Franco, Carolina Andrade. January 2016 (has links)
The plant breeding is considered to be a complex process that generates multiple information sources and in many cases, difficult to understand. The aim of this work was to select progenies with superior characters from bi-parental crosses with soy source of resistance to glyphosate (RR), and identify intersections and parents more efficient through multivariate approaches. Furthermore, aimed to further test the efficiency of the methods in the selection process for multiple traits of interest. The experiment was conducted in the experimental design of type families to witness progress in the agricultural year 2010/2011 and 2011/2012 in Jaboticabal being that in F3 populations selected six plants were phenotypically superior and evaluated for the traits: number of days to flowering (NDF), number of days to maturity (NDM), height of the first pod (AIV), plant height at maturity (APM), lodging (Ac), agronomic value (VA), number of branches (NR), number of pods per plant (NV), one hundred seed weight (PCS), number of seeds per plant (NS) and grain yield (GY). / Abstract: Sweet sorghum (Sorghum bicolor) is featured by the high level of fermentable sugars present in its stalk. The crop is currently considered of high potential to compose the national energetic system, by producing alcohol during the sugarcane off season. The weed interference is one of the most important factors during a crop management. Because not too much have been studied about sweet sorghum, studies about the interaction between weeds and sweet sorghum crop are very welcome for the development of the bioenergy nationwide. The aim of this research were: (I) determine the periods of weed interference and the phytosociological indexes of the weed community on the sweet sorghum crop; (II) analyze the initial growth of sweet sorghum under high weed infestation; (III) study the effect of the sweet sorghum root exudate in other plant species and also evaluate the root extract of an potential stimulant plant on the production of sorgoleone by sorghum; (IV) evaluate the suppressive effect of sweet sorghum crop residues on weeds. For the first objective, the treatments were composed by increasing periods of weed presence and weed absence at two crop seasons; when it was evaluated the sweet sorghum biometry and yield, besides the determination of phytosociological indexes of the weed community (the relative indexes of density, dominance and importance, and the weed community indexes of diversity and equitability). In pots, it was carried out an assay in order to evaluate the third aim, in which hybrids of sweet sorghum were submitted to the presence of five weed species in higher density. / Abstract: Charcoal rot disease, caused by Macrophomina phaseolina, is a disease of significant economic importance in the soybean crop. This disease occurs mainly in tropical and subtropical regions and it is known that the incidence and severity occur mainly when there is stress of water and heat. The evaluation of this pathogen is difficult due to the lack of reliable and efficient methods for the evaluation of soybean genotypes. The present work had the objective of evaluating different methods of inoculation and evaluation in ten soybean genotypes, besides discovering the degree of resistance of the genotypes studied. Infected sorghum seeds, pre-germinated seed in culture medium and infected toothpick test were the tested methods. Colony forming units and root and stem severity were evaluated. There was good correlation between evaluations of colony forming units and root and stem severity (0.87). The infected sorghum seed method was the most efficient for inoculating the pathogen in soybean genotypes. Genotype PI594302 presented higher resistance in the two evaluated parameters, being indicated its introduction in programs of genetic improvement aiming the resistance to the charcoal rot in soybean. / Doutor

Abstract: The gummy stem blight, caused by fungus Didymella bryoniae, is one of the relevant diseases that occurs in melon worldwide. The goals this study were evaluated the reaction of melon genotype of different germplasm banks to D. bryoniae and propose adjustments in the grades scale and resistance reaction class. Two experiments were carried out in a completely randomized design with three replicates and eight plants per plot. The symptoms caused by fungus were evaluate through scale of grades, adopting the average grade of the last evaluation to determine the classes of resistance reaction. Were evaluated 28 genotypes in the Experiment I, of which three were selected as highly resistant (CNPH 05.1020, CNPH 11.1075 e CNPH 11.1063), being again evaluated in Experiment II together with resistance and susceptibility patterns. In both experiments, differences were detected between the genotypes (p < 0.05), indicating the existence of genetic variability for the reaction to gummy stem blight. The genotype CNPH 11.1075 was resistant to D. bryoniae in both experiments and can be incorporate in breeding programs that aim at the transference of resistance into elite lines or cultivars. The adjustments in the methodologies for the evaluation of the experiments conducted in trays, with plants in the seedling stage, allow the adequate differentiation among resistant and susceptible genotypes. / Mestre

The susceptibility of different rubber tree clones to the attack of Calacarus heveae Feres (Acari, Eriophyidae) was analyzed through field, laboratory and greenhouse essays. In the field essay, we studied the population dynamics of phytophagous mites and its influence on the mite community structure in six rubber trees clones, in a farm belonging to the Plantações Edouard Michelin, Itiquitira, MT. For that, 10 leaflets of 10 different plants were sampled from each of the six clonal plots, from March 2004 to March 2005. We observed in essays performed in the Laboratory of Acarology of Depto. Zoology and Botany, UNESP, São José do Rio Preto, the biological cycle, reproduction and population survivorship of 20 C. heveae individuals reared on detached leaflets of clones GT 1, PB 235 and RRIM 600. The essays were repeated four times, in the periods when C. heveae was more abundant in the field. For the greenhouse essay, performed in UNESP, São José do Rio Preto, SP, we verified the influence of C. heveae attack on physiological processes and on the latex yield in 9 and 11 five-months aged seedlings of the clones RRIM 600 and GT 1, respectively. The phytophagous species had different abundances on the six rubber tree clones and, consequently, influenced the occurrence and distribution of other mite species. The most susceptible clones to the attack of C. heveae were PB 260 and RRIM 600, while the most resistant were PB 235, PB 217 and GT 1. In the arenas made of detached leaflets, C. heveae had faster development, high reproductive rate and survivorship increase on PB 235 and RRIM 600. Probably, the different ages of the plants used in field and laboratory essays influenced the conflicting results in relation to PB 235 susceptibility to C. heveae. The C. heveae attack decreased the photosynthetic rate... (Complete abstract click electronic access below)

