Tolerância ao calor em ovelhas de raças de corte lanadas e deslanadas no sudeste do Brasil / Heat tolerance in hair and wool meat breeds ewes in southeast of Brazil

O trabalho, composto de quatro experimentos, teve como objetivo geral avaliar a tolerância ao calor em ovinos, sua relação com a coloração do pelame e a presença ou ausência de lã em ovelhas de raças lanadas e deslanadas. Os experimentos foram conduzidos no Instituto de Zootecnia, localizado em Nova Odessa, Estado de São Paulo (22º42\'S e 47º18\'W, 570m de altitude). Nos primeiros três experimentos, avaliaram-se 83 ovelhas, das raças Santa Inês (31), Morada Nova (15), Texel (14), Suffolk (11) e Ile de France (12). O índice de tolerância ao calor (ITC) foi calculado pela fórmula 10 - (TR2 - TR1), através das temperaturas retais registradas às 13h (TR1), após duas horas em descanso na sombra, e às 15h, após uma hora de exposição à radiação solar direta e uma hora de descanso à sombra (TR2). No quarto experimento, 90 ovelhas, 18 de cada raça, foram avaliadas quanto à temperatura retal (TR) e freqüência respiratória (FR) às 8h, 13h (após duas horas em descanso à sombra), 14h (após uma hora de exposição ao sol), e depois a cada 15 minutos, na sombra, até às 15h, originando quatro TR2 para cálculo dos índices. Verificou-se que não houve diferença na tolerância ao calor entre animais da raça Santa Inês de pelagem clara e escura. A raça Texel (lanada) teve menor variação de temperatura entre os dois horários, e obteve melhor índice de tolerância ao calor (P<0,05) do que a raça deslanada Morada Nova. Ovelhas das raças Suffolk e Ile de France tiveram altos índices de tolerância ao calor, independente do fato de estarem ou não tosquiadas. No experimento 4, ovelhas da raça Santa Inês tiveram TR inferiores (P<0,05) à TR de algumas raças lanadas, às 13h, 14h, 14h30min e 14h45min. Em todas as raças, não houve diferença (P>0,05) entre as TR medidas às 8h e 13h, assim como não foram encontradas diferenças (P>0,05) entre elas quanto à TR da manhã e aos 60 min após o estresse (15h). As ovelhas deslanadas tiveram FR inferiores (P<0,05) às lanadas. Não houve diferença (P>0,05) entre o ITC calculado com base na TR2 aos 45 e 60 min pósestresse. Concluiu-se que o tempo de registro da TR2 para a espécie ovina pode ser antecipado de 60 para 45 minutos após o estresse, e que as ovelhas avaliadas estão adaptadas ao clima do Estado de São Paulo. / This study consisted of 4 experiments and had as main goal evaluate the heat tolerance in ewes, and its relation to hair color and presence or absence of wool in wool and hair breeds. The experiments were conducted at the Instituto de Zootecnia, located in Nova Odessa city, São Paulo State, Brazil (22º42\'S e 47º18\'W, 570m altitude). Three experiments evaluated 83 ewes of the following breeds: (31) Santa Inês; (15) Morada Nova; (14) Texel; (11) Suffolk and (12) Ile de France. The heat tolerance index is calculated on the formula 10 - (RT2 - RT1) through data on rectal temperatures (RT) as follows: RT1 at 13:00h, after 2 hours of rest in the shade and RT2 at 15:00h, after na hour of sun exposure and one hour of rest in the shade. The fourth experiment examined a 90 ewe flock, consisted of 18 ewes of each breed, which were evaluated for rectal temperature (RT) and respiratory frequency (RF) as follows: 8:00h, 13:00h (after 2 hour shade rest); 14:00h (after one hour sun exposure) and in the shade, every 15 minutes till the hour; which provided four RT2 for the index. There was no heat tolerance difference between light or dark hair colored Santa Ines. Texel ewes (wool breed) presented the lowest variation of temperature between the two timeframes and had best heat tolerance index (P<0,05) than the hair breed Morada Nova. Suffolk and Ile de France ewes had high heat tolerance index values, irrespective of having been sheared or not. In the fourth experiment, Santa Inês ewes presented lower RT (P<0,05), than some wool breeds at 13:00h, 14:00h, 14:30h and 14:45h. In all breeds, there was no difference (P>0,05) between the RT values taken at 8h and 13h. In addition, no significant differences (P>0,05) were found among the breeds between the RT values recorded in the morning and the RT taken 60min after heat stress (15h). The hair breeds presented lower RF (P<0,05) than the wool breeds. No difference was found amongst the heat tolerance indexes based on RT2 at 45 and 60min after stress. It was concluded that the registering time of RT2 for sheep can be advanced from 60 to 45 min after stress, and that the ewes in the experiment are all adapted to São Paulo State\'s weather.

