No hospedeiro vertebrado estes parasitos infectam principalmente os macrófagos, onde conseguem escapar dos mecanismos de defesa e se proliferam. Muitos trabalhos relatam que a sinalização purinérgica é modulada durante o desenvolvimento e ativação das células do sistema imune. Para obterem sucesso na infecção, vários patógenos conseguem subverter a resposta imune através da modulação da sinalização purinérgica, promovendo uma diminuição dos níveis de ATP (molécula pró-inflamatória) e levando a um aumento dos níveis de adenosina (molécula imunossupressora). Este desfecho resulta da atividade conjunta de algumas ecto- nucleotidases, das E-NTPDases e da 5’-ecto-nucleotidase, tema que é abordado no segundo capítulo desta tese. Em Leishmania, as E-NTPDases são consideradas importantes porque participam da via de salvação de purinas, além de serem citadas em diversos artigos como responsáveis por aumento de virulência e infectividade em diferentes espécies. Em trabalhos desenvolvidos por nosso grupo de pesquisa, as E- NTPDases 1 e 2 de L. infantum foram caracterizadas em sistema bacteriano, demonstrando resultados promissores. A possibilidade de expressar essas proteínas em sistema de expressão Leishmania (Leishmania Expression System – LEXSY – Jena Bioscience) nos impulsionou a desenvolver o presente projeto, com o intuito de caracterizar essas enzimas em um organismo bem mais próximo ao original e avaliar o seu potencial uso como vacina viva. Portanto, a clonagem das Lic-E-NTPDases sistema LEXSY é o tema abordado no primeiro capítulo desta tese. / Leishmaniasis comprises a broad spectrum of parasitic diseases caused by protozoa of the genus Leishmania. In vertebrate host, these parasites mainly infect macrophages, where they can escape of defense mechanisms and proliferate. Several works have reported that the purinergic signaling is modulated during development and activation of immune cells. To achieve successful infection, many pathogens can subvert the immune response through modulation of purinergic signaling by promoting a reduction in ATP levels (pro- inflammatory molecule) and leading to increased adenosine levels (immunosuppressive molecule). This outcome results from the joint activity of some ecto-nucleotidases, the E- NTPDase and 5'-ecto-nucleotidase, a topic that is discussed in the second chapter of this thesis. In Leishmania, the E-NTPDases are considered important because they participate in purine salvage pathway, and are cited in various articles to be responsible for increased infectivity and virulence in different species. In work carried out by our research group, the E-NTPDase 1 and 2 of L. infantum were characterized in bacterial system, demonstrating promising results. The ability to express these proteins in Leishmania expression system (Leishmania Expression System - LEXSY - Jena Bioscience) prompted us to develop this project, in order to characterize these enzymes in an organism similar to the original and evaluate its potential use as a live vaccine. Therefore, the cloning of Lic-E-NTPDases in LEXSY system is the subject discussed in the first chapter of this thesis.

