Implication des macrophages M1/M2 dans les pathologies vasculaires et valvulaires humaines / M1/M2 macrophages implication in human vascular and valvular diseases

Les maladies cardiovasculaires, conséquence de l’athérosclérose, sont la première cause de morbi-mortalité dans le monde et voient leur incidence et sévérité augmenter avec l’expansion de leurs principaux facteurs de risque, tels que l’âge, l’obésité et le diabète. La sténose valvulaire aortique, valvulopathie la plus fréquemment rencontrée dans les pays occidentaux essentiellement chez le sujet vieillissant, partage de fortes similitudes avec l’athérosclérose vasculaire. En effet, les plaques athéroscléreuses et les lésions valvulaires sont le siège de processus d’inflammation, d’angiogenèse, de fibrose et de calcification. Les macrophages, issus de la différenciation tissulaire des monocytes infiltrés, jouent un rôle clé dans l’apparition des lésions athéroscléreuses vasculaires et leur devenir. Leur rôle dans l’état inflammatoire des lésions est aujourd’hui bien établi avec de récentes publications qui font état des propriétés plastiques des macrophages, selon leur microenvironnement. Deux principaux sous types de macrophages ont été décrits dans les plaques athéroscléreuses, les macrophages M1 dit « classiques » et M2 dits « alternatifs ». Leur rôle respectif dans la thrombogénécité, la protéolyse et l’angiogenèse, processus impliqués dans l’instabilité de la plaque, ont été moins étudiés. En revanche, les macrophages sont peu décrits dans la valve, à l’inverse des cellules interstitielles de valves (VIC), qui sont cruciales pour le maintien de l’homéostasie et la fonction valvulaire et sont impliquées dans la fibrose et la rigidité des feuillets valvulaires. Mon travail de thèse a pour objectif d’étudier les rôles des macrophages M1/M2 dans les pathologies vasculaires et valvulaires chez l’homme. Nous nous sommes focalisés sur leurs rôles dans l’instabilité de la plaque athéroscléreuse (processus de coagulation et de remodelage vasculaire) et dans la fibrose valvulaire ainsi que sur leur modulation phénotypique par d’autres types cellulaires présents dans les lésions, les polynucléaires neutrophiles dans la plaque ou les VIC dans la valve.Nos résultats suggèrent que les macrophages M1 et M2 pourraient moduler différemment des processus physiopathologiques majeurs de l'athérosclérose. Par ailleurs, les macrophages M1 de patients diabétiques présentent un phénotype délétère qui pourrait expliquer la plus grande vulnérabilité des plaques d’athérosclérose observée chez ces sujets. Concernant la pathologie valvulaire, après avoir caractérisé par analyse histologique les M1/M2 dans les valves aortiques humaines, nous avons montré que les macrophages M1 sont impliqués dans la progression de la fibrose via la modulation de leur répertoire sécrétoire par les VIC.Cette thèse apporte de nouveaux indices sur les processus physiopathologiques impliqués dans les maladies vasculaires et valvulaires. Elle met l’accent sur le rôle délétère des macrophages M1 chez les sujets diabétiques en pathologie vasculaire et identifie également une fonction jusque-là méconnue des M1 dans la progression de la fibrose, associée au « cross-talk » avec les VIC. Il conviendra par la suite d’identifier les mécanismes moléculaires sous-jacents à ces intéractions, ce qui devrait permettre d’envisager de nouvelles voies thérapeutiques visant à moduler l’effet de ce sous-type cellulaire dans ces pathologies. / Cardiovascular disease, as a result of atherosclerosis, are the main cause of morbidity and mortality in the world and see their incidence and severity increase with the expansion of their major risk factors, such as age, obesity and diabetes. Aortic valve stenosis, valve disease most frequently encountered in Western countries mainly in the old subject, shares strong similarities with vascular atherosclerosis. Indeed, atherosclerotic plaques and valvular lesions are the site of inflammation, angiogenesis, fibrosis and calcification processes. Macrophages, from monocytes infiltrated tissue differentiation, play a key role in the development of vascular atherosclerotic lesions and their future. Their role in the inflammatory state of the lesions is now well established with recent publications that report on plastic properties of macrophages, according to their microenvironment. Two major subtypes of macrophages have been described in the atherosclerotic plaques, classically (M1) or alternatively (M2) activated macrophages. Their respective role in thrombogenicity, proteolysis and angiogenesis processes involved in plaque instability, have been less studied. In contrast, macrophages are not disclosed in the valve, compared to the valvular interstitial cells (VIC), which are crucial for the maintenance of homeostasis and the valvular function and are involved in the fibrosis and rigidity of the valvular leaflets. My thesis aims to study the roles of macrophages M1/M2 in vascular and valvular pathologies in humans. We focused on their roles in the instability of atherosclerotic plaque (haemostatic or clotting process and vascular remodeling) and valvular fibrosis and their phenotypic modulation by other cell types present in the lesions, neutrophils (PNN) in the plaque or VIC in the valve.Our results suggest that the M1 and M2 macrophages may differently modulate major pathophysiological processes of atherosclerosis. In addition, M1 macrophages from diabetic patients have a deleterious phenotype that could explain the increased vulnerability of atherosclerotic plaques observed in these subjects. About valvular pathology, after characterized histologically M1/M2 in human aortic valves, we have shown that the M1 macrophages are involved in the progression of fibrosis through the modulation of their secretory repertoire by VIC.This work provides new clues about the pathophysiological processes involved in vascular and valvular diseases. It focuses on the deleterious role of M1 macrophages in diabetic subjects in vascular pathology and also identifies an unknown function of M1 in the progression of fibrosis associated with "cross-talk" with VIC. It will be necessary later to identify the molecular mechanisms underlying these interactions, which is expected to consider new therapeutic approaches to modulate the effect of this cell subtype in these diseases.

