Two groups (fungus-treated and control without fungus treatment) consisted of six crossbred mares in each group, aged between 2.5 and 3.5 years, were fed in Cynodon sp. naturally infected with larvae living stages of equine cyathostomin. Each animal of the treated group received oral doses of sodium alginate mycelial pellets 1 g/10 kg live weight per week), during 6 months. In the control group, animals received 1g/10 kg live weight per week of pellets without the fungus. Significant reduction (p< 0.01) in the number of eggs per gram of feces was found for animals of the fungus-treated group compared with the control group in all months of the experiment. The percentage of reduction of EPG were 87.5%, 89.7%, 68.3%, 58.7%, 52.5% and 35.2% during June, July, August, September, October and November, respectively. In coprocultures difference was found (p<0.05) among animals in the group treated with the fungus compared to the control group in all months of the experiment, with percentage reductions of 67.5%, 61.4% and 31.8% for September, October and November, respectively. There was a difference (p<0.01) of 60.9% reduction in herbage samples collected up to 20 cm from fecal pats between the fungus-treated group and the control group, during the experimental period (May October). Difference of 56% (p<0.01) was found between the fungus-treated group and the control group in the sampling distance 20-40 cm from fecal pats. There was no difference (p>0,05) between the mean weight gain of animals in both groups. The treatment with sodium alginate pellets containing the nematode- trapping fungus M. thaumasium reduced cyathostomin in tropical southeastern Brazil and could be an effective tool for biological control of this parasitic nematode in horses. / A viabilidade de uma formulação do fungo predador de nematóides Monacrosporium thaumasium foi avaliada no controle biológico de ciatostomíneos de eqüinos. Dois grupos (tratado com o fungo e controle) constituídos por seis éguas em cada grupo, mestiças, com idade entre 2,5 a 3,5 anos, foram colocados em pastagens de Cynodon sp. naturalmente infectadas com larvas de ciatostomíneos de eqüinos. No grupo tratado, cada animal recebeu 1g/10 kg de peso corporal (0,2g/10kg de peso vivo de fungo) de péletes em matriz de alginato de sódio contendo o fungo M. thaumasium via oral, duas vezes por semana e durante seis meses. No grupo controle, os animais receberam 1g/10 kg de peso corporal de péletes sem o fungo. As contagens de ovos por grama de fezes demonstraram diferença (p<0,01) no grupo de animais tratados com o fungo em relação aos animais do grupo controle em todos os meses do experimento. Os percentuais de redução do OPG foram de 87,5%, 89,7%, 68,3%, 58,7%, 52,5% e 35,2% nos meses de junho, julho, agosto, setembro, outubro e novembro, respectivamente. Nas coproculturas foi encontrada diferença (p<0,05) entre os animais do grupo tratado com o fungo em relação aos animais do grupo controle em todos os meses do experimento, com percentuais de redução de 67,5%, 61,4% e 31,8% para setembro, outubro e novembro, respectivamente. Diferença (p<0,01) foi observada na recuperação de larvas infectantes das pastagens que foram coletadas até 20 cm de distância do bolo fecal no pasto do grupo tratado com o fungo em relação ao grupo controle com uma redução de 60,9% e, entre 20 e 40 cm do bolo fecal a redução (p<0,01) foi de 56% no pasto do grupo tratado com o fungo M. thaumasium em relação ao pasto do grupo controle. Não foi observada diferença (P>0,05) entre as médias de ganho de peso dos animais dos dois grupos. O tratamento de eqüinos com péletes contendo o fungo nematófago M. thaumasium, pode ser efetivo no controle de ciatostomíneos em região tropical do sudeste do Brasil.