Efeitos da Síndrome de Down sobre ajustes termorregulatórios e cardiovasculares durante o exercício físico / Effects of Down Syndrome on thermoregulatory and cardiovascular adjustments during physical exercise

A amostra foi composta por 12 indivíduos, 6 indivíduos com Síndrome de Down (SD) e 6 indivíduos sem Síndrome de Down (SSD). Os indivíduos foram submetidos a um protocolo submáximo de exercício, a 65% da potência (W) máxima, pré-estabelecida em um teste de carga progressivo submáximo no cicloergômetro. Os grupos realizaram 30 minutos de exercício, em um ambiente temperado, temperatura média de 21 ± 0,45 oC e umidade relativa do ar média de 61 ± 1,29%. Os indivíduos foram submetidos a um teste para verificação da densidade da urina e seu nível de hidratação. Além disso, receberam um kit de alimentação para ser consumido na noite anterior ao exercício e na manhã horas antes do exercício. Esse kit foi elaborado por uma nutricionista (CRN9 – 6421). Na noite anterior ao exercício, os indivíduos ingeriram uma cápsula que faz a medição da temperatura gastrointestinal. O exercício foi realizado pela manhã, em uma única sessão experimental. Os indivíduos tiveram as temperaturas do trato gastrointestinal (T tgi ) e da pele (T pele ) avaliadas em três momentos: pré-exercício, exercício e pós-exercício. A comparação entre os grupos foi feita usando-se o programa SigmaPlot 11.0. A normalidade de distribuição dos dados foi verificada através do teste de Shapiro-Wilk. Os dados com distribuição normal foram apresentados como média e erro padrão da média (EPM). Os dados quando não apresentaram distribuição normal, através do Teste t de Student, foi utilizado o teste não paramétrico Mann Whitney. Utilizou-se também ANOVA, mais especificamente ANOVA TWO WAY de medidas repetidas, seguidos do teste post hoc de TUKEY. O cálculo do tamanho do efeito foi obtido para análise das comparações dos parâmetros avaliados. O teste Qui-Quadrado foi obtido para avaliar se havia diferença entre os grupos em relação ao nível de atividade física. O nível de significância adotado foi α = 5%. Os resultados mostraram que não houve diferenças estatísticas entre os grupos SD e SSD nas variáveis: peso, estatura, Área de Superfície Corporal, hidratação e idade. Além disso, os resultados da T amb e UR também não apresentaram diferenças. As variáveis, percentual de gordura (SD 28,67 ± 1,14% e SSD 21,25 ± 1,23%) e IMC (SD 31,72 ± 2,21 Kg/m 2 e SSD 25,5 ± 1,53 Kg/m 2 ) foram diferentes entre os grupos, sendo maiores no SD. Através do cálculo do tamanho do efeito, foi possível detectar um tamanho do efeito pequeno para a idade, médio para peso, estatura, ASC, GEU, IMC, URA e grande para percentual de gordura, além de T SALA. A caracterização dos grupos SSD e SD em relação ao nível de atividade física foi mensurada através do IPAQ. Não houve diferença entre os grupos. Tal resultado foi atestado através da utilização do teste Qui-Quadrado. Não foi detectada nenhuma diferença estatística, tanto na potência do teste progressivo (sessão pré-experimental), quanto nos valores de potência pré- estabelecida para a coleta de dados (sessão experimental). Um tamanho do efeito médio foi encontrado para os dois momentos. Não foi encontrada diferença estatística nos dados entre os grupos em relação à FC. O tamanho do efeito foi calculado. Foi obtido um tamanho do efeito médio para a FC durante a sessão pré-experimental e grande para a FC durante a sessão experimental. Em relação à PA não foi encontrada nenhuma diferença estatística nos dados entre os grupos. Já ao longo do tempo, na PAS, houve aumento em relação ao minuto 0 (pré-exercício) no exercício ( SSD entre o minuto 5 e minuto 30 e SD entre o minuto 10 e minuto 30) e minuto 60 (pós exercício). Na PAD, houve aumento também no exercício em relação ao pré e pós exercício, porém, apenas no grupo SD. Além disso, tal diferença foi registrada entre o minuto 5 e minuto 20. Na PAM, o aumento encontrado foi no exercício em relação ao pré e pós exercício, do minuto 5 ao minuto 30, para os 2 grupos. O tamanho do efeito foi calculado para PAS e PAD. Um tamanho do efeito médio foi encontrado, tanto para PAS quanto para PAD. Houve semelhança da Temperatura do Trato Gastrointestinal (T tgi ) entre os grupos SD e SSD. Efeito do grupo (F= 0,661, p= 0,437), efeito do tempo (F= 41,905, p< 0,001) e interação grupo x tempo (F= 2,048, p= 0,008). O tamanho do efeito foi calculado, sendo obtido um tamanho de efeito médio para os momentos de pré-exercício e pós-exercício e grande para o exercício. Além disso, houve diferença estatística entre os grupos na Temperatura da Pele (T pele ) do minuto 15 até o minuto 20 (SD 29,5 ± 0,02oC e SSD 30,9 ± 0,06 oC p<0,05) e no minuto 30 (SD 29,9 ± 0,8 oC vs SSD 31,5 ± 0,9 oC p<0,05), durante o exercício. Efeito do grupo (F= 47,595, p< 0,001), efeito do tempo (F= 2,901, p< 0,001) e interação grupo x tempo (F= 0,465, p= 0,977). O tamanho do efeito foi calculado, sendo obtido um tamanho de efeito médio para os momentos de pré-exercício e pós-exercício e grande para o exercício. Não houve diferença estatística entre os grupos em relação ao acúmulo de calor (AC) e a taxa de acúmulo de calor (TAC). O tamanho do efeito foi obtido e resultou em um tamanho do efeito grande para AC e TAC. Na Variabilidade da Frequência Cardíaca (VFC) no domínio de tempo, não houve nenhuma diferença para os índices Media