In this thesis, we extend the similar idea to identify multiple outliers for the generalized linear models when part of data are missing. The algorithm starts with imputation method to fill-in the missing observations in the data, and then use the forward search algorithm to confirm outliers. The proposed method can overcome the masking effect, which commonly occurs when multiple outliers exit in the data. Real data are used to illustrate the procedure, and satisfactory results are obtained.

Effects of leaf litter diversity on nutrients and mosquito communities in Neotropical artificial tree holes

Inputs from terrestrial habitats to aquatic habitats are important for structuring aquatic communities. Terrestrial producer diversity in the tropics may decline due to anthropogenic causes. I investigated how tree diversity affects aquatic communities. We used leaves from three timber-producing species (Dalbergia retusa, Pachira quinata, and Tectona grandis) to test the effects of leaf litter species composition and richness on invertebrate aquatic communities in Gamboa, Panama. We quantified macroinvertbrate species richness and abundances, leaf litter mass loss, and dissolved carbon to nitrogen ratios (C:N) after 4 weeks. We found that litter types differed in breakdown and C:N. Tectona grandis had lower dissolved C:N than both native species and supported the fewest number of invertebrates. C:N ratios declined with increasing litter diversity; however breakdown was not affected by litter richness. Mosquito abundance increased with litter species richness. Results of this study highlight the importance of diverse detritus in structuring aquatic treehole communities.

Genetic dissection reveals distinct roles for the transcription factor ZHOUPI in controlling Arabidopsis endosperm cell death and embryonic cuticle development

Angiosperm seed development requires co-ordinated development of the embryo and a second zygotic tissue, the endosperm. In Arabidopsis thaliana, the endosperm is ephemeral and is largely consumed by the embryo during seed development. In addition to a role in embryo nutrition, it is also likely that the endosperm may play a more direct role in signalling to the embryo to regulate development. Despite their importance for embryo development, these processes are very poorly understood. The ZHOUPI (ZOU) gene provides an important tool to address these problems. Firstly, ZOU likely regulates endosperm breakdown. Whereas wild-type seed have a single layer of endosperm at maturity, zou seed has a large persistent endosperm and a correspondingly small embryo. The small zou embryo does not fill the seed so that the seed shrivels as it desiccates during maturation. Secondly, zou embryos have defects in their cuticle, so that the endosperm adheres to the embryo throughout seed development. After seed germination, zou cotyledons develop holes in their epidermis as they expand, probably due to the defects in the cuticle. ZHOUPI (ZOU) encodes a bHLH transcription factor and is expressed in the embryo surrounding region (ESR) of endosperm but not in the embryo itself. The role of ZOU in cuticle development is partly mediated by the ABNORMAL LEAF SHAPE1 (ALE1) gene. Thus, ale1 mutants also show defects in embryonic cuticle development and ALE1 is specifically expressed in ESR in a ZOU-dependent fashion. It was unclear whether the effects of ZOU upon embryo development are an indirect consequence of the persistent endosperm mechanically impeding embryo expansion, or rather reflect a more direct role of the ESR in signalling to the embryo. The main aims of this thesis were 1) to provide evidence that ZOU regulates endosperm cell death and 2) to test whether ZOU function in controlling endosperm cell death could be separated from that in embryonic epidermal cuticle development. To achieve this goal, 1) TUNEL assays were performed in the seeds to confirm the zou endosperm cell death phenotype, 2) ALE1 expression in the ESR in zou mutants was rescued using the ZOU-independent AtSUC5 promoter to investigate whether one or both of zou phenotypes were complemented, 3) Candidate ZOU target genes were validated and characterized to determine their functions in endosperm cell death and/or embryonic epidermal cuticle development. The TUNEL assays revealed that zou mutants display less DNA fragmentation in the ESR than that of the wild-type, but that zou did not have defects in cell death outside the seeds suggesting ZOU specifically regulated endosperm cell death. The AtSUC5::ALE1 transgene partially rescued zou defects in epidermal cuticle but not in endosperm cell death. This shows that the defects in the zou cuticle are not caused by the defective endosperm, rather zou has distinct, separable functions. Lastly, I characterised several novel ZOU targets and showed that RGP3 may be a direct ZOU target as it is expressed in ESR in ZOU dependent fashion, whereas RGP4 is likely indirect as it is expressed in the testa and up-regulated in zou mutants. In conclusion, ZOU has independent roles in endosperm cell death and embryonic epidermal cuticle development. Because ALE1, which largely mediates the role in cuticle development, is less widely conserved than is ZOU, the role in promoting endosperm cell death may be the ancestral function of ZOU.

