Preservation of Nitric Oxide Availability as Nitrite and NitrosothiolsMadrasi, Kumpal J 09 November 2012 (has links)
Nitric Oxide (NO) has been known for long to regulate vessel tone. However, the close proximity of the site of NO production to “sinks” of NO such as hemoglobin (Hb) in blood suggest that blood will scavenge most of the NO produced. Therefore, it is unclear how NO is able to play its physiological roles. The current study deals with means by which this could be understood. Towards studying the role of nitrosothiols and nitrite in preserving NO availability, a study of the kinetics of glutathione (GSH) nitrosation by NO donors in aerated buffered solutions was undertaken first. Results suggest an increase in the rate of the corresponding nitrosothiol (GSNO) formation with an increase in GSH with a half-maximum constant EC50 that depends on NO concentration, thus indicating a significant contribution of ∙NO2 mediated nitrosation in the production of GSNO. Next, the ability of nitrite to be reduced to NO in the smooth muscle cells was evaluated. The NO formed was inhibited by sGC inhibitors and accelerated by activators and was independent of O2 concentration. Nitrite transport mechanisms and effects of exogenous nitrate on transport and reduction of nitrite were examined. The results showed that sGC can mediate nitrite reduction to NO and nitrite is transported across the smooth muscle cell membrane via anion channels, both of which can be attenuated by nitrate. Finally, a 2 – D axisymmetric diffusion model was constructed to test the accumulation of NO in the smooth muscle layer from reduction of nitrite. It was observed that at the end of the simulation period with physiological concentrations of nitrite in the smooth muscle cells (SMC), a low sustained NO generated from nitrite reduction could maintain significant sGC activity and might affect vessel tone. The major nitrosating mechanism in the circulation at reduced O2 levels was found to be anaerobic and a Cu+ dependent GSNO reduction activity was found to deliver minor amounts of NO from physiological GSNO levels in the tissue.

Reduction of Sodium Nitrite and Sodium Chloride in a Cured Meat Product by Using Acid Development to Inhibit Botulinal Toxin FormationAssamongkorn, Angsana 01 May 1985 (has links)
Pediococcus acidilactici as a lactic acid producer, and sucrose were added to breakfast strip products in order to reduce added levels of sodium nitrite and sodium chloride and yet limit the growth of Clostridium botulinum and inhibit toxin production. Sucrose at 0.4 or 0.9% with P. acidilactici at 1.0 x 10 cells/g were added during preparation of breakfast strip products prepared with combinations of 40 or 80 ppm sodium nitrite and 1.0 or 2.0% sodium chloride. Other ingredients were added at levels normally used in cured meat products. C. botulinum types A and B at 1,000 spores/g were inoculated during product preparation. The finished products were sliced, vacuum packed, double bagged, vacuum sealed again, and incubated at 27 C. Samples were analyzed before incubation and every week for 4 weeks for anaerobic plate counts, pH, residual nitrite, Most Probable Number counts, and botulinal toxin. The results revealed that 0.9% sucrose increased the acid production, thus inhibiting the botulinal growth and toxin formation in the products. Lactic acid bacteria and sucrose at 0.4% did not produce enough acid to inhibit toxin formation. Sodium nitrite at 40 ppm and sodium chloride at 1.0% permitted increased growth rate and toxin development of C. botulinum. Addition of 0.9% sucrose and P. acidilactici was necessary to provide antibotulinal properties to breakfast strip products prepared with 40 ppm sodium nitrite and 1.0% sodium chloride.

Chapter1 gives an overview of the basic principles of electrochemistry. A rigorous electrochemical study on the solution phase and solid phase cobalt phthalocyanine (CoPC) is presented in chapter2. The formof CoPC on carbon electrodes was characterized by scanning electron microscope (SEM). The use of CoPC modified edge plane pyrolytic graphite (CoPC-EPPG) for sensing nitrite (NO₂⁻) was also investigated. It was found that the claimed mediator CoPC has no influence on the process. A bare glassy carbon (GC) electrode was successfully applied for the quantitative determination of nitrite as a simple alternative to the modified electrodes reported in the literature (chapter3). Chapter4 compares the voltammetric responses of an edge plane pyrolytic graphite electrode covalently modifed with 2-anthraquinonyl groups (EPPG-AQ2) and solution phase anthraquinone monosulphonate (AQMS) in the presence of a limited concentration of protons. The solution phase and surface bound species show analogous responses resulting in split waves. Digisim™ simulation of the AQMS voltammetry have shown that the pH adjacent to the electrode may be altered by up to 5-6 pH units in low buffered solutions; this is caused by the consumption of protons during the electrochemical reaction. Chapters5 and 6 compare the electrochemical properties of 2-anthraquinonyl groups covalently attached to an edge plane pyrolytic graphite (EPPG) and to a gold electrode. In both cases simulations using newly developedMarcus-Hush-Chidsey theory for a 2e⁻ process assuming a uniform surface did not achieve a good agreement between theory and experiment. Subsequently two models of surface inhomogeneity were investigated: a distribution of formal potentials, E<sup>Ө</sup>, and a distribution of electron tunneling distances, r₀. For both EPPG-AQ2 and Au-AQ2 modified electrodes the simulation involving E<sup>Ө</sup> distribution turned out to be the most adequate. This is the first time that Marcus-Hush-Chidsey theory has been applied to a 2e⁻ system. Chapter7 briefly summarizes the obtained results.