RR, SDNN, RMSSD e PNN50. Efeito do grupo (F= 8,756, p= 0,005), efeito do tempo (F= 0,826, p= 0,536) e interação grupo x tempo (F= 0,520, p= 0,760). No domínio da freqüência não houve diferença estatística nos parâmetros VLF e LF/HF em nenhum dos momentos supracitados. No parâmetro LF, houve diferença estatística nos momentos 50 (pré-exercício) e 20 (pós-exercício) entre os grupos. Já no HF, houve diferença estatística no momento 30 (exercício), sendo o valor para SD menor nos dois parâmetros. Efeito do grupo (F= 5,856, p= 0,036), efeito do tempo (F= 0,556, p= 0,860) e interação grupo x tempo (F= 0,330, p= 0,977). As respostas termorregulatórias no que se refere à T tgi dos grupos SD e SSD são semelhantes nos 3 momentos avaliados, pré-exercício, exercício e pós-exercício. Além disso, a T pele dos indivíduos SD foi estatisticamente menor no exercício e semelhante no pré e pós-exercício em relação aos indivíduos SSD. Os parâmetros cardiovasculares FC, VFC apresentaram resultados semelhantes. Na PA, não houve diferenças estatísticas na PAS e PAM, apenas na PAD. Essa diferença pode explicar a menor T pele dos SD. Isso por conta da ligação dos fatos, uma maior PAD causa um aumento da resistência vascular periférica, que por sua vez prejudica a vasodilatação, já diminuída nos SD por conta de um nível plasmático menor de óxido nítrico. O acréscimo na resistência vascular periférica, somado a uma vasodilatação prejudicada, diminui a dissipação de calor e consequentemente é detectado um menor valor da T pele. / The aim of the study was to measure gastrointestinal temperature and skin temperature in individuals with Down Syndrome during a submaximal physical exercise. The sample consisted of 12 individuals, 6 individuals with Down Syndrome (DS) and 6 individuals without Down Syndrome (SSD). Subjects were submitted to a submaximal exercise protocol, at 65% of maximal power (W), pre-established in a submaximal progressive load test on the cycle ergometer. The groups performed 30 minutes of exercise in a temperate environment, mean temperature of 21 ± 0.45 ° C and relative humidity of 61 ± 1.29%. The subjects underwent a test to verify the density of the urine and its level of hydration. In addition, they received a feeding kit to be consumed the night before exercise and in the morning hours before exercise. This kit was prepared by a nutritionist (CRN9 - 6421). The night before exercise, the subjects ingested a capsule that measures gastrointestinal temperature. The exercise was performed in the morning in a single experimental session. The individuals had the temperatures of the gastrointestinal tract (Ttgi) and skin (Tpele) evaluated at three moments: pre-exercise, exercise and post-exercise. The comparison between the groups was made using the program SigmaPlot 11.0. The normal distribution of the data was verified through the Shapiro-Wilk test. Data with normal distribution were presented as mean and standard error of the mean (EPM). The non-parametric Mann Whitney test was used when the Student's t-test was not normal. ANOVA, more specifically ANOVA TWO WAY of repeated measurements, followed by the post-hoc test of TUKEY was used. The effect size calculation was obtained to analyze the comparisons of the parameters evaluated. The chi-square test was obtained to evaluate if there was difference between the groups in relation to the level of physical activity. The level of significance was α = 5%. The results showed that there were no statistical differences between the SD and SSD groups in the variables: weight, height, Body Surface Area, hydration and age. In addition, the results of Tamb and UR also showed no differences. The variables, fat percentage (SD 28.67 ± 1.14% and SSD 21.25 ± 1.23%) and BMI (SD 31.72 ± 2.21 kg / m2 and SSD 25.5 ± 1.53 Kg / m2) were different between the groups, being higher in SD. By calculating the effect size, it was possible to detect a small effect size for age, mean for weight, height, ASC, GEU, BMI, URA and large for fat percentage, in addition to T SALA. Characterization of the SSD and SD groups in relation to the level of physical activity was measured using the IPAQ. There was no difference between groups. This result was attested through the Chi-Square test. No statistical difference was detected in either the power of the progressive test (pre-experimental session) or the pre-established power values for data collection (experimental session). An average effect size was found for the two moments. No statistical difference was found in the data between the groups in relation to HR. Effect size was calculated. A mean effect size for the HR during the pre-experimental session and large for the HR during the experimental session was obtained. Regarding BP, no statistical difference was found between the groups. Over time, in SBP, there was an increase over minute 0 (pre-exercise) in exercise (SSD between minute 5 and minute 30 and SD between minute 10 and minute 30) and minute 60 (post exercise). In the DBP, there was also an increase in exercise in relation to pre and post exercise, but only in the SD group. In addition, this difference was recorded between minute 5 and minute 20. In the PAM, the increase was found in the exercise in relation to pre and post exercise, from minute 5 to minute 30, for the 2 groups. The effect size was calculated for SBP and DBP. An average effect size was