A. / LIGHT Servi?os de Eletricidade S.A. / The present study looked for alternatives of growth handling and of pruning in plants used in the urban forestation, seeking to the decrease of conflicts of these with electric power spinnings. In the chapter I experiments were accomplished with Flamboyant in soil and simple nutritious solution containing Al, with objective of studying handling alternatives seeking to reduce the growth and development of plantules Flamboyant. The aluminum atrophies roots could affect the growth. Six experiments were accomplished with plantules (soil and simple nutritious solution) in conditions of growth camera and vegetation house, Department of Fitotecnia - UFRRJ. In the soil experiments they were used two sources of sulfate of aluminum, being a commercial one and a pure source analytically, in the concetrations 0, 100, 200, 300 and 400 kg ha-1 in the soils Planossolo and Argissolo, with four repetitions. The experiments accomplished in simple nutritious solution just received calcium (0,001 mM) and they had as source AlCl3 with doses that varied from 0,075 to 5,0 mM. Through the results of the chapter I was ended that in same or superior conditions of soil concentrations to 0,4 Cmolc/dm3 of Al caused toxicity to the plantules of Flamboyant, with effects mainly in the length root, and in solution simple nutritious doses same or upper to 0,075 mM of AlCl3 paralyzed the growth root. Al can be used as inhibitor of vegetable growth in plantules of Flamboyant. The chapter II had as objective studies the effects of the pruning and association with growth inhibitors in arboreal species. Four experiments were accomplished with the plants Sabi?, Sombreiro, and Cucumber (phytotoxicity Evaluation the herbicides). The experiments were installed in the Department of Fitotecnia-UFRRJ, Serop?dica. In the experiment 1 (Sabi?) all the plants were cut off 2 m of height, and they were applied the treatments: control, 2.4-D+Picloram, and Picloram. In the experiment 2 (Sabi?) all the plants were pruned removing 1/3 of the cup and applying the treatments: control, HCl, AlCl3, Picloram and nearby Cut (Without pruning of 1/3 of the cup). The experiment 3 (Sombreiro) had as treatments cut types in relation to the main trunk: nearby, leaving stub of 20 cm and of 40 cm. The experiment 4 (Pepino) it was installed in the originating from soil the first experiment with Sabi?. In the experiment 1, the treatment with Picloram delayed the beginning of the regrowth, important factor for the present study, so that they delay to arrive to the electric power spinning. In the experiment 2, just the treatment Picloram presented length and diameter of sprouts a little smaller in relation to the other treatments. In the experiment 3 it was verified that the nearby cut didn't present new budding until the 145 days after the pruning. In the experiment 4 the cucumber presented more intense toxicity in the treatment with Picloram. Was ended that Picloram and 2,4-D+Picloram were efficient for budding handling in adult plants of Sabi?, because they presented budding with small lengths, delaying the accomplishment of new pruning. In plants of Sombreiro the nearby cut was efficient, not presenting budding. It happened toxicity for Picloram in plants of Pepino. / O presente estudo buscou alternativas de manejo de crescimento e de poda em plantas utilizadas na arboriza??o urbana, visando ? diminui??o de conflitos destas com fia??es de energia el?trica. No cap?tulo I foram realizados experimentos com Flamboyant em solo e solu??o nutritiva simples contendo Al, com objetivo de estudar alternativas de manejo visando diminuir o crescimento e desenvolvimento de pl?ntulas Flamboyant. O alum?nio atrofia ra?zes podendo afetar o crescimento. Foram realizados seis experimentos com pl?ntulas (Solo e solu??o nutritiva simples) em condi??es de c?mara de crescimento e casa de vegeta??o, Departamento de Fitotecnia - UFRRJ. Nos experimentos de solo foram utilizadas duas fontes de sulfato de alum?nio, sendo uma comercial e uma fonte pura analiticamente, nas concentra??es 0, 100, 200, 300 e 400 kg ha-1 nos solos Planossolo e Argissolo, com quatro repeti??es. Os experimentos realizados em solu??o nutritiva simples receberam apenas c?lcio (0,001 mM) e tiveram como fonte AlCl3 com doses que variaram de 0,075 