Vers une amélioration quantitative et qualitative de l'extraction des composés phénoliques du marc de raisin rouge à l'aide d'électrotechnologies / Towards a quantitative and qualitative enhancement of the extraction of phenolic compounds from red grape pomace using electrotechnologies

Ce travail porte sur l’amélioration quantitative et qualitative du procédé de fabrication d’extraits de marc de raisin riches en composés phénoliques. Les effets de technologies alternatives de traitement de la biomasse végétale sur l’amélioration des rendements d’extraction ont été étudiés à l’échelle laboratoire. Leurs impacts sur la qualité biochimique des extraits obtenus ont été évalués via la caractérisation analytique des composés phénoliques.Une étude comparative de différentes technologies alternatives de pré-traitement en milieux aqueux (décharges électriques haute tension (DEHT), champs électriques pulsés (CEP) et ultrasons (US)), d’un marc de raisin issu d’un procédé de vinification en rouge, en amont de la diffusion, a permis d’approfondir la compréhension des phénomènes induits et de leurs effets sur l’amélioration de l’extractabilité des composés. Nous avons montré que le choix du procédé à mettre en oeuvre était relatif au type de composés phénoliques ciblés : la localisation tissulaire de ces composés est apparue comme un facteur-clé pour optimiser leur extraction. La technologie de pré-traitement par CEP, induisant un endommagement localisé des structures végétales, s’est avérée particulièrement intéressante pour extraire sélectivement les anthocyanes situés au niveau des couches de cellules supérieures de l’hypoderme de la pellicule de raisin. Nous avons, par la suite, montré que le pré-traitement par CEP, en milieu sec (sans ajout de solvant) et à intensité de champ électrique modérée, était plus efficace pour la récupération sélective des anthocyanes et moins énergivore qu’un traitement CEP de forte intensité d’une suspension aqueuse de marc. Pour ce faire, nous avons mis en place une nouvelle configuration de cellule qui nous a permis d’étudier la faisabilité d’un pré-traitement en milieu sec, sans ajout de liquide conducteur, et d’optimiser les paramètres opératoires (masse volumique, intensité du champ électrique, énergie spécifique) influençant la récupération subséquente, lors de la diffusion, des composés phénoliques. Afin d’intégrer notre démarche expérimentale dans une approche systémique de type « bioraffinerie », l’accent a été mis sur le traitement du marc réhydraté à posteriori de la diffusion. L’intensification du pressage de ce marc par chauffage ohmique (CO) permet d’améliorer les rendements globaux d’extraction tout en assurant un premier niveau de déshydratation de cette biomasse.Un bilan technico-économique montre que l’application successive des électro-technologies (pré-traitement par CEP en milieu sec et pressage