The use of chemicals in animal anthelmintic treatment, in association with nematophagous fungi used for biological control, is a strategy that has proven to be effective in reducing the nematode population density of farm animals. This study aims to verify the in vitro susceptibility of the nematophagous fungi Arthrobotrys oligospora, Duddingtonia flagrans, Paecilomyces fumosoroseus, Paecilomyces lilacinus, Paecilomyces marquandii and Paecilomyces variotii against the antiparasitic drugs albendazole, thiabendazole, ivermectin (100%), levamisole (7.5%) and closantel (10%) by using the Minimum Inhibitory Concentration (MIC). MICs ranged between 4 and 0,031µg/mL for albendazole, thiabendazole and ivermectin, between 0,937 and 0,117µg/mL for levamisole, and between 0,625 and 0,034 for closantel. The results obtained showed that all antiparasitic drugs tested had an in vitro inhibitory effect on nematophagous fungi, being able to jeopardize the fungus action as a biological control bioagent. / O rápido desenvolvimento de resistência de parasitos do trato gastrintestinal a antihelmínticos tem demonstrado limitada eficiência desse método para o controle de determinadas endoparasitoses em ruminantes, incentivado assim, pesquisas com métodos alternativos de controle parasitário. A utilização de compostos químicos no tratamento anti-helmíntico de animais, em associação com fungos nematófagos usados no controle biológico, é uma estratégia que vem se mostrando eficaz para a redução da densidade populacional de nematódeos nos animais de produção e pouco se sabe sobre seu emprego simultâneo. Este trabalho teve por objetivo verificar, através da Concentração Inibitória Mínima (CIM), a suscetibilidade in vitro dos fungos nematófagos Arthrobotrys oligospora, Duddingtonia flagrans, Paecilomyces fumosoroseus, Paecilomyces lilacinus, Paecilomyces marquandii e Paecilomyces variotii aos antiparasitários albendazol, tiabendazol e ivermectina (100%), levamisol (7,5%) e closantel (10%). As CIMs variaram de 4 a 0,031µg/mL para albendazol, tiabendazol e ivermectina, de 0,937 a 0,117µg/mL para o levamisol e de 0,625 a 0,039µg/mL para o closantel, dependendo do fungo testado. Os resultados mostram que todos os antiparasitários testados tiveram efeito inibitório in vitro sobre os fungos nematófagos, podendo comprometer suas ações como bioagentes de controle biológico.

Estudo do controle biológico dos nematóides dos citros no Estado de São Paulo /

No Brasil, apenas o nematóide dos citros (Tylenchulus semipenetrans Cobb) e o nematóide das lesões radiculares dos citros (Pratylenchus jaehni Inserra et al.) detêm esse status. Entre esses, T. semipenetrans é o mais distribuído, enquanto P. jaehni é o mais agressivo. Essa praga já foi encontrada em pomares de mais de 30 municípios paulistas, dois mineiros e três paranaenses. O objetivo deste estudo foi isolar, identificar, documentar ao microscópio fotônico e eletrônico de varredura e avaliar a eficácia de fungos nematófagos para o controle biológico dos nematóides dos citros (P. jaehni e T. semipenetrans). Foram analisadas 38 amostras coletadas em pomares de citros de municípios paulistas e goiano no Laboratório de Nematologia da UNESP/FCAV, Câmpus de Jaboticabal-SP. Dessas amostras, foram isolados Arthrobotrys robusta de 23,7% das amostras, A. musiformis de 18,4%, A. oligospora de 10,5% e A. conoides de 5,2%. Monacrosporium elegans, M. eudermatum, Dactyllela leptospora e Paecilomices lilacinus foram encontrados em 2,6% das amostras. Do total de 26 isolados dos fungos obtidos, 92,8% foram patogênicos a T. semipenetrans e 82,1% a P. jaehni. Um isolado de cada espécie desses fungos foi incluído na documentação das características mais marcantes ao microscópios fotônico e eletrônico de varredura. Em testes in vitro, foram selecionados isolados de cinco espécies de fungos nematófagos, sendo Monacrosporium eudermatum, Dactylella leptospora, Arthrobotrys musiformis, A. conoides e Paecilomyces lilacinus formulados para T. semipenetrans. In Brazil, only the citrus nematode (Tylenchulus semipenetrans Cobb) and the lesion nematode of citrus (Pratylenchus jaehni Inserra