Estrutura e composição da comunidade de tricladidos em áreas com alto grau de impacto antrópico do sul do Brasil

Para o Rio Grande do Sul, há registros de espécies que podem ser utilizadas como indicadoras de áreas preservadas ou alteradas, mas os trabalhos anteriores têm se concentrado em unidades de conservação, contendo áreas relativamente extensas de floresta nativa. O presente trabalho buscou analisar a estrutura e a composição, ao longo de um ciclo anual, da comunidade de tricladidos terrestres em áreas de mata secundária e áreas ajardinadas do Campus da Universidade do Vale do Rio dos Sinos - UNISINOS, em São Leopoldo, RS. Foram selecionadas quatro áreas de mata secundária e quatro ajardinadas, onde se realizaram 12 amostragens. Para cada área foram demarcados dois transectos de 100m de comprimento, sendo selecionados aleatoriamente dez pontos ao longo de cada transecto. Em cada ponto, foi utilizada uma parcela de 4m², com esforço amostral de 10 minut / Land planarians are bioindicators of the degree of anthropization and preservation of their habitats and areas of high general biodiversity. Some species which occur in Rio Grande do Sul could be used as bioindicators of preserved or altered areas, but the studies have been restricted to legally protected areas showing relatively large fragments of native forest. This paper aims at analysing the structure and composition of land planarian community in areas of secondary forests and grassland of the Universidade do Vale do Rio dos Sinos Campus - UNISINOS, in São Leopoldo, RS along an annual cycle. Four areas of secondary forest and four grassland areas were selected, where 12 samplings were carried out. For each area two 100m-long transects had their boundaries set, where ten points along each transect were randomly chosen. Invasões biológicas representam hoje a segunda maior ameaça à biodiversidade, e o homem desempenha papel fundamental na introdução de espécies exóticas potencialmente invasoras. O cachorro (Canis lupus familiaris) é uma dessas espécies. Presente em todos os continentes, é o carnívoro mais abundante do mundo, e pode causar impactos à fauna nativa através de efeitos letais e não letais da predação, competição, hibridização e transmissão de doenças, além de ser potencialmente importante na epidemiologia de zoonoses. Em áreas rurais, que concentram grande parte dos remanescentes de vegetação nativa no mundo, os cachorros são frequentemente criados soltos e circulam livremente, intensificando a chance de contato com a fauna nativa. Nesse trabalho, avaliamos a importância relativa de fatores associados a atributos individuais (sexo, idade, condição de saúde, comportamento exploratório, tamanho e raça), formas de manejo (incentivo do dono à movimentação, motivo para a criação, confinamento e frequência de alimentação) e contexto ambiental (proximidade à mata nativa) para determinar a chance de cachorros visitarem remanescentes florestais em paisagens fragmentadas de Mata Atlântica. Selecionamos oito paisagens de 2830 ha cada, nas quais visitamos todas as construções em áreas rurais a fim de entrevistar, através da aplicação de questionário, os responsáveis pela criação de cachorros e fotografar os cachorros. Utilizamos a imputação múltipla para estimar os dados faltantes (comuns em dados obtidos via questionários), gerando 10 conjuntos de dados imputados que foram analisados separadamente. Por meio de seleções de modelos através do Critério de Informação de Akaike, comparamos modelos candidatos com até cinco variáveis independentes para determinar a chance de cachorros visitarem remanescentes florestais. Nossos resultados nos permitem afirmar que quatro fatores - dois associados a atributos individuais e dois associados a formas de manejo - atuam em conjunto para determinar a chance de cachorros visitarem remanescentes florestais em paisagens rurais da Mata Atlântica. Cachorros maiores, mais exploradores, que recebem mais incentivo a se movimentar e que passam menos tempo confinados têm maior chance de visitar remanescentes florestais. A maior importância de atributos individuais e da forma de manejo está de acordo com a grande variação fenotípica existente entre cachorros e a variedade de modos como são manejados. Entre os atributos individuais, tanto características físicas como comportamentais são importantes, enquanto os aspectos chave da forma de manejo são aqueles mais diretamente relacionados à movimentação dos cachorros. Independentemente do tempo de confinamento, o incentivo do dono à movimentação do cachorro, em particular o estímulo para que o cachorro o acompanhe em visitas a remanescentes florestais, é fundamental. O contexto ambiental, em especial a proximidade do domicílio a áreas de mata nativa, por sua vez, é irrelevante dada a alta mobilidade dos cachorros. Nossos dados sugerem que a densidade de cachorros em paisagens rurais de Mata Atlântica é uma ordem de magnitude mais alta do que a de carnívoros de médio porte silvestres relativamente comuns, que