Influência do ranelato de estrôncio na reparação de defeitos ósseos em fêmures e nos componentes moleculares do osso em ratas ovariectomizadas / Influence of strontium ranelate on the repair of bone defects in femurs and molecular components of bone in ovariectomized rats

O ranelato de estrôncio (RS) diminui a reabsorção óssea e ao mesmo tempo age como um agente anabólico deste tecido. Este estudo foi realizado para avaliar os efeitos da ovariectomia e do tratamento com RS na reparação de defeitos ósseos em fêmures e nos componentes moleculares do osso em ratas. Vinte e sete ratas adultas foram submetidas a ovariectomia ou cirurgia Sham e, após trinta dias, defeitos ósseos em fêmures foram confeccionados e os animais divididos em três grupos: ovariectomizadas (OVZ), cirurgia Sham (SHAM) e ovariectomizadas + tratamento com 625 mg/kg/dia de RS (RS). A eutanásia foi realizada quatro semanas após a cirurgia do defeito ósseo. A reparação do defeito ósseo foi analisada por microtomografia computadorizada (?CT) e a composição química do tecido ósseo foi verificada por espectroscopia de infravermelho com transformada de Fourier (FTIR) e espectroscopia de energia dispersiva de raios-X (EDS). O grupo SHAM apresentou em média volume ósseo (BV) superior ao grupo OVZ (p=0,014) e o volume ósseo relativo (BV/TV) foi cerca de 8% superior ao grupo OVZ. A comparação entre SHAM e RS apresentou valores limítrofes para essas variáveis. A espessura trabecular (Tb.Th) no grupo RS foi significativamente maior que no grupo OVZ cerca de 1,8% (p = 0,049) e não diferiu em média do grupo SHAM. Não houve diferenças significativas nas relações que avaliam compostos inorgânicos sobre orgânicos nos grupos. A razão das áreas das bandas em 1057 e 1023 cm-1 para o cálculo do IC foi 1.024 no grupo SHAM, 1.015 no grupo RS e 1.108 no grupo OVZ. No estudo da banda de amida III foi observado nos animais ovariectomizados uma possível mudança na estrutura secundária das proteínas da matriz devido a uma diminuição das estruturas ?-hélice e random coil (RC) e aumento de ?-sheet. A maturidade do colágeno estimada pela relação de sub-bandas de amida I (1660/1690) foi menor no grupo RS. Neste grupo houve um aumento significativo de estrôncio (Sr) incorporado ao tecido ósseo e uma diminuição de cálcio (Ca) avaliados pela técnica do EDS. Os resultados demonstram que o tratamento sistêmico com RS promoveu a reparação óssea e melhorou a microarquitetura do osso neoformado no interior do defeito. No entanto, os resultados das análises com FTIR sugerem que o RS não reverteu as alterações na matriz orgânica ocasionadas pela ovariectomia e diminuiu as ligações cruzadas de colágeno maturas. / Strontium ranelate (SR) decreases bone resorption and at the same time acts as an anabolic agent in this tissue. This study was conducted to evaluate the effects of ovariectomy and treatment with SR on the repair of bone defects and molecular components of bones in femurs. Adult female rats (n=27) were subjected to ovariectomy or Sham surgery. Thirty days after surgery, a defect was made in the femur and the animals were divided into three groups: ovariectomy (OVZ), Sham surgery (SHAM) and ovariectomy plus treatment with SR, 625 mg / kg / day (SR). Euthanasia was performed four weeks after surgery to cause the bone defect. Repair in bone defect was assessed by computed microtomography (?CT) and the chemical composition of bone tissue was analyzed by Fourier transform infrared (FTIR) spectroscopy and energy dispersive X-ray spectroscopy (EDS). The SHAM group showed mean values for bone volume (BV) higher than those of the OVZ group (p = 0.014) and the relative bone volume (BV / TV) was about 8% higher than that of the OVZ group. Comparison between SHAM and SR showed borderline values for these variables. In the SR group, the average value for trabecular thickness (Tb.Th) was significantly higher (~1.8%) than that in the OVZ (p = 0.049) group and did not differ from that of the SHAM group. Significant difference was not observed in the relation between inorganic and organic compounds in the three groups. The ratio of the areas of the bands in 1057 and 1023 cm-1 for the calculation of IC was 1,024 in the SHAM group, 1,015 in the RS group and 1,108 in OVZ group. In the study of the amide III band, a change in the proportion of secondary structure of matrix proteins was observed in ovariectomized animals, possibly a decrease in of ?-helical and random coil (RC) and an increase in ?-sheet structures. The collagen cross-linking network maturity estimated by the relation between amide I subbands (1660/1690) showed the smaller number in the SR group. In this group, a significant increase in the amount of strontium (Sr) and a decrease in the amount of calcium (Ca) embedded to bone tissue, as assessed by the EDS technique, were also observed. The results demonstrate that systemic treatment with SR promoted bone repair and improved microarchitecture of the newly formed bone within the defect. However, results of analyses with FTIR suggest that the SR has not reversed the alterations caused by ovariectomy in the organic matrix and decreased cross-linking of collagen mature.

