Inheritance and expression of Cry3Aa and PVY-O coat protein transgenes in diploid and tetraploid potato

The potential benefits of plant genetic engineering for disease and pest resistance have been widely acknowledged in many studies, and although genetically modified crops are still encountering public wariness, these benefits warrant continued exploration. Because of its intrinsic economic benefits, the development of true potato seed (TPS) cropping systems has been instituted in many regions of the world. The incorporation of transgenic resistance could further the economic gain of farmers who are seeking ways to sustain their livelihood in the most efficient way possible. It is, however, largely unresearched how sexual hybridization of a transgenic crop would affect the behavior of a transgene in the resultant progeny. In the initial part of this study, transgenic lines were developed with a Cry3Aa transgene. These plants were then used in 4x-4x reciprocal crosses and 4x-2x hybridization schemes to determine the stability of the transgene after sexual hybridization. There was no observed parent of origin effect on transgene expression; however, a highly significant, non-mendelian inheritance of the Cry3Aa transgene was seen in the maternally inherited transgene of one set of progeny from a reciprocal cross. Additional transgenic lines of potato were developed with a PVY-O coat protein transgene. These plants were challenged with PVY-O and monitored for symptoms visually and for virus serologically. One transgenic line exhibited complete resistance to PVY-O while two others showed a delay in symptom occurrence. Further examination of the expression levels of the PVY-O coat protein transgene will be necessary to determine the type and usefulness of the observed resistance. / Master of Science

Interrelationship of Bacillus thuringiensis Berliner to diamondback moth, Plutella xylostella L. (Lepidoptera: Plutellidaae), and its primary parasitoid, Diadegma insulare Cress (Hymenoptera: Ichneumonidae)

The susceptibility of a population of diamondback moth, Plutella xvlostella L (Lepidoptera: Plutellidae), collected from Montgomery County, Virginia, and its ability to develop resistance to Bacillus thuringiensis was evaluated. The field population of diamondback moths was found to be susceptible to B. thuringiensis. Selection pressure at 153 I.U. of B. thuringiensis endotoxin per ml for nine generations did not cause any significant difference in mean mortality of third instar diamondback moths although there was a trend towards lower mortality, there was significant negative linear correlation between generation and mean mortality of diamondback moth larvae (P = 0.003, r²= 0.73). LC₅₀ increased from 264 I.U. of B. thuringiensis endotoxin per ml in generation I to 514 LU/ml in generation IX. The interrelationship of B. thuringiensis to diamondback moth and its primary parasitoid, Diadegma insulare Cress (Hymenoptera. Ichneumonidae), was studied by determining: the differential response of third instar diamondback moth, parasitized and unparasitized, to B. thuringiensis, and the ability of D. insulare to discriminate between B. thuringiensis-treated and untreated hosts. There was no significant difference (P > 0.05) between mean mortality of parasitized and unparasitized larvae at each of the three concentrations consisting of 154, 334, and 2237 I.U. of B. thuringiensis endotoxin per ml. The regressions of the response of parasitized and unparasitized larvae, however, were highly significant (P = 0.0001). The LC₅₀s of parasitized versus unparasitized larvae were 373 and 176 I.U./ml B. thuringiensis endotoxin, respectively. Female D. insulare did not discriminate between B. thuringiensis-treated and untreated hosts. The percentage of D. insulare females emerging from B. thuringiensis-treated larvae (41.4%) was not significantly different from that of untreated larvae (32.0 %). Mean mortality of third instar diamondback moth subjected to B. thuringiensis endotoxin at 153, 334, and 2237 I.U./ml were not significantly different at temperatures of 15 and 20 °C, but were significantly lower than that at 30 °C. The effects of B. thuringiensis endotoxin residues on leaves under room conditions [27 ± 1 °C, RH 27 ± 72%, and 8:16 (L:D)] were not significantly different at 2 and 192 hours after treatment. / Master of Science

Étude sur les interactions entre les acétophénones de l'épinette blanche et le Btk chez la tordeuse des bourgeons de l'épinette sous diverses conditions d'élevage en milieu artificiel

