Marcação e detecção de proteínas sintetizadas durante o desenvolvimento estrobilar de Echinococcus granulosus IN VITRO

O Echinococcus granulosus é um platelminto parasita da classe Cestoda, causador da hidatidose cística. Durante o ciclo de vida no hospedeiro intermediário (ovinos ou bovinos e, acidentalmente, o homem) há a formação de um cisto hidático, que abriga a forma pré-adulta do parasito, o protoescólex (PSC), que, ao ser ingerida pelo hospedeiro definitivo (geralmente o cão doméstico), desenvolver-se-á no verme adulto. De outra forma, o cisto primário fértil pode romper-se e extravazar seu conteúdo no interior do hospedeiro intermediário, o que induz os PSCs à formação de cistos hidáticos secundários. Embora existam vários relatos na literatura acerca da utilização de proteínas como biomarcadores para fases de desenvolvimento em outras espécies, pouco é conhecido sobre os mecanismos envolvidos nessa plasticidade do parasito. Uma opção de estudo está na resposta do parasito a um evento indutor, sendo interessante a marcação de proteínas recém-sintetizadas (PRS), realizada com aminoácidos radioativos ou, mais recentemente, com a utilização de aminoácidos artificiais. Dessa forma, o presente estudo visa melhor compreender os mecanismos moleculares envolvidos no desenvolvimento estrobilar de E. granulosus, através da análise de PRS nos PSCs com a incorporação do aminoácido artificial azido-homoalanina (AHA). Para isso, estudamos diferentes condições para a diferenciação estrobilar de PSCs in vitro e a incorporação de AHA, utilizando um tratamento inicial com pepsina seguido por cultivo em meio bifásico contendo taurocolato. Dessa forma, foi possível visualizar alterações morfológicas características da estrobilização já descritas na literatura, como a diminuição no número de corpúsculos calcários e a formação do canal excretor e bexiga. Após 72 h de incubação dos PSCs em meio bifásico suplementado com AHA, as PRS foram extraídas e marcadas com tetrametilrodamina. A eficiência da incorporação da AHA foi confirmada por SDS-PAGE 12% e visualização com luz ultravioleta, resultando num padrão de bandas complexo, com proteínas de 10 a 225 kDa, sendo este o primeiro relato de marcação e detecção de PRS com AHA em platelmintos. / The Echinococcus granulosus is a parasite of Class Cestoda, which causes cystic hydatid disease. During its life cycle in the intermediate host (sheep or cattle and accidentally man), initiates the formation of a hydatid cyst, which contains the pre-adult form of the parasite, the protoescolex (PSC). When ingested by a definitive host (usually the domestic dog), the PSC will develop into the adult worm. Another possibility is the rupture of the primary fertile cyst, releasing PSCs in the intermediate host, which may develop into secondary hydatid cysts. Although there are several reports in the literature about the use of proteins as biomarkers for developmental stages, little is known about the mechanisms involved in the plasticity of E. granulosus. A possible way to study this process is to analyze the parasite response to an inducing event, by labeling newly synthesized proteins (NSP), using radioactive amino acids or, more recently, artificial amino acids. This study aims to better understand the molecular mechanisms involved in the estrobilar development of E. granulosus by analyzing NSP in PSCs with the incorporation of the artificial amino acid azidohomoalanine (AHA). For this purpose, we have used different methods for cultivation that led to the in vitro PSCs’ estrobilar differentiation with an initial pepsin treatment, followed by cultivation in biphasic medium containing taurocholate. Using this procedure, it was possible to observe the morphological changes characteristic of the strobilation stages already described in the literature, such as the decrease in the number of calcareous corpuscles or the visualization of the excretory canal and the bladder. Following incubation of PSCs in biphasic medium supplemented with AHA, the NSP were extracted and labeled with tetramethylrhodamine. The incorporation efficiency of AHA was confirmed by analysis on 12% SDS-PAGE and exposure to UV light, showing a complex pattern of bands, with proteins between 10-225 kDa. This is the first report of labeling and detection of NSP with AHA in flatworms.