a 5,0 mM. Atrav?s dos resultados do cap?tulo I conclui-se que em condi??es de solo concentra??es iguais ou superiores a 0,4 Cmolc/dm3 de Al causaram toxicidade ?s pl?ntulas de Flamboyant, com efeitos principalmente no comprimento radicular, e em solu??o nutritiva simples doses iguais ou superiores a 0,075 mM de AlCl3 paralisaram o crescimento radicular. O Al pode ser utilizado como inibidor de crescimento vegetal em pl?ntulas de Flamboyant. O cap?tulo II teve como objetivo estudar os efeitos da poda e da sua associa??o com inibidores de crescimento em esp?cies arb?reas. Foram realizados quatro experimentos com as plantas Sabi?, Sombreiro, e Pepino (Avalia??o de fitotoxidade a herbicidas). Os experimentos foram instalados no Departamento de Fitotecnia-UFRRJ, Serop?dica-RJ. No experimento 1 (Sabi?) todas as plantas foram decepadas a 2 m de altura, e foram aplicados os tratamentos: Testemunha; 2,4-D+Picloram; e Picloram. No experimento 2 (Sabi?) todas as plantas foram podadas retirando 1/3 da copa e aplicando os tratamentos: Testemunha, HCl, AlCl3, Picloram e Corte rente (Sem poda de 1/3 da copa). O experimento 3 (Sombreiro) teve como tratamentos tipos de corte em rela??o ao tronco principal: rente, deixando toco de 20 cm e de 40 cm. O experimento 4 (Pepino) foi instalado no solo proveniente do primeiro experimento com Sabi?. No experimento 1, o tratamento com Picloram atrasou o in?cio da rebrota, fator importante para o presente estudo, a fim de que demorem chegar ? fia??o de energia el?trica. No experimento 2, apenas o tratamento Picloram apresentou comprimento e di?metro de brotos um pouco menores em rela??o aos demais tratamentos. No experimento 3 foi verificado que o corte rente n?o apresentou novas brota??es at? os 145 dias ap?s a poda. No experimento 4 o pepino apresentou toxidez mais intensa no tratamento com Picloram. Conclui-se que o Picloram e 2,4-D+Picloram foram eficientes para manejo de brota??es em plantas adultas de Sabi?, pois apresentaram brota??es com comprimentos pequenos, atrasando a realiza??o de nova poda. Em plantas de Sombreiro o corte rente foi eficiente, n?o apresentando brota??es. Ocorreu toxidez por Picloram em plantas de Pepino

Tacrolimus (TAC) é um fármaco imunossupressor muito utilizado na prevenção de rejeição aguda após o transplante de órgãos. Essa droga apresenta um índice terapêutico muito baixo e grande variabilidade intra e interindividual, sendo necessário programas de monitorização terapêutica para se otimizar a eficácia e limitar a toxicidade. O TAC é um fármaco substrato do CYP3A5 e transportado pela proteína de efluxo PGP e acredita-se que o polimorfismos genéticos (SNPs) destas proteínas estejam relacionados a alta variabilidade farmacocinética desta droga. Neste estudo, investigamos a influência dos polimorfismos destas proteínas sobre alguns parâmetros farmacocinéticos do TAC e também, na incidência de lesões renais e rejeição em receptores de transplante renal. Pacientes recebendo TAC a no mínimo 12 meses (n=108) foram genotipados (PCR real time) para os polimorfismos do CYP3A5*3 (rs776746) e do gene ABCB1 1236C>T (rs1128503), 2677G>T/A (rs2032582) e 3435C>T (rs1045642). Dados da concentração plasmática de vale (Co; ng/mL), dose diária normalizada (mg/dia por Kg do paciente) e a concentração plasmática do fármaco normalizada pela dose ingerida (Co/dose, ng/mL por mg/dia por kg do paciente) de TAC foram obtidos dos prontuários médicos ao longo de três anos após o transplante renal. O desfecho clínico foi analisado avaliando-se a curva de sobrevida o enxerto, a função renal obtida pelo clearance de creatinina (Equação de Cockroft-Gault), e desenvolvimento de lesões renais e rejeição aguda e crônica com diagnósticos estabelecidos mediante suspeita clínica e confirmados pela avaliação das biópsias quanto a presença de necrose tubular aguda (NTA) e nefropatia crônica do enxerto (NFC) de acordo com a classificação de BANFF 07. Os haplótipos do gene ABCB1 foram inferidos estatisticamente utilizando o software PHASE (version 2.1). Diferenças foram consideradas significativas quando p<0,05. Não observamos desvios em relação ao esperado pelo equilíbrio de Hardy-Weinberg em nossa população para nenhum dos genes estudados. Frequências alélicas destes polimorfismos (6986G 74%; 1236C 60%; 3435C 59% and 2677G 64%) e haplótipos (49% 