Obtenção, manutenção e caracterização de células-tronco embrionárias equinas /

Orientador: Fernanda da Cruz Landim Alvarenga / Banca: Rogério Martins Amorim / Banca: José Antonio Visintin / Resumo: A característica inerente às células-tronco embrionárias de se manterem estáveis durante longos períodos de cultivo possibilita que um grande número de células indiferenciadas sejam utilizadas em procedimentos terapêuticos ou realização de pesquisas. Aproximadamente 97 doenças genéticas equinas têm alta homologia com defeitos genéticos de humanos. O estudo de CTE de animais ungulados é útil no estudo de biofarmacos, engenharia genética e elucidação da embriogênese. Trabalhos sobre o isolamento, cultivo e caracterização do perfil genético de CTE equinas são escassos na literatura. No presente estudo, três diferentes métodos do isolamento da MCI foram testados em blastocistos equinos, avaliando o potencial de derivação de linhagens e a expressão do gene Oct-4: 1) separação mecânica (n=26), 2) imunocirurgia (n=6), 3) aspiração por micromanipulação (n=4). Todas as MCI foram cultivadas nas mesmas condições: monocamada de fibroblastos inativadas, meio DMEM/F12 suplementado com LIF, soro fetal bovino, aminoácidos não essenciais, sódio piruvato, l-glutamina e 2-mercaptoetanol. A eficácia do isolamento da MCI e o estabelecimento de linhagens foram avaliados quanto à adesão, aspectos morfológicos dos agregados celulares e velocidade de crescimento. As linhagens estabelecidas foram caracterizadas pela expressão do gene Oct-4 e Nanog. Nenhuma das MCI isoladas do grupo 3 aderiram à placa, e todas morreram entre 48 e 72 horas de cultivo. No grupo 1, uma MCI apresentou crescimento satisfatório apenas durante o cultivo primário. Colônias foram estabelecidas no grupo 2 e, após a segunda passagem, as células foram avaliadas e congeladas. Utilizou-se a RT-PCR para a detecção da expressão dos genes Oct-4 e Nanog em amostras de CTE, anel coriônico e fibroblastos fetais equinos. (Resumo completo, clicar acesso eletrônico abaixo) / Abstract: The ability of Embryonic Stem cells to be maintained during extended culture periods would support the existence of large pools of undifferentiated stem cells for therapeutic and research applications. Approximately 97 equine genetic diseases have high homology to human genetic defects. ES cell lines from domestic ungulates would also be useful for disease resistance, for biopharming, and in genetic engineering. Studies of the isolation of equine embryonic stem cells and their genetic expression are quite limited. In the present study three different experimental approaches were used for ICM isolation from day 8 blastocysts and further ES cell line derivation and OCT-4 and Nanog expression: 1. Mechanical Process (26), 2. Immunosurgery (6) and 3. Aspiration of the ICM (4). All the ICM were maintained at the same conditions: under inactivated murine fibroblast feeder support in medium supplemented with Leukemia Inhibitor Factor, Fetal Calf Serum, Non essential amino acids, Sodium Pyruvate, L-Glutamine and 2-Mercaptoethanol changed every 24 hours. The colonies were evaluated by attachment, size and shape, type of cells and rate of growth. In addition, established ES cell colonies were characterized by OCT-4 expression. Only ICM outgrowths and colonies containing circular, small, and high ratio nucleo:cytoplasm cells were considered and passaged. None ICM from group 3 attached and all died after 48 to 72 hours after isolation. In group 1, one ICM had successful development but died after the first passage. Colonies were established from group 2. And after proliferation for 2 passages the cells were analyzed and frozen. RT-PCR was performed to evaluate the OCT-4 expression from ES cells, fetal fibroblast and day 32-34 chorionic girdle samples. Além disso, a CT apresenta propriedades de auto-renovação, gerando cópias idênticas a si mesma. De acordo com sua origem, as células-tronco podem ser chamadas de adultas e embrionárias. As células-tronco adultas (CTA) mais utilizadas nas clínicas de terapia celular são as células-tronco hematopoiéticas e as células tronco mesenquimais, encontradas principalmente na medula óssea, tecido adiposo e no sangue do cordão umbilical. As células-tronco embrionárias (CTE) são derivadas da massa celular interna de embriões no estágio de blastocisto. Desta maneira este trabalho teve como objetivo desenvolver uma metodologia adequada para o isolamento, cultivo e caracterização de células tronco embrionárias e mesenquimais de eqüinos, além de verificar a capacidade que as células possuem em se diferenciar in vitro em outros tipos célulares. Foi coletado sangue da medula óssea de eqüinos entre 8 e 15 anos de idade. As células tronco mesenquimais foram isoladas após a primeira e segunda passagem. As células foram caracterizadas com marcadores de superfície CD34 (mononucleares) e CD44 (mesenquimais). Após isolamento e caracterização, as células tronco mesenquimais foram diferenciadas para as linhagens osteogênica, adipogênica, condrogênica