assisté par CO à posteriori de la diffusion) permet une amélioration des rendements massiques d’extraction de l’ordre de 30 % comparativement au procédé classique. Ce gain matière compense l’augmentation du coût de production (+ 2.7 % comparativement au procédé classique) et résulte une diminution du coût de production de l’extrait (7.3 €/kg vs 9.4 €/kg pour le procédé classique). / This work aims at studying the effect of alternative technologies for the processing of winery byproducts on the improvement of extraction yields at laboratory scale. Their impacts on the biochemical quality of the extracts obtained from grape pomace were evaluated by the characterization of the extracted compounds. A comparative study on the effects of different alternative pre-treatment (pulsed electric field (PEF), high voltage electric discharges (HVED) and ultrasounds (US)) of aqueous suspensions of red grape pomace, prior to the extraction step, was realized with the specific objective of understanding the induced phenomena. The electroporation induced by PEF may allow the specific recovery of anthocyanins that are located in the upper cell layers of the hypodermis by facilitating the solvent penetration to particular skin tissues of grape. The location of targeted compounds with respect to tissue structures seems to be a key issue to optimize their extraction. As a consequence, we subsequently examined the feasibility of a PEF pre-treatment of relatively low humidity grape pomace for the enhancement of bioactive compounds extraction. We demonstrated that a relatively dry pomace (RH ≈ 55 %) can be effectively treated by PEF after optimal densification using a suitable configuration of the PEF cell. The PEF pre-treatment of a densified fermented grape pomace, at moderate electric field strengths, requires less output current and lower specific energy and is also more efficient for the selective recovery of anthocyanins than Hi-PEF pre-treatment of an aqueous suspension of grape pomace. We finally focused on the treatment of rehydrated grape pomace following the diffusion step. The enhancement of pressing of the grape pomace by ohmic heating allowed the recovery of two fractions: a solid fibrous residue partly dehydrated which could be further valorized as an agro-material or in feeding, and an extract, particularly rich in bioactive components.The technical-economic assessment shows that the successive application of electro-technologies (pre-treatment of grape pomace in dry environment by PEF followed by a pressing step assisted by ohmic heating) lead to an improvement of extraction yields of about 30 % compared to the conventional method. This mass improvement offset the increased production costs (+ 2.7 % compared to the conventional process) by reducing the total production costs of the extract (7.3 €/kg vs 9.4 €/kg for the conventional process).

