Etude des programmes transcriptionnels impliqués dans le développement des neurones somatosensoriels et leur état après axotomie / Transcription programs in the development of somatosensory neurons and their state after axotomy

La sensation du toucher permet de détecter des stimuli mécaniques via des neurones mécanosensitifs dont les corps cellulaires sont localisés dans le ganglion rachidien dorsal. Ces neurones projettent vers la peau en périphérie et font leurs synapses avec les interneurones dorsaux de la moelle épinière. L'étude de ces neurones était difficile à cause du manque de marqueurs spécifiques pour ces neurones, jusqu'à la découverte du gène MafA dans le ganglion rachidien dorsal. En effet, mon équipe a montré que MafA est un marqueur spécifique des neurones mécanosensitifs à bas seuil de type Rapidly adapting. Le gène c-Maf, de la même famille que MafA, est aussi exprimé dans ces neurones et il est l'acteur principal de leur développement et de leur fonction. Afin de comprendre comment c-Maf contrôle le développement des neurones somatosensoriels, la première partie de mon travail visait à identifier de nouveaux gènes cibles du facteur de transcription c-Maf et à savoir comment l'expression de ce dernier est régulée dans les neurones du ganglion rachidien dorsal. Concernant la recherche des gènes cibles de c-Maf, j'ai réalisé l'étude de l'expression de gènes candidats dans le contexte de perte de fonction de c-Maf chez la souris. J'ai pu identifier deux cibles : p-cadhérine et mab21/L2, parmi une liste de gènes candidats. J'ai par la suite analysé l'expression de P-cadhérine au cours du développement et j'ai observé qu'elle est exprimée dans la sous-population de neurones sensoriels myélinisés exprimant c-Maf ainsi que dans certains interneurones des laminae III/IV de la moelle épinière. Une expression particulière de P-cadherine est observée dans les cellules des capsules frontières dans les zones d'entrée et de sortie des racines rachidiennes. Suite à ces observations, nous avons émis l'hypothèse suivante : l'expression de la p-cadhérine est régulée par c-Maf dans les neurones sensoriels ainsi que dans les interneurones pour assurer la connexion synaptique entre eux. Concernant, la régulation du gène c-maf, la question reste ouverte. La deuxième partie de mon étude concernait l'analyse du rôle de facteurs de transcription MafA, c-Maf, Runx3 et Er81 dans la plasticité neuronale induite chez la souris adulte après axotomie du nerf sciatique. Ces facteurs sont impliqués dans le développement des neurones somatosensoriels. Cette analyse a montré que l'expression de MafA et Er81 diminue trois jours après axotomie du nerf périphérique, alors que celle de Runx3 et de c-Maf n'est pas affectée. On peut suggérer que chez l'adulte, la régulation de l'expression de MafA et Er81 dépend des facteurs neurotrophiques libérés par les cibles de ces neurones tandis que celle de c-Maf et Runx3 en est indépendante. / The sense of touch relies on the detection of mechanical stimuli by specialized cutaneous mechanosensory neurons whose cell bodies are located in the dorsal root ganglia. These neurons project peripherally to the skin and synapse on target interneurons in the spinal cord. Until the discovery of MafA expression in the dorsal root ganglion, the lack of molecular markers of mechanoreceptor neurons has made it difficult to analyze the development of these neurons. My team showed that MafA is a specific molecular marker for low-threshold mechanoreceptor neurons RAM. C-Maf gene is a member of the Maf family and it is expressed in the MafA sensory neurons. The transcription factor c-Maf controls touch receptor development and function.In order to understand how c-Maf controls somatosensory neurons development, the first objective of my study was to find new targets for c-Maf transcription factor and to know how c-Maf expression is regulated in the dorsal root ganglion. Therefore, I have analysed the expression of different candidate genes in a loss of function context of c-Maf in the mice. I identified two targets: p-cadherin and Mab21/L2 among a list of candidates. Then, I analysed the p-cadherin expression during development and found that this target of c-Maf is expressed in a sub-population of c-Maf sensory neurons and interneurons of the laminae III/IV of the spinal cord. A particular expression of p-cadherin was noticed in the boundary cap cells at the dorsal root entry zone and the motor exit point of the spinal cord. These observations let us put the following hypothesis: c-Maf regulates p-cadherin expression in the sensory neurons and the interneurons to enable specific connections between these neurons. No identified factors were found to regulate c-Maf expression. In the second part of my study, I focused my efforts on the analysis of the role of MafA, c-MAf, Runx3 and Er81 transcription factors in neuronal plasticity induced in the adult mice three days after sciatic nerve axotomy. These factors are involved in the development of somatosensory neurons. The analysis showed that MafA and Er81 expression are down-regulated after peripheral nerve axotomy but the c-Maf and Runx3 expression did not change. We suggest that at adult stage the regulation of MafA and Er81 expression depend on neurotrophic factors released by the targets of these neurons but it's not the case for c-Maf and Runx3 expression.

