Twenty beagles dogs were selected, divided into two groups: control and treated with a collar containing a combination of deltamethrin 4g and propoxur 12g. Each animal was infested with 50 R. sanguineus ticks (25 males and 25 females) and 100 C. felis felis fleas adults (50 males and 50 females). For the assessment of efficacy, 48 hours after each infestation, the mechanical removal of the parasites was performed. Each animal was infested with fleas and ticks on days -7 (before treatment), -2, +5, +12, +19 and +26. After day 26, every 14 days infestations were performed until the day +208. After this experimental day, the animals were infested with fleas only every 7 days until the termination of the study on day +271. For haematological and biochemical evaluations, blood samples were collected from animals in both groups every seven days until day +14. This experimental day up to +266, samples were collected every 14 days. After removal of the collars (day 274), three collections were made on days: +275, +281 and +288 , to assess the possible occurrence of changes in serum levels of serum enzyme butyrylcholinesterase (BChE). The acaricides effectiveness of the test product was 100 % on days +7, +21 and +28. The efficacy values were above 90% until day 126. As from day +182, the efficacy reached values lower than 80%. Only after day +168, levels declined below 80%. Pulicide efficiencies of the test product was 100 % on days +21, +28, +42, +56, +98 and +112. The efficacy values were above 90% until day +266, reducing 89.48% on day 273. The animals showed no changes in clinical and laboratory examinations that could be associated with adverse effects from the use of the product tested. It is concluded that a collar containing a combination of deltamethrin 4g and propoxur 12g was effective in controlling R. sanguineus ticks for up to 126 days and control C. felis felis fleas for up to 266 days in addition to proving to be safe for use in dogs. The animals showed no clinical and laboratory changes that could be associated with adverse effects from the use of the product tested, thus demonstrating that the association deltamethrin and propoxur in an impregnated collar is safe for use in dogs. / O objetivo do presente estudo foi avaliar a efic?cia de uma coleira impregnada com deltametrina e propoxur no controle de infesta??es por Rhipicephalus sanguineus e Ctenocephalides felis felis em c?es da ra?a beagle e avaliar a seguran?a cl?nica do tratamento atrav?s de exames cl?nicos, hematol?gicos e bioqu?micos, al?m de avaliar a dosagem da colinesterase s?rica em c?es submetidos ao tratamento com um carbamato. Para isso, foram selecionados 20 animais da ra?a beagle em bom estado sanit?rio, distribu?dos em dois grupos: controle e tratado com uma coleira impregnada com 4g de deltametrina e 12g de propoxur. Cada animal foi infestado com 50 carrapatos R. sanguineus (25 machos e 25 f?meas) e 100 pulgas C. felis felis adultas (50 machos e 50 f?meas). Para a avalia??o da efic?cia, 48 horas ap?s cada infesta??o, foi realizada a remo??o mec?nica dos parasitos. Cada animal foi infestado com pulgas e carrapatos nos dias -7 (antes do tratamento), -2, +5, +12, +19 e +26. Ap?s o dia +26 foram realizadas infesta??es a cada 14 dias at? o dia +208. Ap?s este dia experimental, os animais foram infestados somente com pulgas a cada sete dias, at? o encerramento do estudo, no dia +271. Para avalia??o hematol?gica e bioqu?mica, amostras de sangue foram coletadas dos animais de ambos os grupos a cada sete dias at? o dia +14. Deste dia experimental at? o dia + 266, as coletas foram realizadas a cada 14 dias. Ap?s a retirada das coleiras (dia +274), foram realizadas mais tr?s coletas nos dias +275, +281 e +288, para avaliar a poss?vel ocorr?ncia de altera??es nos n?veis s?ricos da enzima butirilcolinesterase s?rica (BChE). A efic?cia carrapaticida do produto em teste foi de 100% nos dias +7, +21 e +28. Os valores de efic?cia foram superiores a 90% at? o dia+126. Apenas a partir do dia +182, a efic?cia atingiu valores inferiores a 80%. Apenas ap?s o dia +168 os n?veis declinaram abaixo dos 80%. A efic?cia pulicida do produto em teste foi de 100% nos dias +21, +28, +42, +56, +98 e +112. Os valores de efic?cia foram superiores a 90% at? o dia+266, reduzindo a 89,48% no dia +273. Os animais n?o apresentaram altera??es em exame cl?nico e laboratoriais que pudessem ser associados a efeitos adversos provenientes do uso do produto testado. Conclui-se que a coleira impregnada com 4g de deltametrina e 12g de propoxur foi eficaz no controle de carrapatos R. sanguineus por at? 126 dias e no controle de pulgas C. felis felis por at? 266 dias. Os animais n?o apresentaram altera??es cl?nicas e laboratoriais que pudessem ser associadas a efeitos adversos provenientes do uso do produto testado, demonstrando assim que a associa??o deltametrina e o propoxur em uma coleira impregnada ? segura para uso em c?es.