2677G-3435C-1236C e 31% 2677T-3435T-1236T) foram consistentes com outros estudos realizados na população brasileira. Indivíduos portadores de ao menos um alelo *1 do gene CYP3A5 necessitavam de maiores doses de TAC para obter níveis plasmáticos semelhantes aos dos indivíduos homozigotos para o alelo *3 (0,09 ± 0,03 vs. 0,06 ± 0,03; mg/dia/kg; p<0,001). Ao final do primeiro ano de transplante pacientes CYP*3/*3 apresentavam praticamente o dobro da razão Co/dose quando comparados com os pacientes CYP*1/*1 (144,60 ± 67,29 vs. 70,44 ± 56,05 ng*mL-1 /mg*kg- 1 /dia; p<0,001). Observou-se semelhante em relação quando indivíduos portadores dos alelos e haplótipo variante da PGP foram avaliados. Não encontramos associações significativas entre os genótipos e a sobrevida do enxerto ou clearance de creatinina. No entanto, observamos que os pacientes portadores do haplótipo GCC apresentaram maior incidência de desenvolvimento de NFC. Este estudo confirma o efeito dos polimorfismos do CYP3A5 e, em menor grau da PGP na farmacocinética do TAC. No entanto, não encontramos associações entre esses polimorfismos e desfechos clínicos significantes, sugerindo que a genotipagem para o CYP3A5 ou ABCB1 ainda não deva ser incorporada na prática clínica como uma ferramenta para o manejo de transplante renal. / Tacrolimus (TAC) is widely used to prevent acute rejection following solid-organ transplantation. This drug is characterized by a narrow therapeutic index and drug monitoring programs are required both to optimize efficacy and to limit toxicity. TAC is known to be substrate of cytochrome P450 (CYP) 3A5 and P-glycoprotein (PGP/ABCB) and its been suggested that genetic polymorphisms (SNPs) of these proteins are highly associated with variations in TAC pharmacokinetics. We investigated the influence of polymorphisms of CYP3A5 and ABCB1 gene on the pharmacokinetic parameters (PK) of TAC and on the incidence of kidney injuries and allograft rejection (AR) in renal transplant recipients. Patients receiving TAC for at least 12 months (n=108) were genotyped (real-time PCR) for CYP3A5*3 (rs776746) and for ABCB1 1236C>T (rs1128503), 2677G>T/A (rs2032582) and 3435C>T (rs1045642) polymorphisms. TAC predose concentration (Co; ng/mL), TAC daily dose (mg/day per kg body weight) and dose-normalized predose concentrations (Co/dose; ng/mL per mg/day per kg body weight) were retrieved from medical records up to 03 years after transplantation. Clinical outcomes were analyzed evaluating renal function in terms of creatinine clearance ( Cockroft-Gault equation) and allograft survival. Kidney injuries and AR diagnostics were established by clinical suspicion and in presence of histological findings in renal biopsies according to the 2007 Banff classification. ABCB1 gene haplotypes were statistically inferred using PHASE software (version 2.1). No deviation from Hardy-Weinberg equilibrium was observed in our study population for the polymorphic loci examined in CYP3A5 and ABCB1. Allelic frequencies of these polymorphisms (6986G 74%; 1236C 60%; 3435C 60% and 2677G 65%) and haplotypes (49% 2677G-3435C-1236C and 30% 2677T- 3435T-1236T) were consistent with other studies in the Brazilian population. Individuals carrying at least one CYP3A5*1 allele required higher TAC dose to achieve similar TAC blood levels as the homozygous individuals for the *3 allele (0.09 ± 0.03 vs.0.06 ± 0.03, mg/day per kg body weight, p<0.001). The presence of the CYP3A5*1 allele was also associated with lower TAC Co/dose compared to CYP*3 homozygous (84.9 ± 43.2 vs. 144.6 ± 66.7 ng/mL per mg/day per kg body weight, p<0.001). Regarding ABCB1 polymorphisms, individuals homozygous for the variant allele of each individual SNP and for the haplotype (TTT) showed higher Co/dose ratio. No associations were found between SNPs or haplotypes and allograft survival or creatinine clearance. We did find, though, that patients carrying GCC haplotype had a higher incidence of chronic rejection. Our findings confirm the effect of CYP3A5 and, less pronounced of ABCB1 polymorphisms, on the TAC pharmacokinetic. On the other hand, we did not find any association between these polymorphisms and relevant clinical outcomes, suggesting that CYP3A5 and ABCB1 genotyping must not be incorporated as a useful clinical tool on the management of kidney transplantation.

