A atividade antimicrobiana do extrato foi analisada em Candida albicans, C. dubliniensis, C. glabrata, C. guilliermondii, C. krusei, C. tropicalis, Staphylococcus aureus, Streptococcus mutans, Escherichia coli e Pseudomonas aeruginosa com a determinação das Concentrações Inibitória Mínima e Microbicida Mínima (CIM e CMM) em culturas planctônicas e posteriormente testado sobre biofilmes monotípicos. A atividade citotóxica foi avaliada em cultura de macrófagos de camundongo (RAW 264.7) e fibroblastos gengivais humanos (FMM-1) após exposição de 5 min e 24 h ao extrato, com o teste MTT e teste imunoenzimático (ELISA) que quantificou a produção das citocinas IL-1β e TNF-α produzidas por RAW 264.7. A genotoxicidade do extrato foi avaliada pelo teste de micronúcleos. A atividade anti-inflamatória foi avaliada nos sobrenadantes coletados de culturas de RAW 264.7 estimuladas com LPS de E. coli e expostas aos extratos (5 min e 24 h) pela quantificação de citocinas IL-1β, TNF-α e IL-10 por ELISA e óxido nítrico (ON) pelo método de Griess. A análise estatística foi realizada por ANOVA e teste de Tukey, com significância de 5%. O extrato teve ação antimicrobiana em todos biofilmes, com redução acima de 39,5% em 5 min e acima de 78% em 24 h. A viabilidade celular foi satisfatória em concentrações abaixo de 50 mg/mL em 5 min tanto para FMM-1, quanto para RAW 264.7, contudo em 24 h concentrações acima de 3,13 e 12,5 mg/mL foram citotóxicas para RAW 264.7 e FMM-1, respectivamente. Não houve indícios de ação genotóxica do extrato. A atividade anti-inflamatória foi evidenciada pela redução significativa na produção de TNF-α e ON nos grupos tratados. Conclui-se que o extrato de bétula tem potencial como agente antimicrobiano e anti-inflamatório. / In order to verify the therapeutic potential of glicolic extract of B. pendula on the threatment of bacterial and fungical infection and on inflammatory diseases, its antimicrobial, anti-inflammatory actions, cytotoxic and genotoxic effects were evaluated. The antimicrobial activity of the extracts was tested on Candida albicans, C. dubliniensis, C. glabrata, C. guilliermondii, C. krusei, C. tropicalis, Staphylococcus aureus, Streptococcus mutans, Escherichia coli and Pseudomonas aeruginosa with determination of Minimum Inhibitory and Minimum Microbicide Concentrations (MIC and MMC) in planktonic growth, following of test in monotypic biofilms. The cytotoxic activity was evaluated in mouse macrophages (RAW 264.7) and human gingival fibroblast (FMM-1) after exposure of 5 min and 24 h to extract, through the MTT test and by Enzyme Linked Immunosorbent Assay (ELISA) to quantify the production of the cytokines IL-1β and TNF-α by RAW 264.7. The genotoxicity of the extracts was evaluated by micronucleus test. The anti-inflammatory activity was evaluated in RAW 264.7 stimulated with LPS from E. coli, after the period of exposure to the extract (5 min and 24 h) the supernatant was removed to quantify pro-inflammatory (IL-1β and TNF-α.) and anti-inflammatory (IL-10) cytokines by ELISA and nitric oxide (NO) by Griess method. Statistical analysis was performed by ANOVA and Tukey's test, with significance level of 5%. The extract has shown antimicrobial activity with reduction above of 39.5% of biofilm in 5 min and more than 78% in 24 h. The cell viability was satisfactory at concentrations below 50 mg/mL in 5 min to RAW 264.7 and FMM-1, however in 24 h concentrations above 3.13 and 12.5 mg/mL were cytotoxic to RAW 264.7 and FMM-1, respectively. There was no evidence of genotoxic action of the extract. The anti-inflammatory activity was evidenced by the significant reduction in the production of TNF-α and NO in the treated groups. It concludes that the birch extract has potential as antimicrobial and anti-inflammatory agent.