e neurogênica. A confirmação da diferenciação das células tronco foi realizada por marcadores teciduais específicos. Estas células também, foram capazes de expressarem marcadores neurais. Para o isolamento das células tronco embrionária eqüina, embriões com oito a nove dias foram coletado e a massa celular interna (MCI) isolada mecanicamente. Após o isolamento, a MCI foi transferida para a placa de cultivo previamente preparada com monocamada de fibroblastos para o desenvolvimento... / The stem cell (SC) is defined as cells with the capacity of generate different cellular types and rebuild various tissues. Moreover, the SC has a selfregenerate ability, generating identical copies of itself. According to its origins, the SC can be named as “adult” or “embryonic”. The adult stem cell (ASC) more often used in clinical trials and cellular therapy, are the hematopoietic stem cells and the mesenchymal stem cells, isolated mainly from the marrow bone, adipose tissue and umbilical cord blood. The embryonic stem cells (ESC) are obtained from the inner cell mass of embryos at the blastocyst stage. In this way the present study had as objective to develop an adequate methodology of isolation, culture and characterization of embryonic and mesechymal stem cells from horses, verifying the capacity of those cells to differentiate in vitro into different cells types. Bone marrow blood was collected from horses, aging from 8 to 15 years and filtered with a donation blood kit filter, to avoid clots. The mesenchymal stem cells were isolated after the first and the second passage. The SC were characterized using surface markers CD34 (monuclear) and CD44 (mesenchymal). After the isolation and characterization, the mesenchymal stem cells were differenced into osteogenic, adipogenic, condrogenic and neurogenic lineage. The cells differentiations were confirmed using specific tissue markers. To isolate the embryonic stem cells equine embryos with 8 to 9 days were used. The inner cell mass (ICM) were extract mechanically and transferred to a culture dish previously prepared with fibroblasts monolayer to colony formation and development. The colonies were characterized with pluripotency markers and then submitted to a differentiation process into neurogenic lineage, confirmed by specific neural tissue markers

Análise biquímica do líquido amniótico e alantoideano do Equus caballus em diferentes fases da gestação

Para a espécie eqüina até o presente momento, não está totalmente definida a composição bioquímica do líquido amniótico no decorrer da gestação. O presente trabalho teve como objetivo verificar a composição bioquímica do liquido amniótico e alantoideano das éguas em diferentes fases da gestação. Para isso analisou-se 60 amostras de fluidos fetais, empregando-se kits comerciais para se determinar a concentração bioquímica da Fosfatase Alcalina, Glicose, Proteínas Totais, Uréia, Creatinina, Cálcio, Cloreto, Sódio e Potássio durante a gestação. A concentração da alfa-fetoproteína foi avaliada empregando-se a eletroforese em gel de poliacrilamida. A concentração da Fosfatase Alcalina no liquido amniótico foi maior quando comparada ao liquido alantoideano nas três fases da gestação (p < 0,05). Para a glicose o valor médio entre os dois fluidos não apresentou variações (p < 0,05). Para a Proteína total o valor médio do liquido amniótico foi maior que o alantoideano (p < 0,05). A Uréia sofreu variações na concentração entre as fases, mas não há diferenças dos valores médios (p > 0,05) entre os fluidos. Para a Creatinina os valores presentes no liquido alantoideano são mais altos que os valores do liquido amniótico (p < 0,05). As concentrações dos íons Cloreto e Sódio apresentaram–se mais elevados (p < 0,05) no liquido amniótico. As concentrações dos íons Cálcio e Potássio foram mais elevadas nos líquidos alantoideanos (p<0,05). A eletroforese identificou duas bandas protéicas que podem ser a alfa-fetoproteína, ela parece aumentar a concentração durante o período gestacional. Porém, faltam estudos na espécie eqüina para a comparação dos resultados do presente trabalho. / Fetal fluids play a vital role in the development of the fetus. The biochemical composition of the amniotic fluid along pregnancy in horses had not been described until this present study. Sixty samples of fetal fluids were collected and the concentrations of Alkaline Phosphatase (FA), Glucose, Total Proteins (PT), Urea, Creatinin, Calcium (Ca), Chloride (Cl), Sodium (Na) and Potassium (K) along pregnancy were determined using commercially available kits. The levels of alpha-fetoprotein (AFT) were measured by polyacrylamide gel electrophoresis. During the three stages of pregnancy the concentrations of FA in the amniotic fluid were higher than those determined in the allantoic fluid (p < 0,05). The glucose levels did not differ between the fluids (p < 0,05). The mean values for the concentrations of PT were higher in the amniotic fluid than in the allantoic (p < 0,05). The urea levels differ among the pregnancy stages, but there were no differences in the mean values of urea (p > 0,05) between the two fluids. The concentrations of creatinin obtained in the allantoic fluid were higher than those obtained in the amniotic fluid (p < 0,05). The concentrations of Cl and Na were elevated (p < 0,05) in the amniotic fluid. The levels of the ions Ca e K were higher in the allantoic fluid (p<0,05). Polyacrylamide gel electrophoresis identified two protein bands that could be alpha-fetoprotein, which appears to have its concentration increased during pregnancy. There is a need for more studies in the biochemical composition of fetal fluids in horses to compare the results obtained in this study.