Variación genética intraespecífica de Echinococcus granulosus en la sierra central y sur del Perú

Echinococcus granulosus es el agente causal de la hidatidosis unilocular, patología de carácter endémico en amplias zonas de Perú. Actualmente la metodología del análisis molecular del ADN se ha utilizado para enriquecer los conocimientos sobre este parásito, a través del aislamiento de sus genes, el estudio de su estructura y organización, así como la expresión de los mismos. La aplicación de estas técnicas ha reconocido en E. granulosus un mínimo de 10 variedades o cepas. La taxonomia y el estatus filogenético de las cepas de E. granulosus es controversial y esta bajo discusión; su variación genética ha sido analizada por la variación de las secuencias de los nucleótidos dentro de las regiones mitocondriales y nucleares del DNA. Estas variantes son referidas como cepas y han sido generalmente asignadas con el nombre del hospedero intermediario involucrado en el ciclo biológico del parásito. Se ha identificado la existencia de 10 genotipos (G1 – G10) de E. granulosus, esta variabilidad tiene implicaciones importantes en la patología de la enfermedad, desarrollo de vacunas, diagnóstico y administración de drogas efectivas. En esta investigación, se han aislado quistes hidatídicos de ovinos, bovinos, porcinos y camélidos sudamericanos procedentes de Ayacucho, Cuzco, Pasco y Puno, departamentos pertenecientes a la sierra central y sur del Perú. La extracción del DNA fue con fenol –cloroformo – alcohol isoamílico; se aplicó un Nested-PCR usando el gen ribosomal ITS-1 y la genotipificación se hizo en base al uso de diferentes enzimas de restricción. De los dos patrones encontrados, los fragmentos producidos en el Nested- PCR fue amplificado y directamente secuenciado y a través de programas bioinformáticos se determinó la presencia de 2 variantes, una de las cuales corresponde a G1 y la otras probablemente restringida a nuestros camélidos sud-americanos. / Unilocular hidatidosis’s disease whose etiological agent is the tapeworm Echinococcus granulosus, affects differents endemic zones in Peru. At this time, the methodology of the molecular analysis of the DNA has been used for the knowledge of this parasite through isolation of its genes, the study of its structure and organization, as well as the expression of the same. The application of these techniques has revolutionized the study of this parasite, recognition a minimum of ten strains. The taxonomic and phylogenetic status of Echinococcus granulosus strains are still controversial and under discussion; genetic variation in E. granulosus has been analyzed for DNA nucleotide sequence variation within of the region mitochondrial and nuclear region of the ribosomal of DNA. These variants are referrend to as strains, and they have been generally named after their typical intermediate hosts. To date, 10 genotypes (G1 – G10) of Echinococcus granulosus have been identified; this variability has important implications for the pathology of the disease, development of vaccines, diagnostic reagents and drugs effective against this parasite. In this research, we isolate different hydatic cysts from ovines, bovines, pork and sudamerican camels, this samples was get from Ayacucho, Cuzco, Pasco and Puno, all belong from central highland and south of Peru. The DNA extraction was done with phenol- cloroform- isoamil alcohol. We did Nested-PCR using primers from ribosomal genes ITS-1, and then the genotyping was done with RFLP PCR using different cutting restriction enzymes. We found two genetics patterns. The representatives patterns, their fragments produced by nested-PCR were amplified and directly sequenced and then with Bioinformatics programs determine one patters belong G1 and others was probably restricted from south-American camels that live in high altitudes in Peru. Although the number of Peruvian isolates examined was not extensive, the G1 genotype (113/120) was far more prevalent in domestic animals.