The aim of this study was to diagnose and treat the infection by T. foetus in cats treated at HVPA of UFRRJ, identify the clinical relevance of this parasite, the risk factors, presence of co-infections and clinical response to treatment. In this study, were admitted 100 felines, from different breeds, both sexes and from 16 weeks of age. As inclusion criteria, patients had to have diarrhea history for more than three weeks, with little success of clinical response to treatments previously used. From the stool samples obtained by colon washing or spontaneous defecation it was conducted direct examination of stool, centrifugal flotation technique and real-time PCR. Of the 100 cats selected for this study, 10% had T. foetus infection. Most positive animals were over one year old (70%) and only 30% had up to 1 year old. Cats were 80% purebred and 20% were animals mongrel. The clinical signs present were diarrhea (60%), weight loss (10%), hemoatochezia (30%) and presence of mucus in the stool (20%). Only two animals were positive for the antigen of FeLV virus, and theses showed more severe case of diarrhea. Three animals showed co-infections with other enteropathogens (30%), one with Giardia, one with Cryptosporidium and one with Cystoisospora. All treated animals showed clinical improvement with the administrations of ronidazole at a dose 30mg/kg every 24 hours during 14 days and showed no recurrence until the recent time. We can conclude that T. foetus infection is an important differential diagnosis in cats with chronic diarrhea, which cats of any age and regardless of whether purebred or not, may have the infection, other enteropathogens needs to be investigated in all cases and the treatment was effective in the resolution of clinical signs of all treated cats. / Tritrichomonas foetus foi recentemente reconhecido como um importante agente etiol?gico de diarreia em diversos pa?ses. O objetivo deste estudo foi diagnosticar e tratar a infec??o por T. foetus nos gatos atendidos no HVPA da UFRRJ, identificar a relev?ncia cl?nica deste parasita, os fatores de risco, a presen?a de co- infec??es e resposta cl?nica ao tratamento. Neste trabalho, foram admitidos 100 animais da esp?cie felina, de diferentes ra?as, de ambos os sexos e a partir de 16 semanas de idade. Como crit?rio de inclus?o, os pacientes deveriam apresentar hist?rico de diarreia a mais de tr?s semanas, com pouco sucesso de resposta cl?nica para terap?uticas previamente institu?das. A partir das amostras de fezes obtidas por coleta via lavagem do colon dos animais e pelo ato de defeca??o espont?nea, foram realizados os m?todos do exame direto das fezes, t?cnica de centrifugo flutua??o e PCR em tempo real. Dos 100 gatos que foram selecionados para esta pesquisa, 10% apresentaram infec??o por T. foetus. A maioria dos animais positivos estavam na faixa et?ria acima de um ano (70%) e apenas 30% tinha at? um ano de idade. Os gatos eram 80% de ra?a pura e 20% eram animais sem ra?a definida. Os sinais cl?nicos apresentados por esses gatos foram diarreia (60%), perda de peso (10%), hematoquezia (30%) e presen?a de muco nas fezes (20%). Apenas dois animais infectados eram positivos para o ant?geno do v?rus da FeLV, e estes apresentaram um quadro mais severo de diarreia. Tr?s animais apresentaram coinfec??es com outros enteropat?genos (30%), um com Giardia sp, um com Cryptosporidium e outro com Cystoisospora. Todos os gatos tratados apresentaram melhora cl?nica com a administra??o do f?rmaco ronidazol na dose de 30 mg/Kg a cada 24 horas por 14 dias, n?o apresentando recidiva da diarreia at? o recente momento. Podemos concluir que a infec??o por T. foetus ? um importante diagn?stico diferencial em felinos com diarreia cr?nica, que gatos de qualquer idade e independente de ser de ra?a pura ou n?o, podem apresentar a infec??o, que a co- infec??o com outros enteropat?genos necessita ser investigada em todos os casos e que o tratamento foi efetivo na resolu??o dos sinais cl?nicos de todos os gatos tratados.

