Chez la levure Saccharomyces cerevisiae, la croissance est orientée et nécessite l’apport de membranes et d’enzymes pour la synthèse de la paroi cellulaire. La régulation du transport des vésicules permettant cet apport est assuré par les GTPases de la famille Ypt/Rab. Sec4p, une Ypt/Rab GTPase, est impliquée dans l’exocytose en assurant la spécificité de l’ancrage des vésicules post-golgiennes envoyées aux sites de croissance. La régulation de son activité GTPase est essentielle pour sa fonction.Nous nous intéressons aux protéines Gyp5p et Gyl1p, deux membres de la famille des protéines activatrices des GTPases Ypt/Rab chez S. cerevisiae. Le laboratoire a montré l’implication du complexe Gyp5p-Gyl1p dans l’exocytose polarisée vraisemblablement par la régulation de Sec4p. Notre étude a montré une interaction directe in vitro de ces deux protéines, ainsi qu’une interdépendance pour une bonne localisation du complexe aux sites de croissance polarisée, c'est-à-dire au sommet du bourgeon durant la croissance apicale et au cou du bourgeon durant la cytocinèse. Cette localisation dépend de deux formines, d’éléments du polarisome et des câbles d’actine. De plus, nous avons montré par des expériences d’immunofluorescence et de microscopie électronique en collaboration avec J.-M. Verbavatz (iBiTec-S, CEA), que ces protéines sont transportées sur des vésicules de sécrétion jusqu’aux sites de croissance polarisée.Notre étude de l’interaction du complexe Gyp5p-Gyl1p avec Rvs167p, une protéine à domaine BAR (Bin1-Amphiphysin-Rvs167p) a montré que Gyp5p et Gyl1p sont nécessaires pour la bonne localisation de Rvs167p au sommet du petit bourgeon et que ces complexes se forment principalement dans des fractions enrichies en membrane plasmique. Pour mieux caractériser ces interactions, nous avons réalisé une mutation de la proline 473 dans le domaine SH3 de Rvs167p et des délétions des séquences riches en proline de Gyp5p et Gyl1p. Ces mutations entraînent un défaut d’interaction de Rvs167p avec Gyp5p et Gyl1p et la perte de la localisation de Rvs167p au sommet du petit bourgeon. Afin de comprendre la fonction de ces interactions, nous avons réalisé des expériences de microscopie électronique et des tests de sécrétion de l’endo-β-1,3-glucanase, Bgl2p dans une souche Δrvs167. Nous avons mis en évidence une accumulation de vésicules de sécrétion au niveau du petit bourgeon et un défaut de sécrétion de Bgl2p à 13°C dans cette souche. De plus, nous avons également observé une accumulation de vésicules de sécrétion dans une souche exprimant Rvs167p mutée pour la proline 473 et un défaut de sécrétion de Bgl2p dans une souche exprimant Gyp5p et Gyl1p dépourvues de leurs séquences riches en proline. Ces résultats montrent que Rvs167p joue un rôle dans l’exocytose polarisée au stade du petit bourgeon et que cette fonction dépend de son recrutement par Gyp5p et Gyl1p au sommet du petit bourgeon. / In Saccharomyces cerevisiae, growth is oriented and requires the contribution of membranes and enzymes for the synthesis of the cell wall. Regulation of vesicles transport allowing this contribution is provided by the Ypt/Rab GTPases family. Sec4p, a Ypt/Rab GTPase, is involved in exocytosis by controlling the tethering of post-Golgi vesicles at sites of growth. Regulation of Sec4p GTPase activity by is essential for its function.We studied the proteins Gyp5p and Gyl1p, two members of the Ypt/Rab GTPases activiting proteins (RabGAP) family in S. cerevisiae. Gyp5p and Gyl1p interact with Sec4p and are involved in the control of exocytosis at the small-bud stage. Our study showed that Gyp5p and Gyl1p interact directly in vitro and are interdependent for their correct localization to the sites of polarized growth, e.g. the bud tip during apical growth and the bud neck during cytokinesis. We showed that the localization of Gyp5p and Gyl1p to the sites of polarized growth depends on the formins Bni1 and Bnr1, but also on polarisome components and actin cables. Moreover, we showed by immunofluorescence and electron microscopy (in collaboration with J.-M. Verbavatz), that Gyp5p and Gyl1p are transported onto secretory vesicles to access the sites of polarized growth.We studied the interaction of Gyp5p and Gyl1p with Rvs167p, a BAR domain (Bin1-Amphiphysin-Rvs167p) protein and showed that Gyp5p and Gyl1p are necessary for the recruitment of Rvs167p to the small-bud tip. Both the mutation of the proline 473 in the SH3 domain of Rvs167p and the deletion of the proline-rich regions of Gyp5p and Gyl1p disrupt the interaction of Rvs167p with Gyp5p and Gyl1p and impair the localization of Rvs167p to the tips of small buds. Electron microscopy experiments unraveled an accumulation of secretory vesicles in small buds of rvs167Δcells and β-1,3-endoglucanase Bgl2p secretion assays showed Bgl2p secretion defects in cultures enriched in small buds at 13°C. In addition, an accumulation of secretory vesicles was observed in Rvs167pP473L strain, and Bgl2p secretion defect were found in strains expressing Gyp5p and Gyl1p deleted of their proline-rich sequences. These results show that Rvs167p plays a role in polarized exocytosis at the small bud stage and that its function in exocytosis depends on its recruitment to the tip of small buds by the RabGAP proteins Gyp5p and Gyl1p.

