Towards a macroinvertebrate sampling protocol for monitoring water quality of wetlands in South Africa.

The degradation of wetlands and loss of their associated ecosystem services is widely recognised in South Africa, however, at present there is no standard method of biologically assessing wetland health in this country. Internationally, particularly in the U.S.A and Australia, wetland bioassessment techniques using macroinvertebrates are well established. A number of these wetland bioassessment protocols have been derived from local river biomonitoring techniques, as there is a belief that river and wetland ecology and macroinvertebrate assemblages at family level are similar. However, some authors consider wetland macroinvertebrate assemblages and ecological processes to differ greatly from those found in rivers, and believe that such techniques are not transferable. South Africa has a well established macroinvertebrate biomonitoring protocol for rivers called SASS5 (South African Scoring System Version 5). This study is a preliminary investigation into the extent to which the SASS5 scoring system is applicable to the assessment of nutrient enriched wetland water quality. Macroinvertebrates are particularly suitable as biomonitoring tools: they respond to a variety of stressors, have life cycles that allow for integrated responses to episodic pollution, and are relatively easy to identify to family level. When selecting wetlands for the development of a biomonitoring protocol, wetlands should all be of the same; classification (Le. palustrine), geomorphological and climate setting, hydrological regime and dominant vegetation class. Sampling was restricted to sedge-dominated palustrine wetlands in the midlands of KwaZulu-Natal, with similar hydro-geomorphological settings. Due to wetlands and rivers having different biotopes (e.g. no riffles present in wetlands), the SASS5 sampling protocol could not be used, thus a pilot investigation was undertaken to derive a suitable sampling technique for \ collecting a representative and diagnostic sample of aquatic macroinvertebrates from a wetland. This technique was developed based on published methods. Both sweep net and activity trap sampling were conducted, and each evaluated for their effectiveness at macroinvertebrate collection. Sweep net sampling was tested over a range of sweep intensities (2-6 sweeps), and activity traps were placed at four different depths: at the water surface, just below the surface, 0.10.15m below surface and on the substrate. A total of 32 taxa identified to family level were identified in the samples. Taxon diversity and composition did not differ in the activity traps placed at the four depth locations. Taxon diversity did not differ significantly between different sweep intensities; however there was a significant difference in taxon composition between the different sweep intensities and between activity trap and sweep net samples (p<0.05). Sixty-eight percent of taxa appeared more frequently in sweep net sampling compared to activity trap sampling. Six taxa were found exclusively in sweep net samples, and two taxa were recorded exclusively in activity traps. There was no trend in either method collecting more or missing any unique trophic group. In conclusion, activity traps are not required to supplement sweep net data, and a technique using a sweep net with a sweep intensity of five would be suitable to collect a representative sample of wetland macroinvertebrates. Using the derived technique, four reference and three wetlands impacted by dairy effluent were sampled. Six macroinvertebrate samples were collected from each of the seven wetland, together with data for selected physico-chemical variables, macrohabitat condition, biotope suitability and organism detectability. For each sample, the macroinvertebrates were identified and assigned a predetermined SASS5 tolerance score between 1 and 15, with higher scores indicating increased sensitivity to poor water quality. 11 A total of 39 taxa, identified to family level, were collected during sampling. SASS5 scores ranged from 15-82. Five of the wetlands had mean SASS5 scores of between 46 and 59. Five of the wetlands had an intra-wetland SASS5 score range greater than 30. ASPT values ranged from 3.3 to 5.5, and few high scoring (~8) taxa were collected. There was no significant difference in SASS5 scores between samples collected above, at and downstream of an effluent discharge point within the same impacted wetland. SASS5 scores for reference wetlands were also not significantly higher than those recorded for impacted wetlands. Comparison of ranked SASS5 scores and environmental data did suggest a relationship between the variables, but was not significant. Based on the SASS5 score water quality guidelines, all sampled wetlands were considered to have impacted water quality; however, this was not supported by the macrohabitat and physico-chemical results. Possible reasons for the low SASS5 scores include: the lack of 'stones in/out current' biotopes in wetlands, lower levels of dissolved oxygen present compared to rivers, and the limited detectability of organisms due to large amounts of substrate in the samples. A wetland adaptation of SASS5 would require the reassignment of modified scores to certain taxa based on their distribution in wetlands of varying water quality. The SASS5 score level of 100 and the ASPT value of 6 (as specified in the SASS5 score water quality guidelines) were found to be inappropriate for wetlands. It is suggested that, either the range of taxa tolerance scores be increased (1 to >15), or the score level of 100 be lowered. The ASPT value should also be reduced. Although SASS5 appears unsuitable for assessing wetlands, variations in taxon composition between sampled wetlands, identified through CA analysis, suggests that macroinvertebrates are responsive to changes in wetland condition, and thus have potential as indicators of wetland water quality. Nine taxa responsive to the presence of nitrogen have been identified as being potentially good indicators. iii Further research should focus on the testing of SASS5 throughout the year, in a range of wetland types, and in wetlands moderately to severely impacted by pollutants other than dairy effluent. The cumulative effect of wetland degradation on water quality at a landscape scale.

