Envolvimento da resposta imune humoral no desenvolvimento da anemia e das alterações quantitativas e qualitativas das plaquetas na erliquiose canina experimental / Involvement of humoral immune response on development of anemia and quantitative and qualitative platelets disorders in experimental canine ehrlichiosis

Com o objetivo de avaliar o envolvimento da resposta imune humoral no desenvolvimento da anemia e das alterações quantitativas e qualitativas das plaquetas na erliquiose canina, sete cães adultos, livres de infecção, foram inoculados com E. canis e acompanhados durante 10 semanas. Três cães permaneceram como controles. Os animais foram submetidos a exame físico diário e avaliação laboratorial (hemograma, contagem de plaquetas, mielograma, bioquímica sangüínea, fragilidade osmótica eritrocitária e teste de Coombs), reação em cadeia da polimerase (PCR), pesquisa de anticorpos anti-E. canis, antiplaquetários e antimegacariocíticos (IFI), e teste da atividade de agregação plaquetária, em diferentes momentos durante o período de observação. Febre, esplenomegalia, linfadenomegalia, trombocitopenia, anemia não regenerativa, aumento das atividades séricas da ALT, AST e FA foram as principais alterações clínicas e de patologia clínica observadas, com pico máximo entre o 21&ordm; e 28&ordm; dias pós-infecção. Houve diminuição da capacidade de agregação plaquetária dos animais infectados (p<0,05) nos dias 14, 21, 28 e 35 pós-infecção e aumento do número das células megacariocíticas, principalmente seus precursores (megacarioblastos e pró-megacariócitos) no 14&ordm; dia pós-infecção. Não foram detectados anticorpos antiplaquetários ou antimegacariocíticos. O teste de Coombs, bem como a fragilidade osmótica eritrocitária, mantiveram-se inalterados, comprovando não ter ocorrido hemólise imunomediada. Conclui-se que a anemia desenvolvida durante a fase aguda da infecção experimental por E. canis, de característica eritroregenerativa, ainda que tardia, pelo menos nos animais estudados, não teve envolvimento de mecanismo imunológico direcionado contra as hemácias, e ainda, que a causa da trombocitopenia e da hipofunção plaquetária observada durante esta fase da doença não parece estar relacionada ao desenvolvimento de mecanismos imunológicos direcionados contra as plaquetas ou seus precursores. / In order to determine the involvement of humoral and immune response on development of anemia and quantitative and qualitative platelets disorders in experimental canine ehrlichiosis, seven adult naïve dogs were inoculated with E. canis and followed during 10 weeks. Three dogs were remained as control. Daily clinical evaluation and laboratorial exams (hematological, platelet counts, bone marrow evaluation, biochemistry, erythrocyte osmotic fragility and Coombs? test) polymerase chain reaction assay (PCR), anti-E.canis, antiplatelet and antimegakaryocyte antibodies detection (IFA) and platelet aggregation studies were evaluated at different moments during all the experimental period. Fever, splenomegaly, lymphadenomegaly, thrombocytopenia, non-regenerative anemia and increase on liver?s enzymes serum activity (ALT, AST and ALP) were the main clinical and laboratorial findings observed, with the highest point between days 21 and 28 post-infection. The infected animals showed a decrease in platelet aggregation responses (p<0.05) on days 14, 21, 28 and 35 post-infection and increase of megakaryocytic cells, mainly its precursors (megakaryoblasts and promegakaryocytes) on day 14 post-infection. It weren?t detected any antiplatelet or antimegakaryocyte antibodies, as well as, there were no variations on erythrocyte osmotic fragility and Coombs? tests results, which confirms the absence of immune-mediated hemolysis. It is concluded that the anemia observed during the experimental acute phase of the E. canis infection, with regenerative pattern, even though late, at least in the studied animals, wasn?t related with immune mechanism against the red blood cells, and that, the thrombocytopenia and platelet dysfunction observed during this phase of the disease don?t seen to be related with immune mechanisms against platelets or its precursors.