最小成本下,規格及X-bar-S管制圖之設計 / The design of economic statistical control charts and specification are both crucial research areas in industry. Furthermore, the determination of consumer and producer specifications is important to producer. In this study, we consider eight cost models including the consumer loss function and/or the producer loss function with the economic statistical X-bar and S charts or Shewhart-type economic X-bar and S charts. To determine the design parameters of the X-bar and S charts and consumer tolerance and/or producer tolerance, we using the Genetic Algorithm to minimizing expected cost per unit time. In the comparison of examples and sensitivity analyses, we found that the optimal design parameters of the Shewhart-type economic X-bar and S charts are similar to those of economic statistical X-bar and S control charts, and the expected cost per unit time may lower than the actual cost per unit time when the cost model only considering consumer loss or producer loss. When considering both consumer and producer tolerances in the cost model, the design parameters of the economic X-bar and S charts are not sensitive to the cost models. If the producer tolerance is smaller than the consumer tolerance, and the producer loss is smaller than the consumer loss, the optimal producer tolerance should be small.

Endocytosis is one of the most fundamental mechanisms by which the cell communicates with its surrounding. Specific signals are transduced through the cell membrane by a complex interplay between proteins and lipids. Clathrin depended endocytosis is one of important signalling pathways which leads to budding of the plasmalemma and a formation of endosomes. The FCHo2 is an essential protein at the initial stage of the this process. In is a membrane binding protein containing BAR (BIN, Amphiphysin, Rvs) domain which is responsible for a membrane binding. Although numerous valuable work on BAR proteins was published recently, the mechanistic description of a BAR domain functionality is missing. In present work we applied in vitro systems in order to gain knowledge about molecular basis of the activity of the FCHo2 BAR domain. In our studies we used supported lipid bilayers (SLBs) and lipid monolayers as s model membrane system.
The experiments were carried out with a minimal number of components including the purified FCHo2 BAR domain. Using SLBs we showed that the BAR domain can bind to entirely flat bilayers. We also demonstrated that these interactions depend on the negatively charged lipid species incorporated in the membrane. We designed an assay which allows to quantify the membrane tubulation. We found out that the interaction of the FCHo2 BAR domain with the lipid membrane is concentration dependent. We showed that an area of the bilayer deformed by the protein depends on the amount of the used BAR domain.
In order to study the relation between the mobility of lipids and the activity of FCHo2 BAR domain we designed a small-volume monolayer trough. The design of this micro-chamber allows for the implementation of the light microscopy. We demonstrated that the measured lipid diffusion in the monolayer by our new approach is in agreement with literature data. We carried out fluorescence correlation spectroscopy (FCS) experiments at different density of lipids at the water-air interface.We showed that the FCHo2 BAR domain binding affinity is proportional to the mean molecular area (MMA). We additionally demonstrated that the increased protein binding is correlated with the higher lipid mobility in the monolayer.
Additionally, by curing out high-speed atomic force microscopy (hsAFM) we acquired the structural information about FCHo2 BAR domains orientation at the membrane with a high spatio-temporal resolution. Obtained data indicate the BAR domains interact witheach other by many different contact sites what results in a variety of protein orientations in a protein assemble.

