Materiálový tisk ozonových dozimetrů / Material printing of ozone dosimeters

This study presents various methods of measuring ozone and consolidates current state of knowledge in the field of disposable printed ozone dosimeters. The paper also identifies prospective dyes for preparation of ozone dosimeters available on the market. Information gathered from literature review was used to prepare dosimeters based on two different dyes. These were calibrated for screen printing technology and their sensitivity to ozone was optimized to achieve visual detectability. Prepared samples were observed and studied while being exposed to ozone. Then, long term mechanical and chemical properties of these were tested in order to identify appropriate conditions for storage.

This diploma thesis deals with ozone measurement methods, especially focusing on opto-chemical methods, when the color change of the sample is examined. Moreover it deals with dyes that are sensitive to ozone and their degradation kinetics. In the experimental part kinetics of dyes Orange I, Orange II and Indigo carmine is examined, These dyes are prepared in the form of ink of different composition. These inks are applied to diverse substrate materials by the material printing method, further by using a Baker film aplicator and in the end by the screen printing method. The properties of the samples are evaluated as well as factors affecting their degradation.

Práce podává přehled o dostupných a využívaných metodách, normách pro testování a hodnocení světlostálosti barevných fotografií a tisků. Bylo provedeno porovnání jednotlivých platných norem a návrhů na hodnocení. Tato práce pojednává o ochraně inkjetových tisků pomocí lakování. Byl připraven lak kompatibilní s fotografickými médii. Připravená kompozice byla obohacena o UV absorbéry EVERSORB ve dvou koncentracích. Pro srovnání byly také testovány tři druhy komerčních ochranných fotografických laků. Výtisky se třemi barvivovými inkousty opatřené lakovou vrstvou, byly vystaveny jak urychlenému světelnému stárnutí v xenonové testovací komoře, tak urychlenému stárnutí v ozonové testovací komoře. Na základě měřených odrazových spekter byl vypočítaný objem barvového gamutu pomocí softwaru Volga. Byl sledován úbytek gamutů v průběhu světelného stárnutí i při expozici ozonem. Stárnutí vzorků probíhalo do ukončení testu, nebo do dosažení kritéria 30% úbytku objemu barvového gamutu. Také bylo pozorováno selhání recipročního zákona, které ukazuje na jistou nepřesnost při předpovědi dlouhé životnosti vzorků. Pomocí změny barvového gamutu byla posuzována ochrana připraveného laku před UV záření a ozonem. Studované vrstvy laků byly dále charakterizovány na prostupnost kyslíku a vodní páry.

Extrakce anthokyanových barviv z bezinkových výlisků / Extraction of anthocyanin pigments from elderberries pomace

This Diploma thesis deals with the production of the concentrate from the Elderberry marc extract (Sambucus nigra L.). The basic chemical characteristics in this concentrate were determined. The theoretical part of the thesis deal with the botanic characteristic of the Elderberry and its berries. The characteristic of the phenolic compounds, mainly anhocyanins was summarized. The use of the anthocyanin pigments in the food industry, the possibilities of their extraction and the concentrate production was discussed. Thereafter the methods of the anthocyanin content and the phenolic content determination were described. In the experimental part of study, the production of the anthocyanin rich extract from Elderberry marc was optimized. The extraction of the marc by the 1:1 ethanol/water (V/V) solvent with the ratio marc/solvent 1:2 (w/V) showed the most convenient. The production of the concentrate followed. During the concentration on the rotary evaporator the pigment degradation was investigated. The degradation decreased with the decrease of the temperature. The final concentrate was prepared from the extract on the rotary evaporator by 45 °C for 15 minutes. This product had the 7times lower volume compared to the initial extract. In the final concentrate come of the chemical characteristic were determined. By liquid chromatography (HPLC) the major pigments cyanidin-3-sambubiosid and cyanidin-3-glucosid were determined. Their total content was determined to 34,9 gl-1. The total anthocyanin content was determined by pH differential method to 7,1 gl-1 (expressed as cyanidin-3-glucoside equivalent). The total phenolic content was determined to 27,1 gl-1 (expressed as gallic acid equivalent) by Folin-Ciocalteu method. By electron paramagnetic resonance the increase in antioxidant activity with the increase in concentration was found. Up to 42 aromatic compounds was determined in Elderberry marc, its extract and the final concentrate.