Estudo de três enzimas da via glicolítica de Echinococcus granulosus com possíveis funções moonlighting na interação da forma larval com o hospedeiro intermediário

A expressão “proteínas moonlighting” refere-se a um subgrupo de proteínas multifuncionais, onde duas ou mais funções são realizadas por uma mesma cadeia polipeptídica. Dentre as proteínas com funções moonlighting estão diversas enzimas glicolíticas que podem desempenhar papeis importantes em processos-chave na biologia de organismos parasitas, como os de motilidade, adesão, invasão e diferenciação. A aldolase, a enolase e a lactato-desidrogenase (LDH) estão entre as enzimas que foram detectadas nos produtos de excreção-secreção (ES) e em tecidos de interface com o hospedeiro de Echinococcus granulosus, o cestódeo causador da hidatidose cística em seres humanos e ungulados domésticos. Neste contexto, a caracterização destas enzimas pode revelar possíveis funções moonligthing por elas desempenhadas e contribuir para a compreensão de aspectos básicos do desenvolvimento do parasito e de possíveis mecanismos de interação com os hospedeiros. Inicialmente, as sequências codificadoras da aldolase (EgAldo), da enolase (EgEno) e da LDH (EgLDH) de E. granulosus foram amplificadas por RT-PCR, clonadas em um vetor de expressão modificado (pGEX-TEV) e expressas em Escherichia coli, para a produção das proteínas recombinantes correspondentes (rEgAldo, rEgEno e rEgLDH, respectivamente). Soros de pacientes humanos com hidatidose reconhecem, em imunoblots, a EgAldo e EgEno nativas mas não reconhecem as proteínas recombinantes correspondentes, sugerindo o envolvimento de epitopos não proteicos no reconhecimento desses antígenos. Antissoros policlonais contra a rEgAldo e a rEgEno evidenciaram, em protoescólices, uma localização predominantemente citoplasmática para as proteínas nativas cognatas. Elas também foram detectadas na camada germinativa e no líquido hidático, que representam interfaces entre o parasito e o hospedeiro. Além disso, tanto a EgAldo como a EgEno foram reconhecidas em frações de proteínas tegumentares e em produtos de ES de protoescólices, localizações sugestivas de funções moonlighting. Proteínas do parasito e do hospedeiro intermediário que interagem com as proteínas rEgAldo, rEgEno e rEgLDH foram recuperadas por experimentos de GST pull-down para posterior identificação destas proteínas por espectrometria de massas. A identificação destas proteínas de interação poderá apontar processos não-glicolíticos nos quais estas enzimas estejam envolvidas. / The term “moonlighting proteins” refers to a subset of multifunctional proteins in which two or more different functions are performed by one polipeptide chain. Among the proteins described as moonlighting, there are many glycolytic enzymes that play important roles in key processes in the biology of parasites, such as motility, adhesion, invasion and cell differentiation. Aldolase, enolase and lactate dehydrogenase are among the enzymes that have been detected in the excretory-secretory (ES) products and in the host-interacting metacestode components from Echinococcus granulosus, a cestode that causes cystic hydatid disease and affects both humans and livestock. In this context, the characterization of these enzymes may reveal possible moonlighting functions they perform and contribute to the understanding of basic aspects of the parasite development and differentiation and possible interaction mechanisms with the intermediate host. Initially, the complete coding sequences from E. granulosus aldolase (EgAldo), enolase (EgEno) and lactate dehydrogenase (EgLDH) were amplified by RT-PCR, cloned into a modified expression vector (pGEX-TEV) and expressed in Escherichia coli for recombinant proteins production (rEgAldo, rEgEno and rEgLDH, respectively). Sera from human hydatid disease patients recognize in immunoblots the native EgAldo and EgEno, but not its recombinant counterparts, which was taken as evidence of involvement of non proteic epitopes in the EgAldo and EgEno recognition. Polyclonal antiserum against rEgAldo and rEgEno have shown, in protoscoleces, a predominantly cytoplasmic localization for the cognate native proteins, as expected for glycolytic enzymes. They have also been detected in the germinal layer and hydatid fluid, which represent host-parasite interfaces. Furthermore, both EgAldo and EgEno were recognized in protoscoleces tegument extracts and in ES products from these parasites in culture, which are indicative of moonlighting functions. Parasite and intermediate host proteins that interact with proteins rEgAldo, rEgEno and rEgLDH in vitro were recovered by GST pull-down assays and the identification of these proteins will be performed using mass spectrometry. Identification of interacting proteins may also be an indication that these enzymes perform nonglycolytic functions.