La connaissance des processus physiologiques, soit dans la phase de repos, soit pendant le développement végétatif, est important dans l'adaptation des angiospermes ligneuses tempérées, plutôt sur la question de la gestion de l'eau par la plante. Les objectifs de cette étude étaient d’évaluer la teneur de l’eau et le métabolisme des hydrates de carbone pendant l'état d'avancement de la dormance chez les poiriers, d'évaluer les stratégies d'utilisation de l'eau chez deux espèces ligneuses (pommier et noyer), qui ont été soumis à trois niveaux de déficit hydrique, et finalement, d’évaluer la fiabilité de la technique d'injection d'air pour provoquer la cavitation dans trois espèces d'angiospermes ligneuses. Lors de la première expérience, menée pendant l'automne et l'hiver 2008 au Brésil, nous avons utilisé les cultivars de poiriers et de ‘Packham Triumph’ et ‘Housui’. La deuxième expérience a été développée en France au cours de l'été 2009, avec des pommiers et des noyers sous stress hydrique. La troisième expérience a été réalisée en France en 2009 avec trois espèces d'angiospermes ligneuses avec différentes longueurs de vaisseaux du xylème, Betula pendula, Prunus persica et Quercus robur. D'après les résultats, on a pu conclure que la teneur de l’eau dans les tissus peut être un marqueur de la progression de dormance de la ‘Packham Triumph’. Cependant, les données ne sont pas concluantes pour la ‘Housui’. La fermeture des stomates peut être un paramètre important dans la prévention de la cavitation du xylème chez les noyers et pommiers soumis à un déficit hydrique du sol. Lors du régime de sécheresses sévères, le pommier conserve les stomates partiellement ouverts, ce qui indique une plus grande tolérance de cette espèce à la sécheresse par rapport au noyer. Enfin, la technique d'utilisation de l'injection d'air avec des caméras d’air à deux sorties est fiable pour les espèces dont le xylème a une porosité diffuse, dès qu’on utilise des caméras courtes et des échantillons de branches qui ont une longueur plus grande que les vaisseaux du xylème. / The knowledge of physiological processes during both dormancy and vegetative growth is important for adaptation processes in temperate zone woody angiosperms, especially in water management by those plants. The objectives of this work were to evaluate the water content and carbohydrate metabolism during the progress of dormancy in pears, to evaluate strategies of water use in two woody species (apple and walnut), which were subjected to three di fferent levels of water deficit, and also to evaluate reliability of air injection technique to induce cavitation in three woody angiosperms. The fi rst experiment was conducted during autumn and winter of 2008 in Brazil, and it was used plants of pears cultivars Packham's Triumph and Housui. The second study was carried out during the summer season of 2009 in France, with apple and walnut trees under water stress. The third experiment was done in France in 2009, and three species of woody angiosperms with different lengths of xylem were used Betula pendula, Prunus persica and Quercus robur. From the results it was concluded that the water content in tissues might be a marker of dormancy progression for cv. Packham's Triumph. However, the data were not conclusive and clear in cv. Housui. Stomatal closure might be an important parameter for preventing xilemian cavitation in walnut and apple trees subjected to soil water deficit. Apple trees stomata grown under severe drought regime remained partially open, indicating a greater tolerance of this species to drought in comparison to walnut. Finally, the doubleended pressure sleeve technique is reliable for species with diffuse-porous xylem by using short chambers and also branches longer than the xylem vessels. / O conhecimento dos processos fisiológicos, tanto na fase de repouso quanto no desenvolvimento vegetativo, é importante na adaptação das angiospermas lenhosas de clima temperado, especialmente na questão da gestão da água pela planta. Os objetivos deste trabalho foram avaliar o conteúdo da água e o metabolismo de carboidratos durante o progresso da dormência em pereiras, avaliar estratégias do uso da água em duas espécies lenhosas (maciei ra e noguei ra), que foram submetidas a três níveis de déficit hídrico, e por fim, avaliar a confiabilidade da técnica de injeção de ar para indução à cavitação em três espécies de angiospermas lenhosas. No primei ro experimento, conduzido durante o outono e inverno de 2008 no Brasil, utilizou-se plantas de pereira cultivares Hous ui e Packham´s Triumph. O segundo experimento foi desenvolvido na França durante o verão de 2009, com macieiras e nogueiras sob estresse hídrico. O tercei ro experimento foi conduzido na França em 2009 com três espécies de angiospermas lenhosas com di ferentes comprimentos dos vasos do xilema, Betula pendula, Prunus persica e Quercus robur. A partir dos resultados concluiu-se que o conteúdo de água nos tecidos pode ser um marcador do progresso da dormência para cv. Packham’s Triumph. Porém os dados não foram conclusivos para cv. Housui. O fechamento estomático pode ser um importante parâmetro na prevenção à cavitação xilemática em nogueiras e macieiras submetidas a déficit hídrico no solo. Em regime de secas severas, a maciei ra mantém os estômatos parcialmente abertos, indicando maior tolerância desta espécie à seca quando comparado à nogueira. E por fim, a técnica de injeção de ar utilizando câmara de ar com duas saídas é fiável para espécies com xilema de porosidade difusa, utilizando câmaras curtas e amostras de ramo que tenha comprimento superior aos vasos do xilema.