muitos cachorros já tiveram contato direto com espécies silvestres, e que a vacinação e outras medidas profiláticas são relativamente incomuns. Todos esses dados indicam o potencial de efeitos negativos, tanto para a fauna silvestre como para o homem. The dog (Canis lupus familiaris) is one of such species. Distributed across all continents, the dog is the most abundant carnivore in the planet, and can impact wildlife through lethal and non-lethal effects of predation, competition, hybridization and disease transmission, besides being potentially important in the epidemiology of zoonoses. In rural areas where most remnants of native vegetation around the world are concentrated, most dogs are free-ranging, enhancing the chance of interactions with wildlife. Here, we evaluated the relative importance of factors associated with individual traits (sex, age, health condition, exploratory behaviour, size and breed), management (owner\'s incentive to movement, motive for raising, confinement and feeding frequency), and environmental context (proximity to native forest) to determine the chance of dogs visiting forest remnants in Atlantic Forest fragmented landscapes. We selected eight landscapes of 2830 ha each, where we visited all constructions in rural areas in order to interview dog owners by applying a questionnaire and to photograph dogs. We used multiple imputation to estimate missing data (common in data obtained through questionnaires), obtaining 10 imputed datasets that were analyzed separately. We compared candidate models with up to five independent variables to determine the chance of dogs visiting forest remnants through the Akaike Information Criterion. Our results indicate that four factors - two associated with individual traits and two associated with management - work together to determine the chance of dogs visiting forest remnants in Atlantic Forest rural landscapes. Larger dogs, and those that exhibit exploratory behaviour, are more stimulated to move or confined for shorter periods have greater chance of visiting forest remnants. The greater importance of factors associated with individual traits and management is in accordance with the ample phenotypic variation among dogs and the variety of ways they can be managed. Among individual traits, both morphological and behaviour characteristics are important, whereas the key aspects of management are those directly related to dog movement. Irrespective of the time confined, the owner\'s incentive to movement, in particular taking the dog to the forest, is a crucial aspect. In contrast, the environmental context, especially the proximity of the household to native forest, is irrelevant given the vagility of dogs. Our results suggest that the density of dogs across rural landscapes in the Atlantic forest is at least one order of magnitude higher than the density of relatively common medium-sized native carnivores, that several dogs have already had direct contact with wild species, and that vaccination and other prophylactic measures are relatively uncommon. All of these highlight the potential for negative effects on both wildlife and human population. Plans to reduce these effects should include not only veterinarian aspects, such as the expansion of public prophylactic campaigns beyond rabies vaccination, but also social aspects, such as the dissemination of information on the problems caused by dogs visiting native vegetation, aiming at changing people\'s beliefs, attitudes and behaviour

Les sols anthropisés, incubateurs d'agents bactériens pathogènes de l'homme : typage génétique, métabolique et antibio-résistance d'agents opportunistes / Human-impacted soils as bacterial pathogen reservoir : genotyping, metabolic properties and antibiotic resistance of infectious agents

Les bactéries pathogènes opportunistes de l'Homme (bpo) sont retrouvées dans le milieu hospitalier où elles sont responsables d'infections nosocomiales ainsi que dans les milieux naturels terrestres et aquatiques. Elles présentent souvent des résistances intrinsèques aux antibiotiques élevées. En milieu clinique, l'usage intensif d'antibiotiques peut conduire à l'émergence de souches dites « Multi Drug Resistant ». L'anthropisation des milieux naturels peut également influencer la prévalence et les propriétés de résistance des bpo. Mes travaux ont porté sur l'impact de l'épandage d'amendements organiques sur la prévalence de bpo dans les sols, leur diversité génétique et leurs propriétés de résistance aux antibiotiques. Une étude des espèces Stenotrophomonas maltophilia, Pseudomonas aeruginosa et Burkholderia du « cepacia complexe » (Bcc) réalisée sur des sites du Burkina-Faso amendés ou non en déchets urbains bruts a mis en évidence des différences dans les propriétés de résistance des 3 modèles. S. maltophila présente fréquemment des phénotypes MDR contrairement à P. aeruginosa et aux Bcc. Une approche de génomique comparative entre souches de S. maltophilia d'origine environnementale ou clinique et de