The role of parasite-derived arginase in murine Leishmania major

The outcome of infection with Leishmania major depends in part on the balance between arginase and inducible nitric oxide synthase in macrophages. These enzymes compete for the substrate L-arginine. Leishmania major also encodes an arginase gene but, the role of this parasite-derived enzyme in infection remains unclear. We hypothesize that parasite-derived arginase influences parasite survival and host immune response to L. major. To examine this hypothesis, we employed an arginase deficient null mutant L. major in in vitro and in vivo experiments. Our results show that deficiency of parasite-derived arginase impaired parasite proliferation and disease pathogenesis. Increased arginase activity however neither affected nitric oxide production, nor did it correlate with IL-4 production. Primary infection of normally resistant hosts causes a chronic infection and does not protect them against re-infection. Thus, parasite-derived arginase is of nutritional importance to L. major, but is not a feasible therapeutic drug target.

O cão se destaca por seu importante papel na cadeia epidemiológica da doença, porquanto é o principal reservatório urbano da infecção, cuja transmissão é dependente de populações de Lutzomyia longipalpis. A enfermidade é multissistêmica e de patogenia complexa, envolvendo diferentes mecanismos imunológicos. A despeito do grande número de estudos sobre a patogenia e lesões que ocorrem na leishmaniose visceral canina, a fisiopatologia da lesão ocular ainda permanece obscura. O objetivo deste estudo foi detectar a presença de células inflamatórias e de formas amastigotas de L. Infantum pela imuno-histoquímica em olhos de cães experimentalmente infectados via ocular tópica. Constituiu-se um grupo inoculado (GI) composto por sete cães, que receberam a instilação de cultura de Leishmania infantum por via conjuntival, e um grupo controle (GC) composto por três cães, nos quais a instilação tópica foi realizada com solução salina. Após 60 dias, os tecidos oculares foram analisados utilizando-se as técnicas de coloração em Hematoxilina e Eosina (HE) e a reação de imuno-histoquímica pela streptoavidina-peroxidase. A análise histopatológica revelou uma resposta inflamatória com predomínio de plasmócitos e linfócitos, perivasculite, congestão vascular e hiperplasia das células caliciformes em conjuntiva. As glândulas lacrimais da terceira pálpebra apresentaram infiltrado inflamatório com atrofia focal e desorganização do estroma. Formas amastigotas de L. infantum foram observadas através da detecção imuno-histoquímica em 100% das amostras do grupo infectado / Abstract: Canine visceral leishmaniasis (CVL) is an infection caused by Leishmania infantum agent. The dog stands out for its important role in the epidemiology of the disease, since it is the main urban reservoir of infection, whose transmission is dependent on populations of Lutzomyia longipalpis. The disease is multisystemic and complex pathogenesis, involving different immunological mechanisms. Despite the large number of studies regarding the pathogenesis and lesions that occur in canine leishmaniasis, the pathophysiology of a ocular lesion still remains obscure. The goal of this study was to evaluate the inflammatory cells and immunohistochemical detection of L. infantum in eyes from dogs experimentally infected by topical ocular infection. Constituted an inoculated group (IG), composed of seven dogs that received instillation of culture of Leishmania infantum by conjunctival route, and a control group (CG) composed of three dogs, in which the topical instillation was performed with saline. After 60 days, the ocular tissues were analyzed using the techniques of staining in hematoxylin and eosin (HE) and immunohistochemical reaction for streptoavidin-peroxidase. Histopathological analysis revealed an inflammatory response characterized by the presence of plasma cells, lymphocytes, congestion, perivasculitis and hyperplasia of the goblet cells in conjuctiva. In the lachrymal gland of the third eyelid were observed inflammatory response with focal atrophy and stromal disorganization. Comparação entre os métodos de ELISA - Antígeno total e ELISA - Ligante de Fucose e Manose em cães sintomáticos e oligossintomáticos para leishmaniose visceral /