The clinical and hematological abnormalities are varied, depending on the strain of Ehrlichia spp., the immune response of the dog and the concomitant presence of other diseases. Among the various hematological disorders, thrombocytopenia is a common feature. The disease can manifest itself through a variety of clinical veterinarians, particularly those working in endemic areas, should always consider the possibility of infection with Ehrlichia spp., especially in dogs with nonspecific signs of illness. This study aimed to evaluate clinical and hematologic recuperation of dogs naturally infected with Ehrlichia sp. and treated with doxycycline. The trial was performided at the Laboratory of Experimental Chemotherapy from the Department of Animal Parasitology of the Institute of Veterinary of the Universidade Federal Rural do Rio de Janeiro. About the total of 90 Beagles dogs kept in kennels experimental laboratory, twelve were selected, with clinical signs such as fever, anorexia, vomiting, weight loss, pale mucous membranes, hepatomegaly and splenomegaly and the presence of ticks R. sanguineus and find hematologic thrombocytopenia. The laboratory diagnosis was confirmed by direct identification of morulae of Ehrlichia sp. in blood smears stained with Giemsa. After weighing all of 12 dogs, they were individually treated with doxycycline at a dose of 10mg/kg body weight every 12 hours for 21 days, during which assessments were made of clinical and hematologic exams. Some clinical changes were observed during the evaluation period as fever, anorexia, vomiting, diarrhea, pale mucous membranes, splenomegaly and lymphadenopathy presenting these standard parameters the 21 days of treatment. The abnormalities found were thrombocytopenia and normochromic normocitic anemia. In the days +14 and +21, the mean counts of red blood cells and hematocrit value were within normal physiological parameters for the species. There was a statistically significant difference between days -1 and +21 by red blood counts and hematocrit values. At the end of treatment, all animals were normalized to platelet count, statistically significant differences between days -1 and +21. It is concluded that there was clinical-hematological normalization at the end of the treatment period and that it administered at a dose of 10mg/kg over 21 days produced no clinical and hematological reactions. Algumas altera??es cl?nicas foram observadas ao longo do per?odo de avalia??o como febre, anorexia, v?mito, diarr?ia, mucosas hipocoradas, esplenomegalia e linfadenomegalia apresentando-se estes par?metros normalizados ao 21? dia de tratamento. As altera??es hematol?gicas encontradas foram trombocitopenia e anemia normocit?ca normocr?mica. Nos dias +14 e +21, as contagens m?dias de hem?cias e valor de hemat?crito estavam dentro dos par?metros fisiol?gicos normais para a esp?cie. Houve diferen?a estat?stica significativa entre os dias -1 e +21 na contagem de hem?cias e nos valores de hemat?crito. Quanto a trombocitopenia, no fim do tratamento, todos animais estavam com plaquetometria normalizada, havendo diferen?a estat?stica significativa entre os dias -1 e +21. Conclui-se que houve uma normaliza??o dos quadros cl?nico-hematol?gicos ao final do per?odo de tratamento e que o mesmo administrado na dose de 10mg/Kg ao longo de 21 dias n?o produziu rea??es cl?nicas e hematol?gicas adversas. Dinâmica da infecção experimental de cães por Ehrlichia canis: aspectos clínicos, laboratoriais e resposta imune humoral e celular / Dynamics of experimental Ehrlichia canis infection of dogs: clinical and laboratorial aspects and humoral and cellular immune response

Com o objetivo de avaliar a evolução clínica, a persistência da infecção e a resposta imune humoral e celular, sete cães adultos desprovidos de anticorpos anti-Ehrlichia canis foram inoculados com E. canis e acompanhados durante vinte semanas. Três cães permaneceram como controle. Exame físico diário e avaliação laboratorial (hemograma, bioquímica sanguínea, fragilidade osmótica eritrocitária, teste de Coombs, teste de redução de tetrazólio nitroazul), reação em cadeia da polimerase (PCR), pesquisa de anticorpos anti-E. canis (IFI) e quantificação dos linfócitos T CD4+ e CD8+ por citometria de fluxo foram realizados, em diferentes momentos, durante o período de observação. A infecção foi comprovada pela presença de mórulas em linfócitos/monócitos no sangue circulante, soroconversão, com títulos de anticorpos entre 1.280 a 20.480 e a reação positiva ao PCR. Febre, esplenomegalia, linfadenomegalia, trombocitopenia, anemia não regenerativa, aumento das atividades séricas de ALT, AST e FA, foram as principais alterações clínicas e de patologia clínica observadas, com pico máximo entre 21 e 28 dias. Também nesses momentos foram