A imunoglobulina E (IgE) tem papel central na patogênese das doenças alérgicas. É parte da resposta do tipo 2, e as citocinas IL-4 e IL-13 são essenciais para que haja produção deste isotipo de imunoglobulina. Produção de IgE é também induzida por parasitas intestinais, particularmente helmintos. O objetivo do presente estudo foi avaliar a contribuição de IgE específica para ácaros, barata, gato, cachorro e para o parasita Ascaris lumbricoides sobre os níveis de IgE total entre crianças de áreas distintas no Brasil. Anticorpos IgE para ácaros Dermatophagoides pteronyssinus e Blomia tropicalis; barata Blattella germanica; gato; cachorro; e A. lumbricoides foram medidos usando o sistema ImmunoCAP, e comparados à IgE total no soro de 150 crianças de 3-6 anos de idade vivendo na cidade de Natal, endêmica para parasitoses intestinais, e de 54 crianças de 3-6 anos de idade com asma e/ou rinite, vivendo em Ribeirão Preto. Níveis de IgE total foram significantemente mais elevados em crianças de Natal quando comparados aqueles em crianças de Ribeirão Preto (média geométrica 630,9 kU/L, faixa 19,6-63.290 kU/L; e 398,1 kU/L, faixa 35,7-4.803 kU/L, respectivamente). Entre as 150 crianças de Natal, 52(34,6%) apresentaram IgE positiva para D.pteronyssinus; 70(46,6%) para B.tropicalis; 45(30%) para barata; 19(12,6%) para gato; 17(11,3%) para cachorro; e 125(83,3%) para A.lumbricoides. Entre as 54 crianças com asma e/ou rinite de Ribeirão Preto, 41(75,9%) apresentaram IgE positiva para D.pteronyssinus; 34(62,9%) para B.tropicalis; 22(40,7%) para barata; 11(20,3%) para gato; 12(22,2%) para cachorro. Embora estas crianças fossem negativas para parasitas à inclusão no estudo, 22(40,7%) tinham IgE para A.lumbricoides. Anticorpos IgE para A. lumbricoides foram mais elevados entre crianças de Natal, quando comparados a IgE para alérgenos inalantes (p<0,01). Níveis de IgE para D. pteronyssinus entre crianças de Ribeirão Preto foram mais altos que IgE para outros inalantes e A.lumbricoides (p<0,01). Em Natal, a porcentagem de IgE para A. lumbricoides em relação à IgE total foi maior em comparação a IgE para D.pteronyssinus e B.germanica (mediana 0,41%; 0,08%; e 0,04% respectivamente, p<0,01). Em Ribeirão Preto, a porcentagem de IgE para D.pteronyssinus e para B.tropicalis em relação à IgE total foi maiorem comparação a IgE para A.lumbricoides e barata (mediana 9,8%; 0,6%; 0,3%; e 0,2%, respectivamente, p<0,05). Regressão linear revelou que a associação mais forte foi para IgE para A.lumbricoides com IgE total em Natal (R²=0,56; p<0,01); associação significante foi também observada para IgE para ácaros com IgE total em Ribeirão Preto (R2=0,35; p<0,01 para D.pteronyssinus; R2=0,33; p<0,01 para B.tropicalis, respectivamente). Nossos resultados demonstraram que anticorpos IgE para ácaros contribuem fortemente para a IgE total entre crianças com asma e/ou rinite, vivendo em uma área de baixa taxa de infecções parasitárias em nosso meio. Por outro lado, entre crianças vivendo em uma área em que parasitas são encontrados em abundância, infecções parasitárias induzem uma forte resposta IgE policlonal, e anticorpos IgE específicos para parasita, além de ácaros, barata, gato e cachorro representam uma modesta proporção da IgE total. A especificidade desta IgE, e os efeitos a longo prazo desta resposta cedo na vida, permanecem desconhecidos. / Immunoglobulin E (IgE) plays a central role in the pathogenesis of allergic diseases. It is part of the type 2 response, and the cytokines IL-4 and IL-13 are essential for the production of this immunoglobulin isotype. IgE production is also induced by intestinal parasites, particularly helminths. The aim of the present study was to evaluate the contribution of specific IgE to mites, cockroach, cat, dog and parasite Ascaris lumbricoides on total IgE levels among children living in different areas in Brazil. IgE antibodies to mites Dermatophagoides pteronyssinus and Blomia tropicalis; cockroach Blattella germanica; cat; dog; and A. lumbricoides were measured using the ImmunoCAP system and