Coordenação de Aperfeiçoamento de Pessoal de Nível Superior / Asteraceae has attracted many embryologists because of its high variability of the embryological features. Nevertheless, nothing is known about the embryology of the species basal subfamily, Barnadesioideae. Thus, this paper aims to describe the processes of ginosporogenesis and ginogametogenesis in two species of Barnadesioideae, Sclechtendalia luzulaefolia, considered basal and sister to the other species that belong to family and Dasyphyllum brasiliense whose genus was also sometimes considered basal. Both species are native to Rio Grande do Sul. S. luzulaefolia was collected in São Pedro do Sul, RS, Brazil and D. brasiliense in Santa Maria, RS, Brazil and in São Pedro do Sul, RS, Brazil. The corolla is the first floral whorl to arise in both species, followed by the androecium, gynoecium and calyx in S. luzulaefolia; calyx, androecium and gynoecium in D. brasiliense. Tenuinucellate incomplete ovule, asymmetric integument and dual origin endothelium (funicule and integument) are new results for Asteraceae. The archesporium is usually unicellular, but ovules with two archesporial cells were observed. In S. luzulaefolia two types of tetrad are present, the linear tetrad being more commom than the tetrad with paired ginospores. The ginophyte development pattern in D. brasiliense can be an indication of its derived position in Barnadesioideae, since it has bisporic ginophyte of the Allium type and in S. luzulaefolia where ginophyte is monosporic of the Polygonum type, integrating these features simultaneity of mitotic cycles and cellularization of ginófito. The mature ginophyte is 4-cell, in both species, because of the degeneration of the antipodes, which can configure a plesiomorphic character for the family. The synergids extend into the micropile canal and they have fibrillar apparatus of typical morphology for Asteraceae. Labyrinthine walls of the middle cell and flange type wall thickening are present only in S. luzulaefolia in cells of the endothelium and micropilar canal. Therefore, common characteristics were identified for S. luzulaefolia and D. brasiliense which may be considered basal for Asteraceae and diagnostic for Barnadesioideae, as well as embryological characters that corroborate the position derived from D. brasiliense in this subfamily. / Asteraceae despertou interesse de muitos embriologistas devido a grande variabilidade das características embriológicas. Apesar disso, nada é sabido sobre a embriologia nas espécies de Barnadesioideae, sua subfamília basal. Sendo assim este trabalho objetiva descrever os processos de ginosporogênese e ginogametogênese em duas espécies de Barnadesioideae, Sclechtendalia luzulaefolia, considerada basal e irmã das demais pertencentes a família, e Dasyphyllum brasiliense cujo o gênero por vezes também foi considerado basal. Ambas as espécies são nativas do Rio Grande do Sul. S. luzulaefolia foi coletada em São Pedro do Sul, RS, Brasil e D. brasiliense em Santa Maria, RS, Brasil e em São Pedro do Sul, RS, Brasil. A corola é o primeiro verticilo floral a surgir em ambas as espécies, seguida pelo androceu, cálice e gineceu em S. luzulaefolia e pelo cálice, androceu e gineceu em D. brasiliense. O rudimento seminal tenuinucelado incompleto, a presença de tegumento assimétrico e de endotélio de origem dupla (funicular e tegumentar) são resultados inéditos para Asteraceae. O arquespório é usualmente unicelular, mas rudimentos seminais com duas células arquesporiais foram observados. Em Sclechtendalia luzulaefolia dois tipos de tétrade estão presentes, sendo a tétrade linear mais comum do que a tétrade com ginósporos pareados. O tipo de desenvolvimento do ginófito em D. brasiliense pode ser um indício de sua posição derivada para Barnadesioideae, já que possui ginófito bispórico do tipo Allium e em S. luzulaefolia o ginófito é monospórico do tipo Polygonum, integrando-se a estas características a simultaneidade dos ciclos mitóticos e da celularização do ginófito. O ginófito maduro é 4-celular, em ambas as espécies, devido degeneração das antípodas, o que pode configurar um caráter plesiomórfico para a família. As sinérgides estendem-se para o interior do canal micropilar e possuem um aparelho fibrilar de morfologia típica para Asteraceae. Somente em S. luzulaefolia foi observada a presença de parede labiríntica da célula média e espessamento de parede tipo flange em células do endotélio e canal micropilar. Portanto, neste trabalho foi possível identificar características comuns à S. luzulaefolia e D. brasiliense que podem ser considerados basais para Asteraceae e diagnósticas para Barnadesioideae, bem como caracteres embriológicos que corroboram a posição derivada de D. brasiliense nesta subfamília.