Com o intuito de conservação deste recurso genético e visando a seleção e o melhoramento genético destas raças diversos estudos estão sendo realizados com estes animais. Neste sentido, objetivou-se com este Experimento avaliar as principais características do ciclo estral das éguas da raça Campeiro e Pantaneiro nas diferentes estações do ano. Foram utilizadas seis éguas de cada raça (n=6). Um ciclo estral de cada égua foi acompanhado por palpação retal e ultrassonografia em cada uma das diferentes estações do ano. As raças foram separadas em capítulos. O capítulo 2 mostra que para as éguas da raça Pantaneiro, houve diferença na duração do ciclo estral. Os meses de Outono e Inverno apresentaram o ciclo estral mais prolongado quando comparados aos meses de Primavera e Verão. Sobre dia de divergência e tamanho folicular, dia de dominância e tamanho folicular, tamanho do folículo pré ovulatório e tamanho da estrutura lútea nas éguas Pantaneiras não houve diferença entre as estações do ano. A maioria das éguas Pantaneiras se mantiveram cíclicas durante as estações do ano no Outono (06/06), Inverno (04/06), Primavera (05/06) e Verão (06/06). O capítulo 3 avalia a raça Campeiro, a duração do ciclo estral das éguas foi menor nos meses de Outono e Verão quando comparado com os meses de Inverno e Primavera (P ≤ 0,05). O dia da divergência folicular para raça Campeiro foi mais tardio no Inverno e Primavera do que no Outono e Verão (P ≤ 0,05). E o dia da dominância foi mais tardio no Outono e Primavera que no Inverno e Verão (P ≤ 0,05). Para os demais parâmetros avaliados não houve diferença (P > 0,05). A maior quantidade de éguas Campeiras cíclicas foi no Outono (06/06), seguido pela Primavera e Verão (05/06) e o Inverno mostrou a menor quantidade de éguas em atividade reprodutiva (04/06). As éguas da raça Pantaneiro e Campeiro apresentam características do ciclo estral similares a de éguas de outros raças. Possuem ondas foliculares maiores e primárias e mostraram pouca influência sazonal já que a maioria dos animais, de ambas as raças, se mantiveram cíclicos em todas as estações do ano. ____________________________________________________________________________ ABSTRACT / The Campeiro and Pantaneiro horse are locally adapted breeds that have important adaptive traits such as rusticity, long walks resistance and traction capacity. With the conservation purpose of this genetic resource and in order to select and the genetic improvement of these breeds several studies are being conducted with these animals. In this sense, the objective was to evaluate with this experiment the main features of the estrous cycle of mares and Campeiro Pantaneiro race in different seasons. Six were selected mares of each race (n = 6). A estrous cycle of each mare was accompanied by rectal palpation and ultrasonography in each of the different seasons. The races were separated into chapters. Chapter 2 shows that for mares Pantaneiro breed was no difference in the estrous cycle. The months of autumn and winter have submitted the most protracted estrous cycle when compared to the months of spring and summer. About the day of divergence and follicular size, day of dominance and follicular size, pre ovulatory follicle size and size of luteal structure in mares not was no difference between the seasons. Most Pantaneiro mares remained cyclical during the seasons in autumn (06/06), winter (04/06), Spring (05/06) and summer (06/06). Chapter 3 evaluates the Campeiro breed, the length of the estrous cycle of the mares was lower in the months of autumn and summer compared to the winter months and spring (P ≤ 0.05). The day of follicle deviation for Campeiro was slower in winter and spring than in autumn and summer (P ≤ 0.05). And the day of dominance was later in the autumn and spring than in winter and summer (P ≤ 0.05). For other parameters there was no difference (P > 0.05). The largest amount of cyclic mares was foraging in autumn (06/06), followed by the spring and summer (05/06) and Winter showed the least amount of reproductive activity in mares (04/06). The mares of the Pantanal and Campeiro race have similar estrous cycle characteristics of the other breeds mares. Have larger and primary follicular waves and showed little seasonal influence since the majority of the animals of both breeds, remained cyclical in all seasons.