Wetlands have been described as “one of the most globally threatened and important ecosystems”, with most wetlands providing a variety of important ecosystem services, including water quality enhancement. Despite their importance the loss of wetlands is more rapid than that of any other ecosystem, which is of particular concern in South Africa, where many communities are directly and indirectly dependent on wetlands for survival. Two useful wetland assessment tools are currently used in South Africa, but a system that assesses the extent to which wetland ecosystem services (water quality enhancement in particular) are lost as a result of wetland degradation in a landscape context does not currently exist. This study therefore aims to develop a method to determine the cumulative effect of wetland degradation on water quality, which involves the exploration and integration of a number of issues, including land-cover and its effects on water quality, wetland health and its influence on the provision of ecosystem services such as water quality enhancement, and the spatial configuration of wetlands in a landscape, and its effect on water quality at a landscape scale. The method that has been developed is applied to a case study that comprises a quaternary catchment of the upper reaches of the Goukou River Wetlands in the Western Cape of South Africa. Prioritisation criteria are also explored in a series of scenarios, and the criterion and rehabilitation method that gives the best outcome in terms of water quality enhancement is applied to the case study catchment. The workings of the method are scrutinized and benefits and limitations are subsequently highlighted. An important benefit of the methodology is that many previously inadequately explored issues are integrated into a single tool that allows for prioritisation of wetlands for rehabilitation and conservation. This was achieved with South African contexts in mind. Limitations include poor responses by potential questionnaire respondents, while the scope of the study limits the inclusion of detailed aspects which would have further enhanced the accuracy of the tool and of the level of water quality enhancement explored. The methodology that is developed in this research has also not been applied to catchments with good long term water quality data in order to improve its validity. Recommendations for future research are made, which include possible refinement of the system by accounting for factors not included in the current methodology, validation of the system by applying it to a catchment with good water quality data, and the creation of software to make the system easier to use. / Thesis (M.Sc.)-University of KwaZulu-Natal, Westville, 2009.

An investigation of the seasonal and spatial occurrence of coliform bacteria in a distribution system

Bacteriological data from 1980 to 1991 were reviewed to determine whether coliform bacteria occurred seasonally and spatially within a midwestern city's distribution system. Coliform bacteria are used as microbiological indicator organisms to determine if a public water supply is safe for consumption. The public water_ distribution system examined had at least a twelve year history of the presence of coliform bacteria. Previous investigations have described the occurrence of the coliform bacteria as sporadic because there were no apparent patterns to their presence. An analysis of bacteriological data has not previously been performed to specifically detect seasonal and spatial occurrences of coliform bacteria.This study attempted to determine if seasonal or spatial patterns of coliform occurrences exist within the in the dominant coliform species. Data indicate that the highest percentage of coliform positive samples occurred in the summer, followed by fall, then winter, with spring having the lowest percentage of coliform positive samples. While Enterobacter cloacae was the dominant coliform species during the spring, summer and fall, Klebsiella oxytoca was the dominant coliform during the winter. Coliform occurrence throughout the distribution system was variable among the 43 sample sites. The percentage of positive samples from the various sites ranged from 0% to 10.5%. The five sites with the highest percentage of coliform positive samples were at the extremities of the distribution system. E. cloacae was dominant at 88% of the sites. K. oxytoca was dominant at 9% sites, which typically had a low percentage of coliform positive samples. / Department of Biology