found for both SBP and DBP. There was similarity of the Temperature of the Gastrointestinal Tract (Ttgi) between the SD and SSD groups. Effect of the group (F = 0.661, p = 0.437), time effect (F = 41,905, p <0.001) and group x time interaction (F = 2.048, p = 0.008). The effect size was calculated, obtaining an average effect size for the pre-exercise and post-exercise moments and great for the exercise. In addition, there was a statistical difference between the groups at Skin Temperature (Tpele) from minute 15 to minute 20 (SD 29.5 ± 0.02 ° C and SSD 30.9 ± 0.06 ° C p <0.05) and at minute 30 (SD 29.9 ± 0.8 ° C vs SSD 31.5 ± 0.9 ° C p <0.05), during exercise. Effect of the group (F = 47.595, p <0.001), time effect (F = 2.901, p <0.001) and group x time interaction (F = 0.465, p = 0.977). The effect size was calculated, obtaining an average effect size for the pre-exercise and post-exercise moments and great for exercise. There was no statistical difference between the groups in relation to the accumulation of heat (AC) and the rate of accumulation of heat (CAT). Effect size was obtained and resulted in a large effect size for AC and TAC. In the Time Domain Variability (HRV) in the time domain, there was no difference for the RR, SDNN, RMSSD and PNN50 indices. Effect of the group (F = 8.756, p = 0.005), time effect (F = 0.826, p = 0.536) and group x time interaction (F = 0.520, p = 0.760). In the frequency domain there was no statistical difference in the VLF and LF / HF parameters in any of the above mentioned moments. In the LF parameter, there were statistical differences at moments 50 (pre- exercise) and 20 (post-exercise) between groups. In HF, there was statistical difference at the 30th moment (exercise), the value for SD being lower in both parameters. Effect of the group (F = 5.856, p = 0.036), time effect (F = 0.556, p = 0.860) and group x time interaction (F = 0.330, p = 0.977). The thermoregulatory responses regarding Ttgi from the SD and SSD groups are similar at the 3 evaluated moments, pre-exercise, exercise and post-exercise. In addition, the Tpele of SD individuals was statistically lower in exercise and similar in pre and post exercise compared to SSD subjects. The cardiovascular parameters HR, HRV presented similar results. In PA, there were no statistical differences in SBP and MAP, only in DBP. This difference may explain the lower Tpele of SD. Due to the fact of the facts, a higher PAD causes an increase in peripheral vascular resistance, which in turn impairs the already reduced vasodilatation in SD because of a lower plasma level of nitric oxide. The increase in peripheral vascular resistance, coupled with impaired vasodilation, decreases heat dissipation and consequently a lower Tpele value is detected. / Lattes não encontrado.

Levando em conta que a maior parte do território nacional se encontra em regiões de clima tropical, a adaptabilidade desses animais a temperaturas ambientais aumentadas é extremamente importante, tanto do ponto de vista produtivo como reprodutivo. O objetivo do presente estudo foi avaliar o efeito da presença ou ausência de sombra, nos parâmetros fisiológicos e seminais de bovinos de raças localmente adaptadas. Touros Curraleiro Pé-Duro, Crioulo Lageano, Pantaneiro e Nelore, foram submetidos a presença ou ausência de sombra e coletados mensalmente durante o período do verão na região Centro-Oeste do Brasil, para avaliação de parâmetros fisiológicos e seminais. As raças apresentaram resultados semelhantes para as características fisiológicas e seminais tanto na sombra quanto no sol, e as raças Curraleiro Pé- Duro e Nelore do grupo sol, apresentaram aumento na quantidade de defeitos menores dos espermatozoides no sêmen fresco, na última coleta quando comparados as coletas anteriores na mesma raça e tratamento, entretanto, não apresentaram diferença significativa entre as raças e tratamentos na última coleta. As raças estudadas demonstram alta adaptabilidade as condições climáticas presentes no verão da região Centro-Oeste do Brasil. / Brazil is one of the largest cattle producers in the world, and much of the reproduction of these animals is still carried out by natural breeding. Most of the national territory is in regions of tropical climate, then the adaptability of these animals to increased environmental temperatures is extremely important, both from productive and reproductive point of view. The objective of the present study was to evaluate the effect of the presence or absence of shade on physiological and seminal parameters of cattle of locally adapted breeds. Curraleiro Pé-Duro, Crioulo Lageano, Pantaneiro and Nelore buuls, were submitted to the presence or absence of shade and collected monthly during the summer period in the Center-West region of Brazil, for the evaluation of physiological and seminal parameters. Breeds presented similar results for physiological and seminal characteristics in both under shade and under the sun, Curraleiro Pé-Duro and Nelore of the sun group showed an increase in the number of minor sperm defects in fresh semen, in the last collection when compared to previous collections in same breed and treatment, however, showed no significant difference between breeds and treatments in the last collection. Breeds studied show high adaptability to the climatic conditions present in the summer of the Center-West region of Brazil.