O presente trabalho analisou as etapas de formação de rudimentos seminais e grãos de pólen de Passiflora elegans e P. haematostigma. As espécies foram analisadas fazendo-se o uso de microscopia fotônica e microscopia eletrônica de varredura e transmissão. O gineceu apresenta um estigma com emergências estigmáticas, estilete sólido e ovário tricarpelar e unilocular. O ovário apresenta inúmeros rudimentos seminais que formam-se a partir de divisões periclinais da camada central. Os rudimentos são crassinucelados e os tegumentos têm origem dérmica. A célula arquesporial divide-se mitoticamente antes da meiose. A meiose resulta em uma tétrade linear, com ginósporo calazal viável. A ginogametogênese é do tipo Poligonum e, durante a diferenciação do ginófito, os estratos parietais do nucelo são parcialmente consumidos e a epiderme nucelar divide-se periclinalmente na porção apical. As antípodas são efêmeras e o ginófito maduro apresenta duas sinérgides com aparelho fibrilar desenvolvido, oosfera e núcleo secundário no pólo micropilar. O rudimento seminal é anátropo, arilado, bitegumentado e com micrópila em “zig-zag”. O androceu é constituído por cinco anteras tetrasporangiadas. Os estratos parietais desenvolvem-se do tipo Dicotiledôneo e as camadas médias são persistentes na antera madura de P. haematostigma. As células do tapete são poliplóides e colapsam liberando os conteúdos no interior do lóculo. A citocinese é do tipo simultânea e as tétrades são tetraédricas na maioria. O pólen é liberado na fase bicelular, apresenta número de colpos variável em P. elegans e seis colporos em P. haematostigma. A esporoderme não apresenta camada basal em P. elegans e é mais espessa do que em P. haematostigma. Plastídios e mitocôndrias são englobados durante a formação da célula generativa e persistem até a fase de pólen maduro. / The present work analyzed the different developmental stages of the ovule and of the pollen grain in Passiflora elegans and P. haematostigma. The species were analyzed using photonic microscopy and scanning and transmission electron microscopy. The gynoecium has a stigma with stigmatic outgrowths, solid style and tricarpelar and unilocular ovary. The ovary has several seminal rudiments that are originated from the periclinal divisions of the central layer. The ovule is crassinucelate and the dermal layer originates the integuments. The arquesporial cell divides mitotically before meiosis. The meiosis results into a linear tetrad and only the chalazal gynospore is viable. The gynogametogenesis is of the Poligonum type, the nucellus parietal layers are partially degraded during the gynophyte differentiation, and nuclear epidermis divides periclinally in the apical portion. The antipodal cells are ephemeral and the mature gynophyte has two synergids with a developed filiform apparatus, egg cell and secondary nuclei in the micropilar pole. Nas últimas décadas tem se dado maior atenção à reprodução de animais silvestres, visto que para se conhecer e preservar uma espécie se faz necessário o conhecimento da história natural aliado aos processos da fisiologia e morfologia reprodutivas. Ciclos reprodutivos podem ser sazonais ou assazonais e apresentar variações em ambos os padrões. Por sua natureza e diversidade de fatores associados (e.g., atividade, dieta, temperatura), as estratégias reprodutivas vêm sendo investigadas sob uma abordagem evolutiva. Estudos do comportamento reprodutivo e dimorfismo sexual também têm sido destacados nesse âmbito. No entanto, poucos trabalhos são realizados para se conhecer as estratégias reprodutivas em serpentes neotropicais. Na maioria das vezes tais trabalhos abordam apenas alguns aspectos do ciclo reprodutivo de serpentes, como época de nascimentos, cópula e gestação, podendo ainda ser descritos de forma divergente pelo fato de muitos destes estudos serem norteados por dados de serpentes em cativeiro. Estudos microscópicos e macroscópicos, delineados com animais recém-chegados da natureza e preservados em coleções científicas, pode fornecer uma descrição e uma análise mais efetiva do ciclo reprodutivo permitindo a integração destes fatores. Assim, este trabalho investigou diversos aspectos da biologia reprodutiva de fêmeas e machos de populações de Bothrops alternatus do Brasil. Além disso, foram analisadas e descritas as estratégias reprodutivas empregadas por ambos os sexos durante o ciclo reprodutivo anual. Para isso, os espécimes foram dissecados ventralmente retirando-se