The development of new solid forms of active pharmaceutical ingredients (API) is relevant both from fundamental as well as industrial perspectives. To this end, Crystal Engineering plays an ever-increasing important role in pharmaceutical sciences. Among the crystal engineering strategy, salt formation is the most important and implemented approach. The salt forms of API could be used to modulate and tuned the solubility and stability of API to provide optimal practical uses. Herein, we report pharmaceutical salts of two Selective Serotonin Reuptake Inhibitor antidepressants used in the treatment of depression and anxiety disorders, Paroxetine (PRX) and Fluoxetine (FLX). For this purpose, salt formers, supramolecular synthesis and crystallization protocols have been driven by the systematization of structural and supramolecular data of molecules and analogues from the Cambridge Structural Database. Paroxetine bromide hemihydrate ((PRXBr)0.5H2O), Paroxetine Nitrate hydrate (PRXNO3H2O) and two polymorphs of Fluoxetine Nitrate (FLXNO3) have obtained. All were characterized by a combination of techniques including Single Crystal X-ray Diffraction, Differential Scanning Calorimetry (DSC), Thermogravimetry analysis (TGA), Hot Stage Microscopy, Fourier transform infrared spectroscopy (IR) and solubility measurements. Since the hydration/dehydration process in APIs induces phase transitions that compromise its efficiency, the structural characterization of (PRXBr)0.5H2O help to understand its reversible dehydration process. Also, this study has implication in the understating of dehydration of isostructural PRX hydrochloride salt. Additionally, the (PRXNO3)H2O have shown the conformational flexibility and supramolecular diversity of PRX. On the other hand, the chirality of FLX is related to two nitrate salt polymorphs. A racemate and a non-centrosymmetric structure with independent enantiomers in the asymmetric unit were obtained for FLXNO3. Their packing have shown the existence of different racemic motifs, resulting in different enantiomer orientations The rare occurrence of racemic systems in non-centrosymmetric space groups becomes this event a noteworthy case. By their physicochemical properties, the polymorphs were monotropically related. The scientific contributions of this thesis show the diversity of the solid forms and define candidates to new antidepressants APIs solid formulations. / O desenvolvimento de novas formas sólidas de ingredientes farmacêuticos ativos (API) é relevante tanto numa perspectiva fundamental como industrial. Para tal, a Engenharia de cristais tem desempenhado um papel importante nas ciências farmacêuticas. Dentre as estratégias, a formação de sais é a abordagem mais importante e implementada. Os sais de APIs são capazes de modular e ajustar a solubilidade e a estabilidade, a fim de proporcionar uso prático. Nesta tese, são reportados sais de dois fármacos Inibidores Seletivos de Recaptação de Serotonina, consolidados no tratamento da depressão e distúrbios de ansiedade, a Paroxetina (PRX) e a Fluoxetina (FLX). Brometo de Paroxetina hemiidratado ((PRXBr)0.5H2O), Nitrato de Paroxetina hidratado (PRXNO3H2O) e polimorfos de Nitrato de Fluoxetina (FLXNO3), síntese e protocolos de cristalização foram cuidadosamente delineados, com base na sistematização de dados estruturais e supramoleculares das moléculas e seus análogos, depositados no Cambridge Structural Database. Todos os sais foram caracterizados por Difração de Raios-X por Monocristal, Calorimetria Explanatória Diferencial (DSC), Análise termogravimétrica (TGA), Termomicroscopia, Espectroscopia vibracional na região do infravermelho (IR) e solubilidade. Considerando que a hidratação/desidratação induz mudanças de fases que comprometem a eficiência do API, a caracterização do (PRXBr)0.5H2O auxiliou no entendimento do processo de desidratação reversível que ocorre para esse fármaco. Estas mudanças de fase resultam também em implicações sobre a compreensão do processo de desidratação do sal isoestrutural de cloreto de PRX hemiidratado. Além disso, por meio da elucidação estrutural do (PRXNO3)H2O, foi possível analisar a diversidade conformacional e supramolecular da PRX. Quanto à FLX, verificou-se que sua quiralidade está relacionada com seu polimorfismo. Um racemato e uma estrutura não centrossimétrica com dois enatiômeros independentes na unidade assimétrica foram obtidos para o FLXNO3. A comparação destas estruturas permitiu mostrar a existência de arranjos supramoleculares racêmicos, constituídos por diferentes orientações de enatiômeros. A rara ocorrência de sistemas racêmicos em grupos espaciais não-centrossimétricos tornou este evento um caso notável. A partir das propriedades físico-químicas, os polimorfos puderam ser monotropicamente relacionados. Os resultados desta tese trazem importantes contribuições científicas para diversidade de formas sólidas e também define novas formulações sólidas para utilização como antidepressivos.