Variação intraespecífica e biogeografia de isolados brasileiros de Leishmania infantum chagasi baseado em genes nucleares e mitocondriais / Intraspecific variation and biogeography of Brazilian isolates of Leishmania infantum chagasi based on nuclear and mitochondrial genes

As espécies do gênero Leishmania parasitam mamíferos do Novo e Velho Mundo e possuem ciclos de vida com alternância entre vertebrados e invertebrados. A maioria das espécies se desenvolve em artrópodes hematófagos, que podem pertencer a diversas ordens e famílias. A Leishmaniose visceral é uma importante zoonose e possui canídeos silvestres e domésticos como importantes reservatórios conhecidos na diversidade genética de Leishmania infantum chagasi no Brasil ainda não é conhecida. Leishmaniose é uma doença severa com ampla distribuição geográfica com uma incidência de dois milhões de casos por ano e 350 milhões de pessoas em áreas de risco de infecção. O objetivo deste projeto foi avaliar a variação intraespecífica e biogeografia de isolados brasileiros de Leishmania infatum chagasi baseados em genes nucleares e mitocondriais. Os marcadores SSUrDNA, gGAPDH, Citocromo b, Hsp 70, Quitinase e ITS1rDNA foram amplificados, sequenciados e comparados filogeneticamente pelos métodos de parcimônia e bayesiana através de análises concatenadas. As sequencias obtidas apresentaram variabilidade entre 0 e 1% e a topologia gerada não evidenciou diferenças intraespecíficas entre os isolados brasileiros de Leishmania infantum chagasi e consequentemente padrões biogeográficos. Apesar dos resultados obtidos não refletirem a variabilidade esperada, as diversas sequencias obtidas darão subsídios para a determinação e padronização de novos testes diagnósticos da Leishmaniose visceral canina. / The Leishmania species parasite mammals in the New World and have life cycles with alternating between vertebrates and invertebrates. Most species develops in blood-sucking arthropods, which may belong to different orders and families. Visceral leishmaniasis is an important zoonosis and has wild and domestic canids as important known reservoirs and genetic diversity of Leishmania infantum chagasi in Brazil is not yet known. Leishmaniasis is a severe disease with wide geographical distribution with an incidence of two million cases per year and 350 million people at risk in endemic areas. The aim of this study was to evaluate the intraspecific variation and biogeography based on nuclear and mitochondrial genes. The SSUrDNA markers, gGAPDH, cytochrome b, hsp 70, chitinase and ITS1rDNA were amplified, sequenced and compared phylogenetically by maximum parsimony and Bayesian methods in concatenated analysis. The variability of sequences was 0 to 1%, and the topology generated showed no intraspecific differences between Brazilian strains of Leishmania infantum chagasi and consequently biogeographic patterns. Although the results do not reflect the expected variability, the various sequences obtained will provide subsidies for the establishment and standardization of new diagnostic tests of canine visceral leishmaniasis.

Papel de linfócitos Th17 durante a infecção experimental por Leishmania infantum/chagasi / Role of Th17 lymphocytes during Leishmania infantum/chagasi infection

Leishmaniose visceral (LV) é uma doença crônica e potencialmente fatal causada pelas espécies Leishmania infantum/chagasi e Leishmania donovani. O desenvolvimento da resposta Th1 é classicamente associado à proteção contra esses parasitos, mas também há uma correlação positiva entre a produção de citocinas relacionadas com o padrão Th17 e a proteção contra LV por L. donovani em seres humanos. No entanto, a participação de Th17 durante a infecção por L. infantum/chagasi permanece desconhecida. O objetivo desse estudo foi avaliar a participação de Th17 e citocinas relacionadas, além do mecanismo pelo qual tais células operam durante a resposta imune do hospedeiro contra L. infantum/chagasi. Os resultados mostraram que o parasito é capaz de induzir grandes quantidades de TGF-, IL-1, IL-6 e IL-23 por células dendríticas derivadas da medula óssea (BMDC), citocinas envolvidas na indução e/ou manutenção do perfil Th17. Assim, co-cultivando células do baço de camundongos C57BL/6 naves com BMDCs