Identification of Epo-independent red cell progenitors : the E-cad+ progenitors

Erythrozyten zählen zu den am häufigsten vorkommenden terminal differenzierten Zelltypen des menschlichen Körpers. Durchschnittlich werden täglich ca. 2 x 1011 von ihnen im Körper eines erwachsenen Menschen produziert. Die reifen Erythrozyten entstehen aus multipotenten hämatopoetischen Stammzellen, die über Stadien von erythroiden Vorläuferzellen, erst den sogenannten burst forming units-erythroid (BFU-E) und später den colony forming units-erythroid (CFU-E), zu kernlosen hämoglobinisierten Zellen differenzieren. <br /> <br /> Für die Untersuchung der molekularen Mechanismen der humanen Erythropoese ist die effektive in vitro Amplifizierung einer weitgehend homogenen Population der Vorläuferzellen der einzelnen Entwicklungsstadien notwendig. Den Wachstumsfaktoren stem cell factor (SCF) und Erythropoietin (Epo) fällt dabei eine entscheidende Rolle zu. Unter ihrem synergistischen Einfluß lassen sich Epo-abhängige Zellpopulationen, die sich aus BFU-E und CFU-E Typ Zellen zusammensetzen, ausreichend amplifizieren (Panzenböck et al., 1998). Freyssinier et al., 1999 beschrieb erstmals die Isolierung einer Epo-unabhängigen Population von Vorläuferzellen (CD36+ Vorläuferzellen), die ebenfalls erythroide Eigenschaften aufweisen.<br /> <br /> Ziel dieser Arbeit war die Isolierung und Charakterisierung von Epo-unabhängigen Vorläuferzellen, die eine frühe erythroide und möglichst homogene Vorläuferzellpopulation darstellen und möglicherweise ein höheres Proliferationspotential aufweisen.<br /> <br /> Für die Identifizierung der Epo-unabhängigen Vorläuferzellen, wurden CD34+ Zellen aus Nabelschnurblut aufgereinigt und unter serumfreien Kulturbedingungen und unter Zusatz der Wachstumsfaktoren SCF, Interleukin 3 (IL-3) und eines Fusionsproteins aus IL-6 und löslichem IL-6 Rezeptor (hyper-IL-6) über einen Zeitraum von 8 Tagen kultiviert. Anschließend wurde eine Population von E-cadherin positiven (E-cad+) Zellen über immunomagnetische Selektion isoliert. Diese neu gewonnenen Epo-unabhängigen E-cad+ Vorläuferzellen wurden hinsichtlich ihres proliferativen Potentials und ihrer Differenzierungseigenschaften mit SCF/Epo-Vorläuferzellen und CD36+ Vorläuferzellen verglichen. Von allen drei Zelltypen wurden des weiteren detailierte molekulargenetische Analysen mittels DNA microarray Technologie durchgeführt und die resultierenden Genexpressionsmuster miteinander verglichen.<br /> <br /> Die Ergebnisse zeigen, dass die E-cad+ Vorläuferzellen eine frühe, weitgehend homogene Epo-unabhängige Population vom BFU-E Typ darstellen und durch entsprechende Änderungen der Kulturbedingungen zu einer in vitro Differenzierung angeregt werden können. Die E-cad+ Vorläuferzellen sind hinsichtlich ihres proliferativen Potentials, ihrer Reaktion auf verschiedene Wachstumsfaktoren, der Expression spezifischer Oberflächenmoleküle und ihrer Genexpressionsmuster mit SCF/Epo-Vorläuferzellen und CD36+ Vorläuferzellen vergleichbar.<br /> <br /> Aufgrund der Identifizierung unterschiedlich exprimierter Gene zwischen den Epo-unabhängigen E-cad+ und den Epo-abhängigen SCF/Epo Vorläuferzellen konnten Kanditatengene wie Galectin-3, Cyclin D1, der Anti-Müllerian Hormonrezeptor, Prostata-Differenzierungsfaktor und insulin-like growth factor binding protein 4 identifiziert werden, die als potentielle Regulatoren der Erythropoese in Betracht kommen könnten. Es konnte weiterhin gezeigt werden, dass CD36+ Vorläuferzellen, die aus der selben Zellpopulation wie die E-cad+ Vorläuferzellen immunomagnetisch selektioniert wurden, eine heterogene Population darstellen, die sowohl E-cadherin positive als auch negative Zellen enthält. Die Analyse der Genexpressionsmuster zeigte, dass in den CD36+ Vorläuferzellen zwar auch die Expression erythroid-spezifischen Gene nachgewiesen werden kann, hier aber im Gegensatz zu den E-cad+ Vorläuferzellen auch für Megakaryozyten spezifische Gene stark exprimiert sind.<br /> <br /> Die Ergebnisse dieser Arbeit tragen zu einem neuen Modell der in vivo Abläufe der Entwicklung roter Blutzellen bei und werden der weiteren Untersuchung der molekularen Mechanismen der Erythropoese dienen. / Red cell development in adult humans results in the mean daily production of 2x1011 erythrocytes. Mature hemoglobinized and enucleated erythrocytes develop from multipotent hematopoietic stem/progenitor cells through more committed progenitor cell types such as BFU-E and CFU-E. The studies on the molecular mechanisms of erythropoiesis in the human system require a sufficient number of purified erythroid progenitors of the different stages of erythropoiesis. Primary human erythroid progenitors are difficult to obtain as a homogenous population in sufficiently high cell numbers. Various culture conditions for the in vitro cell culture of primary human erythroid progenitors have been previously described. Mainly, the culture resulted in the generation of rather mature stages of Epo-dependent erythroid progenitors. In this study our efforts were directed towards the isolation and characterization of more early red cell progenitors that are Epo-independent.