Detecção de potenciais patógenos de ostras de cultivo em Santa Catarina e investigação de mecanismos de defesa imune e antioxidante

Contudo, mortalidades massivas de ostras têm aumentado em frequência e distribuição geográfica ao redor do mundo. Desta forma, estudos básicos a respeito da biologia desses animais, incluindo seus sistemas de defesa, são de fundamental importância. O presente trabalho investigou a presença de patógenos de bivalves em ostras cultivadas em Santa Catarina, os quais têm sido associados a mortalidades de ostras em outros países (Estudo 1). Além disso, buscou-se compreender melhor a respeito da relação entre os sistemas antioxidante e imune de ostras (Estudos 2 e 3). No Estudo 1, ostras do Pacífico Crassostrea gigas e nativas C. gasar (= C. brasiliana)foram coletadas ao longo da costa de Santa Catarina e avaliadas quanto à presença do vírus Ostreid herpesvirus 1 (OsHV-1) e das bactérias Vibrio aestuarianus e V. splendidus. Análises moleculares revelaram amostras positivas para V. splendidus (15 %) e OsHV-1 (28 %), sendo confirmadas por análises de sequenciamento. Este foi o primeiro trabalho demonstrando a presença de bactérias e vírus potencialmente patogênicos em ostras na costa brasileira, servindo de alerta para que medidas preventivas sejam adotadas para evitar o surgimento de mortalidades massivas de ostras no país. No Estudo 2, hemócitos de ostras C. gigas foram expostos ao agente eletrofílico 1-cloro-2,4-dinitrobenzeno (CDNB). O CDNB não afetou a viabilidade mitocondrial, entretanto, diminuiu a estabilidade da membrana lisossomal dos hemócitos (= 12,5 µM). O CDNB causou um forte distúrbio no sistema antioxidante, promovendo depleção da glutationa (= 0,1 µM) e inibição das enzimas glutationa redutase (2,5 e 50 µM) e tioredoxina redutase (50 µM). Importantes funções desempenhadas pelos hemócitos (adesão, fagocitose e produção de espécies reativas de oxigênio após imunoestimulação) foram comprometidas somente por 50 µM de CDNB. Os resultados indicam que a depleção total de glutationa não afeta a imunocompetência dos hemócitos, porém ocorre mediante uma inibição adicional de tiol redutases. No Estudo 3, a importância da glutationa para o sistema imune das ostras foi avaliada utilizando-se butionina sulfoximina (BSO), inibidor da síntese de glutationa. Ostras C. gigas foram expostas via intramuscular ao BSO (15 µmol/animal) por 1, 2 3 ou 6 dias, sendo observados menores níveis de glutationa (21-46 %) nas brânquias, glândula digestiva e hemócitos. O tratamento de 3 dias não afetou o hemograma das ostras, nem a viabilidade e capacidade de adesão e fagocitose dos hemócitos. Contudo, aumentou significativamente a susceptibilidade das ostras ao patógeno V. anguillarum (33 %), e de forma marginalmente significativa às espécies V. alginolyticus e V. harveyi. A depleção parcial da glutationa também aumentou a susceptibilidade das ostras ao peróxido de hidrogênio e hidroperóxidode cumeno. Os Estudos 2 e 3 sugerem que a glutationa não é indispensável para a função dos hemócitos, porém, quando outros componentes do sistema antioxidante são afetados, ou quando um segundo fator de estresse é aplicado, a homeostase destas células ou do organismo é comprometida. De forma geral, estes estudos básicos abrem caminho para vários outros nas áreas de aquicultura e ecotoxicologia, e contribuem para uma melhor compreensão dos mecanismos envolvendo respostas imunes e antioxidantes de bivalves. <br> / Abstract: Oysters are organisms that play important role economically andfor the natural ecosystems. However, oyster massive mortalities have increased in intensity and geographic distribution worldwide. Therefore, studies aiming basic knowledge regarding the biology of oysters are pivotal, especially regarding their defense mechanisms.Thus, the aim of this study was to investigate the presence of oyster pathogens in Santa Catarina, Brazil, which have been associated to oyster massive mortalities in other countries (study 1). Moreover, another aim was tocontribute to basic knowledge regarding the interrelationship of the immune and antioxidant system in oysters (studies 2 and 3). For the 1st study, Pacific oysters Crassostrea gigas and native oysters C. gasar (=C. brasiliana) were collected throughout the coast of Santa Catarina, and the presence of the virus Ostreid herpesvirus (OsHV-1), and the bacteria Vibrio aestuarianus and V. splendidus were evaluated. PCR analyses revealed positive samples for V. splendidus (15 %) and OsHV-1 (28 %), which was confirmed by sequencing analyses. This was the first study to investigate viral and bacterial pathogens to oysters in Brazil, and enlightens the need of preventive measures to be undertaken in order to avoid oyster massive mortalities in Santa Catarina. For the 2nd study, C. gigas hemocytes were in vitro exposed to the electrophilic agent 1-chloro-2,4-dinitrobenzene (CDNB). CDNB did not affect hemocyte mitochondrial viability, however decreased lysosomal membrane stability (=12.5 µM).CDNB strongly affected hemocyte antioxidant system by depleting glutathione (=0.1 µM) and by decreasing the activity of the enzymes glutathione reductase (2.5 and 50 µM) and thioredoxin reductase (50 µM). Hemocyte function (adhesion, phagocytosis, and reactive oxygen species production upon immunostimulation) were jeopardized only with CDNB 50 µM. These results revealed that total glutathione depletion does not affect hemocyte immunocompetence, however this may occur upon inhibition of thiol reductases. For the 3rd study, the importance of glutathione to the immune system of oysters was investigated by using butionine sulfoximine (BSO), an inhibitor of the glutathione synthesis.Oysters C. gigas were exposed to BSO through intramuscular injections (15 µmol/animal/day) for 1, 2, 3 or 6 days, which promoted reduced glutathione levels (21-46 %) in the gills, digestive gland and hemocytes. The injection protocol of 3 days did not affect oyster hemogram, hemocyte viability, adhesion and phagocytosis. Nevertheless, it significantly increased oyster susceptibility to the pathogenV. anguillarum (33 %), while only a trend was observed for thespeciesV. alginolyticusandV. harveyi. Glutathione partial depletions also increased oyster susceptibility to cumene and hydrogen peroxides. Studies 2 and 3 suggest that glutathione is not imperative for hemocyte function, however when other components associated to the antioxidant system are affected, or when a second stress challenge is applied, these cells or the whole organism homeostasis is impaired.Overall, these basic studies can make way for many others in the field of aquaculture and ecotoxicology, and contribute to a better understanding of the mechanisms guiding the immune and antioxidant responses of bivalves.