No experimento 4, ovelhas da raça Santa Inês tiveram TR inferiores (P<0,05) à TR de algumas raças lanadas, às 13h, 14h, 14h30min e 14h45min. Em todas as raças, não houve diferença (P>0,05) entre as TR medidas às 8h e 13h, assim como não foram encontradas diferenças (P>0,05) entre elas quanto à TR da manhã e aos 60 min após o estresse (15h). As ovelhas deslanadas tiveram FR inferiores (P<0,05) às lanadas. Não houve diferença (P>0,05) entre o ITC calculado com base na TR2 aos 45 e 60 min pósestresse. Concluiu-se que o tempo de registro da TR2 para a espécie ovina pode ser antecipado de 60 para 45 minutos após o estresse, e que as ovelhas avaliadas estão adaptadas ao clima do Estado de São Paulo. / This study consisted of 4 experiments and had as main goal evaluate the heat tolerance in ewes, and its relation to hair color and presence or absence of wool in wool and hair breeds. The experiments were conducted at the Instituto de Zootecnia, located in Nova Odessa city, São Paulo State, Brazil (22º42\'S e 47º18\'W, 570m altitude). Three experiments evaluated 83 ewes of the following breeds: (31) Santa Inês; (15) Morada Nova; (14) Texel; (11) Suffolk and (12) Ile de France. The heat tolerance index is calculated on the formula 10 - (RT2 - RT1) through data on rectal temperatures (RT) as follows: RT1 at 13:00h, after 2 hours of rest in the shade and RT2 at 15:00h, after na hour of sun exposure and one hour of rest in the shade. The fourth experiment examined a 90 ewe flock, consisted of 18 ewes of each breed, which were evaluated for rectal temperature (RT) and respiratory frequency (RF) as follows: 8:00h, 13:00h (after 2 hour shade rest); 14:00h (after one hour sun exposure) and in the shade, every 15 minutes till the hour; which provided four RT2 for the index. There was no heat tolerance difference between light or dark hair colored Santa Ines. Texel ewes (wool breed) presented the lowest variation of temperature between the two timeframes and had best heat tolerance index (P<0,05) than the hair breed Morada Nova. Suffolk and Ile de France ewes had high heat tolerance index values, irrespective of having been sheared or not. In the fourth experiment, Santa Inês ewes presented lower RT (P<0,05), than some wool breeds at 13:00h, 14:00h, 14:30h and 14:45h. In all breeds, there was no difference (P>0,05) between the RT values taken at 8h and 13h. In addition, no significant differences (P>0,05) were found among the breeds between the RT values recorded in the morning and the RT taken 60min after heat stress (15h). The hair breeds presented lower RF (P<0,05) than the wool breeds. No difference was found amongst the heat tolerance indexes based on RT2 at 45 and 60min after stress. It was concluded that the registering time of RT2 for sheep can be advanced from 60 to 45 min after stress, and that the ewes in the experiment are all adapted to São Paulo State\'s weather.

A melatonina desempenha funções fisiológicas importantes, como o aumento da capacidade de termogênese do tecido adiposo marrom. Além dos depósitos adiposos branco e marrom, há um terceiro tipo, as células adiposas beges. Presentes no tecido adiposo branco, apresentam características semelhantes aos adipócitos marrons via um processo denominado browning que está relacionado com a termogênese e aumento do gasto energético. Diante disso, o objetivo deste estudo foi investigar o papel da melatonina sobre a regulação da temperatura corporal em animais idosos obesos da linhagem Wistar que apresentam produção de melatonina prejudicada. Por 16 semanas os animais foram divididos nos grupos controle e suplementado com melatonina e viu-se que os adipócitos marrons e beges atuam de forma coordenada e complementar para garantir a produção de calor de maneira adequada e que a melatonina tem papel importante nos mecanismos termorregulatórios, assegurando maior capacidade de suportar ao frio e, ainda, participando da regulação do balanço energético. / The hormone melatonin plays several physiological functions, one of them is the increase of thermogenesis capacity of brown adipose tissue. In addition to white and brown adipose deposits, there is also a third type, the beige adipocyte, which presents similar characteristics to brown adipocytes, induced in a process called browning. This process is related to thermogenesis and the increase of energy expenditure. Thus, the aim of this study was to investigate the role of melatonin on the regulation of body temperature in elderly obese animals that have impaired production of melatonin. Male-Wistar were divided into control group and group supplemented with melatonin, and the experimental protocol lasted 16 weeks. The main results showed that brown and beige adipocytes act in a coordinated and complementary way to ensure adequate heat production and that melatonin plays an important role in the thermoregulatory mechanisms, ensuring greater capacity to withstand the cold and, also, participating in the regulation of the energy balance.

Anfíbios tem a habilidade de manifestar febre comportamental em ambientes heterotermais durante infecção a um custo metabólico associado à elevação da temperatura corpórea e à ativação do sistema imune. Apesar do custo metabólico, a temperatura corpórea febril otimiza a resposta imune no combate à infecção e aumenta as chances de sobrevivência do indivíduo. Contudo, devido à limitada capacidade de termorregular, os anfíbios enfrentam variações diárias e sazonais na temperatura corpórea e na resposta metabólica de reação à infecção. O nosso objetivo foi medir a variação da resposta metabólica à infecção dentro da variação de temperaturas ecológicas relevantes do sapo Cururu. Testamos a hipótese de que a infecção aumenta as taxas metabólicas do sapo Cururu, mas o custo energético da resposta imune deve ser menor na temperatura febril dos sapos infectados. Para testarmos as hipóteses, nós medimos a temperatura operacional dos sapos no campo, a preferencia termal dos sapos hígidos e a temperatura preferencial dos sapos infectados. Depois, medimos a taxa metabólica e a resposta metabólica dos sapos antes e depois da infecção por LPS nessas temperaturas. Nossos resultados mostraram que as temperaturas ecológicas relevantes dos sapos variaram entre 17°C e 26°C. A temperatura influenciou a taxa