observados o aumento de linfócitos T CD8+ e a diminuição dos linfócitos T CD4+. O aumento do metabolismo oxidativo dos neutrófilos, quando estimulados, entre o 28&ordm; e 35&ordm; dia p.i., o aumento de reticulócitos na circulação sanguínea nessa ocasião, a redução dos linfócitos T CD4+ e o aumento de linfócitos T CD8+ indicaram a resposta do hospedeiro, na tentativa de eliminar a infecção. No auge da infecção entre o 21&ordm; e 49&ordm; dia p.i., houve a inversão da relação CD4:CD8, principalmente o aumento dos linfócitos T CD8+. A fragilidade osmótica eritrocitária manteve-se inalterada, bem como o teste de Coombs, comprovando não ter ocorrido a hemólise imunomediada. Nos cães infectados não foram detectados a presença de anticorpos IgM anti-E. canis em nenhum momento de observação. O aumento de reticulócitos na circulação sanguínea a partir do 28&ordm; dia p.i. sugere ser temporária a supressão da atividade eritropoética. O baixo número de linfócitos T CD4+, coincidindo com a esplenomegalia e a PCR positiva até a vigésima semana p.i., sugere a relação entre a persistência da infecção, com a localização esplênica de E. canis e ativação de um mecanismo de escape pelo microrganismo, representado pelo baixo limiar de ativação de linfócitos T CD4+. Por outro lado, o aumento de linfócitos T CD8+ no auge da infecção coincidindo com a melhora clínica, sugere o envolvimento do mecanimo de citotoxicidade na patogenia da erliquiose canina. O bloqueio parcial da atividade dos linfócitos T CD4+, constitui-se provavelmente em um dos mecanismos de escape da E. canis, enquanto a contínua liberação de TNF-&alpha; pelas células citotóxicas, como parte do mecanismo do hospedeiro para a eliminação do parasita pode resultar na pancitopenia terminal observado freqüentemente nos cães cronicamente infectados por E. canis. / The clinical evolution, the persistence of infection and humoral and cellular immune response were evaluated on seven adult naïve dogs experimentally inoculated with an E. canis strain and followed during 20 weeks. Three dogs were remained as control. Daily clinical evaluation and laboratorial exams (hematological, biochemistry, erythrocyte osmotic fragility, Coombs´ test, neutrophils oxidative metabolism by nitroblue tetrazolium reduction test), polymerase chain reaction assay (PCR), anti-E. canis antibodies detection by indirect immunofluorescence assay (IFA) and quantification of CD4+ and CD8+ T lymphocytes by flow cytometry technique were evaluated, at different moments during all the experimental period. The presence of E. canis morulae, seroconversion with high antibodies titers (1,280 to 20,480) and positive PCR had confirmed the infection in all the inoculated animals. Fever, splenomegaly, lymphadenomegaly, thrombocytopenia, non-regenerative anemia and increase on liver´s enzymes serum activity (ALT, AST and ALP) were the main clinical and laboratorial changes observed, with the highest point between days 21 and 28 post-infection (p.i.). Also at these moments were observed increase of CD8+ T lymphocytes and reduction of the CD4+ T lymphocytes. The increase in the stimulated neutrophils oxidative metabolism between days 28 and 35 p.i., and the increase of reticulocytes in the same period, allied to the reduction of the CD4+ T lymphocytes and the increase of CD8+ T lymphocytes, indicated the host´s immune response in the attempt to eliminate the infection. On the peak of the infection, from the 21st to the 49th post-infection day, there was an inversion in the CD4:CD8 ratio of blood lymphocytes, mainly caused by increase of the CD8+ T lymphocytes. There was no variation on erythrocyte osmotic fragility and Coombs? test results, which suggested the absence of immune-mediated hemolysis. It wasn´t detected any anti-E. canis IgM antibodies in the infected group of dogs. The increase of reticulocytes only from the 28th post-infection day suggests a temporary erythropoietic suppression. The low number of CD4+ T lymphocytes, coinciding with the splenomegaly and the positive PCR until the twentieth week p.i., suggest that there is a relation between the persistent infection, the splenic localization of E. canis and an evasion mechanism, represented for the low threshold of activation of CD4+ T lymphocytes. On the other hand, the increase of CD8+ T lymphocytes in the peak of infection, coinciding with clinical improvement, suggests the involvement of the cytotoxicity mechanism on canine ehrlichiosis pathogenesis. The partial blockage of CD4+ T lymphocytes activity could probably be one of the E. canis escape mechanisms, while the continue TNF-&alpha; release by host´s cytotoxic cells, as a mechanism to eliminate the parasite, could result in terminal pancytopenia commonly observed in dogs chronically infected with E. canis.

