The conidia production for the isolates was analyzed in the means of WA 2%, potato-dextrose-agar 2% (PDA 2%), corn-meal-agar 2% (CMA 2%) and yeast-phosphate-sulphate-sucrose-agar (YPSSA). In relation to the control without fungi there was significant reduction (P <0,05) of 89,89%, 97,75% and 88,76% in the average of L3 of Ancylostoma spp. recovered from medium WA 2% of the plates of Petri in the treatments with isolates CG768, I31 and NF34A, respectively. The isolate I31 was the most effective in the capture of the L3. In the fecal cultures, the reduction in average of L3 of Ancylostoma spp. in the treatments was not significant (P> 0,05). The isolated I31 was also the most effective, reducing the average of recovered L3 in 32,04% while isolated NF34A and CG768 reduced in 15,36% and 7,30% the average of infective larvae, respectively in relation to the control without fungi. There was larger conidia production of the isolate CG768 in the medium PDA 2% while in isolates I31 and NF34A this production was larger in the medium YPSSA. The results evidence that these trapping fungi can be used in the control of the L3 of Ancylostoma spp. of dogs, however, the doses and the time of interaction fungus-nematode should be studied better. / A capacidade predatória dos isolados fúngicos de Arthrobotrys robusta (I31), Duddingtonia flagrans (CG768) e Monacrosporium thaumasium (NF34A) sobre larvas infectantes (L3) de Ancylostoma spp. foi avaliada em condições laboratoriais durante 10 dias em meio ágar-água 2% (AA 2%) e em culturas fecais de fezes de cães. Analisou-se a produção de conídios pelos isolados fúngicos nos meios ágar-água (AA 2%), batata-dextrose-ágar 2% (BDA 2%), corn-meal-agar 2% (CMA 2%) e yeast-phosphate-sulphate-sacarose-agar (YPSSA). Em relação ao controle sem fungos, houve redução significativa (P < 0,05) de 89,89%, 97,75% e 88,76% na média de L3 de Ancylostoma spp. recuperadas do meio AA 2% das placas de Petri nos tratamentos com os isolados fúngicos CG768, I31 e NF34A, respectivamente. O isolado fúngico I31 foi o mais eficiente na captura das L3. Nas culturas fecais, a redução nas médias de L3 de Ancylostoma spp. nos tratamentos não foi significativa (P > 0,05). O isolado fúngico I31 também foi o mais eficaz, reduzindo a média das L3 recuperadas das coproculturas em 32,04%, enquanto que os isolados fúngicos NF34A e CG768 reduziram a média de L3 em 15,36% e 7,30%, respectivamente, em relação ao controle sem fungos. Houve maior produção de conídios do isolado fúngico CG768 no meio BDA 2% enquanto que os isolados fúngicos I31 e NF34A produziram maior número de conídios no meio YPSSA. Os resultados evidenciam que estes fungos predadores podem ser utilizados no controle de L3 de Ancylostoma spp. de cães, no entanto, a dosagem e o tempo de interação fungo-nematóide nas coproculturas devem ser melhor estudadas.

Contaminación de las playas urbanas de la provincia de Ilo con huevos de nemátodo de importancia zoonótica (Toxocara canis y Ancylostoma spp)

El presente estudio se realizó en la provincia de Ilo, departamento de Moquegua, que cuenta con cuatro playas urbanas, la problemática corresponde a la gran cantidad de perros vagabundos y con dueño, que existen en la ciudad y que acuden a estas playas, eliminando ahí sus heces. El objetivo del presente estudio fue determinar la contaminación por huevos de nematodos (Toxocara canis y Ancylostoma spp) y determinar el grado de contaminación con huevos de nematodos (Toxocara canis y Ancylostoma spp) en las playas urbanas de la Provincia de Ilo. El estudio comprendió la identificación de huevos de Toxocara canis y Ancylostoma spp, en las muestras de arena de las 4 playas urbanas que comprende la provincia de Ilo, a través de la prueba de flotación sobresaturada con Na Cl, en positivos y negativos y grado de contaminación, teniendo como resultado playas positivas a presencia de huevos Toxocara canis: la playa del Diablo 12%, la playa Boca del Río 14.26% y la playa Media Luna 16.6%, prevalece el grado de contaminación ligero con un 90% seguido del grado de contaminación moderado con un 10% y el grado de contaminación intenso con un 0%y negativo en presencia de huevo y en grado de contaminación para Ancylostoma spp, en la totalidad de las playas muestreadas. Con los resultados obtenidos se puede concluir: la presencia de huevos Toxocara canis en las playas mencionadas y ausencia de Ancylostoma spp y el grado de contaminación es ligero en las playas que presentan huevos de Toxocara canis.