compared to total serum IgE of 150 children 3-6 year-old living in the city of Natal, endemic for intestinal parasites, and 54 children 3-6 years of age with asthma and /or rhinitis, living in Ribeirão Preto. Total IgE levels were significantly higher in children from Natal as compared to those among children in Ribeirão Preto (geometric mean 630,9 kU/L, range 19,6-63.290 kU/L; e 398,1 kU/L, range 35,7-4.803 kU/L, respectively). Among the 150 children from Natal, 52(34.6%) presented IgE positive to D.pteronyssinus; 70(46.6%) to B. tropicalis; 45(30%) to cockroach; 19 (12.6%) to cat; 17(11.3%) to dog; and 125(83.3%) to A. lumbricoides. Among the 54 children with asthma and /or rhinitis from Ribeirão Preto, 41(75.9%) had IgE positive to D.pteronyssinus; 34(62.9%) to B.tropicalis; 22(40.7%) to cockroach; 11(20.3%) to cat; 12(22.2%) to dog. Although these children were negative for parasites at inclusion in the study, 22(40.7%) had IgE to A.lumbricoides. IgE antibodies to A.lumbricoides were higher among children from Natal, as compared to IgE to inhalant allergens (p<0.01). IgE levels to D.pteronyssinus among children living in Ribeirão Preto were higher than IgE to other inhalants and A.lumbricoides (p<0.01). In Natal, the percentage of IgE to A.lumbricoides in relation to total IgE was higher in comparison to IgE to D.pteronyssinus e B.germanica (median 0.41%, 0.08%, and 0.04% respectively, p<0.01). In Ribeirão Preto, the percentage of IgE to D.pteronyssinus and to B.tropicalis in relation to total IgE was higher in comparison to IgE to A.lumbricoides and cockroach (median 9.8%, 0,6%, 0.3%, and 0.2%, respectively, p <0.05). Linear regression analysis revealed that the strongest association was for IgE to A.lumbricoides with total IgE in Natal (R²=0.56, p<0.01); significantassociation was also observed for IgE to mites with total IgE in Ribeirão Preto (R2=0.35, p<0.01 for D. pteronyssinus, R2=0.33, p<0.01 for B.tropicalis, respectively). Our results demonstrated that IgE antibodies to mites contribute strongly to total IgE among children with asthma and /or rhinitis, living in an area of low parasite infection rates in our country. On the contrary, among children living in an area where parasites are found in abundance, parasitic infections induce a strong polyclonal IgE response, and IgE antibodies specific for parasite, and also for mites, cockroaches, cat and dog represent a modest proportion of total IgE. The specificity of these IgE antibodies and the implications of this response occurring early in life remain unknown.

La Llorona Don’t Swim The LA River is a collection of poems that explore the issues of growing up in East LA as a young, bilingual Chicana. These poems are an attempt to capture my experiences through story telling by blending Spanish, English, street slang, and the casual spoken diction. The speaker embodies several versions of the trickster figure: linguistic tricksterism, cultural tricksterism, gender tricksterism, and religious tricksterism. She navigates linguistic tricksterism through the code switching of English and Spanish, using Spanglish as her main dialect, and often blending in slang, creating Slanglish. In cultural tricksterism, she focuses on the experiences of the working class, the migrant workers, the bilingual speakers, and the overall generational changes in the Mexican culture. The speaker constantly searches for the meaning of what it is to be a woman and what it means to love through gender tricksterism. She looks to her mother, la Virgen De Guadalupe, and La Llorona to try and understand her complex identity. La Llorona Don’t Swim The LA River works to form a different type of spirituality through sacred humanism from the point of view of the religious trickster. These poems pose the question: where else can God be found other than church and the bible? The speaker seeks God in tattoos, in punk music, in the moshpit, in the city, and in people. She forms a new approach to tradition through the persistence of kind rebellion, her own personal version of love.