La tordeuse des bourgeons de l’épinette (Choristoneura fumiferana (Clems.)) est le principal défoliateur des forêts de conifères de l’Amérique du Nord. Récemment, des épinettes blanches (Picea glauca (Moench) Voss) naturellement résistantes à cet insecte ont été découvertes dans une plantation subissant une forte infestation de ce ravageur. Des analyses chimiques du feuillage ont révélé la présence de deux acétophénones, le picéol (4’-hydroxy-acetophenone) et le pungénol (3’, 4’-dihydroxy-acetophenone) chez ces épinettes résistantes. Les objectifs de cette étude étaient dans un premier temps de valider l’effet de ces deux molécules sur la survie des larves de la tordeuse selon plusieurs conditions d’élevage, et dans un deuxième temps de vérifier la compatibilité entre le picéol et le principal moyen de lutte utilisé au Québec contre ce ravageur, le Btk (Bacillus thuringiensisvar. kurstaki). Les larves de la tordeuse des bourgeons de l’épinette ont été élevées en laboratoire sur de la diète artificielle, et les composées acétophénones leur ont été administrés en solution ou en les mélangeant à la diète artificielle. Les résultats ont montré que l’absorption directe d’une dose ponctuelle de picéol et de pungénol ne provoque pas de mortalité chez les larves alors que la toxicité du picéol chez la tordeuse des bourgeons de l’épinette soumise à une exposition chronique du produit varie selon les conditions d’élevage. De plus, un effet antagoniste entre le picéol et le Btk a également été détecté au laboratoire. Cette recherche a permis de faire avancer la compréhension des interactions complexes entre les molécules de défense de l’épinette blanche et les conditions d’élevage en laboratoire. D’autres travaux en laboratoire et sur le terrain sont nécessaires afin de mieux comprendre le mode d’action de ces composés de défense et ainsi les utiliser à leur plein potentiel.

Molecular mode of action of Cry6Aa1, a new insecticidal Bacillus thuringiensis toxin

Cry6Aa1, une nouvelle toxine produite par Bacillus thuringiensis (Bt), agit comme insecticide sur la chrysomèle du maïs (WCRW). Dans cette étude, on démontre que Cry6Aa1 est une toxine formeuse de pores (TFP) en bicouches lipidiques planes (BLP). Contrairement aux autres toxines de Bt étudiées jusqu’à présent, la formation de pores par Cry6Aa1 ne requiert pas de prétraitement par protéases et se produit à des doses de toxine deux à trois ordres de grandeur plus faibles que celles nécessaires pour les autres toxines de Bt dans les mêmes conditions. La formation de pores par la forme non traitée de Cry6Aa1 dépend du pH; les pores obtenus ont des conductances comprises entre 31 et 689 pS en conditions symétriques de 150 mM de KCl; ils sont cationiques avec un comportement cinétique complexe. Les propriétés biophysiques des pores ne changent pas lorsque la toxine est traitée avec le suc du mésenthéron de l’insecte (Cry6Aa1 WCR1). Par contre, un traitement à la trypsine (Cry6Aa1 TT) modifie la conductance et la sélectivité des pores à pH 5,5 (le pH physiologique de l’intestin de WCRW). La reconstitution en BLP de fraction de membrane native du mésenthéron de WCRW affecte les propriétés des pores formés par Cy6Aa1. Les déterminants moléculaires du mode d’action de cette nouvelle toxine formeuse de pores semblent donc différer de ceux décrits précédemment pour d’autres toxines de Bt. La structure atomique tridimensionnelle de Cry6Aa1 vient tout juste d’être élucidée. Elle montre que la toxine adopte une conformation riche en hélices α qui ressemble fortement à celle de la TFP ClyA produite par E. coli. En se fondant sur les données disponibles pour ClyA, on a étudié l’effet de divers changements dans les régions N et C terminales de Cry6Aa1 sur sa capacité de former des pores en BLP. / Cry6Aa1 is a new toxin produced by Bacillus thuringiensis (Bt), which displays insecticidal activity against the Western corn rootworm (WCRW). The present work demonstrates that Cry6Aa1 is a pore-forming toxin (PFT) in planar lipid bilayers (PLBs). Contrary to other Bt toxins tested so far, pore formation by Cry6Aa1 does not require protease pretreatment and takes place at doses that are two to three orders of magnitude lower than those required for other Bt toxins under similar conditions. Pore formation by Cry6Aa1 is pH-dependent; the conductances of the pores range between 31 and 689 pS under symmetrical 150 mM KCl conditions; they are cationic and display a complex kinetic behaviour. The treatment of the toxin with midgut juice (Cry6Aa1 WCR1) does not change the biophysical properties of the pores. However, the treatment with trypsin (Cry6Aa1 TT) affects their conductance and selectivity at pH 5.5 (the WCRW gut physiological pH). The incorporation in PLBs of native membrane material from WCRW midgut affects the behaviour of the Cry6Aa1 pores. The molecular determinants of the mode of action of this new PFT appear therefore to differ from those reported before for other Bt toxins. The three-dimensional (3-D) atomic structure of Cry6Aa1 has just been elucidated. It shows that the toxin assumes an α-helix-rich configuration, which is quite similar to that of the ClyA PFT produced by E. coli. Based on the data available for ClyA, we have studied how different changes in the N- and C-terminal regions of Cry6Aa1 affect its pore formation ability in PLBs.