L'interférence entre la désintégration du méson $\Bs$ vers un état final propre de CP, directement ou via le mélange $\Bs-\Bsb$, donne lieu à une phase violant CP mesurable $\phi_s$, dont la valeur dans le Modèle Standard est $\phi_s^{MS} = (-0.0370\pm 0.0006)~\mathrm{rad}$. Cependant, des contributions de Nouvelle Physique peuvent intervenir dans ce processus et modifier la valeur de $\phi_s$. À l'heure actuelle, la mesure la plus précise de $\phi_s$ est donnée par l'expérience LHCb et la valeur moyenne mondiale est $\phi_s^{\rm{exp}} = (-0.021 \pm 0.032)~\mathrm{rad}$, dominée par l'incertitude statistique.Dans ce contexte, l'étude de la désintégration $\Bs \to \etac \phi$ est effectuée en utilisant les données de collisions $pp$ correspondant à une luminosité intégrée de~3.0\invfb, collectées avec le détecteur LHCb avec le Run~1 du LHC. However, such process may receive contributions from New Physics and change the value of $\phi_s$. At present, the most precise measurement of $\phi_s$ is given by the LHCb experiment and the world average is $\phi_s^{\rm{exp}} = (-0.021\pm 0.032)~\mathrm{rad}$, with uncertainty still dominated by the statistics.In this context, a study of $\Bs \to \etac \phi$ decays is performed using $pp$ collision data corresponding to an integrated luminosity of~3.0\invfb, collected with the LHCb detector during the Run~1 of the LHC. The observation of the decay $\Bs \to \etac \phi$ is reported, where the \etac meson is reconstructed in the $p\bar p$, $K^+K^-\pi^+\pi^-$, $\pi^+\pi^-\pi^+\pi^-$ and $K^+K^-K^+K^-$ decay modes and the $\phi(1020)$ in the $K^+ K^-$ decay mode. The decay $\Bs \to \jpsi \phi$ is used as a normalisation channel.The measured branching fraction is $\mathcal B (B^{0}_{s} \to \eta_{c} \phi) = \left(5.01 \pm 0.53 \pm 0.27 \pm 0.63 \right) \times 10^{-4}$, where the first uncertainty is statistical, the second systematic and the third uncertainty is due to the limited knowledge of the external branching fractions.

An exploratory study was undertaken to investigate the relationship between
personality preference groupings, as described by Jung’s (1959) type theory, and
emotional intelligence, as measured by Bar-On’s emotional intelligence quotient
(Bar-On, 1997). The sample group consisted of 1 121 recruitment candidates for
a South African investment bank. The sixteen personality types, as measured by
the Myers-Briggs Type Indicator, were represented in the sample. The statistical
analysis conducted for this study included comparison of means, correlation
analysis and analysis of variance. The results indicated statistically significant
relationships between the preferences of Extroversion, Judgement, their
combined preference grouping and emotional intelligence. No statistically
significant relationships were found between the preference groupings of Intuition
and Thinking, Sensing and Thinking, Intuition and Feeling, and Sensing and
Feeling. The preferred Feeling preference type consistently scored the lowest in

Previous issue date: 2011-03-03 / Eventually, violations of voltage limits at buses or admissible loadings of transmission
lines and/or power transformers may occur by the power system operation. If violations are
detected in the supervision process, corrective measures may be carried out in order to
eliminate them or to reduce their intensity. Loading restriction is an extreme solution and
should only be adopted as the last control action. Previous researches have shown that it is
possible to control constraints in electrical systems by changing the network topology, using
the technique named Corrective Switching, which requires no additional costs. In previous
works, the proposed calculations for verifying the ability of a switching variant in eliminating
an overload in a specific branch were based on network reduction or heuristic analysis. The
purpose of this work is to develop analytical derivation of linear equations to estimate current
changes in a specific branch (due to switching measures) by means of few calculations. For
bus-bar coupling, derivations will be based on short-circuit theory and Relief Function
methodology. For bus-bar splitting, a Relief Function will be derived based on a technique of
equivalent circuit. Although systems of linear equations are used to substantiate deductions,
its formal solution for each variant, in real time does not become necessary. A priority list of
promising variants is then assigned for final check by an exact load flow calculation and a
transient analysis using ATP Alternative Transient Program. At last, results obtained by

