O papel do trânsito de cálcio e de compostos da matriz extracelular no modelo de insuficiência aórtica aguda experimental em ratos

Objetivos: a) Comparar alterações morfofuncionais, celulares (hipertrofia) e intersticiais (fibrose) em diferentes momentos da evolução da insuficiência aórtica; b) Verificar quais dos seguintes mecanismos estão envolvidos na remodelação cardíaca induzida pela IAo e em qual momento esta alteração acontece: alteração na metaloprotease 2 (MMP2) e do inibidor tecidual de metaloprotease 1 (TIMP1), alterações dos RNAs mensageiros específicos para a codificação das proteínas envolvidas na homeostase do cálcio (Fosfolambam, Ryr e Serca2a), alterações da expressão de proteínas fosfolambam e Serca2a do trânsito de cálcio. Casuística e Métodos: Estudo experimental com 64 ratos Wistar machos, 32 animais submetidos à insuficiência aórtica aguda (grupo IAo) e 32 animais a procedimento simulado (grupo Controle). Todos os animais foram seguidos com 1, 4, 8 e 12 semanas através de ecocardiogramas seriados e, após eutanásia, foram analisada morfometria do tecido cardíaco, atividade da MMP2 e TIMP-1, expressão gênica por Reação em Cadeia da Polimerase em Tempo Real das proteínas do trânsito de cálcio, expressão das proteínas Serca2a e fosfolambam pela técnica Western Blot. A análise estatística foi efetuada pela ANOVA de dois fatores; o pós teste de Holm Sidak; teste t grupo a grupo; Anova de 1 via, complementada por Tukey; correção por Bonferroni; teste de correlação de Spearman e teste de correlação de Pearson. Em todos os casos, o nível de significância adotado foi p<0,05. Resultados: Observou-se, na primeira semana, que o peso do VE e a pressão diastólica foi maior no grupo IAo. Na quarta semana, MMP2, TIMP-1, iMVE e fração de colágeno foram maiores no grupo IAo. Objectives: a) to compare morphological, cell (hypertrophy) and interstitial (fibrosis) changes at different times of the evolution of aortic regurgitation (AR); b) to analyze which of the following mechanisms are involved in cardiac remodeling induced by aortic failure and at what moment this change takes place: change in the metalloprotease 2 (MMP 2) and tissue inhibitor of metalloproteinase 1 (TIMP1); changes of messenger RNAs to code proteins involved in calcium homeostasis (phospholamban, Ryr and SERCA2a), changes in expression of phospholamban and SERCA2a in the calcium transit. Methods: Experimental study with 64 male Wistar rats, 32 animals submitted to acute aortic regurgitation (AR group) and 32 animals sham procedure (Sham group). All animals were followed with 1, 4, 8 and 12 weeks by serial echocardiography and after euthanasia, were analyzed morphometry of cardiac tissue, the activity of MMP2 and TIMP-1, gene expression by RT-PCR of calcium homeostasis protein, SERCA2a and phospholamban expression by Western Blotting. In all cases, the significance level was set at p <0.05. Results: In the first week it was observed that weight of the LV and diastolic blood pressure were higher in the AR group. A proteína BthA-I é uma fosfolipase A2 (PLA2) ácida com elevada atividade catalítica e que possui a capacidade de inibir a agregação de plaquetas e reduzir a pressão arterial. Cristais da BthA-I nativa e de sua forma quimicamente modificada pelo pBPB (brometo de p-bromofenacila), um conhecido inibidor de a PLA2s que também é capaz de suprimir os mecanismos farmacológicos próprios desta proteína, foram obtidos e submetidos a experimentos de difração de raios-X. A estrutura cristalina da BthA-I foi elucidada a 1,9 Å de resolução e o processamento dos dados experimentais revelou a presença de dois monômeros na unidade assimétrica. A análise estrutural destes monômeros revelou que a BthA-I dimérica possui uma conformação oligomérica distinta das demais PLA2s, onde os sítios catalíticos da molécula encontram-se expostos ao solvente. Apesar das condições de cristalização da BthA-I nativa, é possível sugerir que a forma dimérica desta proteína seja aquela encontrada predominantemente in vivo. A determinação estrutural da BthA-I quimicamente modificada pelo pBPB a 1,85 Å de resolução revelou a presença inequívoca de moléculas de inibidor covalentemente ligadas aos resíduos His48, localizados nos sítios de ligação de substrato dos dois monômeros do complexo. Três regiões do complexo BthA-I/pBPB são estruturalmente alteradas pela ligação do inibidor: os loops de ligação de Ca++ e as regiões de β-wing C-terminais. Estas mudanças estruturais são provavelmente derivadas da conformação oligomérica apresentada pelo complexo, cuja disposição pode explicar a supressão das atividades farmacológicas da BthA-I... BthA-I, a high catalytic acidic phospholipase A2 (PLA2) with platelet aggregation inhibition, anticoagulant and hypotensive properties, was crystallized in its native form and complexed to pBPB (p-bromophenacyl bromide), a well-known inhibitor of PLA2s which also abolishes the biochemical