Mode d’action moléculaire de la toxine anti-tumorale : PS1Aa2 du bacille de Thuringe

Les parasporines sont des toxines Cry du bacille de Thuringe actives contre des cellules tumorales. Ce travail montre que la parasporine PS1Aa2 (Cry31Aa2) forme des pores dans des membranes artificielles, comme de nombreuses toxines Cry. Ceux-ci ont plusieurs niveaux de conductance dont les plus fréquents étaient de 11, 16 et 21 pS dans une solution de 150 mM KCl. Nos résultats de microspectrofluorométrie avec la sonde Fura-2 montrent que la présence de la PS1Aa2 peut produire des augmentations du calcium intracellulaire, la plupart du temps sous la forme d’oscillations calciques et parfois des augmentations soutenues. Ces réponses ont été observées en présence et en absence de calcium extracellulaire, dans les lignées tumorales HeLa et HepG2 et dans la lignée non tumorale HEK 293. Bien que quelques études aient montré que le calcium semble intervenir dans leur mode d’action, de telles oscillations calciques n’ont jamais été décrites auparavant pour des toxines Cry. Les expériences ont dû être faites à des concentrations beaucoup plus élevées de toxine que prévues sur la base des résultats publiés de cytotoxicité. Malgré la présence des fragments identifiés auparavant comme actifs, sa faible efficacité semble liée à la présence d’ADN dans les préparations qui entraîne la précipitation de la protéine. Les travaux futurs sur cette toxine seraient donc grandement facilités par une amélioration de sa méthode de préparation. / Parasporins are Cry toxins from Bacillus thuringiensis that are active against tumor cells. This work shows that parasporin PS1Aa2 (Cry31Aa2) forms pores in artificial membranes like many Cry toxins. These pores have several levels of conductance; the most frequently seen in 150 mM KCl solutions were of 11, 16 and 21 pS. Our microspectrofluorometric results with the Fura-2 probe showed that the presence of PS1Aa2 can induce changes in intracellular calcium levels, most often in the form of calcium oscillations and sometimes as sustained increases. Such responses were observed in the presence and absence of extracellular calcium, with the tumor cell lines HeLa and HepG2, and with the non-tumorous cell line HEK 293. Calcium oscillations have not been described previously for Cry toxins even though some studies have shown that calcium appears to be involved in their mode of action. Our experiments required the use of much higher concentrations of toxin than suggested from the published cytotoxicity results. Despite the presence of fragments previously identified as active, its low efficacy appears to be related to the presence of DNA in the preparations causing the protein to precipitate. Future work on this toxin would therefore be greatly facilitated by an improvement in its method of preparation.

Viabilização do uso de Bt para o manejo do HLB dos citros via redução da população de Diaphorina citri / Feasibility of the Bt use for reduction of Diaphorina citri population and improved citrus HLB management

