IgA1 (immunoglobulin A1) functions as an immune molecule, against microbial invaders and other pathogens, on the surface of animal and human cells, especially on the mucosal membrane. To impair the function of immunoglobulin the human immune system some bacteria secrete site-specific IgA1 proteases to cleave IgA1 at hinge region. The protease has therefore been implicated as a putative virulence factor. Although the alignment of the DNA for the proteases predicted the IgA1 protease domain and catalytic site, lack of experimental evidence hindered in both structural and functional enzymic studies. In order to elucidate and understand the exact activity, this study aims at to analyze catalytic site of IgA1 protease from H. influenza, and thus PCR was used to amplify the DNA for putative reactive site domain (1-885) and protease domain (1-2373), respectively, were cloned in pGEM-T and transferred to the expression vector, pTrcHis-A for production of the recombinant proteins. Analysis of the recombinant proteins shows that the Human IgA1 heavy chain hydrolyzed by the protease domain recombinant protein but putative reactive site. This demonstrates that protease domain is enough for protease activity. The putative reactive site domain alone is incapable in hydrolyzing Human IgA1.

On Modelling and Estimation of Curl and Twist in Multi-Ply Paperboard

<p>This thesis describes a grey-box model for the dimensionalstability properties (i.e. curl and twist) of the carton boardproduced at AssiDomän Frövi paper mill in Sweden.AssiDomän Frövi AB is one of Sweden major cartonboard manufacturer, and produces some 350000 ton of board peryear.</p><p>Curl is defined as the departure from a at form, and it mayseriously affect the processing of the paper. For this reason,customers impose quite restrictive limits on the allowedcurvatures of the board. So, it is becoming more and moreimportant to be able to produce a carton board with a curlwithin certain limits. Due to the economic significance of thecurl problem, much research has been performed to find sheetdesign and processing strategies to eliminate or reducecurl.</p><p>The approach we used to tackle this problem is based ongrey-box modelling. The reasons for such an approach is thatthe physical process is very complex and nonlinear. The inuenceof some inputs is not entirely understood, and besides itdepends on a number of unknown parameters andunmodelled/unmesurable disturbances.</p><p>One of the main part of the model is based on classicallaminate theory which is used to model the dimensionalstability of multi-ply board. The main assumption is that eachlayer is considered as an homogeneous elastic medium.</p><p>The model is then complemented with a sub-model forunmodelled/umeasurable disturbances which are described asstates of a dynamical system, and estimated by means of anextended Kalman filter.</p><p>The simulated curvatures show a general agreement with themeasurements. However, the prediction errors are too large forthe model to be used in an effective way, and a bigger efforthas to be carried out in order to improve the physicalsub-models.</p><p>A chapter of this thesis discusses the modelling of thewet-end part of the paper machine with Dymola, a modelling toolfor simulation of large systems based on Modelica language.</p>

Efficacy of direct restorative materials in proximal box elevation on the margin quality and fracture resistance of molars restored with CAD/CAM onlays

The purpose of this study was to investigate the effect of four direct restorative materials that can be used in the proximal box elevation (PBE) technique. Materials and Methods: Seventy-five molar teeth were randomly assigned to one of five groups (n=15): Type II glass-ionomer (GI), Type II resin-modified glass-ionomer (RMGI), resin-based composite (RBC), bulk fill (BF) resin-based composite, and a control with no box elevation procedure. Specimens were prepared for a standard CAD-CAM ceramic onlay preparation with mesial cervical margins located 1 mm above CEJ (cemento-enamel junction) and distal cervical margins located 2 mm below the CEJ. PBE was used to elevate the distal margins to 1 mm above the CEJ in all groups except the control group. For the control group the onlay margin was placed directly on the prepared distal tooth without PBE. A Lava UltimateTM, CAD/CAM Resin, nano-Ceramic onlay Restorative (LAVU) was milled and bonded on all specimens with RelyX UltimateTMAdhesive Resin Cement. The margin quality of the tooth-PBE material and PBE material-onlay interface was evaluated with scanning electron microscopy (SEM) using epoxy replicas before and after mechanical loading (100,000 cycles, 1.2 Hz at 65N). In addition to margin quality, the fracture resistance of each group was measured using a universal testing machine. Fracture pattern was recorded by visual examination. One-way ANOVA was performed followed by Least Square Means. Results: For dentin margins, a statistically significant difference was detected between RMGI and control group at baseline (p=0.0442). All other groups GI, RBC, and BF showed no difference to control at baseline (p>0.05). No statistical significance was observed among groups for post-mechanical fatigue (p=0.8735). For onlay margins, no statistical significance was observed among groups for pre-mechanical fatigue, post-mechanical fatigue, or change (p=0.9713, p=0.528, p= 0.4385 respectively). No significant difference was observed for the fracture resistance among groups or for the type of break by material used (p=0.1593, p=0.77 respectively). Conclusion: Within the parameters of this study, following mechanical fatigue, the materials used for PBE: resin-modified glass-ionomer and glass-ionomer, resin-based composite and bulk-fill composite, did not influence results in terms of margin quality and fracture resistance. Therefore, collective findings suggest that these materials might be suitable for proximal box elevation procedures. Nevertheless, clinical caution is recommended with any PBE procedure and further testing of GI materials is needed.