Os peitos de galinhas matrizes pesadas de corte, poedeiras comerciais brancas e frangos foram desossados manualmente, com o objetivo de avaliar e comparar as propriedades tecnológicas - capacidade de retenção de água e de emulsificação - bem como pH e composição centesimal, além de elaborar e avaliar sensorialmente um produto reestruturado empanado, tipo nugget. Os resultados das análises indicaram que não houve diferença significativa entre as carnes analisadas, nem entre produtos processados, implicando utilização regular desse tipo de matéria-prima na indústria de alimentos. O restante das carcaças das galinhas foi utilizado para extração de carne mecanicamente separada (CMS) para avaliação da estabilidade microbiológica. As CMSs foram submetidas a dois tratamentos: I) com 150ppm de nitrito, embaladas em saco de polietileno; e II) com 150ppm de nitrito e 500ppm de eritorbato, embaladas em saco de polietileno. Como controle, as amostras de CMS foram apenas acondicionadas em sacos de polietileno. Todas as embalagens foram estocadas e congeladas a -18ºC por 99 dias. Os resultados da análise de caracterização microbiológica indicaram que tanto as amostras de CMS de galinha matriz quanto de galinha poedeira atenderam os padrões de contagem requeridos pela legislação Brasileira. A avaliação da estabilidade microbiológica das CMSs no primeiro e no último dia para mesófilos, Escherichia coli, Staphylococcus aureus, Clostridium perfringens e Pseudomonas, bem como as análises quinzenais de microrganismos psicrotróficos, indicaram que não houve uma redução significativa nas contagens, independentemente do tratamento aplicado. Não foi detectada Salmonella em nenhuma das amostras analisadas. Tais resultados apontaram o tempo de estocagem como o principal fator na redução da carga bacteriana. / The breasts of heavy fowls, white commercial egg-layers and chickens were manually deboned in order to evaluate and compare technological propertieswater hold and emulsifying capacityand pH and centesimal composition, as well as to prepare and assess a restructured coated product. The results of the analyses indicated that there was not a significant difference between the meats analyzed, nor among the processed products, implying the regular use of such meat in the food industry. The remaining parts of the hens carcasses were mechanically deboned for the evaluation of microbiological stability. The mechanically deboned meat (MDM) was submitted to two treatments: I) 150ppm of nitrite, packed in polyethylene bags; II) 150ppm of nitrite and 500ppm of erythorbate, packed in polyethylene bags. The control meats were just packed in polyethylene bags. All samples were frozen-stored at -18ºC for 99 days. The results of microbiological characterization indicated that both samples from heavy fowls and egg-layers were in accordance with the Brazilian legislation. MDM microbiological stability evaluation on the first and last days for fecal coliforms, mesophiles, Staphylococcus aureus, Clostridium perfringens and Pseudomonas, as well as the analyses carried out every fifteen days for psychrotrophics bacteria indicated that there was not a significant reduction in the presence of the above, irrespective of the treatment applied. Salmonella was not detected in any of the samples. Such results indicated that freezing storage period was the main factor responsible for the reduction of the bacterial count in MDM.