A citricultura se destaca como uma das mais importantes atividades do agronegócio brasileiro, sendo o estado de São Paulo o principal produtor mundial de laranja e o maior exportador de suco concentrado e congelado. O psilídeo dos citros (Diaphorina citri) tornou-se nos últimos anos um dos mais importantes vetores na cultura, devido à transmissão de \'Candidatus Liberibacter asiaticus\' e \'Ca. L. americanus\', bactérias associadas ao huanglongbing (HLB), principal doença da citricultura atual. Uma vez que não há variedades comerciais de citros resistentes ao HLB, seu manejo é baseado no uso de mudas sadias, erradicação de plantas infectadas e pelo controle químico do vetor. No entanto, o custo elevado e os significativos danos ambientais causados pelos inseticidas químicos, associados à possibilidade da seleção de populações de psilídeos resistentes a esses produtos, têm levado à busca por estratégias alternativas de manejo do vetor do HLB que sejam mais adequadas e sustentáveis. Nosso grupo tem tentado contribuir nesse sentido, com a prospecção e caracterização de estirpes de Bacillus thuringiensis (Bt) patogênicos ao psilídeo. Sendo assim, os objetivos deste estudo foram: (a) Confirmar a capacidade endofítica e a patogenicidade de estirpes de Bt em seedlings e mudas de citros com diferentes combinações de copa/porta-enxerto; (b) Determinar a concentração letal (CL50) necessária para ocasionar mortalidade em populações de D. citri; (c) Estudar a interação entre toxinas Cry e receptores de membrana do intestino de ninfas e (d) Avaliar a compatibilidade das estirpes selecionadas com agrotóxicos comumente utilizados na citricultura. Os bioensaios realizados com seedlings e mudas de citros demonstraram que as estirpes de Bt (Cyt1A, Cry2Aa, Cry4A, Cry10, Cry11, S1302, S1450 e S1989) translocaram endofiticamente nas plantas, mantendo sua viabilidade e, em sua maioria, o potencial patogênico para ninfas de D. citri. Para seedlings, os valores de mortalidade passaram de 80% para as estirpes S1302 e S1450. Foram observados esporos e células vegetativas de B. thuringiensis subsp. kurstaki expressando green fluorescent protein (Btk::GFP), visualizados por microscopia de fluorescência, aderidos principalmente aos elementos de vasos e no xilema, obtidas de amostras seccionadas transversal e longitudinalmente, de caule e raiz de seedlings e mudas de laranjeira Pera. A CL50 e CL80 para a estirpe S1302 foi de 4,92 × 104 esporos/mL e 6,63 × 107 esporos/mL, respectivamente. Já para a estirpe S1450, 50% de mortalidade das ninfas testadas foi estimada em 2,19 × 104 esporos/mL, e a CL80 foi de 6,18 × 108 esporos/mL, quando utilizadas suspensões de esporos da bactéria diretamente no substrato de seedlings de laranjeira Pera. Os ensaios de ligação demonstraram que todas as toxinas Cry presentes nas estirpes S1302, S1450 e S1989 (Cry1Aa, Cry1Ab, Cry1Ac, Cry2Aa, Cry2Ab2, Cry1B e Cry11) interagiram com os receptores de membrana intestinal, brush border membrane vesicles (BBMV\'s), obtidos de ninfas de D. citri. Ensaios in vivo evidenciaram que as estirpes S1302 e S1450 mostraram-se compatíveis com todos os agrotóxicos comumente utilizados