activities of this protein. The crystal structure of BthA-I native was solved at 1.9 Å and the data processing showed two monomers in the unit cell. A novel dimeric conformation of this native protein with the active sites of the monomers exposed to the solvent was observed. Despite of the conditions used in the crystallization experiments, it is possible to suggest that the dimeric form must be predominantly found in vivo. The structural determination of BthA-I chemically modified by pBPB at 1.85 Å revealed the unambiguous identification of inhibitor molecules covalently bound to residues His48 in the substrate-binding clefts of both monomers of the complex. Three regions of BthA-I/pBPB complex are structurally altered by the binding of the inhibitor: the Ca++ -binding loops, the β-wings and the C-terminal regions. These structural changes are probably derived from the oligomeric state presented by the complex, which could explain the abolition of pharmacological activities of the pBPB-chemically modified BthA-I. A residue present in the "pancreatic" loops (Lys69), which is usually related to the anticoagulant effect of the BthA-I, is in the dimeric interface of the complex, supporting this hypothesis regarding the abolition of this and other activities caused by pBPB. Metaloproteases de carrapato : perspectiva de antígeno para vacina

As metaloproteases (MPs) são proteínas que participam em diversos processos fisiológicos e patológicos. Neste trabalho, relatamos a identificação de MPs em três carrapatos de importância econômica: Ixodes persulcatus (Ip-MPs), Rhipicephalus sanguineus (Rs-MPs) e Rhipicephalus microplus (BrRm-MPs). O perfil transcricional em vários tecidos e fases de vida mostra que MPs são transcritas em glândula salivar durante a alimentação de fêmeas e não são transcritos no macho (com exceção de uma MP, BrRm-MP4) e ovos, o que sugere que esta família da proteínas são componentes funcionais necessários para interferir nas defesas do hospedeiro, apoiando o processo de hematofagia. A presença de um sítio de ligação de zinco, uma dobra“Met-turn” e um domínio rico em cisteína na região C-terminal indica que todas os transcritos obtidos codificam para a família de MPs reproplisina (metzincina). Uma das MPs de R. microplus (BrRm-MP4) foi selecionada para uma investigação mais aprofundada quanto ao potencial como um antígeno vacinal, devido ao seu padrão de transcrição ubíquos e devido àprevisão in silico de epítopos imunogênicos, em comparação com as demais MPs identificadas. BrRm-MP4 recombinante (rBrRm-MP4) foi expressa em Escherichia coli e testadas como um antígeno contra infestação de R. microplus. Imunoblot mostrou que o soro de bovinos imunizados com rBrRm-MP4 reconhece BrRm-MP4 da glândula salivar, ovário e larvas de R. microplus. Comparando com o controle, a vacinação com rBrRm-MP4 proporcionou uma redução de 43% no número de fêmeas adultas, redução de 14,80% na postura de ovos e redução de 17,53 % na capacidade de eclosão da larva, realizando um proteção total de 60% estatisticamente significativa. Os resultados indicam que rBrRm-MP4 é um potencial candidato a ser incluído como imunógeno em uma vacina anti-carrapato. / Metalloproteases (MPs) are proteins participating in several physiological and pathological processes. Here, we report the identification of MPs in three economically important ticks: Ixodes persulcatus (Ip-MPs), Rhipicephalus sanguineus (Rs-MPs) and Rhipicephalus microplus (BrRm-MPs). Transcriptional profile in various tissues and life stages revealed the presence of all MPs transcripts in salivary glands during feeding stages and absence in males (except one MP, BrRm-MP4) and eggs, suggesting this family proteins are functional components required to interfere with host defenses, supporting tick hematophagy. The presence of a zinc binding site, a “Met-turn” and C-terminal cysteine rich domain indicates all obtained transcripts encode for proteins belonging to the reproplysin (metzincin) family of MPs. A R. microplus MP (BrRm-MP4) was selected for further investigation concerning its potential as a vaccinal antigen due to its ubiquitous transcription pattern and in silico prediction of immunogenic epitopes, comparing to other obtained MPs. Recombinant BrRm-MP4 (rBrRm-MP4) was expressed in Escherichia coli and tested as an immune-protective antigen against R. microplus infestation. Immunoblot showed specific antibodies against rBrRm-MP4 in immunized calves sera. Sera from immunized calves recognized BrRm-MP4 in R. microplus salivary gland, ovary and larvae. Comparing to ticks reared non-vaccinated calves, rBrRm-MP4 provided 43% reduction in adult female number, 14.80% reduction in egg laying and 17.53% reduction in larva hatching capacity, performing an overall 60% statistically significant protection. We assume rBrRm-MP4 is a potential candidate to be included as immmunogen for further development in an anti-tick vaccine.