Variations on a Theme: Forty years of music, memories, and mistakes

How did music play a consistent role through various memories? In this memoir, I look at the sweet, the traumatic and troubling. I use specific songs as connections to lost loved ones. I pin the power of music to the loss of three important people in my life: my sister, father, and mother. Who were their musical touchstones? Did I share them? Did music run through them as it has always run through me? The memoir is sandwiched by a brief extended metaphor that props up the conceit that we are entering a live concert performance. It is billed as a "letter to a lost loved one" because it is indeed meant to address that lost one, my sister, my guide. In the opening section I've lost my voice. I eventually reclaim it and vow that I will perhaps meet my sister at some point in the future.

Electric rock breaking for south african ore bodies

Student Number : 9803381J - MSc Dissertation - Faculty of Engineering and the Built Environment / Although pulsed power has been used in many parts of the world over the last few decades to initiate high-voltage discharges through rock, no systematic test work on South African ore bodies and related rock types has been done so far. As part of CSIR Miningtek’s integrated approach of combining underground comminution with a novel Tore© hydrotransport system, which has been shown to operate well with coarse particles up to 10 mm, various rock types were fragmented in single discharge mode under laboratory conditions. The work was conducted at the University of the Witwatersrand’s high-voltage laboratory with a custom-designed test rig. The rig configuration was based on a critical review and analysis of the literature and on assessments of existing test facilities elsewhere. Core samples with diameters ranging from 16 to 48 mm were cut from test specimens with thicknesses ranging from 8 to 48 mm. Rock types included Ventersdorp Contact Reef, Carbon Leader, Elsburg Formation, UG2 and Merensky, as well as pure quartz, shales, lava and dykes. A six-stage Marx generator provided a voltage rise time of 2 000 kV/μs to create a discharge through the rock, in preference to a discharge through the surrounding water, which acts as an insulator at ramp-up times faster than 0,5 μs. High-speed photography, and an analysis of the voltage and current signals for various rock types and for water alone, were used to quantify the potential benefits of rock breaking by electric discharge. It was found that some Kimberlite specimens and mineralised gold-bearing reefs were much easier to fragment than hanging wall or footwall material. Merensky reef appeared to be more susceptible than the less brittle UG2 material. A correlation was derived between the dynamic resistivity of various rock types, measured at 16 MHz excitation frequency, and the electrical breakdown strength at which discharge took place. The fragments created had a more cubical shape than would be created by conventional impact crushing. However, the high voltage requirements of about 30 to 35 kV per millimetre of rock thickness would necessitate not only efficient mechanical and electrical contact between the electrodes and the rock, but also considerable safety features for underground installations. The clearly identified, preferential fracturing of reef rock types, compared with the hanging or footwall materials, suggests that the greater benefit of electric rock breaking may lie in primary rock breaking as a mining method, rather than in secondary comminution of broken rock to enable hydraulic transportation by pipeline to surface.