et al.) have that status. Contudo, foram estudados os fatores determinantes da freqüência de fungos totais e nematófagos, da atividade enzimática e da distribuição de nematóides de solos não rizosféricos e rizosféricos sob alface (Lactuca sativa L.); banana (Musa cavendishii L.); grama batatais (Paspalum notatum) e Impatiens (Impatiens valleriana). A atividade predadora dos fungos nematófagos foi avaliada contra o nematóide Panagrellus redivivus. O número de fungos totais variou de 6,9 x 105 a 31,2 x 105 UFC g-1 solo seco no solo não-rizosférico e de 6,9 x 105 a 25,6 x 105 UFC g-1 solo seco no solo rizosférico. As contagens dos fungos nematófagos corresponderam a 23-41 % e 23-34 % dos fungos totais encontrados no solo rizosférico e não-rizosférico, respectivamente. Enquanto as contagens dos fungos totais do solo rizosférico se distribuíram na seguinte seqüência: alface > banana > grama-batatais > Impatiens, os nematófagos diminuíram na seguinte ordem: alface > Impatiens > banana > grama-batatais. A matéria orgânica e a umidade do solo influenciaram a distribuição tanto dos fungos totais como dos nematófagos do solo não-rizosférico. We investigated the factors affecting the frequency of the total and the nematophagous fungi, soil enzymatic activity as well as the distribution of the nematodes in rhizospheric and bulk soil of lettuce (Lactuca sativa L.); Impatiens (Impatiens valleriana); banana (Musa cavendishii L.) and Bahiagrass (Paspalum otatum). The effect of the nematophagous fungi were evaluated against the nematode Panagrellus redivivus. The total fungal number ranged from 6,9 x 105 to 31,2 x 105 UFC g-1 dry soil in the bulk soil and from 6,9 x 105 to 25,6 x 105 UFC g-1 dry soil in the rhizosphere. Nematofagous fungi counts were to 23-41 % and 23-34 % of the total fungi found in the rhizosphere and bulk soil, respectively. While the total fungi counts of the rizosphere were distributed in the following sequence: lettuce > banana > Bahiagrass > Impatiens, the nematophagous fungi decreased in the following order: lettuce> Impatiens >banana > Bahiagrass. Moisture and organic matter contents of the bulk soil influenced the distribution of the total and nematophagous fungi of the soil. The aims of this study were to evaluate the ovicidal activity of the isolates VC1 and VC4 of the nematophagous fungus Pochonia chlamydosporia on solid medium containing 2% water-agar(2% WA) on eggs of E. paraensei, selecting the most efficient isolate (assay A), evaluate the ovicidal effect (destruction of eggs) of most effective isolate on supplemented agar culture media (assay B), and to evaluate the ovicidal capacity of crude enzymatic extract of most effective isolate on E. paraensei eggs (assay C). One thousand eggs of E. paraensei (assay A) were placed in Petri dishes containing 2% WA with one isolate of fungus P. chlamydosporia (VC1 or VC4) grown for 10 days, and without fungus as control. Both the treated and control group consisted of six replicates. After 15 days, approximately one hundred eggs were removed from each dish and classified according to the following parameters: type 1 effect, physiological effect without morphological damage to eggshell, where hyphae are observed attached to the shell; type 2 effect, lytic effect with morphological alteration of embryo and egg shell with hyphal penetration through the shell; and type 3 effect, lytic effect with morphological alteration of embryo and eggshell, in addition to hyphal penetration and internal egg colonization. In assay B, E. paraensei eggs were placed in Petri dishes with different supplemented agar culture media: 2% chitin- agar (2% AC), 2% corn-meal-agar (2% CMA), 2% WA and 2% starch- agar (2% SSA) with isolate VC4 (the most effective); in control group were not used fungus. Each treatment consisted of six replicates. After 25 days, one hundred eggs were removed from each dish and the ovicidal effect was evaluated according to the parameters already mentioned. In assay C, one thousand E. paraensei eggs were poured into Petri dishes with 5 ml of isolate VC4 crude extract constituting the treated group. The control group