Resumo: A Leishmaniose visceral canina (LVC), conhecida como calazar, é uma antropozoonose endêmica no Brasil, causada pela Leishmania L. chagasi. O teste sorológico de ELISA tem sido empregado na rotina de inquéritos epidemiológicos e no auxílio diagnóstico clínico de cães suspeitos. O método de ensaio imunoenzimático em fase sólida (ELISA) usando o antígeno total de Leishmania L. chagasi (ELISA-AgT), assim como, o ELISA - Ligante de Fucose e Manose (ELISA-FML), tem demonstrado boa sensibilidade e especificidade para detectar a doença em cães assintomáticos e oligossintomáticos. O presente trabalho teve como objetivo comparar dois antígenos pelo método de ELISA no sorodiagnóstico de cães naturalmente infectados pela LVC, positivos no exame parasitológico, e agrupados em: grupo sintomático e, grupo oligossintomáticos, tendo como grupo controle cães de área não endêmica. Nos animais oligossintomáticos, a sensibilidade observada para ELISA-AgT foi de 86,7%, já para o método ELISA-FML apresentou valor de 90%. A especificidade foi de 100% no método de ELISA-AgT e 96,7 % para o ELISA-FML. Nos animais sintomáticos, a sensibilidade e especificidade para o ELISA-AgT foram de 90% e 93,3%, respectivamente; já o método de ELISA-FML apresentou sensibilidade e especificidade de 86,7% e 96,7%. No teste ELISA-AgT o valor preditivo positivo foi de 93,1% nos sintomáticos e 100% nos oligossintomáticos, enquanto que nos animais submetidos ao teste ELISA-FML, foi observado 96,3% para os sintomáticos e 96,4% para os oligossintomáticos. O índice Kappa foi utilizado para medir o grau de concordância real entre os dois métodos imunoenzimáticos empregados e o exame parasitológico direto, mostrando boa concordância entre os métodos realizados. / Abstract: Visceral canine leishmaniasis or calazar is Brazilian an endemic antropozoonosis caused by Leishmania L. chagasi. ELISA sorologycal test has been used in routine of epidemiological studies and in diagnosis of clinical suspect dogs. The immunoenzymatic assay in solid phase (ELISA) using Leishmania L. chagasi total antigens AgT-ELISA and Fucose Manose ligant-ELISA (FML-ELISA) has been showed good sensibility and specificity for detection disease in asymptomatic and oligosymptomatic dogs. The present paper has the goal of compare the efficiency of two antigens by ELISA methods in dogs naturally affect by leishmaniasis with positive parasitological exam and grouped by symptoms. Group composed by symptomatic dogs, and Group by oligosymptomatic dogs. Control group was constituted of dogs from Leishmania free areas. Dogs of Group oligosymptomatics presented 86,7% of sensibility in AgT-ELISA and 90% at FML-ELISA. The specificity was 100% at AgT-ELISA and 96.7% for FML-ELISA. At Group symptomatics the sensibility and specificity for AgTELISA and FML-ELISA were respectively 90% and 93.3% and, the FMLELISA showed 86.7% and 96.7% corresponding to sensibility and specificity. On ELISA-AgT the positive predictive valor was 93.1% at symptomatic and 100% at oligosymptomatic, moreover in the dogs tested by FML-ELISA the valor was 96.3% for the symptomatic and 96.4% for oligosymptomatics. The Kappa was used and showed a good concordance between both methods tested. Utilidad de muestras clínicas de pacientes para realizar estudios de variabilidad genética de Leishmania (V.) braziliensis, procedentes de diferentes departamentos del Perú

Manifiesta que los estudios de variabilidad genética describen la variabilidad usando cultivos como fuente primaria de muestra lo que permite obtener una alta calidad de ADN del parásito. Sin embargo el establecimiento del cultivo requiere un procedimiento invasivo de muestreo y personal altamente entrenado para evitar la contaminación bacteriana de los medios usados para aislar el parásito. Pocas investigaciones se han realizado usando muestras alternativas en estudios de variabilidad genética demostrando su utilidad frente a los cultivos, por ello, se determinó la utilidad de improntas en papel filtro, raspados por lanceta y biopsias para evaluar los parámetros de genética de poblaciones en Leishmania. El material genético fue extraído de todos los tipos de muestra y se caracterizó los aislados de leishmaniasis como L. (V.) braziliensis usando la técnica de energía por resonancia de fluorescencia (FRET) en una PCR en tiempo real anidada de 42 cultivos aislados de pacientes y 78 muestras clínicas (improntas, aspirados y biopsias). Luego se realizó la PCR de microsatélites a cada muestra clínica usando 9 marcadores polimórficos marcados con compuestos fluorescentes de tipo 6-FAM, HEX y NED mediante electroforesis capilar en el analizador genético 3130xl. Finalmente, los electroforegramas fueron analizados usando el software Gene mapper v4.0; los índices de heterocigosidad observada y esperada, número promedio de alelos, estructura poblacional y distancias genéticas fueron realizadas usando el software Cervus v3.0.6, Structure v2.1 y Population v1.2.32 La comparación entre los tipos de muestra mostró que las improntas en papel filtro, raspados y biopsias tuvieron menor resolución que los cultivos para obtener análisis genéticos precisos, este hallazgo puede deberse al escaso material del parásito en muestras clínicas primarias. Nuestros resultados indican que la utilidad de muestras clínicas primarias para el análisis de microsatélites es limitada en comparación a los cultivos que presentan mayor resolución y facilitan las pruebas de variabilidad genética. / Tesis

En artículos publicados, relacionados a esto, existe mucha información sobre el tratamiento tópico de diferentes enfermedades usando grasas naturales, principalmente animales y vegetales administrados bajo la forma de pomadas, debido a que estas como la manteca de cerdo son compatibles con la piel humana. Se ha estudiado la manteca, como componente del cerdo y se ha determinado que la proveniente de la grasa que se ubica alrededor de los riñones es de mejor calidad y con características muy similares a la de los humanos. / At present, the treatment of the diseases in animals and humans using natural resources is a very used custom. In the published articles in reference to this fact there are plenty of publications for the topical treatment of different diseases using natural resources mostly animal and vegetables which are administrated under the form of ointments due to the fact that the materials such as lard are very compatible with the human skin. Lard is the grease which is a normal component of the pork. It has been studied and it has been determined that lard which surrounds the kidneys is the due to its characteristics very similar to the humans.