La present tesi vol explicar la implantació de la Reforma Catòlica en una sèrie de parròquies rurals dels bisbats de Girona (valls de Ridaura, Bas, Hostoles i Amer) i Vic (El Collsacabra i les valls de Susqueda i Sau), entre 1587 i 1800, des dels bisbes posttrentins Jaume Caçador i Pedro Jaime als il·lustrats Tomàs de Lorenzana i Francisco de Veyan. La documentació principal són les sèries de les visites pastorals conservades a l'Arxiu Diocesà de Girona i l'Arxiu Episcopal de Vic; paral·lelament, s'ha reforçat amb documentació parroquial (llibres sagramentals, consuetes, llibres d'obra i confraries), protocols notarials (notaries de Rupit, Sant Feliu de Pallerols, El Mallol i Amer) i impresos episcopals. Tots ells han servit per demostrar la lentitud en la implantació del programa tridentí, que s'assoleix, de fet, amb força retard (ben entrat el segle XVIII). / The present thesis tries to explain the implantation of the Catholic Reform in a few rural parish churches in the bishoprics of Girona (Valleys of Ridaura, Bas, Hostoles and Amer) and Vic (El Collsacabra and Valleys of Susqueda and Sau), from 1587 to 1800, from the post-Trent bishops Jaume Caçador and Pedro Jaime to Illustrated Tomàs de Lorenzana and Francisco de Veyan. The main sources of information are the pastoral bishop visits. These series are well preserved in the Arxiu Diocesà de Girona and the Arxiu Episcopal de Vic. We also have added some other documents: parish registers, notary books (from Rupit, Sant Feliu de Pallerols, El Mallol i Amer) and bishop printed material. We have contrasted the orders from the bishops visits with the decrees from the Council of Trent, the Constitutions of the Tarraconense Province and the Synodal Constitutions of Girona and Vic, and the artistic, architectural and archaeological evidences. All of them have shown the slow pace and the delay of the enterprise.

A systematic assessment of fault seal risk to hydrocarbon exploration in the Penola Trough, Otway Basin, South Australia.