<br /> <br /> To identify such progenitors, CD34+ cells were purified from cord blood and cultured under serum free conditions in the presence of the growth factors SCF, IL-3 and hyper-IL-6, referred to as SI2 culture conditions. By immunomagnetic bead selection of E-cadherin (E-cad) positive cells, E-cad+ progenitors were isolated. These Epo-independent E-cad+ progenitors have been amplified under SI2 conditions to large cell numbers. The E-cad+ progenitors were characterized for surface antigen expression by flow cytometry, response to growth factors in proliferation assay and for their differentiation potential into mature red cells. Additionally, the properties of E-cad+ progenitors were compared to those of two other erythroid progenitors: Epo-dependent progenitors described by Panzenböck et al. (referred to as SCF/Epo progenitor), and CD36+ progenitors described by Freyssinier et al. (Panzenböck et al., 1998; Freyssinier et al., 1999). Finally, the gene expression profile of E-cad+ progenitors was compared to the profiles of SCF/Epo progenitors and CD36+ progenitors using the DNA microarray technique.<br /> <br /> Based on our studies we propose that Epo-independent E-cad+ progenitors are early stage, BFU-E like progenitors. They respond to Epo, despite the fact that they were generated in the absence of Epo, and can completely undergo erythroid differentiation. Furthermore, we demonstrate that the growth properties, the growth factor response and the surface marker expression of E-cad+ progenitors are similar to those of the SCF/Epo progenitors and the CD36+ progenitors. By the comparison of gene profiles, we were also able to demonstrate that the Epo-dependent and Epo-independent red cell progenitors are very similar. Analyzing the molecular differences between E-cad+ and SCF/Epo progenitors revealed several candidate genes such as galectin-3, cyclin D1, AMHR, PDF and IGFBP4, which are potential regulators involved in red cell development. We also demonstrate that the CD36+ progenitors, isolated by immunomagentic bead selection, are a heterogeneous progenitor population containing an E-cad+ and an E-cad- subpopulation. Based on their gene expression profile, CD36+ progenitors seem to exhibit both erythroid and megakaryocytic features.<br /> <br /> These studies led to a more updated model of erythroid cell development that should pave the way for further studies on molecular mechanisms of erythropoiesis.

Die Rolle des N-Cadherin im Mammakarzinom

Trotz aller Fortschritte in Diagnostik und Behandlung bleibt die Therapie von BrustkrebspatientInnen – gerade bei tripelnegativen Mammakarzinomen, die nicht auf eine hormonelle Therapie ansprechen – eine Herausforderung. Eine wachsende Zahl von Belegen spricht dafür, dass Cadherine nicht nur in physiologischen Prozessen wie der Embryogenese, sondern auch in pathologischen Prozessen wie der Tumorprogression eine Rolle spielen, was diese Moleküle zu potenziellen Zielen einer targetspezifischen Tumortherapie macht. Im Mausmodell konnte bislang gezeigt werden, dass eine Herabregulierung von N-Cadherin das Mammakarzinomwachstum in vivo hemmt. N-Cadherin wird auch in humanem Brustkrebsgewebe aberrant exprimiert. In nur schwach invasiven Brustkrebszelllinien führte eine Überexpression von N-Cadherin in vitro zu einer Steigerung des migratorischen und invasiven Verhaltens der Zellen. In dieser Arbeit wurde an der humanen Mammakarzinomzelllinie SUM149PT der Einfluss einer Defizienz von N-Cadherin auf die Expression von E- und VE-Cadherin, sowie auf das Proliferations-, Migrations- und Invasionsverhalten der Zellen in vitro untersucht. Eine Charakterisierung der humanen Brustkrebszelllinien SUM149PT, der aus ihr hervorgegangenen mit shRNA transduzierten N-Cadherin-Knock-down-Klone und der Scramble-Kontrollen fand mittels Western Blot, RT-PCR, q-PCR und Immunfluoreszenzanalysen statt. Das Proliferationsverhalten der Zelllinien wurde mit Hilfe von Verdopplungszeitbestimmungen und Sphäroidversuchen analysiert. Um den Einfluss einer N-Cadherin-Defizienz auf die Migration zu untersuchen, wurden auf dem Prinzip der Boyden-Chamber basierende Versuche, sowie in vitro Wundheilungsversuche durchgeführt. Auch die Invasionsversuche basierten auf dem Prinzip der Boyden-Chamber. Eine zusätzliche Beschichtung mit Matrigel simulierte hier die extrazelluläre Matrix. Bei den Untersuchungen mittels Western Blot, RT-PCR und q-PCR wurde deutlich, dass die N-Cadherin defizienten Klone keine Veränderung in der E-Cadherin-Expression, jedoch interessanterweise eine starke Herabregulierung von VE-Cadherin aufweisen. Bei den Immunfluoreszenzanalysen wiesen SUM149PT und Scramble-Kontrollen eine Expression von N-Cadherin in der Zellmembran auf, während die N-Cadherin-defizienten Klone keine N-Cadherin-Expression zeigten. Wie schon in Western-Blot, RT-PCR und q-PCR zeigten die N-Cadherin defizienten Klone im Vergleich zu den SUM149PT und den Scramble-Kontrollen keine veränderte E-Cadherin-Expression. Bei Einsatz eines VE-Cadherin-Anitkörpers zeigten die N-Cadherin defizienten Klone keine Expression von VE-Cadherin, während SUM149PT und Scramble eine deutliche VE-Cadherin-Expression in der Zellmembran zeigten. Daraus lässt sich schließen, dass N-Cadherin möglicherweise die Expression von VE-Cadherin beeinflussen kann. In der Verdopplungszeitbestimmung ließen sich keine signifikanten Änderungen des Wachstumsverhaltens zwischen N-Cadherin defizienten Klonen und Kontrollzelllinien nachweisen. Um auch das dreidimensionale Wachstum der Zellen zu untersuchen, wurden Sphäroidversuche durchgeführt. Alle untersuchten Zelllinien bildeten stabile Sphäroide aus, die jedoch einen großen Saum an nicht integrierten Zellen aufwiesen. Es zeigte sich kein signifikanter Unterschied im Sphäroidwachstum zwischen N-Cadherin defizienten Klonen und Kontrollzelllinien. Die Herabregulierung von N-Cadherin scheint in humanen Mammakarzinomzelllinien in vitro keinen Einfluss auf