The Diploma Thesis "Evaluation of the Financial Situation in the Water Rescue Services and Proposals to Its Financing" I have chosen, because this issue is very actual and affects a large part of the population. Water Rescue Service cares about the safety and health of people in more than 80 locations in the Czech Republic. Thanks to learning the methods of the financial analysis will be possible in practical part to evaluate the situation in the Water Rescue Service. The main proposal is analysis of the one local group and the other part is presenting the functioning of a few selected groups. All found data will lead to the creation of the proposals and solutions of the financing that will help this organization continue to operate and take care about the health of many people.

Algorithme de recherche incrémentale d'un motif dans un ensemble de séquences d'ADN issues de séquençages à haut débit / Algorithms of on-line pattern matching in a set of highly sequences outcoming from next sequencing generation

Dans cette thèse, nous nous intéressons au problème de recherche incrémentale de motifs dans des séquences fortement similaires (On-line Pattern Matching on Highly Similar Sequences), issues de technologies de séquençage à haut débit (SHD). Ces séquences ne diffèrent que par de très petites quantités de variations et présentent un niveau de similarité très élevé. Il y a donc un fort besoin d'algorithmes efficaces pour effectuer la recherche rapide de motifs dans de tels ensembles de séquences spécifiques. Nous développons de nouveaux algorithmes pour traiter ce problème. Cette thèse est répartie en cinq parties. Dans la première partie, nous présentons un état de l'art sur les algorithmes les plus connus du problème de recherche de motifs et les index associés. Puis, dans les trois parties suivantes, nous développons trois algorithmes directement dédiés à la recherche incrémentale de motifs dans un ensemble de séquences fortement similaires. Enfin, dans la cinquième partie, nous effectuons une étude expérimentale sur ces algorithmes. Cette étude a montré que nos algorithmes sont efficaces en pratique en terme de temps de calcul / In this thesis, we are interested in the problem of on-line pattern matching in highly similar sequences, On-line Pattern Matching on Highly Similar Sequences, outcoming from Next Generation Sequencing technologies (NGS). These sequences only differ by a very small amount. There is thus a strong need for efficient algorithms for performing fast pattern matching in such specific sets of sequences. We develop new algorithms to process this problem. This thesis is partitioned into five parts. In the first part, we present a state of the art on the most popular algorithms of finding problem and the related indexes. Then, in the three following parts, we develop three algorithms directly dedicated to the on-line search for patterns in a set of highly similar sequences. Finally, in the fifth part, we conduct an experimental study on these algorithms. This study shows that our algorithms are efficient in practice in terms of computation time.

Orientador: Anna Silvia Palcheco Peixoto / Resumo: A erosão é um problema ambiental em que a perda de solo pode acarretar problemas econômicos e, quando próximos a urbanizações, problemas sociais. A cidade de São Manuel está situada no interior de São Paulo e apresenta, tanto solos argilosos com baixa suscetibilidade a erosão, como solos arenosos com alta suscetibilidade a erosão. Uma vez que existem áreas suscetíveis à erosão próximas à área urbana capazes de colocar a população em risco, surgiu a necessidade de analisá-las a fim de auxiliar políticas públicas para minimizar suas consequências. Com isso, a presente pesquisa propôs o desenvolvimento de uma metodologia para gerar índices de erosão, por Análise de Componente Principal (PCA) e por Análise Fatorial, fundamentado em alguns dos principais fatores influenciadores nos processos erosivos que ocorrem na área urbana do município. Nessa etapa foram considerados: textura do solo; declividade; permeabilidade; uso e ocupação; pluviosidade; e erodibilidade dos solos. Inicialmente, foram levantadas as feições erosivas existentes na área urbana, espacializadas e classificadas. Entre as 9 feições espacializadas, duas eram provenientes de processos fluviais, duas estavam recuperadas, restando cinco feições erosivas lineares para serem estudadas. Uma das cinco, apesar de estar estabilizada, apresentou um avanço significativo em um dos braços. São Manuel town is located in the interior of São Paulo and has both clayey soils with low susceptibility to erosion and sandy soils with high susceptibility to erosion. Since there are areas susceptible to erosion close to the urban area capable of putting the population at risk, the need arose to analyze them in order to assist public policies to minimize their consequences. Therefore, this research proposed the development of a methodology to generate erosion indexes, by Principal Component Analysis (PCA) and Factorial Analysis, based on some of the main factors influencing erosion processes that occur in the urban area of the municipality. At this stage the following factors were considered: soil texture; slope; permeability; use and occupation; rainfall; and soil erodibility. Initially, the erosive features existing in the urban area were surveyed, spatialized and classified. Among the 9 spatialized features, two were from fluvial processes, two were recovered, leaving five linear erosive features to be studied. Comparação da acurácia diagnóstica de índices dinâmicos e estáticos de pré-carga para predição da fluido-responsividade em cães anestesiados com isoflurano, sob ventilação mecânica