Efficacy evaluation of Mohlolo Insecticide Bait on German Cockroach, Blattella Germanica LMbowane, Clarah January 2014 (has links)

(Plant Production)) -- University of Limpopo, 2014 / In most cases the discipline plant protection is restricted to the protection of propagules, the plants and their produce in storage, while excluding the protection of processed (cooked) food from contaminants. Generally, the German cockroach (Blattella germanica L.) is a serious contaminant of processed food and had recently been viewed as a pest worthy of control in plant protection when viewing the entire value chain of agricultural commodities. Due to the nature of the pest, it is quite difficult to control using registered agrochemicals on the market. An entrepreneur in the rural areas of Limpopo Province developed bait for cockroach control, which was code named Mohlolo during efficacy trials. The use of baiting in cockroach management is currently the sought after approach in the world where cockroaches are problematic. Bait has the advantage over liquid or dust formulations because baiting requires shorter service, has shown increased efficacy, and has reduced environmental contamination. The objectives of this study were to determine whether the effect of MIB would be age related in the suppression of populations of B. germanica under laboratory conditions, and also to investigate whether MIB would reduce populations of B. germanica in residential areas. Five concurrent B. germanica age-related experiments were run in a complete randomized design. 1-d, 2-d, 3-d, 4-d, 5-d old nymphs and adult German cockroach experiments were conducted at Limpopo Agro-Food Technology Station (LATS) laboratory, (53’21,41” S/29 23o 44’19,95”E) hosted at the University of Limpopo. Each age group consisted of 10 nymphs and was put in 250 cm3 container that contained

Mohlolo insecticide bait (MIB). One gram (1 g) of Mohlolo insecticide bait and moistened cotton wick were put at the bottom centre of the 250 cm3 containers containing nymphs based according to their respective ages. Residential experiments were conducted at UL student apartments. Two Bait containers were then placed in the kitchen, lodge, bathroom and bedroom. In the kitchen MIB containers were put behind the cooking stove and refrigerator, in the lodge the bait was placed next to the study table (because most study tables are also used during the eating time). In the bathroom the baits were put next to toilet seat and lastly in the bedroom, the bait was placed next to study table. According to the results, 100% mortality of 1-, 2-, 3-, 4-, 5-days nymphs and adult B. germanica was obtained after 7th, 2nd 3rd 3rd and 4th day, whereas LT50 values ranged from half a day to one and half (1.5) day. The study also demonstrated that B. germanica populations had high LT50 and LT100 values in residential areas compared to LT50 and LT100 values obtained in the laboratory experiments. This is probably due to the fact that residential areas that have low levels of sanitation availed the cockroaches with an opportunity to choose from many foods hence, it reduced the probability for cockroaches to feed on Mohlolo insecticide bait in the respective apartments. Results of this study showed that, Mohlolo bait toxicity and effectiveness as a control agent can reduce infestation of cockroaches in all developmental stages when applied at small quantity. Providing results of this study would provide information on resistance and tolerance of B. germanica nymphs and adult population against MIB in the

laboratory and residential areas. It would also provide biological information on the efficacy of MIB against the German cockroaches.

Die neuronale Kontrolle der Speicheldrüse von Periplaneta americana / The neuronal control of the salivary glands of Periplaneta americanaRotte, Cathleen January 2009 (has links)
Die acinösen Speicheldrüsen der Schabe Periplaneta americana sind reich durch serotonerge, dopaminerge und GABAerge Fasern innerviert. Die biogenen Amine Serotonin (5-HT) und Dopamin (DA) induzieren die Sekretion eines NaCl-haltigen Primärspeichels. Die physiologische Rolle der GABAergen Innervation des Drüsenkomplexes war bislang unbekannt. Weiterhin wurde vermutet, dass Tyramin (TA) und Octopamin (OA) an der Speichelbildung beteiligt sind. Mittels intrazellulärer Ableitungen von sekretorischen Acinuszellen mit und ohne Stimulierung des Speicheldrüsennervs (SDN) sollte daher die Wirkung von GABA, TA und OA im Speicheldrüsenkomplex untersucht werden. Intrazelluläre Ableitungen aus Acinuszellen zeigten, dass sowohl DA als auch 5 HT biphasische Änderungen des Membranpotentials induzierten. Diese bestanden aus einer initialen Hyperpolarisation und einer darauf folgenden transienten Depolarisation. Stimulierung des SDN mittels einer Saugelektrode verursachte ebenfalls biphasische Änderungen des Membranpotentials der Acinuszellen, die mit den DA- bzw. 5-HT-induzierten Änderungen kinetisch identisch waren. Dieses Ergebnis zeigte, dass die elektrische Stimulierung des SDN im Nerv-Speicheldrüsenpräparat eine verlässliche Methode zur Untersuchung der Wirkungen von Neuromodulatoren auf die dopaminerge und/oder sertotonerge Neurotransmission ist.