Comparação entre doses constantes e decrescentes de extrato de pituitária eqüina na indução de superovulacão em éguas

O extrato de pituitária eqüina (EPE) é um preparado parcial de gonadotrofina eqüina, e o único composto que consistentemente induz ovulações múltiplas em éguas (Squires et al, 1999); contudo, a resposta superovulatória tem sido baixa (1-3 ovulações/égua) (Mc Cue, 1996). Estudos mais recentes demonstram uma melhora no percentual de ovulações múltiplas em éguas superovuladas com a administração do EPE duas vezes ao dia (4-7ovulações/égua), entretanto com baixa taxa de recuperação embrionária (Scoggin et al., 2002; Alvarenga et al., 2001). Este fato pode estar relacionado a produção de oócitos de baixa qualidade (anormalidade na maturação folicular e oocitária) (Dippert et al., 1994; Palmer et al., 1993), a falha do folículo em liberar o oócito e a dificuldade na captação do mesmo para o interior do oviduto, assim como o trânsito deste ou do embrião pelo oviduto (Dippert et al., 1994). O extrato de pituitária eqüina (EPE) foi preparado no laboratório do Departamento de Reprodução Animal e Radiologia Veterinária da Universidade Estadual Paulista (UNESP), campus de Botucatu - SP, através do método proposto por Guillon & Combarnous, (1983). Seis éguas em bom estado nutricional, com idade variando entre quatro e 15 anos e massa corpórea de 400 a 500 Kg, além de um bom histórico reprodutivo, foram monitoradas diariamente durante os períodos compreendidos entre os meses de novembro (2001) a março (2002). Foram utilizados dois ciclos estrais de cada égua subdivididos em dois grupos: doses constantes e doses decrescentes; porém entre os tratamentos esperou-se um intervalo de dois ciclos estrais. O experimento foi conduzido no posto de monta... / Several studies have reported the is unresponsive to all the routinely used drugs aiming the superovulation in other species, such as the FSH and the eCG. The equine pituitary extract (EPE) is a partial equine gonadotropin preparation, and the only compound which consintenty induces multiple ovulation in mares; however, the superovulatory response has been low (1-3 ovulations/mare). More recent studies demonstrate some improvement in the percentage of multiple ovulation in superovulated mares with the EPE administration twice a day (4-7 ovulations/mare), nevertheless with a low rate of embryo recovery. This fact can be related to the low quality oocyte production (abnormality in the follicular and oocytary maturation), the follicular failure in releasing the oocyte and its collection towards the inner part of the oviduct, as its or the embryo traffic through the oviduct. The improvement on embryo recovery rate has been shownin cows with the utilization of decreasing doses of FSH. The present experiment aimmed to compare the ovulatory answer and embryo production of mares treated twice daily with constant and decreasing doses of EPE. Six mares in good nutrition status, with ages ranging from four and 15 years old and weighing from 400 to 500 Kg, together with a good reproductive history, were monitored daily through the preiods between the months of november/2001 to march/2002. Two estrous cytrous cycles were used from each mare and subdivided into two groups: constant doses and decresing doses;however between the treatments na interval of two estrous cycles was given. Resposta ovariana e taxa de recuperação embrionária em éguas tratadas com FSH suíno

