Adhesive organs are widespread structures among vertebrate larvae. They allow the larvae to attach to a substrate, so that the time for the development of mouth or motoric apparatus could be prolonged. Similar structures in ascidians, larvaceans and lancelets are known too. Thus, it might be hypothesized that the presence of some type of adhesive gland could indeed represent the ancestral state for chordate larvae. Interestingly, however, whilst in most species these glands take their developmental origin in ectodermal layer, in bichir, a member of a primitive actinopterygian lineage, their origin was suggested to be endodermal already at the beginning of 20th century. Since then, however, the former study has become almost forgotten and even recent analyses do not come with new findings on this topic. Because of the essential importance of study of bichir cement glands for understanding the relationship between these structures among chordates, I have decided to focus on this subject. To obtain appropriate comparative data Xenopus, Weather loach and Ribbed newt embryos were included in this study as well. Aquatic larvae of many vertebrate lineages develop specialized, cranially located cement or attachment glands which allow them to remain attached to a substrate by means of polysaccharide secretion. The larvae can thus remain still and safe in well-oxygenated water out of reach of any predators until the digestive and locomotory apparatus fully develops. Xenopus cement gland is the most thoroughly studied example of this type of glands, since it was used as a model for the anteriormost patterning of the developing head. Based on shared expression patterns of key transcription factors and a similar ectodermal origin it has been repeatedly suggested that Xenopus cement gland is homologous to adhesive organs of teleosts and adhesive papillae of ascidians. The lack of comprehensive knowledge on this type of glands in other lineages however rendered any considerations of homology among such a distant lineages rather inconclusive. In the present work I have focused on a detailed study of the cement glands and other corresponding structures in three representatives of basal actinopterygian lineages: Senegal bichir (Polypterus senegalus), sterlet (Acipenser ruthenus), and tropical gar (Atractosteus tropicus). Dynamika encystace střevního prvoka Giardia intestinalis. / Dynamics of Giardia intestinalis encystation.Vinopalová, Martina January 2018 (has links)
Giardia intestinalis is an anaerobic parasite, that colonizes the small intestine of humans and other vertebrate hosts. This cosmopolitan parasite, which causes diarrhoea, is transmitted by contaminated water or food via a resistant stage, the cyst. The encystation process involves a number of events that lead to a complete reconstruction of the cell into the form of infectious cyst. The aim of this work was to visualize these modifications in vivo by means of enzymatic labelling of proteins. For the purposes of this work, enzymatic tags Y-FAST and HaloTag were chosen, as they enable visualizing live cells under anaerobic conditions. Chimeric protein constructs were created to visualize the dynamics of the encystation vesicles, the structures of endoplasmic reticulum, the adhesive disc and mitosis. Using the developed constructs, we successfully followed the dynamics of the encystation vesicles and the adhesive disc in vivo. Finally, this work has provided novel molecular tools, which will be used to follow the overall redesign of the parasite cell during encystation.

Introduction: Adhesive technology represents a significant advancement among different treatment options of dental caries. Its advantages involve not only highly aesthetic reconstructions, but also ability to preserve hard dental tissues compared to more traditional filling materials such as amalgam, where the material retention was achieved through undercuts. A potential of adhesive technology to arrest progression of an incipient caries has triggered its wider use in dental caries management, potentiated by an ongoing development of new adhesive systems and composite materials that assure an acceptable long-term stability of adhesive reconstructions. Adhesive bonds of dental reconstructions are in the oral cavity constantly challenged by various mechanical, chemical and biological factors. One of the chemical agents with detrimental potential is peroxide bleaching gel used for teeth brightening. Currently bleaching belongs to the most frequent cosmetic dental procedures. The active ingredient of bleaching gels is hydrogen peroxide or its precursors. It is assumed that free oxygen radicals released from peroxides attack double bonds of chromophore molecules captured within the tooth tissues. Introduction: An adhesive bond between hard dental tissues and composite reconstructions is exposed to mechanical load, water, bacteria, enzymes and different chemical compounds. These factors could impair durability and resistance of composite reconstructions. In contrast with extensive research of negative influence of water on degradation of adhesive bonds, the influence of chemical substances on adhesive bond has not been investigated to a greater extent. Aim: The aim of the experimental part of the thesis was to evaluate the influence of hydrogen peroxide containing tooth-whitening products and mouth rinses containing chlorhexidine and fluoride on a long-term stability of class V composite restorations created using four contemporary adhesive systems by a microleakage method. Materials and methods: The following adhesive systems were investigated: total-etch Gluma Comfort Bond (GLU), and self-etch adhesives Clearfil SE Bond (CLF), Adper Prompt (ADP) and iBond (IBO). The theoretical part deals with solid dispersions, their classification and methods of preparation, methods of drug dissolution testing, properties, effect, use of terbinafine and preparations containing terbinafine. In the experimental part, solid dispersions were prepared by the melting method. The drug was incorporated into the polyester in the form of a hydrochloride or a base in a concentration of 10 % or 20 %. Some systems were plasticized with 30% triethyl citrate. Thin layers were formulated from solid dispersions, and drug dissolution tests were performed on phosphate buffer pH 7.4 at 37 řC. Assay of terbinafine released was performed spectrophotometrically at 223 nm and ultra-high performance liquid chromatography. Juraj Martiška The diploma thesis is focused on biodegradable polymer nanoparticles loaded by terbinafine based on the copolymer of glycolic and lactic acid branched on polyacrylic acid. The nanoprecipitation method was employed, and the influence of formulation factors on nanoparticle characteristics was studied. The following formulation factors were the concentration of the polymer, the amount of terbinafine, and the concentration of surfactant. Nanoparticles of 120 nm to 300 nm were obtained depending on the preparation conditions. The nanoparticle polydispersity was in all cases from 0.080 to 0.230. The prepared nanoparticles were stable, as evidenced by zeta potential values above 38 mV. A positive zeta potential is desirable for dermal and mucosal adhesion in the topical and ocular application of nanoparticles with terbinafine.