infectadas com L. infantum/chagasi observou-se uma significativa produção de IL-17 por células T. Esses achados foram confirmados por experimentos in vivo onde se constatou a produção de IL-17 no fígado e no baço de camundongos WT infectados, sendo o pico de produção dessa citocina observado na 4ª e 6ª semanas após a infecção. O padrão de resposta do tipo Th17 é crítica para a imunidade protetora contra L. infantum/chagasi, uma vez que camudongos IL-17R-/-, IL-23p19-/- e IL-6-/- mostraram aumento da carga parasitária nos órgãos alvo da infecção, sendo que a susceptibilidade observada em camundongos IL-17R-/- foi associada com o aumento da produção de IL-10 por linfócitos, sugerindo que a IL-17 regula negativamente a produção de IL-10 levando ao controle da infecção causada pelo parasito. Ainda, a ausência da sinalização via IL-17R gerou uma diminuição da inflamação hepática, decorrente de uma menor capacidade proliferativa de linfócitos frente ao estímulo com conA. De maneira interessante, na ausência de IL-10 houve potencialização na produção de IL-17 por camundongos infectados, e esses foram mais resistentes à infecção, apresentando números reduzidos parasitos no baço e no fígado. Além de promover proteção através da modulação de IL-10, a IL-17 foi capaz de potencializar a produção de NO in vitro e in vivo. Juntos, nossos resultados demonstram que a L. infantum/chagasi é capaz de desencadear o padrão Th17 de resposta imune, o qual promove proteção do hospedeiro durante a infecção. / Visceral leishmaniasis (LV) is a chronic and potentially fatal disease caused by Leishmania donovani and Leishmania infantum/chagasi. In this study, our aim was to evaluate the Th17 and related cytokines participation, besides the mechanism by which these cells playing during the L. infantum/chagasi infection.The results showed that the parasite induces high amounts of TGF-, IL-1, IL-6, and IL-23 by bone marrow-derived dendritic cells (BMDC), cytokines involved with Th17 induction and/or maintenance. Accordingly, naïve-C57BL/6 spleenocytes co-cultured with L. infantum/chagasi-infected BMDC produced significant amounts of IL-17 by TCD4+. Interestingly, IL-17 was produced in high amounts in the liver and spleen of WT infected-mice, being peaked at 4th and 6th weeks after infection. The Th17 is critical for protective immunity against L. infantum/chagasi, since that IL-17R-/-, IL-23p19-/- and IL-6-/- infected-mice showed increasing of parasite load associated with enhancement of IL-10 production in IL-17R-/- compared to WT infected-mice. Strictly, in the absence of IL-17 signaling, a smaller inflammatory infiltrate was observed in the liver. Interestingly, IL-17 production was potentiated in IL-10-/- infected-mice, and they were more resistant to infection, showing reduced parasites numbers in the spleen and liver. In addition to promoting protection through the downmodulation of IL-10, IL-17 enhanced the NO production. Together, our results demonstrate that L. infantum/chagasi trigger Th17 response that promotes the host protection during infection.

Não existem vacinas licenciadas para uso em humanos e o antimônio pentavalente ainda é o fármaco de primeira escolha para o tratamento da doença, apesar de sua toxicidade. Em um estudo prévio foram realizados ensaios sobre a participação de proteína ligante de heparina de L. infantum chagasi (PLHLc) no processo infeccioso do parasito, considerando os processos de adesão, penetração e internalização em macrófagos, demonstrando que a proteína nativa está envolvida na interação entre parasito e hospedeiro. Também foram demonstrados níveis de imunogenicidade e de proteção contra L.infantum chagasi quando camundongos BALB/c foram imunizados com a PLHLc nativa. A partir disso, foi feito o sequenciamento dessa proteína e a produção da sua forma recombinante (rPLHLc). Nesse trabalho, utilizamos a rPLHLc em experimentos de imunização de camundongos BALB/c para avaliar se, assim como a proteína nativa, a forma recombinante também confere imunogenicidade e proteção após desafio com as formas promastigotas do parasito. Nossos resultados mostraram que no teste de imunogenicidade, os grupos imunizados com a rPLHLc, com AIF + rPLHLc e com SAP + rPLHLc produziram níveis altos de IgG1 e IgG2a