phénotypes sensibles à MDR a été réalisée afin d'élucider l'origine génétique de l'hétérogénéité des phénotypes de résistance. Une variation dans le contenu en pompes à efflux et la présence de pompes souche spécifique chez des souches environnementales ont été observées. L'étude de l'expression d'une de ces pompes confirme son implication dans la résistance aux antibiotiques et dans l'adaptation à des paramètres environnementaux tels que la température / Opportunistic bacterial pathogens (obp) of Man are found in hospital setting where they are responsible for nosocomial infections as well as in terrestrial and aquatic natural environments. Obp often show high intrinsic antibiotic resistance level. Moreover, the intensive use of antibiotics in clinical settings can lead to the emergence of "Multi Drug Resistant" strains. The anthropisation of the natural environment leads to modifications in bacterial diversity of these environments and can affect the prevalence and the antibiotic resistance properties of obp. My research focused on the impact of organic amendments on the prevalence, genetic diversity and antibiotic resistance properties of obp. A study on the species Stenotrophomonas maltophilia, Pseudomonas aeruginosa and the “Burkholderia cepacia complex" (Bcc) was conducted on sites in Burkina Faso amended or not with raw urban wastes. This study showed differences in antibiotic resistance properties between the 3 models. S. maltophila frequently showed MDR phenotypes unlike P. aeruginosa and Bcc. A comparative genomics study between S. maltophilia strains from environmental or clinical origin showing sensitive or MDR phenotypes was performed to elucidate the genetic origins of heterogeneity in the resistance phenotypes. A variation in the efflux pumps content was observed between strains. The expression of an efflux pump specific to an environmental MDR strain was then evaluated and confirmed its likely involvement in antibiotic resistance and adaptation to environmental parameters such as temperature

L’objectif principal de la thèse était de caractériser les impacts de différents types de gestion pastorale (bovins, équins, caprins) sur les communautés végétales de « nouveaux écosystèmes » fortement anthropisés que sont les digues et remblais de la réserve de Donzère-Mondragon dans la basse vallée du Rhône. Différents outils ont été utilisés (relevés de végétation, analyses de sols, analyses minérales des herbages, télédétection, SIG, etc.) à des échelles spatiales différentes afin de dégager les interrelations entre les différents niveaux d’organisation et fonctionnalités de la végétation. En effet, l’émergence des « nouveaux écosystèmes » oblige les gestionnaires à reconsidérer les principes initiaux de la gestion conservatoire. Dans notre étude, il a pu être démontré que les systèmes de pâturage faisant appel à des troupeaux d’herbivores dont l’organisation est adaptée pour maintenir l’ouverture des milieux, augmentent la richesse spécifique, la diversité et l’hétérogénéité de la végétation à court terme (3-5 années) par rapport à l’absence de gestion ou aux interventions mécaniques. Les analyses montrent également que les espèces annuelles qui apparaissent sont appétentes et contribuent à augmenter la valeur fourragère de certains faciès du site pâturé par les chevaux. Ces herbivores ont également un impact sur la banque de graines permanente du sol en augmentant son hétérogénéité et la densité de graines viables sous les faciès les plus pâturés. Le pâturage des vaches et des chevaux confirme une corrélation entre l’indice de végétation (NDVI) obtenu après l’analyse de photographies aériennes, la biomasse et la richesse en espèces végétales. Les estimations d’indices de consommation de la ronce sous SIG ont permis d’apprécier l’efficacité de l’action des chèvres. Ces différents résultats, pour les modalités testées, correspondent bien à ceux déjà obtenus lors d’applications de systèmes pastoraux en milieux naturels. Des expérimentations devraient également être menées à l’avenir via des systèmes de pâturage mixtes notamment bovin-caprin et équin-caprin pour accroître l’effet de la restauration aux dépens de la colonisation d’arbustes compétitifs dont la ronce. La gestion pastorale appliquée devrait néanmoins être pensée sur le moyen terme, si possible via des contrats pluriannuels, favorisant la durabilité de cette méthode de gestion et de ses effets dans le temps. / The main objective of the thesis was to characterize the impacts of different types of pastoral management (cattle, horses, goats) on "new ecosystems" plant communities. Our experimentations were conducted in highly anthropized dykes and embankments of the Donzère-Mondragon reserve in the lower Rhone valley. Various tools were used (i.e. vegetation surveys, soil analyzes, grassland mineral analyzes, remote sensing, GIS, etc.), and at different spatial scales in order to identify the interrelationships between the different levels of vegetation organization and