A madeira é a principal variável de custo na produção de polpa celulósica. A polpa celulósica obtida a partir da madeira de folhosas é principalmente utilizada para produção de papéis de escrita e impressão assim como, a produção de papéis para fins sanitários; para estes usos normalmente a polpa celulósica deve ser branqueada. Mundialmente, várias espécies de madeira de folhosas são utilizadas para produção de polpa celulósica de fibra curta, estando entre as principais o híbrido Eucalyptus grandis x Eucalyptus urophylla (Brasil), Betula pendula (Finlândia) Eucalyptus globulus (Portugal). Este projeto teve por objetivo: i-avaliar e comparar o desempenho de madeiras de principais espécies utilizadas mundialmente, para produção de polpa celulósica de fibra curta, considerando os aspectos relacionados à qualidade da madeira e polpa, o consumo de reagentes químicos na polpação e também os aspectos relacionados ao rendimento e a capacidade de produção de polpa celulósica; ii-fornecer informações estratégicas para o setor celulósico nacional com relação as possíveis ameaças e as oportunidades apresentadas pelos materiais não utilizados no Brasil, visando o conhecimento e posicionamento em competitividade destas madeiras no mercado mundial de celulose e papel; iii-fornecer subsídios para o programa genético e de melhoramento florestal; iv-posicionar a polpa celulósica de Eucalyptus grandis x Eucalyptus urophylla em relação as suas principais concorrentes. Para realização deste trabalho foram coletadas 5 árvores de todas as espécies, sendo considerado o diâmetro médio do povoamento, transportando a madeira em forma de discos e transformadas em cavacos de forma manual; com relação à madeira foram determinadas, densidade básica, dimensões de fibras, composição química e análise anatômica através da microscopia eletrônica de varredura; as condições de cozimento foram ajustadas visando à obtenção de polpa branqueável (número kappa de 17±0.5). Os resultados obtidos mostraram que as espécies de Eucalyptus globulus e E. grandis x E. urophylla 2 apresentaram melhor desempenho no processo de polpação; atingiram a mesma intensidade deslignificação (expressa como número kappa), sendo necessário menor nível de álcali ativo. As matériasprimas citadas apresentaram maior rendimento, maior viscosidade e menor teor de ácidos hexenurônicos. As diferentes matérias-primas estudadas neste trabalho apresentaram características bastante distintas que implicam em diferentes eficiências nos processos de polpação e branqueamento. / Wood is the main variable in the cost of pulp production, besides being a strong component in the final quality of the paper product. Pulps obtained from hardwoods are mainly used in the production of printing and writing papers as well as tissue papers; for these uses, the pulp must be bleached. Several species of hardwoods are used in the production of pulp worldwide. The main ones are the hybrid Eucalyptus grandis x Eucalyptus urophylla (Brazil), E. globulus (Portugal), Betula pendula (Finland). The objectives of this research was: 1) to evaluate and compare the performance of woods from the main species used worldwide for hardwood cellulose pulp production, considering the aspects regarding the quality of the wood and pulp, chemicals consumption during pulping, and aspects related to the yield and capacity pulp production; 2) to supply strategic information for the Brazilian pulp and paper sector regarding possible threats and opportunities presented by materials which are not used in Brazil in order to know and compete with these woods in the world market of pulp and paper. In order to accomplish this work, chips were manually obtained from disks. Regarding the wood, the basic density, fibers dimensions, chemical composition and anatomic analysis were conducted. The cooking conditions were adjusted in order to obtain the bleachable pulp (kappa number 17±0.5). The results had shown that the species of Eucalyptus globulus from Portugal and hybrid of Eucalyptus grandis x Eucalyptus urophylla 2 had better performance in the kraft pulping process; they had reached the same delignification level, as number kappa, requiring lesser active alkali. The mentioned raw materials showed higher yield, greater viscosity and minor levels of acid hexenurônicos. The raw-materials considered in the research showed clearly distinct characteristics that lead to differences in the pulping and bleaching processes efficiencies and strategies.