Pesquisa de Rickettsia sp. e Ehrlichia spp. em canídeos e felídeos selvagens de vida livre e cães domésticos da região do Parque Nacional das Emas, Goiás / Survey of Rickettsia sp. and Ehrlichia spp. in free ranging wild canids and felids and domestic dogs in the region of Emas National Park, Goiás, Brazil

Este estudo teve como objetivo pesquisar a presença de Rickettsia sp. e Ehrlichia sp. em canídeos e felídeos selvagens de vida livre e cães domésticos da região do Parque Nacional das Emas (PNE), Goiás. Os anticorpos detectados através da Reação de Imunofluorescência Indireta utilizando 6 espécies de riquétsias. Amostras de soros de canídeos (140) e felídeos (38) selvagens de vida livre, incluindo lobos-guarás (76), cachorros-do-mato (57), gatos-palheiros (19), jaguatiricas (10), raposinhas-do-campo (7), onças-pintadas (4), onças-pardas (4), gato-mourisco (1), e cães domésticos (134) das propriedades rurais do entorno do PNE. Para a detecção de Ehrlichia sp. foi realizada a técnica de PCR, utilizando amostras de sangues totais de canídeos selvagens (44), sendo cachorros-do-mato (26), lobos-guarás (16) e raposinhas-o-campo (2) e cães domésticos (17). Amostras de lobos-guarás (29), cachorros-do-mato (12), onças-pardas (2), gatos-palheiros (2), raposinha-do-campo (1) e cachorro doméstico (1) apresentaram títulos para uma ou mais riquétsias testadas. Para Ehrlichia sp., 6 cães domésticos apresentaram resultados positivos, com Ehrlichia canis com semelhança de 100% com a cepa Uberlândia, pelo sequenciamento do gene dsb. Este é o primeiro relato de detecção de anticorpos, sugerindo provável exposição a R. amblyommii, R. rickettsii e R. parkeri em lobos-guarás, cachorros-do-mato e onças-pardas; R. parkeri em raposinhas-do-campo; R. amblyommii e R. parkeri em gatos-palheiros; R. amblyommii, R. rickettsii e R. rhipicephali em jaguatiricas. Não houve evidências que comprovassem ou mesmo sugerissem a transmissão desses patógenos entre animais silvestres e domésticos. / This study aimed to investigate the presence of Rickettsia sp. and Ehrlichia sp. in free-ranging wild canids and felids, and domestic dogs in the region of Emas National Park (ENP), Goiás. The antibodies were detected through Immunofluorescence Assay using 6 rickettsiae species. Serum samples of free-ranging wild canids (140) and felids (38), including maned wolves (76), crab-eating foxes (57), pampas cats (19), ocelots (10), hoary foxes (7), jaguars (4), pumas (4) and jaguarondi (1), and domestic dogs (134) from surrounding farms were tested. For detection of Ehrlichia sp., PCR was performed on whole blood samples of canids (44), among crab-eating foxes (26), maned wolves (16) and hoary foxes (2), and domestic dogs (17). Samples of maned wolves (29), crab-eating foxes (12), pumas (2), pampas cats (2), hoary fox (1) and domestic dog (1) presented one or more antibody titers for rickettsiae testing. For Ehrlichia spp. detection, 6 domestic dogs showed positive results for Ehrlichia canis with 100% similarity to the Uberlândia strain, by sequencing of dsb gene. This is the first report of antibodies detection, suggesting probable exposure to R. amblyommii, R. rickettsii, R. parkeri in maned wolves, crab-eating foxes and pumas; R. parkeri in hoary foxes; R. amblyommii and R. parkeri pampas cats; R. amblyommii, R. rickettsii, R. rhipicephali in ocelots. There was no evidence to conclusively prove or even suggest the transmission of pathogens between wild and domestic animals.

CERTAINES D'ENTRE ELLES SUREXPRIMEES CHEZ LES SOUCHES ATTENUEES SEMBLENT ETRE DE BONS CANDIDATS VACCINAUX ET DEVRAIENT ETRE ETUDIEES / Transcriptomic study of gardel and senegal both virulent and attenuated e. ruminantium strains was conducted during my phd. an analysis of transcriptome at different stages of development has been first conducted for virulent gardel strain. at reticulate body stage (intracellular form non-infectious), over-expression of genes coding for proteins involved in metabolism, transport and exchange of nutrients and resistance to oxidative stress was observed. at this stage of development, e. ruminantium seems to activate mechanisms for its survival and development within the host cell. at elementary body stage, dksa the gene encoding for a transcription factor was over-expressed. this gene has been shown to be involved in the regulation of virulence factors. it seems, therefore, at the elementary body stage, e. ruminantium induces its virulence factors. secondly, we compare the transcriptome of elementary body between virulent and attenuated strains. our results showed an important membrane modification of attenuated and virulent strains. for attenuated strains, we observed an over-expression of genes involved in membrane biogenesis and a diminution of expression of map multigenic family. it seems that map proteins subvert the protective immune response. hypothetical membrane proteins are over-expressed in both virulent and attenuated strains. some over-expressed proteins in attenuated strains seem to be good vaccine candidates and willstudied.