Esses indivíduos apresentam, desde o nascimento, dificuldades na alimentação, disturbios ósseos maxilar e mandibular, na arcada dentária, na fala e, em alguns casos, na audição. Além disto, fissurados e seus familiares enfrentam sérios problemas psicológicos decorrentes do comprometimento estético causado pela anomalia. Sem dúvida alguma, a morfogênese do palato secundário é um dos pontos mais críticos na formação da face e do palato. Depende de uma sequência complexa de eventos que possuem em comum o remodelamento de matriz extracelular (MEC), regulado, principalmente, pelas metaloproteinases de matriz (MMPs). Esta éa primeira vez que RECK, uma proteína indutora da reversão tumoral, rica em cisteína e com motivos kazal, é pesquisada neste processo. O primeiro objetivo deste estudo foi investigar se há expressão de RECK durante os eventos envolvidos na formação do palato secundário em camundongos. A partir dos resultados obtidos, indicando a presença de RECK nesses eventos, objetivou-se estabelecer um padrão temporal e espacial dessa expressão, assim como analisar o padrão de distribuição de MMP-2 e MMP-9, comparando os resultados obtidos entre si. Para tanto, cabeças inteiras e palatos dissecados de embriões de camundongos com 13; 14; 14,5 e 15 dias de desenvolvimento embrionário (DE) foram submetidos a reações de PCR em tempo real, hibridização in situ, imunoistoquímica e zimografia. As reações de PCR em tempo real indicaram a presença de mRNA de RECK (mRECK) em todos os períodos analisados, os níveis do transcrito aumentaram gradativamente e foram significativamente diferentes entre 13 e 15DE e entre 14 e lSDE. Os resultados de hibridização in situ e imunoistoquímica apontaram, consecutivamente, um padrão variável de distribuição de mRECK e da proteína RECK no mesênquima, epitélio e linha de junção (LI) formada entre as paredes palatais, no transcorrer dos períodos analisados. Com relação ao padrão de expressão das MMPs, a análise imunoistoquímica revelou a presença de MMP-2 no mesênquima, epitélio e LI em todos os períodos. A MMP-9 foi expressa no 14DE, 14,SDE e lSDE na LI e mesênquima palatal. A análise zimográfica confirmou a atividade de MMP-2 e MMP-9. Dessa forma, concluímos que RECK está presente nos principais eventos de formação do palato secundário, apresentando um padrão de expressão variável no tempo e espaço, que quando compar~o ao padrão de " expressão de MMP-2 e MMP-9 apresentam, na maioria das vezes, similaridades entre si. Esses , achados sugerem um provável envolvimento dessas moléculas na organogênese do palato secundário e uma possível interação/modulação entre elas / Abstract: The oral clefts show a great influence in the size of their hosts. They have, since birthy, difficulties in alimentation, problems in the maxilar bones, in dental arch, in speech, and in some cases, in hearing. In addition, the hosts and their families have serious psychological problems due to the esthetic lenght caused by anomalia. With no doubt, the formations of the secondary palate is one of the most critial phases during the formation of the surface and of the palate. Depending on a sequente of complex events that have in common the remodeling of the extracellular matrix (ECM), mainly regulated by matrix metalloproteinases (MMPs). This is first time that the presence of RECK, a reversion-inducing cystein-rich protein with kazal motifs, this is process is researched. The first objective of this study was to investigate the RECK expression during events involved in formation of the secondary palate in mice. Based on the results obtained, which indicated the presence of RECK in these events, the objective was to establish a temporal and spatial pattern of such expression, as well as to analyze the pattern of distribution of MMP-2 and MMP-9, comparing the results obtained to each other. For that purpose, whole heads and dissected palates of mice aged 13, 14, 14.5 and 15 days of embryonic development (ED) were submitted to real-time PCR, in situ hybridization, immunohistochemistry and zymography. The real-time PCR indicatedthe presence of mRNA of RECK (mRECK) in all periods analyzed; the transcript levels were gradually increased and were significant1y different between 13 and 15ED and between 14 and 15ED. The results of in situ hybridization and immunohismchemistry consecutively indicated a variable pattern of distribution of mRECK and RECK protein in the mesenchyme, epithelium and in midline edge seam (MES) formed between the palatal shelves during the study periods. With regard to the pattern of expression of MMPs, immunohistochemical analysis revealed the presence of MMP- 2 in the mesenchyme, epithelium and MES in all periods. The MMP-9 was expressed in the MES and palatal mesenchyme at 14ED, 14.5ED and 15ED. Zymographic analysis confirmed the activity of MMP- 2 and MMP-9. Therefore, it was concluded that RECK is present in the main events of formation of the secundary pala te, presenting a variable pattern of expression along time and space, which often present similarities to each other compared to the pattern of expression of MMP-2 and MMP-9. Novos enfoques no estudo dos mecanismos de ação de metaloproteinases e outras proteínas tóxicas envolvidas no envenenemento ofídico e lonômico