metabólica dos sapos, mas só na temperatura preferencial dos sapos hígidos houve custo metabólico associado à infecção. Contudo, na temperatura corpórea dos sapos infectados a resposta metabólica de reação à infecção foi menor, indicando que o controle regulado no ponto de ajustes \"set-point\" da temperatura corpórea durante a infecção coevoluiu com um custo energético otimizado da resposta imune / Anphibians have the ability of manifested behavioral fever in heterothermal environments during infection with a metabolic cost associated to elevated body temperature set-point and due to activation of immune system. Despite the metabolic cost, fever body temperature optimizes immune response to combat infection and increase the survival of the host. However, because of the limited capacity for thermoregulation, amphibians can confront daily and seasonal variation in body temperature and in the metabolic response of reaction to combat infection. So, we measured the variation in metabolic response of reaction to infection at ecology relevant body temperature range in Cururu toads. We hypothesized that infection increases metabolic rates of the Cururu toads due to the activation of the immune system at different temperatures, but the energetic cost of immune response is lower at preferred body temperature of infected toads (behavioral fever). To test these hypotheses we measured the operative body temperature in the field, the preferred body temperature of higid toads, and the preferred body temperature of infected toads. After, we measured metabolic rate and metabolic response of the toads before and after injection of LPS at these temperatures. Our results showed that the ecology relevant temperature range of Cururu toads (R. icterica) varies between 17°C and 26°C, respectively, at operative temperature and at preferred body temperature in infected toads when exposed to heterothermal environment. The temperature had the major impact on metabolic rate of the toads during infection. But, at fever body temperature toads decrease the metabolic response of reaction to infection, indicating that the regulated control of body temperature set-point during infection coevolved with an optimized energetic cost of immune response

Estudo sobre respostas comportamentais à infecção no anfíbio anuro Proceratophrys boiei / A study of behavioral responses to infection in the anuran amphibian Proceratophrys boiei

Esta tese tem como tema central o conceito de febre comportamental, que tem-se definido como o aumento da temperatura corporal pela efetivação da mudança nas temperaturas preferidas devido ao reconhecimento, por parte do corpo, de uma infecção ou patógeno. O trabalho está composto por três partes. Na Introdução Geral é discutida a febre comportamental em sete pontos fundamentais no entendimento desta resposta dentro da Ecofisiologia, começando pela definição, as pesquisas de laboratório e campo existente até o momento, até as limitações ecológicas dos indivíduos e as implicações do tema na conservação. No capítulo 1, com formato de texto científico, apresentamos a pesquisa na qual estudamos o comportamento e a preferência termal da espécie Proceratophrys boiei em condições experimentais com indivíduos injetados com lipopolisacáridos (LPS), para simular uma infecção, e indivíduos intactos (injetados com salina, grupo controle). Nessa pesquisa consideramos duas alternativas de respostas no comportamento, tal como discutido na introdução geral: a) febre comportamental, que é caracterizada por um aumento da temperatura corporal dos indivíduos pela mudança nas temperaturas preferidas dentro de uma paisagem termal; b) comportamento de doente, que no contexto do desenho experimental seria reconhecido pela diminuição da atividade dos indivíduos. Assim, registramos os seguintes tratamentos durante 24 horas com uma câmara termográfica: 1) indivíduos intactos no gradiente termal desligado, 2) indivíduos intactos no gradiente termal ligado, 3) indivíduos injetados com salina, no gradiente termal ligado 4) indivíduos injetados com LPS no gradiente termal ligado. Para cada um dos tratamentos foi registrada a distância de locomoção e as preferências termais, junto com outros detalhes do comportamento e as preferências termais. A partir de nossos resultados, concluímos que os indivíduos de P. boiei apresentam comportamento de doente como resposta dominante ao ser injetados com LPS, e que as preferências termais destes são consequência do comportamento de doente e não da termorregulação comportamental. Finalmente, a discussão geral explica como o capitulo 1 contribui na discussão de cada um dos sete pontos tratados na introdução geral tentando propor metodologias e estudos mais completos para manter o diálogo entre a fisiologia e a ecologia dos indivíduos no contexto de infecção e doenças / The central theme of this thesis is the concept of behavioral fever, which has been defined as the increase of body temperature by effecting the change in preferred temperatures due to the recognition by the body of an infection or pathogen. The thesis is composed of three parts. In the General Introduction, behavioral fever is presented around seven fundamental points to the understanding of this response within Ecophysiology, starting with the definition, the laboratory and field research until now, to the ecological limitations of individuals and the implications of this theme in conservation. In the first chapter, with scientific text format, we present the research in which we studied the behavior and thermal preference of Proceratophrys boiei species under experimental conditions in individuals injected with lipopolysaccharides (LPS), to simulate an infection, and in intact individuals (injected with Saline, a control group). In this research we considered two alternatives of behavioral responses, as discussed in the General Introduction: a) behavioral fever, which is characterized by an increase in the individuals body temperature by changing the preferred temperatures within a thermal landscape; B) patient behavior, which, in the context of experimental design, would be recognized by the decrease in the activity of individuals. Thus, we recorded the following treatments for 24 hours with a thermographic camera: 1) intact individuals in the