anti-rPLHLc quando comparados aos seus grupos controles, caracterizando, de acordo com a razão IgG1/IgG2a, resposta mista nos grupos imunizados com rPLHLc e SAP + rPLHLce um perfil de resposta Th2 no grupo imunizado com AIF + rPLHLc. Já no teste de desafio, nossos resultados mostraram que houve redução da carga parasitária no baço e no fígado dos grupos imunizados com a rPLHLc, com AIF + rPLHLc e com SAP + rPLHLc. Os grupos imunizados com AIF + rPLHLc e com SAP + rPLHLc produziram as citocinas IFN-γ, TNF e IL-2, que direcionam o padrão de resposta imune para Th1, as citocinas IL-17 e IL-6 que direcionam para Th17 e a citocina reguladora IL-10, sendo que a média de produção dessas citocinas foi maior no grupo que tinha como adjuvante a SAP. Na produção de IgG1 e IgG2a anti-rPLHLcos grupos imunizados com a rPLHLc, com AIF + rPLHLc e com SAP + rPLHLc produziram níveis altos dos dois isotipos comparado aos seus grupos controles e de acordo com a razão IgG1/IgG2a conclui-se que encontramos perfil de resposta Th1 nos grupos imunizados com rPLHLc e com SAP + rPLHLc e perfil de resposta Th2 no grupo imunizado com AIF + rPLHLc. Portanto, concluímos que houve imunogenicidade e proteção contra L. infantum chagasi quando os grupos experimentais são imunizados com a proteína recombinante, mostrando a viabilidade de uso da mesma como vacina e que o grupo imunizado com SAP + rPLHLc foi o que apresentou resultados mais promissores, o que torna essa preparação uma possível vacina contra LV. / Visceral Leishmaniasis (VL) is widely distributed in 98 countries, with more than 350 million people at risk. There are no licensed vaccines for use in humans and the pentavalent antimony is still the drug of choice for treatment of this disease despite its toxicity. In a previous study, tests were performed on the involvement of the heparin binding protein of L.infantum chagasi (HBPLc) in the infectious process of the parasite, considering the processes of adhesion, penetration and internalization in macrophages, demonstrating that the native protein is involved in the interaction between parasite and host. Levels of immunogenicity and protection against L. infantum chagasi were also demonstrated when BALB/c mice were immunized with native HBPLc. From that, the sequencing of this protein and the production of its recombinant form (rHBPLc) was carried out. In this work, we used rHBPLc in experiments for immunization of BALB/c mice to evaluate whether, the recombinant protein form also confer immunogenicity and protection after challenge with the promastigote forms of the parasite. Our results showed that in the immunogenicity test, the groups immunized with rHBPLc, FIA + rHBPLc and ASP + rHBPLc produced high levels of anti-rHBPLc IgG1 and IgG2a when compared to their control groups, characterizing, according to the IgG1/IgG2a ratio, mixed response in the immunized groups with rHBPLc and ASP + rHBPLc and a Th2 response profile in the group immunized with FIA + rHBPLc. In the challenge test, our results showed that there was a reduction of the parasite load in the spleen and liver of the groups immunized with rHBPLc, with FIA + rHBPLc and with ASP + rHBPLc. The groups immunized with FIA + rHBPLc and ASP + rHBPLc produced the cytokines IFN-γ, TNF and IL-2, which direct the immune response pattern for Th1, the cytokines IL-17 and IL-6 directed for the Th17 and for the regulator cytokine IL-10, and the mean production of these cytokines was higher in the adjuvant group containing ASP. In the production of anti-rHBPLc IgG1 and IgG2a the groups immunized with rHBPLc, with FIA + rHBPLc and with ASP + rHBPLc produced high levels of the two isotypes compared to their control groups and according to the IgG1/IgG2a ratio we found that Th1 response profile in the group immunized with rHBPLc and with ASP + rHBPLc and Th2 response profile in the group immunized with FIA + rHBPLc. Therefore, we concluded that there was immunogenicity and protection against L.infantum chagasi when the experimental groups were immunized with the recombinant protein, showing the feasibility of using it as a vaccine and that the group immunized with ASP + rHBPLc presented the most promising results, which makes this preparation a potencial vaccine against VL.