Microbial enzymes implicated in plant cell hydrolysis may have several potential aplications such as biomass degradation biocatalysts or with biofuel production. Bark beetles establish symbiosis with several microbial strains which play different roles benifitting the beetle, as the production of hydrolytic enzymes to degrade the ingested wood, the protection against mirobial antagonist or the detoxification of the environment. Fungal symbionts have been traditionally the best studied, but several recent research with bacterial symbionts of several bark beetle species show that bacterial also display important functions for the host. In this study, the bacterial communities of the bark beetle species Cryphalus piceae and Pithophtorus pithophtorus, collected in the Czech Republic from pine and fir trees, respectively, were isolated and 55 out of 89 samples were identified by 16S rRNA gene amplification and sequencing. Members of the genera Erwinia, Pantoea, Curtobacterium, Yersinia, Pseudomonas and Staphylococcus were detected. The isolates were object of study for their possible biotechnological potential in (ligno)cellulose materials degradation by screening several enzymes implicated in plant cell hydrolysis, as cellulases, xylanases, amylases, laccases, as well as their capability for colorant...

Diafragmový výboj v roztocích organických barviv / Diaphragm discharge in organic dye solutions

This Diploma thesis was focused on the degradation of dyes Saturn Red L4B (Direct Red 79) and Saturn Blue LB (Direct Blue 106) by DC diaphragm discharge (DC-DD). Supplied power was between 160 and 180 W. Conductivity and pH were changing at each electrode area during the DC diaphragm discharge, therefore the effect of pH and conductivity changes on the dye solution itself were examined. All samples were measured by UV-VIS spectrometer in the wavelength range of 300–800 nm. No significant dependence of dye absorption spectra on conductivity was observed, while pH significantly affected the absorption curves of dyes. Ageing of dye spectra showed significant changes of Saturn Red L4B. Next task was the comparison of dyes destruction efficiency by DC-DD, audiofrequency diaphragm discharge (AF-DD) and electrolysis. Dye decomposition by AF-DD was not observed at set conditions (voltage of 80–120 V, current of 2.2 A and frequency of 2 kHz). In the case of DC diaphragm discharge the electrolysis played an important role. Decomposition efficiency of the dyes by electrolysis was up to 15 %. The pumping effect in the DC diaphragm discharge was also investigated. Although the individual electrode areas were linked only by a small pinhole in a nonconductive barrier (the pinhole diameter of 0.3 mm), solutions of the anode and cathode compartment interacted with each other up to 10 %. Finally, the degradation products of Saturn Red L4B treated by DC-DD in two electrodes parts are also described. Decomposition products were determined by high performance liquid chromatography (HPLC) combined with the mass spectrometer.

Rozklady organických barviv metodami AOP / Organic dye decomposition by AOP´s methods