contained one thousand eggs in 10 ml of distilled water in Petri dishes. Each treatment consisted of six replicates. After 7 days, the total number of E. paraensei eggs present on each dish of the treated and control groups were counted according to the methodology described by Mukhtar and Pervaz. In assay A, there was no difference (p > 0.05) in ovicidal activity among the tested isolates; however, the highest percentage for ovicidal activity (type 3 effect) was demonstrated by the isolate VC4. In assay B, the culture medium starch agar showed the best results for the destruction of the eggs at the end of 25 days of interaction, with a percentage of 46.6%. In assay C, the crude extract of VC4 was effective in the destruction of E. paraensei eggs at the end of 7 days, with a percentage reduction of 53%. Results of in vitro experimental assay A, B and C have shown that P. chlamydosporia (VC1 and VC4) had negative influence on the E. paraensei eggs, and thus can be considered as a potential candidate for the biological control of this helminth. / Echinostoma paraensei é um trematóide do gênero Echinostoma que pode causar a equinostomíase em humanos. Os objetivos desse trabalho foram avaliar a atividade ovicida dos isolados VC1 e VC4 do fungo nematófago Pochonia chlamydosporia em meio sólido ágar- água 2% sobre ovos de Echinostoma paraensei, selecionando o isolado mais eficaz (ensaio A); avaliar o efeito ovicida (destruição dos ovos) do isolado mais eficaz em meios de cultura ágar suplementados (ensaio B) e avaliar a capacidade ovicida do extrato bruto enzimático do melhor isolado sobre ovos de E. paraensei (ensaio C). Mil ovos de E. paraensei (ensaio A) foram colocados em placas de Petri contendo ágar-água 2% (AA 2%) com um isolado do fungo P. chlamydosporia crescido (VC1 ou VC4) durante 10 dias, e sem fungo como controle. Seis repetições foram feitas para ambos o grupo tratado e controle. Após 15 dias, cerca de cem ovos foram retirados de cada placa e classificados de acordo com os seguintes parâmetros: tipo 1, efeito fisiológico sem prejuízo morfológico à casca do ovo, onde hifas são observadas aderidas à casca; tipo 2, efeito lítico com alteração morfológica da casca e embrião do ovo, com penetração de hifas através da casca; e tipo 3 efeito lítico com alteração morfológica do embrião e da casca, além de penetração de hifas e colonização interna do ovo. No ensaio B, ovos de E. paraensei foram colocados em placas de Petri com diferentes meios de cultura ágar suplementados: ágar-quitina 2% (AQ 2%), corn-meal-ágar 2% (CMA 2%), ágar-água 2% (AA 2%) e ágar- amido 2% (SSA 2%) com o isolado VC4 (o mais eficaz), no grupo controle não foram utilizados fungos. Seis repetições foram feitas para cada tratamento. Depois de 25 dias, cem ovos foram retirados de cada placa e o efeito ovicida foi avaliado de acordo com os parâmetros já citados. No ensaio C, mil ovos de E. paraensei foram vertidos para placas de Petri com 5ml de extrato bruto do isolado VC4 constituindo o grupo tratado. O grupo controle tinha 1.000 ovos em 10 ml de água destilada nas placas de Petri. Cada tratamento foi constituído por seis repetições. Após 7 dias, foram contados o número total de ovos de E. paraensei presentes em cada placa dos grupos tratado e controle de acordo com a metodologia descrita por Mukhtar e Pervaz. No ensaio A, foi observado que não houve diferença (p>0,05) na atividade ovicida entre os isolados testados, contudo, o maior percentual para a atividade ovicida (efeito do tipo 3) foi demonstrado pelo isolado VC4. No ensaio B, o meio de cultura ágar- amido 2% (SSA 2%) apresentou os melhores resultados para a destruição dos ovos ao final de 25 dias de interação, com um percentual de 46.6%. No ensaio C, o extrato bruto de VC4 demonstrou ser eficaz na destruição dos ovos de E. paraensei ao final de 7 dias com um percentual de redução de 53%. Os resultados dos ensaios experimentais in vitro A, B e C demonstraram que P. chlamydosporia (VC1 e VC4) influenciou de forma negativa sobre os ovos de E. paraensei, e assim pode ser considerado como um potencial candidato a controlador biológico desse helminto.