Distribution and prevalence of Ehrlichia chaffeensis, the causative agent of human monocytic ehrlichiosis, in Indiana and Ohio

Human monocytic ehrlichiosis (HME) is a tick-borne infectious disease caused by the bacterium Ehrlichia chaffeensis and transmitted by the ixodid tick Amblyomma americanum. The first confirmed case of HME in Indiana occurred in 1994. Since then, there have been an additional 17 confirmed cases reported from 11 counties.A total of 498 A. americanum and 25 Dermacentor variabilis ticks were collected from counties in southern Indiana during May and June 1998, pooled, and examined for the presence of E. chaffeensis using nested PCR with primers HE 1 and HE3, which are specific for the 16S rRNA gene of E. chaffeensis. Ten pools of adult A. americanum specimens tested positive for E. chaffeensis DNA. This represented a minimum infection rate (MIR) of 3.82%. None of the A. americanum nymphs or adult D. variabilis ticks tested positive.In addition, 325 white-tailed deer blood samples from Indiana and 327 from Ohio were collected during November, 1998 and tested for the presence of E. chaffeensisreactive antibodies using an indirect immunofluoescence assay (IFA). Evidence of such antibodies was found in deer killed in six Indiana counties where infection rates ranged from 43% - 64% and four Ohio counties where infection rates ranged from 4% - 25%.The results from this study support the view that the distribution of E. chaffeensis closely follows that of A. americanum in the North Central United States. This is the first report of E. chaffeensis-reactive antibodies in white-tailed deer from Ohio. / Department of Biology

Esses parasitas são responsáveis pela transmissão de agentes infecciosos como as bactérias dos gêneros Rickettsia e Ehrlichia e também protozoários dos gêneros Hepatozoon e Babesia. Relatos mostram que carrapatos do gênero Amblyomma, Ixodes e Rhipicephalus podem atuar como vetores na transmissão destes agentes. Esses carrapatos, nos estágios imaturos geralmente parasitam pequenos mamíferos terrestres enquanto os adultos parasitam mamíferos de médio e grande porte. Levando em conta a diversidade de pequenos mamíferos terrestres do Pantanal e sendo eles hospedeiros para os carrapatos vetores desses agentes infecciosos, o presente estudo teve como objetivo verificar a ocorrência de infecção por Rickettsia spp., Ehrlichia spp., Anaplasma spp., Hepatozoon spp. e Babesia spp., bem como o parasitismo por carrapatos em pequenos mamíferos terrestres da subregião do Pantanal de Poconé. As coletas foram realizadas em oito habitats distintos onde foram instaladas grades amostrais contendo 100 armadilhas cada. Dos animais coletados foram retiradas amostras de sangue, fígado e baço. A detecção dos agentes foi realizada por meio de Reação em Cadeia pela Polimerase (PCR). Foram coletados 64 indivíduos entre marsupiais e roedores, dos quais dez apresentavam parasitismo por carrapatos, sendo um animal parasitado por Amblyomma cajennense, um por Amblyomma triste e oito por Argasideos. Nas PCRs, um roedor (1,6%) foi positivo para o gênero Anaplasma; três (4,7%) foram positivos para Hepatozoon spp., sendo todos eles roedores e quatro (6,2%) foram positivos para Babesia spp., dos quais três (75%) eram roedores e um (25%) era marsupial. Nenhum animal apresentou resultado positivo para Rickettsia spp. ou Ehrlichia spp. Após o sequenciamento das amostras positivas, observou-se para Hepatozoon um fragmento de 560 pb do gene 18S rRNA, que apresentou 99% de similaridade com Hepatozoon sp. (FJ719819) detectado em marsupial do Chile. Para Babesia observou-se à amplificação de dois fragmentos distintos do gene 18S rRNA, um de 532 pb detectado em roedor e outro de 556 pb detectado em marsupial. Ambos apresentaram similaridade de 95% entre si e foram 97% similares com B. bicornis (AF419313). Os resultados sugerem que novas espécies ou genótipos ainda não descritos dos protozoários dos gêneros Hepatozoon e Babesia estão presentes na área estuda e que os pequenos mamíferos podem estar atuando como hospedeiros para esses agentes na região do Pantanal mato-grossense. / Ticks are parasites of a wide range of hosts that can transmit several pathogens. These parasites are responsible for the transmission of infectious agents such as bacteria of Rickettsia and Ehrlichia genus and also protozoa of Babesia and Hepatozoon genus. Previous reports show that Amblyomma, Ixodes and Rhipicephalus ticks genus can act as vectors in the transmission of these agents. Larvae and nymphs stages of the related ticks usually parasitize terrestrial small mammals while adults prey on large mammals. Taking into account the diversity of small terrestrial mammals of the Pantanal, the present study aimed to verify the occurrence of infection by Ehrlichia spp., Anaplasma spp., Rickettsia spp., Hepatozoon spp., Babesia spp. and parasitism by ticks in terrestrial small mammals of the Pantanal of Poconé region. Samples were collected in eight different habitats where sampling grids were installed containing 100 traps each. Samples of blood, liver and spleen were collected from animals captured. The detection of agents was performed by the Polymerase Chain Reaction (PCR). Sixty-four animals, marsupials and rodents were collected, and ten presented parasitism by ticks, one parasitized by Amblyomma cajennense, one by Amblyomma triste and eight by Argasidae. In the PCR analysis, one rodent (1.6%) was positive for the genus Anaplasma; three rodents (4,7%) were positive for Hepatozoon spp. and four (6,2%) were positive for Babesia spp., three of them (75%) were rodents and one (25%) was a marsupial. No animal presented positive result for Rickettsia spp. or Ehrlichia spp. After the sequencing analysis of positive samples, there was observed a fragment of 560 pb of 18S rRNA of Hepatozoon showed 99% of similarity with Hepatozoon sp. (FJ719819) detected in a marsupial species from Chile. For Babesia two distinct fragments of the 18S rRNA gene were amplified, one of 532 pb detected in a rodent and one of 556 pb detected in a marsupial. Both showed 95% of similarity with each other and were 97% similar to B. bicornis (AF419313).The results suggest that new genotypes or not described protozoa of the Babesia and Hepatozoon genus are present in the study area and small mammals may be acting as reservoirs for these agents in the Pantanal region.