Venenos e outras secreções animais vêm sendo explorados como fonte de substâncias ativas na hemostasia em mamíferos. Esses princípios ativos, com funções primárias na alimentação e defesa, afetam elementos chave de quase todas as vias fisiológicas animais, tendo um enorme potencial como base para o desenvolvimento de novas drogas. No envenenamento ofídico, hemorragias locais e sistêmicas são causadas em grande parte pela ação de metaloproteinases hemorrágicas presentes nos venenos (SVMPs). Os estudos com metaloproteinases até então descritos baseiam-se principalmente em metodologias bioquímicas clássicas. Neste trabalho, as metaloproteinases e suas interações com outras proteínas são abordadas através de técnicas de dinâmica molecular, ressonância plasmônica de superfície e proteômica quantitativa. O perfil de inibição da atividade proteolítica de SVMPs por inibidores teciduais de metaloproteinases (TIMPs) foi avaliado. Experimentos de modelagem e dinâmica molecular mostraram similaridades nas bases moleculares da interação das TIMPs tanto com SVMPs quanto com metaloproteinases endógenas (MMPs e ADAMs), seus alvos fisiológicos. O domínio rico-em-cisteínas das SVMPs da classe PIII está envolvido na interação com proteínas de matriz extracelular. Os sítios de interação do domínio rico-em-cisteínas de jararragina com fator de von Willebrand (vWF) foram identificados por ressonância plasmônica de superfície. Um modelo de interação entre os domínios das duas proteínas foi proposto. Considerando a presença de domínios A de vWF em diversas proteínas de matriz extracelular, o domínio de rico-em-cisteínas funcionaria direcionando as metaloproteinases para substratos específicos, promovendo hemorragia. Além disso, através de uma metodologia de proteômica quantitativa, foi verificada pela primeira vez a ação proteolítica de metaloproteinases em proteínas de matriz extracelular íntegra de células em cultura. Já no envenenamento lonômico, um quadro clínico hemorrágico é observado apesar da ausência de metaloproteinases no veneno. O estudo de proteínas tóxicas da lagarta Lonomia obliqua baseia-se no isolamento e caracterização bioquímica dos princípios ativos relacionados com aspectos da patologia do envenenamento. Nos últimos anos, esses estudos se focaram em uma única secreção da lagarta, identificando o seu potencial pró-coagulante, fibrin(ogen)olítico, hemolítico, edematogênico e nociceptivo. Neste trabalho, foram identificadas diferenças quali-quantitativas em quatro secreções venenosas de L. obliqua. A diferente participação de cada uma dessas secreções durante o contato com as lagartas pode levar a grande variabilidade de sintomas e gravidade do acidente. Utilizando a metodologia de microarranjos de DNA, analisamos o efeito do veneno da lagarta no padrão de expressão gênica de células em cultura e identificamos a expressão aumentada de genes possivelmente relacionados ao quadro clínico do envenenamento. Proteínas potencialmente tóxicas de L. obliqua descritas em um estudo de transcriptoma, juntamente com os dados de expressão gênica aqui demonstrados possibilitam uma maior compreensão dos efeitos do veneno na hemostasia e sintomas do lonomismo. / Animal venoms and other secretions have been explored as source of active substances on the mammal hemostasis. These active principles, which have primary function in feeding and defense, act on key elements of almost all physiologic pathways and have an enormous potential in the development of new therapeutic drugs. In ophidic envenomations, local and systemic hemorrhages are caused mainly by the action of hemorrhagic metalloproteinases from the venom (SVMPs). Previous studies with metalloproteinases were based mostly in classical biochemical methodologies. Here, the metalloproteinases and their interactions with other proteins were examined by molecular dynamics, surface plasmon resonance and quantitative proteomics methodologies. The inhibition profile of the SVMPs proteolytic activity by tissue inhibitors of metalloproteinases (TIMPs) was evaluated. Modeling and molecular dynamics procedures showed similarities in the molecular bases of the interaction of TIMPs both with SVMPS and endogenous metalloproteinases (MMPs and ADAMs). PIII SVMP cysteine-rich domain is involved in the interaction with extracellular matrix proteins. Using surface plasmon resonance, we identified the interaction sites between jararhagin cysteine-rich domain and von WIllebrand factor (vWF). An interaction model of these two proteins was proposed. Considering the presence of vWF A domains in several matrix proteins, the cysteine-rich domain probably works targeting the metalloproteinases to specific substrates, leading to hemorrhage. Moreover, using a quantitative proteomic approach, it was shown for the first time the metalloproteinases proteolytic action upon organized extracellular matrix proteins in culture cells. In lonomic envenomation, a hemorrhagic clinical profile is observed regardless the absence of metalloproteinases. The caterpillar's toxic proteins studies are based on isolation and biochemical characterization of active principles related to the patophisiology of the envenomation. In the last few years, these studies were focused on one caterpillar's secretion, containing pro-coagulant, fibrin(ogen)olytic, hemolytic, edematogenic and nociceptive activities. In this work, we identified quali-quantitative differences in four Lonomia obliqua venomous secretions. The different participation of each of these secretions during the accident could lead to the variability of symptoms and severity of the envenomation. Using a microarray methodology, we analyzed the effects of the caterpillar venom on the cultured cells gene expression profile and identified increased expression of genes possibly involved with the clinical manifestations. Potentially toxic proteins from L. obliqua identified in a transcriptome study, together with the gene expression data described here allow a better understanding of the venom effects in hemostasis during lonomism.