The Immunoepidemiology of Hookworm Infection in the Peruvian Amazon

Hookworm infection caused by Ancylostoma duodenale or Necator americanus is a significant global health threat, causing chronic anemia, malnutrition, developmental delay, enteritis, and increased susceptibility to non-parasitic diseases. One of the most prevalent of the neglected tropical diseases, hookworm infection affects almost 1 billion people worldwide, particularly in developing countries. Current methods for diagnosis and treatment of hookworm infection are largely the same as they have been for the past century. However, several recent advances in the molecular characterization of hookworm virulence factors now provide researchers with an improved understanding of disease pathogenesis, potential targets for treatment and novel antigens for vaccine development. In order to better understand hookworm pathophysiology and immunology in human populations, a comprehensive, cross-sectional immunoepidemiologic survey of approximately 200 villagers in a remote area of the Peruvian Amazon was conducted. Hookworm prevalence rates were found to approach 40% by microscopic diagnosis. Additionally, molecular speciation techniques showed that both A. duodenale and N. americanus are endemic to this region. Reagents from a laboratory model of hookworm disease were then utilized to characterize human immune responses to hookworm specific antigens. By studying the immunoepidemiology of an endemic community we have found that a laboratory strain of hookworm, Anyclostoma ceylanicum, is a useful tool for describing species specific immune responses to disease. This work lays the foundation for future development of improved hookworm diagnostic techniques by molecular and immunologic methods.