Caracterização das proteínas 14-3-3ζ expressas na fase larval de Echinococcus granulosus

As proteínas 14-3-3 formam uma família altamente conservada, expressas em todos os organismos eucariotos já estudados. Através da interação com seus alvos, as proteínas 14-3-3 participam de importantes processos celulares, como controle do ciclo celular, apoptose, transdução de sinal e diferenciação celular. Neste trabalho, essa família gênica foi estudada em Echinococcus granulosus e em Echinococcus multilocularis, as espécies de maior importância do gênero Echinococcus. E. granulosus e E. multilocularis, na fase larval, são agentes etiológicos da hidatidose cística e da hidatidose alveolar, respectivamente, zoonoses associadas a grandes prejuízos na área da pecuária e a gastos no tratamento de pacientes humanos. Por meio da análise dos bancos de dados disponíveis e comparação com os dados genômicos, neste estudo, seis potenciais genes codificadores de proteínas 14-3-3 foram identificados para ambas as espécies, sendo denominados Eg14-3-3 e Em14-3-3 para E. granulosus e E. multilocularis, respectivamente. Análises filogenéticas das sequências preditas de aminoácidos codificadas por esses genes demonstram que as proteínas Eg14-3-3 e Em14-3-3 retêm características ancestrais, não se agrupando claramente com proteínas ortólogas de outros organismos quanto ao tipo de isoforma (zeta e épsilon), como ocorre, por exemplo, em mamíferos. A fim de caracterizar funcionalmente as proteínas Eg14-3-3, o padrão de expressão das isoformas Eg14-3-3ζ2 e Eg14-3-3ζ3 foi avaliado. A identificação dessas isoformas em componentes do metacestódeo de E. granulosus que representam interfaces parasito-hospedeiro sugere a possível participação dessas proteínas em mecanismos de interação com o hospedeiro. Estudos para a identificação dos ligantes protéicos que interagem com as proteínas Eg14-3-3ζ2 e Eg14-3-3ζ3 também foram realizados. Entre os ligantes identificados estão chaperonas, proteínas associadas à conversão de energia, citoesqueleto, e transporte e metabolismo de carboidratos. Essas interações apontam o envolvimento da família proteica 14-3-3 em mecanismos que promovem o estabelecimento e sobrevivência de E. granulosus. A caracterização das interações identificadas neste estudo poderá auxiliar na elucidação de aspectos biológicos básicos do parasito, e mecanismos moleculares utilizados para o desenvolvimento e manutenção do metacestódeo nas vísceras do hospedeiro intermediário por longo período de tempo. / The 14-3-3 proteins form a highly conserved family, and have been shown to be actively expressed in all eukaryotic organisms studied so far. Through interaction with their targets, the 14-3-3 proteins participate in some of the major eukaryotic cellular processes, such as cell cycle regulation, apoptosis, signal transduction, and cell differentiation pathways. In the present work, this gene family has been studied in two species of the genus Echinococcus, namely Echinococcus granulosus and Echinococcus multilocularis, which, in their larval stage, are the causative agents of the cystic hydatid disease and the alveolar hydatid disease, respectively, two economically relevant zoonotic diseases associated with livestock production losses and human health care expenditures. Through analysis and comparison of available database, and searches in Echinococcus sequencing genome data, six candidate genes encoding 14-3-3 proteins were identified in both species, being named Eg14-3-3 and Em14-3-3 to E. granulosus and E. multilocularis, respectively. Interestingly, the phylogenetic analyses of deduced amino acid sequences encoded by these genes showed that Eg14-3-3 and Em14- 3-3 proteins retain more ancestral characteristics, and do not group clearly with orthologous proteins from other organisms relative to isoform types (zeta and epsilon), as occurs, for example, in mammals. In order to functionally characterize the Eg14-3-3 proteins, the expression patterns of the Eg14-3-3ζ2 e Eg14-3-3ζ3 isoforms were analyzed. The identification of both isoforms in E. granulosus metacestode components, that represent host–parasite interface, suggests a possible participation of these proteins on mechanisms of interaction with the host. Additionally, studies aiming the identification of proteins able to bind to Eg14-3-3ζ2 and Eg14-3-3ζ3 isoforms were carried out. Among the ligands identified, chaperones, proteins associated with energy production and conversion, cytoskeleton and carbohydrate transport and metabolism, were present, suggesting the involvement of 14-3-3 protein family in mechanisms that promote the establishment and survival of E. granulosus. Further characterization of the interactions identified in this study can help to elucidate basic biological aspects of these parasites, as well as molecular mechanisms involved in the development and maintenance of hydatid cyst in within intermediate host tissues for long time.