Esses parasitos possuem forte tropismo por órgãos viscerais, especialmente baço, fígado e medula óssea, onde se proliferam e provocam lesões podendo ser letal caso não sejam tratadas. O parasito possui moléculas consideradas fatores de virulência que vem sendo intensamente investigadas. Entre elas destacamos as proteínas ligante de heparina (PLH), glicoproteínas relacionadas em diversos trabalhos na literatura com o processo de adesão e internalização do parasito à célula hospedeira. Nesse trabalho utilizamos a PLH de L. infantum chagasi (PLHLc) em experimentos de imunização de camundongos BALB/c para avaliar a proteção conferida a esses animais após desafio com as formas promastigotas do parasito. Foram avaliadas a carga de parasitos no baço e no fígado, produção de citocinas (IFN- , TNF, IL-2, IL-17, IL-6, IL-10 e IL-4) e de óxido nítrico (NO) após imunização com PLHLc isolada ou associada ao Adjuvante Incompleto de Freund (AIF) e Adjuvante Saponina (SAP), quatro semana após o desafio. Os camundongos foram imunizados por via intraperitoneal ou subcutânea, sendo submetidos a duas doses de reforço utilizando o mesmo protocolo da primeira imunização. Quinze dias após a última dose os camundongos foram infectados com 1 x 10 7 promastigotas de L. infantum chagasi em fase final logarítmica de crescimento. Quatro semanas após a infecção os animais foram eutanasiados e o baço e o fígado foram coletados para ensaio da quantificação da carga parasitária e avaliação da produção de citocinas e NO por citometria de fluxo e pelo método de Griess, respectivamente. Nossos resultados mostraram que o grupo imunizado apenas com PLHLc não apresentou redução da carga parasitária e os esplenócitos estimulados in vitro com antígeno particulado de L. infantum chagasi (AgLc) produziram citocinas IL-6 e IL-17, ambas de padrão de resposta imune Th17 e a citocina IL-10. O grupo imunizado com PLHLc + AIF apresentou redução da carga de parasitos apenas no baço e produziram citocinas IFN- e IL-2 (Th1), IL-6 e IL-17 (Th17) e IL-10. O grupo imunizado com PLHLc + SAP apresentou redução da carga parasitária no baço e no fígado. Os esplenócitos desse grupo estimulados in vitro com AgLc produziram maiores médias de citocinas de padrão de resposta imune Th1 e Th17 e a citocina reguladora IL-10 comparado com os grupos anteriores. Em relação a produção de NO por esplenócitos estimulados in vitro com AgLc níveis basais foram detectados pelo método de Griess em todos os grupos avaliados. Os resultados alcançados nos mostra que a PLHLc associada ao adjuvante Saponina representa uma forte candidata a vacina contra LV e nos direciona para uma nova etapa de investigação, a proteção conferida pela PLHLc recombinante. / Visceral leishmaniasis (VL) is a disease caused by the protozoan intracellular parasite Leishmania infantum chagasi specie in the Americas. These parasites exhibit a strong tendency to invade visceral organs, especially the spleen, liver and bone marrow, where they proliferate and cause injuries and can be lethal if not treated. The parasite exhibit molecules considered as virulence factors that have been intensively searched. Among then we highlight the heparin-binding proteins (HBP), glycoproteins related in several studies in the literature with the process of adhesion and internalization of the parasite to the host cell. In this work, we used L. infantum chagasi HBP (HBPLc) in immunization experiments using BALB/c mice to evaluate the protection given to the animals after challenge with promastigote forms of the parasite. Were evaluate the parasite load in the spleen and liver, cytokine production (IFN- , TNF, IL-2, IL-17, IL-6, IL-10 e IL-4) and nitric oxide (NO) after immunization with isolated HBPLc or associated with Incomplete Freund’s Adjvant (IFA) and Saponin Adjuvant (SAP), four weeks after challenge. The mice were immunized by intraperitoneal or subcutaneous route and submitted two times to booster doses using the same protocol of the first immunization. Fifteen days after the last dose the mice were infected with 1 x 10 7 L. infantum chagasi promastigotes end logarithmic phase of growth. Four weeks after infection the animals were euthanized and spleen and liver were collected for assay quantification of the parasite load and evaluation of the cytokine and NO production by flow cytometry and by Griess method, respectively. Our results showed that the group immunized with only HBPLc showed no reduction in parasite load and spleen cells stimulated in vitro with crude extract of L. infantum chagasi (AgLc) produced cytokines IL-6 and IL-17, both standard immune response Th17 and IL-10 cytokine. The group immunized with HBPLc + IFA decreased the parasite load only in the spleen and produced cytokines IFN- and IL-2 (Th1), IL-6 and IL-17 (Th17) and IL-10 cytokine. The group immunized with HBPLc + SAP decreased the parasite load in the spleen and liver. The spleen cells this group in vitro stimulated with AgLc generated pattern cytokines of Th1 and Th17 and regulatory cytokine IL-10 compared to previous groups. About NO production by spleen cells stimulated in vitro with AgLc baseline levels were detected by Griess method in all groups evaluated. The results achieved shows that the HBPLc associated with Saponin adjuvant is a strong candidate against VL vaccine and directs us to a new stage of research, the protection given by recombinant HBPLc.