Estudo comparativo de características físico-químicas e nutricionais da soja preta e amarela / A comparative study of chemical and nutritional characteristics of black and yellow soybeans

O Brasil é o segundo maior produtor de soja (Glycine max, L.), sendo responsável por quase 30% da colheita mundial. Mais de 80% da produção total é destinada à extração de óleo e proteína para a alimentação humana, sendo que o consumo na forma de grão e de produtos industrializados vem crescendo, como alternativa a proteínas de origem animal e devido aos potenciais benefícios à saúde. Grande parte da literatura científica refere-se à soja amarela, enquanto estudos sobre soja preta ainda são escassos. Este projeto visou estudar e comparar as características físicas e de composição química, incluindo os compostos fenólicos e atividade antioxidante da soja preta e amarela, cultivadas no Brasil em condições climáticas e ambientais similares. Fez parte do escopo do projeto avaliar o efeito do cozimento sobre os compostos polifenólicos e a atividade antioxidante. Não foi observada diferença significativa no conteúdo de nutrientes e no perfil de ácidos graxos entre a soja preta e amarela, nem nas características físicas, como massa, capacidade de hidratação e tempo de cozimento dos grãos. Porém, a soja preta apresentou teores muito mais elevados de compostos fenólicos totais (CFT) e de flavonóides em relação à soja amarela. Os teores na soja preta foram, em média, de 4,78 mg eq. ácido gálico/g e de 1,75 mg eq. catequina/g, de CFT e de flavonoides,respectivamente. A soja amarela apresentou em média 40% a menos de CFT e aproximadamente 60% a menos de flavonóides. Ao contrário da soja amarela que é desprovida de antocianinas, na soja preta foi encontrado um teor médio de 0,92 mg eq. cianidina-3-O-glicosídeo/g. Empregando a técnica de HPLC-DAD-MS/MS foram identificadas duas antocianinas, a cianidina-3-O-glicosídeo e a peonidina-3-Oglicosídeo, além da cianidina. A soja preta apresentou atividade antioxidante 70% superior à soja amarela pelo método de DPPH e 50% superior, pelo método de ORAC. O cozimento dos grãos reduziu em pelo menos 40% o teor de compostos fenólicos e a atividade antioxidante, tanto na soja preta como na amarela. Foi observada uma forte correlação entre os teores de CFT, flavonóides e a atividade antioxidante, antes e após o cozimento. Assim, o consumo de soja preta ou mesmo a produção de novos alimentos a partir desses grãos poderá oferecer ao consumidor a possibilidade de diversificar a sua dieta, introduzindo um alimento com a mesma qualidade nutricional da soja amarela e, possivelmente, maiores benefícios à saúde. Vislumbra-se a possibilidade de expansão do cultivo da soja preta, inclusive por pequenos produtores, criando um novo nicho de mercado para um alimento diferenciado. / Brazil is the second largest producer of soybean (Glycine max, L.), accounting for almost 30% of the world\'s production. Over 80% of the total production is destined to oil extraction and protein for human consumption. Soybean and soy-product consumption has been rising as an alternative to animal protein and due to its potential health benefits. Most of the scientific literature refers to yellow soybeans, while studies on black soybeans are still scarce. This project aimed to study and compare black and yellow soybeans, cultivated in Brazil in similar climatic and environmental conditions, as to their physical characteristics and chemical composition, including phenolic compounds and antioxidant activity. The effect of cooking on phenolic compounds and antioxidant activity was also investigated. No significant difference was observed between black and yellow soybeans as to nutrient content, fatty acid composition and physical characteristics, such as seed-weight, hydration capacity and cooking time. However, black soybeans had a much higher content of total phenolic compounds (TPC) and flavonoids than yellow soybeans. TPC and flavonoid contents in black soybeans were 4.78 mg gallic acid equivalents/g and 1.75 mg (+)-catechin equivalents/g, respectively. Yellow soybeans had on average 40% less TPC and approximately 60% less flavonoids. Unlike yellow soybeans which lack anthocyanins, black soybeans were found to have an average content of 0.92 mg cyanidin-3-O-glucoside equivalents/g. High performance liquid chromatography coupled to photodiode array and mass spectrometry detectors (HPLCDADMS/MS) was used to identify two anthocyanins, namely cyanidin-3-Oglucoside and peonidin-3-O-glucoside, and cyanidin. Black soybeans also showed a 70% higher DPPH-free radical scavenging activity than yellow soybeans, and a 50% higher ORAC value. Cooking reduced at least 40% of phenolic compounds and antioxidant capacity in all soybean samples, both black and yellow. High correlations between phenolic compositions and antioxidant activities were observed, before and after cooking. These results suggest that black soybean consumption or new processed food products from these crops may offer consumers an opportunity to diversify their diet with food nutritionally equivalent to yellow soybeans, but possibly with greater health benefits. The possibility of expanding black soybean cultivation, including small-scale production, may create a new market niche for a value-added food product.