O crescimento folicular foi acompanhado diariamente e os tratamentos (i.m. a cada 12h) foram mantidos até que o maior(es) folículo(s) atingiu 32mm, a indução da ovulação (2500 UI de hCG i.v.) foi realizada quando o maior(es) folículo(s) atingiu 35mm e a colheita de embriões no D8. No experimento 1, a dose de 25mg de FSHp foi testada em 28 éguas para determinação do melhor momento após aspiração folicular: 13mm (n=7, salina, controle), 13mm (n=7, FSHp-F13) e 20mm (n=7, FSHp-F20); ou em um dia determinado do ciclo: D6 (n=7, FSHp- D6). No experimento 2, a dose de 50mg de FSHp foi testada em 17 éguas quando o maior folículo atingiu 13mm (n=7, FSHp-13) e comparada ao grupo controle (n=10). No experimento 3, em 26 éguas BH inciou-se os seguintes tratamentos no D6: salina (n=7, controle), 25mg de FSHp (n=7, 25FSHp-D6), 12,5mg de EPE (n=7, EPE-D6) e 50 mg de FSHp (n=5, 50FSHp-D6). Usou-se a análise de variância de perfil, seguida do método de Tukey e para os dados de embriões/ovulação foi realizado o teste exato de Fisher (5% de significância). Os tratamentos com 25mg ou 50mg não promoveram aumento significativo no número de ovulações e de embriões recuperados, mas promoveu aumento no diâmetro máximo atingido por F3. O início do tratamento quando o maior folículo da onda induzida atingiu 20mm reduziu o tempo de tratamento. Conclui-se que os tratamentos com FSHp não promoveram resposta satisfatória e que o início do tratamento próximo ao desvio permitiu redução no tempo de tratamento. / The present study aims to verify the ovulatory response and embrionary recover in mares treated at different moments with pFSH (Folltropin-V®), with different doses and in BH breeding mares. The follicular growth was daily accompanied and treatments (i.m. BID) were kept until major follicles reached 32mm, ovulation induction (2500 UI of hCH i.v.) was done when major follicles reached 35mm and embryo collection at D8. In experiment 1, 25mg of pFSH was tested in 28 mares for determination of the best moment for start of treatment after follicle ablation: 13mm (n=7, saline, control), 13mm (n=7, FSHp-F13) e 20mm (n=7, FSHp-F20); or in a previously determined cycle day: D6 (n=7, FSHp-D6). In experiment 2, 50mg of pFSH was tested in 17 mares when the largest follicle achieved 13mm (n=7, FSHp-13) and compared to a control grupo (n=10). In experiment 3, in 26 BH mares was initiated the following treatments beginning at D6: saline (n=7, control), 25mg of pFSH (n=7, 25FSHp-D6), 12,5mg of EPE (n=7, EPE-D6) e 50 mg of pFSH (n=5, 50FSHp-D6). For statistical analysis, Profile analysis followed by Tuckey method and, for embryo/ovulation, Fisher test were used (significance at 5%). Treatments with 25mg or 50mg did not promote a significative increase on ovulations and embryo recover, but increased maximum diameter of F3. The start of treatment when the largest follicle of an induced follicular wave achieved 20mm reduced time of treatment. It is concluded that pFSH treatment did not promote a satisfactory response and that beginning of treatment close to deviation allowed the reduction of treatment time.