fragmentos das gônadas e vias genitais de fêmeas e machos, em diferentes épocas do ano, a fim de se identificar as principais fases do ciclo vitelogênico, espermatogênico e do segmento sexual renal. Igualmente, para determinar se há estocagem de esperma nas fêmeas e nos machos, será investigada o útero aglandular e região do infundíbulo, vias genitais e ductos deferentes respectivamente. Tais dados permitirão compor um padrão reprodutivo, para essa espécie, levando-se em conta processos morfofisiológicos em ambos os sexos. Também foi realizada uma comparação dos ciclos reprodutivos entre populações de Bothrops alternatus e análises de possíveis influências ambientais na duração dos eventos reprodutivos / Over the last decades a greater attention has been given to the reproduction of wild animals, as the knowledge of natural history combined with the processes of reproductive physiology and morphology is essential for the preservation of a species. Reproductive cycles may be seasonal or not, and both patterns may have variations. Due to its nature and to the diversity of the associated factors (e.g., activity, diet, temperature), reproductive strategies have been investigated in an evolutionary approach. Studies of reproductive behavior and sexual dimorphism have also been highlighted in this context. However, few studies have been conducted to determine reproductive strategies in neotropical snakes. Most studies address only some aspects of the reproductive cycle of snakes, such as time of birth, copulation and pregnancy, and may present divergent descriptions as many of them are based on data from snakes in captivity. Microscopic and macroscopic studies carried out with animals newcomers from nature and preserved in scientific collections can provide a more effective analysis of the reproductive cycle allowing the integration of these factors. In this context, this project aims to investigate various aspects of the reproductive biology of female and male populations of Bothrops alternatus from Brazil. The reproductive strategies employed by both sexes during the annual reproductive cycle was analyzed and described. The specimens was dissected ventrally and fragments of gonads and genital tracts of females and males was removed at different times of the year in order to identify the main phases of the vitellogenic cycle, spermatogenesis and sexual segment of the kidney. Also, to determine whether sperm storage is done by females and males, we will investigate the aglandular uterus, the posterior infundibulum, genital tracts and vas deferens respectively. Such data was compose a reproductive pattern for the species that will take into account the morphophysiologic processes of both sexes. It was also performed a comparison of reproductive cycles between populations of Bothrops alternates and analysis of possible environmental influences in the duration of reproductive events

Uma das espécies de coral pétreo zooxantelado com mais ampla distribuição no litoral brasileiro é Madracis decactis Lyman, 1859. M. decactis forma colônias incrustantes nodulares que podem atingir até 30 cm de diâmetro. O estudo da gametogênese foi realizado através de coletas bimensais na Baía de Ilha Grande, RJ durante 21 meses (agosto/2004-maio/2006), totalizando 10 coletas (12 colônias/coleta) (Licença IBAMA no. 201/2004). Foram realizados cortes histológicos de 7 µm, e de 10 até 16 pólipos por colônia foram analisados, totalizando mais de 1800 pólipos. A análise dos pólipos indicou que M. decactis é hermafrodita, com gametas localizados no mesmo lóculo gástrico, porém em mesentérios diferentes. A gametogênese durou cerca de sete meses. A ovogênese iniciou-se ao redor de outubro, enquanto que a espermatogênese teve inicio no final de fevereiro, ambas terminando em sincronia no final do mês de maio. O exame dos pólipos férteis indicou a presença dos estágios I, II e III de desenvolvimento para a ovogênese e dos estágios I, II, III e IV para a espermatogênese. Não foram encontrados embriões ou plânulas nos cortes histológicos, indicando talvez que estes estágios permaneçam pouco tempo no interior do pólipo. O pico da atividade reprodutiva ocorre entre os meses de fevereiro e abril com todas as colônias férteis contendo ovócitos principalmente no estágio III de maturação. A provável época de liberação de plânulas ocorre entre os meses de abril e maio, sob influencia das condições ambientais como a temperatura da água do mar, a