Adults and immature of some muscoid dipteran groups have little morphological differences, making the identification process more difficult. Besides, the available dichotomous keys are based on adults, but in the case of myiasis and cadaver colonization, only immature are found. These insects are known pathogen vectors, their larvae parasite humans and animais and they play an important role in forensic entomology. Therefore, the correct identification of these species is very important. This study aims to characterize features of medical, veterinary and forensic important dipteran species using two new approaches. The first chapter uses scanning electron microscopy which gives detailed information on the morphological characters that can help identify the immature forms of the flies. Immatures belonging to three muscoids with forensic and sanitary importance were studied using this methodology: Peckia (Eubaettcheria) cal/usar (Diptera: Sarcophagidae) (larvae and puparia), Luc?lia cuprina (larvae and puparia) e Cachliamyia macellaria (eggs, larvae and puparia) (Diptera: Calliphoridae). The size ofthe chorionic cells as well as the absence of spines at the posterior spiracle allowed to differentiate C. mace/faria from C. haminivarax, both myiasis causing species. The maxillary palp complex composed by a group of papillae inside a cavity seem to be specific to the subgenus Eubaeucheria. The anterior spiracular openings displaced in an irregular row are found exclusively in Sarcophagidae. The filiform spines at the anal segments are found only in the genus Lucilia. In the second chapter, barcading is used to provide a gene sequence database of medical, veterinary and forensically important Calliphoridae flies. This technique is based on the sequencing of a portion of the genome of the mitochondrial cytochrome oxidase l (cal) gene. Nine species of flies were collected in four regions in Brazil, corresponding to 346 specimens. Two other gene fragments ( elongation factor 1 alpha and white) were sequenced for the first time for some species. The barcade proposal was successfully achieved for Hemilucilia segmentaria, Hemi/ucilia semidiaphana, Chlarapracla idiaidea, Cachliamyia macellaria, Chrysamya megacephala and Chrysamya albiceps, for which the intraspecific variation was less than 3%. However, for Lucilia eximia, Chrysamya putaria and Chrysamya rufifacies, this methodology brought incipient results. Intraspecific variation among Lucilia eximia samples was 4.6%, suggesting that the threshold proposed should not be used for ali dipteran species without previous analysis of the studied group. Chrysamya putaria is polyphyletic according to nuclear and mitochondrial genes. In addition, a set of samples grouped with samples of C. albiceps and C. rufifacies. Therefore, more studies need to be clone to better explain these taxonomic problems. / A ordem Diptera ? composta por aproximadamente 153 mil esp?cies descritas, mas acredita-se que o n?mero de esp?cies ainda desconhecidas ? muito grande. Alguns grupos de d?pteros musc?ides possuem imaturos e adultos com poucas varia??es morfol?gicas, dificultando muito o processo de identifica??o. Al?m disso, as chaves dicot?micas existentes s?o baseadas nos adultos, entretanto, s?o os imaturos mais encontrados causando mi?ases ou colonizando cad?veres. Estes inselos s?o conhecidos vetores de pat?genos, suas larvas s?o parasitas de humanos e animais e apresentam papel importante como auxilio a entomologia forense. Assim, a correta identifica??o das esp?cies que desempenham estas fun??es ? muito importante. O presente estudo tem como objetivo principal caracterizar esp?cies de d?ptcros mnscoidcs de import?ncia m?dica-veterin?ria e forense, atrav?s da utiliza??o de duas t?cnicas modernas de identifica??o. O primeiro cap?tulo utiliza a microscopia eletr?nica de varredura que permite a visualiza??o e a determina??o de caracteres que possam ser utilizados na identifica??o das esp?cies. Foram estudados imaturos de tr?s esp?cies de d?pteros de duas fam?lias diferentes: Peckia (Eubaettcheria) callusar (Diptera: Sarcophagidae) (larvas e pup?rios), Lucilia cuprina (larvas e pup?rios) e Cachliamyia macellaria (ovos, larvas e pup?rios) (Diptera: Calliphoridae) atrav?s desta t?cnica. O tamanho das c?lulas do exoc?rion permitiu diferenciar entre C. macellaria e C. haminivarax, duas esp?cies causadoras de mi?ases. A aus?ncia de espinhos no espir?culo posterior das larvas de C. macellaria tamb?m a diferencia de C. haminivarax. As papilas do complexo do palpo maxilar em uma cavidade parecem ser espec?ficas do subg?nero Eubaettcheria. As aberturas respirat?rias do espir?culo anterior dispostas em fileiras irregulares s?o exclusivas da fam?lia Sarcophagidae. A presen?a de espinhos filamentosos no ?ltimo segmento larval ? caracter?stica espec?fica do g?nero Lucilia. No segundo cap?tulo, a t?cnica de harcade ? utilizada para a forma??o de um banco de dados contendo sequ?ncias g?nicas de esp?cies de import?ncia m?dica-veterin?ria e forense da fam?lia Calliphoridae. Esta t?cnica baseia-se no sequenciamento de uma por??o do gene mitocondrial citocromo oxidase I (cal). Foram analisadas nove esp?cies de d?pteros, coletadas em quatro regi?es do Brasil, em um total de 346 esp?cimes utilizados. Dois segmentos do genoma nuclear (fator de elonga??o 1 alfa e white) tamb?m foram sequenciados pela primeira vez para algumas esp?cies. A proposta de barcade foi bem sucedida em Hemilucilia segmentaria, Hemilucilia semidiaphana, Chlaraprocta idiaidea, Cachliamyia macellaria, Chrysamya megacephala e Chrysamya albiceps, onde a varia??o intraespec?fica foi menor do que 3%. Entretanto, em Lucilia eximia, Chrysamya putaria e Ch1ysamya rufifacies esta metodologia trouxe resultados incipientes. A varia??o intraespec?fica em Lucilia eximia foi 4,6%, sugerindo que o limiar proposto n?o deve ser aplicado em todos os d?pteros sem an?lise pr?via do grupo estudado. Chrysamya pularia ? uma esp?cie polifi!?tica comprovada tanto pelos genes nucleares quanto mitocondrial, al?m isso, uma parte das amostras forma um agrupamento com as amostras de C. albiceps e C. nijifacies. Assim, acrcditase que mais estudos precisam ser conduzidos com o objetivo de esclarecer melhor estas quest?es taxon?micas.