Quandoa perda de sangue excede as reservas nutricionais, há deficiência de ferro e desenvolvimento de anemia, relacionada ao baixo rendimento escolar em crianças, apatia, adinamia e queda da produtividade em adultos. O enfoque atual para o tratamento da ancilostomíase eoutras helmintoses transmitidas pelo solo consiste em administrações periódicas de anti-helmínticos da classe dos benzimidazólicos, tanto albendazol quanto mebendazol. Contudo, estudos têm demonstrado que a reinfecção ocorre rapidamente após o tratamento, o que na maioria das vezes resulta em administrações regulares de anti-helmínticos predispondo ao risco de aparecimento de resistência às drogas e manutenção de níveis endêmicos da doença. Diante disso, nosso objetivo nesse trabalho foi avaliar a evolução da intensidade de infecção pelo ancilostomídeo em crianças entre 1 a 10 anos de idade de seis comunidades em dois municípios na região nordeste de Minas Gerais, Brasil, após o tratamento com a droga mais comumente utilizada, o Albendazol, num período de dois anos, além da possibilidade de avaliar a velocidade com que a reinfecção ocorre. Nossos resultados mostraram que a intensidade da infecção pelo ancilostomídeo medida em ovos por grama de fezes foi reduzida pela administração do medicamento, com queda parcial na prevalência que se manteve estável durante o seguimento. Houve uma superutilização do albendazol além do esperado pelo protocolo em grande parte devido ao encontro de outros parasitas cujo tratamento também se baseia no uso dessa droga. A velocidade de reinfecção não pôde ser mensurada em virtude de limitações metodológicas do protocolo. Pudemos inferir com nossos resultados, a tendência encontrada na literatura de que isoladamente o tratamento farmacológico em massa interfere na intensidade das infecções, mas não controla os níveis de doença, conforme foi observado nos 24 meses de seguimento. São necessários novos estudos com melhor abordagem metodológica e tempo de seguimento para avaliar com mais precisão o efeito da terapia farmacológica sobre a velocidade em que a reinfecção se faz. / The hookworm infection is caused by parasitic nematode species Ancylostoma duodenaleand Necator americanus. It forms one of the most common chronic infectionin humans with an estimated 740 million cases especially in poor rural areas of the tropics and subtropics according to the World Health Organization the main clinical manifestations of the disease result from chronic intestinal blood loss caused by fixing adult worms from the intestinal mucosa and submucosa. When blood loss exceeds the nutritional reserves, appear iron deficiency and development of anemia, related to poor school performance in children, apathy, adynamia and decreased productivity in adults. The current approach for the treatment of helminthiasis hookworm and other soil-transmitted helminthiasis consists of periodic administration of anthelmintic class of benzimidazole, mebendazole so as albendazole. However, studies have shown that reinfection occursrapidly after treatment, which in most cases results in regular administration of anthelmintic predisposing risk of the emergence of drug resistance and maintenance of the disease endemic levels. Therefore, our goal in this work was to evaluate the intensity of parasite infection in children aged 1-10 years from six communities in the municipalities of Novo Oriente de Minas e Caraí in northeastern of Minas Gerais, Brazil, after treatment with the most commonly used drug, the Albendazole , a period of two years, plus the ability to assess the speed with which reinfection occurs. Our results showed that the intensity of hookworm infection measured in eggs per gram of feces was reduced by administration of the drug, with partial fall in prevalence that remained stable during follow-up. On the other hand, there was an overuse of albendazole, which according to the protocol should be dispensed for the treatment of heavy infections, largely due to the gathering of other parasites which treatment also based on the use of this drug. The rate of reinfection could not be measured due to methodological limitations of the protocol. We conclude our results with the tendency found in theliterature of the pharmacological treatment in mass alone interfere with the intensity of the infection, but not controls the levels of disease, as was observed during the 24 month follow up. Further studies are needed to better methodological approach and follow-up time to more accurately assess the effect of drug therapy on the speed of reinfection.

Intestinal granules found in the parasitic nematodes Ancylostoma caninum, and Oesophagostomum radiatum

The parasitic nematodes Ancylostoma caninum and Oesophagostomum radiatum were collected and analyzed for intestinal inorganic granules. Three means of identification were utilized to determine the composition granules, including birefringence, x-ray diffraction and energy dispersive spectrometric (EDS) analysis. Initial x-ray diffraction results of the two worms showed a calcium sulfide presence within the worms. Closer examination of the granules within Ancylostoma caninum however, utilizing EDS analysis revealed their composition to be zinc sulfide. These results concur with those of Rogers (1940) and Clark (1956) who found zinc sulfide granules in several species of Strongylus. The ZnS granules seem to be a result of a detoxification function that binds excess zinc and sulfhydryl groups present from the ingestion and breakdown of dietary blood meals.

