An investigation into the biology and function of protein Icb-1

In this thesis I describe an investigation into the function of the protein Icb-1, a homologue of Themis1 in B cells and monocytes. Themis1 is important for T cell positive and negative selections. Yet its function in T cell development is not clear. Although it shows characteristics of an adaptor protein and involvement in TCR-induced signalling, the exact signalling defects in Themis1-/- T cells remain obscure. Icb-1 is similar to Themis1 in sequence, function and binding partners. It has been studied in human tumour and macrophage cell lines, leading to limited conclusions. Its role in B cells has never been published. Given the link with Themis1, it is of great interest to investigate the function of Icb-1. My study has been focused on the comparison between Icb-1 knockout mice with wild-type controls. I characterised the B cell development in Icb-1-/- mice, either naturally born or produced as mixed adult bone marrow chimeras reconstituted from WT and Icb-1-/- donor cells. I examined the possible compensation and redundancy of Themis1 and Icb-1, by characterising Thems1/Icb-1 double knockout mice. The Ig-HEL mouse models were used to examine the change in B cell repertoire due to negative and positive selections. The mice were challenged with SRBCs or NP-CGG to examine the germinal centre response to foreign antigen when Icb-1 is absent. In vitro stimulation of B cells with soluble and membrane-bound antigens was used to investigate early B cell responses in detail and to give insights into the defects found in in vivo challenges. Finally, I examined the BCR-induced phosphorylation of key signalling molecules and Ca2+ flux in splenic B cells. The study revealed largely normal B cell development with subtle selection impairments, but a partially defected B cell immune response to antigens in Icb-1-/- mice. The marginal zone B cell population was enlarged in the absence of Icb-1, while the positive selection of B1 B cells induced by intracellular self-antigen was impaired. The deficient mice showed a reduction in germinal centre B cell generation. The defects are associated with impaired BCR-induced cell signalling to low abundance and/or low avidity antigens. In particular, Ca2+ flux and Erk1/2 phosphorylation were clearly reduced under certain conditions. The results shine a light on the function of protein Icb-1, and also improve our knowledge of Themis1 and the Themis family. They provide a new avenue of investigation into the regulation of BCR signalling, especially in Ca2+ flux induction and Erk1/2 activation. They also provide insight into how differential signalling is controlled within cells during activation and differentiation in response to antigens that vary in terms of affinity, avidity and frequency.

This thesis deals with certification of project managers according to the ICB standard, which focuses on the competence of the project manager. The theoretical part introduces the historical development and approaches to project management, basic concepts in project management and widespread standards and certification in project management. The practical part is devoted to detailed description of the version 3 and 4 of the ICB standard. Subsequently comparison of both versions according to selected criteria and on the basis of this comparison are evaluated strengths and weaknesses of each version. The conclusion is formulated to benefit ICB 4 for the practice of project management and is designed to possibly form the structure of the standard.

The objective of this diploma thesis is to design a proposal for managing projects in small and mid-sized companies that will be part of reference Model of Business Informatics MBI. MBI is a reference model for business IT, which is being developed by Department of Information Technologies at the University of Economics in Prague. MBI contains best practices in combination with theoretical background. The proposal includes prepared document templates ready to use in project management. This thesis also compares well-known and most widely used project management standards. The theoretical part is focused on the most widely used project management standards and methodologies. First chapter compares PMBOK, ICB and PRINCE2. This chapter is focused on the most important characteristics, used approaches and differences of these standards. Next chapter is closely focused on PRINCE2 described in terms of defined processes. In the practical part of this diploma thesis is a proposal for managing projects in small and mid-sized companies. The proposal takes the form of tables that matches the MBI model structure. This proposal is available on MBI website and is ready to use in practice. The theoretical part of this thesis is mainly based on the official publications and information from official websites of companies focused on project management. Practical part of this thesis is based on theoretical and practical experience. The usability of the results is evaluated in conclusion.

The second part is composed of questionnaires’ evaluation and further comparison, which were made by using statistic calculations and coefficients. Findings acquired through questionary assessment are described as follows and on their basis a recommendation for IPMA development is given.

Adquiriu seus conhecimentos anatômicos convivendo com Renato Locchi e Odorico Machado de Sousa, consagrados anatomistas e discípulos de Afonso Bovero. Contratado como docente do Departamento de Anatomia da FMUSP foi orientado pelo professor Odorico que, com auxílio financeiro da Fundação Humboldt enviou-o para a realização de um estágio na Alemanha no Instituto Anatômico de Kiel, sob a tutela de Wolfgang Bargmann, discípulo de Alfred Benninghoff, um dos fundadores da Anatomia Funcional da Escola Alemã, quando se especializou na Anatomia Funcional de Conotação Secretória Celular e Subcelular. Em um novo estágio, agora no Instituto de Anatomia da Universidade de Mainz, coordenado por Adolf Dabelow, um discípulo de Alfred Benninghoff, obteve fundamentos sólidos para o desenvolvimento da Anatomia Funcional de Conotação Mecânica. Como resultado dessas experiências desenvolveu diversos trabalhos no âmbito da Anatomia Funcional, tornando-se o principal representante dessa vertente no Brasil, uma ciência de cunho dinâmico, que visa estabelecer a relação entre a estrutura e a sua função sem, contudo, enveredar para o campo da Fisiologia. The diploma thesis is focused on the topic of managerial competencies and competency models, as a particular combination of competencies that are necessary for the effective performance of the work. Managerial competencies are in this thesis specifically focused on the competencies of the project manager. The aim of the diploma thesis is based on an analysis to identify the competencies of the project manager and on this basis to develop generic competency model of project manager. The theoretical basis of the work is focused mainly on the issues of the concept of competence, the development, use and types of competencies. Further are processed concepts of the area of managerial competencies and project management on the basis of literature and Internet resources on this topic. Own part is based on analysis of documents, which is the basis for compiling the survey, whose result is a list of selected competencies important for work as a project manager. Based on the list of important competencies is compiled generic competency model project manager. In conclusion summarizes the results from own part.

Manažerské kompetence v projektovém řízení / Managerial Competencies in Project Management

In project management, the role of the person in charge the project manager is absolutely crucial. Depending on the requirements of the company, all kinds of demands can be placed on them. The first part of this dissertation is focused on important milestones in the development of project management, leading to the concept of the project manager itself. Drawing on the relevant literature, there is an emphasis on the basic definition of his/her role and how it is currently perceived. The role is theoretically defined in a competence model, which is discussed in the practical part of the dissertation. The second part deals with a questionnaire, which is evaluated using STATISTICA12 software. According to outputs from the quantitative method of research, there are then interviews with people who work in the field of project management - which provide additional background and explanation to the results of the research. In the conlusion of thesis is proposed a own competency model,which is based on an analysis of theoretical background and the results of the applied methods and which is also inspired by IPMA Competence Baseline 3.
