SMALL MOLECULE INHIBITORS IN EGFR/HER-2 AND MUTANT RAS/RAF BREAST CANCER MODEL SYSTEMS

Dysregulation of receptor tyrosine kinase (RTK) signaling has been implicated in the pathogenesis of breast cancer. Both EGFR and HER-2 regulate tumor cell proliferation and survival through the Ras-Raf-Mek-Erk (MAPK) pathway. Cyclin-dependent kinases (CDKs) are implicated in regulating proliferative and survival signaling downstream of the MAPK pathway. Here I show that CDK inhibitors exhibit an order-of-magnitude greater cytotoxic potency than a suite of inhibitors targeting RTK and Ras-MAPK signaling in cell lines representative of clinically recognized breast cancer subtypes. Drug combination studies showed that the pan CDK inhibitor, Flavopiridol, synergistically potentiated cytotoxicity induced by the Raf inhibitor, Sorafenib. In vivo mouse models of breast cancer treated with both drugs exhibited reduced primary tumor growth rates and metastatic tumor load in the lungs compared to treatment with either drug alone, and this correlated with greater reductions in Rb signaling and Mcl-1 expression in resected tumors. In addition, Mek inhibition with U0126 potentiated the cytotoxic efficacy of Sorafenib, showing added benefits of drug combination to reduce doses while achieving greater therapeutic efficacy in a mutant Ras/Raf triple-negative breast cancer cell line. In vivo mouse models showed targeted Raf (Sorafenib) and Mek (AZD6244) inhibition resulted in reduced primary tumor growth and metastatic tumor load in the lungs, with the combined targeted Raf and Mek treatment showing greater inhibition than targeting either Raf or Mek alone. Synergistic anti-tumorigenic effects of the Sorafenib-Flavopiridol and Sorafenib-U0126/AZD6244 combinations across breast cancer cells driven by EGFR/HER-2 or Ras-MAPK signaling suggest the broad applicability of these combinations to treat multiple breast cancer subtypes. Interaktionsbedömning vid kommunikativ funktionsnedsättning : Översättning, prövning och utvärdering av ett verktyg för bedömning av interaktion mellan barn med kommunikativ funktionsnedsättning och deras föräldrar

Interaktion är det komplexa samspel mellan interaktörer där meningsskapande skerinom kommunikativa sekvenser. Föreliggande studie syftar till att översätta ettbedömningsinstrument till svenska för bedömning av interaktion mellan barn medkommunikativ funktionsnedsättning och deras föräldrar. Instrumentet Coding of Parent-Child Interaction and Communication Skills Assessment är hämtat från studier avPennington et al. (1999; 2009) och prövas på två interaktionsdyader (barn i interaktionmed en förälder) före och efter genomgången föräldrakurs för att utvärdera ominstrumentet är användbart vid bedömningen av interaktion. Den svenska versionen avbedömningsinstrumentet innehåller koder med definitioner för identifiering avinteraktionsmönster. Strukturella bidrag och pragmatiska funktioner utgör kategorierför koderna. En jämförelse av kodningsresultaten gjordes genom kvalitativkommunikationsanalys med grund i de principer för samtalsanalys som arbetats framinom Conversation Analysis (CA). Resultaten visade att det krävs utveckling ochförtydligande av definitioner, avgränsningar och utökning av koder ochkodningsschema för fortsatt användning. Den komparativa analysen visar hur störrehänsyn måste tas till den bredare samtalskontexten, passiva respektive aktiva bidragsamt mottagarens tolkning av det aktuella bidraget vid användning avbedömningsinstrumentet. Man kan dock på tydligt sätt demonstrera interaktionsmönstergenom att använda bedömningsinstrumentet och dess kodningsstruktur. The purpose of the study at hand is totranslate an instrument into Swedish for assessment of interaction between children withcommunicative disorders and their parents. The instrument Coding of Parent-ChildInteraction and Communication Skills Assessment was originally published inPennington et al. (1999; 2009) and is here tested on two interactional dyads (child ininteraction with one parent) pre and post intervention, to evaluate whether theinstrument can be used for assessment of interaction. The Swedish version of theinstrument consists of codes and definitions for identification of interactional patterns ofthe participants. Code categories are structural moves and pragmatic functions. Resultsof the coding are compared with qualitative communication analysis using methodsbased on conversation analysis (CA). Results showed development and clarification ofdefinitions, limitations and additions of the codes and coding scheme are essential forfurther usage. The comparative analysis demonstrates how the instrument must developin regard of the conversational context, passive and active moves and the listener’sinterpretation and response of the move in question. However, the coding function andits structure is a good way to clarify and demonstrate interactional patterns. Dyskalkyli i vardagen : – svårigheter, konsekvenser och strategier