Résistance acquise chez les Burkholderia pseudomallei : analyse de l'expression de l'efflux et de son inhibition / Acquired resistance in Burkholderia pseudomallei : analysis of efflux expression and inhibitionSchnetterle, Marine 03 December 2018 (has links)
Burkholderia pseudomallei est l’agent causal de la mélioïdose, une maladie tropicale endémique dans le Nord de l’Australie et en Asie du Sud-Est. Nous avons analysé le système d’efflux, mécanisme majoritairement impliqué la multi-résistance aux antibiotiques. Nous avons chercher à identifier des mutations dans les pompes d’efflux et des modulation de l’expression de ces dernières afin d’expliquer ces phénotypes de résistance. Les techniques de séquençage de l’ADN et de transcriptomique par RT-qPCR nous ont permis d’identifier deux mécanismes chez des souches cliniques. Un mécanisme transitoire responsable d'une résistance croisée du Cotrimoxazole, avec les quinolones, et le chloramphénicol, pour lequel nous suspectons une modulation de l’expression de l’efflux. Le second, impliqué dans la résistance au méropénème, par surexpression de l'efflux suite à une mutation dans le régulateur de la pompe AmrAB-OprA.Dans un second axe de recherche, nous avons également criblé plusieurs molécules afin d'identifier des candidats inhibiteurs de l'efflux, dérivant de la famille des phénothiazines et capables de restaurer une sensibilité aux antibiotiques. Nous avons analysé l’impact de ces molécules sur des souches modèles de multi-résistance (Burkholderia thailandensis) et sur des souches cliniques et environnementales de B. pseudomallei. Ces molécules sont capables d’impacter l’expression de l’efflux, mais nous pensons que le mécanisme majeur d’inhibition de cette famille de molécules reste l’entrée en compétition avec les antibiotiques efflués. Nous avons identifié une molécule, AST17, capable de restaurer la sensibilité au Cotrimoxazole, ainsi qu'aux quinolones. / Burkholderia pseudomallei is thecausal agent of melioidosis, a tropical disease, endemic in Notrhern Australia and South-East Asia. We have analyzed efflux systeme, known to be one of the main mecanism implicated in antibiotic resistance phenotypes. We have looked for mutations in efflux pumps and for transient modulations of the efflux pumps expression, that could explain resistance phenotypes. Whole genome sequencing and a the targeted method of RT-qPCR allowed us to identified two mecanisms in clinical strains. A transient mecanism, responsible of a cross-resistance to Cotrimoxazole, quinolones and chloramphenicol, and we suspect an implication of modulation of efflux. The second one is implicated in meropenem resistance by an overexpression of the AmrAB-OprA efflux pumps, due to a mutation of its regulator. In a second time, we also have screened several compounds, all derivated from phenothiazines, in order to identify efflux pump inhibitors for a restoration of the antibiotic susceptibility. We have analyzed the impact of these molecules in multi-resistant strain models, and on several clinical and environnemental strains. These molecules are able to modulate efflux pumps expression, however, we think that the main inhibition mecanism of these derivatives is about a competition between the molecule and the antibiotics. We have identified one molecule, AST17, that is able to restore Cotrimoxazole and quinolones susceptibilities.

Weltweit zählt die Tuberkulose zu den tödlichsten und am weitesten verbreiteten Infektionskrankheiten. Missstände in der ohnehin komplexen Therapie einerseits und fehlende Entwicklung neuartiger adäquater Wirkstoffe andererseits, führten zur Entstehung von multi- und sogar total-resistenten Keimen. Der Haupterreger ist das Mycobacterium tuberculosis. Charakteristisch für Mykobakterien ist eine dicke und undurchlässige wachsartige Zellwand mit einem großen Anteil an bestimmten Fettsäuren. Die mykobakterielle Biosynthese dieser Fettsäuren unterscheidet sich stark von eukaryotischen Zellen. Die selektive Beeinflussung dieses Systems führt zu nicht überlebensfähigen Mykobakterien und stellt somit ein idealer Angriffspunkt für Arzneistoffe dar.
Die vorliegende Arbeit befasst sich mit der Entwicklung neuartiger direkter Hemmstoffe von InhA, einem für den Zellwandaufbau des Mycobacterium tuberculosis essenziellem Enzym.