irradiação solar e a pluviosidade. O presente trabalho forneceu informações básicas a respeito da biologia reprodutiva de Madracis decactis presente em Ilha Grande, sendo uma contribuição para outros estudos mais específicos na área de manejo e conservação de ambientes marinhos. / One of the most wide distributed coral species along the Brazilian cost is Madracis decactis Lyman, 1859. M. decactis growths as nodular incrusting colonies that can reach up to 30 cm in diameter. In order to study the gametogenesis cycle, bimonthly collections were done at Ilha Grande Bay, Rio de Janeiro, during 21 months (August/2004- May/2006), totaling 10 collections (12 colonies/collection) (License no. 201/2004). Histological sections of 7 µm thick were done to analyze 10 to 16 polyps per colony (more than 1800 polyps in total). The analysis indicated that M. decactis is hermaphroditic, with male and female gametes developing at the same gastric loculi, but in different mesenteries. The gametogenesis lasted about 7 months, the oogenesis starts at October , while spermtogenesis starts at the end of February, both reaching the maturity in synchrony at the end of May. The exam of fertile polyps indicated the presence of stages I, II and III for oogenesis and I, II, III and IV for spermatogenesis. No embryo or planula were observed in the histological sections, indicating that maybe these stages stay for a short period inside the polyp cavity. The peak of reproductive activity was between February and April when all the polyps were fertile containing manly stage III oocytes. The releasing of planulae may happen between April and May, under the influence of environmental conditions as sea water temperature, solar insolation and rainy season. The present study has provided basic information about the reproductive biology of Madracis decactis from Ilha Grande Bay, and it may be a contribution to further studies about management and conservation of marine environments.

The reproductive and physiological condition of a deep-sea mussel (Bathymodiolus septemdierum Hashimoto & Okutani, 1994) living in extremely acidic conditions

Oceanic uptake of anthropogenic CO2 emissions is causing wholesale shifts seawater carbonate chemistry towards a state of decreased carbonate ion concentration and reduced ocean pH. This change in water chemistry has potentially dire implications for marine organisms, especially those that build and maintain calcium carbonate structures. Our understanding of how ocean acidification may affect marine organisms is limited, as most studies have been short-term laboratory experiments. The CO2 flux from hydrothermal vent fluids on NW Eifuku submarine volcano (Mariana Volcanic Arc) provides a natural setting to investigate the effects of acidification. Here, the vent mussel, Bathymodiolus septemdierum, lives in water with pH as low as 5.22. This study was designed to examine the consequences of a low pH environment on reproduction, calcification and somatic growth in B. septemdierum, since the presumed elevated cost of acid-base regulation may diminish available energy for these processes. Histological analysis reveals both females and males display synchronous gametogenesis across collection sites with spawning occurring between late winter and early spring. Mussels are functionally dioecious, although evidence of protogynous hermaphroditism was found– a first record for the genus. In comparison with mussels at near normal pH, we find no evidence that the pattern of gametogenesis is affected by low pH conditions. However, calcification is compromised: at a given shell volume, shells from NW Eifuku weigh about half those from sites with near normal pH mussels. The condition index (CI = body ash free dry weight/ shell volume) was assessed in mussels collected from four low pH sites on Northwest Eifuku and two control sites from Lau Basin and Nifonea Ridge; we show that low pH conditions negatively affect CI, especially when energy availability is limited. Bathymodiolus septemdierum acquires energy from chemoautotrophic symbionts in the specialized gill epithelial cells. Using a gill condition index (GCI = gill ash free dry weight/ shell volume) and transmission electron microscopy to determine symbiont abundances in gill tissues, we show that NW Eifuku mussels with healthy gills and abundant symbionts have a higher CI than mussels from NW Eifuku with unhealthy gills. Optimal environmental sulphide concentrations appear to sustain higher symbiont abundances. While the survival of mussels on NW Eifuku is remarkable, it can come at a considerable cost to body and shell condition when during periods of energy limitation. Bathymodiolus septemdierum shows substantial resilience to low pH conditions when energy availability is sufficient due to energy budget adjustments that maximize fitness. / Graduate / 0329, 0472, 0416