na citricultura, ainda que os produtos à base de cobre e o inseticida Dimetoato tenham sido deletérios em aplicações diretas na bactéria in vitro. Esses dados evidenciam a possibildade do uso de Bt como bioinseticida no manejo integrado do HLB. / Citriculture is one the most important activities of Brazilian agribusiness, and the State of São Paulo being the world\'s leading orange producer and the largest juice exporter. Asian citrus psyllid - ACP (Diaphorina citri) has become one of the most important vectors in the citriculture in recent years, due to the transmission of \'Candidatus Liberibacter asiaticus\' and \'Ca. L. americanus\', bacteria associated with huanglongbing (HLB), the main citrus disease worldwide. Nowadays there are no commercial citrus varieties resistant to HLB, and its management is based on the use of healthy nursery citrus trees, eradication of symptomatic planst from the orchards, and vector chemical control. However, excessive cost and environmental damage due to application of chemical insecticides associated with the possibility of selection of ACP resistant populations, have led researchers to persue alternative strategies for the management of HLB. Our group has contributed with the screening of Bacillus thuringiensis (Bt) strains with potential against ACP. Therefore, our objectives were: (a) to confirm Bt endophytic translocation in citrus seedlings and nursery trees with different scion-rootstock combinations and to evaluate their pathogenicity against D. citri; (b) to estimate the lethal concentration (LC50 and LC80) to D. citri nymphs; (c) to study the interaction between Cry toxins and brush border membrane vesicle (BBMV) of the midgut D. citri nymphs, and (d) to evaluate the compatibility of pesticides with the selected Bt strains. The bioassays with citrus seedlings and nursery trees demonstrated that the Bt strains (Cyt1A, Cry2Aa, Cry4A, Cry10, Cry11, S1302, S1450 and S1989) translocated from roots to young leaves, maintaining their viability and pathogenicity against D. citri nymphs. For the seedlings, we found mortality values up to 80% for strains S1302 and S1450. Btk::GFP spores and vegetative cells were visualized by fluorescence microscopy in citrus seedlings and nursery trees adhered mainly to vessels and xylem, from roots to stems, in cross-section analyses. LC50 and LC80 for strain S1302 were 4.92 × 104 spores/mL and 6.63 × 107 spores/mL, respectively. For strain S1450, 50% mortality of nymphs tested was estimated at 2.19 × 104 spores/mL, and LC80 was 6.18 × 108 spores/mL, when bacterial spore suspensions were applied to citrus seedlings. Binding assays demonstrated that all Cry toxins present in strains S1302, S1450 and S1989 (Cry1Aa, Cry1Ab, Cry1Ac, Cry2Aa, Cry2Ab2, Cry1B and Cry11) interacted with the BBMV obtained from D. citri nymphs. In vivo assays showed that strains S1302 and S1450 were compatible with all pesticides commonly used in citrus orchards, althoug copper-based pesticides and dimethoate insecticide were incompatible in vitro with Bt strains. Our results demonstrate the potential of using Bt as systemic bioinsecticide in the future in HLB management.