The Brazilian has low rates of productivity. The performance of the animals in pastures is reasonable during the rainy season, being in the dry food deficiency, caused by the low availability and quality of grass, which results in weight loss of animals. Thus ensiled foods are a recently solution to this situation in the production process. But we have a very few studies on the food substrate in Brazil and Latin America. The objectives of this study were primarily to establish the natural frequency of species of the genera Aspergillus, Penicillium and Fusarium potentially produce mycotoxins in samples of vegetable and silage components intended for cattle and other fungal genera present and detect mycotoxins presents in the same (aflatoxins, ochratoxin A, fumonisin B1, deoxinivalenol and glyotoxin). We are evaluated a total of 168 samples of corn and sorgum silos, from Janerie 2009 to October 2012. These farms creators of beef cattle were located in the state of S?o Paulo. We evaluated the pre and post fermented samples during this period. In all substrates had evaluated more than 50% of samples contaminated above the recommended limits feed of 1.0 x 104 ufc.g-1 (GMP, 2006). As in other studies the genera Aspergillus and Penicillium have high occurrence following related studies in the Americas and worldwide. Aspergillus flavus was the species of greatest occurrence in the samples evaluated, followed by A. niger aggregate, A. fumigatus and A. parasiticus. Presence was detected in high levels of toxins produced by these genera mainly: aflatoxins (up to 70 ppb) glyotoxin (up to 30 ppm) and ochratoxin A (up to 10 ppm). The Fusarium sp. showed a low variability of species in the samples evaluated, even in pre-fermented samples where counts were higher in this genera, had the occurrence of only two species F. verticillioides and F. gramineraum. Next to it was also the detection of toxins produced by these genera at low levels (levels of 0.32-2 ppm).T hese observations creates the need to expand studies related to ensiled food, which the animals were feed during major periods of the year. / Apesar de ter o segundo maior rebanho do mundo a bovinocultura de corte brasileira, apresenta modestos ?ndices de produtividade. O desempenho dos animais em pastagens ? razo?vel durante a esta??o chuvosa, observando-se na esta??o seca defici?ncia alimentar, causada pela baixa disponibilidade e qualidade do pasto, o que resulta em perda de peso dos animais. Assim, alimentos ensilados s?o uma solu??o atualmente utilizada a esta situa??o no processo produtivo. A avalia??o deste substrato alimentar ? escassa tanto no Brasil quanto no restante da Am?rica Latina, assim, os objetivos deste trabalho foram os de estabelecer principalmente a freq??ncia natural de esp?cies dos g?neros Aspergillus, Penicillium e Fusariu potencialmente produtoras de micotoxinas em amostras de componentes vegetais e silagens destinadas ao consumo bovino, bem como de outros g?neros f?ngicos presentes e detectar micotoxinas presentes nas mesmas (aflatoxinas, ocratoxina A, fumonisina B1, deoxinivalenol e gliotoxina). Foram avaliados um total de 168 amostras de ensilados de milho e sorgo no per?odo de janeiro 2009 a outubro 2012 (avaliado o material pr? e p?sfermentado nesse per?odo), provenientes de tr?s fazendas criadoras de gado de corte no estado de S?o Paulo. Foi observado que em todos os substratos avaliados tiveram mais de 50% de amostras contaminadas acima dos limites recomendados para alimenta??o animal de 1,0 x 104 ufc.g-1 (GMP, 2006). Sendo Aspergillus flavus ? esp?cie de maior ocorr?ncia nas amostras avaliadas, seguida por A. niger agregado, A. fumigatus e A. parasiticus. Detectou-se presen?a em n?veis altos de toxinas produzidas por esses g?neros principalmente: aflatoxinas (n?veis acima de 70 ppb), gliotoxina (n?veis acima de 30 ppm) e ocratoxina A (n?veis de at? 10 ppm). O g?nero Fusarium apresentou uma variabilidade baixa de esp?cies nas amostras avaliadas, mesmo em amostras pr?-fermentadas onde ocorreram contagens maiores deste g?nero, tiveram a ocorr?ncia de apenas duas esp?cies F. verticillioides e F. gramineraum. Junto a isso tamb?m tiveram a detec??o de toxinas produzidas por esses g?neros em n?veis baixos (n?veis de 0,32-2 ppm), principalmente no milho pr?-fermentado. Tais achados refor?am a avalia??o mais ampla desse substrato que ? base alimentar durante grandes per?odos do ano para o nosso rebanho.