das Wachstumsverhalten der Zellen zu haben. Im Boyden-Chamber-Assay zeigte sich, dass eine Herabregulierung von N-Cadherin bei der humanen Mammakarzinomzelllinie SUM149PT in vitro zu einer signifikanten Verringerung des Migrationspotenzials führt. Dieses Ergebnis wurde beim in vitro Wundheilungsversuch bestätigt. Hier zeigten die N-Cadherin defizienten Klone eine signifikant verringerte Flächenbedeckungsrate – also eine signifikant verringerte Geschwindigkeit mit der die aufeinander zuwanderndern Zellen eine freie Fläche bedecken – als die Kontrollzelllinien. Im Invasionsversuch zeigten die N-Cadherin defizienten Klone im Vergleich zu den Kontrollen eine hochsignifikant verringerte Invasivität. Eine Herabregulierung von N-Cadherin bei der humanen Mammakarzinomzelllinie SUM149PT führt also in vitro zu einer signifikanten Verringerung des Invasionspotenzials. Die Ergebnisse dieser Arbeit lassen den Schluss zu, dass N-Cadherin bei humanen Mammakarzinomzelllinien durch seinen positiven Einfluss auf Migration und Invasion der Zellen eine große Rolle beim Prozess des invasiven Wachstums und der Metastasierung spielt. Eine Anti-N-Cadherin-Therapie könnte auch bei BrustkrebspatientInnen, gerade mit tripelnegativen Mammakarzinomen, neue Möglichkeiten der Behandlung eröffnen. / Breast cancer is the most common cancer type for women and the most frequent type after lung cancer overall. In spite of all improvements in clinical diagnostics and treatment, the therapy of patients with breast cancer remains a difficult task. Therefore it is important to find new strategies of treatment, especially for patients with triple negative breast cancer that does not respond to hormone therapy. There is growing evidence that cadherins are not only important in physiological processes like embryogenesis but also in pathological processes like tumour progression. A so called cadherin switch has been implicated in carcinogenesis, in particular the loss of E-cadherin and the upregulation of N-cadherin. Thus, these molecules are an interesting subject of studies and a potential target for specific cancer therapy. It was shown previously that efficient silencing of N-cadherin in murine breast cancer cells inhibits tumour growth in vivo. N-cadherin is also aberrantly expressed in human breast cancer tissue. An overexpression of N-Cadherin in breast cancer cells enhances tumour cell motility, migration and invasion in vitro. In this thesis the impact of N-cadherin deficiency on the expression level of E- and VE-cadherin as well as its effect on proliferation, migration and invasion behaviour of breast cancer cells in vitro was investigated. The human breast cancer cell line SUM149PT, its N-cadherin deficient clones and scramble controls were characterised by Western Blot, RT-PCR, q-PCR and immunofluorescence staining. To analyse the impact of N-cadherin on the proliferation behaviour doubling time proliferation assays and spheroid assays were performed. A Boyden chamber assay and an in vitro wound healing assay were performed to investigate the effect of N-cadherin deficiency on cell migration. Furthermore, a Boyden chamber assay with a coating of Matrigel was used to study the invasion potential of the N-cadherin deficient cell clones and control cell lines. The results of Western Blot, RT-PCR and q-PCR show that silencing of N-cadherin expression in SUM149PT cells had no influence on E-cadherin expression, but significantly decreased VE-cadherin expression. With immunofluscence staining it was evidenced that SUM149PT and scramble controls express N-cadherin in the cell membrane, whereas N-cadherin deficient clones do not show any N-cadherin expression. There was no difference seen in E-cadherin expression between N-cadherin deficient clones and control cell lines. While SUM149PT and scramble controls expressed VE-cadherin in the cell membrane, N-cadherin deficient clones did not express VE-cadherin. These results suggest that N-cadherin may have an influence on the expression of VE-cadherin. No significant alteration in proliferation behaviour between N-cadherin deficient clones and controls could be shown with the doubling time proliferation assay. To investigate the impact of N-cadherin on the three-dimensional growth spheroid assays were performed. All cell lines formed stable speroids, however fringes of many not incorporated cells were found. There was no difference in spheroidgrowth between N-cadherin deficient clones and control cell lines. No influence of N-cadherin on cell proliferation in vitro could be seen. With the help of Boyden chamber assays it was shown that silencing of N-cadherin in the human breast cancer cell line SUM149PT results in a significant decrease of migration potential in vitro. This was confirmed with an in vitro wound healing assay. N-cadherin deficient clones showed a significantly reduced surface coverage rate than SUM149PT and scramble controls. A significant decrease of invasive cells in N-cadherin deficient clones in comparison to the controls was revealed in invasion assays. Thus silencing of N-Cadherin in the human breast cancer cell line SUM149PT leads to a significantly decreased invasion potential in vitro. The results presented this thesis provide evidence that N-cadherin is involved in invasive tumour progression and metastasis of the human breast cancer cell line SUM149PT by promoting cell migration and invasion. Anti-N-cadherin strategies could thus be a potential target specific therapy of cancer patients, particularly with triple negative breast cancer.

