The Phenomenon Of Blastocyst Hatching : Role Of COX-2 And NF-kBRoy, Shubhendu Sen 06 1900 (has links) (PDF)
The zona-pellucida (zona, ZP) is an adhesion-refractory, acellular coat enclosing the rapidly growing, free-living mammalian preimplantation embryo which undergoes successive cleavage divisions to form the blastocyst, composed of ICM-cells surrounded by outer TE-cells. For further development, the blastocyst must ‘hatch’ or ‘escape’ out of the zona before it can implant into the endometrium for further development (Fig. 5.1A). Hence, the event of hatching or ‘zona escape’ assumes critical importance for the establishment of a successful pregnancy. The golden-hamster blastocyst offers a very unique paradigm to understand hatching, whereby upon attainment of a fully-expanded state, the blastocyst undergoes a dramatic (and molecularly unexplained) deflation event, followed by appearance of TE-derived dynamic cellular projections called TE-projections, whose appearance in an embryonic-stage and -time dependant manner suggest an intimate association with the hatching phenomenon (Fig. 5.1B). Thirdly, embryo-derived zonalytic proteases have been shown to bring about a focal-lysis of the ZP followed by global zona dissolution. Earlier work in the laboratory had demonstrated the intimate involvement of signaling molecules like LIF, HB-EGF, TGF-β and (ER)-α with hatching (Seshagiri et al., 2002, 2009). Investigations also revealed the involvement of cysteine-proteases of the cathepsin (cts) family, especially cts-L, -B and-P to be involved in zona lysis (Sireesha et al., 2008).
In order to achieve a better understanding of mammalian preimplantation development, especially hatching, it was important to investigate the role and impact of other critical regulators of developmental and reproductive physiology. COX-2 is one such key signaling moiety and it was decided to investigate the role, if any, of COX-2 and its derived PGs in hamster peri-implantation events. COX-2 transcripts and immunoreactive COX-2 protein were detected in the different preimplantation stages, from 8-cell onwards. COX-2 protein was abundant in both the ICM and TE, but was especially enriched in the TE-cells of the late blastocyst. In order to investigate the function of this enzyme in preimplantation development and hatching, two very-specific inhibitors of COX-2 catalytic action, NS-398 and CAY-10404, were tested in identical concentrations of 25, 50 and 75 μM on in vitro cultured hamster blastocysts. In order to assess the impact of COX-2 inhibition on an embryo-stage and time-dependant manner, inhibitors were tested on freshly recovered 8-cell embryos or early blastocysts, continuously for 72 h or 48 h, respectively. COX-2-selective inhibitors inhibited hamster blastocyst hatching in a dose-dependant manner with maximum inhibition observed in the 75 μM dose. Surprisingly, there was a profound dose-dependent failure of deflation of late-blastocysts upon inhibitor treatment and embryos which hatched, did so in an inflated state and retained intact zonae in cultures. Moreover, embryos subjected to NS-398 treatment phenocopied those subjected to CAY-10404 treatment. Results demonstrate that the effect of inhibitors, and hence the need for COX-2 mediated signaling events is more pronounced in 8¬cell embryos than with early-blastocysts, indicating that COX-2 dependant molecular and cellular processes required for blastocyst morphogenesis and ZP-lysis may have been initiated prior to compaction and cavitation.
The reversal of effects of COX-2 inhibition on hatching with exogenous addition of PGE2 and Iloprost (a stable PGI2 analogue) to inhibitor cultures, show that COX-2-derived eicosanoids could, in effect bring about hamster hatching, which is in agreement with previous reports (Davis et al., 1999) and augment peri-implantation development including hatching (Huang et al., 2003). Additionally, it has been successfully demonstrated that PGE2 was superior to PGI2 in augmenting blastocyst hatching in inhibitor-cultures. In this study, the modulation of critical cts-L, -B, -P proteases in COX-2 mediated hamster zona hatching has been verified by quantifying cts in transcripts in control and inhibitor-subjected embryonic samples which was further substantiated by the decreased intra-embryonal protein levels of cts-L and -P. These results demonstrate that COX-2 mediated signaling components directly and effectively modulate hamster preimplantation development, especially zona-hatching phenomenon by transcriptional-regulation of the critical zonalytic proteases.