Die Hyperpolarisation der DA-induzierten Potentialänderungen wurde durch eine intrazelluläre Ca2+-Freisetzung und die Öffnung basolateral lokalisierter Ca2+-gesteuerter K+-Kanäle verur-sacht. Die DA- und 5-HT-induzierte Depolarisation hing kritisch von der Aktivität eines basolateral lokalisierten Na+-K+-2Cl--Symporters ab.
GABA, TA und OA potenzierten die elektrischen Antworten der Acinuszellen, wenn diese durch SDN-Stimulierung hervorgerufen wurden. Dabei war OA wirksamer als TA. Dieses Ergebnis zeigte, dass diese Substanzen als im Drüsenkomplex präsynaptisch und erregend als Neuromodulatoren wirken. Pharmakologische Untersuchungen ergaben, dass die erregende Wirkung von GABA durch einen G-Protein-gekoppelten GABAB-Rezeptor vermittelt wurde. Messungen der durch SDN-Stimulierung induzierten Flüssigkeits- und Proteinsekretionsraten zeigten, dass beide Parameter in Anwesenheit von GABA verstärkt waren. Dies ließ auf eine verstärkte serotonerge Neurotransmission schließen, da nur 5-HT die Bildung eines Protein-haltigen Speichels verursacht.
Immuncytochemische Untersuchungen zeigten, dass die Drüsen tyraminerge und octopaminerge Innervation empfangen. Weiterhin wurde der erste charakterisierte TA-Rezeptor (PeaTYR1) der Schabe auf einem paarigen, lateral zur Drüse ziehenden Nerv markiert, der auch tyraminerge Fasern enthielt.
Die vorliegende Arbeit trug zum Verständnis der komplexen Funktionsweise der Speicheldrüse der Schabe bei und erweiterte das lückenhafte Wissen über die neuronale Kontrolle exokriner Drüsen in Insekten. / The cockroach Periplaneta americana has acinar type salivary glands. The secretory acini consist of P-cells, responsible for electrolyte and water secretion and C-cells that secrete protein into the saliva. Salivation is controlled by the dopaminergic and GABAergic salivary neurons SN1 and SN2, and by several smaller serotonergic neurons. Dopamine (DA) and serotonin (5-HT) induce the secretion of a NaCl-rich saliva. The physiological role of the GABAergic innervation was unknown. Furthermore, the cellular actions of the biogenic amines DA and 5-HT were poorly understood. Based on studies on other insect salivary glands a role for octopamine (OA) and tyramine (TA) acting as neuromodulators was suggested.
In this study, intracellular recordings of the basolateral membrane potential of acinar cells were performed to examine direct and modulating actions of the biogenic amines DA, 5-HT, OA, TA and of GABA. A nerve-gland preparation was developed and used to investigate the actions of neuromodulators, namely GABA, OA and TA.
DA and 5-HT induced biphasic membrane potential changes, consisting of an initial hyperpolarization and a transient depolarization. The DA-induced hyperpolarization was mediated by intracellular Ca2+-release and subsequent opening of basolateral Ca2+-dependent K+-channels. The DA- and 5-HT-induced depolarization was dependent on the presence of extracellular Na+ and the activity of a basolateral Na+-K+-2Cl--cotransporter.