Bromeliaceae is one of the important neotropical families. It has approximately 3501 species. Pitcairnioideae has about 170 species. The present study aims to analyze and characterize the sporogenesis and gametogenesis in anthers and seminal rudiments of Dyckia polyclada L.B. Sm. (Pitcairnioideae-Bromeliaceae). Dyckia polyclada L.B.Sm. is native and endemic to Rio Grande do Sul and it is found in the central area of the state. The species presents perfect trimerous and heterochlamydous flowers. The androecium consists of six stamens, tetrasporangiate and dorsifixed anthers. Nine different phases of development were defined when it comes to the chemical constituents of the anthers. Only starch was detected in the connective. Starch and reserve lipids were observed in sporangia, microspores and pollen grains. The pollenkitt presents neutral lipids, essential oils and oleoresins in its chemical composition. The ovary in D. polyclada is superior, tricarpellate, trilocular and polispermic. Placentation is axial. The stigma is trífid of the conduplicate spiral. The ovules are anatropous, bitegmic and crassinucellate. The teguments are from dermic origin. The micropyle is formed by the inner tegument. The gametophyte is monosporic of the Polygonum type. The fibrillar aparattus is from pectic nature. Infralocular, labyrinthine and unstructured septal nectaries are present. The chalazal appendix is short and curved toward the raphe and features elongated cells with intracellular spaces. The obturator in D. polyclada has elongated dome-shaped cells and dense, vacuolated cytoplasm, coating the entire placental region. The presented data are unprecedented to the species and contribute to the understanding of the phylogenetic relationships within the family and for better understanding of the functioning of the reproductive process of Dyckia polyclada, given that, for lack of studies, the species is in the category of insufficient data and one cannot diagnose the degree of extinction. / Bromeliaceae é umas das importantes famílias neotropicais. Possui aproximadamente 3501 espécies. Pitcairnioideae apresenta cerca de 170 espécies. O presente trabalho tem por objetivo analisar e caracterizar a esporogênese e a gametogênese em anteras e em rudimentos seminais em Dyckia polyclada L.B. Sm. (Pitcairnioideae-Bromeliaceae). Dyckia polyclada L.B. Sm. é nativa e endêmica do Rio Grande do Sul, sendo encontrada na região central do estado. A espécie apresenta flores perfeitas, trímeras e heteroclamídeas. O androceu é composto por seis estames, anteras tetraesporângiadas e dorsifixas. Quanto a investigação sobre os contítuintes químicos da antera foram definidos nove estádios distintos de desenvolvimento. Apenas amido foi detectado no conectivo. Amido e lipídios de reserva foram observados nos esporângios, micrósporos e grãos de pólen. O pollenkitt apresenta lipídios neutros, óleos essenciais e oleorresinas em sua composição química. O ovário de D. polyclada é súpero, tricarpelar, trilocular e plurispérmico. A placentação é do tipo axial. O estigma é trífido do tipo espiral conduplicado. Os rudimentos seminais são anátropos, bitegumentados e crassinucelados. Os tegumentos são de origem dérmica. O canal micropilar é formado pelo tegumento interno. O gametófito é de origem monospórica e do tipo Polygonum. O aparelho fibrilar é de natureza péctica. Nectários septais infraloculares, labirínticos e não estruturados estão presentes. O apêndice calazal é curto e curvo em direção a rafe e apresenta células alongadas e com espaços intracelulares. O obturador em D. polyclada apresenta células alongadas em forma de domo e citoplasma denso e vacuolado, revestindo toda a região placentária. Os dados apresentados são inéditos para a espécie e contribuem para a compreensão das relações filogenéticas dentro da família e para melhor entendimento do funcionamento do processo reprodutivo de Dyckia polyclada, tendo em vista que a espécie, por falta de estudos, encontra-se na categoria de dados insuficientes e não se pode diagnosticar o grau de risco de extinção.