A enzima metabólica ácido graxo sintase (FASN, EC2.3.1.85) é a responsável por catalisar a síntese de ácidos graxos saturados de cadeia longa. FASN é um homodímero com massa molecular de 250 kDa responsável pela produção do palmitato endógeno. Em tecidos normais, a atividade metabólica de FASN é mínima, uma vez que a maior parte dos ácidos graxos usados pelas células provém da dieta, com exceção dos tecidos lipogênicos. Por outro lado, tem sido demonstrado que nas células malignas a maior parte dos ácidos graxos provém da biossíntese endógena conseqüente ao aumento da atividade de FASN e que existe uma associação positiva entre a expressão desta enzima metabólica e o comportamento agressivo de tumores malignos, pois sua alta expressão ocorre principalmente em casos com prognóstico ruim. Inibidores específicos da atividade FASN bloqueiam a síntese de DNA e causam apoptose em linhagens celulares derivadas de neoplasias malignas de próstata, mama, cólon, estômago, intestino, endométrio, cavidade bucal, ovário e melanoma. A droga orlistat (Xenical®), aprovada pela FDA e utilizada para o tratamento de obesidade, foi descrita como tendo propriedades anti-neoplásicas em câncer de próstata, mama, cólon, estômago e melanoma, devido a sua capacidade de bloquear especificamente a atividade de FASN. Este trabalho teve como objetivo principal estudar o efeito do tratamento de camundongos C57BL6 com a droga orlistat sobre as atividades de MMPs, expressão de integrinas por células B16F10, adesão destas últimas à macromoléculas da MEC e formação de colônias pulmonares a partir da inoculação na veia caudal de camundongos C57BL6. Através de ensaios zimográficos, não observamos alterações das atividades de MMP-2 e -9 em células B16F10 tratadas com orlistat. No entanto, a inibição de FASN aumentou a adesão das células B16F10 aos componentes de matrix extracelular laminina e fibronectina. Através de ensaios de imunofluorescência observamos uma redução das regiões de adesão focal das integrinas ?v?3 nas células B16F10 tratadas com orlistat. Finalmente, o tratamento com orlistat reduziu em 53,6% o número de colônias metastáticas pulmonares, em comparação aos grupos controle. Em conjunto, os resultados aqui descritos sugerem que esta FASN é um alvo terapêutico em potencial para estes tumores. / Abstract: Malignant melanoma has poor prognosis due to its high metastatic potential and resistance to the existing chemotherapeutic agents. Fatty acid synthase (FASN, EC2.3.1.58) is a metabolic enzyme with molecular mass of 250 kDa responsible for the endogenous biosynthesis of saturated long chain fatty acids. FASN activity is relatively low in most normal human tissues, since most of the fatty acids used by the cells come from the diet, except in liver, adipose tissue, fetal lung and lacting breast. On the other hand, it has been demonstrated that in several cultured malignant cells fatty acids are mostly produced by FASN. A similar phenomenon is also observed in melanoma cells and overexpression of FASN has been associated with a poor prognosis for patients with this malignancy. Specific inhibitors of FASN activity block DNA synthesis and cause apoptosis in prostate, breast, colon, stomach, endometrial, oral cavity, ovary and melanoma cancer cells lines. Orlistat (Xenical®), approved for FDA and used for the treatment of obesity, has antitumor properties in prostate, breast, colon, gastric cancers and melanoma, due to its capacity to block the FASN activity. This work had as main objectives to study the effect of orlistat on the expression and activity of MMPs and expression of integrins in B16F10 cells, as well as on the adhesion of these cells to ECM macromolecules fibronectin and laminin. Moreover, we sought to verify the effect of this drug on the lung colonization by B16F10 innoculated in the tail vein of C57BL6 mice. The treatment with orlistat did not change MMP-2 and -9 gelatinolytic activities in B16F10 cells and enhanced the adhesion of these cells on laminin or fibronectin. Interestingly, treatment of B16F10 cells with orlistat promoted a reduction on the number of integrin ?v?3 focal adhesion plates observed in the immunofluorescence assay. Finally, orlistat promoted an inhibition of 53,6% in the number of lung metastatic foci, in comparison with the control groups, further confirming the anticancer potential of FASN inhibitors. Esta inflamação pode estar associada a uma degradação das proteínas da matriz extracelular pelas metaloproteinases da matriz (MMPs). Dessa forma, o objetivo deste estudo foi avaliar os efeitos de exposição de cimentos endodônticos sobre a atividade gelatinolítica das MMP-2 e -9, produzidas por fibroblastos humanos. Fibroblastos da linhagem MRC5 (3x105 células/poço) foram incubados diretamente ou indiretamente com os cimentos AH Plus, Endomethasone N, Pulp Canal Sealer EWT e Sealapex nos períodos de 1/2h, 1h, 4h e 24h. A citotoxicidade dos cimentos foi determinada pela contagem de células viáveis, utilizando para isso o teste do azul de tripan. Sobrenadantes da cultura de células incubadas com os cimentos endodônticos, nas duas formas testadas, foram coletadas após cada período de exposição, com o objetivo de determinar os níveis de atividade gelatinolítica de MMP-2 e -9, pela técnica da zimografia. Os dados foram submetidos à análise de variância (ANOVA) e avaliados estatisticamente através do teste t (p<0,05). Os resultados mostraram haver uma maior atividade gelatinolítica de MMP-2 após os períodos de 4 e 24 horas, sem haver diferença entre os cimentos testados. Uma maior atividade gelatinolítica pode ser observada nas células que foram expostas ao cimento de forma direta, quando comparadas com aquelas que de receberam o contato indireto com o cimento (p<0,05). Nos períodos de tempo testados nenhuma atividade gelatinolítica pode ser observada no grupo controle, que não