Estudo de diferentes sistemas e curvas de congelação na eficiência da congelabilidade e fertilidade de sêmen equino

Comparou-se a eficácia do período de estabilização do sêmen em diferentes sistemas de transporte refrigerado (Botutainer® e Equitainer®) em relação à estabilização feita em refrigerador Minitüb® previamente à congelação. No experimento I e II foram utilizados 4 garanhões com sêmen de alta congelabilidade. As amostras foram refrigeradas durante 20 minutos previamente à congelação em refrigerador Minitüb® (GM), Botutainer® (GB) e Equitainer® (GE). Não houve diferença entre os grupos quanto a motilidade total (MT) e motilidade progressiva (MP): GM (67,1;32,1%); GB (65,2;31,9%); GE (63,4;30,4%) demonstrando que os dispositivos podem ser utilizados como uma alternativa aos refrigeradores. Os valores de integridade de membrana plasmática (IMP%) foram GM(49,43a); GB(48,70a); GE(44,90b). No experimento II foram utilizadas 3 curvas programadas na TK3000® (C1, C2, C3) e a metodologia em caixa de isopor (GC). As comparações foram realizadas em relação ao GC e não apresentaram diferença significativa. Valores em porcentagem de MT, MP e IMP: GCxC1 (70 x 67,5; 35,7 x 32; 48,3 x 46); GCxC2 (63,3 x 67,66; 33,3 x 34,8; 45,4 x 48,3); GCxC3 (63,2 x 65; 32,2 x 32,4; 48,09 x 49,3). No teste de fertilidade, as inseminações foram realizadas com amostras congeladas com C2 e GC apresentando índices de prenhez de 65 e 60% respectivamente, sem diferença significativa. A TK3000® foi tão eficaz quanto à metodologia de congelação em caixa de isopor em relação à congelabilidade e fertilidade. Todas as curvas testadas foram eficientes na manutenção da viabilidade... / This present study compared freezing ratios in a programmable freezer (TK3000) performed by usual methodology, using good freezers and poor freezers stallions. The semen stabilization period efficiency was compared in cooled semen transport systems (Botutainer® e Equitainer®) in relation with Minitüb® fridge before the cryopreservation. Four good freezers stallions was used in experiments I and II. Samples were cooled during 20 minutes before the cryopreservation in a Minitüb® fridge (GM), Botutainer® (GB) and Equitainer® (GE). There was not difference between groups in relation with total motility (TM) and progressive motility (PM): CM (67,1;32,1%); GB (65,2;31,9%); GE (63,4;30,4%) showing that transport systems can be used as an alternative instead of fridges. Plasma membrane integrity (MPI%) values were CM (49,43a); GB (48,70a); GE (44,90b). Three frozen rations were used in experiment II in the TK3000® (C1, C2, C3) and the isothermic box (CG). Results from each freezing ratio were compared to the control group and no difference was observed. Percentage values from TM, PM and MPI were the following: CGxC1(70x67,5; 35,7x32; 48,3x46); CGxC2 (63,3x67,66; 33,3x34,8; 45,4x48,3); CGxC3 (63,2x65; 32,2x32,4; 48,09x49,3). In the fertility test, artificial inseminations were performed with C2 and GC frozen samples showing pregnancy rates of 65 and 60%, respectively, without significant difference. The TK3000® was as efficient as the isothermic box frozen methodology in relation to freezability and fertility. All freezing ratios were efficient in semen viability maintenance. Ten Mangalarga Marchador breed stallions were used in the experiment III. O objetivo deste estudo foi avaliar as concentrações plasmáticas de estradiol, inibina, FSH e LH em éguas superovuladas com eFSH e EPE (em doses constantes ou decrescentes): antes, durante e imediatamente após o término da administração hormonal. Foram utilizadas seis éguas as quais foram submetidas aos seguintes tratamentos: Grupo I (GI) recebeu 25mg de EPE intramuscular (I.M.), Grupo II (GII) foram tratadas com EPE (I.M.) em doses decrescentes. No Grupo IIl (GIII), o EPE foi substituído por 12,5mg de eFSH (I.M.). O Grupo IV (GIV) foi utilizado como controle. Sangue foi coletado diariamente para avaliar as concentrações hormonais por radioimunoensaio. As coletas foram iniciadas dois dias antes da primeira aplicação medicamentosa, encerrando 48horas após a última ovulação do estro. As diferenças nos níveis plasmáticos hormonais foram comparadas utilizando-se o ANOVA, sendo considerada diferença estatística quando P0,05. O número médio de ovulações foi de 3,3; 4,8; 5,0 e 1,0 nos grupos 1 a 4, respectivamente, tendo sido maior (P<0,05) em todos os grupos superovulados em relação ao controle. As concentrações de estrógeno, inibina, FSH e LH não diferiram entre os grupos durante o período 1. No período 2, os valores de estradiol foram maiores (P<0,05) em éguas sob tratamento superovulatório com eFSH (GIII) 29,52pg/ml, os grupos I e II, apresentaram valores intermediários (GI: 16,75pg/ml, GII: 16,26pg/ml) e o GIV (7,78pg/ml) menor estatisticamente aos demais. Foi verificada uma correlação altamente positiva (R=0,615; P=0,01) entre os valores de estradiol e inibina, os quais, durante os tratamentos, foram maiores (P<0,05)... / Satisfactory superovulatory responses have been reported in mares with the use of equine pituitary extract (EPE) and purified equine FSH (eFSH), however