A diminuição da biodisponibilidade do óxido nítrico (NO) e o aumento na atividade das metaloproteinases da matriz extracelular (MMPs) são alguns dos principais mecanismos fisiopatogênicos envolvidos nas doenças cardiovasculares (DCV). Foi demonstrado que o NO pode reduzir a expressão e atividade de MMPs em células musculares lisas vasculares, células mesangiais, entre outras. Em outro estudo, foi mostrado que drogas inibidoras da NO sintase (NOS) podem aumentar a expressão de MMPs. Apesar de o NO apresentar-se diminuído e as MMPs aumentadas durante as DCV, não há evidência clara de que os níveis de NO possam modular diretamente a atividade de MMPs no aparelho cardiovascular. Também não se sabe se este possível efeito seria mediado pelo NFB, nem se este possível efeito é dependente da ativação da guanilato ciclase [que promove a formação de GMP cíclico (GMPc)]. Desta maneira, este estudo teve como objetivos: A) investigar os efeitos de drogas ativadoras da via NO-GMP sobre os aumentos da atividade e expressão de MMP-9 em células endoteliais que acontecem sob efeito de phorbol 12-miristato 13-acetato (PMA, droga indutora da expressão de MMP-9); e B) determinar se a inibição de NOS em células endoteliais é acompanhada por aumento da atividade e expressão de MMPs, e C) determinar se estes efeitos são dependentes da ativação de NFB ou da formação de GMPc. Células endoteliais de veia umbilical humana (HUVECs) foram cultivadas em DMEM e tratadas por 24 horas com 10 nmol/L de PMA ou diferentes concentrações de drogas ativadoras da via NO-GMP ou de inibidor da NOS. Meio de cultura condicionado ou lisado celular foram coletados e submentidos aos ensaios de zimografia, ELISA, immunoblotting ou análise da concentração de nitrito. Os tratamentos com detanonoato, SNAP, atorvastatina e nitrito de sódio diminuíram os aumentos da atividade gelatinolítica e expressão de MMP-9 estimulados por PMA sem afetar as concentrações do inibidor tecidual da metaloproteinase da matriz-1 (TIMP-1). Esses efeitos não foram modificados pelos tratamentos com ODQ (inibidor da guanilato ciclase solúvel) ou 8- bromo-cGMP (um análogo de GMPc) ou hemoglobina (um sequestrador de NO). Enquanto o PMA aumentou a concentração de fosfo-NFB p65, os tratamentos com SNAP, atorvastatina ou nitrito não apresentaram influência sobre esse efeito. O tratamento com L-NAME, um inibidor da NOS, não apresentou efeito sobre a atividade gelatinolítica de MMP-9. Em conclusão, foram demonstrados que os efeitos inibitórios de drogas ativadoras da via NO-GMPc sobre a produção estimulada de MMP-9 em células endoteliais são independentes de mecanismos mediados por GMPc e NFB, e a inibição da NOS não altera a atividade de MMP-9. / Impaired nitric oxide (NO) bioavailability and imbalanced matrix metalloproteinases (MMPs) activity have important roles in the pathophysiological mechanisms involved in cardiovascular disease (CVD). It was shown that NO can reduce MMPs expression and activity in vascular smooth muscle cells, mesangial cells, and others. In another study, a NO synthase (NOS) inhibitor has increased MMPs expression. Although NO was decreased and MMPs was increased during CVD, there is clear evidence that NO levels can directly modulate MMPs activity in the cardiovascular system. Also, it is not known whether this effect would be mediated by NFB, nor whether this effect is dependent on guanylate cyclase activity (which promotes the formation of cyclic GMP). Thus, this project aims to study whether A) the effect of NO donors might decrease MMP-9 activity and expression in endothelial cells stimulated by phorbol 12-myristate 13-acetate (PMA) (a well-known inducer of MMP-9), and B) the effect of NOS inhibitors might increase MMPs activity and expression in endothelial cells, and C) to determine whether those effects are mediated by the NFB activation or cGMP levels. Endothelial cells from human umbilical vein (HUVECs) were grown in modified DMEM and were treated for 24 hours with 10 nmol/L PMA or different concentrations of NO-GMPc pathway stimulating drugs or NOS inhibitor. Conditioned medium or cell lysate were collected after treatments and analyzed by zymography, ELISA, immunoblotting or to determine nitrite concentration. Detanonoate, SNAP, atorvastatin or sodium nitrite treatments attenuated PMA-induced increases in MMP- 9 gelatinolytic activity and expression, but they had no effect on tissue inhibitor of matrix metalloproteinase-1 (TIMP-1) concentrations. These effects were not modified by ODQ (a soluble guanylate cyclase inhibitor), or 8-bromo-cGMP (cGMP analogue), or hemoglobin (a NO scavenger). While PMA increased phospho-NFB p65 concentration, SNAP, atorvastatin or nitrite had no influence on this effect. The treatment with L-Name, a NOS inhibitor, had no effect on MMP-9 activity. In conclusion, this study shows that the inhibitory effects of NO-GMPc pathway stimulating drugs on MMP-9 production by endothelial cells are independent of cGMP- and NFB-mediated mechanisms, and NOS inhibitor had no effect on MMP-9 levels