Bei den Immunfluoreszenzanalysen wiesen SUM149PT und Scramble-Kontrollen eine Expression von N-Cadherin in der Zellmembran auf, während die N-Cadherin-defizienten Klone keine N-Cadherin-Expression zeigten. Wie schon in Western-Blot, RT-PCR und q-PCR zeigten die N-Cadherin defizienten Klone im Vergleich zu den SUM149PT und den Scramble-Kontrollen keine veränderte E-Cadherin-Expression. Bei Einsatz eines VE-Cadherin-Anitkörpers zeigten die N-Cadherin defizienten Klone keine Expression von VE-Cadherin, während SUM149PT und Scramble eine deutliche VE-Cadherin-Expression in der Zellmembran zeigten. Daraus lässt sich schließen, dass N-Cadherin möglicherweise die Expression von VE-Cadherin beeinflussen kann. In der Verdopplungszeitbestimmung ließen sich keine signifikanten Änderungen des Wachstumsverhaltens zwischen N-Cadherin defizienten Klonen und Kontrollzelllinien nachweisen. Um auch das dreidimensionale Wachstum der Zellen zu untersuchen, wurden Sphäroidversuche durchgeführt. Alle untersuchten Zelllinien bildeten stabile Sphäroide aus, die jedoch einen großen Saum an nicht integrierten Zellen aufwiesen. Es zeigte sich kein signifikanter Unterschied im Sphäroidwachstum zwischen N-Cadherin defizienten Klonen und Kontrollzelllinien. Die Herabregulierung von N-Cadherin scheint in humanen Mammakarzinomzelllinien in vitro keinen Einfluss auf das Wachstumsverhalten der Zellen zu haben. Im Boyden-Chamber-Assay zeigte sich, dass eine Herabregulierung von N-Cadherin bei der humanen Mammakarzinomzelllinie SUM149PT in vitro zu einer signifikanten Verringerung des Migrationspotenzials führt. Dieses Ergebnis wurde beim in vitro Wundheilungsversuch bestätigt. Hier zeigten die N-Cadherin defizienten Klone eine signifikant verringerte Flächenbedeckungsrate – also eine signifikant verringerte Geschwindigkeit mit der die aufeinander zuwanderndern Zellen eine freie Fläche bedecken – als die Kontrollzelllinien. Im Invasionsversuch zeigten die N-Cadherin defizienten Klone im Vergleich zu den Kontrollen eine hochsignifikant verringerte Invasivität. Eine Herabregulierung von N-Cadherin bei der humanen Mammakarzinomzelllinie SUM149PT führt also in vitro zu einer signifikanten Verringerung des Invasionspotenzials. Die Ergebnisse dieser Arbeit lassen den Schluss zu, dass N-Cadherin bei humanen Mammakarzinomzelllinien durch seinen positiven Einfluss auf Migration und Invasion der Zellen eine große Rolle beim Prozess des invasiven Wachstums und der Metastasierung spielt. In spite of all improvements in clinical diagnostics and treatment, the therapy of patients with breast cancer remains a difficult task. Therefore it is important to find new strategies of treatment, especially for patients with triple negative breast cancer that does not respond to hormone therapy. There is growing evidence that cadherins are not only important in physiological processes like embryogenesis but also in pathological processes like tumour progression. A so called cadherin switch has been implicated in carcinogenesis, in particular the loss of E-cadherin and the upregulation of N-cadherin. Thus, these molecules are an interesting subject of studies and a potential target for specific cancer therapy. It was shown previously that efficient silencing of N-cadherin in murine breast cancer cells inhibits tumour growth in vivo. N-cadherin is also aberrantly expressed in human breast cancer tissue. An overexpression of N-Cadherin in breast cancer cells enhances tumour cell motility, migration and invasion in vitro. In this thesis the impact of N-cadherin deficiency on the expression level of E- and VE-cadherin as well as its effect on proliferation, migration and invasion behaviour of breast cancer cells in vitro was investigated. The human breast cancer cell line SUM149PT, its N-cadherin deficient clones and scramble controls were characterised by Western Blot, RT-PCR, q-PCR and immunofluorescence staining. To analyse the impact of N-cadherin on the proliferation behaviour doubling time proliferation assays and spheroid assays were performed. A Boyden chamber assay and an in vitro wound healing assay were performed to investigate the effect of N-cadherin deficiency on cell migration. Furthermore, a Boyden chamber assay with a coating of Matrigel was used to study the invasion potential of the N-cadherin deficient cell clones and control cell lines. The results of Western Blot, RT-PCR and q-PCR show that silencing of N-cadherin expression in SUM149PT cells had no influence on E-cadherin expression, but significantly decreased VE-cadherin expression. With immunofluscence staining it was evidenced that SUM149PT and scramble controls express N-cadherin in the cell membrane, whereas N-cadherin deficient clones do not show any N-cadherin expression. There was no difference seen in E-cadherin expression between N-cadherin deficient clones and control cell lines. While SUM149PT and scramble controls expressed VE-cadherin in the cell membrane, N-cadherin deficient clones did not express VE-cadherin. These results suggest that N-cadherin may have an influence on the expression of VE-cadherin. No significant alteration in proliferation behaviour