We have optimizeda processing route consisting in entangling and shape-setting a spring bythermomechanical treatments. The samples were characterized in compression andtension, using optical and x-ray tomographic observations. The structures thus obtainedare homogeneous, isotropic, superelastic at room temperature up to strains ofat least 30%, and can become ferroelastic with a shape memory effect up to at least16% strain by an additional heat treatment. The mechanical behavior in compressionis first consolidating and then dilating, while in tension, the samples are slightlyauxetic. A comparison with similar media made of ductile and viscoelastic fibers,as well as with discrete element simulations on friction-free elastic fibers, show thatthe mechanical properties of these self-entangled structures are controlled by theirunique architecture, in-between continuous and discrete media.

Étude de la rhéologie des suspensions de fibres non-newtoniennes par imagerie et simulation numérique 3D à l'échelle des fibres. / 3D Micro-Rheology of non-Newtonian fibre suspensions using fast X-ray tomography and Finite Element simulations at fibre scale

Ce travail porte sur la mise en forme des matériaux composites à matrice polymère renforcée par des fibres courtes dont les performances physiques et mécaniques sont directement reliées à la distribution spatiale et à l’orientation des renforts employés. Il se focalise sur l’étude des mécanismes de déformation se produisant au cours de l’écoulement de ces systèmes qui se comportent comme des suspensions de fibres non-newtoniennes. Le problème est abordé par une procédure originale combinant images 3D acquises en temps réel et simulations numériques avancées, réalisées à l’échelle des fibres. Dans le premier cas, des suspensions modèles avec fluide suspensif non-newtonien ont été déformées en compression dans des conditions confinées dans un microtomographe à rayons X synchrotron. Cette technique a permis l’acquisition en temps réel de clichés 3D à forte résolution spatiale de l’écoulement des suspensions. Dans le deuxième cas, un code de calculs éléments finis 3D a été utilisé, celui-ci étant capable de décrire finement des objets immergés dans des fluides non-newtoniens, par des level-sets et des techniques de remaillage anisotrope. La pertinence des simulations numériques dans les régimes de concentration dilués à semi-dilués a été jaugée par une comparaison expériences-simulations avancée.De là, dans le régime de concentration dilué, nous montrons que le confinement de l’écoulement et le comportement rhéofluidifiant du fluide suspensif ont une influence mineure sur la cinématique des fibres, si ces dernières sont suffisamment éloignées des plateaux de compression. Si ce prérequis n’est pas respecté, l’effet du confinement devient important. Des modifications au modèle heuristique d’haltère de la littérature ont été proposées pour corriger la cinématique de fibres. Dans le régime semi-dilué, des déviations de la cinématique de fibres sont également observées au cœur des suspensions. Ces déviations sont principalement liées aux interactions hydrodynamiques entre fibres suffisamment voisines. La cinématique des fibres prédite par le modèle de Jeffery et les approximations de champ affine sont mises en défaut. Dans le régime concentré, si l’évolution de l’orientation globale de la suspension est étonnamment bien décrite par l’équation de Jeffery, de très importantes fluctuations des champs de translation et de rotation des fibres sont observées à l’échelle des fibres. Celles-ci sont induites par les nombreux contacts entre fibres qui peuvent par ailleurs être correctement prédits par le modèle de tube. / This study focuses on the processing of short fibre-reinforced polymer composites. The physical and mechanical properties of these materials are mainly affected by the position and orientation distribution of fibres induced during their forming. Thus, we analysed the flow-induced micro-mechanisms that arose at the fibre scale during the forming stage of these complex systems which behave as non-Newtonian fibre suspensions. For that purpose, an original approach was developed by combining 3D imaging technique and direct numerical simulation, both performed at the fibre scale. Hence, several