Desenvolvimento de um sensor eletroquímico planar modificado com 1-2 Diaminobenzeno (DAB) para monitoração de nitrito por FIA-automatizada. / Development of a planar electrochemical sensor modified with 1-2 Diaminobenzene (DAB) for nitrite measuring by automatic FIA.Almeida, Fernando Luis de 11 September 2009 (has links)
A presente dissertação de mestrado tem o propósito de relatar o desenvolvimento de um sensor planar eletroquímico com três eletrodos para o monitoramento de nitrito e seus interferentes (ácido úrico, ácido ascórbico e paracetamol). Para tal, no procedimento experimental é descrito o desenvolvimento do aparato extracorpóreo (mini-bomba compressora, válvulas solenóides e cela de análise). Esse é acoplado a um sistema de Análise por Injeção em Fluxo Automatizada (do inglês, Flow-injection Analysis) FIA-automatizada. Este trabalho também descreve a fabricação dos sensores em substrato de alumina, o processo de limpeza padrão dos eletrodos, o processo de obtenção dos eletrodos de referência em HCl (0,1 mol L-1) e ativação eletroquímica da superfície do eletrodo de trabalho com H2SO4 (0,1 mol L-1). Em seguida, é mostrada a estabilização dos eletrodos de referência Ag/AgCl em solução salina contendo cloretos, o estudo da secagem dos eletrodos em temperatura ambiente com exposição à luz e a caracterização dos mesmos por três métodos distintos, a saber: i) teste de reversibilidade (corrente redox vs. potencial); ii) variação do potencial com o tempo (coeficiente de degradação termodinâmica) e iii) variação do potencial com a concentração de cloretos. Também, apresenta-se a caracterização da área efetiva exposta do eletrodo de medição (trabalho) e os resultados da deposição de 1-2 Diaminobenzeno (DAB) como polímero seletivo. Além disso, são apresentados os resultados e discussões dos pseudo-eletrodos (recoberto ou não com Náfion® 117). Um ponto relacionado, a histerese associada à irreversibilidade dos eletrodos de referência resultou da ordem de 40 mVAg/AgCl Náfion® 117. Os potencias termodinâmicos observados foram da ordem de 100 ± 6 mVAg/AgCl Náfion® 117 para pseudo-eletrodo de referência sem e com Náfion® 117. Depois, foi realizado um estudo criterioso dos potenciais de resposta ao nitrito e aos interferentes por meio da técnica de DPV (do inglês, Differencial Pulse Voltammetry). Para evitar degradação acelerada (corrosão) do polímero sobre o eletrodo de trabalho, optou-se por realizar as medições com potencial igual a 0,50 VAg/AgCl Náfion® 117. Nessa condição, apresenta-se a monitoração de nitrito por FIA-automatizada na faixa de concentração de 50 a 250 mol L-1. Os resultados para a medição de nitrito mostraram que o sensor planar eletroquímico amperométrico desenvolvido é promissor, pois esse apresentou ótimo desempenho de reprodutividade (99,66%), sensibilidade (90 microA mol-1 L mm-2), seletividade (0,32%), repetitividade (91,28%) e estabilidade (15 ± 0,3 pA). / In this Master of Science, it has been developed an electrochemical planar sensor defined with three electrodes for nitrite measuring and its interferents (uric acid, ascorbic acid and paracetamol). In the experimental procedure, it is shown the development of an extracorporis set up (mini-pump, solenoid valves and analysis cell) which is coupled to a system of Flow-injection Analysis (FIA). This work also describes the sensor fabrication using alumina substrates, the standard cleaning of the electrodes, the process to obtain Ag/AgCl reference electrodes in HCl (0.1 mol L-1) and electrochemical activation of the working-electrode surface with H2SO4 (0.1 mol L-1). Following, it is shown the study of drying for electrodes at room temperature with exposition to room light and their characterization using three different methods: i) reversibility test (redox current vs. potential); ii) variation of the potential with the time (thermodynamics degradation coefficient) and iii) variation of the potential with the chloride concentration. Also, it is presented the characterization of the effective exposed area of the working electrode and the results of the deposited 1-2 Diaminobenzene (DAB) is presented as a selective polymer. In addition, the results and discussions of the pseudo-reference (recovered or not with Nafion® 117) are presented. Related to irreversibility is the hysteresis associated to the reference electrodes which resulted in the order of 40 mVAg/AgCl Nafion® 117. The observed thermodynamic potentials were of (100 ± 6) mVAg/AgCl Nafion® 117 for pseudo-electrodes recovered with Nafion® 117. Later on, a careful study of the response to nitrite and to its interferents was performed by means of the DPV technique (Differential Pulse Voltammetry). In order to avoid accelerated degradation (corrosion) of the polymer, measurements were performed at a potential of 0,5 VAg/AgCl Nafion® 117. In this condition, nitrite was monitored using the automatized-FIA (Flow Injection Analysis) for concentration in the range of 50 to 250 mol L-1. The results have shown that the planar amperometric sensor for nitrite measuring is usefull since it was observed excellent performance related to reproducibility (99.66%), sensitivity (90 microA mol-1 L mm-2), selectivity (0.32%), repetibility (91.28%) and stability (15.0 ± 0.3 pA).