Caracterização estrutural de oligômeros de subunidades recombinantes do antígeno B de Echinococcus granulosus

O antígeno B (AgB) de Echinococcus granulosus é um dos principais componentes do líquido hidático. O AgB é um proteína oligomérica de 120-160 kDa composta de subunidades de 8 kDa, que em SDS-PAGE dissocia-se em componentes de 8, 16, 24 e 32 kDa. Embora diferentes subunidades do AgB tenham sido isoladas, pouco se sabe sobre a estrutura da proteína e do seu mecanismo de oligomerização. Neste trabalho foi realizada a caracterização estrutural de homo-oligômeros de três subunidades recombinantes do AgB, AgB8/1, AgB8/2 e AgB8/3, expressadas a partir de seqüências previamente clonadas Essas subunidades associam-se em homo-oligômeros com características semelhantes às da proteína purificada de líquido hidático, como massa molecular, conteúdo de estrutura secundária, tendência agregativa e termoestabilidade, fazendo deles ferramentas importantes para o estudo da estrutura do AgB. Diferentes graus de estabilidade e compactação foram verificados entre os oligômeros recombinantes, com o oligômero de AgB8/3 apresentando-se mais estável e compacto. Através da modelagem molecular das subunidades do AgB, foi possível calcular a superfície de potencial eletrostático das moléculas e propor um mecanismo de oligomerização envolvendo interações eletrostáticas e hidrofóbicas. Foram também realizadas tentativas de cristalização de oligômeros de AgB8/3 para ensaios de difração de raios X, e cristais foram obtidos em três condições do screening inicial. Estes cristais difrataram à resoluções máximas de 8 Å, não permitindo a coleta de dados estruturais da proteína. Entretanto, estas condições podem ser refinadas para a obtenção de cristais com melhor qualidade. / Echinococcus granulosus antigen B is one of the major components of the hydatid fluid. AgB is a oligomeric protein of 120-160 kDa composed by 8 kDa subunits, that in SDS-PAGE dissociates in components of 8, 16 , 24 and 32 kDa. Although different AgB subunits have been isolated, little is known about AgB structure and its oligomerization mechanism. In this work we have performed the homo-oligomers structural characterization of three AgB recombinant subunits, AgB8/1, AgB8/2 and AgB8/3, expressed from previously cloned genes. These subunits self-assemble in homo-oligomers with similar characteristics to that of hydatid fluid purified protein, such as molecular mass, secondary structure content, aggregative tendency and thermostability, making them valuable tools for AgB structure study. Different degrees of stability and compactness were verified between the recombinant oligomers, with the AgB8/3 one showing a more stable and compact structure. Using molecular modelling it was possible to calculate the surface electrostatic potencial of AgB subunits and to propose a mechanism of oligomerization involving electrostatic and hydrophobic interactions. Also crystallization attempts had been carried through with AgB8/3 oligomer to X-ray diffraction, and crystals have been obtained in three conditions of initial screening. These crystals difracted to maximum resolutions of 8 Å, and it was not possible to collect protein structural data. However, these crystallization conditions can be refined in order to obtain crystals of better quality.