This diploma thesis is focused on the study of the influence of advanced oxidation processes on degradation of organic dyes. The field of AOP – very effective physically-chemical methods of wastewater treatment – includes application of strong oxidation agents, UV and ionizing radiation and electrical discharges. For this thesis, two of these methods were chosen – the application of oxidation agent (ozone) and UV radiation. Along with electrolysis, which is mentioned rather marginally in this thesis (it is the subject of the previous thesis), these phenomena are products of electrical discharge in water, where they participate in the processes of degradation in a different way. For all measurement series, two direct azo dyes were chosen as model substances – C.I. Direct Blue 106 and C.I. Direct Red 79. The ozonizer, in which either oxygen or synthetic or technical air were loaded as carrier gases, was used for degradation of dyes by ozone. The generated ozone was loaded into the bubbling vessel with dye solution of different initial concentration (10–130 mg.dm-3), which was followed by other bubbling vessel with KI solution for the next analytical determination of the amount of generated ozone. The other parameters changed were the gas flow (1–2 dm3.min-1), ozonizer output (minimal and maximal), type of dye, pH value of the solution (neutral or acid) and additional electrolyte (NaCl, Na2SO4 or any). The reactor for the study of the influence of UV radiation on degradation of dyes was an UV sterilizer into which the equivalent volume of dye solution was added. The possibilities of experimental settings were limited and only the type of dye, an additional electrolyte and pH value of the dye solution were adjusted (as in the case of ozone). Several series of samples were measured with various input conditions which more or less influenced the degradation of investigated dyes in this experiment. It was found that for both used methods the Direct Blue 106 dye was more degradable (with significantly better results for ozone than for UV radiation). The degradation of Direct Red 79 dye proceeded only by ozone treatment, in the case of the application of UV radiation no degradation occurred. By investigation of the influence of initial concentration of dye on its degradation, it was confirmed that with the initial concentration enhancement the final concentration rises as well, whereas in low concentrations (10–50 mg.dm-3) the initial concentration has no effect. The addition of an electrolyte had an accelerating effect on dye degradation in both methods (NaCl and also Na2SO4 showed similar results though the degradation proceeded in different ways). The addition of HCl accelerated the degradation only in the case of UV radiation; during the application of ozone the pH level of the system did not have any influence on the degradation. Oxygen and synthetic air had the strongest effect on ozone degradation (comparable results); in the case of technical air the final dye concentration was higher up to 30 %. The gas flow of 1.5 dm3.min-1 was stated as optimal with the ozonizer output 30 W (maximal). At minimal power the ozonizer produced very low amount of ozone.

Diafragmový výboj v roztocích organických barviv z hlediska elektrolytického rozkladu / Diaphragm discharge in organic dye solutions with focus on electrolytic decomposition

This Diploma thesis is focused on physical and chemical effects which contribute to the decomposition of organic dyes by diaphragm discharge generated in water solutions. Due to the application of DC high voltage source in continuous regime, there is an effect of electrolysis contributing to the dye decomposition by diaphragm discharge. The aim of this work was to find out when the electrolysis is running (or when is the moment of discharge breakdown) and which factors influence the breakdown. The other goal was decomposition of selected textile and food organic dyes by electrolysis itself. In the theoretical part, theory about creation of electrical discharges in aqueous solutions is noted and various types of underwater discharges are described. Background researches about underwater electrical discharges used in the world are mentioned as well as the use of diaphragm discharges and various ways how to remove organic dyes from wastewater. Finally, theories of electrolysis, UV-VIS spectroscopy and basis of other analytical methods useful for detection of organic molecules are described. Experimental part is oriented to experiment procedure which was carried out in a reactor with separated electrode areas. Separation was made by dielectric diaphragm with a pinhole in the centre. Its initial diameter was 0.4 mm. Used chemicals and course of experiments are described in this part, too. First, the breakdown moment in the reactor was investigated (i. e. determination, when only electrolysis was operating) by formation of hydrogen peroxide and measurement of dynamic (time resolved) electrical characteristics. Next, decomposition of selected dyes by electrolysis was carried out. As the decomposition was related to decolorization of the solution, UV-VIS spectroscopy in the range of 350–700 nm was used for determination of dye concentration. Next part focused on results presents various factors which had an effect on breakdown of diaphragm discharge. These factors are kind of used electrolyte, initial conductivity of solution, kind of dye, temperature of solution and type of reactor (or solution volume). From the result, the most important factor is initial solution conductivity. After the determination of the breakdown moment, the electrolysis of organic dyes was performed. The applied current was 10 mA, initial conductivity was 500 µS/cm and used electrolyte was NaCl. Moreover comparison of dye decomposition in dependence on the different applied power was realized. From this comparison one can assume, there is no significant contribution of electrolysis (the efficiency is approximately 15 %) to the diaphragm discharge in aqueous solution.