A new depth-based method of seismic imaging is used to provide insights into the 3D structural geometry of faults, and to facilitate a detailed structural interpretation of the Penola Trough, Otway Basin, South Australia. The structural interpretation is used to assess fault kinematics through geological time and to evaluate across-fault juxtaposition, shale gouge and fault reactivation potential for three selected traps (Zema, Pyrus and Ladbroke Grove) thus providing a full and systematic assessment of fault seal risk for the area. Paper 1 demonstrates how a depth-conversion method was applied to two-way time seismic data in order to redisplay the seismic in a form more closely representative of true depth, here termed ‘pseudo-depth’. Some apparently listric faults in two-way time are demonstrated to be planar and easily distinguishable from genuine listric faults on pseudo-depth sections. The insights into fault geometry provided by pseudo-depth sections have had a significant impact on the new structural interpretation of the area. Paper 2 presents the new 3D structural interpretation of the area. The geometry of faulting is complex and reflects variable stress regimes throughout structural development and the strong influence of pre-existing basement fabrics. Some basement-rooted faults show evidence of continual reactivation throughout their structural history up to very recent times. Structural analysis of all the live and breached traps of the area demonstrate that traps associated with a basement rooted bounding fault host breached or partially breached accumulations, whereas non-basement rooted faults are associated with live hydrocarbon columns. Papers 3 and 4 demonstrate that for all the traps analysed (Zema, Pyrus and Ladbroke Grove), initial in-place seal integrity was good. The initial seal integrity was provided by a combination of both favourable across fault juxtaposition (Ladbroke Grove) and/or sufficiently well developed shale gouge over potential leaky sand on sand juxtaposition windows to retain significant hydrocarbon columns (Zema, Pyrus). The palaeocolumns observed at Zema and Pyrus indicate that there has been subsequent post-charge breach of seal integrity of these traps while Ladbroke Grove retains a live hydrocarbon column. Evidence of open, permeable fracture networks within the Zema Fault Zone suggest that it is likely to have recently reactivated, thus breaching the original hydrocarbon column. Analysis of the in-situ stress tensor and fault geometry demonstrates that most of the bounding faults to the selected traps are at or near optimal orientations for reactivation in the in-situ stress tensor. The main exception being the Ladbroke Grove Fault which has a NW-SE trending segment (associated with a relatively high risk of fault reactivation and possible leakage at the surface) and an E-W trending segment (associated with a relatively low risk of fault reactivation and a present day live column). Signage & sense of place : preserving the experience of historic illuminated signage

The purpose of this thesis is to raise awareness about the contribution historic signs make to the experience of a place. Illuminated signage has played a key role in the development of the American landscape. The relationship between illuminated signage and sense of place is complex. Because of the ephemeral nature of signs and the public perception of them as advertising tools, the preservation community frequently overlooks signs. By examining three case studies, this thesis presents three different approaches to the preservation of illuminated signage. The first case study is the 2002 New Mexico Route 66 Neon Sign Restoration Project. This case uses Route 66 to examine the preservation of original material in original location. The second case study is the 2002 Amendment to the New York City Zoning Regulations, which mandates signage saturation in Times Square. This case examines the preservation of the experience of Times Square by protecting the historical use of innovative signage. The third case study is the 1996 installation of restored illuminated signs by the Neon Museum in Las Vegas at the Fremont Street Experience. This case examines the collections approach to preserving original Las Vegas illuminated signs in an outdoor museum setting as public works of art. While these three case studies evaluate iconic locations, the lessons are broadly applicable. The preservation approaches outlined here illustrate that every situation is unique and requires a full analysis of the context of the sign. Preservationists should evaluate signage within their local communities by examining the artistry and materiality of the signs. However, it is equally important to evaluate the overall community context of the signs. In order to preserve the experience of historic illuminated signage, it is imperative that preservationists and the general public understand that there are numerous approaches to safeguarding these works and that the time to take action is now. / text