Följande studie syftar till att beskriva hur vuxna personer med diagnosen dyskalkyli påverkas i vardagen av sina matematiska svårigheter. Studien är kvalitativ och bygger på fem semistrukturerade intervjuer. Den kartlägger vilka strategier som används för att komma runt de matematiska svårigheterna, hur deltagarna har blivit bemötta av omgivningen och vilka förslag deltagarna själva har på hur man kan underlätta deras vardag. Dyskalkyli, eller specifika räknesvårigheter, förekommer hos 3-6% av befolkningen. Forskningen kring diagnosen är begränsad i jämförelse med andra inlärningssvårigheter. Forskarna har ännu inte enats kring orsakerna och forskningen kompliceras av att diagnosgruppen är heterogen. Studiens resultat visar att dyskalkyli, i de flesta fall, får stora konsekvenser för vardagslivet såväl i skolan och på jobbet som på fritiden. Detta trots användandet av en mängd strategier för att underlätta vardagen som exempelvis att ta hjälp av sin omgivning och att använda miniräknare. Räknesvårigheterna gör bland annat att det är svårt att göra inköp, att följa vägbeskrivningar och att passa tider. Deltagarna upplever också att de har fått ett okunnigt och oförstående bemötande från omgivningen och önskar mer hjälp och förståelse i framtiden. Dyskalkyli visar sig i hög grad kunna påverka både känslolivet, välbefinnandet och, hos vissa, även stora livsval. Vilka konsekvenser som orsakas av just dyskalkylin går inte att fastslå med säkerhet. Deltagarnas uppfattning är dock att räknesvårigheterna har påverkat ovan nämnda faktorer.   Nyckelord: dyskalkyli, räknesvårigheter, strategier, upplevelser, bemötande / The following study aims to describe how mathematical difficulties affect adults diagnosed with dyscalculia in their everyday life. It is a qualitative study which is based on five semi-structured interviews. It maps what strategies are used to get around the mathematical difficulties, how the participants have been treated by people in their environment and the participants’ suggestions of how to facilitate their everyday life. Dyscalculia, or mathematical learning disabilities, is found in 3-6% of the population. Compared to other learning disabilities, there is a limited amount of research on the diagnosis. The researchers have yet to achieve consensus concerning the causes and the research is further complicated by the heterogeneity of the group. The results of the study show that dyscalculia, in most cases, have great impact on everyday life, in school, as well as at work and during spare time. This despite the usage of several strategies to facilitate everyday life, such as asking others for help and using a calculator. The mathematical difficulties complicate for example making purchases, following directions and being on time. The participants’ experience is that people in their environment have treated them with ignorance and a lack of understanding, and they request more help and understanding in the future. As it shows, dyscalculia may have a serious impact on emotions and well-being and, for some, even major choices in life. What consequences are caused by the dyscalculia per se is not possible to establish. However, it is the opinion of the participants that the mathematical difficulties have affected the factors mentioned above.   Keywords: dyscalculia, mathematical disabilities, strategies, experiences, treatment