model fibre suspensions with a non-Newtonian suspending fluid and with a concentration regime that ranged from dilute to concentrated were prepared . They were subjected to confined lubricated compression loadings using a rheometer mounted on a synchrotron X-ray microtomograph. Thanks to very short scanning times, 3D images of the evolving fibrous microstructures at high spatial resolution were recorded in real-time. These experiments were also simulated using a dedicated Finite Element library enabling an accurate description of fibre kinematics in complex suspending fluids thanks to high performance computation, level sets and adaptive anisotropic meshing. The efficiency of the numerical simulation from the dilute to semi-dilute concentration regimes was assessed through experimental and numerical comparisons.Then, we showed that the confinement effect and the non-Newtonian rheology of the suspending fluid had a weak effect on the fibre kinematics, if the fibres were sufficiently far from the compression platens, typically the fibre-platen distance should be larger than twice the fibre diameter. Otherwise, confinement effects occurred. Some extensions of the dumbbell model were proposed to correct the fibre kinematics in this flow conditions. In semi-dilute concentration, deviations of the fibre kinematics compared to the Jeffery’s predictions were also observed and related to hydrodynamic interactions between fibres. In this case, the predictions of Jeffery’s model and the related assumption of affine fibre motions are less relevant. In the concentrated regime, even if the overall orientation of fibre suspension could be astonishingly well described by using the Jeffery’s model, strong fluctuations on each fibre motion and rotation were observed. These deviations were induced by the numerous fibre-fibre contacts, which could be correctly predicted by the tube model.

Rhéologie des suspensions concentrées de fibres : application à la mise en forme des matériaux composites / Rheology of concentrated fibre suspensions : Application to polymer composite forming

Cette étude porte sur la mise en forme des matériaux composites renforcés par des fibres ou des mèches de fibres courtes tels que les SMC ou les BMC. Un travail expérimental a dans un premier temps été réalisé à l’échelle macroscopique. Ce travail a permis de mettre au point un rhéomètre de compression lubrifiée ainsi que des méthodes d’essais et de dépouillement. Ceci permet de mieux caractériser la rhéologie des compounds SMC et BMC en traitant les problématiques de leur compressibilité et des frottements éventuels entre les parois du rhéomètre et la matière déformée. Un travail numérique a ensuite permis de simuler la mise en forme d’un BMC après l’identification des paramètres d’un modèle rhéologique simple à partir des données expérimentales obtenues sur le rhéomètre. Enfin, un travail expérimental à l’échelle microscopique a permis d’une part de caractériser finement les microstuctures de SMC modèles à partir de microtomographies à rayons X, et d’autre part de caractériser et de modéliser par le biais d’essais d’extraction de fibres les interactions entre les mèches formant le renfort fibreux de ces matériaux. / This study focuses on the processing of composite materials reinforced with short fibres or fibres bundles such as SMC or BMC. Firstly, an experimental work was carried-out at the macroscopic scale. This work led to the development of a lubricated compression rheometer and associated analysis methods to better characterize the rheology of SMC and BMC compounds, by accounting for the compressibility of compounds and the possible friction between the rheometer wall and the flowing composite. Numerical simulation was then achieved in order to simulate the forming of a BMC. For that purpose, the constitutive parameters of a simple tensorial rheological model were determined from experimental data obtained with the rheometer. Finally, an experimental work at the microscopic level allowed (i) the microstuctures of SMC models from X-ray microtomography micrographs and (ii) fibre pull-out experiment to be characterized, and the interaction mechanisms between the fiber bundles forming the fiber reinforcement of these materials to be modelled.