Electrical stimulation of the salivary duct nerve (SDN) by means of a suction electrode induced membrane potential changes with the same kinetics as those induced by bath application of DA and 5-HT. These results suggested that electrical nerve stimulation is a adequate method to investigate presynaptic effects of neuromodulators.
GABA, OA and TA affected neither the resting membrane potential of the acinar cells, nor the DA- or 5 HT- induced potential changes. When GABA was applied during SDN-stimulation, it enhanced the amplitudes of the membrane potential changes of the acinar cells as well as fluid- and protein secretion rates of the glands. Pharmacological experiments revealed that the excitatory action of GABA in the gland complex is mediated by a metabotropic GABA receptor (GABAB-type).
OA and TA enhanced the membrane potential changes of the acinar cells when these were induced by SDN-stimulation, suggesting presynaptic excitatory roles for both amines in the gland complex. Immunocytochemistry revealed rich innervation of the salivary glands with octopamine- immunoreactive fibers that were also stained by the tyramine-antibody, and with tyramine-immunoreactive fibers lacking octopamine-immunoreactivity. Since the tyramine receptor PeaTYR1 is expressed in the salivary gland complex, its distribution was investigated by using a specific antibody. Immunoreactivity was detected in a paired nerve of unknown root. This nerve innervated only few acini lying in the periphery of the gland complex and contained tyraminergic fibers.
This study extends our knowledge about the complex neuronal control and function of insect salivary glands.

Reduced order multi-legged mathematical model of cockroach locomotion on inclinesPeterson, Delvin E. 11 July 2011 (has links)
While the locomotion performance of legged robots over flat terrain or known obstacles has improved over the past few decades, they have yet to equal the performance of their animal counterparts over variable terrain. This work analyzes a multi-legged reduced order model of cockroach locomotion on variable slopes which will be used as an inspiration for a future sprawled posture legged robot. The cockroach is modeled as a point mass, and each leg of the cockroach is modeled as a massless, tangentially rigid, linearly elastic spring attached at the center of mass. All of the springs are actuated to allow changes in energy to the system. This is accomplished by varying the force free length of each leg in a feed-forward manner without reliance on feedback to change the actuation scheme. Fixed points of the model are found using a numerical solver that varies the velocity and phase shift parameters while leaving all other parameters at fixed values selected to match true cockroach motion. Each fixed point is checked for stability and robustness representing how effective the model is at staying on the predetermined gait, and transport cost as a measure of how efficient this gait is. Stable and robust fixed points were successfully found for the range of heading angles encompassing those of representative cockroach motion at each slope. Cockroaches may select the gait
used based on stability or efficiency. Thus, additional fixed points were found in combination with a search routine that varies the leg actuation parameters in order to optimize either stability or metabolic efficiency, gaining insights into why cockroaches use the gaits that they do. Optimized fixed points were found based on four different leg functional combination families depending on whether each leg pushes or pulls. Optimized fixed point gaits exist for every incline slope studied between level ground and vertical slopes, at a range of initial heading angles that encompass those typically used by cockroaches. The selected gaits using both a stability based and an efficiency based optimization on the modeled cockroach are very similar. Both are also similar to gaits used by real cockroaches. The forces generated by the model are qualitatively similar