January 2013 (has links)
Abstract: Resveratrol is a phytoalexin produced under biotic and abiotic stresses. It has been found in a restricted number of plant species including peanut (Arachis hypogaea) and its wild relatives. This phytochemical has antioxidant properties, being considered a promising antitumour, cardioprotective and neuroprotective agent. Peanut is an allotetraploid specie with an AABB genomic constitution. Arachis ipaënsis and A. duranensis are the donors of the B genome and the A genome, respectively. Previous studies have shown that wild species of the genus Arachis can produce resveratrol. Therefore, the evaluation of other species of thus genus and the analysis of the variation in the production of resveratrol between accessions is essential for better exploitation of wild species in the genus Arachis. It also important the understanding of the variation in resveratrol concentration between species and accessions and that could be reached through the better understating of the role resveratrol synthase.. . The aims of this work were to evaluate the variation in resveratrol production between species (A. hypogaea, A. stenosperma, A. valida, A. wiliamsii, A. duranensis, A. ipaënsis and in a synthetic amphidiploid derived from these two wild species A. duranensis and A. ipaënsis) and between accessions of the same specie (A. hypogaea and A. stenosperma) through HPLC, and to quantify the expression of resveratrol synthase by RT-qPCR in A. hypogaea, A. duranensis, A. ipaënsis and in a synthetic amphidiploid. Leaves for the test and control groups were collected from plants cultivated in a greenhouse and three biological replicates were evaluated for each specie. The synthesis of resveratrol in leaves was induced by treatment with UV for 2 hours and thirty minutes and then the difference in the concentration of resveratrol between species and between the accessions and cultivars of the same species was analyzed. Brazil is the world's largest crop producer and exporter of sugar and ethanol. However, this crop is exposed to different kinds of stress, among which; the availability of water is noticeable as the main factor that negatively influences the vegetable growth and, therefore, the productivity. In order to identify the possible genes involved in the responses to drought tolerance in sugarcane crops, the comparative analysis of transcripts profile from two tolerant cultivars, SP83-5073, RB867515 and the sensitive cultivar SP86-5073, after has been submitted to the interruption of irrigation for 1, 3, 5 and 10 days were performed using the cDNA-AFLP technique. With 14 combinations of EcoRI/MseI selective primers, there was detected on a polyacrylamide gel, 1,316 transcript-derived fragments (TDFs) differently expressed between tolerant and sensitive cultivars submitted to water deficit (1, 3, 5 and 10 days). From these, 630 TDFs were identified on the cultivar SP83-5073 and 686 TDFs corresponded to the cultivar RB867515. The comparative analysis of genotype dependent gene expression profile in sugarcane revealed fragments similarity with genes involved in many molecular events related to the responses of plants to different stresses, including those caused by drought. These genes work in hormonal signaling, regulation of gene transcription, senescence, DNA transposition, transport of amino acids, respiration and metabolism of carbohydrates. However, nine TDFs possibly represent still unknown genes, which may be developing important roles on the responses to drought and contributing to the trait of tolerance / Doutor

Abstract: The velvetbean caterpillar Anticarsia gemmatalis Hübner and the fall armyworm Spodoptera frugiperda (J. E. Smith) are two of the major defoliating pests of soybeans. Among the methods available for their control, the use of genotypes possessing moderate and high resistance stands out. However, numerous factors, inherent to the plant or to the insect, may influence the expression of resistance in plants. Thus, this work aimed to evaluate potential factors influencing the expression of resistance in soybeans, and we tested the hypotheses that flavonoids, nutrients, oxidative enzymes, phenolics, proteins, and proteinase inhibitors may be some of the mechanisms responsible for resistance to the velvetbean caterpillar and fall armyworm. Three experiments were performed in order to elucidate these issues. The first experiment evaluated the influence of the following factors on the expression of antixenosis in the resistant genotype IAC 100 and susceptible genotype BRSGO 8360 to both insects in free-choice and no-choice feeding assays: one larva versus two larvae per leaf disc; use of non-starved larvae versus larvae starved for three hours prior to the assay; leaf disc versus entire leaflet; upper part versus lower part of plants; vegetative growth stage versus reproductive stage. The second experiment assessed the influence of the upper part versus lower part of the plant, and plants at vegetative growth stage versus reproductive stage on the expression of antibiosis to A. gemmatalis and S. frugiperda in the resistant genotype PI 227687 and susceptible genotype IGRA RA 626 RR. In addition, the concentration of nutrients and flavonoids were evaluated as potentital mechanisms responsible for resistance in soybeans. The macronutrientes quantified in the leaves of the soybean genotypes from the respective treatments were N, P, K, Ca, Mg, and S; the quantified micronutrients were B, Cu, Fe, Mn, and Zn; among the flavonoids, we quantified ... / Doutor