Es wurden zwei photometrische gekoppelt-enzymatische Assay-Systeme im 96-Well-Format entwickelt, die sich das Absorptions- bzw. Fluoreszenzverhalten des Coenzyms NADH zu Nutze machen.
Das hierzu benötigte Enzym InhA wurde überexprimiert und aufgereinigt. Mehrere Synthesemethoden für das im Testverfahren verwendete Substrat 2-trans-Octenoyl-CoA (2toCoA) wurden etabliert.
Die etablierten Assay-Systeme wurden mit Hilfe von Positivkontrollen validiert. Grundlegende Experimente zur Errichtung einer substratunabhängigen orthogonalen Methode mittels MST wurden getätigt.
Basierend auf den Ergebnissen eines in Vorarbeiten durchgeführten virtuellen Screenings wurden erste potenzielle Inhibitoren kommerziell erworben und getestet. Nachfolgend wurde mit der Synthese von Derivaten begonnen, welche auf iterativem Wege optimiert wurden (Testung – Docking – Synthese neuer Derivate). Hierdurch wurde eine umfassende Substanzbibliothek bestehend aus insgesamt 254 Verbindungen aufgebaut. Diese setzte sich aus unterschiedlich substituierten Thiazolidin-2,4-dionen- und Thiazolin-2-on-Derivaten, Derivaten der ähnlich strukturierten Fünfring-Heterozyklen Rhodanine, Thiohydantoine und Hydantoine und weiteren Strukturklassen bestehend aus Biphenylether-, Pyrrolidoncarboxamid-, Pyridon- und Sulfonamid-Derivaten zusammen. Die Verbindungen wurden entweder selbst synthetisiert, kommerziell erworben oder von Kooperationspartnern bezogen. Neben der Etablierung zuverlässiger und effizienter Syntheserouten stand hierbei ebenso die strukturelle Aufklärung der stereochemischen Verhältnisse der Produkte im Mittelpunkt.
Die Verbindungen der aufgebauten Substanzbibliothek wurden mit dem etablierten InhA-Testsystem auf ihre inhibitorischen Eigenschaften gegenüber InhA untersucht. Soweit möglich wurden Struktur-Aktivitätsbeziehungen abgeleitet. Insbesondere einige disubstituierte Thiazolidindione zeigten eine schwache Hemmung von bis zu 25 %. Die zur Aufklärung des Inhibitionsmechanismus durchgeführten Experimente deuten auf eine unkompetitive Hemmung hin. Bei den direkten Testungen an Mykobakterien konnten die inhibitorischen Eigenschaften hingegen nicht bestätigt werden.
Weiterhin wurden Testungen an Cystein- und Serin-Proteasen von Erregern anderer Infektionskrankheiten durchgeführt. Das Thiazolinon SV102 wurde hierbei als nicht-kompetitiver Hemmstoff von Cathepsin B mit einem Ki-Wert von 1.3 µM identifiziert. Die Synthese und Testung weiterer Thiazolin-2-on-Derivate sowie Cokristallisationsversuche mit Cathepsin B sind somit in Betracht zu ziehen. Die getesteten Thiazolidindion-Derivate der Substanzbibliothek zeigten hierbei mittelstarke bis gute Hemmeigenschaften, die ebenfalls an den Erregern beobachtbar waren. Relativiert werden diese vielversprechenden Ergebnisse allerdings durch eine ebenfalls zu beobachtende Zytotoxizität. Weiterhin konnte eine antibakterielle Wirkung der untersuchten Verbindungen in zellulären Assay-Systemen nicht gezeigt werden.
Abschließend wurde die Eignung der Thiazolidindione und verwandter Fünfringheterozyklen als Leitstruktur für potenzielle InhA-Inhibitoren, aber auch die Eignung dieser Verbindungsklasse als potenzielle Leitstruktur per se diskutiert. / Tuberculosis is one of the most deadly infectious diseases and it is highly prevalent world-wide. The issues arising from the complexity of the current treatments schemes as well as the lacking development of effective new drugs have led to the formation of multi- or even totally drug-resistant strains of Mycobacterium tuberculosis which is known as the major microbial species causing tuberculosis.
Mycobacteria are characterized by a unique, thick and waxy cell wall that functions as a nearly impermeable barrier due to its high concentration of mycolic acids. The biosynthesis of these fatty acids requires the presence of a specific set of mycobacterial enzymes that differ markedly from their eukaryotic counterparts. Disturbance in the proper formation of this essential cell wall unvariably interferes with mycobacterial survival. Thus, the mycobacterial fatty acid synthesis pathway is an attractive target for the development of selective new drugs against Mycobacterium tuberculosis.