Bases para a implementação de um programa de manejo da resistência de Blattella germanica (L.) a inseticidas foram coletadas no presente trabalho. Técnicas de bioensaio de aplicação tópica e de contato tarsal foram comparadas inicialmente para a detecção da resistência a deltametrina e clorpirifós. O bioensaio de aplicação tópica possibilitou uma melhor discriminação entre a linhagem suscetível e as populações de campo de B. germanica testadas. Resultados do monitoramento da suscetibilidade de B. germanica a inseticidas evidenciaram a presença de variabilidade genética que confere resistência a deltametrina, clorpirifós e fipronil em populações coletadas em alguns estabelecimentos comerciais dos Estados de São Paulo e Rio de Janeiro. As razões de resistência encontradas para deltametrina e clorpirifós foram de aproximadamente 43 e 6 vezes, respectivamente. A resistência de B. germanica a deltametrina foi instável, ou seja, houve uma redução significativa na freqüência de resistência na ausência de pressão de seleção. Estudos de relações de resistência cruzada revelaram uma baixa intensidade de resistência cruzada (aproximadamente 2 vezes) entre fipronil e os inseticidas deltametrina e clorpirifós. E por último, a mistura de deltametrina e clorpirifós foi avaliada para o controle de linhagens de B. germanica resistentes a deltametrina. A mistura proporcionou um melhor controle da linhagem suscetível do que os produtos utilizados isoladamente. No entanto, a discriminação entre as linhagens suscetível e resistente a deltametrina foi evidente com a mistura nas doses recomendadas comercialmente, o que inviabilizaria o emprego desta estratégia no manejo da resistência de B. germanica a deltametrina. / Bases for implementing a resistance management program of Blattella germanica (L.) to insecticides were collected in this study. Initially, we compared the topical and residual tarsal-contact bioassays for detecting resistance to deltamethrin and chlorpyrifos. The topical bioassay gave better discrimination between susceptible and field-collected populations of B. germanica. Results from a survey of the susceptibility of B. germanica to insecticides revealed the presence of genetic variability that confers resistance to deltamethrin, chlorpyrifos and fipronil in field-collected populations of B. germanica from the States of São Paulo and Rio de Janeiro. The resistance ratios to deltamethrin and chlorpyrifos were approximately 43 and 6-fold, respectively. The resistance of B. germanica to deltamethrin was unstable; that is, a significant reduction in the frequency of resistance was detected under absence of selection pressure. Studies on cross-resistance relationships showed a low intensity of cross-resistance (approximately 2-fold) between fipronil and the insecticides deltamethrin and chlorpyrifos. And finally, the viabilility of the use of deltamethrin and chlorpyrifos mixture was evaluated for controlling deltamethrin-resistant strains of B. germanica. The use of mixtures gave better control of the susceptible strain than the use of the products by itself. However, the discrimination between susceptible and resistant strains was clear with the use of mixtures at recommended rates, which impairs the use of this strategy for managing B. germanica resistance to deltamethrin.

Níveis de IgE total foram significantemente mais elevados em crianças de Natal quando comparados aqueles em crianças de Ribeirão Preto (média geométrica 630,9 kU/L, faixa 19,6-63.290 kU/L; e 398,1 kU/L, faixa 35,7-4.803 kU/L, respectivamente). Entre as 150 crianças de Natal, 52(34,6%) apresentaram IgE positiva para D.pteronyssinus; 70(46,6%) para B.tropicalis; 45(30%) para barata; 19(12,6%) para gato; 17(11,3%) para cachorro; e 125(83,3%) para A.lumbricoides. Entre as 54 crianças com asma e/ou rinite de Ribeirão Preto, 41(75,9%) apresentaram IgE positiva para D.pteronyssinus; 34(62,9%) para B.tropicalis; 22(40,7%) para barata; 11(20,3%) para gato; 12(22,2%) para cachorro. Embora estas crianças fossem negativas para parasitas à inclusão no estudo, 22(40,7%) tinham IgE para A.lumbricoides. Anticorpos IgE para A. lumbricoides foram mais elevados entre crianças de Natal, quando comparados a IgE para alérgenos inalantes (p<0,01). Níveis de IgE para D. pteronyssinus entre crianças de Ribeirão Preto foram mais altos que IgE para outros inalantes e A.lumbricoides (p<0,01). Em Natal, a porcentagem de IgE para A. lumbricoides em relação à IgE total foi maior em comparação a IgE para D.pteronyssinus e B.germanica (mediana 0,41%; 0,08%; e 0,04% respectivamente, p<0,01). Em Ribeirão Preto, a porcentagem de IgE para D.pteronyssinus e para B.tropicalis em relação à IgE total foi maiorem comparação a IgE para A.lumbricoides e barata (mediana 9,8%; 0,6%; 0,3%; e 0,2%, respectivamente, p<0,05). Regressão linear revelou que a associação mais forte foi para IgE para A.lumbricoides com IgE total em Natal (R²=0,56; p<0,01); associação significante foi também observada para IgE para ácaros com IgE total em Ribeirão Preto (R2=0,35; p<0,01 para D.pteronyssinus; R2=0,33; p<0,01 para B.tropicalis, respectivamente). Nossos resultados demonstraram que anticorpos IgE para ácaros contribuem fortemente para a IgE total entre crianças com asma e/ou rinite, vivendo em uma área de baixa taxa de infecções parasitárias em nosso meio. Por outro lado, entre crianças vivendo em uma área em que parasitas são encontrados em abundância, infecções parasitárias induzem uma forte resposta IgE policlonal, e anticorpos IgE específicos para parasita, além de ácaros, barata, gato e cachorro representam uma modesta proporção da IgE total. A especificidade desta IgE, e os efeitos a longo prazo desta resposta cedo na vida, permanecem desconhecidos. / Immunoglobulin E (IgE) plays a central role in the pathogenesis of allergic diseases. It is part of the type 2 response, and the cytokines IL-4 and IL-13 are essential for the production of this immunoglobulin isotype. IgE production is also induced by intestinal parasites, particularly helminths. The aim of the present study was to evaluate the contribution of specific IgE to mites, cockroach, cat, dog and parasite Ascaris lumbricoides on total IgE levels among children living in different areas in Brazil. IgE antibodies to mites Dermatophagoides pteronyssinus and Blomia tropicalis; cockroach Blattella germanica; cat; dog; and A. lumbricoides were measured using the ImmunoCAP system and compared to total serum IgE of 150 children 3-6 year-old living in the city of Natal, endemic for intestinal parasites, and 54 children 3-6 years of age with asthma and /or rhinitis, living in Ribeirão Preto. Total IgE levels were significantly higher in children from Natal as compared to those among children in Ribeirão Preto (geometric mean 630,9 kU/L, range 19,6-63.290 kU/L; e 398,1 kU/L, range 35,7-4.803 kU/L, respectively). Although these children were negative for parasites at inclusion in the study, 22(40.7%) had IgE to A.lumbricoides. IgE antibodies to A.lumbricoides were higher among children from Natal, as compared to IgE to inhalant allergens (p<0.01). IgE levels to D.pteronyssinus among children living in Ribeirão Preto were higher than IgE to other inhalants and A.lumbricoides (p<0.01). In Natal, the percentage of IgE to A.lumbricoides in relation to total IgE was higher in comparison to IgE to D.pteronyssinus e B.germanica (median 0.41%, 0.08%, and 0.04% respectively, p<0.01). In Ribeirão Preto, the percentage of IgE to D.pteronyssinus and to B.tropicalis in relation to total IgE was higher in comparison to IgE to A.lumbricoides and cockroach (median 9.8%, 0,6%, 0.3%, and 0.2%, respectively, p <0.05). Linear regression analysis revealed that the strongest association was for IgE to A.lumbricoides with total IgE in Natal (R²=0.56, p<0.01); significantassociation was also observed for IgE to mites with total IgE in Ribeirão Preto (R2=0.35, p<0.01 for D. pteronyssinus, R2=0.33, p<0.01 for B.tropicalis, respectively). Our results demonstrated that IgE antibodies to mites contribute strongly to total IgE among children with asthma and /or rhinitis, living in an area of low parasite infection rates in our country. On the contrary, among children living in an area where parasites are found in abundance, parasitic infections induce a strong polyclonal IgE response, and IgE antibodies specific for parasite, and also for mites, cockroaches, cat and dog represent a modest proportion of total IgE. The specificity of these IgE antibodies and the implications of this response occurring early in life remain unknown.

