Copula Deletion in English as a Lingua Franca in AsiaLeuckert, Sven 11 July 2019 (has links)
Non-standard features such as copula deletion have long been dismissed as learner errors or were interpreted as results of simplification processes in English as a Lingua Franca (ELF), and only recent publications tend to acknowledge the influence of language contact in ELF settings (cf. Schneider 2012). The present paper analyses tokens of copula deletion in the Asian Corpus of English (ACE 2014) with respect to speaker L1s, situational context and syntactic environment, with our results suggesting a correlation between copula usage patterns in the speakers’ L1s and constructions involving copula deletion found in ACE. Thus, opening up the field to ELF settings, our data confirm findings of previous studies such as Sharma (2009) that point to contact-induced copula usage in non-standard English(es).

Im Zentrum meiner folgenden Darstellung stehen Charakteristika der Filme von Mara Mattuschka, die in Kooperationen mit Chris Haring und liquid loft entstanden sind, und die Frage, in welcher Weise sie zur Transformation der Bühnenperformances beitragen. Eine Dokumentation der Tanz-Perfomances, die in der gängigen Ausformung in einer Totalen und in einem Take oder mit mehreren Kameras auf der Bühne gefilmt wird, stand nie zur Diskussion. Vielmehr versuchte Mattuschka mit dem Medium Film Verknüpfungen von Bild,- Sound,- und Körperchoreografien zu entwickeln, die die Qualitäten der jeweiligen Dramaturgien akzentuieren.

Filmische Aufzeichnungen von Aufführungen – sei es Theater, Tanz oder Performance – sind Transformationen und folgen deren medialen Bedingungen, auch wenn sie sich als Dokumentation verstehen. Sie überführen Raum und Dauer eines performativen Ereignisses in filmische Parameter, konstruieren sie neu und anders. Am Beispiel von Babette Mangoltes Film „Marina Abramovic – Seven Easy Pieces“ untersucht der Text einen solchen Transformationsprozess. Und stellt ihm eine andere intermediale Bewegung zur Seite, konfrontiert ihn mit der Frage: was interessiert Filmkünstler_innen an einer deutlich als theatral markierten mise en scène? Arbeiten der Wiener Künstlerin Mara Mattuschka bilden die Basis für die Suche nach Antworten auf diese Frage. Mit dem Begriff der Maskerade und seinen verschiedenen Bedeutungskontexten werden Verfahren der Schichtung verschiedener künstlerischer Techniken und Gestaltungsparameter beschrieben.

Objective: Brain-computer interfaces (BCIs) aimed at restoring communication to people with severe neuromuscular disabilities often use event-related potentials (ERPs) in scalp-recorded EEG activity. Up to the present, most research and development in this area has been done in the laboratory with young healthy control subjects. In order to facilitate the development of BCI most useful to people with disabilities, the present study set out to: (1) determine whether people with amyotrophic lateral sclerosis (ALS) and healthy, age-matched volunteers (HVs) differ in the speed and accuracy of their ERP-based BCI use; (2) compare the ERP characteristics of these two groups; and (3) identify ERP-related factors that might enable improvement in BCI performance for people with disabilities. Methods: Sixteen EEG channels were recorded while people with ALS or healthy age-matched volunteers (HVs) used a P300-based BCI. The subjects with ALS had little or no remaining useful motor control (mean ALS Functional Rating Scale-Revised 9.4 (±9.5SD) (range 0-25)). Each subject attended to a target item as the items in a 6. ×. 6 visual matrix flashed. The BCI used a stepwise linear discriminant function (SWLDA) to determine the item the user wished to select (i.e., the target item). Offline analyses assessed the latencies, amplitudes, and locations of ERPs to the target and non-target items for people with ALS and age-matched control subjects. Results: BCI accuracy and communication rate did not differ significantly between ALS users and HVs. Although ERP morphology was similar for the two groups, their target ERPs differed significantly in the location and amplitude of the late positivity (P300), the amplitude of the early negativity (N200), and the latency of the late negativity (LN). Conclusions: The differences in target ERP components between people with ALS and age-matched HVs are consistent with the growing recognition that ALS may affect cortical function. The development of BCIs for use by this population may begin with studies in HVs but also needs to include studies in people with ALS. Their differences in ERP components may affect the selection of electrode montages, and might also affect the selection of presentation parameters (e.g., matrix design, stimulation rate). Significance: P300-based BCI performance in people severely disabled by ALS is similar to that of age-matched control subjects. At the same time, their ERP components differ to some degree from those of controls. Attention to these differences could contribute to the development of BCIs useful to those with ALS and possibly to others with severe neuromuscular disabilities.