recebeu contato com os cimentos. Os resultados de citotoxicidade mostraram que os cimentos testados foram citotóxicos em ambas as formas de contato sendo que o Sealapex apresentou menor citotoxicidade e que o AH Plus foi o cimento mais citotóxico. Pode-se concluir que todos os cimentos endodônticos podem induzir a expressão de MMP-2 em fibroblastos MRC5 e que apesar de o AH Plus possuir a maior citotoxicidade, todos os cimentos testados apresentaram efeitos citotóxicos. / Abstract: Root canal sealers might be into contact with periapical tissues during root canal filling. This inflammation can be associated with extracellular matrix proteins degradation by matrix metalloproteinases (MMPs). The aim of this study was to investigate the effects of root canal sealers on the gelatinolytic acitivity of MMP-2 and -9 produced by human fibroblast cells. Human fibroblast cells MRC5 (3x105 cells/well) were incubated directly or indirectly with AH Plus, Endomethasone N, Pulp Canal Sealer EWT or Sealapex for 1/2h, 1h, 4h or 24h (timepoints). The cytotoxicity of all root canal sealers was determined by counting viable cells using the trypan blue assay. Supernatants of cell cultures incubated with root sealers, directly or indirectly, were collected after each time point to determine the levels of MMP-2 and MMP-9 gelatinolytic activity by gelatin zymography. Data were analyzed using ANOVA and t tests (p<0.05). The results showed that the cells secreted MMP-2 after the periods of 4 and 24 hours. However, there were no statistical differences between the sealers. Secretion of gelatinases was found to be elevated by the sealers in direct contact with the cell monolayer, when compared to the indirect contact (p<0.05). In the timepoints tested no MMP activity could be detected in the control group without the sealers. The cytotoxicity results showed that all the sealers were cytotoxic in both contact forms. These results indicated that Sealapex had a lower cytotoxicity while AH Plus was the most citotoxic endodontic sealer. In conclusion all root canal sealers can induce the expression of MMP-2 in MRC5 fibroblast cells. A placa bacteriana é seu agente etiológico principal, embora uma resposta imune exacerbada seja apontada como fator preponderante para o agravamento das lesões. Já nos primeiros estágios da doença é possível notar uma grande degradação da matriz extracelular local, principalmente de fibras colágenas. Dessa forma, o envolvimento das metaloproteinases de matriz (MMPs), uma família de enzimas com atividade dependente de íons metálicos que degradam proteínas estruturais da matriz extracelular, tem sido amplamente estudado nesta patologia. Baseado nestas evidências, o objetivo deste projeto foi caracterizar as principais modificações teciduais associadas ao modelo de doença periodontal induzida em ratos com ligaduras durante três, cinco ou sete dias. Foi analisada a microbiota associada à progressão inicial da doença, a disposição de fibras colagênicas e reticulares no tecido gengival e o perfil de células inflamatórias migrantes. Também foi avaliada a expressão e atividade enzimática de MMP-2 e -9, além da expressão gênica de MMP-2, -7, -9 e -14, e do inibidor RECK. A indução da doença com ligaduras pennitiu a formação de placa bacteriana caracterizada pela substituição de microorganismos Oram-positivos por Oram-negativos. O tecido gengival apresentou uma diminuição na área ocupada por fibras colagênicas, com a deposição de fibras mais espessas, características de um processo inicial de fibrose, aos sete dias de indução da resposta inflamatória. Não foram observadas alterações na disposição de fibras reticulares próximas à membrana basal do epitélio. Aos três e cinco dias de inflamação, respectivamente, foram observados picos de migração de neutrófilos e macrófagos. A MMP-2 apresentou um aumento crescente na atividade enzimática e expressão gênica até os sete dias de inflamação induzida, com a marcação de células epiteliais e fibroblastos na imunohistoquímica. A MMP-9 apresentou um pico na atividade enzimática aos três dias e um pico na expressão gênica aos cinco dias de inflamação, com a marcação imunohistoquímica de células epiteliais, fibroblastos e células inflamatórias migrantes para o sítio gengiva!. As MMPs-7 e -14, assim como o inibidor RECK, não apresentaram mudanças significativas em suas expressões gênicas. Os amplificados derivados de RT-PCR para MMP-7 apresentaram um fragmento correspondente a um íntron, indicando um possível mecanismo de regulação relacionado ao processamento do RNA mensageiro. Podemos concluir que o modelo de doença periodontal induzida por ligadura em ratos apresentou uma dinâmica bastante similar aos resultados já descritos para a evolução da doença no homem. A degradação de matriz extracelular observada foi coerente com o aumento na expressão e atividade enzimática de MMP-2 e -9, sugerindo que estas enzimas estejam relacionadas a este processo. Os resultados obtidos para a MMP-2 sugerem uma produção vinculada predominantemente a células próprias do tecido afetado pelo processo inflamatório. A MMP-9, no entanto, parece ter um aumento diretamente relacionado com a chegada de células inflamatórias no sítio gengival. Nossos estudos foram realizados com o intuito de elucidar os mecanismos moleculares da doença periodontal, buscando encontrar caminhos para o desenvolvimento de aplicações de potencial utilização diagnóstica ou terapêutica / Abstract: Periodontal disease is the major chronic disease affecting the human dentition. Bacteri plaque is its main etiological factor, although an exacerbated immune response is point< preponderant factor to the aggravation of the lesions. In the early stages of disease there is a gre degradation of the local extracellular matrix, mainly of the collagen fibers. That way, ti involvement of matrix metalloproteinases (MMPs), a family