embryo recovery rates are still inconsistent. Recent studies demonstrated that the superovulatory treatment disturbs oocyte maturation and transport. The main objective of the present study was to evaluate the plasmatic concentrations of estradiol, inhibin, FSH and LH in super ovulated mares treated with eFSH and EPE (with constant or decreasing doses): Six mares were used in a crossover experiment in four different groups: Group I mares received 25mg of EPE every twelve hours, Group II received EPE in decreasing doses and Group IlI mares received 12.5mg of eFSH (IM). Group IV served as control. Embryo flushes were performed 8 days pos ovulation. Blood was collected daily to evaluate hormonal concentrations through radioimuneassay, starting two days before the first administration, until 48 hours post-ovulation. Mean and daily hormonal concentrations of estradiol, inhibin, FSH and LH were evaluated. Hormonal concentrations were submitted to statistical analysis using ANOVA. Significance levels were set as P<0.05. Mean number of ovulations was 3.3; 4.8; 5.0 and 1.0 in groups 1 to 4, respectively, being higher (P<0.05) in all super ovulated groups, when compared to control. Concentrations of estrogen, inhibin, FSH and LH did not differ between groups during the first period. In period 2, estradiol levels were higher (P<0.05) in mares with superovulatory treatment eFSH (GIII) 29.52pg/ml, the groups GI and GII (GI: 16.75pg/ml, GII: 16.26pg/ml) values intermediary, and GIV (7.78pg/ml) levels lower. A positive correlation between estradiol levels and number of follicles ≥30mm (R=0.917; P<0.05), recovered embryos (R=0.945; P<0.05) and ovulations (R=0.75; P<0.05) was identified in super ovulated mares. A positive correlation (R=0.615; P=0.01) between estradiol and inhibin concentrations.

Criopreservação e fertilidade de espermatozóides recuperados da cauda do epidídimo de garanhões

Sendo assim, o objetivo do experimento I foi comparar os parâmetros espermáticos dos espermatozóides da cauda do epidídimo recuperados imediatamente após orquiectomia e em diferentes momentos após armazenamento a 5°C e a temperatura ambiente. Já o experimento II, teve o objetivo de comparar os parâmetros espermáticos e fertilidade dos espermatozóides do ejaculado e do epidídimo recuperados logo depois da orquiectomia e após armazenamento por 24 horas a 5°C. No experimento I, 48 garanhões foram submetidos à orquiectomia bilateral, sendo que em oito a colheita dos espermatozóides do epidídimo foi realizada imediatamente após orquiectomia (grupo controle). Nos outros 40 garanhões, um epidídimo foi armazenado a temperatura ambiente e o contralateral armazenado a 5°C por 6, 12, 18, 24 e 30 horas, de acordo com o grupo. No experimento II, dois ejaculados de oito garanhões foram colhidos com vagina artificial e congelados (grupo EJ-0h). Uma semana após, os garanhões foram submetidos à orquiectomia bilateral, sendo que os espermatozóides de um epidídimo foram congelados imediatamente após orquiectomia (grupo EP-0h) e o epidídimo contralateral foi previamente armazenado por 24 horas a 5°C (EP-24h). O teste de fertilidade demonstrou que o grupo EP-0h (92,3%; n=13) tendem a ser maior que os grupos EJ-0h (61,5%; n=13) e EP-24h (61,5%; n=13). Conclui-se que o armazenamento dos testículos-epidídimos a 5°C proporcionou melhor preservação espermática e que, independente da temperatura, a motilidade progressiva é o parâmetro espermático mais sensível ao tempo de armazenamento. / Cauda epididymis sperm recovery and cryopreservation may be the last chance to obtain genetical material when sudden death or serious injury occur in valuable stallions. Thus, the aim of experiment I was to compare sperm parameters of epididymal sperm recovered immediately after orchiectomy and at different moments after storage at 5°C and at room temperature. Experiment II aimed to compare sperm parameters and fertility of ejaculated sperm and epididymal sperm recovered immediately after orchiectomy and after storage for 24h at 5°C. In experiment I, 48 stallions were submitted to bilateral orchiectomy, in eight of those the sperm collection were done immediately after orchiectomy (control group). In the other 40 stallions, one epididymis was stored at room temperature and the other was stored at 5°C for 6, 12, 18, 24 and 30 hours accorded to groups. In experiment II, two ejaculated from eight stallions were obtained with artificial vagina and cryopreserved (group EJ-0h). One week later, the stallions were submitted to bilateral orchiectomy, and the sperm of one epididymal were frozen immediately after orchiectomy (group EP-0h) and the contralateral epididymal was previously storage for 24h at 5°C (EP-24h). Fertility test showed that group EP-0h (92,3%; n=13) tended to be higher than groups EJ-0h (61,5%; n=13) and EP-24h (61,5%; n=13). These results allowed concluding that storage of the testis-epididymis complex at 5°C is more efficient in preserving sperm parameters than room temperature storage and that progressive motility is the parameter that is more affected by storage period, regardless of the temperature.