Orientador: Francisco José Teixeira Neto / Resumo: Objetivo: Comparar a acurácia diagnóstica da variação da pressão de pulso (VPP), variação do volume sistólico mensurada através do análise de contorno de pulso (VVSACP), índice de variabilidade pletismográfica (IVP), pressão venosa central (PVC) e índice do volume diastólico final global mensurado pela técnica de termodiluição transpulmonar (GEDVITDTP) para predizer a fluido-responsividade em cães. Animais: Quarenta cadelas saudáveis (13,8–26,8 kg) submetidas a ovário-salpingo-histerectomia. Métodos: A anestesia foi mantida com isoflurano sob ventilação mecânica com volume controlado (volume corrente 12 mL/kg, pausa inspiratória 40%, relação inspiração/expiração: 1:1,5). O débito cardíaco e o volume sistólico foram obtidos pela técnica de termodiluição transpulmonar através de um cateter inserido na artéria femoral. A fluido-responsividade (FR) foi avaliada por uma prova de carga (solução de Ringer com lactato, 20 mL/kg durante 15 minutos), administrada uma vez (n = 21) ou duas vezes (n = 18) antes da cirurgia. Respondedores a volume foram definidos como indivíduos onde o VS mensurado pela técnica de termodiluição transpulmonar, elevou-se acima de 15% após a última prova de carga. Animals: A group of 40 bitches (13.8–26.8 kg) undergoing ovariohysterectomy. Methods: Anesthesia was maintained with isoflurane under volume-controlled ventilation (tidal volume 12 mL/kg; inspiratory pause during 40% of inspiratory time; inspiration: expiration ratio 1:1.5). Transpulmonary thermodilution cardiac output was recorded through a femoral artery catheter. FR was evaluated by a fluid challenge (lactated Ringer's, 20 mL kg over 15 minutes) administered once (n = 21) or twice (n = 18) before surgery. Individuals were responders if stroke volume index measured by transpulmonary thermodilution increased >15% after the last fluid challenge. Orientador: Augusto Batagin Neto / Resumo: Compostos baseados em carbono têm se mostrado materiais de grande interesse tecnológico, principalmente devido à sua alta flexibilidade de síntese, baixo custo relativo e propriedades únicas. Graças a isto, tem-se observado um número crescente de trabalhos teóricos e experimentais acerca da compreensão de características básicas destes sistemas e a proposição de novos compostos com propriedades otimizadas para aplicações específicas. No presente trabalho são apresentados estudos teóricos acerca de dois temas relacionados à reatividade e propriedades estruturais de materiais orgânicos e baseados em carbono: i) estudo e análise de reatividade de subunidades de melanina; e ii) estudo estrutural e de reatividade de nanoflocos (ou nanoflakes em inglês) de carbono para aplicações em sensores químicos. Melaninas são pigmentos naturais com propriedades biológicas e eletrônicas que as tornam promissoras para aplicações bio-eletrônicas. Contudo não há ainda entendimento pleno acerca de sua estrutura macromolecular e conexão entre suas unidades básicas. Neste estudo cálculos de estrutura eletrônica, combinados com análise de reatividade, foram realizados para melhor compreender os processos de oligomerização. (Resumo completo, clicar acesso eletrônico abaixo) / Abstract: Carbon-based materials have been considered compounds of great technological interest, manly due to their flexibility of synthesis, relative low cost and unique opto-electronic properties. A crescent number of theoretical and experimental studies has been reported regarding the comprehension of basic features of these materials and the proposition of new compounds with optimized properties. This thesis presents studies about the reactivity and structural properties of carbon based and organic materials according to two themes: i) reactivity study and oligomerization analysis of melanin; and ii) reactivity and structural analysis of carbon nanoflakes and their application as chemical sensors. Melanins are natural pigments with biological and electrical properties that turn them promising compounds for bioelectronic applications. However, despite of their promising properties, up to date the macromolecular structure of melanin and the connection between its basic units have not been understood in detail. In this study, electronic structure calculations, combined with reactivity analysis were conducted to better understand the oligomerization process of this compound. The obtained results allow us to propose the most probable dimeric structures and identify relevant reactions that occur during melanin oligomerization process. Multidimensional Highway Construction Cost Indexes Using Dynamic Item Basket

A highway construction cost index (HCCI) is an indicator of the purchasing power of a highway agency. Thus, it must reflect the actual construction market conditions. However, current methods used by most state departments of transportation are not robust enough to meet this primary goal due to (1) a significantly insufficient sample size of bid items used in HCCI calculation; and (2) inability to address the need to track highway construction market conditions in specific submarket segments such as, but not limited to, various project types, sizes, and locations. This study proposes an advanced methodology to overcome these apparent limitations using two new concepts: (1) dynamic item basket; and (2) multidimensional HCCIs. The dynamic item basket process identifies and utilizes an optimum amount of bid-item data to calculate HCCIs in order to minimize the potential error due to a small sample size, which leads to a better reflection of the current market conditions. Multidimensional HCCIs dissect the state highway construction market into distinctively smaller sectors of interest and thus, allow state Departments of Transportation to understand the market conditions with much higher granularity. A framework is developed to integrate these two concepts and a standalone prototype system, named the Dyna-Mu-HCCI System, is developed to automate the data-processing part of the framework. The historical bid data of the Montana Department of Transportation are used to evaluate the performance of the Dyna-Mu-HCCI System and measure the effects of the dynamic item basket (DIB) and multidimensional HCCIs. The results show an eightfold increase in terms of the number of bid items used in calculating HCCIs and at least a 20% increase in terms of the total cost of bid items used. In addition, the multidimensional HCCIs reveal different cost-change patterns from different highway sectors. For example, the bridge construction market historically shows a very different trend compared with the overall highway construction market. The new methodology is expected to aid state Departments of Transportation in making more-reliable decisions in preparing business plans and budgets with more accurate and detailed information about the construction market conditions. Further, the prototype Dyna-Mu-HCCI System is expected to significantly facilitate the HCCI calculation process and rapidly implement this new system.