Another potential hatching-associated molecule i.e., NF-κB which is known to exert a great deal of influence on overall reproductive and developmental biology, was investigated in this study. Its specific effects on mammalian preimplantation development, especially hatching, remain totally uninvestigated. This formed the rationale to investigate the reach and impact of NF-κB signaling network in the modulation of peri-hatching events. Transcripts and immunoreactive NF-κB protein of crucial pathway-components like IKK, IκB-β and RelA were detected from 8-cell embryo to the zona-free blastocyst. In order to ascertain the impact of NF-κB signaling on peri-hatching events, two very-specific inhibitors of the NF-κB pathway, BAY-11-7082 and JSH-23 were employed which acted at two strategic signaling points. In order to assess the impact of NF-κB inhibition on an embryo-stage and time-dependant manner, inhibitors were tested on freshly recovered 8-cell embryos or early blastocysts, continuously for 72 h or 48 h, respectively. NF-κB-selective inhibitors inhibited blastocyst hatching in a dose-dependent manner. Interestingly, a profound dose-dependent failure of deflation of late-blastocysts upon inhibitor treatment was observed and embryos which hatched did so in an inflated state and also retained intact zonae in cultures. Moreover, embryos subjected to BAY-11-7082 treatment phenocopied those subjected to JSH-23 treatment, indicating specificity of inhibitor action. Time-course experiments demonstrated that the need for efficient NF-κB mediated signaling is distinctively more for 8-cell embryos than early-blastocysts, indicating that NF-κB dependant molecular and cellular processes required for blastocyst morphogenesis and ZP-lysis may have been initiated prior to compaction and cavitation.
Moreover, modulation of zonalysins cts-L, -B and -P by NF-κB-signaling, during the event of zona lysis, both by real-time quantitation of its transcripts and intracellular protein levels has been demonstrated. These results demonstrate that NF¬κB mediated signaling components directly and effectively modulate hamster preimplantation development, especially zona-hatching phenomenon by transcriptional-regulation of the critical zonalytic proteases. The profound inhibition of hatching and effects on blastocyst morphogenesis observed by inhibition of COX-2 and NF-κB signaling systems demonstrate a fundamental need of the growing embryo for these critical signaling moieties. Moreover, the underlying similarity of consequences obtained upon inhibition of both signaling networks i.e., NF-κB and COX-2, perhaps indicate a linear mode of signaling between these principles. It remains to be tested, though, if it really is the case.
A striking observation made in this study was the detection of immunoreactive signals for critical signaling moieties like ER-α, COX-2 and RelA onto TEPs of the deflated hamster blastocyst, in addition to earlier TEP-localisation of cathepsins.

Preliminary results indicate that TEPs could be the site of membrane lipid-rafts, focal points of membrane-based signaling. The definitive role of TEPs in peri-hatching events is yet to be confirmed, but it is presumed that these actin-based undulating structures, harboring several key molecules involved in peri-implantation events in the embryo as well as the maternal uterus could be instrumental in successfully bringing about the concomitant processes of hatching and implantation. Interestingly, during rodent implantation (hamster, guinea-pig, mouse and rat), the blastocyst orients in such a way that the ICM is oriented away from the endometrium and, at least in the hamster, the TEP-carrying abembryonic (mural) pole remains closest to the luminal epithelium (LE) (Gonzales et al., 1996b; Seshagiri et al., 2009; Fig. 5.1C). In contrast, in humans and other primates, the embryonic pole is closest to LE before implantation (Kirby, 1971; Lee and DeMayo, 2004). Although direct evidence is lacking, but these observations gives rise to a possibility that both hatching and implantation could be intimately related to the polar appearance of TEPs in the embryo.
Several key signaling molecules like ER-α, LIF, HB-EGF and TGF-β have been already demonstrated to play crucial roles in mammalian hatching. In this thesis, we have exemplified the need for COX-2 mediated prostanoid signaling and the pleotropic NF-κB signaling system in bringing about mammalian blastocyst hatching. How exactly do these molecular entities communicate among themselves and with cellular principles like TEPs thereby effectively enabling peri-implantation development, remain to be understood.