La simulation à l'échelle mésoscopique de la déformation des renforts composites fournit des informations importantes. En particulier, elle donne la direction et la densité de fibres qui conditionne la perméabilité du renfort textile et les propriétés mécaniques du composite final. Ces analyses mésoscopiques par éléments finis dépendent fortement de la qualité de la géométrie initiale du modèle. Certains logiciels ont été développés pour décrire ces géométries de renforts composites. Mais, les géométries obtenues impliquent une simplification (notamment dans la section transversale de mèche) qui peut perturber le calcul de déformation du renfort. Le présent travail présente une méthode directe utilisant la microtomographie à rayon X pour générer des modèles éléments finis, basée sur la géométrie réelle de l'armure textile. Le modèle EF peut être obtenu pour tout type de renfort, plus ou moins complexe. Les problèmes d’interpénétrations de mèches sont évités. Ces modèles sont utilisés avec deux lois de comportement : une loi hypoélastique et une loi hyperélastique. Les propriétés de chacune d'entre elles, ainsi que les grandeurs caractéristiques nécessaires à leur implémentation dans le logiciel ABAQUS sont développées. Enfin, une identification des paramètres matériau à l'aide d'une méthode inverse est proposée. Les résultats obtenus pour les simulations de mise en forme sont comparés avec les résultats expérimentaux et montrent une bonne correspondance entre les deux. / The simulation at meso-scale of textile composite reinforcement deformation provides important information. In particular, it gives the direction and density of the fibres that condition the permeability of the textile reinforcement and the mechanical properties of the final composite. These meso FE analyses are highly dependent on the quality of the initial geometry of the model. Some software have been developed to describe composite reinforcement geometries. The obtained geometries imply simplification that can disrupt the reinforcement deformation computation. The present work presents a direct method using computed microtomography to determine finite element models based on the real geometry of the textile reinforcement. The FE model is obtained for any specificity or variability of the textile reinforcement, more or less complex. The yarns interpenetration problems are avoided. These models are used with two constitutive laws : a hypoelastic law and a hyperelastic one. An analysis of their properties is presented and their implementation in the software ABAQUS is detailed. Finally, an identification method is presented and the results of forming simulations are compared to experimental tests, which shows a good fit between the both.

Application de la microtomographie par rayons X à l'étude des dépôts de tsunamis / Application of X-ray microtomography to the study of tsunami deposits