thermal gradient switched off, 2) intact individuals in the connected thermal gradient, 3) individuals injected with saline, in the bound thermal gradient 4) individuals injected with LPS in the thermal gradient on. For each of the treatments it was recorded the locomotion distance and the thermal preferences, along with other details of the behavior and the thermal preferences. From our results, we conclude that the individuals of P. boiei present a patient\'s behavior as a dominant response when injected with LPS and their thermal preferences are a consequence of patient behavior and not behavioral thermoregulation. Finally, the general discussion explains how chapter 1 contributes to the discussion of each of the seven points highlighted in the general introduction attempting to propose a complete methodology and studies to maintain the dialogue between the physiology and the ecology of individuals in the context of infection and disease

Em mamíferos, a temperatura corporal (Tc) varia ao longo do dia, de forma rítmica, bem como a atividade locomotora. Essa ritmicidade diária é gerada endogenamente, por osciladores circadianos. Em geral, os valores mais elevados de Tc concentram-se nos mesmos horários em que há atividade locomotora. No entanto, variações diárias de Tc são observadas mesmo em indivíduos imóveis, evidenciando que o ritmo de Tc não é uma mera consequência dos efeitos agudos da atividade locomotora. Este trabalho teve o objetivo de estudar o ritmo circadiano de Tc no roedor Ctenomys aff. knighti (tuco-tuco) e sua relação temporal com o ritmo de atividade locomotora. A caracterização do ritmo de Tc nesse animal é particularmente interessante, pois ele habita o ambiente subterrâneo, que impõe desafios tanto à expressão rítmica (pois os ciclos ambientais são ausentes ou têm baixa amplitude) quanto à termorregulação (devido à características como alta umidade e atmosfera hipóxica e estagnada). Medimos a Tc e a atividade locomotora dos tuco-tucos em condições controladas de laboratório. Na caracterização inicial, constatamos que o ritmo de Tc dos tuco-tucos persiste mesmo em escuridão e temperatura constantes. Quando houve exposição a um ciclo diário de claro/escuro, o ritmo de Tc foi sincronizado, e os valores mais altos ocorreram na fase de escuro. Os ritmos de Tc e de atividade locomotora mostraram-se fortemente associados no tempo. Investigamos, então, se a corrida na roda de atividade modificaria a amplitude do ritmo de Tc, por envolver atividade muscular intensa. Observamos, no entanto, que a amplitude é pouco alterada com a retirada da roda e que na sua ausência os animais substituem a corrida por outras atividades que também causam efeitos agudos na Tc. Em seguida, estudamos a variação da sensibilidade da Tc aos efeitos agudos da atividade ao longo do dia. Através de um método estatístico, que também foi utilizado para filtrar os dados de Tc, verificamos que há maior correlação entre variações de Tc e de atividade no início da noite. Por fim, investigamos se o controle do ritmo de Tc é exercido pelos núcleos supraquiasmáticos, os quais são os osciladores circadianos do ritmo de atividade locomotora. Nossa abordagem utilizou animais neurologicamente intactos, comparando os padrões do ritmo de Tc e de atividade locomotora durante a ocorrência do fenômeno de \"partição do ritmo\". Constatamos que na maioria dos casos os padrões de partição, tanto da Tc como da atividade locomotora são idênticos, sugerindo um controle temporal comum. No entanto, em um dos animais observamos diferenças entre os dois ritmos, sugerindo que esse controle circadiano pode ser ainda mais complexo. / In mammals, body temperature (Tb), as well as locomotor activity, changes during the day, exhibiting a rhythmic pattern. This daily rhythmicity is generated endogenously by circadian oscillators. Usually, the highest Tb values occur simultaneously to locomotor activity. Nevertheless, daily Tb changes are observed even in immobile individuals. This fact is evidence that the Tb rhythm is not a mere consequence of the acute effects of locomotor activity. The present work,which os performed in the research center CRILAR, in Argentina, had the objective of studying the circadian rhythm of Tb. in the rodent Ctenomys aff. knighti (tuco-tuco) and its temporal relationship with the locomotor activity rhythm. The characterization of the Tb rhythm in this animal is especially interesting because it inhabits the subterranean environment, which poses challenges to both rhythmic expression (because environmental cycles are either absent or have low amplitude) and thermoregulation (due to high relative humidity, hypoxic atmosphere and limited ventilation). We measured Tb and locomotor activity of tuco-tucos under controlled laboratory conditions. An initial characterization showed that the tuco-tuco\'s Tb rhythm persisted even in constant darkness and constant temperature. This rhythm synchronized to a daily light/dark cycle, with highest values occurring during the dark phase. Tb and locomotor activity rhythm were robustly associated in time. Then, we investigated whether the wheel running would modify the Tb rhythm amplitude, because this behavior involves intense muscular activity. However, we observed that the amplitude is only slightly altered upon running-wheel removal and that in the absence of the wheel tuco-tucos substitute running with other behaviors which also have acute effects on Tb. Applying a statistical method, which was also used to filter Tb data, we verified that there is a stronger correlation between Tb changes and activity in the beginning of the night. Finally, we investigated whether the Tb rhythm is also under control of the suprachiasmatic nuclei, which are knowingly the circadian oscillators for the locomotor activity rhythm. We used an approach which involved neurally intact animals, by comparing the rhythmic patterns of Tb and locomotor activity rhythms during the occurrence of a phenomenon called \"splitting\". In most cases, splitting patterns of both Tb and locomotor activity rhythms were identical, indicating a common temporal control of these two variables. Nevertheless, we observed, in one animal, differences between the two rhythms, which suggest that the circadian control might be even more complex.