Envolvimento da resposta imune humoral no desenvolvimento da anemia e das alterações quantitativas e qualitativas das plaquetas na erliquiose canina experimental / Involvement of humoral immune response on development of anemia and quantitative and qualitative platelets disorders in experimental canine ehrlichiosis

Com o objetivo de avaliar o envolvimento da resposta imune humoral no desenvolvimento da anemia e das alterações quantitativas e qualitativas das plaquetas na erliquiose canina, sete cães adultos, livres de infecção, foram inoculados com E. canis e acompanhados durante 10 semanas. Três cães permaneceram como controles. Os animais foram submetidos a exame físico diário e avaliação laboratorial (hemograma, contagem de plaquetas, mielograma, bioquímica sangüínea, fragilidade osmótica eritrocitária e teste de Coombs), reação em cadeia da polimerase (PCR), pesquisa de anticorpos anti-E. canis, antiplaquetários e antimegacariocíticos (IFI), e teste da atividade de agregação plaquetária, em diferentes momentos durante o período de observação. Febre, esplenomegalia, linfadenomegalia, trombocitopenia, anemia não regenerativa, aumento das atividades séricas da ALT, AST e FA foram as principais alterações clínicas e de patologia clínica observadas, com pico máximo entre o 21&ordm; e 28&ordm; dias pós-infecção. Houve diminuição da capacidade de agregação plaquetária dos animais infectados (p<0,05) nos dias 14, 21, 28 e 35 pós-infecção e aumento do número das células megacariocíticas, principalmente seus precursores (megacarioblastos e pró-megacariócitos) no 14&ordm; dia pós-infecção. Não foram detectados anticorpos antiplaquetários ou antimegacariocíticos. O teste de Coombs, bem como a fragilidade osmótica eritrocitária, mantiveram-se inalterados, comprovando não ter ocorrido hemólise imunomediada. Conclui-se que a anemia desenvolvida durante a fase aguda da infecção experimental por E. canis, de característica eritroregenerativa, ainda que tardia, pelo menos nos animais estudados, não teve envolvimento de mecanismo imunológico direcionado contra as hemácias, e ainda, que a causa da trombocitopenia e da hipofunção plaquetária observada durante esta fase da doença não parece estar relacionada ao desenvolvimento de mecanismos imunológicos direcionados contra as plaquetas ou seus precursores. / In order to determine the involvement of humoral and immune response on development of anemia and quantitative and qualitative platelets disorders in experimental canine ehrlichiosis, seven adult naïve dogs were inoculated with E. canis and followed during 10 weeks. Three dogs were remained as control. Daily clinical evaluation and laboratorial exams (hematological, platelet counts, bone marrow evaluation, biochemistry, erythrocyte osmotic fragility and Coombs? test) polymerase chain reaction assay (PCR), anti-E.canis, antiplatelet and antimegakaryocyte antibodies detection (IFA) and platelet aggregation studies were evaluated at different moments during all the experimental period. Fever, splenomegaly, lymphadenomegaly, thrombocytopenia, non-regenerative anemia and increase on liver?s enzymes serum activity (ALT, AST and ALP) were the main clinical and laboratorial findings observed, with the highest point between days 21 and 28 post-infection. The infected animals showed a decrease in platelet aggregation responses (p<0.05) on days 14, 21, 28 and 35 post-infection and increase of megakaryocytic cells, mainly its precursors (megakaryoblasts and promegakaryocytes) on day 14 post-infection. It weren?t detected any antiplatelet or antimegakaryocyte antibodies, as well as, there were no variations on erythrocyte osmotic fragility and Coombs? tests results, which confirms the absence of immune-mediated hemolysis. It is concluded that the anemia observed during the experimental acute phase of the E. canis infection, with regenerative pattern, even though late, at least in the studied animals, wasn?t related with immune mechanism against the red blood cells, and that, the thrombocytopenia and platelet dysfunction observed during this phase of the disease don?t seen to be related with immune mechanisms against platelets or its precursors.