Nos frutos, a cor é fundamental para a aceitabilidade inicial do consumidor, especialmente para os chamados \"pequenos frutos\" como morangos e framboesas, sendo um critério tradicional para a apreciação e seleção do estado de maturação. Em framboesas (Rubus idaeus), as pesquisas têm focado a síntese de antocianinas, os pigmentos mais abundantes nesta espécie. Seus precursores provem da via de biossíntese dos fenilpropanóides, porém os fatores que regulam a atividade dessa via metabólica ainda são pouco conhecidos. Sabe-se que alguns hormônios como o etileno, o ácido indol-3-acético (AIA), o ácido abscísico (ABA) e o metil-jasmonato (MJ) estão relacionados à inibição da síntese ou aumento no acúmulo destes flavonóides. Neste contexto, o objetivo do trabalho é avaliar o efeito destes fatores hormonais sobre a biossíntese de antocianinas em framboesas da cultivar (cv.) Autumn Bliss. Foram avaliados os efeitos dos hormônios sobre as características físico-químicas e fisiológicas dos frutos (açúcares solúveis, ácidos orgânicos, respiração, síntese de etileno, cor), conteúdo de compostos fenólicos e flavonóides. Também foram quantificados os níveis de AIA livre, ABA e MJ e a expressão relativa das enzimas antocianidina sintase (ANS), dihidroflavonol redutase (DFR) e do fator de transcrição MYB10, regulador da transcrição de genes da via de biossíntese de antocianinas em framboesas. As alterações mais significativas foram observadas no tratamento com MJ, para o qual os frutos apresentaram acentuada produção de etileno (sem efeito sobre a respiração) e também na cor, mais acentuada nos frutos maduros deste grupo em comparação com os frutos do grupo controle. O MJ também induziu aumento nos níveis de expressão relativa de MYB, ANS e DFR, nos dois primeiros dias pós-colheita. Outro grupo que se destacou foi o tratado com AIA por apresentar inibição na síntese de antocianinas e compostos fenólicos, a qual foi correlacionada à inibição da transcrição do MYB e ANS. O ABA por sua vez, inibiu o acúmulo de transcritos da DFR, de forma correlata ao menor acúmulo de antocianinas. Exceto pelo ACC, que não produziu efeitos significativos em nenhum dos parâmetros avaliados, os resultados indicam que o metabolismo de antocianinas deve ser regulado por múltiplos sinais hormonais. / The color is fundamental for the fruits, for the initial acceptability of the consumers, is a factor that influence the appreciation and selection of the maturity stage, especially for berries, like strawberries and raspberries. In raspberries (Rubus idaeus), the research have focused on the anthocyanin biosynthesis, the pigments most frequent on this species. Their precursors came from the phenylpropanoid pathway but the factors that regulate this metabolic pathway remain unclear. There are evidences that some phytohormones as ethylene, indole-3-acetic acid (IAA) abscisic acid (ABA) and methyl jasmonate (MJ) are related to the inhibition or increase of these flavonoids on fruits. In this context, the aim of this study is to evaluate the effect of these phytohormones on the anthocyanin pathway in raspberries Autumn Bliss. It was evaluated the effects of these phytohormones in some physico-chemical and physiologic characteristics of these fruits (soluble sugars, organic acids, respiration, ethylene biosynthesis, color), phenolic compounds and flavonoid content. Also, it was quantified the auxin, ABA and MJ levels, and the relative transcript levels of anthocyanidin synthase (ANS), dihydroflavonol 4-reductase (DFR) and the transcriptional factor MYB10, a transcriptional regulator of the anthocyanin pathway in raspberries. The most significant alterations were observed on MJ treatment, in which the ethylene production in berries reach the highest levels (without effect on the respiration) and the most intense red color on ripe fruits in comparison with the control group. MJ also induced the increase on relative expression of MYB, ANS and DFR two days after harvest prior to the anthocyanin accumulation. Other noticeable effect was detected in the IAA group in which anthocyanin synthesis and phenolic compound content were inhibited and related to the inhibition of MYB and ANS transcription. ABA inhibited the DFR transcript accumulation, related to the delay in anthocyanins synthesis in relation to control group. Except by ACC, that did not have significant effects in any of the evaluated parameters, the results suggest that the anthocyanin metabolism might be regulate for multiple hormonal signs.