Cette thèse étudie deux modèles de réalisabilité pour la logique classique construits sur la sémantique des jeux HO, interprétant la logique, l'arithmétique et l'analyse classiques directement par des programmes manipulant un espace de stockage d'ordre supérieur.La non-innocence en jeux HO autorise les références d'ordre supérieur, et le non parenthésage révèle la CPS des jeux HO et fournit une catégorie de continuations dans laquelle interpréter le lambda-mu calcul de Parigot. Deux modèles de réalisabilité sont construits sur cette interprétation calculatoire directe des preuves classiques.Le premier repose sur l'orthogonalité, comme celui de Krivine, mais il est simplement typé et au premier ordre. En l'absence de codage de l'absurdité au second ordre, une mu-variable libre dans les réaliseurs permet l'extraction. Nous définissons un bar-récurseur et prouvons qu'il réalise l'axiome du choix dépendant, utilisant deux conséquences de la structure de CPO du modèle de jeux: toute fonction sur les entiers (même non calculable) existe dans le modèle, et toute fonctionnelle sur des séquences est Scott-continue. La bar-récursion est habituellement utilisée pour réaliser intuitionnistiquement le « double negation shift » et en déduire la traduction négative de l'axiome du choix. Ici, nous réalisons directement l'axiome du choix dans un cadre classique.Le second, très spécifique au modèle de jeux, repose sur des conditions de gain: des ensembles de positions d'un jeu munis de propriétés de cohérence. Un réaliseur est alors une stratégie dont les positions sont toutes gagnantes. / This thesis investigates two realizability models for classical logic built on HO game semantics. The main motivation is to have a direct computational interpretation of classical logic, arithmetic and analysis with programs manipulating a higher-order store.Relaxing the innocence condition in HO games provides higher-order references, and dropping the well-bracketing of strategies reveals the CPS of HO games and gives a category of continuations in which we can interpret Parigot's lambda-mu calculus. This permits a direct computational interpretation of classical proofs from which we build two realizability models.The first model is orthogonality-based, as the one of Krivine. However, it is simply-typed and first-order. This means that we do not use a second-order coding of falsity, and extraction is handled by considering realizers with a free mu-variable. We provide a bar-recursor in this model and prove that it realizes the axiom of dependent choice, relying on two consequences of the CPO structure of the games model: every function on natural numbers (possibly non computable) exists in the model, and every functional on sequences is Scott-continuous. Usually, bar-recursion is used to intuitionistically realize the double negation shift and consequently the negative translation of the axiom of choice. Here, we directly realize the axiom of choice in a classical setting.The second model relies on winning conditions and is very specific to the games model. A winning condition is a set of positions in a game which satisfies some coherence properties, and a realizer of a formula is then a strategy which positions are all winning.

São estudados alguns aspectos das álgebras alternativas, como o bar-radical de uma álgebra bárica alternativa e as identidades de grau 4 e 5 nas álgebras de Cayley-Dickson. Neste estudo fazemos uso da decomposição de Peirce e de diversas propriedades importantes das álgebras alternativas. Concluímos mostrando que as únicas identidades de grau 4 são as triviais e as de grau 5 são conseqüência de outras duas identidades conhecidas / We studied some aspects of alternative algebras, in special the bar radical of an alternative baric algebra and identities of degree 4 and 5 of Cayley-Dickson algebras. We made significant use of the Peirce decomposition and several properties of the alternative algebras, in order to show that the only identities of degree four are the trivial ones, and the identities of degree five are consequences of other two known identities.

The aim of my master´s thesis is a new sports center project in Havlíčkův Brod in neighborhood of sports complex Na Losích on plot no. 561/1, 564/17. The land is slightly sloping and their total area is 12574 square meters. The building of the sports center is designed as a two-storey, with a cellar, with a dominant part of the multipurpose sports hall. The vertical supports construction consists of a reinforced concrete monolithic combined system. In the sports hall, the supports construction consists of a skeletal structure made of steel rolled elements in combination with reinforced concrete monolithic columns. Vertical peripheral construction are equipped with thermal insulation system with perforated sheet metal. The foundation building construction is designed as a monolithic foundation slab serves as a white bath. The horizontal supports construction are designed from prestressed SPIROLL slabs. The roof construction is designed as a single-layer vegetation flat roof. The roof supports construction of the sports hall is designed from steel-truss girder with supporting trapezoidal sheet. The designed sports center has 3 functional parts. It is a multifunction sports hall with facilities for athletes on the underground floor. The second functional part on the first floor is a sports bar connected with the main entrance hall and a bleacher with a capacity of 400 persons, which is located in the sports hall. The third part consists of a separate section on the first floor for athletes using the adjacent outdoor playground and the main part is located on the second floor, which serves as a facilities for TJ Jiskra club management, then there is a VIP zone, a clubroom and a small exercise room with facilities.