O presente trabalho inclui duas etapas da pesquisa. Na primeira fase do projeto, a oxidação eletroquímica do nitrito em eletrodo de carbono vítreo modificado com um filme obtido eletrostaticamente de Co-TRP/Zn-TPPS, foi investigada em pH 4,7. Um aumento de corrente foi detectado em 0,75V, cujo máximo foi proporcional à concentração do íons nitrito, exibindo uma dependência inversa com a espessura do filme. A velocidade de transferência de carga foi avaliada por cronoamperometria, sendo que a velocidade de difusão da carga eletroquímica através do filme é mais alta do que a velodicade de difusão do íon em solução. O processo catalítico envolvendo a oxidação do íon nitrito no filme de pares de depósitos alternados foi também investigado com as técnicas da voltametria cíclica e de disco rotativo. Em concentrações relativamente baixas de substrato, o processo mediado foi tratado como um sub-caso, SR, na notação de Saveant, permitindo o cálculo da constante de velocidade para a reação de troca na interface filme-solução, sendo que tal valor foi estimado em 3,4x103 dm3 mol-1 s-1. Na segunda parte da investigação, a potencialidade analítica do eletrodo modificado foi confirmada por experimentos de injeção em fluxo usando solução carregadora tampão pH 4,7. Devido à atividade catalítica da camada imobilizada, a determinação de nitrito foi realizada em potencial menos positivo do que aqueles obtidos em eletrodos polidos (0,75 V vs. Ag/AgCl) e os dados obtidos demostraram o melhor desempenho do eletrodo modificado, indicando excelente repetibilidade e maior sensibilidade. Na condição experimental otimizada, o nitrito foi determinado na configuração FIA com limite de detecção de 0,1 µmol L-1 e a faixa analítica de aplicabilidade do método foi de 1 a 1000 µmol L-1. Nitrato também foi quantificado após sua redução estequiométrica a nitrito pela passagem da solução através da coluna de cádmio/cobre. Ambos analitos foram quantificados em água mineral e amostras de saliva e os resultados foram concordantes com aqueles obtidos usando método oficial. / This present work includes two research steps. ln the first step the electrochemical oxidation on nitrite on a glassy carbon disc electrode, modified with an electrostatically assembled film of tetraruthenated cobalt porphyrin/(meso-tetra(4-sulphonatephenyl)porphyrinate zinc(II), was investigated at pH = 4.7. A current enhancement was observed by using the modified electrode, the signal being proportional to the concentration of nitrite ions and exhibiting an inverse dependence with the film thickness. The charge transfer rate was evaluated by chronoamperometry, showing that the rate of diffusion of electrochemical charge through the film is higher than the rate of diffusion of substrates in solution. The catalytic process involving the oxidation of the nitrite ions at the bilayered film was also investigated based on cyclic and rotating disc voltammetry. At relatively low substrate concentrations the mediated process has been dealt with in terms of the SR sub-case in Saveant\'s notation, allowing the calculation of the rate constant for the cross-exchange reaction at the film-solution interface as 3.4x103 dm3 mol-1 s-1. In the second part the analytical potentiality of the modified electrode was studied by flow injection experiments carried out by using a pH 4.7 buffer as carrier solution. Owing to the catalytic activity of the immobilized layer, the determination of nitrite was performed at less positive potentials (0.75 V versus Ag/AgCl) and the data obtained showed a better performance indicated by both an excellent repeatability and increased sensitivity. At the optimized experimental condition, nitrite was determined in the FIA configuration with a limit of detection of 0.1 µmol L-1. Nitrate was also quantified after its stoichiometric reduction to nitrite by passing the solution through a cadmium/copper column. Both analytes were quantified in mineral water and saliva samples and the results were in agreement with the ones obtained by using an official method.