Dinâmica da infecção experimental de cães por Ehrlichia canis: aspectos clínicos, laboratoriais e resposta imune humoral e celular / Dynamics of experimental Ehrlichia canis infection of dogs: clinical and laboratorial aspects and humoral and cellular immune response

Com o objetivo de avaliar a evolução clínica, a persistência da infecção e a resposta imune humoral e celular, sete cães adultos desprovidos de anticorpos anti-Ehrlichia canis foram inoculados com E. canis e acompanhados durante vinte semanas. Três cães permaneceram como controle. Exame físico diário e avaliação laboratorial (hemograma, bioquímica sanguínea, fragilidade osmótica eritrocitária, teste de Coombs, teste de redução de tetrazólio nitroazul), reação em cadeia da polimerase (PCR), pesquisa de anticorpos anti-E. canis (IFI) e quantificação dos linfócitos T CD4+ e CD8+ por citometria de fluxo foram realizados, em diferentes momentos, durante o período de observação. A infecção foi comprovada pela presença de mórulas em linfócitos/monócitos no sangue circulante, soroconversão, com títulos de anticorpos entre 1.280 a 20.480 e a reação positiva ao PCR. Febre, esplenomegalia, linfadenomegalia, trombocitopenia, anemia não regenerativa, aumento das atividades séricas de ALT, AST e FA, foram as principais alterações clínicas e de patologia clínica observadas, com pico máximo entre 21 e 28 dias. Também nesses momentos foram observados o aumento de linfócitos T CD8+ e a diminuição dos linfócitos T CD4+. O aumento do metabolismo oxidativo dos neutrófilos, quando estimulados, entre o 28&ordm; e 35&ordm; dia p.i., o aumento de reticulócitos na circulação sanguínea nessa ocasião, a redução dos linfócitos T CD4+ e o aumento de linfócitos T CD8+ indicaram a resposta do hospedeiro, na tentativa de eliminar a infecção. No auge da infecção entre o 21&ordm; e 49&ordm; dia p.i., houve a inversão da relação CD4:CD8, principalmente o aumento dos linfócitos T CD8+. A fragilidade osmótica eritrocitária manteve-se inalterada, bem como o teste de Coombs, comprovando não ter ocorrido a hemólise imunomediada. Nos cães infectados não foram detectados a presença de anticorpos IgM anti-E. canis em nenhum momento de observação. O aumento de reticulócitos na circulação sanguínea a partir do 28&ordm; dia p.i. sugere ser temporária a supressão da atividade eritropoética. O baixo número de linfócitos T CD4+, coincidindo com a esplenomegalia e a PCR positiva até a vigésima semana p.i., sugere a relação entre a persistência da infecção, com a localização esplênica de E. canis e ativação de um mecanismo de escape pelo microrganismo, representado pelo baixo limiar de ativação de linfócitos T CD4+. Por outro lado, o aumento de linfócitos T CD8+ no auge da infecção coincidindo com a melhora clínica, sugere o envolvimento do mecanimo de citotoxicidade na patogenia da erliquiose canina. O bloqueio parcial da atividade dos linfócitos T CD4+, constitui-se provavelmente em um dos mecanismos de escape da E. canis, enquanto a contínua liberação de TNF-&alpha; pelas células citotóxicas, como parte do mecanismo do hospedeiro para a eliminação do parasita pode resultar na pancitopenia terminal observado freqüentemente nos cães cronicamente infectados por E. canis. / The clinical evolution, the persistence of infection and humoral and cellular immune response were evaluated on seven adult naïve dogs experimentally inoculated with an E. canis strain and followed during 20 weeks. Three dogs were remained as control. Daily clinical evaluation and laboratorial exams (hematological, biochemistry, erythrocyte osmotic fragility, Coombs´ test, neutrophils oxidative metabolism by nitroblue tetrazolium reduction test), polymerase chain reaction assay (PCR), anti-E. canis antibodies detection by indirect immunofluorescence assay (IFA) and quantification of CD4+ and CD8+ T lymphocytes by flow cytometry technique were evaluated, at different moments during all the experimental period. The presence of E. canis morulae, seroconversion with high antibodies titers (1,280 to 20,480) and positive PCR had confirmed the infection in all the inoculated animals. Fever, splenomegaly, lymphadenomegaly, thrombocytopenia, non-regenerative anemia and increase on liver´s enzymes serum activity (ALT, AST and ALP) were the main clinical and laboratorial changes observed, with the highest point between days 21 and 28 post-infection (p.i.). Also at these moments were observed increase of CD8+ T lymphocytes and reduction of the CD4+ T lymphocytes. The