functions. Indeed, the emergence of "new ecosystems" requires managers to reconsider the initial principles of conservation management. Our study showed that, in the short term (3-5 years), grazing systems using herds of herbivores, whose organization is adapted to maintain open habitats, increase species richness, diversity and heterogeneity of plant communities compared to lack of management or mechanical interventions. Moreover, the annual species that appear are appetizing and contribute to increase the forage value of the site grazed by horses. These herbivores also have an impact on the permanent seed bank by increasing both its heterogeneity and the density of viable seeds under the most grazed patches. Grazing of cattle and horses confirm a correlation between the vegetation index (NDVI), biomass and plant species richness. Finally, we evaluated the efficacy of goats’ action on bramble via consumption indices calculated using GIS. Our different results are in the line with those already obtained in natural environments. Future experiments should test the effects of mixed grazing systems, particularly cattle, goats and horses, on the colonization dynamic of competitive shrubs such as brambles. Applied pastoral management should be considered in the medium term, via multi-year contracts to promote the sustainability of this management method and its effects over time.

Fonctionnement et évolution pédogénétiques de Technosols issus d'un procédé de construction de sol / Functionning and pedogenic evolution of technosols coming from a soil contruction process

La restauration des fonctions des sols est une étape clé de la requalification des sites dégradés par les activités humaines. Ces opérations nécessitent de disposer de procédés de remise en état du couvert pédologique favorisant la résilience écologique. Ils génèrent de nouveaux sols dont il faut prévoir l’évolution. Notre objectif est de caractériser le fonctionnement et la pédogenèse de sols construits à l’aide d’un nouveau procédé fondé sur la formulation de déchets et sous-produits (terre industrielle traitée, sous-produit papetier, compost). Dans ce but, des colonnes en laboratoire ainsi que des parcelles lysimétriques in situ ont été mises en place. Des paramètres descriptifs des fonctionnalités et de la pédogenèse des sols construits ont été mesurés. Les résultats ont montré que les matériaux parents présentent des propriétés aptes à remplir des fonctions comme la fourniture d’éléments ou la rétention en eau. Les sols construits remplissent les rôles de support de végétation, d’échange/filtre et de support de la biodiversité comme des sols naturels. Les premiers stades de leur évolution pédogénétique se caractérisent par leur intensité, leur rapidité et leur caractère non habituel (e.g. dissolution conjointe de gypse et de calcite). La prospective sur l’évolution des sols construits démontre que, de Technosols, ils évoluent vers des types de sols analogues aux sols naturels. Ce cheminement nous conduit à proposer une évolution de la classification des sols très anthropisés sur des critères génétiques. Enfin, les bases de la modélisation de la pédogenèse des sols très anthropisés sont posées ainsi que celles d’un outil d’aide à la décision pour le génie pédologique / Human activities induce growing impacts on soils. This has produced derelict lands that need thorough reclamation to minimize their negative effect on the environment. A better knowledge and control of the evolution of highly anthropized soils is needed in order to achieve a sustainable management of these sites. This work emphasizes on both the development of a process of soil construction with wastes and by-products (paper sludge, treated industrial soil, compost) and the study of the functioning and pedogenic evolution of the constructed soils. The project relies on both in situ pilot scale application and lab-scale experiments. The characterization of the parent materials and their compounding highlighted their properties and their originalities compared to geological or pedological materials. The study of the functioning of constructed soils demonstrated that they could fulfil the soil basic functions. In this way, the process has been able to achieve the reclamation of the pedological cover and initiate the ecological resilience. The nature, the intensity and the kinetics of the pedogenic processes have been identified. It appeared that the reactions were very fast and that they sometimes strongly differ from the natural local pedo-climate (e.g. dissolution of gypsum, amorphous silica). We suggest that they will spontanuously evolve to analogous natural form of soils. Our results are confronted with the present classification of the Technosols and some propositions are made in favour of a more genetic-oriented way. Some thoughts about the modelling of the pedogenesis of very anthropized soils are proposed. At last, this work evocates the basis for the development of a decision tool for the pedo-engineering approach.