Echinococcus. granulosus endémico en algunas áreas de Perú, evidencia que los animales infectados con este parásito puede ser un potencial reservorio para infecciones humanas, además nuestros resultados constituyen un aporte importante para los programas de control que realizan una vigilancia continua de sus áreas endémicas a través de la implementación de un adecuado tratamiento de los animales infectados y el adiestramiento de los pobladores acerca de los riesgos de contraer la enfermedad y de las conductas adecuadas para el manejo de órganos infectadas. Although the number of Peruvian isolates examined was not extensive, the G1 genotype (113/120) was far more prevalent in domestic animals. Besides ours dates constitute an important contribution for the Control programs that realize a continuous monitoring of endemic areas through the implementation of a suitable treatment of the animal infected and the training of the settlers about the risks of contracting the disease in contaminated areas and of the conducts adapted for the handling of infected organs. In particular, the existence of different genotypes has to contribute to take measures from importance epidemiologist and to establish strategies where the objective is the prevention, treatment and control of the infections caused by E. granulosus in their different hosts from the different endemic zone from Peru. / Tesis

Caracterização estrutural de oligômeros de subunidades recombinantes do antígeno B de Echinococcus granulosus

O antígeno B (AgB) de Echinococcus granulosus é um dos principais componentes do líquido hidático. O AgB é um proteína oligomérica de 120-160 kDa composta de subunidades de 8 kDa, que em SDS-PAGE dissocia-se em componentes de 8, 16, 24 e 32 kDa. Embora diferentes subunidades do AgB tenham sido isoladas, pouco se sabe sobre a estrutura da proteína e do seu mecanismo de oligomerização. Neste trabalho foi realizada a caracterização estrutural de homo-oligômeros de três subunidades recombinantes do AgB, AgB8/1, AgB8/2 e AgB8/3, expressadas a partir de seqüências previamente clonadas Essas subunidades associam-se em homo-oligômeros com características semelhantes às da proteína purificada de líquido hidático, como massa molecular, conteúdo de estrutura secundária, tendência agregativa e termoestabilidade, fazendo deles ferramentas importantes para o estudo da estrutura do AgB. Diferentes graus de estabilidade e compactação foram verificados entre os oligômeros recombinantes, com o oligômero de AgB8/3 apresentando-se mais estável e compacto. Através da modelagem molecular das subunidades do AgB, foi possível calcular a superfície de potencial eletrostático das moléculas e propor um mecanismo de oligomerização envolvendo interações eletrostáticas e hidrofóbicas. Foram também realizadas tentativas de cristalização de oligômeros de AgB8/3 para ensaios de difração de raios X, e cristais foram obtidos em três condições do screening inicial. Estes cristais difrataram à resoluções máximas de 8 Å, não permitindo a coleta de dados estruturais da proteína. Entretanto, estas condições podem ser refinadas para a obtenção de cristais com melhor qualidade. / Echinococcus granulosus antigen B is one of the major components of the hydatid fluid. AgB is a oligomeric protein of 120-160 kDa composed by 8 kDa subunits, that in SDS-PAGE dissociates in components of 8, 16 , 24 and 32 kDa. Although different AgB subunits have been isolated, little is known about AgB structure and its oligomerization mechanism. In this work we have performed the homo-oligomers structural characterization of three AgB recombinant subunits, AgB8/1, AgB8/2 and AgB8/3, expressed from previously cloned genes. These subunits self-assemble in homo-oligomers with similar characteristics to that of hydatid fluid purified protein, such as molecular mass, secondary structure content, aggregative tendency and thermostability, making them valuable tools for AgB structure study. Different degrees of stability and compactness were verified between the recombinant oligomers, with the AgB8/3 one showing a more stable and compact structure. Using molecular modelling it was possible to calculate the surface electrostatic potencial of AgB subunits and to propose a mechanism of oligomerization involving electrostatic and hydrophobic interactions. Also crystallization attempts had been carried through with AgB8/3 oligomer to X-ray diffraction, and crystals have been obtained in three conditions of initial screening. These crystals difracted to maximum resolutions of 8 Å, and it was not possible to collect protein structural data. However, these crystallization conditions can be refined in order to obtain crystals of better quality.