Atividade inibitória de bovicina HC5 sobre bactérias deterioradoras de polpa de manga / Inhibitory activity of bovicin HC5 against spoilage bacteria from mango pulp

Nisin is the bacteriocin that has been most used in foods. Recently, a new bacteriocin, bovicin HC5, produced by Streptococcus bovis HC5, was characterized. Previous work indicated that this bacteriocin has activity against Listeria monocytogenes and Clostridium sporogenes. In this study, the activity of this bacteriocin was tested against Bacillus cereus, Bacillus thuringiensis and Clostridium tyrobutyricum isolated form spoiled mango pulp. The addition of bovicin HC5 (40 to 160 AU/mL) into BHI media resulted in reduced specific growth rate and maximal optical densities of B. cereus, B. thuringiensis and C. tyrobutyricum. Concentrations of 40 and 80 AU/mL increased lag phase duration for at least 10 h. When 160 AU/mL was used, growth was not observed even after 144 h. The effect of bovicin HC5 against vegetative cells inoculated into mango pulp was bactericidal and more pronounced at acidic conditions. After 24 h of incubation with the bacteriocin, the viable cell number was bellow the detection level. Similar results were obtained when nisin was used. When C. tyrobutyricum was inoculated into mango pulp with 100 AU/mL of bovicin HC5, gas production was not observed for at least 10 days of incubation. Bovicin HC5 reduced spore germination of B. cereus and B. thuringiensis inoculated into mango pulp, and the number of non-germinated spores after 122 h of incubation was at least 100-fold greater than control treatments. Bovicin HC5 did not affect the thermal resistance of B. cereus and B. thuringiensis spores. However, if added to the mango pulp, bovicin HC5 could remain stable during the heat treatment, and reduce spore germination of these microorganisms. Cultures of B. cereus, B. thuringiensis and C. tyrobutyricum that were transferred successively in the presence of subletal doses of bovicin HC5 did not become resistant. Considering the results obtained in this study and the fact that bovicin HC5 was stable in culture supernatants and in mango pulp, it seems that this bacteriocin could be useful to prevent the spoilage of mango pulp by B. cereus, B. thuringiensis and C. tyrobutyricum. / Bacteriocinas de bactérias do ácido láctico têm sido propostas como alternativa aos métodos tradicionais de conservação de alimentos, como tratamento térmico, que interferem nas características naturais do alimento. A nisina é a bacteriocina que tem sido mais utilizada em alimentos. Recentemente, uma nova bacteriocina, bovicina HC5 produzida por Streptococcus bovis HC5, foi caracterizada e apresentou efetividade contra Listeria monocytogenes e Clostridium sporogenes. Neste estudo, a atividade desta bacteriocina foi testada contra Bacillus cereus, Bacillus thuringiensis e linhagens de Clostridium tyrobutyricum isolados da polpa de manga deteriorada. A adição de 40 a 160 UA/mL de bovicina HC5 em caldo BHI resultou na diminuição da velocidade específica de crescimento e da DO máxima atingida por B. cereus, B. thuringiensis e C. tyrobutyricum. Concentrações de 40 e 80 UA/mL de bovicina HC5 resultaram no aumento da fase lag dos isolados em pelo menos 10 h e quando a concentração utilizada foi de 160 UA/mL, o crescimento não foi observado por, pelo menos, 144 h. Concentração de 100 UA/mL de bovicina HC5 foi bactericida para células vegetativas inoculadas em polpa de manga e este efeito foi mais pronunciado em condições acídicas. Após 24 horas de incubação na presença da bacteriocina, o número de células viáveis das bactérias avaliadas ficou abaixo do limite de detecção. Resultados semelhantes foram obtidos com a nisina. Quando as linhagens de C. tyrobutyricum foram inoculadas em polpa de manga contendo 100 UA/mL de bovicina HC5, a produção de gás não foi observada por até 10 dias de incubação. Bovicina HC5 reduziu a germinação de esporos de B. cereus e B. thuringiensis e o número de esporos no estado dormente após 122 h de incubação foi, pelo menos, 100 vezes maior do que o observado no tratamento controle, sem bacteriocina. Esta bacteriocina não teve efeito na resistência térmica de esporos de B. cereus e B. thuringiensis. Entretanto, uma vez que a bovicina HC5 apresenta resistência a altas temperaturas (121 ºC/20 min), ela pode permanecer estável na polpa de manga após o tratamento térmico e reduzir a germinação dos esporos sobreviventes. A transferência dos isolados por, aproximadamente, 40 gerações na presença de 20 UA/mL de bovicina HC5 não causou adaptação dos microrganismos deterioradores. Esta bacteriocina permaneceu estável após incubação em mistura com o sobrenadante das culturas e com a polpa de manga. Considerando os resultados obtidos neste trabalho, a bovicina HC5 parece ser útil para reduzir a deterioração de polpa de manga causada por B. cereus, B. thuringiensis e C. tyrobutyricum.