Taken together, these results demonstrate, for the first time, the involvement of embryo-derived signaling molecules, like COX-2 and NF-κB in an embryo stage-and time-dependant manner in mammalian peri-implantation events, especially blastocyst hatching. The association of TEPs with key molecules common to embryonic and maternal preparation for hatching and implantation, respectively, indicates towards a molecular and cellular continuity between the concomitant events. These fundamental findings on hamster blastocyst biology have profound clinical implications in the management of human infertility.

Distributed space-time coding, including the Golden Code, with application in cooperative networksGe, Lu January 2015 (has links)
This thesis presents new methodologies to improve performance of wireless cooperative networks using the Golden Code. As a form of space-time coding, the Golden Code can achieve diversity-multiplexing tradeoff and the data rate can be twice that of the Alamouti code. In practice, however, asynchronism between relay nodes may reduce performance and channel quality can be degraded from certain antennas. Firstly, a simple offset transmission scheme, which employs full interference cancellation (FIC) and orthogonal frequency division multiplexing (OFDM), is enhanced through the use of four relay nodes and receiver processing to mitigate asynchronism. Then, the potential reduction in diversity gain due to the dependent channel matrix elements in the distributed Golden Code transmission, and the rate penalty of multihop transmission, are mitigated by relay selection based on two-way transmission. The Golden Code is also implemented in an asynchronous one-way relay network over frequency flat and selective channels, and a simple approach to overcome asynchronism is proposed. In one-way communication with computationally efficient sphere decoding, the maximum of the channel parameter means is shown to achieve the best performance for the relay selection through bit error rate simulations. Secondly, to reduce the cost of hardware when multiple antennas are available in a cooperative network, multi-antenna selection is exploited. In this context, maximum-sum transmit antenna selection is proposed. End-to-end signal-to-noise ratio (SNR) is calculated and outage probability analysis is performed when the links are modelled as Rayleigh fading frequency flat channels. The numerical results support the analysis and for a MIMO system maximum-sum selection is shown to outperform maximum-minimum selection. Additionally, pairwise error probability (PEP) analysis is performed for maximum-sum transmit antenna selection with the Golden Code and the diversity order is obtained. Finally, with the assumption of fibre-connected multiple antennas with finite buffers, multiple-antenna selection is implemented on the basis of maximum-sum antenna selection. Frequency flat Rayleigh fading channels are assumed together with a decode and forward transmission scheme. Outage probability analysis is performed by exploiting the steady-state stationarity of a Markov Chain model.

Chromosomal evolution and phylogeny of golden moles and tenrecs (Mammalia : Afrosoricida)Gilbert, Clement 03 1900 (has links)
Thesis (PhD (Botany and Zoology)) -- University of Stellenbosch, 2008. / Afrosoricida is a 65 million years old (my) eutherian order that together with the
Tubulidentata (aardvark) and Macroscelidea (elephant shrews) form the
Afroinsectiphillia, a subclade of Afrotheria. It includes two families – Chrysochloridae
(nine genera of golden moles) and Tenrecidae (11 genera of tenrecs) – that collectively
represent ~59% of the afrotherian generic diversity. This study presents the first
comprehensive cytogenetic comparison between members of these two families (seven
genera and 11 species/subspecies of golden moles, and two genera and 11 species of
tenrecs) using G- and C-banding and chromosome painting. All detected
rearrangements are interpreted in a strict cladistic framework. In the case of
Chrysochloridae, this provides evidence for a sister relationship between Chrysochloris
and Cryptochloris, the monophyly of the Amblysomus genus, and for the elevation of A.