However, coccidiosis or eimeriosis is a disease that causes a health problem of growing economic importance in Brazilian cattle. The aim of this study was to identify the species of Eimeria present in the faeces of buffalo, their distribution, frequency and prevalence in the State of Rio de Janeiro, as well as in the regions studied. Regarding morphotypes found observed the presence of three specific species such as buffalo E. bareillyi, E. ankarensis and E. gokaki, and the latter two species were noted for the first time in Brazil. And the morphotypes common to cattle and buffalo, buffalo were found in E. alabamensis, E. auburnensis, E. bovis, E. brasiliensis, E. cylindrica, E. ellipsoidalis, E. subspherica and E. zuernii. The most prevalent species in the state of Rio de Janeiro was E. zuernii, but this is not observed when analyzing the frequencies of each species per meso studied, and E. auburnensis more prevalent in the regions North Fluminense, Metropolitan and Northwest Fluminense, E. ellipsoidalis more prevalent in Central Fluminense and Coastal Lowlands, E. zuernii mesoregion in South Fluminense. The distribution of the genus Eimeria was not homogeneous, even within a meso, having been observed buffalo with a single species or with up to eight species of the genus Eimeria, these results may characterize properties of extensive creations where the animals have remained independent of age and sex persist for a long time. The morphometric characteristics observed in both the oocysts and sporocysts we were better detailed in this study. / A bubalinocultura ? considerada como uma grande op??o para pecu?ria nacional. No entanto, a coccidiose ou eimeriose ? uma enfermidade que gera um problema sanit?rio de import?ncia econ?mica crescente na pecu?ria brasileira. O objetivo deste estudo foi identificar as esp?cies de Eimeria presentes nas fezes de b?falos, sua distribui??o, frequ?ncia e preval?ncia no estado do Rio de Janeiro, assim como, nas mesorregi?es estudadas. The flagellate Pentatrichomonas hominis has been reported in association with domestic cats since the early 20th century. As a eurixenic parasite has been described in several hosts, including humans, dogs, bovines, rats, and a variety of wild animals. Another parabasalid with great importance in livestock, Tritrichomonas foetus has been also described in cats, pigs, and humans. These observations raise questions about the zoonotic potential of both species and their host specificity. This study aimed to diagnose parabasalid species found in cats using morphological and molecular analysis. Therefore, this study was divided into two parts to assess two different cat populations. In the first part, 41 animals in a trial cattery were evaluated. Twenty-six percent of the animals (11) were positive for P. hominis, at both techniques as fresh examination and culture. The DNA was extracted from the samples in culture and rRNA genes were amplified by polymerase chain reaction (PCR) using universal primers (TFR1 and 2) and other two specific species for T. foetus (TFR3 and 4) and P. hominis (TH3 and 5). Morphological analysis of trophozoites revealed the presence of five previous plagues core round and axostyle tapering uniform, characteristic way of P. hominis. The results of morphology were confirmed by molecular study. The sequencing of the isolates revealed a sequence with 100% similarity to P. hominis isolated from cats and dogs deposited in Genbank. This is the first study in Brazil with pointed out the presence of Parabasalids in cats by using morphological and molecular data and it is the one in the literature where P. hominis was isolated. In the second part of this study, 77 samples of feces from cats from the clinical care of HVPA-UFRRJ were examined. Only four of 77 samples tested were positive. Morphological analysis showed predominantly pear-shaped protozoa with three previous scourges, elongated nucleus and ax?stilo abruptly ending in characteristic fillet in T. fetus. In scanning electron microscopy and transmission, were visualized the identifying characters were similar to those previously reported for T. foetus. Molecular analysis confirmed the morphological diagnosis in the organism from four samples showed a sequence with 99.7 to 100% of similarity. It was deposited in Genbank as T. foetus. Despite the morphological analysis have recognized only T. foetus in the four samples examined, three of them were also positive for P. hominis in molecular analysis used as a differential diagnosis using species-specific primers (TH3 and TH5). The molecular analysis was used as a confirmatory tool for the presence of only one species present in evaluated feces. This demonstrated that not only T. foetus was identified in this study, but a co-infection by P. hominis cats could be considered. This indication was only confirmed as a diagnostic techniques when the morphological analysis and molecular biology were used to confirm both species. / O estudo sobre parabasal?deos em animais de companhia vem ganhando cada vez mais aten??o por sua associa??o a quadros de diarreia. O flagelado Pentatrichomonas hominis foi relatado em associa??o com gatos