O trabalho objetivou isolar e caracterizar fungos com potencial nematofágico a partir de amostras de solo, provenientes de municípios do Rio Grande do Sul Os isolados foram testados quanto a eficiência nematofágica diante do nematóide Panagrellus redivivus sendo eleito um dos fungos para a realização de ensaios in vitro e in vivo. Os ensaios consistiram em avaliar a atividade do isolado sobre larvas infectantes em condições ideias (estufa) e condições adversas (campo), a partir de administrações orais do fungo em substrato milho, em três doses distintas, em ovinos naturalmente infectados com nematóides gastrintestinais. Também foram avaliadas as taxas de integridade física e a viabilidade do referente fungo após passagem pelo trato digestório desses animais. Os resultados demonstraram a existência de fungos com potencial nematofágico no estado. Foi isolado o fungo nematófago Duddingtonia flagrans, considerado um dos mais eficientes pela grande produção de clamidósporos, o qual foi utilizado nos testes. D. flagrans comprovou integridade e viabilidade após atravessar o trato gastrintestinal dos ovinos e os resultados tanto em ambiente controlado como adverso, demonstraram uma diminuição no número de larvas em relação ao grupo controle. Diante do exposto, o Rio Grande do Sul possui fungos com poder nematofágico, e D. flagrans quando colocado em desafio, atestou sua eficácia nematicida sendo, portanto, passível sua utilização como controlador biológico de larvas gastrintestinais de ovinos. / The biocontrol with nematophagous fungi has been a promising alternative for use against gastrointestinal nematodiosis of sheep, due to the adversities that suppressive use of chemical molecules has occasioned. This study aimed to isolate and characterize fungi with nematophagic potential from soil samples from the municipalities of Rio Grande do Sul. The isolates were tested for nematophagic efficiency against the nematode Panagrellus redivivus being one of the fungi elected for in vitro and in vivo testing. The tests consisted of evaluation of the activity of the isolated on infective larvae in ideal conditions (bacteriological oven) and adverse conditions (field) from oral administrations of the fungus in corn substrate at three different doses in sheep naturally infected with gastrointestinal nematodes. Rates of physical integrity and viability of the related fungus were also evaluated after passage through the digestive tract of these animals. The results demonstrated the existence of fungi with nematophagic potential in the state. There has been the isolation of Duddingtonia flagrans nematophagous fungi, considered one of the most efficient due to its large-scale production of chlamydospores, which was used in the tests was isolated. D. flagrans demonstrated integrity and viability after crossing the gastrointestinal tract of sheep and the results in both environment such as controlled and adverse , demonstrated a decrease in the number of larvae when compared to the control group. Given the above, Rio Grande do Sul has fungi with nematophagic potential and D. flagrans when challenged, attested its nematocide efficacy being therefore likely its usage as a biological controller of gastrointestinal sheep larvae.

Os objetivos deste estudo foram avaliar o efeito do subproduto sólido da indústria vinícola (SSIV) sobre o desenvolvimento, in vitro, de Pochonia chlamydosporia e o efeito da associação do SSIV a esse fungo no manejo de Meloidogyne javanica em tomateiros, em casa de vegetação. Para avaliar o efeito direto do SSIV sobre o desenvolvimento do fungo, adicionaram-se as concentrações (0; 5; 10; e 15%) do extrato aquoso do SSIV em meio de cultivo batata-dextrose-ágar (BDA) fundente. Em seguida, a mistura foi vertida em placas de Petri e um disco de meio de cultivo ‘Corn Meal Ágar’ (CMA) colonizado pelo fungo foi colocado no centro de cada placa. Em outro estudo avaliou-se o efeito dos compostos voláteis do SSIV sobre o desenvolvimento de P. chlamydosporia. Para isto, utilizaram-se placas de Petri bipartidas, adicionando em um dos compartimentos as concentrações do SSIV (0,000; 0,065; 0,125; 0,250; e 0,500 g placa-1) e 2 mL de água destilada esterilizada placa-1. No centro do outro compartimento contendo meio de cultivo BDA, adicionou-se um disco de meio de cultivo CMA colonizado pelo fungo. As placas dos dois estudos foram armazenadas em câmara de crescimento, no escuro, a 21°C. Os dois estudos foram realizados duas vezes. Após 14 dias avaliaram-se o diâmetro das colônias e a produção