Développement d’un système analytique pour la datation in situ des roches martiennes par la méthode K-Ar / Development of a K-Ar dating instrument for in situ measurement of the martian rocks

Sur Mars, la datation par comptage de densité de cratères est actuellement la seule méthode utilisée pour dater les surfaces planétaires. Cependant, sur Mars, elle n’a pas encore été corrigé et complété par des datations absolues. Ce travail consiste à développer un prototype démontrant le potentiel d’une nouvelle approche expérimentale basée sur la méthode K-Ar pour dater les roches martiennes in situ. L’objectif à terme est de proposer une solution instrumentale de datation absolue pour un futur rover d’exploration.Un laser Nd :YAG quadruplé pour tirer à 266 nm ablate un échantillon basaltique mis sous vide secondaire. L’observation du plasma par « Laser Induced Breakdown Spectroscopy » apporte des informations sur la concentration en K et sur la nature chimique et minéralogique de la cible. Puisque l’ablation est faite par un laser UV et sous vide secondaire, l’ablation est reproductible par minéralogie. La reconnaissance stoechiométrique permet donc d’estimer la masse vaporisée. Après purification des gaz libérés, un spectromètre de masse quadripolaire détermine la quantité d’argon.L’ensemble de ces mesures pourvoit un âge avec une incertitude théorique de 13% dans les meilleures conditions.Les calibrations du dispositif expérimental ont apporté de nombreuses informations sur des effets sur les spectres LIBS provoqués par l’ablation sous vide secondaire. L’augmentation de la pression e tla variation de géométrie du cratère d’ablation ont des effets opposés sur les pics des éléments.Nous avons aussi démontré que l’instrument était capable de mesurer l’âge de la mésostase de roche basaltique et qu’il offre des perspectives intéressantes sur certaines phases minérales comme la biotite. / Crater counting is the only method used on Mars to give relative geochronological information but it never had been fitted and corrected by absolute geochronology. This work is about the development of a new prototype demonstrating the ability of a protocol using in situ K-Ar dating. The goal is to propose a solution of an absolute geochronology for the next explorations rovers. A quadrupled Nd:YAG laser at 266 nm ablates a basaltic sample under high vacuum. The light collection by a spectrometer (Laser Induced Breakdown Spectroscopy) gives the rate of potassium and the chemical or the mineralogy of the target. Thanks to the specificities of the ablation in highvacuum and with a UV laser, the ablated mass has a good reproducibility per mineralogy. Thus, theLIBS identification gives an estimation of the ablated mass. After the purification of the released gas,a quadrupole mass spectrometer determines the quantity of argon. All these measures give an age with a theoretical uncertainty of 13% in the best conditions.The calibration of the experiment had given new information about the effects on LIBS spectrarelated to the ablation under high vacuum. The rise of the pressure and the variation of geometry of the pit have opposite effects on the elements peaks. We also demonstrated that the instrument was capable of measuring the age of the groundmass of basalt and has some interesting perspectives on some mineral phases such as biotite.

Este trabalho investiga a quebra da aproximação modulacional na interação não-linear de três ondas, a interação do tripleto de ondas. Um modo comum de descrever a interação de três ondas portadoras de alta frequência é a partir da aproximação modulacional, que assume que as amplitudes e fases são lentamente moduladas. Isto apenas é verdade quando o acoplamento entre as três ondas é fraco. Ao se analisar os tipos de dinâmica envolvidas quando o valor do acoplamento é alterado, para grandes valores de acoplamento é detectada uma transição abrupta onde a amplitude limitada do regime modulacional chega a outras regiões do espaço configuracional. Também é investigado o caso onde uma das ondas do tripleto possuir uma frequência muito menor que as outras duas. / This work investigates the breakdown of the traditional modulation approximation in the three wave nonlinear interactions, the wave triplet interaction. A common way to describe the interaction of three high-frequency carriers is by the modulacional approximation, which assume that amplitude and phases are slowly modulated. This is only accurate when the three-wave coupling is weak. When analyzing the different types of dynamics when the coupling is changed, for large values of coupling it is detected an abrupt transition where the limited amplitude of the modulacional region reaches to other regions of the configuration space. It is also investigated the case where one of the waves of the triplet has a much lower frequency than the other two.