between N-cadherin deficient clones and controls could be shown with the doubling time proliferation assay. To investigate the impact of N-cadherin on the three-dimensional growth spheroid assays were performed. All cell lines formed stable speroids, however fringes of many not incorporated cells were found. There was no difference in spheroidgrowth between N-cadherin deficient clones and control cell lines. No influence of N-cadherin on cell proliferation in vitro could be seen. With the help of Boyden chamber assays it was shown that silencing of N-cadherin in the human breast cancer cell line SUM149PT results in a significant decrease of migration potential in vitro. This was confirmed with an in vitro wound healing assay. N-cadherin deficient clones showed a significantly reduced surface coverage rate than SUM149PT and scramble controls. A significant decrease of invasive cells in N-cadherin deficient clones in comparison to the controls was revealed in invasion assays. Thus silencing of N-Cadherin in the human breast cancer cell line SUM149PT leads to a significantly decreased invasion potential in vitro. The results presented this thesis provide evidence that N-cadherin is involved in invasive tumour progression and metastasis of the human breast cancer cell line SUM149PT by promoting cell migration and invasion. Investigating the potentially expanded target repertoire of murinized Internalin of Listeria monocytogenes

The ability of intracellular pathogens to invade and spread from non-phagocytic cell to another is an imperative mechanism broadly investigated in cellular biology. Listeria monocytogenes (Lm) –one example of intracellular pathogens, invades specifically human epithelial cells using its surface proteins Internalin A (InlA) and InlB, respectively. InlA alone is sufficient to internalise the pathogen into the host cells by interacting with human E-cadherin –specifically the N-terminal domain 1 (hEC1). The InlA variant (InlAm) that was previously made to increase the binding affinity to hEC1 was successfully engineered in this study. This variant was found to interact with N-terminal domain 1 of murine E-cadherin (mEC1) by isothermal titration calorimetry (ITC). Previously, the InlAm was reported to allow Lm invasion into M villous cells that express murine N-cadherin –possibly via the N-terminal domain 1 (mNC1). In this study, InlAm did not have affinity for mNC1 or N-terminal domain 1 of human N-cadherin (hNC1) when analysed by ITC –possibly due to amino acid sequences variation from both mEC1 and hEC1. However, by structurally engineering the complexes (InlAm/mNC1 and InlAm/hNC1) and studying their interaction interfaces, it was revealed that mNC1 and hNC1 can be recognised by InlAm just like hEC1. This was supported by the distances between interacting amino acid residues in InlAm/hEC1 crystal structure complex, which were also conserved in the engineered complexes. These observations related to the fact that the N-terminal domains of E- and N-cadherin are structurally conserved, therefore that could have attributed to similarities observed in the engineered complexes. Transcriptional and Epigenetic Regulation of Epithelial-Mesenchymal Transition

The transforming growth factor beta (TGFβ) is a cytokine that regulates a plethora of cellular processes such as cell proliferation, differentiation, migration and apoptosis. TGFβ signals via serine/threonine kinase receptors and activates the Smads to regulate gene expression. Enigmatically, TGFβ has a dichotomous role as a tumor suppressor and a tumor promoter in cancer. At early stages of tumorigenesis, TGFβ acts as a tumor suppressor by exerting growth inhibitory effects and inducing apoptosis. However, at advanced stages, TGFβ contributes to tumor malignancy by promoting invasion and metastasis. The pro-tumorigenic TGFβ potently triggers an embryonic program known as epithelial-mesenchymal transition (EMT). EMT is a dynamic process whereby polarized epithelial cells adapt a mesenchymal morphology, thereby facilitating migration and invasion. Downregulation of cell-cell adhesion molecules, such as E-cadherin and ZO-1, is an eminent feature of EMT. TGFβ induces EMT by upregulating a non-histone chromatin factor, high mobility group A2 (HMGA2). This thesis focuses on elucidating the molecular mechanisms by which HMGA2 elicits EMT. We found that HMGA2 regulates a network of EMT transcription factors (EMT-TFs), such as members of the Snail, ZEB and Twist families, during TGFβ-induced EMT. HMGA2 can interact with Smad complexes to synergistically induce Snail expression. HMGA2 also directly binds and activates the Twist promoter. We used mouse mammary epithelial cells overexpressing HMGA2, which are mesenchymal in morphology and highly invasive, as a constitutive EMT model. Snail and Twist have complementary roles in HMGA2-mesenchymal cells during EMT, and tight junctions were restored upon silencing of both Snail and Twist in these cells. Finally, we also demonstrate that HMGA2 can epigenetically silence the E-cadherin gene. In summary, HMGA2 modulates multiple reprogramming events to promote EMT and invasion.