dom?sticos desde o in?cio do s?culo 20. Por ser um parasito eurix?nico, j? foi descrito em diversos hospedeiros, incluindo os seres humanos, c?es, bovinos, ratos e uma variedade de animais selvagens. Assim tamb?m outra esp?cie de parabasal?deo com grande import?ncia na pecu?ria, Tritrichomonas foetus j? foi descrito em gatos, su?nos, e em humanos tamb?m. Tais observa??es levantam d?vidas sobre o potencial zoon?tico de ambas as esp?cies e sua inespecificidade hospedeira. Este estudo teve por objetivos diagnosticar esp?cies de parabasal?deos encontrados em gatos utilizando de an?lise morfol?gica e molecular. Para tanto, este estudo foi dividido em duas partes para avaliar duas popula??es felinas distintas. Na primeira parte, 41 animais de um gatil de experimenta??o foram avaliados. Vinte e seis por cento dos animais (11) foram positivos para P. hominis, tanto no exame a fresco quanto na cultura. O DNA foi extra?do das amostras em cultura e os genes de rRNA foram amplificados por rea??o em cadeia da polimerase (PCR), utilizando iniciadores universais (TFR1 e 2) e outros dois esp?cies espec?ficos para T. foetus (TFR3 e 4) e P. hominis (TH3 e 5). A an?lise morfol?gica dos trofozo?tos revelou a presen?a de cinco flagelos anteriores, n?cleo redondo e ax?stilo afunilando de maneira uniforme, caracter?stico de P. hominis. Os resultados da morfologia foram confirmados pelo estudo molecular. O sequenciamento dos isolados revelou 100% de similaridade de sequ?ncia com P. hominis isolado de gato e de c?o depositados no Genbank. Este ? o primeiro estudo realizado no Brasil sobre a presen?a de parabasal?deos em gatos utilizando dados morfol?gicos e moleculares e o ?nico na literatura onde somente P. hominis foi isolado. Na segunda parte deste estudo, 77 amostras de fezes de gatos provenientes do atendimento cl?nico do HVPA-UFRRJ foram examinadas. Somente quatro amostras apresentaram positivas. Burk. (Leguminosae, Papilionoidae, Millettiedae) on samples of Boophilus microplus (Canestrini, 1887) in bovines of the Para?ba Valley region ? S?o Paulo, Brasil. The ?in vitro? trials were carried out using immersion of engorged females, and ?in vivo? on bovines in the field by means of artificial infestation. The standard dilution was made from one part of root powder and three parts of ethanol this was considered the 100% solution. To determine the more efficient solvent to extract rotenone from the root, laboratory trials were conducted on B. microplus engorged females with in three solvents: water, ethanol a.p. and acetone a.p. The laboratory trials permitted the calculation of lethal doses at 90% (LD90) and 50% (LD50) for larvae between seventh and fourtheenth days of application and efficient concentrations at 90% (EC90) and 50% (EC50) on strain local and Mozo with B. microplus engorged females. For the larvae of the local strain (Para?ba Valley Regional Hub) LD50 calculated was 1:31,37 mL and LD90 was 1:85,24 mL. The EC90 for engorged females, of the same strain was of 1:10,19 mL and the EC50 1:34,94 mL. For the engorged females on the hypersersitive strain Mozo the EC90 was 1:23,91 mL and the EC50 1:60,46 mL. The mortality of engorged females in relation to the different kinds of solvents, was analyzed. In the field, the best results (76,10% of control) were obtained three days after application of the product extracted in ethanol, in 1:10, on animals. Then gradually between the seventh and fourteenth days the products lost efficiency, there was no significant difference between treatments and the control group after 21 days. / Foram realizados testes laboratoriais e de campo para avaliar a efici?ncia do extrato de ra?zes da planta Dahlstedtia pentaphylla (Taub.) Para as larvas da cepa local (Polo Regional do Vale do Para?ba) a DL50 calculada foi de 1: 231,37 mL e DL90 1: 85,24 mL. A CE90 para as tele?ginas, da mesma cepa, foi de 1:10,19 mL e a CE50 1: 34,94 mL. Para tele?ginas da cepa sens?vel Mozo a CE90 foi de 1: 23,91 mL e a CE50, 1: 60,46 mL. Analisou-se tamb?m a mortalidade das tele?ginas frente aos diferentes solventes. No campo os melhores resultados obtidos (76,10% de controle) foram obtidos tr?s dias ap?s a aplica??o do produto extra?do em etanol, na dilui??o 1: 10, sobre os animais. A range of 101 blood samples were collected from Gallus gallus in poultry rustic breeds of in Rio de Janeiro, Brazil. The collected bloods were used to prepare blood smears and to extract the deoxyribonucleic acid (DNA) of these samples. There with, molecular protocols were tested, such as the already published conventional PCR (cPCR) and nested PCR (nPCR), designed for Plasmodium spp. In this study two qPCR protocols were developed using primers targeting the Cyt b and 18S rDNA genes. The qPCR detection limit for both genes was 10 copies of the target DNA, which were higher than the detection limit observed in nPCR and cPCR. In qPCR, 69.30% (n = 70/101) samples were positive targeting 18Sr DNA gene and 59.40% (n = 60/101) samples were positive targeting Cyt b gene. In nPCR and cPCR, 54.45% (n = 55/101) and 52.47% (n= 53/101) samples were positive, respectively. In blood smear microscopy, 31 (30.69%) samples were positive. There