Reproduction and Metabolic Responses to Acute and Chronic Hypoxia in Ovoviviparous Blaberid Cockroaches, with a Focus on Blaptica dubiaMallery, Christopher Sean 08 1900 (has links)
The major components of the tracheal system of insects are an extension of the exoskeleton, and the size of the exoskeleton is fixed in the adult stage, so any increase in metabolic demand that may accompany reproduction must be met by a relatively unaltered tracheal system that the female set in place at ecdysis, when entering adulthood. Acute hypoxia tends to elicit an increase in ventilation in insects, and here, I observe increased interburst VCO2 release, and a tendency towards a more continuous gas exchange pattern being preferred over discontinuous gas exchange when Blaptica dubia and Eublaberus posticus are exposed to a descending regime of hypoxia. Additionally, higher temperatures appear to increase sensitivity to hypoxia in these species, an expected result because both species, like most ectothermic animals, display a Q10 effect, increasing metabolic rates as temperature increases. The reproductive mode of B. dubia is considered to be lecithotrophic pseudoviviparity (or type A ovoviviparity), and by the time the embryos are born, they have more than doubles in volume from the time of oviposition. This gain is apparent in the wet mass of the embryo, with no change occurring in dry mass. The egg mass that can be attributed to water begins at 39% at oviposition and increased to nearly 82% at hatching. The metabolic rates of females and embryos increase as embryonic development progresses, but bomb calorimetry reveals that energy content of the embryos does not change. It is possible that these embryos gain some nutrition from the mother during embryonic development, but direct evidence remains to be demonstrated. Blaptica dubia and Eublaberus posticusare both blaberid species that display the same reproductive mode, pseudoviviparity, with incubation occurring in a brood sac. Comparisons between the reproductive traits of B. dubia and E. posticus reveal that the two species have similar reproductive periods, interbirth periods, and clutch sizes to one another, and that both have reproductive cycles and incubation times intermediate to oviparous species and the one species of cockroach that some consider to be truly viviparous (Diploptera punctata). However, whereas adult female E. posticus are larger than female B. dubia (E. posticus non-gravid female mass: 2.91 +/- 0.42 g, 20; B. dubia non-gravid female mass: 2.60 +/- 0.40 g, 20), the offspring of B. dubia are larger than those of E. posticus at the time of birth (B. dubia neonate mass: 24.70 +/- 4.01 mg, 19; E. posticus neonate mass: 22.40 +/- 1.36 mg, 19). Both gravid and non-gravid female B. dubia respond similarly to acute hypoxia exposure, increasing mean total VCO2. However, the reproductive state does not appear to exacerbate, nor dull, the acute response to hypoxia. Gravid and non-gravid female B. dubia were exposed to chronic hypoxia for 30 days and 45 days. Relatively high mortality was observed in nearly all chronic hypoxic treatment groups as compared to controls at 21 kPa O2. Comparing treatment groups to controls maintained at 21 kPa O2, embryo mass was not decreased, nor was embryo VCO2 at day 30. Adult female B. dubia metabolic rates did not show a consistent change in response to chronic hypoxia, but decreased metabolic rate was observed in the non-gravid B. dubia exposed to chronic 4 kPa O2, an observation that is consistent with past studies in insects. Survival rates were lower for both gravid and non-gravid females in 4, 8, and 12 kPa O2, as well as in 15 kPa O2 in gravid females, as compared to 21 kPa O2. Gravid females experiences a decreased survival rate compared with non-gravid females at 12 and 15 kPa O2, but gravid and non-gravid females had similarly low survival rates at 4 and 8 kPa O2. This difference in survival rates suggests there is a cost associated with carrying an ootheca in a brood sac, resulting in an increased sensitivity to hypoxia, at least over a long period of time.

The class Kinetoplastea contains free-living and parasitic species. One of the most dominant group within the class is the order Trypanosomatida which includes obligate parasites (Trypanosoma, Leishmania) infecting a wide range of hosts. Some species are serious pathogens of humans and domestic animals and cause considerable losses. However, the majority of trypanosomatids belongs to monoxenous parasites of insect which are usually harmless to their hosts. Monoxenous trypanosomatids predominantly infect Hemiptera and Diptera. This diploma thesis is focused on the detection of monoxenous trypanosomatids in cockroaches captured in the Czech Republic and cockroaches from different breedings. Cockroaches are very suitable mechanical vectors of many different pathogens (including parasites) and are significant health threat for humans and animals. First trypanosomatids in cockroaches were documented at the beginning of the 20th century, but there is no study focused on this topic specifically. Another aim of this thesis is morphological and ultramicroscopic analysis and the study of the host specificity of the recently described species Herpetomonas tarakana, isolated from a cockroach.