hottentotus meesteri to specific rank. The detection of telomeric-like repeats in the
centromeres of all chromosomes of the Amblysomus species/subspecies but not in those
of A. h. meesteri further strengthens its recognition as a distinct species. Parsimony
analysis of chromosomal rearrangements within Tenrecidae, the second Afrotheria
assemblage studied, showed that rearrangements which could be interpreted as Whole
Arm Reciprocal Translocations (WARTs) were more likely to be the result of
Robertsonian translocations. Four interspecific associations are recovered within
Microgale that are consistent with morphological and molecular characters. It was also
possible to infer ancestral karyotypes for the Chrysochloridae, Oryzorictinae and the
two tenrecid genera, Oryzorictes and Microgale. Given the relatively high karyotypic
diversity observed among some Microgale species and the prevailing debates on
chromosomal evolution and regional palaeoenvironmental fluctuations, it is suggested
that Microgale be added to the list of taxa where structural rearrangements are likely to
have played a role in speciation. Using Genbank sequences and a relaxed Bayesian
clock method, we estimate the age of the family Chrysochloridae at ~28.5 my and that
of the genus Microgale at ~9.9 my. Based on these dates, it can be shown that most of
the evolutionary branches are characterized by a slow rate of chromosomal change, but
that markedly high rates are observed in some Microgale species and to a lesser extent
in the lineage leading to A. robustus. The rates of chromosomal evolution and other
cytogenetic features highlighted in this study are discussed in light of recent advances in
understanding the molecular mechanims that underpin changes to genomic architecture.

Theory of intrinsic and extrinsic tunnelling in cupratesBeanland, Joanne January 2010 (has links)
This thesis addresses the tunnelling of charge carriers in different materials. First looking at the simplest case of electron tunnelling in metals at zero, then finite temperature, the current is obtained using the Fermi-Dirac golden rule and then the conductance is obtained. This is extended to take into account the spatial dependence of one of the metals being a tip since experimentally this is done by scanning tunnelling microscopy where a tip traces over the surface of a sample. The next step is to look at tunnelling between a metal and a semiconductor, again the current is found. Semiconductors can be doped and the effect this has on tunnelling is examined. Next superconductors are introduced. The purpose of my research has been to look at the tunnelling spectra of high-temperature superconducting cuprates for both extrinsic (metal-superconductor) and intrinsic (superconductor-superconductor) tunnelling. The main features seen experimentally with cuprate tunnelling are identified and then a theory capable of explaining these features is discussed. The theory is compared to experimental results and we find good agreement.

The expansion of wind power comes with additional risks for bird populations, one of which is the collision with wind turbine components, mainly the rotating blades. In order to mitigate this risk, few systems on the market propose solutions with modules that deter the birds from the wind turbine or trigger the temporary shutdown of the turbines.The thesis seeks to obtain a common view from different stakeholders in order to create a list of requirements that the bird protection systems must have so as to be accepted by all the parties involved in the wind industry in Sweden. In order to meet this objective, interviews were conducted with interested stakeholders. Consequently, the interviews were summarized and common points were extracted to create a list of requirements and additional suggestions.Firstly, a comprehensive literature review was done that has pointed out issues like impacts of wind farms on bird species, especially raptors, causes of bird mortality, environmental legislation in European Union and Sweden, and current solutions on mitigating the risk of collisions with wind turbines onshore. Additionally, the author looked up into the most efficient ways to engage the stakeholders in constructive discussions and develop an easy framework for presenting the requirements.The primary data was collected through interviews with stakeholders representing 12 organizations. In the next steps, the data was analyzed by summarizing each interview and identifying the common and differentiating points in respondents’ views and suggestions over automated bird mitigation systems. The identified points laid the basis for a list of requirements considered important for safe and efficient bird mitigation systems in Sweden. Among others, stakeholders identified that the most crucial issues regarding bird mitigation systems are the response time to shutdown of the turbine, distance from the birds’ species living areas, need of species identification in bad weather conditions and night time and overall financial feasibility of such technologies.

Conspecific Attraction in a Low-Density Population of a Threatened SongbirdAlbrecht-Mallinger, Daniel James 01 May 2014 (has links)
Many organisms use both vegetation structure and social cues in selecting habitats. Many species of songbirds use the presence of breeding conspecifics as a social cue and sign of habitat quality, and can be induced to settle in unoccupied habitats by artificially broadcasting breeding song, a process referred to as “conspecific attraction”. In our study, we tested response to conspecific attraction a low-density population of the threatened Golden-winged Warbler (Vermivora chrysoptera) in Highland County, VA. Response to broadcast song was observed, with a modest increase in mean male abundance at survey points within 250 meters of treatments, and mean abundance decreasing outside of this range throughout the study area. Recruitment to conspecific playback was lower in our study than observed in previous research on high-density systems. Our results suggest more research is needed of the effectiveness of conspecific attraction in low density species and that its use should be tailored to the spatial and demographic conditions of the managed population.
