layer (SL), sem smear layer impregnada com água, sem smear layer impregnada com CLX em água, sem smear layer impregnada com etanol e sem smear layer impregnada com CLX em etanol. Ângulos de contato entre a superfície de dentina e o sistema adesivo Single Bond 2 foram mensurados por meio de um goniômetro, e os dados submetidos aos testes de ANOVA e Tukey ( =0,05). No segundo estudo foi avaliado o efeito da solução de CLX em diferentes excipientes, etanol ou água, na presença de colágeno exposto na união resina-dentina hígida e dentina afetada por cárie. Foram produzidas superfícies planas de dentina em 24 molares humanos hígidos, sendo metade delas cariadas artificialmente. Os dentes foram divididos em 8 grupos (n=3) segundo o tratamento da dentina após o condicionamento ácido (CLX associada a etanol, CLX associada a água, somente etanol ou somente água) e segundo o tipo de substrato utilizado para a adesão. The overall purpose of this work, divided into three studies, was to evaluate the effect of chlorhexidine (CHX) in different excipients, ethanol and water, on the production and mechanical stability of resin-caries affected dentin bonds. Materials and Methods. In the first study, it was evaluated the influence of aqueous and alcoholic solution of CHX on the wettability of noncarious and caries-affected dentin by an adhesive system. Flat dentin surfaces (n=100) were produced, and 50 were submitted to a biological method of artificial caries induction. Within each type of substrate, noncarious and caries-affected dentin, the surfaces were divided into 5 groups (n=10): with smear layer, smear layer-free impregnated with water, smear layer-free impregnated with CHX in water, smear layer-free impregnated with ethanol and smear layer-free impregnated with CHX in ethanol. Contact angles between the dentin surface and the adhesive system Single Bond Plus were measured by using a goniometer and the data were submitted to ANOVA and Tukey tests ( =0.05). In the second study, it was evaluated the effect of CHX in different excipients, ethanol or water, on the collagen exposure in interfaces produced on noncarious and caries-affected dentin. Flat dentin surfaces were prepared in 24 noncarious human molars, and half of them were artificially decayed.