Identificação e caracterização de proteínas expressas pelo metacestódeo de Echinococcus granulosus durante a infecção do seu hospedeiro intermediário

A hidatidose cística é uma zoonose helmíntica endêmica causada pela infecção com a forma larval ou metacestódeo de Echinococcus granulosus. Neste trabalho, foi realizada uma análise proteômica do metacestódeo de E. granulosus durante a infecção do seu hospedeiro intermediário bovino. A identificação de proteínas do parasito e do hospedeiro presentes nos diferentes componentes da larva do parasito (protoescólices, camada germinativa e líquido hidático) gerou informações valiosas sobre a relação parasitohospedeiro. Os resultados obtidos permitiram a caracterização da resposta imune montada pelo hospedeiro contra a infecção por E. granulosus, bem como de potenciais estratégias moleculares adotadas pelo parasito para evadir essa resposta e promover a sua sobrevivência dentro do corpo do hospedeiro. Além disso, novas proteínas-alvo foram identificadas para o desenvolvimento ou melhora de ferramentas de diagnóstico, tratamento e controle da hidatidose. Nesse trabalho, foi também estudada a estrutura do antígeno B (AgB) de E. granulosus, uma proteína implicada em múltiplas interações parasito-hospedeiro durante a infecção. Medidas de espalhamento de luz dinâmico utilizando subunidades recombinantes (AgB8/1, AgB8/2 e AgB8/3) demonstraram que o AgB forma diferentes estruturas moleculares sob condições fisiológicas, incluindo oligômeros e agregados de alta massa molecular. As subunidades recombinantes do AgB mostraram diferentes tendências agregativas, sendo os agregados de alta massa molecular formados por AgB8/3 os mais similares, em morfologia e tamanho, àqueles do AgB produzido pelo parasito. Experimentos de dissociação induzida por pressão revelaram que a formação de pontes dissulfeto confere maior estabilidade aos homo-oligômeros de AgB8/2 e AgB8/3. A composição de subunidades do AgB foi avaliada por espectrometria de massas (MS), identificando as subunidades AgB8/1, AgB8/3 e AgB8/4, o que indica uma contribuição principal dessas subunidades nas propriedades estruturais, biológicas e imunológicas do AgB de E. granulosus. A análise de MS indicou também uma associação entre AgB e Ag5, os principais antígenos secretados pelo metacestódeo de E. granulosus. / Cystic hydatid disease (CHD) is an endemic helminthic zoonosis caused by infection with the larval stage or metacestode of the tapeworm Echinococcus granulosus. Here, we performed a proteomic analysis of the E. granulosus metacestode during infection of its intermediate bovine host. Identification of parasite and host proteins present in different components of parasite larvae (protoscoleces, germinal layer and hydatid cyst fluid) provided valuable information on host-parasite interplay. Our results allowed the characterization of host immune response mounted against E. granulosus infection, as well as potential molecular strategies adopted by the parasite to avoid this response and promote its survival inside the host body. Moreover, new protein targets were identified for the development or improvement of CHD diagnosis, treatment, and control tools. In this work, we also study the structure of E. granulosus antigen B (AgB), a protein implicated in multiple host-parasite interactions during infection. Dynamic light scattering experiments with recombinant subunits (AgB8/1, AgB8/2 and AgB8/3) demonstrated that AgB form different molecular assemblies under physiological conditions, including oligomers and high-molecular-weight aggregates. AgB recombinant subunits showed different aggregative tendencies, being AgB8/3 high-molecular-weight aggregates most similar, both in morphology and size, to those of parasite-produced AgB. Pressure-induced dissociation experiments revealed that disulfide bonds formation confers greater stability to AgB8/2 and AgB8/3 homo-oligomers. AgB subunit composition was evaluated by mass spectrometry (MS), identifying AgB8/1, AgB8/3 and AgB8/4 subunits, which indicates a major contribution for these subunits on E. granulosus AgB structural, biological, and immunological properties. The MS analysis also indicated an association between AgB and Ag5, the major antigens secreted by E. granulosus metacestode.