was no disagreement between the results (p > 0.05) of qPCR for 18Sr DNA and Cyt b genes. Additionally, qPCR was more sensitive than the other techniques discussed, mostly related to blood smear microscopy (p < 0.05). Therefore, the two qPCR developed in the study showed more sensitivity than other techniques and enabled the detection of Plasmodium spp. in poultry even in low parasitemia. / Este estudo teve como objetivo desenvolver um ensaio de PCR em tempo real (qPCR) para o diagn?stico de Plasmodium spp. utilizando os genes 18S rDNA e cyt b. Foram coletadas 101 amostras de sangue de aves da esp?cie Gallus gallus de cria??o r?stica ou org?nica no munic?pio de Serop?dica, Rio de Janeiro. Os sangues coletados foram utilizados na prepara??o de esfrega?os de sangue e para extra??o do ?cido desoxirribonucl?ico (DNA) destas amostras. Protocolos de ensaios moleculares foram testados, tal como o PCR convencional (cPCR), nestedPCR (nPCR) e qPCR para Plasmodium spp. Neste estudo dois protocolos de qPCR foram desenvolvidos utilizando oligoiniciadores desenhados com alvo no citocromo b e 18S rDNA. O limite de detec??o para os dois genes qPCR foi de 10 c?pias do alvo de DNA, que foram maiores do que o limite de detec??o observado em nPCR e cPCR. Em qPCR, 69,30% (n = 70/101) amostras foram positivas com alvo no gene 18SrDNA e 59,40%(n = 60/101) amostras positivas com alvo no gene cyt b. The objectives of this study were to delineate a multiplex PCR technique for the simultaneous diagnostic of microorganisms of Babesia and Ehrlichia genera in canine blood samples, and to realize the partial characterization of fragments of the 16S rRNA gene of the family Anaplasmataceae agents and, of 18S rRNA gene of Babesia detected in some samples PCR-positive, comparing the sequences obtained with sequences of other strains previously deposited in GenBank. Total DNA of 119 blood samples was extracted, of these, 40 were selected by showing cytoplasmatic inclusions in leukocytes and/or platelets suggesting infection by agents of Anaplasmataceae family (1E to 40E), 37 by showing piroplasms (1B to 37B), and two by presenting structures of both agents (M1 and M2), and finally, 40 samples with negative parasitological diagnostic and hematological exam without alterations. All these samples were tested by PCR to confirm the absence or presence of these hemoparasites, and them utilized in the multiplex PCR delineation. In multiplex PCR reactions the primers A17/EC3 were used to amplify an approximately 600bp region of the 16S rRNA gene of Ehrlichia species and the primers PIRO-A1/PIRO-B were used to amplify an approximately 450bp region of the 18S rRNA gene of Babesia species. Validation of multiplex PCR was performed by real time multiplex PCR. The multiplex PCR was able to simultaneously detect both agents in a DNA sample of a dog naturally co-infected and all the single infections by Babesia, but does not detected all the Ehrlichia infections. The real-time multiplex PCR was more sensitive in detect both single and also co-infections, as well as positive DNA mixtures for the two agents. The sequencing results confirmed the isolates identity, and that the primers PIRO-A1/PIRO-B also amplified the DNA of Hepatozoon canis. Phylogenetic analysis indicated that E. canis, A. platys, B. canis and H. canis species found in this study showed close similarities with sequences previously deposited in GenBank, forming monophyletic groups. / Os c?es podem se infectar com diversos hemoparasitos, sendo muito comum a ocorr?ncia de coinfec??es entre as esp?cies Ehrlichia canis, Babesia canis, Anaplasma platys e Hepatozoon canis, visto que possuem o mesmo carrapato vetor. Este estudo teve como objetivos delinear uma t?cnica de PCR multiplex para diagnosticar simultaneamente microrganismos dos g?neros Ehrlichia e Babesia em amostras de sangue de c?es e realizar a caracteriza??o parcial de fragmentos do gene 16S rRNA de agentes da fam?lia Anaplasmataceae e do gene 18S rRNA de Babesia detectados em algumas amostras positivas pela PCR, comparando as sequ?ncias obtidas com as sequ?ncias de outras cepas depositadas previamente no GenBank. Nas rea??es de PCR multiplex utilizou-se os oligonucleot?deos iniciadores A17/EC3 que amplificam um produto de aproximadamente 600pb de uma por??o do gene 16S rRNA de esp?cies de Ehrlichia e os oligonucleot?deos iniciadores PIRO-A1/PIRO-B que amplificam um produto de aproximadamente 450pb de uma por??o do gene 18Sr RNA de esp?cies de Babesia. A valida??o da PCR multiplex foi realizada por PCR multiplex em tempo-real. A PCR multiplex foi capaz de detectar simultaneamente os dois agentes em uma amostra de DNA de um c?o naturalmente coinfectado e todas as infec??es individuais por Babesia, mas n?o detectou todas as infec??es por Ehrlichia. A PCR multiplex em tempo real foi mais sens?vel em detectar tanto infec??es ?nicas quanto coinfec??es, al?m de misturas de DNA positivo para os dois agentes. As an?lises filogen?ticas indicaram que as esp?cies de E. canis, A. platys, B. canis e H. canis encontradas neste estudo possuem similaridades pr?ximas com sequ?ncias previamente depositadas no GenBank, formando grupos monofil?ticos.