Zisk a komplexní charakterizace extraktů aronie / Obtaining and complex charaterization of Aronia spp. extracts

Aronia (Aronia melanocarpa) is a berry fruit with distinctive sensory characteristics and health-promoting properties. Polyphenols are the main bioactive compounds found in aronia including natural pigments – anthocyanins. Bioactive compounds are usually obtained from natural materials by extraction, in this work, two methods of extraction were compared – maceration and PHWE. Based on the total phenolic content, maceration was chosen as a more suitable method and then was optimized with statistical model – Design of Experiment. Optimal conditions were set to temperature of 30 °C, extraction agent 50 % ethanol, solid-solvent ratio 10 g per 50 ml and time of extraction for 30 minutes. Extract obtained under these conditions was characterised by total phenolic content – 1441 ± 90 mg/100 g DW, total anthocyanin content 943 ± 8 mg/100 g DW and antioxidation activity by ABTS 24,78 ± 0,09 molTE/g. In total of 25 volatile compounds were indentified in the optimal extract with content > 0,5 %. Major part created aldehydes and esters, with the main compounds being benzaldehyde (32,25 %) and methyl hexanoate (21,37 %).

Anthokyanová barviva ve vybraném ovoci / Anthocyan pigments in selected fruits

This diploma thesis is concerned with determination anthocyanins dyes in elderberry (Sambucus nigra L.). In theoretical part there is described general characterization of the plant, its chemical composition, short view of elderberry breeding and possibilities of usage in food and medical industry. Bigger attention is paid to anthocyanins dyes. There is described their characterization, occurrence, factors that have an impact on their stability. Biosynhesis of the pigments and metabolism, benefit effects and their usage in food chemistry are then shortly sumarized. Methods of determination of anthocyanins are discused too. Next chapter deals with high performance liquid chromatography. At the end of theoretical part, there is sumarized findings of anthocyan dyes analysis by expert scientists in various matrix. In experimental part, total of 16 species of elderberry were analyzed. These species are from research and breeding institute of pomology in Holovousy. The dyes were identification and quantification by high performace liquid chromatography coupled with a diode array detector. Detection wavelenght was 520 nm. Gradient of mobile phase was optimilized because of shortening analyses time and resolution improvement. Four dyes were identified by HPLC: cyanidin-3-sambubioside, cyanidin-3-glukoside, cyanidin-3,5-diglukoside a cyanidin-3-sambubioside-5-glukoside. Total amount of anthocyanins ranged from 33627167,0 to 5525284,0 mAU•min-1. The most anthocyanins dyes were determined in Mammut species (33627167,0 mAU•min-1), the lowest amount was measured in Aurea species (5525284,0 mAU•min-1). The most abundant anthocyanin in all species of eldeberry fruit except Haschebrg and Aurea species was cyanidin-3-sambubioside. The concentrations of cynidin-3-glucoside in all of the berries were quantified using regression equation of calibration curve. Mammut species had the most amount of this anthocyan (0,486 mg•ml-1) and the lowest amount had Aurea species (0,033 mg•ml-1).

Diploma thesis deals with the new varieties of gooseberries. The theoretical part characterizes gooseberry from a biological point of view, what is the content of healthful nutrients and states food technology aspects. Furthermore, the theoretical part deals with the main parameters examined nutrition: vitamin C, anthokyanům and antioxidant activity. In the experimental part was measured both weight solids content and soluble solids. Vitamin C was measured by HPLC, anthocyanins using a pH differential method and for antioxidant activity determination was selected method by Kanedy known also as DPPH.

Charakterizace plodů aronie (Aronia melanocarpa) a muchovníku (Amelanchier alnifolia) / Characterisation of chokeberry and saskatoon berry fruits

Diploma thesis deals with the characterization of chokeberry (Aronia melanocarpa) and saskatoon berry (Amelanchier alnifolia) fruits. The theoretical part focuses on the botanical classification, chemical composition, curative effects and the use of chokeberry and saskatoon berry in the food industry. Further, the attention is also devoted to a review of methods for the determination of important antioxidants, specialized on the determination of polyphenols, anthocyanins, ascorbic acid and total antioxidant activity. The experimental part of the work deals with the determination of selected parameters in different varieties of chokeberry and serviceberry, harvested in 2012 and 2013 using spectroscopic and chromatographic techniques with an emphasis on effect of solvent (50% ethanol, destilled water, DMSO). Specifically total content of polyphenols and anthocyanins, color characteristics using the CIE L*a*b* system and the ability of the sample to reduce the iron are determined using the UV-VIS-NIR spectroscopy. Further ascorbic acid concentration in individual extracts is quantified by High performance liquid chromatography coupled with a diode array detector and the total antioxidant activity is characterized by electron paramagnetic resonance involving •DPPH and ABTS•+ assays.

Impacts de l'oxygène sur les évolutions chimiques et sensorielles du vin rouge / Oxygen impacts on chemical and sensory evolutions in red wine