Uma característica dos insetos sociais, entre elas as abelhas, é o controle da temperatura de seus ninhos. Abelhas sociais são denominadas como animais heterotérmicos, ou seja, são endotérmicas quando realizam atividades motoras (adultas) e ectotérmicas quando apresentam inatividade (cria e abelhas jovens). Essa característica está entrelaçada com o comportamento social. As abelhas melíferas (Apidae; Apini: Apis mellifera) são conhecidas por apresentarem uma temperatura ótima dentro de seus ninhos, regulando a temperatura independente da variação de temperatura ambiente (externa). Entretanto, as abelhas sem ferrão (Apidae; Meliponini) não mantém a temperatura dentro de seus ninhos rigorosamente como as abelhas melíferas. Apesar disso, essas abelhas conseguem manter uma temperatura mais ou menos estável para a emergência perfeita da prole. Porém, se ocorrer algum tipo de alteração de temperatura durante a fase do desenvolvimento ontogênico, esses indivíduos podem não desempenhar suas funções com eficiência, podendo trazer malefícios para a colônia. O objetivo do presente trabalho foi registrar essas temperaturas internas e compará-las com as temperaturas externas (ambiente), como também analisar a termorregulação passiva nas abelhas sem ferrão. Além disso, foi verificado também se estas abelhas podem apresentar uma alteração no comportamento adulto quando desenvolvidas em temperaturas consideradas inadequadas. Foram utilizadas colônias de Melipona scutellaris mantidas em caixas de madeira em uma sala. Para os registros das temperaturas foram utilizados 2 sensores para cada colônia, um na área de cria (dentro do invólucro) e outro na periferia do ninho (fora do invólucro). No momento dos registros internos das colônias, foram anotadas também as temperaturas externas, ou seja, as temperaturas da sala (onde estavam mantidas as colônias) e do ambiente externo. Para controle, foram feitos os registros de temperatura em uma caixa vazia (sem abelhas) com as mesmas dimensões das colônias. Para analisar a termorregulação das abelhas sem ferrão Melipona scutellaris foi utilizada uma estufa, onde foram incubados favos e cera (colônia) em diferentes temperaturas, a fim de verificar se a cera da colônia ou se a cria presente nos favos auxiliava na manutenção da temperatura do ninho. Os resultados obtidos demonstraram que a temperatura na área de cria é sempre maior que na periferia do ninho. Também foi verificado que a temperatura do ambiente influencia na variação da temperatura dentro do ninho. Para verificar a influência da temperatura no desenvolvimento das abelhas, foram realizados em estufa desenvolvimentos de crias em fase de pupa até a emergência dos indivíduos. Foi observado o tempo dos desenvolvimentos, taxas de mortalidade, medidas do corpo e asas como também a análise do comportamento adulto, a fim de avaliar o limiar gustativo. Os indivíduos emergidos das condições experimentais de temperatura (influência de temperatura por meio da estufa) demonstraram de acordo com as medidas do corpo e das asas, tamanhos menores que os indivíduos da colônia, porém não emergiram indivíduos deformados. Com relação ao comportamento adulto, avaliado pelo limiar de resposta gustativa, utilizando o Reflexo de Extensão da Probóscide, os placares gustativos foram semelhantes para os grupos de indivíduos emergidos na situação controle (colônia) e na situação experimental de 32ºC, sugerindo que a temperatura de 32ºC não causa nenhuma deficiência (neural e morfológica) à cria. Porém, os tratamentos de 28 e 30ºC não apresentaram bons desenvolvimentos, e assim, a taxa de mortalidade foi alta e o tempo de desenvolvimento foi longo. Este trabalho foi relevante para os estudos de influência de temperatura no desenvolvimento de imaturos e para a termorregulação colonial. / One of the characteristics of social insects, the bees among them, is the temperature control of their nests. Social bees are called heterothermic animals, e.g. they are endothermic when they carry out motor activities (adults) and ectothermic when they present inertness (hatch and young bees). This characteristic is correlated with their social behavior. Honeybees (Apidae; Apini: Apis mellifera) are known by presenting an optimum temperature inside their nests, correcting temperature independently of environmental temperature variation (external). Meanwhile, stingless bees (Apidae; Meliponini) do not keep temperature inside their nests as rigorously as honeybees. Nevertheless, these bees are able to maintain temperature balanced for a perfect emergency of their offspring. However, in case there is any temperature change during the ontogenic development phase, they may not perform their duties efficiently, bringing harm to their colony. The present study aims to record the internal temperatures and compare them to the environmental ones (external), along with the analysis of passive thermoregulation of the stingless bees. Besides, it was checked out if those bees can reveal an adult behavior alteration when raised under inadequate temperatures. It was placed Melipona scutellaris colonies in wooden boxes in a room. For measurement temperature recording, it was used 2 (two) sensors for each colony, one of them in the foal area that is inside the casing and another in the nest periphery that is outside the casing. Both internal temperatures of the colonies and external ones were recorded, i.e. room temperatures, where boxes were kept in external environment. For comparison control, an empty box without bees was recorded under similar colony dimensions. On analyzing the stingless thermoregulation Melipona scutellaris, it was used a greenhouse for honeycomb and wax incubation (colony) in different temperatures for the purpose of verifying whether the colony wax or the hatch, present in the honeycomb, helps in the nest temperature maintenance. The results show the brood area temperature is always higher than the nest periphery which presents lower temperatures. Likewise, it was seen that the environmental temperature influences the temperature variation inside the nest. Verifying temperature influence in the bees, it was carried out greenhouse developments (pupa) until the emergency of subjects. Development time, mortality rate, body and wing measurements as well as adult behavior were observed with the view to assessing taste threshold. Subjects emerged from experimental condition of temperature influenced by greenhouse showed, according to body and wing measurements, smaller size than the colony subjects, although there were no malformed subjects. Concerning adult behavior evaluated by taste threshold answer using Proboscis Extension Reflex, gustatory scores were similar to all groups emerged from the control situation (colony) and under the experimental condition of 32°C, suggesting that 32°C does not produce any neural and morphological damage in the hatch. But, 28º and 30°C treatments did not come up with good developments, therefore mortality rate was high and development period of time was long. This work was relevant to the temperature influence on immature development and colonial thermoregulation.