Is important to use alternative ways that can be used to fighting the spread of these parasites and their environmental infective forms, such as the use of natural antagonists of nematodes The biological control with nematophagous fungi has been suggested as an alternative to reduce this and other gastrointestinal infections caused by nematodes. The genus Duddingtonia stands out by control of nematodes in the environment. Duddingtonia flagrans is considered as the most promising species, due to the abundance of chlamydospores, which are structures of resistance. This study evaluated a fungus isolated from D. flagrans (AC001) in vitro for their ability to prey infective L3 larvae of A. ceylanicum, and its effectiveness in capturing larvae. Then, evaluated the ability to withstand the passage through the gastrointestinal tract of hamsters and ability to prey on the larvae. Was assessed on the dose of the mycelium would be more effective in relapse of the infection, were used 50 hamsters that received orally 100 L3 of A. ceylanicum. After, were formed five groups of 10 animals each, receiving fungal mycelia (25grams by weight) for a period of 14 days, every two days, and four groups treated with the fungus D. flagrans in different concentrations: group 1 (0.04 grams), group 2 (0.08 grams), group 3 (0.16 grams), group 4 (0.32 grams) and a control group that don t received this treatment. The animals were placed in separate cages and after the period of patency by A.ceylanicum (20 days) every two days, 2 grams of faeces from each group were collected from beds of hamsters for further proceed with the count of eggs per gram of feces (OPG).The results showed that there was difference (p<0.05) in reduction of the OPG of treated groups when compared to the control group.There was no difference (p>0.05) in reducing the OPG in the treated groups at different concentrations. The results of this study suggest that the fungus D. flagrans could be used as a tool in biological control of A. ceylanicum in natural conditions. / Ancylostoma ceylanicum é um nematoide gastrointestinal de cães e gatos, podendo parasitar o ser humano. Medidas alternativas que possam ser empregadas no combate à disseminação ambiental destes parasitos e suas formas infectantes são importantes, como o uso de antagonistas naturais de helmintos. O controle biológico com fungos nematófagos tem sido sugerido como uma alternativa para a redução desta e de outras infecções causadas por helmintos gastrintestinais. O gênero Duddingtonia se destaca pelo controle de nematoides no ambiente. Duddingtonia flagrans é considerada como a espécie mais promissora, devido à abundância de clamidósporos, que são estruturas de resistência. O presente estudo avaliou um isolado do fungo D. flagrans (AC001) in vitro quanto à capacidade de predar larvas infectantes L3 de A. ceylanicum, e sua eficácia na captura das larvas. Em seguida, foi avaliada a capacidade de suportar a passagem pelo trato gastrointestinal de hamsters, e habilidade de predar as larvas. Foi avaliada qual a dose do micélio seria mais eficaz na recidiva da infecção. Foram utilizados 50 hamsters que receberam por via oral 100 L3 de A. ceylanicum e formados 5 grupos com 10 animais cada um, que receberam micélio fúngico por um período de 14 dias, a cada dois dias, sendo 4 grupos tratados com o fungo D. flagrans em diferentes concentrações: grupo 1 (0,04g); grupo 2 (0,08g); grupo 3 (0,16g); grupo 4 (0,32g) e o grupo controle que não recebeu esse tratamento. Os animais foram colocados em gaiolas separadas e, após o período de patência do A. ceylanicum (20 dias), a cada dois dias, 2g de fezes de cada grupo foram coletados das camas dos hamsters para posterior realização da contagem de ovos por grama de fezes (OPG). Os resultados demonstraram que houve diferença (p<0,05) na redução do OPG dos grupos tratados em relação ao grupo controle. Não foi observada diferença (p>0,05) na redução do OPG nos grupos tratados nas diferentes concentrações. Os resultados do presente trabalho sugerem que o fungo D. flagrans poderia ser utilizado como uma ferramenta no controle biológico de A. ceylanicum em condições naturais.