L’impact de l’oxygène a été suivi pendant 4 mois pour 3 vins de Merlot d’IPT 61, 63 et 76. Le vin d’IPT 63 était enrichi par une fraction d’ellagitannins de bois de chêne. Les vins ont été ajustés à pH 3, pH 3,5 et pH 4. Pour chaque pH, des doses de 0, 2, 8 et 20 mg/L d’oxygène ont été fournies à l’aide d’oxygène pur, puis des analyses mensuelles ont permis de suivre les évolutions chimiques et sensorielles des vins afin d’apprécier les impacts liés à l’oxygène.Les résultats montrent pour tous les vins, que la consommation de l’oxygène augmente de 2 à 4 fois plus lorsque l’on passe de pH 3 à pH 4. La présence d’ellagitannins dans le vin favorise la consommation d’oxygène, ainsi qu’en solutions modèles hydro-alcooliques de fractions phénoliques. Les teneurs en tannins totaux sont globalement stables. Les polymérisations analysées par la méthode DMACH se produisent majoritairement à pH 3 et pH 3,5 pour le vin d’IPT 61, alors que les différences liées à l’oxygène sont moins fortes sur le vin d’IPT 76. Pour le vin d’IPT 63 + ellagitannins, les polymérisations sont favorisées à pH 3. Les teneurs en anthocyanes totales et moléculaires montrent généralement des diminutions en fonction du temps, de l’oxygène ainsi que de l’acidité. Pour le vin d’IPT 76 et d’IPT 63 + ellagitannins, les anthocyanes se polymérisent davantage aux tannins polymères en présence d’oxygène, et à pH plus acide. 13 pyranoanthocyanes ont pu être identifiées et quantifiées, dont deux molécules de pyranomalvidine-di-(épi)catéchine quantifiées pour la première fois dans des vins rouges secs qui peuvent être considérées comme des marqueurs en lien avec l'apport en oxygène subi par le vin. Les analyses sensorielles réalisées sur le vin d’IPT 76 en fonction des pH montrent que l’amertume est plus forte à pH 4, et pH 3,5 dans une moindre mesure. Cette tendance se retrouve aussi sur la perception d’oxydation.Pour les deux autres vins, d’IPT 61 et d’IPT 63 enrichi par une fraction d’ellagitannins, l’intensité du fruité diminue globalement avec les doses d’oxygène, tandis que le caractère oxydé est bien corrélée avec les doses croissantes d’oxygène, ainsi que l’amertume dans une moindre mesure. / The impact of oxygen has been followed-up during 4 months for 3 Merlot red wines of TPI 61, 63 anf 76. The wine of TPI 63 was supplied by a fraction of ellagitannin from oak wood. Wines have been adjusted at pH 3, pH 3.5 and pH 4. For each pH, quantities of 0, 2, 8 and 20 mg/L of pure oxygen have been added. Then, anlyses have been performed each month to follow the wine chemical and sensory evolutions in order to determine the impact of oxygen.For all wines, the results show that oxygen consumption is 2 to 4 fold higher from pH 3 to pH 4. The presence of ellagitannin in wine favors oxygen consumption, as well as in wine model solutions with phenolic fractions. Total tannin contents remain quite constant. Polymerisations analysed by DMACH index occur mainly at pH 3 and pH 3.5 for the wine of TPI 61, while the differences related to oxygen are less important in the wine of TPI 76. For the wine of TPI 63 + ellagitannins, polymerisations are favored at pH 3. Total and molecular anthocyanins contents generally show a decrease according to time, oxygen and acidity. For the wines of TPI 76 and TPI 63 + ellagitannins, the anthocyanins polymerise more to polymers of tannins with oxygen and acidity. 13 pyranoanthocyanins have been identified et quantified. Among them, two molecules of pyranomalvidin-di-(epi)catechin quantified for the first time in a dry red wine. They can be considerated as marker related to oxygen amount consumed by wine. Sensory analyses realized on wine of TPI 76 according to pH show that bitterness is higher at pH 4, and pH 3.5 in a lesser extent. There is the same trend with the oxidation perception.For the two other wines, of TPI 61 and 63 with ellagitannin fraction, fruity intensity globally decreases with oxygen quantities, while the oxidized parameter is well related to increasing oxygen amounts, as wella s bitterness in a lesser extent.

Total of 17 varietes of gooseberries, which come from growers Vondráček and the Research and Breeding Institute of Pomology Holovousy Ltd. were analyzed in the experimental part. pH differential method was used to determination total monomeric anthocyanins. Total content of polyphenols were determined by using the Folin-Ciocalteu reagent. Next was determined content of vitamin C in all varieties by HPLC and was evaluated the total antioxidant aktivity using radical DPPH. In conclusion, individual varieties of gooseberries were compared by the contents of the determined chemical parameters.