On the importance of oxidizable structures in bleached kraft pulps

After cooking, kraft pulps always contain not only residual lignin but also significant amounts of hexenuronic acid and other non-lignin structures oxidizable by permanganate under the standard kappa number determination conditions. These here referred to as false lignin. Like ordinary lignin, the false lignin also consumes bleaching chemicals, thus increasing both the production costs and the environmental impact of bleach plant effluents. The false lignin also has an effect on pulp properties such as brightness stability. This necessitates the development of efficient experimental routines for the determination of false lignin in different types of unbleached and bleached kraft pulps, together with studies of its formation, chemical behaviour, and ultimate fate. The main aim of this work has been to establish a method for the quantification of various types of oxidizable structures in bleached kraft pulps and to study their impact on pulp quality, particularly, on the brightness stability of pulps bleached in elemental-chlorine-free (ECF) and a totally-chlorine-free (TCF) processes. Part of this research deals with the relationship between the kappa number and the lignin content in the case of partly oxidized lignins. Spruce and birch kraft pulps processed according to the ODEQP and OQ(OP)Q(PO) bleaching sequences, respectively, have been analyzed. It has been found that the oxidation equivalent of the residual lignin decreases with increasing degree of oxidation along each bleaching sequence. This finding has been further supported by experiments with a number of model compounds. The Ox-Dem kappa number method has been shown to be an accurate means of determining the residual lignin content and of monitoring the efficiency of lignin removal along different bleaching sequences. It has been demonstrated that the kappa number can always be fractioned into partial contributions, the first of which comes from the residual lignin and is measured by the Ox-Dem kappa number, and the second from the false lignin and is given by the difference between the standard kappa number and the Ox-Dem kappa number. The effect of false lignin on the pulp kappa number is most pronounced in unbleached and oxygen-delignified kraft pulps. The extractability of residual and false lignin in different solvents has been investigated. The changes that occurred in the kappa number following different extraction steps have been compared with corresponding changes in the chemical composition and the conclusion has been drawn that the hemicellulose component of a kraft pulp is a major sourse of non-lignin structures contributing to the kappa number. The influence on the brightness stability of various oxidizable structures, viz.: residual lignin, hexenuronic acid and other non-lignin structures, in spruce, birch and eucalyptus kraft pulps bleached in ECF and TCF type processes was studied. It was demonstrated that the selective removal of all false lignin structures significantly improves the brightness stability. The degree of yellowing was found to be proportional to the content of HexA groups in pulps. It has been shown that 2-furancarboxylic acid, 5-formyl-2furancarboxylic acid and reductic acid are formed during the course of thermal yellowing. The influence of two bleaching sequences, D0(EP)D1 (ECF-type) and Q1(OP)Q2(PO) (TCF)-type, on the content of different oxidizable structures in eucalyptus kraft pulp was studied in relation to the brightness stability of the pulp. It was shown by kappa number fractionation that pulp bleached to full brightness with ECF- and TCF-type sequences contains different amounts of HexA. The most significant discoloration was observed in the case of TCF-bleached pulp having an especially high content of HexA. The mechanism of the moist (8 % moisture) thermal yellowing of fully bleached kraft pulps was further studied using dissolving pulp impregnated with a set of model compounds representing the most likely HexA degradation products, viz. as 2-furancarboxylic acid (FA), 5-formyl-2-furancarboxylic acid (FFA) and reductic acid (RA), either alone or in combination with Fe(II) or Fe(III) ions. It was found that the latter two acids take part in reactions leading to colour formation whereas 2-furancarboxylic acid does not. The effect of iron ions on the colour formation appears to vary with their oxidation state. The brightness loss caused by either FFA or RA, present in an amounts similar to the content of HexA in industrial pulps, was of the same order of magnitude as that observed in industrial pulps aged under the same conditions. Based on these findings, it is suggested that the overall mechanism of moist thermal yellowing involves several stages, including the degradation of hexenuronic acid and the formation of reactive precursors, such as 5-formyl-2-furancarboxylic acid and reductic acid. Treeline dynamics in short and long term perspectives : observational and historical evidence from the southern Swedish Scandes