Nas Américas, em especial na América do Sul, a infecção por Leishmania (L.) infantum chagasi causa manifestações subclínicas e leishmaniose visceral (LV), a qual quando não tratada é potencialmente fatal. Na América Central, especialmente em Honduras, a leishmaniose cutânea não ulcerada ou atípica e a leishmaniose visceral são causadas pelo mesmo agente etiológico Leishmania (L.) infantum chagasi e ocorrem na mesma região geográfica. Pouco se conhece sobre o perfil da infecção humana por Leishmania (L.) infantum chagasi em Honduras, especialmente sobre a forma não ulcerada ou atípica, sendo assim, no sentido de compreender melhor a patogênese da infecção causada por esta espécie de parasito no município de Amapala (Valle) e Orocuina (Choluteca), Honduras, o presente estudo visou a caracterização histopatológica e da resposta imune celular in situ em lesões de pacientes com leishmaniose cutânea não ulcerada ou atípica. Foram utilizadas 20 biópsias de pele de pacientes com diagnóstico parasitológico confirmado por raspado de lesão corado por Giemsa e observado em microscópio ótico. A histopatologia foi avaliada em cortes histológicos corados por hematoxilina-eosina (HE) e a resposta imune por meio da reação de imunoistoquímica utilizando anticorpos primários para macrófagos (CD68), linfócitos T (CD4, CD8), células natural killer - NK (CD56), linfócitos B (CD20), óxido nítrico sintetase (NOS2) e interferon-gama (IFN-y. Microscopicamente as alterações histopatológicas mais significativas foram observadas na derme superficial e profunda caracterizando-se por um infiltrado inflamatório predominantemente linfohistiocitário de intensidades variável com arranjo difuso e, por vezes, associado à formação de granulomas epitelioides. A observação de formas sugestivas do parasito ocorreu em apenas 55 % dos casos. A análise imunoistoquímica das lesões confirmou a presença de células mononucleares no infiltrado inflamatório caracterizada principalmente pela presença de linfócitos TCD8+, seguido por TCD4+, macrófagos, linfócitos B e células NK. A grande densidade de células INF-y+ e a presença da enzima iNOS, responsável pela produção de óxido nítrico, molécula com atividade leishmanicida, em quase a totalidade dos macrófagos do infiltrado inflamatório, sugere uma resposta imune local bastante efetiva, que colabora para o controle do número de parasitos e consequentemente a evolução do tamanho da lesão / In America, especially in South America, infection by Leishmania (L.) infantum chagasi causes subclinical manifestations and visceral leishmaniasis (VL), which when untreated is potentially fatal. Little is known about the profile of human infection by Leishmania (L.) infantum chagasi in Honduras, especially on the non-ulcerated or atypical form. In order to better understand the pathogenesis of the infection caused by this species of parasite in the municipality of Amapala (Valle) and Orocuina (Choluteca), Honduras, the present study aim the characterization of histopathological changes and cellular immune response in situ in lesions of patients with non-ulcerated or atypical cutaneous leishmaniasis. Twenty skin biopsies from patients with parasitological diagnosis confirmed by scraping of Giemsa-stained lesion and observed using an optical microscope. Histopathology, was evaluated by hematoxylin-eosin (HE) and the immunohistochemistry using the primary antibodies to macrophages (CD68), T lymphocytes (CD4, CD8), natural killer cells - NK (CD56), B lymphocytes CD20), nitric oxide synthase (NOS2) and interferon-gamma (IFN-y). Microscopically, the most significant histopathological changes were observed in the superficial and deep dermis, characterized by a predominantly lymphohistiocytic inflammatory infiltrate of variable intensities with diffuse arrangement and sometimes associated with the formation of epithelioid granulomas; the observation of suggestive forms of the parasite occurred in only 55 % of the cases. The immunohistochemical analysis of the lesions confirmed the presence of mononuclear cells in the inflammatory infiltrate characterized mainly by the presence of T-CD8+ lymphocytes, followed by T-CD4+, macrophages, B lymphocytes and NK cells. The high density of INF-y+ cells and the presence of iNOS, enzyme responsible for the production of nitric oxide, molecule with activity leishmanicide, in almost all of the macrophages in the inflammatory infiltrate, suggest an effective local immune response that collaborate to the control in the number of parasites and consequently the evolution of the lesion