of enzymes with activity dependir on metallic ions that can degrade structural proteins of the extracelular matrix, it has been studit thoroughly in this pathology. Based on these evidences, the aim of this project was characterize the main tissue modifications associated to the model of periodontal disease induc~ in rats with ligatures during three, five or seven days. It was analyzed the microbiota associat~ to the initial development of the disease, the arrangement of collagen and reticulin fibers in ti gingival tissue and the migration of inflammatory cells. It was also evaluated the expression ar enzymatic activity ofMMP-2 and -9, besides the gene expression ofMMP-2, -7, -9 and -14, ar of the RECK inhibitor. The induction of disease using ligatures allowed the formation I bacterial plaque, characterized by substitution of Gram-positive for Gram-negati1 microorganisms. The gingival tis sue presented a decrease in the area occupied by collagen fibel with the formation of thicker fibers at seven days on induction of inflammatory response, usual found in fibrosis processes. Alterations were not observed for the organization of reticulin fibe elose to the basement membrane of the epithelium. To the three and five days of inflammatio respectively, peaks of neutrophils and macrophages migration were observed. MMP-2 present~ an increase in the enzymatic activity and gene expression until the seven days of induc~ inflammation, with the demarcation of epithelial cells and fibroblasts by immunohistochemistr MMP-9 presented a peak of enzymatic activity at the third day of inflammation and a peak I gene expression at the fifth day, with the demarcation of epithelial cells, fibroblasts ar migrating inflammatory cells to the gingival area. MMP-7 and -14, as well as the inhibitl RECK, did not present significant changes in their gene expressions. The amplified from R.' PCR for MMP-7 presented a fragment corresponding to an intron, indicating a possib regulation mechanism related to an altemative splicing of mRNA. The degradation ( extracellular matrix was coherent with the increased gene expression and enzymatic activity I MMP-2 and -9, suggesting that these enzymes are related to this processo The results for MMI suggest that its production may be related to the resident cells grom gingival tissue. In the otl hand MMP-9 may be produced by migrating inflammatory cells. As metaloproteinases (MMPs) são enzimas envolvidas na manutenção da homeostase da matriz extracelular e um desequilíbrio entre suas concentrações e as de seus inibidores endógenos (TIMPs) pode ter um papel importante nas modificações cardiovasculares associadas com a MetS. Além disso, um estado inflamatório crônico tem sido descrito em pacientes com MetS e podem levar ao desenvolvimento de aterosclerose. A hipótese deste estudo é que aumento nas concentrações de alguns marcadores pró-inflamatórios e um desequilíbrio entre as concentrações de metaloproteinases de matriz extracelular (MMPs) e seus inibidores endógenos (TIMPs) possam estar envolvidos na fisiopatologia da MetS. Para abordar esta questão, foram estudados 25 indivíduos saudáveis (controles) e 25 pacientes com MetS. As concentrações plasmáticas de pró-MMP-2 e de pró-MMP-9 foram determinadas por zimografia. A concentração plasmática de MMP-8, MMP-3, TIMP-1, TIMP-2, proteína quimioatraente de monócitos-1 (MCP-1), interleucina-6 (IL-6), molécula de adesão intracelular-1 (sICAM-1) e sP-selectina foram medidas por kits de ELISA. Os resultados revelaram um aumento nas concentrações de sP-selectina, sICAM-1, MCP-1, IL-6, pró-MMP-9, MMP-8 e TIMP-1 em pacientes com MetS comparados com controles saudáveis. Além disso, nenhuma diferença nos níveis de pró-MMP-2, MMP- 3 e TIMP-2 foi encontrada. Nossos resultados sugerem que as MMPs podem ter um papel no aumento do risco cardiovascular em pacientes com MetS e que intervenções farmacológicas no alvo das MMPs, especialmente a MMP-9 e a MMP-8 merecem maior atenção em pacientes com MetS / Abstract: The metabolic syndrome (MetS) denotes a clustering of cardiovascular and diabetes risk factors. The metalloproteinase (MMPs) are enzymes involved in maintaining the homeostasis of the extracellular matrix and an imbalance between their concentrations and their endogenous inhibitors (TIMPs) may play important role in the cardiovascular modifications associated with MetS. In addition, a chronic inflammatory state has been described in patients with MetS, thus leading to the development of atherosclerosis. We hypothesized that increased concentrations of pro-inflammatory mediators and imbalanced concentrations of matrix metalloproteinases (MMPs) and their endogenous inhibitors (TIMPs) may reflect the pathophysiology of MetS. To address this issue, we studied 25 healthy subjects and 25 MetS patients. The plasma levels of pro-MMP-2 and pro-MMP-9 were determined by gelatin zymography. The plasma concentrations of MMP-8, MMP-3, TIMP-1, TIMP-2, monocyte chemoattractant protein-1 (MCP-1), interleukin-6 (IL-6), intercellular adhesion molecule (sICAM-1), and sP-selectin were measured by ELISA kits. The results revealed an increase in sP-selectin, sICAM-1, MCP-1, IL-6, pro-MMP-9, MMP-8, and TIMP-1 in MetS patients compared with healthy controls. In addition, no differences in pro-MMP-2, MMP-3, and TIMP-2 levels were found. These findings suggest that MMPs may have a role in the increased cardiovascular risk of MetS patients and pharmacological interventions targeting MMPs, especially MMP-9 and MMP-8 deserve further investigation in MetS patients / Mestrado / Mestre em Farmacologia