Against the background of past, recent and future climate change, the present thesis addresses elevational shifts of alpine treelines in the Swedish Scandes. By definition, treeline refers to the elevation (m a.s.l.) at a specific site of the upper trees of a specific tree species, at least 2 m tall. Based on historical records, the first part of the thesis reports and analyzes the magnitude of treeline displacements for the main trees species (Betula pubescens ssp. czerepanovii, Picea abies and Pinus sylvestris) since the early 20th century. The study covered a large and heterogeneous region and more than 100 sites. Concurrent with temperature rise by c. 1.4 °C over the past century, maximum treeline advances of all species amount to about 200 m. That is virtually what should be predicted from the recorded temperature change over the same period of time. Thus, it appears that under ideal conditions, treelines respond in close equilibrium with air temperature evolution. However, over most parts of the landscape, conditions are not that ideal and treeline upshifts have therefore been much smaller. The main reason for that discrepancy was found to be topoclimatic constraints, i.e. the combined action of geomorphology, wind, snow distribution, soil depth, etc., which over large parts of the alpine landscape preclude treelines to reach their potential thermal limit. Recorded treeline advance by maximum 200 m or so over the past century emerges as a truly anomalous event in late Holocene vegetation history. The second part of the thesis is focused more on long-term changes of treelines and one specific and prevalent mechanism of treeline change. The first part of the thesis revealed that for Picea and Betula, treeline shift was accomplished largely by phenotypic transformation of old-established stunted and prostrate individuals (krummholz) growing high above the treeline. In obvious response to climate warming over the past century, such individuals have transformed into erect tree form, whereby the treeline (as defined here) has risen. As a means for deeper understanding of this mode of positional treeline change, extant clonal spruces, growing around the treeline, were radiocarbon dated from megafossil remains preserved in the soil underneath their canopies. It turned out that Picea abies in particular may attain almost eternal life due to its capability for vegetative reproduction and phenotypic plasticity. Some living clones were in fact inferred to have existed already 9500 years ago, and have thus persisted at the same spot throughout almost the entire Holocene. This contrasts with other tree species, which have left no living relicts from the early Holocene, when they actually grew equally high as the spruce. Thereafter they retracted by more than 300 m in elevation supporting that also on that temporal scale, treelines are highly responsive to climate change. The early appearance of Picea in the Scandes, suggests that Picea “hibernated” the last glacial phase much closer to Scandinavia than earlier thought. It has also immigrated to northern Sweden much earlier than the old-established wisdom. The experiences gained in this thesis should constitute essential components of any model striving to the project landscape ecological consequences of possible future climate shifts.

Life-history consequenses of host plant choice in the comma butterfly

There is much evidence that herbivory is a key innovation for the tremendous success of insect. In this thesis I have investigated different aspects of host plant utilization and phenotypic plasticity using the polyphagous comma butterfly, Polygonia c-album. Even though external conditions affect a phenotypic plastic response, the outcome is often influenced by a genetic background which may differ among populations. In Paper I we suspected the genetic background to seasonal polymorphism to be X-linked. However, results from interspecific hybridization between two populations suggested that diapause response is instead inherited in a mainly autosomally additive fashion, with a possible influence of sexual antagonism on males. In Paper II we showed that female oviposition preference is not a plastic response influenced by larval experience, but has a genetic background coupled to host plant suitability. Further, there is a strong individual correlation between larval host plant acceptance and female host plant specificity (Paper III). We believe this to be a larval feed-back genetically linked to female host specificity: offspring to ‘choosy’ specialist mothers benefit by remaining on the original host while offspring to less discriminating generalist mothers should risk inspecting the surroundings, thus compensating for potential poor female choice. In the larval mid-gut, genes are differentially expressed depending on host plant diet (Paper IV). Therefore, we expected to find fitness consequences of host plant switch. However, although growth rate was affected in a few treatments, larvae were generally surprisingly good at adjusting to new diets (Paper V). To conclude, host plant choice in both female and larval life stage is connected to performance. Combined with increased understanding about the plastic response to diet intake and seasonal polymorphism we have gained further insights into the processes of local adaptations and speciation in the Lepidoptera. / At the time of the doctoral defense, the following papers were unpublished and had a status as follows: Paper 3: Submitted Manuscript; Paper 5: Manuscript