Role of biological monitoring in water quality assessment and a case study on the Crocodile River, Eastern Transvaal

M.Sc. (Zoology) / National water quality monitoring in South Africa has in the past mainly focused on measuring physical and chemical variables. However, it is increasingly realised that measuring physical and chemical variables on their own cannot provide an accurate account of the general "health" of an aquatic ecosystem. Biological communities, on the other hand, are accurate indicators of overall environmental conditions. Water quality management must, therefore, rely on comparative data for both chemical composition and biological effects. In fact, it appears as if biological monitoring (biomonitoring) is worldwide becoming a primary tool in assessing environmental condition and verifying compliance with effluent discharge. This study classified different biomonitoring approaches and techniques under bioassessment (referring to the field oriented biomonitoring protocols which make use of biotic indices to assess water quality); bioassays (toxicity tests which is usually laboratory-based); behavioural bioassays (including aspects such as early warning systems, and preference and avoidance studies); bacteriological studies (the monitoring of certain microbes to allow the detection of faecal contamination); measurement of bioaccumulation (referring to the methods by which the uptake and retention of chemicals in the body of an organism can be monitored); and fish pathology (fish health studies dealing with the causes, processes and effects of disease). Habitat assessment and evaluation was identified as an essential part of any biosurvey. There can be little uncertainty about the mutual dependence of habitat quality, biological health and chemical characteristics of water in the environment. Relative habitat condition, as the principal determinant of attainable biological potential, should set the context for interpreting the results of a biosurvey and can be used as a general predictor of biological condition. Chemistry can further help to explain and characterise certain impacts. The Crocodile River, Eastern Transvaal, was selected for conducting a case study. The SASS2 rapid bioassessment protocol, as well as a habitat quality index (Hal) was used during five consecutive biosurveys. From the results obtained in this study, it appears as if biomonitoring can be used to good effect in overall environmental assessment. The SASS2 index appeared to be both a robust and sensitive indicator of environmental condition. Application of the SASS2 technique on a regional or even national basis should be feasible with regard to simplicity and practicality. It is also cheaper and less labour intensive than comprehensive chemical monitoring. However, bioassessments should not replace but rather compliment chemical and physical monitoring.

Development of a passive, in situ, integrative sampler for monitoring biotoxins in aquatic environments

(Chemistry) / This thesis focuses on functionalising the silicone membrane by filling it with synthesised γ-Fe2O3 nanoparticles for the purpose of using it as a passive sampling device for monitoring microcystins and cylindrospermopsin in aquatic environments. The identity and purity of the synthesised nanoparticles was verified by X-ray diffraction (XRD), transmission electron microscopy (TEM), scanning electron microscope (SEM), surface zeta potential analyser and Brunauer-Emmet-Teller (BET). There was no chemical uptake of microcystin compounds into the silicone membrane without a suspension of maghemite nanoparticles. But, a linear uptake of microcystin compounds was observed into the functionalised silicone membrane passive sampler throughout the exposure period. These findings suggest that the silicone membrane filled with a suspension of synthesised γ-Fe2O3 nanoparticles can be an efficient, green and less costly instrument useful in estimating biotoxin concentrations in South African waters and the world at large.