L’étude des dépôts de tsunamis est une science relativement jeune, et si de nombreux progrès ont été faits, en particulier au cours des trois dernières décennies, les techniques disponibles à l’heure actuelle ne permettent pas l’étude exhaustive des dépôts, que ce soit à cause de problèmes de conservation, fréquemment rencontrés, ou de limites liées aux techniques analytiques elles-mêmes. En effet, leur résolution spatiale s’avère insuffisante pour les dépôts fins (fractions sableuses et inférieures), soit par leur principe même (absence de visualisation en trois dimensions sur des lames minces, par exemple), soit à cause des techniques d’échantillonnage dont elles dépendent. Ce travail a permis d’explorer les applications d’une technique analytique jusqu’à présent inutilisée dans ce domaine, la microtomographie par rayons X, appliquée à trois dépôts de tsunamis : les dépôts du tsunami de Lisbonne en 1755 sur les côtes Andalouses, ceux d’un tsunami causé en 1996 dans le Lac Karymskoye au Kamchatka, et un potentiel dépôt de tsunami datant de l’Holocène à Ninety Mile Beach, sur le littoral ouest de l’Australie. Les problématiques liées aux techniques d’échantillonnage et au traitement des données brutes et à leur exploitation sont passées en revue et les solutions retenues sont exposées. Les résultats obtenus sont comparés à ceux obtenus par l’utilisation de techniques analytiques dont la pertinence et l’utilité ont été démontrées lors de précédentes études. Cette technique se révèle extrêmement utile pour les analyses structurales (organisation interne des dépôts), texturales (distributions de tailles de grains, fabrique sédimentaire et dynamiques d’écoulements), ainsi que les analyses de composition (abondances en bioclastes et en minéraux lourds par exemple). Le croisement des études structurales et texturales du dépôt du tsunami de 1755 mettent en avant des dynamiques de mise en place allant parfois à l’encontre des scenarii généralement admis et offre un nouveau point de vue sur les dépôts de tsunami. / The study of tsunami deposits is a relatively young science, and even if substantial progress has been made (especially during the last three decades) the techniques available at present day do not allow exhaustive studies of the deposits, weither poor conservation of the deposits, which is a common problem, is to blame, or because of limitations of the techniques themselves. Their spatial resolution often appears to be insufficient for fine deposits (sand size fractions and finer) because of their working principle (lack of three dimensional visualisation of structures on thin sections for instance) or because of the sampling techniques they depend on. This work allowed exploring the applications of an analytic technique, X-ray computed microtomography, which had to this day never used before for the study of tsunami deposits. Three distinct case studies have been made: deposits from the 1755 Lisbon tsunami on the western coast of Spain (Andalusia), deposits from the 1996 tsunami triggered in Karymskoye Lake, and a potential mid-Holocene tsunami deposit at Ninety Mile Beach, western Australia. Problematic linked to sampling techniques and raw data processing and exploitation are addressed. Chosen solutions are exposed. The results are compared to those obtained using techniques whose relevance and usefulness have been proven in previous studies. This technique proves to be extremely useful for structural (deposits inner organisation), textural (grain size distribution, sedimentary fabrics and flow dynamics), and composition (bioclasts and heavy minerals abundances) analyses. Crossing structural and textural analysis of the 1755 Lisbon tsunami deposit unravels deposition modes which go against the usually admitted scenario and offers a new point of view on tsunami deposits.

Este trabalho inicialmente descreve a formação das imagens por contraste de fase pelo método da propagação. Apresenta simulações de primeiros princípios para esta técnica comparando estes com a literatura. Em seguida reporta o desenvolvimento da instrumentação para uma estação experimental de microtomografias de raios X bem como os métodos de processamento de dados para reconstruções tomográficas. Microtomografias com alta resolução foram obtidas e são apresentadas com o intuito de caracterizar a instrumentação e suas aplicações. Essa técnica de instrumentação foi aplicada, em particular, no estudo da histomorfometria óssea em ratos Sprague-Dawley com o objetivo de quantificar os efeitos da dieta alimentar na estrutura óssea trabecular e compacta. Argumenta-se que ocorre uma ligeira tendência a diferenciação nesses tecidos em função da dieta alimentar. Além disso, neste trabalho foi implementado, caracterizado e aplicado a técnica de imagens harmônicas por espalhamento em amostras biológicas, detecção de fraturas e espalhamento anisotrópico. Também são descritos os esforços na melhoria da qualidade sinal-ruído dessa técnica / Abstract: The main scope of this work was the study, implementation, characterization and application of x-ray phase contrast and scattering constrast imaging techniques. First we describe the production of phase constrast images and early attempts to simulate phase contrast phenomena following the known literature. We also report efforts toward development of an experimental x-ray microtomography station as well as imaging processing techniques for tomography reconstruction. We present results on high resolution x-ray microtomography. This technique was applied in the study of Sprague-Dawley bone morphometric properties aiming quantification of high-fat diet effects on compact and trabecular bone structuture. It is argued about a tendency to differentiation on these tissues according to the diet. INVESTIGATING DAMAGE IN SHORT FIBER REINFORCED COMPOSITES