Analysis of the role of the E-(Epithelial) Cadherin in murine lung tumorigenesis / Aanlyse der Rolle des E-(Epithelischer) Cadherin in der Lunge Tumorigenesis

Beim humanen nichtkleinzelligen Bronchialkarzinom ist die schrittweise Progression vom gutartigen Tumor zur malignen Metastasierung weitestgehend ungeklärt. In einem transgenen Mausmodell für das humane nichtkleinzellige Bronchialkarzinom, in dem in Lungenepithelzellen eine onkogene Mutante der Proteinkinase C-RAF exprimiert wird, können einzelne Schritte im Prozess der malignen Progression entschlüsselt werden. Die durch C-RAF induzierten Adenome zeichnen sich durch eine hohe genomische Stabilität in den Tumorzellen, durch starke interzelluläre Adhäsionskontakte zwischen den Tumorzellen und das Fehlen einer malignen Progression aus. Hier wurde demzufolge untersucht, ob die Auflösung der E-Cadherin-vermittelten Zellkontakte zwischen den einzelnen Tumorzellen eine Metastasierung auslösen könnte. Es wurden zwei genetische Ansätze verfolgt, um die Rolle der Tumorzelladhäsion im C-RAF Modell zu bewerten, die konditionelle Eliminierung des E-Cadheringens Cdh1 sowie die regulierbare transgene Expression von dominant-negativem E-Cadherin. Die Auflösung der E-Cadherin-vermittelten Zelladhäsion führte zur Neubildung von Tumorgefäßen, welche in der frühen Phase der Gefäßbildung durch Wiederherstellung des Zellkontakts reversibel war. Die vaskularisierten Tumore wuchsen schneller, bildeten invasive Fronten aus und führten zur Ausbildung von Mikrometastasen. Es konnte gezeigt werden, dass Beta-Catenin für die Induktion der Angiogenesefaktoren VEGF-A und VEGF-C in Lungentumorzellinien des Menschen und der Maus essentiell war. Lungentumorzellen aus den in situ Tumoren mit aufgelösten E-Cadherin-vermittelten Zellkontakten exprimierten Gene endodermaler und anderer Zellabstammung, was epigenetische Reprogrammierung in Tumorzellen als den Mechanismus bei der malignen Progression vermuten lässt. / Steps involved in the progression of non-small cell lung cancer (NSCLC) to metastasis are poorly understood. Expression of oncogenic C-RAF in lung epithelial cells has yielded a model for non-small cell lung cancer (NSCLC). The induced adenomas are characterised by high genomic stability, a lack of tumor progression and pronounced cell-cell contacts raising the question whether disruption of E-cadherin complexes would promote progression to metastasis. Two genetic approaches were used to evaluate the role of adherens junctions in a C-RAF driven mouse model for NSCLC: conditional ablation of the Cdh1 gene and expression of dominant negative (dn) E-cadherin. Disruption of E-cadherin function caused massive formation of intratumoral vessels that was reversible in the early phase of induction. Vascularized tumors grew more rapidly, developed invasive fronts and gave rise to micrometastasis. ß-catenin was identified as a critical effector of E-cadherin disruption leading to up-regulation of angiogenic inducers (VEGF-A and VEGF-C) in mouse and human lung tumor cell lines. In vivo, lung tumor cells with disrupted E-cadherin expressed ß-catenin target genes of endodermal and other lineages suggesting that reprogramming may be involved in metastatic progression.

Coordenação de Aperfeiçoamento de Pessoal de Nível Superior / O câncer colorretal representa uma das maiores causas de morbidade e mortalidade relacionadas ao câncer. No Brasil, é o terceiro tipo de câncer mais frequente em homens e mulheres. Muitos estudos estão sendo desenvolvidos no sentido de esclarecer os diversos aspectos moleculares que regulam as alterações fenotípicas exibidas pelas células que constituem o câncer colorretal, no entanto, comparativamente, ainda são poucos os que são dedicados a investigar o papel de modificações co- e pós-traducionais neste processo. Entre os vários tipos destas modificações que ocorrem em proteínas, a glicosilação é a mais comum. Cogita-se que aproximadamente cinquenta por cento de todas as proteínas são glicosiladas. Durante a transformação maligna, mudanças no perfil de expressão de glicanos (carboidratos covalentemente ligados a proteínas ou lipídios) estão envolvidas em uma variedade de mecanismos celulares, tais como: perda da adesão célula-célula e célula matriz, migração, invasão e evasão da apoptose. Neste estudo, foi investigada a atividade antitumoral de inibidores da biossíntese de N-glicanos, swainsonina e tunicamicina, em células derivadas de câncer colorretal (Caco-2, HCT-116 e HT-29). Os resultados obtidos mostram que o tratamento das células HCT-116 com tunicamicina inibe mecanismos celulares relacionados ao fenótipo maligno, como formação de colônia dependente e independente de ancoragem, migração e invasão. Estes resultados sugerem que modulação da biossíntese de N-glicanos parece ser uma potencial ferramenta terapêutica para o tratamento do câncer colorretal. Em outra etapa do trabalho, foram avaliados também o impacto da estimulação com insulina e IGF-1 na expressão N-glicanos bissectados em células tumorais MDA-MB-435. Os resultados obtidos confirmaram também a existência de uma relação entre a estimulação dos receptores de insulina e IGF-1 e a regulação da expressão de N-glicanos bissectados em células tumorais MDA-MB-435, fornecendo assim informações relevantes sobre o papel