A citricultura se destaca como uma das mais importantes atividades do agronegócio brasileiro, sendo o estado de São Paulo o principal produtor mundial de laranja e o maior exportador de suco concentrado e congelado. O psilídeo dos citros (Diaphorina citri) tornou-se nos últimos anos um dos mais importantes vetores na cultura, devido à transmissão de \'Candidatus Liberibacter asiaticus\' e \'Ca. L. americanus\', bactérias associadas ao huanglongbing (HLB), principal doença da citricultura atual. Uma vez que não há variedades comerciais de citros resistentes ao HLB, seu manejo é baseado no uso de mudas sadias, erradicação de plantas infectadas e pelo controle químico do vetor. No entanto, o custo elevado e os significativos danos ambientais causados pelos inseticidas químicos, associados à possibilidade da seleção de populações de psilídeos resistentes a esses produtos, têm levado à busca por estratégias alternativas de manejo do vetor do HLB que sejam mais adequadas e sustentáveis. Nosso grupo tem tentado contribuir nesse sentido, com a prospecção e caracterização de estirpes de Bacillus thuringiensis (Bt) patogênicos ao psilídeo. Sendo assim, os objetivos deste estudo foram: (a) Confirmar a capacidade endofítica e a patogenicidade de estirpes de Bt em seedlings e mudas de citros com diferentes combinações de copa/porta-enxerto; (b) Determinar a concentração letal (CL50) necessária para ocasionar mortalidade em populações de D. citri; (c) Estudar a interação entre toxinas Cry e receptores de membrana do intestino de ninfas e (d) Avaliar a compatibilidade das estirpes selecionadas com agrotóxicos comumente utilizados na citricultura. Os bioensaios realizados com seedlings e mudas de citros demonstraram que as estirpes de Bt (Cyt1A, Cry2Aa, Cry4A, Cry10, Cry11, S1302, S1450 e S1989) translocaram endofiticamente nas plantas, mantendo sua viabilidade e, em sua maioria, o potencial patogênico para ninfas de D. citri. Para seedlings, os valores de mortalidade passaram de 80% para as estirpes S1302 e S1450. Foram observados esporos e células vegetativas de B. thuringiensis subsp. kurstaki expressando green fluorescent protein (Btk::GFP), visualizados por microscopia de fluorescência, aderidos principalmente aos elementos de vasos e no xilema, obtidas de amostras seccionadas transversal e longitudinalmente, de caule e raiz de seedlings e mudas de laranjeira Pera. A CL50 e CL80 para a estirpe S1302 foi de 4,92 × 104 esporos/mL e 6,63 × 107 esporos/mL, respectivamente. Já para a estirpe S1450, 50% de mortalidade das ninfas testadas foi estimada em 2,19 × 104 esporos/mL, e a CL80 foi de 6,18 × 108 esporos/mL, quando utilizadas suspensões de esporos da bactéria diretamente no substrato de seedlings de laranjeira Pera. Os ensaios de ligação demonstraram que todas as toxinas Cry presentes nas estirpes S1302, S1450 e S1989 (Cry1Aa, Cry1Ab, Cry1Ac, Cry2Aa, Cry2Ab2, Cry1B e Cry11) interagiram com os receptores de membrana intestinal, brush border membrane vesicles (BBMV\'s), obtidos de ninfas de D. citri. Ensaios in vivo evidenciaram que as estirpes S1302 e S1450 mostraram-se compatíveis com todos os agrotóxicos comumente utilizados na citricultura, ainda que os produtos à base de cobre e o inseticida Dimetoato tenham sido deletérios em aplicações diretas na bactéria in vitro. Esses dados evidenciam a possibildade do uso de Bt como bioinseticida no manejo integrado do HLB. / Citriculture is one the most important activities of Brazilian agribusiness, and the State of São Paulo being the world\'s leading orange producer and the largest juice exporter. Asian citrus psyllid - ACP (Diaphorina citri) has become one of the most important vectors in the citriculture in recent years, due to the transmission of \'Candidatus Liberibacter asiaticus\' and \'Ca. L. americanus\', bacteria associated with huanglongbing (HLB), the main citrus disease worldwide. Nowadays there are no commercial citrus varieties resistant to HLB, and its management is based on the use of healthy nursery citrus trees, eradication of symptomatic planst from the orchards, and vector chemical control. However, excessive cost and environmental damage due to application of chemical insecticides associated with the possibility of selection of ACP resistant populations, have led researchers to persue alternative strategies for the management of HLB. Our group has contributed with the screening of Bacillus thuringiensis (Bt) strains with potential against ACP. Therefore, our objectives were: (a) to confirm Bt endophytic translocation in citrus seedlings and nursery trees with different scion-rootstock combinations and to evaluate their pathogenicity against D. citri; (b) to estimate the lethal concentration (LC50 and LC80) to D. citri nymphs; (c) to study the interaction between Cry toxins and brush border membrane vesicle (BBMV) of the midgut D. citri nymphs, and (d) to evaluate the compatibility of pesticides with the selected Bt strains. The bioassays with citrus seedlings and nursery trees demonstrated that the Bt strains (Cyt1A, Cry2Aa, Cry4A, Cry10, Cry11, S1302, S1450 and S1989) translocated from roots to young leaves, maintaining their viability and pathogenicity against D. citri nymphs. For the seedlings, we found mortality values up to 80% for strains S1302 and S1450. Btk::GFP spores and vegetative cells were visualized by fluorescence microscopy in citrus seedlings and nursery trees adhered mainly to vessels and xylem, from roots to stems, in cross-section analyses. LC50 and LC80 for strain S1302 were 4.92 × 104 spores/mL and 6.63 × 107 spores/mL, respectively. For strain S1450, 50% mortality of nymphs tested was estimated at 2.19 × 104 spores/mL, and LC80 was 6.18 × 108 spores/mL, when bacterial spore suspensions were applied to citrus seedlings. Binding assays demonstrated that all Cry toxins present in strains S1302, S1450 and S1989 (Cry1Aa, Cry1Ab, Cry1Ac, Cry2Aa, Cry2Ab2, Cry1B and Cry11) interacted with the BBMV obtained from D. citri nymphs. In vivo assays showed that strains S1302 and S1450 were compatible with all pesticides commonly used in citrus orchards, althoug copper-based pesticides and dimethoate insecticide were incompatible in vitro with Bt strains. Our results demonstrate the potential of using Bt as systemic bioinsecticide in the future in HLB management.