Prefabricerade sandwichelement kommer att produceras till Linnéskolan i Älmhult av Torps Byggelement AB. Ett sandwichelement består av två lager betong (fasad och bärande skikt) med isolering mellan de två skikten. Dessa sandwichelement hade till en början förankringssystemet Thermopin tillverkade av B.T. Innovation, men på grund av produktion- och leveransproblem så ersattes detta förankringssystem mot Isolink som tillverkas av Schöck. Isolink och Thermopin är två förankringssystem tillverkade av glasfiberarmerade plaststänger (GFRP). Sandwichelementen som kommer att användas i Linnéskolan använder sig av isoleringen Kooltherm tillverkad av Kingspan. I detta arbete jämförs förankringssystemens bärförmågor i form av utdragskapacitet. Beräkningar för kapacitet mot vidhäftningsbrott, utdragsbrott och brott i stångmaterial görs för de båda systemen. Utöver detta testas även bärförmågan i form av utdragskapacitet för isolering Kooltherm. 12 testprover, som tillverkades och levererades av Torps Byggelement AB, utsätts för utdragslaster till brott uppstod. Endast de förankringssystem som bär vinkelrät mot skikten testades. De 12 testproverna var uppdelade på följande vis: Fyra tester med Thermopin, fyra tester med Isolink samt fyra tester med endast isoleringen Kooltherm. De testprover med Isolink-systemet gav ett utdragsbrott vid 18.9 kN. Thermopin gav ett spjälkningsbrott vid 14.9 kN. Kooltherm gav brott i isoleringen vid 1.3 kN. Då Isolink fick ett vidhäftningsbrott är det viktigt med en god vidhäftning mellan förankringssystemet och omkringliggande betong. Vibrering av betongen i flera steg vid användning av Isolink kan därför anses vara viktig. Thermopin fick ett spjälkningsbrott då tillverkarens minsta avstånd till kant inte uppfylldes med testproverna. Det går med stor sannolikhet att säga att Thermopin inte hade fått ett utdragsbrott även om minsta avstånd till kant uppfylldes, mest troligt är att ett konbrott hade uppstått. / Prefabricated sandwich elements will be produced to Linnéskolan in Älmhult by Torps Byggelement AB. A sandwich element is built up by two layers of concrete (facade- and load bearing layer) with insulation separating the two layers. These sandwich elements were intended to have the anchors Thermopin produced by B.T. Innovation but due to delivery problems, this connection system was replaced with Isolink made by Schöck. Isolink and Thermopin are two anchors consisting of glass fiber reinforced plastic bars (GFRP). The sandwich elements that will be used in Linnéskolan will have the insulation Kooltherm made by Kingspan. This work is comparing the two connection systems in load bearing capacity by pull-out load tests. Calculations for pull-out failure, concrete failure and anchor failure were done for both of the connection systems. The load bearing capacity was also to be checked for the Kooltherm insulation by pull-out load tests. 12 test specimens were manufactured and delivered by Torps Byggelement AB. Only the anchors perpendicular to the concrete layers were tested. The 12 test specimens were divided as follows: four tests with Thermopin, four tests with Isolink and four tests without a connection system with only the Kooltherm insulation. The test specimens with Isolink gave a pull-out load at 18.9 kN. Thermopin gave a splitting failure at 14.9 kN. Kooltherm insulation gave a failure in the insulation at 1.3 kN. Due to Isolink getting a pull-out load failure, it is important that the bond between the anchor and the surrounding concrete is strong. Compacting of the concrete in multiple steps when using the Isolink-system can be considered important. The test specimens with the Thermopin-system got a splitting failure due to the manufacturers requirement on minimum edge distance not being achieved in the test specimens. It is proper to state that it is very unlikely that a pull-out failure would occur in the Thermopin test specimens even if the minimum edge distance was achieved. Most likely a concrete-cone failure would have occurred.
