Mikrobiální aktivity půdy ovlivněné různou mírou kontaminace ropnými uhlovodíky

Bioremediation is a method of reviving enviroment which is using natural processes. These processes, the modern technology is trying to make faster and more effective. This diploma thesis deals with a theme about Microbial Activity of Soil influenced by different measurements of crude oil hydrocarbon contamination and observing of microbial consorcia activity in contamined, not contamined and sterile soil. The initial chapter deals with crude oil contamination and bacterial metabolism which is able to remove this contamination. There was executed a container trial in the experimental part of this thesis. The plants were planted into different types of adjusted soil (crude oil application, sterilization and so on). There was compared a production of biomass and there are a few conclusion from these results. The elementary thing is that the soil micro-organisms which appear in oil soil can make a life strategy in this enviroment and can also prosper which reflects on the production of biomass.The container trial was determined as the most exact method of soil activity valuation because it is nearest to the real soil proportion. Followingly there were executed analysis of store soil and analysis of so after finishng of the container trial. These results brought similar conclusions, however the cultivation on nutrient soil and the cultivation like that it is not comparable. The store soil underwent a water cress trial. This trial confirmed toxic effects of crude oil but it also showed the fact that crude oil is a natural substance and micro-organisms can adapt to it. The mineralization of soil was measured with help of ionic measurers.

Hodnocení vlivu doplňkových závlah s přídavkem bioaditiv v kulturách zeleniny

The aim of this study was to evaluate the influence of the bioaditivs on the pepper (Capsicum annuum). For a given experiment were selected products based on bacteria strains , exactly : Azospirillum brasilense K2012 242/9 , Azospirillum largimobile B41 K2012, K2009 Leuconostoc mesenteroides 25/4 , Pseudomonas chlororaphis K2009 13/4, Pseudomonas lundensis K2009 9/4-B and algal bio-preparation R49S/2011 companies Rawat . Furthermore, the variant treated with arbuscular fungi , the inoculum was composed of Glomus intraradices BEG140, Glomus claroideum BEG210 , Glomus microaggregatum BEG56, Glomus intraradices S7, Glomus versiforme a saprophytic fungus Trichoderma harzianum. Next experiment contained a variant treated with phenyl and seven untreated control variant. The aim of this study was to evaluate the influence of the additives, namely the action of bacterial additives on the yield and nutritional parameters. The entire experiment was carried out in the Slovak village Závod. The results were analyzed statistically using programme STATISTICA 12. Based on the obtained results, it is evident that the variant algae + bacteria was found to increase revenues , compared to control treatment. The highest yield was achieved in the variations of Mycorrhiza + Trichoderma. The nutritional parameters were determined total antioxidant capacity in fruits, vitamin C and dry matter content . High levels of vitamin C were achieved in variants Phenyl 7, at least variants Algae + bacteria between these variants was statistically significant difference of almost 50 %. Total antioxidant capacity was clearly highest in control untreatment variant. Among the variants and Algae Control + bacteria was 37 %. In determining the content of dry matter was found that a minimum content of dry matter showed variant Algae + bacteria, 4.75%.

Isolace mikroorganismů z mlék a mléčných výrobků schopných tvořit biogenní aminy

The aim of the dissertation thesis, The Isolation of Microorganisms Producing Biogenic Amines from Milks and Dairy Products, was to isolate pure bacterial cultures of the genus Enterococcus and coliform bacteria from milks and cheeses and to test them for ability to produce biogenic amines. Isolated strains were identified and decarboxylase activity was tested on several amino-acids (lysine, histidine, tyrosine, tryptamine, ornithine, phenylalanine and arginine) at four different temperatures. At the end of the experiment, PCR was processed for the presence of tyrosine decarboxylase gene in enterococci strains. Identification showed higher variability of microorganisms isolated from cheeses rather than from milks. Decarboxylation activity test has revealed ability of genus Enterococcus to decarboxylate tyrosin and arginine. Coliform bacteria were found to be possibly able to decarboxylate arginine, tyrosin, ornithine, lysine and histidine. The subsequent PCR reaction confirmed most of the enterococci isolates to be producers of tyramine.

Studies on Antimicrobial Resistance in Pathogenic Bacteria

Bacterial infections caused by pathogenic microorganisms are classified among one of the major global health problems. In recent decades, bacterial resistance to antimicrobials has been seen as a major problem in the human and veterinary fields. The dissertation thesis is focused on the development and evaluation of the biological activity of antimicrobial agents, followed by describing the mechanism of antibiotic resistance from a molecularly microbiologically aspects. In the experimental part the nanomaterials, antimicrobial peptides, antibiotics and also the composites from these materials have been used for development of new antimicrobial agents and subsequently tested for their inhibitory effect on bacterial growth. The synthesized antimicrobials were physically and chemically characterized and their biological activity was established by basic microbiological and molecular methods. The results show that the produced composites can overcome existing drug resistance mechanisms, show excellent biocompatibility, and can be used in clinical practice in the future.

Ověření vlivu aplikace přípravků na bakteriální bázi pro ovocné dřeviny

The diploma thesis „The Effect of Application of Bacterial Preparations on Fruit Trees“ compares quantitative and qualitative properties of apricots treated and nontreated with bacterial praparations. The theoretical part of the thesis is divided into three parts; the first is focused on description of the species apricot (Prunus armeniaca L.), the next part deals with nutrition of fruit trees. The last part focuses on importance of soil microorganisms, especially PGPR bacteria, for plants. PGPR bacteria is group of plant growth promoting bacteria. At the end of the theoretical part there is a description of the most important genera of plant growth promoting bacteria. The practical part focuses on application of bacterial preparations in an apricot orchard. Variants with bacterial applications were compared to control variants in yield, size and weight of apricot, analytical evaluation of fruits (content of elements, vitamin C, flavonoids and antioxidant capacities) and monitoring of storage parametres, such as weight loss, content of refractometric dry matter, total titratable acidity and fruit firmness.