<div>In contrast to traditional steel and aluminum, short fiber reinforced polymer composites (SFRCs) provide promising alternatives in material selection for automotive and aerospace applications due to their potential to decrease weight while maintaining excellent mechanical properties. However, uncertainties about the influence of complex microstructures and defects on mechanical response have prevented widespread adoption of material models for</div><div>SFRCs. In order to build confidence in models’ predictions requires deepened insight into the heterogenous damage mechanisms. Therefore, this research takes a micro-mechanics standpoint of assessing the damage behavior of SFRCs, particularly micro-void nucleation at the fiber tips, by passing information of microstructural attributes within neighborhoods of incipient damage and non-damage sites, into a framework that establishes correlations between the microstructural information and damage. To achieve this, in-situ x-ray tomography of the gauge sections of two cylindrical injection molded dog-bone specimens, composed of E-glass fibers in a polypropylene matrix, was conducted while the specimens were monotonically loaded until failure. This was followed by (i) the development of microstructural characterization frameworks for segmenting fiber and porosity features in 3D images, (ii) the development of a digital volume correlation informed damage detection framework that confines search spaces of potential damage sites, and (iii) the use of a Gaussian process classification framework to explore the dependency of micro-void nucleation on neighboring microstructural defects by ranking each of their contributions. Specifically, the analysis considered microstructural metrics related to the closest fiber, the closest pore, and the local stiffness, and the results demonstrated that less stiff resin rich areas were more relevant for micro-void nucleation than clustered fiber tips, T-intersections of fibers, or varying porosity volumes. This analysis provides a ranking of microstructural metrics that induce microvoid nucleation, which can be helpful for modelers to validate their predictions on proclivity of damage initiation in the presence of wide distributions of microstructural features and</div><div>manufacturing defects. </div>

[pt] A indústria mineral vem presenciando, ao longo das últimas décadas, uma redução da qualidade de minério de ferro extraído e o surgimento de novas demandas ambientais. Esta conjuntura fortalece a busca por produtos provenientes do minério de ferro que atendam aos requisitos da indústria siderúrgica, como é o caso de novos aglomerados de minério de ferro. A Microtomografia de Raios-X (microCT) permite a caracterização da estrutura tridimensional de uma amostra, com resolução micrométrica, de forma não-destrutiva. Entretanto, tal técnica apresenta diversas limitações. Quanto melhor a resolução, maior o tempo de análise e menor o volume de amostra adquirido. Modelos de Super Resolução (SR), baseados em Deep Learning, são uma poderosa ferramenta para aprimorar digitalmente a resolução de imagens tomográficas adquiridas em pior resolução. Este trabalho propõe o desenvolvimento de uma metodologia para treinar três modelos de SR, baseados na arquitetura EDSR, a partir de imagens tomográficas de briquetes de redução direta: Um modelo para aumento de resolução de 16 um para 6 um, outro para aumento de 6 um para 2 um, e o terceiro para aumento de 4 um para 2 um. Esta proposta tem como objetivo mitigar as limitações do microCT, auxiliando o desenvolvimento de novas metodologias de Processamento Digital de Imagens para os aglomerados. A metodologia inclui diferentes propostas para avaliação do desempenho da SR, como comparação de PSNR e segmentação de poros. Os resultados apontam que a SR foi capaz de aprimorar a resolução das imagens tomográficas e mitigar ruídos habituais da tomografia. / [en] The mining industry has been witnessing a reduction of extracted iron ore s quality and the advent of new environmental demands. This situation reinforces a search for iron ore products that meet the requirements of the steel industry, such as new iron ore agglomerates. X-ray microtomography (microCT) allows the characterization of a sample s three-dimensional structure, with micrometer resolution, in a non-destructive analysis. However, this technique presents several limitations. Better resolutions greatly increase analysis time and decrease the acquired sample’s volume. Super-Resolution (SR) models, based on Deep Learning, are a powerful tool to digitally enhance the resolution of tomographic images acquired at lower resolutions. This work proposes the development of a methodology to train three SR models, based on EDSR architecture, using tomographic images of direct reduction briquettes: A model for enhancing the resolution from 16 um to 6 um, another for enhancing from 6 um to 2 um, and the third for enhancing 4 um to 2 um. This proposal aims to mitigate the limitations of microCT, assisting the development and implementation of new Digital Image Processing methodologies for agglomerates. The methodology includes different proposals for SR s performance evaluation, such as PSNR comparison and pore segmentation. The results indicate that SR can improve the resolution of tomographic images and reduce common tomography noise.
