Coupled Dynamic Analysis of Large-Scale Mono-Column Offshore Wind Turbine with a Single Tether Hinged in Seabed

The increased interest in the offshore wind resource in both industry and academic and the extension of the wind field where offshore wind turbine can be deployed has stimulated quite a number of offshore wind turbines concepts. This thesis presents a design of mono-column platform supported for 5 MW baseline wind turbine developed by the National Renewable Energy Laboratory (NREL), with a single tether anchored to the seabed. The design, based on the pioneer concept SWAY, results from parametric optimized design processes which account for important design considerations in the static and dynamic view, such as the stability, natural frequency, performance requirements as well as the economic feasibility. Fully coupled aero-hydro-servo-elastic model is established in the time-domain simulation tool FAST (Fatigue, Aerodynamics, Structures, and Turbulence) with the hydrodynamic coefficients from HydroGen, an indoor program providing same outputs as the commercial software WAMIT. The optimized model is verified by imitating the frequency-domain approach in FAST and thus comparing the results with the frequency-domain calculations. A number of simulations with various wind and wave conditions are run to explore the effect of wind speed and wave significant height in various water depths. By modifying the optimized model to a downwind turbine with the nacelle rigidly mounted on the tower and the single tether connected to the platform by a subsea swivel, the modified models are more closed to the original SWAY-concept wind turbine. These models are compared based on the platform motion, tether tension, displacement, nacelle velocity and acceleration, resonant behavior as well as the damping of the coupled systems. The results of these comparisons prove the advantage of the modified model in performance. The modified model has also clarified itself a good candidate for deep water deployment.

Affinity chromatographic purification of recombinant human growth hormone

The purpose of the study is to purify human growth hormone from the fermentation broth by affinity chromatography. For this purpose, human growth hormone specific oligonucleotide aptamers are selected among an aptamer library / selected oligonucleotides were synthesized and used as ligands. Effect of pH on ligand-human growth hormone complex formation was investigated and the highest complex formation was obtained at pH= 7.0. Human growth hormone is separated from the fermentation broth with 99.8% purity and 41% overall yield. The equilibrium data obtained was described by Langmuir type isotherm where saturation constant (q0) and affinity constant (K) are calculated as 0.338 mg hGH/&micro / mol aptamer and 0.059 mg hGH/ml, respectively. Further, equilibrium data obtained using aptamer affinity column was described by Langmuir type isotherm where saturation constant (q0) and affinity constant (K) are 0.027 mg hGH/&micro / mol aptamer and 1.543 mg hGH/ml, respectively. It is possible that, selected aptamer can be used for purification of bulk amounts of recombinant human growth hormone by using aptamer affinity chromatography.

[pt] Atualmente o mundo enfrenta uma grave crise ambiental e elevados índices de pobreza. Com a intenção de minimizar estes problemas esta Tese propõe a alternativa do uso do bambu como armadura em pilares de concreto armado, substituindo o aço em construções civis. O comportamento dos pilares de concreto armado é descrito através de uma revisão bibliográfica e de ensaios realizados em pilares curtos submetidos à carga axial, dos quais três foram armados com bambu e um com aço. Abordam-se aspectos relativos à taxa de armadura longitudinal, a resistência do bambu à compressão e a aderência entre bambu e concreto, para isto ensaios complementares em ripas pequenas de bambu sob carga de compressão centrada e testes de arrancamento utilizando o produto sikadur 32 gel para acrescentar aderência ao bambu e constatar sua eficiência, foram realizados e comparados com um corpo de prova armado com aço. Os pilares de concreto armados com bambu e aço foram comparados e analisados. Segundo recomendações das normas trabalhos futuros foram propostos. / [en] Nowadays the world faces a serious environmental crisis and high poverty rates. Intending to minimize these problems, this thesis proposes the alternative usage of bamboo as reinforcement on columns of reinforced concrete, substituting steel in civil constructions. The reinforced concrete columns performance is described through a bibliographical revision and experiments done on short columns submited to axial load: three were built with bamboo and one with steel. Aspects related to the surcharge of longitudinal reinforcement, bamboo resistance to compression and adherence between bamboo and concrete are discussed extensively. With this purpose, subsequent experiments were made using small chips of bamboo submited to strong centered compression and also, Pull Out tests, utilizing the product Sikadur 32 gel to add adherence to bamboo and appraise its efficiency, were performed and compared to a body of proof built with steel. The concrete columns reinforced with bamboo and steel were thoroughly compared and analyzed. According to the pattern experiments, future developments on this research were proposed.

Planning an aquifer storage and recovery scheme in the Sherwood Sandstone aquifer

Aquifer Storage and Recovery (ASR) involves the injection of water into an aquifer for subsequent recovery from the same well. Whilst ASR provides a competitive alternative to reservoir storage, a lack of precedence of successful schemes and uncertainties with respect to regulatory requirements, and abstracted water quality and quantity have limited its implementation in the UK. The ambition of this research is to improve understanding of these impediments with particular reference to the Sherwood Sandstone aquifer. Drawing on existing project review and risk management approaches, a decision support tool to help scheme designers scope ASR scheme potential and challenges was developed and tested. The tool provides practitioners with a systematic method for early stage evaluation of ASR schemes with testing confirming broad value and alignment with business processes. Limitations on the recovery of recharged water was investigated through a critical literature review on clogging with geochemical, biological, physical and gaseous binding processes identified as the most likely mechanisms to impact an ASR scheme in this aquifer. Water quality changes during storage and the impact of storage period on recovered water quality were investigated in the laboratory using ASR simulating columns, with storage times of 15, 20, 30 and 60 days. Water quality improvements such as biodegradation of disinfection by-products, denitrification and sulphate reduction were observed. However recovered water quality deteriorated with respect to iron, arsenic, manganese, total organic carbon and nickel. These results, together with the review of regulations conducted as part of decision support tool development, suggest that the current interpretation of the Water Framework Directive requirements is overly restrictive and is deterring wider implementation of ASR in the UK. Conclusions focus on the need for a more appropriate approach to regulating ASR schemes, in particular, one which adopts a risk based approach to determining water quality standards.