Efeito do tratamento com extrato de pituitaria equina (EPE) e hCG no índice de recuperação de oócitos e maturação folicular em equinos /

Orientador: Fernanda da Cruz Landim e Alvarenga / Banca: Cezinande de Meira / Banca: Marcelo Marcondes Seneda / Resumo: Em éguas, o aprimoramento e a aplicação de técnicas como a ICSI, TO e transferência nuclear é restrita devido à dificuldade de obtenção de oócitos in vivo, atribuída a particularidades dos folículos ovarianos da espécie. Em folículos pré-ovulatórios, a expansão do cumulus que ocorre com a progressão da maturação folicular e deposição de ácido hialurônico aumenta os índices de recuperação de oócitos por folículo, enquanto o tratamento superovulatório, embora não seja eficiente em aumentar os índices de recuperação por folículo, melhora os índices de recuperação por égua. Este trabalho teve como objetivo avaliar o efeito da administração de hCG na expansão das células da granulosa e do cumulus e no índice de recuperação de oócitos a partir de folículos imaturos de éguas superovuladas. Para isso 10 éguas foram submetidas à OPU após três diferentes tratamentos hormonais (G1: EPE, G2: EPE/hCG e G3:controle). Cada égua foi submetida aos três tratamentos, tendo sido alternados um ciclo onde era permitido que houvesse ovulação, sem interferência, e um ciclo com OPU. No ciclo onde devia ocorrer aspiração, as éguas recebiam aplicações de prostaglandina no sétimo e oitavo dia após a ovulação. Para os grupos 1 e 2, iniciava-se o tratamento com EPE no D7. Quando a maioria dos folículos atingia entre 22 e 27 mm, somente o grupo 1 recebia hCG, a EPE era suspensa e efetuava-se OPU 24 horas depois para ambos os grupos. Para o grupo controle, após a aplicação de prostaglandina a OPU era efetuada 24 horas após o folículo dominante atingir 27-30 mm. Em todos os grupos foram aspirados folículos entre 10 e 35 mm. Não houve diferença entre os 3 grupos quanto aos índices de recuperação de oócitos por folículo, com médias de 15% a 16,7%. Entretanto, a hCG foi capaz de induzir uma expansão e luteinização precoce das células foliculares. / Abstract: In mares, the use of artificial reproductive techniques (ART) such as intracitoplasmatic sperm injection (ICSI), oocyte transfer (OT), and cloning is limited by the low availability of oocytes obtained either in vitro or in vivo. The low recovery rate of oocytes from immature follicles seems to be linked with anatomical particularities of the follicular wall. In pre-ovulatory follicles (>35mm in diameter), final oocyte maturation is accompanied by the expansion of the cumulus, which releases the oocyte from the follicular wall resulting in increased recovery rates after follicular aspiration. The number of oocytes recovered in each OPU (Ovum Pick Up) section should be higher with superovulation. However, although the total number of oocytes recovered by each individual mare is increased, the recovery rate per follicle is still low. Treatment with hCG modifies the intra-follicular environment by inducing precocious expansion of the granulosa cells, leading to a higher recovery rate during OPU of pre-ovulatory follicles. Therefore, this study aimed to evaluate the effect of hCG treatment on the recovery rate of oocytes obtained from immature follicles in mares superovulated with Equine Pituitary Extract (EPE). Ten mares were rotated in three different treatments: G1 - superovulated with EPE; G2 - Superovulated with EPE and treated with hCG, and G3 - Control. For all treatments, when the biggest follicle reached 27 - 30 mm, all follicles bigger than 10 mm were aspirated. Each OPU cycle was alternated with a rest cycle when a spontaneous ovulation occurred. Although treatment with hCG increased the number of follicles presenting expanded granulosa and high intra-follicular progesterone concentration, no statistically significant differences were observed between groups regarding oocyte recovery. / Mestre