Gametogenesis and spawning of the elliptical star coral, Dichocoenia stokesi (Cnidaria: Scleractinia) in Southeast Florida

Reproductive characters of the elliptical star coral Dichocoenia stokesi (Cnidaria: Scleractinia) were investigated based in histological examination of tissues collected in southeastern Florida between September 1999 and September 2000. The study population was predominantly gonochoric with a small incidence of hermaphroditism. Dichocoenia stokesi exhibits intermediate egg size (312.2 ± 40.4μm; mean ± SD) compared with other scleractinian species, suggesting a moderately lecithotrophic larval stage. Although spawning was not directly observed in this study, two separate spawning events per year are inferred from histological slide analyses; the first in late August or early September, and the second in early October. Sea water temperature was significantly positively correlated with female gonadal development in D. stokesi, whereas insolation was significantly inversely correlated with gonadal development. A recent epizootic of White Plague Type II in south Florida appears to be skewing local D. stokesi populations toward smaller - and potentially less fecund - colony sizes, although colony size and fecundity were not correlated in this study. The mean fecundity of the study population is estimated as 1138.1 eggs cm-2 yr-1.

Signaling mechanisms involved in regulated vesicle discharge in Plasmodium falciparum / Mécanismes de signalisation impliqués dans l'écoulement des vésicules régulées chez Plasmodium falciparum

L'activation des gamétocytes de Plasmodium falciparum par exposition à l'acide xanthurénique à basse température conduit à la sortie de gamètes mâles et femelles matures des érythrocytes de l’hôte, ce qui est essentiel pour la fertilisation et la formation ultérieure de zygotes. Les gamètes possèdent des vésicules cytosoliques contenant une protéine formant des pores connue sous le nom de PfPLP2. Durant la sortie, les vésicules ayant PfPLP2 sont redistribuées du cytoplasme des gamètes à la périphérie. Puis ces vésicules vont sécréter PfPLP2 permettant ainsi la sortie des gamètes par perforation de la membrane érythrocytaire. Dans cette étude, nous avons identifié le rôle d'une phospholipase de type patatine A2 (PfPATPL1) dans la sortie de gamètes de P. falciparum. Nous avons utilisé une approche de génétique inverse pour générer un knock-down conditionnel de PfPATPL1 par fusion à un domaine de déstabilisation (DD). Nous avons modifié l’expression PfPATPL1 dans les gamétocytes de stade V en supprimant le ligand de Shield-1, ce qui nous a permis de constater l’importance de PfPATPL1 durant les différentes étapes de la gamétogenèse, ainsi que dans l'arrondissement des gamétocytes, la sortie des gamètes et l'exflagellation des gamètes mâles. Nous avons également trouvé que PfPATPL1 est nécessaire pour la redistribution des vésicules portant PfPLP2 à la périphérie des gamètes pendant sortie. De plus, nous avons utilisé un inhibiteur connu de la phospholipase du nom de bromure de 4-bromophénacyle (4-BPB) pour étudier l'importance de l'activité de la PLA2 dans la gamétogenèse. Nous avons découvert que le traitement par le 4-BPB entraîne également des anomalies dans l'arrondissement des gamétocytes, inhibe la sortie des gamètes et l'exflagellation des gamètes mâles. Et que ce même traitement au 4-BPB entravait la redistribution des vésicules contenant PfPLP2 à la périphérie des gamètes. Lorsque les moustiques ont été nourris avec des gamétocytes traités au 4-BPB dans un test d'alimentation membranaire standard (SMFA), nous