Detection and quantification of post-translational modifications in non-invasive samples : Phosphoproteins as biomarkers and a market analysis of protein quantification technologiesBaudin, Sammi, Fjellström, Hillevi, Kraft, Aron, Lamberg, Erica, Rosenbaum, Måns, Sjöstrand, Hanna January 2023 (has links)
Post-translational modifications (PTMs) of proteins can be a sign and/or cause of disease. These modified proteins have the potential to be used as biomarkers for diagnostic purposes. However, research in the field is limited. The challenge of having an accessible way of diagnosing patients in time and at a low cost is crucial to improve public health. Blood samples or other non-invasive methods to detect diseases such as Alzheimer’s disease, Parkinson’s disease, Amyotrophic lateral sclerosis and cancers are of urgent need. This report investigates PTMs as possible biomarkers measurable in biofluids, such as blood, for diagnosis and prognosis. Biomarkers like phospho-tau and amyloid-beta are examined in the context of neurodegenerative diseases, as well as phosphorylations on neurofilaments, TAR DNA-binding protein 43 and α-synuclein. All of these are detectable in blood. Several PTMs with connection to different types of cancers are also investigated, such as F3-phosphopeptide and AFP-L3. It was found that many biomarkers for the detection of cancers can potentially be found in extracellular vesicles in blood. Methods such as ELISA, PEA, SomaScan, xMAP, SIMOA and mass spectrometry (MS) are all now available on the market to quantify these PTMs. MS has revolutionized the fields of protein detection in the past and has further evolved to being capable of protein quantification. ELISA has been prevalent for decades and laid the groundwork for improved methods such as xMAP and SIMOA that are easy to use and provide adequate sensitivity. SomaScan and PEA lead the way in dynamic range and multiplexing capacity with around 7000 and 3000 protein assays. The soon-to-be-released technology NULISA, with promising values in sensitivity and dynamic range, is also investigated here. Additionally, a written ethical analysis regarding the process and consequences of biomarker quantification through these technologies was performed. Although the investigated biomarkers are detectable in biofluids, using them as clinical diagnostic markers still poses a challenge, which is why further research in the field is needed. Through an increased knowledge of PTMs of proteins and the right use of platforms, clinical diagnostics and population screenings can be done more efficiently improving public health around the world.

Als offen angelegtes Experiment brachte Echohaus 2009 ein klassisch-modernes Ensemble mit einen elektronischen DIY-Musiker und einem Produzenten für Underground-Popmusik zusammen. Für die einwöchigen Aufnahmen in dem Hamburger Studio Electric Avenue galten einige Regeln: keine künstlichen Halleffekte oder digitalen Plug-Ins verwenden, nur die akustischen Reflexionen echter Räume, keine Overdubs. 2010 erschien aus dem Projekt ein Tonträger beim Hamburger Avantgarde-Label Dekorder. Im gleichen Jahr gab es eine Uraufführung beim Märzmusik Festival in den Berliner Sophiensaelen. Aus Anlaß des 10-jährigen Jubiläums des Raum-Klang-Projektes Echohaus sprachen die beiden Komponisten und Musiker Felix Kubin und Burkhard Friedrich über dieses Projekt und die grundlegende Bedeutung von Raum als Klangkörper, vom Komponieren durch Bewegung im Raum und von technisch generierten parallelen Räumen.

Characterizing the Function of PAS kinase in Cellular Metabolism and Neurodegenerative DiseasePape, Jenny Adele 01 June 2019 (has links)
The second identified substrate of PAS kinase discussed is Pbp1. The human homolog of Pbp1 is ataxin-2, mutations in which are a known risk factor for amyotrophic lateral sclerosis (ALS). As diet and sex have been shown to be important factors regarding PAS kinase function, they also are strong contributing factors to ALS and are extensively reviewed herein. Pbp1 is known to be sequestered by PAS kinase under glucose depravation, and it can sequester additional proteins along with it to regulate different cellular pathways. To shed light on the pathways affected by Pbp1, we performed a yeast two-hybrid assay and mass spectrometry, identifying 32 novel interacting partners of Pbp1 (ataxin-2). We provide further analysis of the direct binding partner Ptc6, measuring mitophagy, mitochondrial content, colocalization, and respiration. This work elucidates novel molecular mechanisms behind the function of PAS kinase and yields valuable insights into the role of PAS kinase in disease.

Characterizing Stress Granule Regulation by PAS Kinase, Ataxin-2 and Ptc6 and Investigating the Lifespan of Covid-19 Virus on CurrencyNewey, Colleen R 07 December 2023 (has links) (PDF)
The protein Ataxin-2 is a known positive regulator of stress granules in humans, mice and yeast (known as yeast PBP1). Due to the role that stress granules play in diseases including Amyotrophic Lateral Sclerosis (ALS) and cancer, this thesis investigates the role of Ataxin-2 and its protein binding partners in stress granule development and its effects on various metabolic phenotypes of the cell. PAS kinase is a sensory protein kinase, conserved from yeast to man, which regulates respiration and lipid biosynthesis. Our lab discovered that PAS kinase phosphorylates and activates Ataxin-2 in yeast, and that PAS kinase overexpression enhances localization of Ataxin-2 to stress granules. Our preliminary results from yeast show that PAS kinase positively regulates stress granule formation in response to metabolic stress. Ataxin-2 normally functions to promote stress granule formation and it has been specifically shown to sequester and inhibit mammalian target of rapamycin complex I (mTORC1), a major player in the regulation of cell growth, to stress granules in both yeast and mammalian cells. To build upon this knowledge we performed a large-scale yeast interactome to identify Pbp1 binding partners through yeast-two hybrid and mass spectrometry. We identified 32 novel putative binding partners. A protein of note was Ptc6, a known regulator of mitophagy with human homolog PPM1K, which is not known to be involved in stress granules. Through colocalization with Ppb1 we determined that Ptc6 is sequestered to stress granules under glucose depravation. Under Pbp1 overexpression, Ptc6 was shown to increase localization to a stress granule marker, Pab1, showing that Pbp1 may be actively promoting Ptc6 to stress granules. We investigated the effects of eliminating Pbp1 and Ptc6 in yeast cells, including on mitophagy, mitochondrial quantification, whole cell respiration and mitochondrial reactive oxidative species. In a separate project, due to the outbreak of a worldwide pandemic and early concerns that currency could be a potential SARS-CoV-2 fomite, we investigated whether the virus could survive on varying types of currency. We conducted environmental studies and found no viable virus on bank notes or money cards. In vitro studies with live virus suggested SARS-CoV-2 was highly unstable on banknotes, however SARS-CoV-2 displayed increased stability on money cards with live virus detected after 48 hours.