The experiment was divided into two assays: (i) The first assay was carried out to determine the effects of different concentrations of piperine (0, 60, 120, 180 ppm) and it?s possible toxicity in broiler chickens diets. Ninety six male chicks (Cobb), seven days old were used, being randomly allocated into four experimental groups (n=24) during 35 consecutive days. The following parameters were evaluated: biochemical, hematological, histopathological (proventriculus, gizzard, liver, kidney), histomorphometric (small intestine) and zootecnic. The concentration of 60 ppm of piperine in the diet was safe for broilers, showing better performance of broilers on period from 36 to 42 days old. The concentration of 180 ppm caused leukopenia and concentrations of 120 and 180 ppm was observed decrease in the number of heterophils and monocytes. Hepatotoxicity was observed by elevated AST enzyme activity, histopathological changes and decreased absorption surface in the segments (jejunum and ileum) of small intestine were observed for both 120 and 180 ppm concentrations. (ii) In the second assay, 60 broilers with nine days old divided into four groups: control, piperine (60 ppm added to diet), aflatoxin B1 (0.5 mg of aflatoxin B1.Kg-1 of body weight, orally) and piperine associated aflatoxin B1, were evaluated by effect chemoprotector of piperine against toxics effects of aflatoxin B1 being evaluated for zootecnic, biochemical, histopathological and histomorphometric parameters, toxic heterophils in peripheral blood and genotoxic by comet assay and micronucleus were also determined. No changes in the performance parameters were observed after this experiment. Broiler chickens intoxicated with AFB1 (0.5 mg of aflatoxin B1.kg-1 of body weight ) showed: decreased body weight gain and increased feed conversion; reduced carcass and cuts yields; liver toxicity, with increased relative weight of the liver and heart, macroscopic variations of hepatic parenchyma and increase of liver enzymes activity; kidney enzymes increase without evidence of renal tissue damage macroscopic or microscopic; leukopenia with significant reduction of lymphocytes and heterophils; reduction in absorptive surface due to the reduction of the length and width of the villi of all studied segments of small intestine; presence of toxic heterophils. The cytotoxic and genotoxic effects of aflatoxin B1 described above were significantly reduced or absent in the group of broiler intoxicated with aflatoxin B1 and fed with with piperine. No significant difference between piperine associated aflatoxin B1 in control and piperine groups were observed. The addition of 60 ppm of piperine in the diet of broiler chickens was safe, promoting beneficial effect both in zootecnic parameters and in poultry health, preventing toxic effects of aflatoxin B1in broiler chickens. / A interfer?ncia da piperina (amida extra?da da pimenta do reino) adicionada ? ra??o de frangos de corte intoxicados experimentalmente por aflatoxina B1 (micotoxina de grande relev?ncia no setor av?cola) e sua capacidade quimioprotetora foram o principal objetivo deste trabalho. O experimento foi dividido em dois ensaios: (i) O primeiro ensaio foi realizado para determinar os efeitos de diferentes concentra??es de piperina (0, 60, 120 e 180 ppm) foram avaliados e sua poss?vel toxicidade. Noventa e seis pintos (Cobb), com 7 dias de idade foram divididos aleatoriamente em 4 grupos (n=24), por 35 dias consecutivos. Os par?metros avaliados foram: hematol?gicos, bioqu?micos, histopatol?gicos (proventr?culo, moela, f?gado e rim), histomorfom?trico (intestino delgado) e par?metros zoot?cnicos. A concentra??o de 60 ppm de piperina adicionada ? ra??o foi segura para frangos de corte, tendo ainda resultado em melhor desempenho dos frangos na fase final (36-42 dias de idade). A concentra??o de 180 ppm promoveu leucopenia e nas concentra??es de 120 e 180 ppm foi observada diminui??o do n?mero de heter?filos e mon?citos; hepatotoxicidade, com eleva??o da enzima AST e altera??es histopatol?gicas em ambas as concentra??es; diminui??o da superf?cie de absor??o nos segmentos (jejuno e ?leo) do intestino delgado, por?m, sem altera??o dos par?metros zoot?cnicos. (ii) Para o segundo ensaio com a concentra??o de 60ppm de piperina: 60 frangos com 9 dias de idade, foram divididos em 4 grupos (n=15): grupo controle, grupo aflatoxina B1 (0,5 mg aflatoxina B1.kg-1 de peso vivo por via oral), grupo piperina (60 ppm adicionada ? ra??o) e grupo piperina associada a aflatoxina B1, determinando-se a capacidade quimioprotetora da piperina sendo avaliados os par?metros zoot?cnicos, hematol?gicos, bioqu?micos, histopatol?gicos, histomorfom?tricos, os efeitos genot?xicos da aflatoxina B1 pelo teste do cometa e do micron?cleo, presen?a de heter?filos t?xicos no sangue perif?rico. Os frangos intoxicados com aflatoxina B1 (0,5 mg de aflatoxina B1.Kg-1 de peso vivo) apresentaram: diminui??o do ganho m?dio de peso e piora da convers?o alimentar; diminui??o do rendimento de carca?a e cortes; hepatotoxicidade, com aumento de peso relativo do f?gado e cora??o, varia??es macrosc?picas do par?nquima hep?tico e eleva??o das enzimas hep?ticas; aumento das enzimas renais, sem evid?ncia de les?es macrosc?picas e microsc?picas no tecido renal; leucopenia, com diminui??o significativa de linf?citos e heter?filos; diminui??o da superf?cie de absor??o em fun??o da redu??o do comprimento e largura das vilosidades de todos os segmentos estudados do intestino delgado; presen?a de heter?filos t?xicos. Os efeitos citot?xicos e genot?xicos da aflatoxina B1 foram significativamente reduzidos ou ausentes no grupo piperina associada a aflatoxina B1, sem diferen?a significativa entre o grupo controle e piperina. A ra??o de frangos de corte com 60 ppm de piperina foi segura, promovendo efeito ben?fico tanto nos par?metros zoot?cnicos avaliados, como na sanidade av?cola, por impedir os efeitos t?xicos da aflatoxina B1 em frangos de corte.