The aim of this work was the synthesis and optimization of thiazolidindiones and related five membered heterocycles as lead structures for the development of novel, direct inhibitors of InhA, an essential enzyme in the biosynthesis of mycolic acids.
Two coupled photometric enzyme assays that monitor the absorption of the involved cofactor NADH were developed in a 96-well-plate format. For this purpose, the enzyme InhA was recombinantly expressed and purified from E.coli. Several routes of synthesis for its substrate 2-trans-octenoyl-CoA were established.
Assay systems were validated by characterizing positive controls known from the literature, and an orthogonal analysis method was introduced by using microscale thermophoresis.
Thiazolidindiones as lead compound structure were discovered by performing a virtual screening campaign in preliminary works. Several substances were commercially acquired and tested in the established InhA-assay-system. Based on these results the syntheses of further compounds were started and optimized in an iterative manner (testing – docking – synthesis of new derivatives). Thus, a large compound library of 254 substances was built up. It consists of different substituted thiazolidindiones, thiazolinons and related five membered heterocycles such as rhodanines, thiohydantoines and hydantoines as well as further compound classes, namely, derivatives of biphenylethers, pyrrolidoncarboxamides, pyridines and sulfonamides. The compounds were either synthesized, received from collaboration partners, or acquired commercially. Concerning the synthetic work, the focus was on developing effective routes of synthesis, elucidating reaction mechanisms and determining the stereochemical properties of the received products.
The compound library was subsequently tested against InhA by using the previously established assay systems. As far as possible, structure-activity relationships were derived. In particular, some disubstitued thiazolidindiones showed moderate inhibitory properties of up to 25 % when tested against the purified enzyme. Kinetic experiments performed to obtain information about the mode of inhibition indicated that thiazolidinediones acted as uncompetitive inhibitors of InhA. However, these results could not be confirmed in direct measurements using mycobacteria.
Further measurements against various cysteine- and serine-proteases were performed. The thiazolinone SV102 was identified as non-competitive inhibitor of cathepsin B (Ki = 1.3 µM). Consequently, synthesis of new derivates as well as co-crystallization experiments should be taken into consideration. Thiazolidinedione derivatives also showed proper inhibition of isolated proteases. This inhibitory activity also was also observed in direct measurements against trypanosoma and leishmania but was actually accompanied by a certain extent of cytotoxicity.
Finally, the question was addressed of whether thiazolidindiones and related five membered heterocycles should be seen as a privileged scaffold in drug development, or just as promiscuous binders that should be excluded from drug discovery.

Small molecules regulated bone resorption and enzyme activity in osseous cells / Petites molécules régulant la résorption osseuse et l’activité enzymatique dans les cellules osseusesRen, Zhongyuan 05 December 2014 (has links)
La Cathepsine K est parmi la plus efficace des collagénases de mammifère pour cliver la triple hélice de collagène de type-1. Nous avons développé une série d'azanitriles, (CKI-8 and CKI-13) inhibiteurs de cathepsine K. CKI-8 (un isomère de CKI-13) et CKI-13 ne sont pas toxiques sur les osteoblastes Saos-2 et les cellules RAW 264.7 jusqu' à une concentration de 1000 nM, tandis qu'ils ne le sont pas jusqu'à une concentration de 100 nM sur les osteoclastes. CKI-8 n'affecte pas l'activité de la phosphatase alkaline ainsi que la minéralisation induite par les Saos-2 et par les osteoblastes primaires. CKI-13 diminue de 35 % la minéralisation induite par les Saos-2 tandis qu'il n'affecte pas la minéralisation induite par les osteoblastes primaires. L'addition de CKI-13 diminue l'activité de la phosphatase alkaline d'environ 20% (Saos-2) et de 40 % (osteoblastes primaires). La résorption osseuse sur des tranches d'os d'origine bovine est diminuée avec 10 nM de CKI-13, 100 nM de CKI- 8 et 100 nM d'inhibiteur commercial E64. CKI-8 et CKI-13 diminuent la mobilité des osteoclastes. Nous avons développé un dosage d'hydrolyse de PPi par la phosphatase alkaline au moyen de l'IR, ayant l'avantage de fonctionner sur des vésicules matricielles et des cellules avec des substrats naturels à un pH physiologique. La bande de PPi localisée à 1107 cm-1 (∑= 2158 ± 211 M-1.cm-1) et celles de Pi localisées à 1076 cm-1 (∑= 1346 ± 116 M-1.cm- 1) et à 991 cm-1 (∑= 493 ± 49 M-1.cm-1) ont servis à mesurer les concentrations du substrat et du produit / Cathepsin K is among the most potent mammalian collagenase, capable of cleaving the triple helix in type-I collagen. We developed a series of azanitriles (CKI-8 and CKI-13) which are inhibitors of cathepsin K. CKI-8 (an isomer of CKI-13) and CKI-13 did not induce significant toxicity on osteoblasts Saos-2 and RAW 264.7 cells up to 1000 nM, while they were not toxic on mature osteoclasts up to 100 nM. Commercial E64 inhibitor was not toxic in primary osteoclast cells up to 1000 nM. CKI-8 did not affect alkaline phosphatase activity as well the mineralization induced by Saos-2 cells and by primary osteoblasts. CKI-13 decreased by 35% the mineralization induced by Saos-2 cells while it did not on mineralization induced by primary osteoblasts. Addition of CKI-13 decreased alkaline phosphatase activity by around 20% (Saos-2 cells) and 45% (primary osteoblasts). Bone resorption on bovine slices decreased significantly with 10 nM of CKI-13, with 100 nM of CKI-8 and commercial inhibitor E64. Our findings indicated that CKI-8 and CKI-13 inhibited bone resorption and affected the mobility of osteoclast. To monitor directly the PPi hydrolytic activity by alkaline phosphatase, we developed an infrared (IR) assay taking the advantage to use natural substrate under physiological pH in matrix vesicles and in living cells. Synthesis and functionalization of ring-fused 2-pyridones : Targeting pili formation in <i>E. coli</i>Pemberton, Nils January 2007 (has links)
<p>Bicyclic dihydrothiazolo fused 2-pyridones have been studied as a new class of antibacterial agents, termed pilicides, that target the formation of adhesive bacterial surface organelles (pili) in pathogenic bacteria. Synthetic methods to further functionalize the bicyclic 2-pyridone scaffold have been developed in order to increase water-solubility and thereby facilitate biological evalua-tions. This was accomplished by introducing aminomethylenes at the open position C-6. Tertiary amines were introduced via a microwave–assisted Mannich reaction and a synthetic route based on a formyl intermediate gave access to primary, secondary and tertiary amines, but also to other interesting functionalities. Biological evaluation confirmed that several of the function-alized compounds inhibited pili formation in uropathogenic <i>E. coli</i>., as dem-onstrated by assays of hemagglutination, biofilm formation and adherence to bladder cells. Co-crystallizing one of the pilicides with the target protein gave information about the binding site and based on this a mechanism of action was proposed, which was supported experimentally by surface plas-mon resonance and single point mutations in the protein.</p><p>Furthermore, the previously developed acylketene imine reaction used to prepare bicyclic thiazolo fused 2-pyridone pilicides has been developed to allow preparation of other ring-fused 2-pyridone systems. Benzo[a]quinolizine-4-ones and indolo[2,3-a]quinolizine-4-ones could be prepared in a fast and simple manner starting from dihydroisoquinolines and a β-carboline. Finally, this method could also be applied for the preparation of heteroatom analogs of the previously studied sulfur containing pilicides. Biological evaluations established that the sulfur atom can be replaced by oxygen and still maintain the ability to prevent pili assembly.</p>

<p>This thesis describes the design and synthesis of compounds that are</p><p>intended to inhibit serine and aspartic proteases. The first part of the text deals with preparation of inhibitors of the hepatitis C virus (HCV) NS3 serine protease. Hepatitis C is predominantly a chronic disease that afflicts about 170 million people worldwide. The NS3 protease, encoded by HCV, is essential for replication of the virus and has become one of the main targets when developing drugs to fight HCV. The inhibitors discussed here constitute surrogates for the widely used <em>N</em>-acyl-hydroxyproline isostere designated 4-hydroxy-cyclopentene. The stereochemistry of the 4-hydroxy-cyclopentene scaffold was determined by nuclear overhauser effect spectroscopy (NOESY) and the regiochemistry by heteronuclear multiple bond correlation (HMBC). The scaffold was decorated with different substituents to obtain both linear and macrocyclic HCV NS3 protease inhibitors that display low nanomolar activity. The second part of the thesis describes the design and synthesis of potential aspartic protease inhibitors. The hydroxyethylene motif was used as a noncleavable transition state isostere. The synthetic route yielded a pivotal intermediate with excellent stereochemical control, which was corroborated by NOESY experiments.