São poucos os relatos da função dessas moléculas nos tecidos que sustentam, constituem e dão origem aos dentes. Entretanto, já foi mostrado que antibióticos como a doxiciclina podem reduzir a atividade dessas enzimas. Considerando que o incisivo de rato apresenta crescimento e erupção continuados ao longo da vida do animal assegurada por constante proliferação e diferenciação das células localizadas na região apical (odontogênica) em esmalte, dentina e ligamento periodontal, este se torna um excelente modelo para o estudo da atividade de MMPs e seus inibidores sobre o processo proliferativo e eruptivo do mesmo. Assim, os objetivos deste estudo foram: 1-Avaliar, na região odontogênica, o efeito da doxiciclina e da hipofunção sobre a expressão e a atividade da MMP-2 e MMP-9; a expressão dos TIMP-1,-2,-3 por meio das técnicas de zimografia, zimografia reversa, imunohistoquimica e a expressão de MT1-MMP pela técnica de Western Blot, 2- Avaliar o efeito da doxiciclina e a condição hipofuncional sobre a taxa de erupção dos dentes incisivos, 3- Avaliar a proliferação celular da região odontogênica, na presença de doxiciclina e hipofunção, por meio de marcador de proliferação celular, 4- Estabelecer relação entre os efeitos da doxiciclina, a expressão das MMPs, TIMPs, a proliferação celular da região odontogênica e o processo de erupção. Foram usados de 5 a 7 ratos machos adultos divididos em 4 grupos: controles - normofuncional (NF) e doxinormofuncional (DNF) e tratados - hipofuncional (HP) e doxhipofuncionais (DHP). Os grupos HP e DHP tiveram os dentes esquerdos inferiores cortados com broca de alta rotação e medidos a cada dois dias durante 12 dias. Nos dentes dos grupos NF e DNF uma marca foi feita para estimar a taxa de erupção realizada da margem gengival até a marca ou até o final do dente nos grupos HP e DHP. Os grupos DNF e DHP receberam dose diária matutina, de 80mg/kg de doxiciclina diluída em água por agulha de gavagem durante 14 dias. Os ratos foram sacrificados por deslocamento cervical e as hemimandibulas foram processadas para os métodos de imunohistoquimica, zimografia e Western Blot. Os resultados mostraram que os tratamentos não alteraram a atividade e expressão de MMP-2 e MMP-9 e nem de TIMP-1 e TIMP-3. Entretanto, a hipofunção aumentou a taxa de erupção, a expressão de MT1-MMP, TIMP-2 e a proliferação celular quando comparadas com os grupos controles. Conclui-se que: 1- MMP-9 não participa da remodelação da matriz extracelular na região odontogênica, 2- A doxiciclina não altera a erupção, atividade e expressão das moléculas avaliadas e nem a proliferação celular; 3- A condição hipofuncional altera; aumentando a erupção, a expressão de MT1-MMP, TIMP-2 e a proliferação celular, 4- Existe relação entre o aumento da erupção e proliferação celular bem como entre a expressão de MT1-MMP/TIMP-2 e a proliferação celular sugerindo papel importante dessas moléculas nos processos de proliferação e erupção / Abstract: Metalloproteinases (MMPs) are enzymes that degrade extracellular matrix components and are regulated by molecules called tissue inhibitors of metalloproteinases (TIMPs) and there is few reports of the function of these molecules in the tissues that support and give rise to teeth. However, it has been shown that antibiotics such as doxycycline can reduce the activity of these enzymes. Whereas the rat incisor has erupted and continued growth over the life of the animal provided by continuing proliferation and differentiation of cells in the apical region (odontogenic) on enamel, dentin and periodontal ligament, it becomes an excellent model for studying the activity of MMPs and their inhibitors on the proliferative and eruption process. Thus the aims of this study were: 1-To study in odontogenic region, the effects of doxycycline and hypofunctional condition on the expression and activity of MMP-2 and MMP-9, the expression of TIMP-1, -2, -3, using techniques of zymography, reverse zymography, immunohistochemstry and the expression of MT1-MMP by Western Blot; 2 - To evaluate the effect of doxycycline and hypofunctional condition on the eruption rate of incisors. 3 - To estimate the cellular proliferation of odontogenic region in the presence of doxycycline and hypofunctional eruption condition, using cell proliferation marker, 4 - Establish relationship between the effects of doxycycline, the expression of MMPs, TIMPs, the cellular proliferation of the odontogenic region and the process of eruption. Were used 5 to 7 adult male rats divided into 4 groups: control-functioned normally (NF) and doxinormofuncional (DNF) and treated-hipofuncional (HP) and doxihipofuncional (DHP). The groups HP and DHP had left lower teeth cut with high-speed drill every two days until for 12 days. In the teeth of groups NF and DNF a mark was made to estimate the rate of eruption of the gingival margin held up to the mark or until the end of the tooth in groups HP and DHP. Groups DNF and DHP received daily morning dose of 80mg/kg of doxycycline diluted in water by needle gavage for 14 days. The rats were sacrificed by cervical dislocation, and the hemimandibles were processed for immunohistochemical methods, zymography and Western Blot. The results showed that the treatments did not alter the activity and expression of MMP- 2 and MMP-9 nor TIMP-1 and TIMP-3. However, the hypofunction increased the rate of eruption, the expression of MT1-MMP, TIMP-2 and cell proliferation when compared with control groups. We conclude that: 1 - MMP-9 does not participate in remodeling the extracellular matrix in the odontogenic region 2 - Doxycycline does not alter the eruption, activity and expression of the molecules assessed and not cell proliferation, 3 - The condition hipofuncional changes, increasing the eruption, the expression of MT1-MMP, TIMP-2 and cell proliferation, 4 - There is a relationship between the eruption and increased cell proliferation and between the expression of MT1-MMP/TIMP-2 and cell proliferation suggesting an important role of molecules in the cell proliferation and eruption processes / Doutorado / Histologia e Embriologia / Doutor em Biologia Buco-Dental