desempenhado pela sinalização de insulina e IGF-1 durante a progressão de carcinomas. / Colorectal cancer is a major cause of cancer-related morbidity and mortality. In Brazil, it is the third most common cancer. Many studies have been developed to clarify several molecular features which regulate phenotypic changes exhibited by cells that constitute colorectal cancer, however, comparatively, there are few studies dedicated to investigate co- and post-translational modifications of proteins in this process. Glycosylation is the most common post-translational modification of proteins. Approximately fifty percent of all proteins are glycosylated. During malignant transformation, changes in the expression profile of glycans (carbohydrates covalently bound to proteins or lipids) may be involved in a variety of events, including the loss of cellcell and cellmatrix adhesion, migration, invasion, and evasion of apoptosis. In this study, we investigated the in vitro anticancer activity of the N-glycan biosynthesis inhibitors, swainsonine and tunicamycin, in cells derived from colorectal cancer (Caco-2, HCT-116 e HT-29). Our results show that treatment with tunicamycin inhibits cellular mechanisms related to the malignant phenotype, such as anchorage-dependent and anchorage-independent colony formation, migration and invasion, in HCT-116 colon cancer cells. Given these results, we suggest that the modulation of N-glycan biosynthesis appears to be a potential therapeutic tool for CRC treatment. Moreover, we confirmthe existence of an interplay between stimulation with insulin and IGF-1 and bisecting GlcNAc N-glycans expression in MDA-MB435 cancer cells, providing new insights into the role that Insulin/IGF-I signaling play during carcinoma progression.

Melanomas melânicos e amelânicos da cavidade bucal de cães: aspectos epidemiológicos, morfológicos e moleculares / Melanotic and amelanotic melanomas from the buccal cavity of dogs: epidemiological, morphological and molecular aspects

Melanoma é uma neoplasia maligna com comportamento agressivo considerado o câncer mais comum da cavidade bucal de cães. Ele pode ser classificado quanto a morfologia celular em epitelióide, fusiforme e misto ou quanto ao fenótipo: melânico ou amelânico. Estudos têm sugerido que os melanomas amelânicos são mais agressivos. O objetivo deste estudo foi avaliar o comportamento dos melanomas da cavidade bucal dos cães, quantificando a expressão das Cx26 e 43, caderina-E, MMPs 2 e 9 e proliferação celular. Para tanto foram coletados 25 melanomas provenientes de cães atendidos no HOVET FMVZ-USP (16 melânicos e 9 amelânicos). Após a cirurgia os animais foram acompanhados até a morte, sendo que 5 animais foram eutanasiados (2 antes da cirurgia e 3 após o procedimento cirúrgico, devido ao sofrimento físico). Os tumores eram diagnosticados através do histopatológico e classificados de acordo com OMS (1998). Para confirmação dos melanomas amelânicos foi utilizado à técnica de imuno-histoquímica com o perfil de anticorpos pré-estabelecidos. A proliferação celular foi quantificada através do uso de PCNA, índice apoptótico e caspase-3. O comportamento tumoral foi avaliado através da técnica de imuno-histoquímica para caderina-E e MMPs 2 e 9. E a expressão das Cxs foram avaliadas através da imunofluorescência e western blot. Cães com melanma amelânico apresentaram menor sobrevida, com aumento do número de metástase, fraca marcação para caderina-E e alta intensidade para as MMPs 2 e 9, aumento de células positivas para PCNA e índice mitótico, e diminuição da caspase-3, não havendo diferença significante quanto aos tipos histotógicos. Houve diminuição de expressão das Cxs entre os amelânicos, no entanto, aumento da síntese do gene de CX 26 entre o mesmo grupo, o que se verificou através do PCR em Tempo Real. It can be classified as cell morphology in epithelioid, spindle and mixed or on the phenotype, in melanotic or amelanotic. Studies have suggested that amelanotic melanomas are more aggressive. The aim of this study was to evaluate the behavior of melanoma of the buccal cavity of dogs, quantifying the expression of Connexins 26 and 43, E-cadherin, MMPs 2 and 9 and cell proliferation. For both melanomas were collected from 25 dogs attended at HOVET FMVZ (USP) (16 melanotic and 9 amelanotic melanomas). After the surgery, the animals were followed until death, so 5 animals were euthanized (2 before surgery and 3 after surgery, due to physical suffering). Cell proliferation was quantified by using PCNA, apoptotic index and caspase-3. The tumor behavior was evaluated using the technique of immune-histochemistry for E-cadherin and MMPs 2 and 9. And the expression of Cxs was evaluated by immune-fluorescence and western blot. Dogs with amelanotic melanoma had lower survival with increasing number of metastatic, weak immunoreactivity for E-cadherin and high intensity for MMP 2 and 9, an increase of cells positive for PCNA and mitotic index, and decreased caspase-3, no significant difference in the morphologic subtypes. There was a decrease of expression of Cxs between amelanotic, however, increased synthesis of CX 26 gene among the same group, which was verified by real time PCR. Our findings suggest that melanomas of the buccal cavity of dogs exhibited more aggressive behavior.