increase in the stimulated neutrophils oxidative metabolism between days 28 and 35 p.i., and the increase of reticulocytes in the same period, allied to the reduction of the CD4+ T lymphocytes and the increase of CD8+ T lymphocytes, indicated the host´s immune response in the attempt to eliminate the infection. On the peak of the infection, from the 21st to the 49th post-infection day, there was an inversion in the CD4:CD8 ratio of blood lymphocytes, mainly caused by increase of the CD8+ T lymphocytes. There was no variation on erythrocyte osmotic fragility and Coombs? test results, which suggested the absence of immune-mediated hemolysis. It wasn´t detected any anti-E. canis IgM antibodies in the infected group of dogs. The increase of reticulocytes only from the 28th post-infection day suggests a temporary erythropoietic suppression. The low number of CD4+ T lymphocytes, coinciding with the splenomegaly and the positive PCR until the twentieth week p.i., suggest that there is a relation between the persistent infection, the splenic localization of E. canis and an evasion mechanism, represented for the low threshold of activation of CD4+ T lymphocytes. On the other hand, the increase of CD8+ T lymphocytes in the peak of infection, coinciding with clinical improvement, suggests the involvement of the cytotoxicity mechanism on canine ehrlichiosis pathogenesis. The partial blockage of CD4+ T lymphocytes activity could probably be one of the E. canis escape mechanisms, while the continue TNF-&alpha; release by host´s cytotoxic cells, as a mechanism to eliminate the parasite, could result in terminal pancytopenia commonly observed in dogs chronically infected with E. canis.

Este estudo teve como objetivo pesquisar a presença de Rickettsia sp. e Ehrlichia sp. em canídeos e felídeos selvagens de vida livre e cães domésticos da região do Parque Nacional das Emas (PNE), Goiás. Os anticorpos detectados através da Reação de Imunofluorescência Indireta utilizando 6 espécies de riquétsias. Amostras de soros de canídeos (140) e felídeos (38) selvagens de vida livre, incluindo lobos-guarás (76), cachorros-do-mato (57), gatos-palheiros (19), jaguatiricas (10), raposinhas-do-campo (7), onças-pintadas (4), onças-pardas (4), gato-mourisco (1), e cães domésticos (134) das propriedades rurais do entorno do PNE. Para a detecção de Ehrlichia sp. foi realizada a técnica de PCR, utilizando amostras de sangues totais de canídeos selvagens (44), sendo cachorros-do-mato (26), lobos-guarás (16) e raposinhas-o-campo (2) e cães domésticos (17). Amostras de lobos-guarás (29), cachorros-do-mato (12), onças-pardas (2), gatos-palheiros (2), raposinha-do-campo (1) e cachorro doméstico (1) apresentaram títulos para uma ou mais riquétsias testadas. Para Ehrlichia sp., 6 cães domésticos apresentaram resultados positivos, com Ehrlichia canis com semelhança de 100% com a cepa Uberlândia, pelo sequenciamento do gene dsb. Este é o primeiro relato de detecção de anticorpos, sugerindo provável exposição a R. amblyommii, R. rickettsii e R. parkeri em lobos-guarás, cachorros-do-mato e onças-pardas; R. parkeri em raposinhas-do-campo; R. amblyommii e R. parkeri em gatos-palheiros; R. amblyommii, R. rickettsii e R. rhipicephali em jaguatiricas. Não houve evidências que comprovassem ou mesmo sugerissem a transmissão desses patógenos entre animais silvestres e domésticos. / This study aimed to investigate the presence of Rickettsia sp. and Ehrlichia sp. in free-ranging wild canids and felids, and domestic dogs in the region of Emas National Park (ENP), Goiás. For detection of Ehrlichia sp., PCR was performed on whole blood samples of canids (44), among crab-eating foxes (26), maned wolves (16) and hoary foxes (2), and domestic dogs (17). Samples of maned wolves (29), crab-eating foxes (12), pumas (2), pampas cats (2), hoary fox (1) and domestic dog (1) presented one or more antibody titers for rickettsiae testing. For Ehrlichia spp. detection, 6 domestic dogs showed positive results for Ehrlichia canis with 100% similarity to the Uberlândia strain, by sequencing of dsb gene. This is the first report of antibodies detection, suggesting probable exposure to R. amblyommii, R. rickettsii, R. parkeri in maned wolves, crab-eating foxes and pumas; R. parkeri in hoary foxes; R. amblyommii and R. parkeri pampas cats; R. amblyommii, R. rickettsii, R. rhipicephali in ocelots. There was no evidence to conclusively prove or even suggest the transmission of pathogens between wild and domestic animals.