Estudo de três enzimas da via glicolítica de Echinococcus granulosus com possíveis funções moonlighting na interação da forma larval com o hospedeiro intermediário

A expressão “proteínas moonlighting” refere-se a um subgrupo de proteínas multifuncionais, onde duas ou mais funções são realizadas por uma mesma cadeia polipeptídica. Dentre as proteínas com funções moonlighting estão diversas enzimas glicolíticas que podem desempenhar papeis importantes em processos-chave na biologia de organismos parasitas, como os de motilidade, adesão, invasão e diferenciação. A aldolase, a enolase e a lactato-desidrogenase (LDH) estão entre as enzimas que foram detectadas nos produtos de excreção-secreção (ES) e em tecidos de interface com o hospedeiro de Echinococcus granulosus, o cestódeo causador da hidatidose cística em seres humanos e ungulados domésticos. Neste contexto, a caracterização destas enzimas pode revelar possíveis funções moonligthing por elas desempenhadas e contribuir para a compreensão de aspectos básicos do desenvolvimento do parasito e de possíveis mecanismos de interação com os hospedeiros. Inicialmente, as sequências codificadoras da aldolase (EgAldo), da enolase (EgEno) e da LDH (EgLDH) de E. granulosus foram amplificadas por RT-PCR, clonadas em um vetor de expressão modificado (pGEX-TEV) e expressas em Escherichia coli, para a produção das proteínas recombinantes correspondentes (rEgAldo, rEgEno e rEgLDH, respectivamente). Soros de pacientes humanos com hidatidose reconhecem, em imunoblots, a EgAldo e EgEno nativas mas não reconhecem as proteínas recombinantes correspondentes, sugerindo o envolvimento de epitopos não proteicos no reconhecimento desses antígenos. Antissoros policlonais contra a rEgAldo e a rEgEno evidenciaram, em protoescólices, uma localização predominantemente citoplasmática para as proteínas nativas cognatas. Elas também foram detectadas na camada germinativa e no líquido hidático, que representam interfaces entre o parasito e o hospedeiro. Além disso, tanto a EgAldo como a EgEno foram reconhecidas em frações de proteínas tegumentares e em produtos de ES de protoescólices, localizações sugestivas de funções moonlighting. Proteínas do parasito e do hospedeiro intermediário que interagem com as proteínas rEgAldo, rEgEno e rEgLDH foram recuperadas por experimentos de GST pull-down para posterior identificação destas proteínas por espectrometria de massas. A identificação destas proteínas de interação poderá apontar processos não-glicolíticos nos quais estas enzimas estejam envolvidas. / The term “moonlighting proteins” refers to a subset of multifunctional proteins in which two or more different functions are performed by one polipeptide chain. Among the proteins described as moonlighting, there are many glycolytic enzymes that play important roles in key processes in the biology of parasites, such as motility, adhesion, invasion and cell differentiation. Aldolase, enolase and lactate dehydrogenase are among the enzymes that have been detected in the excretory-secretory (ES) products and in the host-interacting metacestode components from Echinococcus granulosus, a cestode that causes cystic hydatid disease and affects both humans and livestock. In this context, the characterization of these enzymes may reveal possible moonlighting functions they perform and contribute to the understanding of basic aspects of the parasite development and differentiation and possible interaction mechanisms with the intermediate host. Initially, the complete coding sequences from E. granulosus aldolase (EgAldo), enolase (EgEno) and lactate dehydrogenase (EgLDH) were amplified by RT-PCR, cloned into a modified expression vector (pGEX-TEV) and expressed in Escherichia coli for recombinant proteins production (rEgAldo, rEgEno and rEgLDH, respectively). Sera from human hydatid disease patients recognize in immunoblots the native EgAldo and EgEno, but not its recombinant counterparts, which was taken as evidence of involvement of non proteic epitopes in the EgAldo and EgEno recognition. Polyclonal antiserum against rEgAldo and rEgEno have shown, in protoscoleces, a predominantly cytoplasmic localization for the cognate native proteins, as expected for glycolytic enzymes. They have also been detected in the germinal layer and hydatid fluid, which represent host-parasite interfaces. Furthermore, both EgAldo and EgEno were recognized in protoscoleces tegument extracts and in ES products from these parasites in culture, which are indicative of moonlighting functions. Parasite and intermediate host proteins that interact with proteins rEgAldo, rEgEno and rEgLDH in vitro were recovered by GST pull-down assays and the identification of these proteins will be performed using mass spectrometry. Identification of interacting proteins may also be an indication that these enzymes perform nonglycolytic functions.