The control these nematodes in adult stage is based on the use of anthelmintics. However, the use of biocontrol agents may be an additional action to reduce the population in pre-parasitic stages developing in the environment. This study aimed to evaluate the Duddingtonia flagrans predator fungus for the control of the larval form (L3) of Ancylostoma spp., in beach sand. We evaluated the in vitro infectivity of two isolates of the nematophagous fungus D. flagrans (AC001 and CG768) on infective larvae (L3) of Ancylostoma spp. dogs. Vegetative structures (mycelium), reproductive (conidia) and survival (chlamydospores) were used as fungal inoculum. The interaction was evaluated at the end of 10 days of incubation in Petridishes containing agar-water 2% medium in a temperature of 25 ° C. The antagonism in semi-natural conditions was assessed by use of a mass production of fungal inoculum in grains of milled maize. The fungus was introduced into the sand in colonized milled maize at the concentration of 15,000 chlamydospores / gram of sand. This concentration was the most effective in the preliminary in vitro assay (reduction of 59.2%). The results showed the efficiency of the fungus D. flagrans in the control of infective larvae of Ancylostoma spp. in beach sand. This suggests that isolates of this fungus may be used as part of a control program of Ancylostoma spp. in the environment. / Os nematóides do gênero Ancylostoma são endoparasitas de cães e também geohelmintos zoonóticos que podem infectar o ser humano. O controle destes nematóides em estágio adulto é baseado na utilização de anti-helmínticos. No entanto, o uso de agentes biocontroladores pode ser uma medida complementar para reduzir a população em estágios pré-parasitários em desenvolvimento no ambiente. Este estudo objetivou avaliar o fungo predador Duddingtonia flagrans no controle da forma larval infectante (L3) de Ancylostoma spp., em areia de praia. Foi avaliada a infectividade in vitro de dois isolados do fungo nematófago D. flagrans (AC001 e CG768) sobre larvas infectantes (L3) de Ancylostoma spp. de cães. Utilizou-se como inóculos fúngicos estruturas vegetativas (micélio), reprodutivas (conídios) e de sobrevivência (clamidósporos). A interação foi avaliada ao final de 10 dias de incubação em placas de Petri contendo meio ágar-água 2% em temperatura de 25 °C. O antagonismo em condições semi-naturais foi avaliado por meio da utilização de uma produção massal de inóculo fúngico em grãos de milho moído. O fungo foi incorporado à areia, em grãos colonizados de milho moído na concentração de 15.000 clamidósporos/grama de areia. Essa concentração se mostrou a mais efetiva em ensaio in vitro preliminar (redução de 59,2%). Os resultados mostraram a eficiência do fungo D. flagrans no controle de larvas infectantes de Ancylostoma spp. em areia de praia. Isso sugere que isolados desse fungo podem ser utilizados como parte de um programa de controle de Ancylostoma spp. no ambiente.

O objetivo deste estudo foi verificar a ocorrência de parasitos causadores de zoonoses em gatos entregues ao Centro de Controle de Zoonoses (CCZ) do Município de Andradina, SP, no período de março a novembro de 2007. Amostras de sangue, medula óssea, linfonodos e fezes foram submetidas a análises laboratoriais. Para o diagnóstico coproparasitológico associou-se as técnicas de Willis e Faust, observando-se uma ocorrência de 96,1% dos animais para Ancylostoma spp., 43,1% para Toxocara spp., 43,1% para Cystoisospora spp., 21,6% para Dipylidium caninum e 5,9% para Giardia spp. Oocistos de Cryptosporidium spp. foram detectados em 3,9% das amostras pela técnica de verde malaquita. Por meio do ELISA direto, constatou-se uma ocorrência de 5,8% para Cryptosporidium spp. e 13,7% para Giardia spp. Dos animais, 3,9% foram reagentes para anticorpos anti-Leishmania spp. pela técnica de ELISA indireto. Foi observada presença de forma amastigota de Leishmania spp. em “imprint”de linfonodo de um único animal. Não foi verificada associação significativa entre a ocorrência de endoparasitos e a raça, sexo e idade dos animais analisados. Os resultados obtidos confirmam que os felinos de Andradina, SP, são importantes hospedeiros de parasitos, alguns destes com elevado potencial zoonótico. / Forms evolving parasitic are eliminated in the feces of animals, promoting the environmental contamination. The purpose of this study was to verify the occurrence of the parasites that cause zoonoses in cats delivered to the Center for Zoonoses Control (CCZ) of the Andradina City, SP, in the period from March to November 2007. Samples of blood, bone marrow, lymph nodes and feces were subjected to laboratory tests. For the diagnosis coproparasitológics was associated with the techniques of Willis and Faust, observing the occurence of 96.1% of the animals to Ancylostoma spp., 43.1% for Toxocara spp., 43.1% for Cystoisospora spp. , 21.6% to Dipylidium caninum and 5.9% for Giardia spp. Oocysts of Cryptosporidium spp. were detected in 3.9% of the samples by the technique of malachite green. By the technique of direct ELISA, it is 5.8% of positive for Cryptosporidium spp. and 13.7% for Giardia spp. Of the animals, were reagents in 3.9% for antibodies anti- Leishmania spp. the technique of indirect ELISA. It was observed presence of amastigote form of Leishmania spp. in lymph nodes. There was no significant association between the occurrence of endoparasite and breed, gender and age of animals tested.