avons observé une diminution significative du taux d'infection des moustiques et une chute drastique de la densité des oocystes et du nombre de moustiques infectés. Ces données suggèrent que PfPATPL1 est important pour le développement des gamètes chez les moustiques et peut constituer une cible prometteuse pour les stratégies d'intervention de transmissivité. / The activation of Plasmodium falciparum gametocytes by exposure to low temperature and xanthurenic acid leads to the egress of mature male and female gametes from host erythrocytes, which is essential for fertilization and subsequent zygote formation. Gametes contain cytosolic vesicles bearing a pore forming protein known as PfPLP2. During egress, PfPLP2 containing vesicles gets redistributed from gamete cytoplasm to periphery. Subsequently, PfPLP2 is secreted from these vesicles leading to perforation of host erythrocyte membrane resulting in gamete egress. In this study, we have identified the role of a patatin-like phospholipase A2 (PfPATPL1) in P. falciparum gamete egress. We have taken a reverse genetics approach to generate a conditional knock down of PfPATPL1 by fusion to a destabilization domain (DD). We have knocked down PfPATPL1 in Stage V gametocytes by removal of Shield-1 ligand and found that PfPATPL1 is required during different steps of gametogenesis including gametocyte rounding up, gamete egress and male gamete exflagellation. We have also found that PfPATPL1 is needed for redistribution of PfPLP2 bearing vesicles to the gamete periphery during egress. Additionally, we have utilized a known inhibitor of phospholipase A2 known as 4- bromophenacyl bromide (4-BPB) to study the importance of PLA2 activity in gametogenesis. We have found that 4-BPB treatment also leads to defects in gametocyte rounding up, inhibits gamete egress and male gamete exflagellation. 4-BPB treatment also hampers the redistribution of PfPLP2 containing vesicles to gamete periphery. When the mosquitoes were fed with 4-BPB treated gametocytes in a standard membrane feeding assay (SMFA), we observed a significant decline in mosquito infection rate and a drastic drop in oocyst density and number of infected mosquitos. These data suggests that PfPATPL1 is important for gamete development in mosquitos and may serve as a promising target for transmmision intervention strategies.

The Phylogeny of Land Plants: A Cladistic Analysis Based on Male Gametogenesis

A cladistic analysis was carried out to resolve phylogenetic pattern among bryophytes and other land plants. The analysis used 22 taxa of land plants and 90 characters relating to male gametogenesis. Coleochaete or Chara/Nitella were the outgroups in various analyses using HENNIG86, PAUP, and MacClade, and the land plant phylogeny was unchanged regardless of outgroup utilized. The most parsimonious cladograms from HENNIG86 (7 trees) have treelengths of 243 (C.I. = 0.58, R.I. = 0.82). Bryophytes are monophyletic as are hornworts, liverworts, and mosses, with hornworts identified as the sister group of a liverwort/moss assemblage. In vascular plants, lycophytes are polyphyletic and Selaginella is close to the bryophytes. Lycopodium is the sister group of the remaining vascular plants (minus Selaginella). Longer treelengths (over 250) are required to produce tree topologies in which either lycophytes are monophyletic or to reconstruct the paraphyletic bryophyte phylogeny of recent authors. This analysis challenges existing concepts of bryophyte phylogeny based on more classical data and interpretations, and provides new insight into land plant evolution.