Em se tratando de uma das sete endemias de maior impacto mundial, a Leishmaniose é uma doença negligenciada causada pelo parasita do gênero Leishmania, com estimativas alarmantes de novos casos e óbitos por ano, se tornando um grande problema de saúde pública. No Brasil, a Leishmaniose Visceral em áreas urbanas se destaca pelo fato de ser uma zoonose que possui como principal reservatório também o cão, que sempre esteve presente e em estreito contato com grande parte das famílias brasileiras. O mesmo, quando infectado pela doença, é submetido à eutanásia como norma do Ministério da Saúde, por apresentar riscos à população. Utilizando-se a Lei da Similitude, instruída por Hahnemann, chegou-se à medicação Antimonium crudum para possível tratamento e profilaxia da Leishmaniose Visceral Canina. Portanto, no presente trabalho, visou-se o estudo da ação da medicação homeopática Antimonium crudum, in vitro, diretamente em promastigotas de Leishmania (Leishmania) infantum chagasi, através do teste de MTT (brometo de 3-[4.5 dimetitiazol- 2- il) 2,5 difenil- 2 H- tetrazólio), que visa mensurar a atividade metabólica e a observação da curva de crescimento do parasito na forma promastigota, bem como a ação na infecção das Leishmanias em macrófagos da linhagem J774, em sua forma amastigota. Os resultados obtidos para a forma promastigota mostram que o estímulo homeopático não teve influência direta sobre os parasitos, tanto no teste de MTT quanto na curva de crescimento, corroborando os estudos que sugerem que a homeopatia não age diretamente sobre o parasito e sim em conjunto com o sistema imune do hospedeiro. Já nos resultados dos ensaios infectivos, observa-se, em geral, maior porcentagem de macrófagos infectados, assim como maior taxa de fagocitose nos grupos tratados com Antimonium crudum, o que sugere uma maior atividade dos mesmos levando à uma resposta imune secundária mais representativa e consequentemente, à destruição do antígeno. When it comes to one of the seven largest worldwide impact endemics, Leishmaniasis is a neglected public health complex with alarming estimates of new cases and deaths per year. In Brazil, urban Visceral Leishmaniasis also stands out because the reproductive cycle of this zoonotic disease can include the dog, which has always been present and in close contact with most Brazilian families. Dogs, when infected by the disease are euthanized as a rule from the Ministry of Health to prevent risks to the population. Using the Law of Similars, developed by Hahnemann, the medication Antimonium crudum was identified as possible treatment and prophylaxis for Canine Visceral Leishmaniasis. Therefore, the present work was to study the direct action of the homeopathic medicine Antimonium Crudum in vitro against the promastigotes of Leishmania (Leishmania) infantum chagasi, through the MTT test (3- [4.5 - dimetitiazol- 2-yl) - 2.5 diphenyl tetrazolium H 2), which aims to measure the metabolic activity; the promastigote growth curve; as well as the performance of promastigote infectivity in J774 macrophages. The results from both the MTT assay as well as the growth curve show that the homeopathic stimulus did not directly influence the promastigote form of the parasites, corroborating studies that suggest that homeopathy has no direct action on the parasite, but may function together with the host\'s immune system.