Bioaccumulation of metals and the general health of fish from the Vaal Dam and Vaal River Barrage

M.Sc. / The water quality in the Vaal River system has deteriorated due to increasing industrial, mining and agricultural activities in the Vaal Dam and Vaal River Barrage catchment areas. This study was undertaken to determine the water quality in the Vaal Dam, and three localities within the Vaal River Barrage reservoir (i.e. the Riviera Aquatic Club, Abrahamsrust Pleasure Resort and at the Barrage structure). The objectives of this study, was to determine the fish population species composition, evaluate the general health of the selected fish species and obtain site specific metal bioaccumulation data for each locality. Suggestions and proposals for future monitoring and management of these waterbodies were also made. Metereological information on the Vaal River Barrage area was supplied by Rand Water, as well as physical and chemical data of fixed localities, sampled fortnightly (January 1997 to September 1998). In addition, water samples were collected at the Vaal Dam and the three Vaal River Barrage localities mentioned above, during February, May, August and November 1997. These samples were analysed by the Chemistry Department of the Rand Afrikaans University in accordance with standard methods. The chemical and physical characteristics and metal concentrations (aluminium, cadmium, calcium, copper, iron, magnesium, manganese, nickel, strontium and zinc) of these separate sets of data were then compared with Rand Water's Raw Water Quality Criteria. The aluminium, ammonia, chromium, copper, dissolved organic carbon, fluoride, iron, lead, manganese, nitrate, sulphate, total hardness and turbidity variables did not comply with the recommended guideline values. No guideline levels for strontium are available, however. Sedimentation, turbidity, salination and eutrophication within the Vaal River system play an important role in the bioaccumulation of metals by benthic organisms. Metals that adsorb to sediment particles, through various processes, are generally more persistent and less mobile than those in the overlying water and pose a severe long-lasting threat to the aquatic environment. Generally, the highest metal concentrations (AI, Cd, Cu, Fe, Mn, Ni, Pb, Sr and Zn) in the sediment samples collected at the four localities and seasons, were found in the finer sediment classes (class D). The Riviera Aquatic Club locality had the highest overall concentrations of the metals analysed. The adult fish communities within the Vaal Dam and Vaal River Barrage were assessed, as it is believed that the fish community structure is indicative of environmental conditions. Adult Labeo capensis, Labeo umbratus, Cyprinus carpio, Clarias gariepinus, Barbus aeneus and Micropterus sa/moides, were collected (using gill nets) to determine the adult fish community composition and age. When compared with a study conducted by Mulder (1971), it is clear that Labeo capensis has remained the most abundant species within the system, but that Barbus aeneus has now been replaced by Labeo umbratus as the second most abundant species. Furthermore, age determination tables (Mulder, 1971) were used to classify the fish population between the ages of 2 and 7 years. This indicates a healthy, dynamic community structure where recruitment is taking place. The Health Assessment Index (HAl) was used to determine the general health of the two fish species selected for metal analysis (i.e. Labeo capensis and Labeo umbratus). Except for moderate to severe gill damage recorded, no damage to external variables, were of concern. However, both Labeo capensis and Labeo umbratus had high percentages (89% and 93%, respecii tively) of abnormal livers. Using the Plascon Colour Chart (Watson, personal communication), these livers were classified as ranging from tan to "coffee with cream" in colour. High haematocrit and plasma protein levels were observed and could be due to environmental stress, while the White Blood Cell Counts were within normal ranges. Generally speaking, only mild to moderate parasite infections were observed and probably do not pose a serious health threat to the fish host. Total HAl values for Labeo capensis indicate that this this fish population was in relatively good health, but too few Labeo umbratus individuals were collected to make accurate assumptions about this population. Metal analysis (AI, Cd, Cu, Fe, Mn, Ni, Pb, Sr and Zn) was carried out on the tissues and organs (skin, muscle, gill filaments, gill arches, liver, hindgut content, gonads and vertebrae) of Labeo capensis and Labeo umbratus, collected during the study period. Only aluminium, iron and strontium are discussed. Aluminium concentrations were relatively high at all of the localities, but as pH values were mostly alkaline, aluminium is rendered biologically unavailable and as for iron, was mostly accumulated in the non-edible portion and viscera of the fish. Strontium accumulates in bony tissues due to its similarity with calcium, but is not essential for bone formation. The human health risks of these metals in the edible portion of the fish still have to be refined and communicated to the general public of South Africa. In conclusion, this baseline study has determined the metal concentrations in the aquatic environment (due to anthropogenic activities) that may be accumulated by fish and may affect the adult fish community structures and their general health.