de conídios do fungo. O SSIV reduziu de 9 a 21% o crescimento do fungo nos dois estudos, bem como reduziu a produção de conídios no primeiro estudo. Em casa de vegetação, avaliou-se a associação do SSIV (30 g kg-1 de substrato) às concentrações de P. chlamydosporia com clamidósporos (500; 1.500; 2.500; 3.500; 4.500; e 5.000 clamidósporos g-1 de substrato) ou sem clamidósporos (2,5; 5; 10; e 15 g de inóculo do fungo kg-1 de substrato). Três tratamentos foram utilizados como controle em cada ensaio: adição de P. chlamydosporia (5.000 clamidósporos g-1 de substrato ou 15 g de inóculo do fungo kg-1 de substrato), adição de SSIV (30 g do SSIV kg-1 de substrato) e apenas o substrato. Em seguida, cada saco contendo 4 kg de substrato esterilizado e os respectivos tratamentos foi infestado com 6.000 ovos de M. javanica kg-1 de substrato, homogeneizado e umedecido até 60% de capacidade de campo e armazenado por 14 dias no escuro, a 25 ºC. Depois, o substrato foi transferido para vasos de polipropileno de 500 mL, e uma muda de tomateiro foi transplantada para cada vaso. Após 60 dias foram avaliados a altura, a massa da parte aérea e das raízes frescas e o número de galhas e de ovos do nematoide. A associação do SSIV ao fungo, independentemente do tipo de inóculo do fungo, potencializou o desenvolvimento dos tomateiros e o controle do nematoide. As concentrações mais indicadas do fungo para se associar ao SSIV foram 3.500 clamidósporos g-1 de substrato e 15 g do inóculo sem clamidósporos kg-1 de substrato, pois, além de controlarem efetivamente o nematoide, também proporcionaram o desenvolvimento dos tomateiros. Conclui-se que a associação do SSIV ao fungo P. chlamydosporia potencializa o manejo do nematoide-de-galhas. / The biological control and the addition of organic matter to the soil represent the main alternatives for the control of root-knot nematode. The objectives of this study were to evaluate the effect of solid wine industry by-product (SSIV) on the development, in vitro, of Pochonia chlamydosporia and the effect of combining this fungus with the SSIV on the management of Meloidogyne javanica in tomatoes grown in a greenhouse. To evaluate the direct effect of the SSIV on fungal development, concentrations (0, 5, 10, and 15%) of the aqueous SSIV extract were applied to potato-dextrose-agar (BDA) culture medium. The mixture was then poured into Petri dishes and a disk of 'Corn Meal Agar' (CMA) culture medium colonized by the fungus was placed in the centre of each plate. A further study evaluated the effect of volatile SSIV compounds on the development of P. chlamydosporia. For this, twopart Petri dishes were utilised, to one of the compartments the concentrations of SSIV (0.000, 0.065, 0.125, 0.250, and 0.500 g plate-1) and 2 mL plate-1 sterile distilled water were added. To the other compartment, which contained BDA culture medium, a disc of CMA culture medium colonised by the fungus was added in the centre. The plates from the two studies were stored in a growth chamber in the dark at 21 °C. The two studies were performed twice. After 14 days, the diameter of the colonies and the production of fungal conidia were evaluated. The SSIV reduced fungal growth by 9 to 21% in the two studies, and reduced conidia production in the first study. In the greenhouse, the combination of SSIV (30 g kg-1 of substrate) with the concentrations of P. chlamydosporia with chlamydospores (500; 1,500; 2,500; 3,500; 4,500; and 5,000 chlamydospores g-1 of substrate) or without chlamydospores (2.5, 5, 10, and 15 g of inoculum of fungus kg-1 substrate) were evaluated. Three treatments were used as controls in each assay: the addition of P. chlamydosporia (5,000 chlamydospores g-1 substrate or 15 g of inoculum of fungus kg-1 of substrate), the addition of SSIV (30 g SSIV kg-1 of substrate) and only substrate. After 60 days the height, the shoot and fresh root masses, and the numbers of nematode galls and eggs were evaluated. The combination of SSIV and the fungus, independent of the type of fungal inoculum, increased the development of the tomato plants and the control of the nematodes. The fungal concentrations most recommended to combine with the SSIV were 3,500 chlamydospores g-1 of substrate and 15 g of inoculum without chlamydospores kg-1 of substrate, because, besides controlling effectively the nematode, they also improved the development of the tomato plants. It was concluded that the combination of SSIV and the fungus P. chlamydosporia improves the management of this root-knot nematode.