Étude in vitro des changements physiologiques des cellules épithéliales du moustique Aedes aegypti en réponse à une exposition aux toxines du bacille de Thuringe

Bacillus thuringiensis sérotype israelensis (Bti) produit quatre toxines entomocides utilisées à grande échelle pour le biocontrôle des populations de diptères nuisibles et vecteurs de maladies : Cry4Aa, Cry4Ba, Cry11Aa et Cyt1Aa. Chacune de ces toxines présente un effet létal sur différents insectes mais, lorsqu’elles sont combinées, on observe un effet synergique et l’absence de résistance. Bien que cette synergie soit bien documentée par des tests de toxicité, il existe très peu d’information sur son mécanisme aux niveaux cellulaire et moléculaire. À l’aide d’intestins isolés des larves du moustique Aedes aegypti, le principal vecteur du paludisme, et de microélectrodes, nous avons observé une dépolarisation membranaire en présence de Cyt1Aa et de Cry4Aa individuellement. Cette dépolarisation se produit cependant plus rapidement lorsque la Cyt1Aa est utilisée en même temps que la Cry4Aa. D’autre part, des expériences réalisées avec la sonde calcique Fura-2 sur une lignée cellulaire provenant d’Anopheles gambiae (Ag55), ont révélé une forte activité lytique de la Cyt1Aa, mais très peu d’effets des autres Cry, et ce même en combinaison. Nous avons dissocié les cellules de l’épithélium intestinal isolé du moustique pour des expériences de Fura2. Nos résultats, quoique préliminaires, montrent les effets variables de ces toxines lorsqu’elles sont administrées seules sur les cellules dissociées : une augmentation du calcium intracellulaire, ou une fuite de la sonde se traduisant par une perte du signal fluorescent, ou la lyse cellulaire. On observe également en présence de Cyt1Aa et de Cry4Ba, que les effets sont presque instantanés. / Bacillus thuringiensis var israelensis (Bti) produces four insecticidal toxins used around the world to control disease-borne and harmful dipterans populations: Cry4Aa, Cry4Ba, Cry11Aa and Cyt1Aa. They each present their lethal effect on different dipterans, but combined, they generate a synergistic activity and a reduced resistance is observed. Though these synergies are well documented and supported by toxicity bioassays, little is known regarding the cellular and molecular mechanisms of these synergies. Here, by using freshly isolated midguts from the mosquito Aedes aegypti, an important malaria vector, and glass microelectrodes, we measured the electrical potential of the apical membrane when exposed to these toxins alone or in combination. We observed a depolarisation when treated with Cyt1Aa and Cry4Aa. Toxin mixture assays only revealed a faster depolarisation of the membrane when the above two toxins were combined together, and a variety of responses with other toxin mixtures. Microspectrofluometry using the calcium probe Fura-2 on an immortal cell line from Anopheles gambiae (Ag55) showed massive effect of Cyt1Aa, but very little effect of the Cry toxins alone or in mixture. Microspectrofluometry experiments were also conducted on freshly dissociated cells from Aedes aegypti. Though these experiments are innovative and the results preliminary, it was observed that some cells responded differently to Cyt1Aa and Cry4Ba, showing the various ways these toxins affect cells, by inducing either intracellular calcium change, or by entirely losing the probe, or by cell lysis. The mixture of these toxins is very efficient and almost instantaneous.

Transfer of Microorganisms from Fomites to Hands and Risk Assessment of Contaminated and Disinfected Surfaces

It is now widely accepted that surface contamination plays an important role in the transmission of both respiratory and gastrointestinal infections in the domestic environment and community setting. The efficiency of transfer of a pathogen to the hand from a fomite is important in modeling transmission in microbial risk assessment models. The objective of this study was to use published literature to assess the role of fomites and hands in disease transmission, and to conduct fomite-to-finger transfer studies from various porous and nonporous fomites under different relative humidity condition using non-pathogenic strains of Escherichia coli, Staphylococcus aureus, MS2 coliphage, Bacillus thuringiensis spores, and poliovirus 1; to evaluate the persistence of bacteria and viruses on surfaces; to examine bacteria and virus transfer from treated surfaces; and to conduct a foodborne quantitative microbial risk assessment using Campylobacter jejuni from the data obtained in these studies. It was found that numerous factors influence the transfer efficiency of microorganisms, with moisture being the most important, with greater transfer under humid conditions. Other factors influencing transfer include drying time, contact time, pressure, friction, type of material, and porosity of the fomite. Percent transfer was greater under high relative humidity for both porous and nonporous surfaces. Most organisms on average had greater transfer under high relative humidity (40 - 65%) compared to low relative humidity (15 - 32%). Relative humidity and fomite type influenced the survival of all studied organisms; survival was greater on nonporous surfaces than those for porous surfaces. Test organisms were reduced up to 99.997% on the fomites after the surfaces were wiped with a disinfectant wipe. Microbial fomite-to-finger transfer from disinfectant wipe-treated surfaces were, lower than from non-treated surfaces. The disinfectant-wipe intervention reduced the risk of Campylobacter infection, illness, and death by 2 to 3 orders on all fomites. The disinfectant-wipe intervention reduced the annual risk of illness below the reported national average of diagnosed Campylobacteriosis cases 1.3E-04. This risk assessment demonstrates that the use of disinfectant wipes to decontaminate surface areas after chicken preparation reduces the risk of C. jejuni infections up to 99.2%.