Parâmetros hemato-imunológicos e perfil histológico de alguns tecidos de ostra do mangue Crassostrea rhizophorae

No. of bitstreams: 0 / O objetivo do presente trabalho foi o de realizar um estudo inicial dos hemócitos e de alguns parâmetros hemolinfáticos da ostra do mangue Crassostrea rhizophorae, além de caracterizar histologicamente sua glândula digestiva e brânquias. Os animais foram coletados sazonalmente (n=30) no período de fevereiro a novembro de 2001, de uma estação de cultivo e de dois habitats naturais da ilha de Santa Catarina, Santa Catarina, Brasil. Foram identificados dois tipos celulares, hemócitos hialinos (HH) e hemócitos granulares (HG) na hemolinfa das ostras. Estas células mostraram conter carboidratos e glicogênio em seu citoplasma, quando submetidas ao método de PAS. A presença de fosfatase ácida intracelular, detectada pelo método de Gomori, sugeriu a presença de lisossomos e o potencial envolvimento dos hemócitos nas reações de fagocitose. Efetivamente, os hemócitos foram capazes de fagocitar in vitro partículas de zymosan, além de produzir moléculas citotóxicas, como ânion superóxido (redução do NBT) durante este processo. A contagem total de hemócitos (THC) variou de 1.100 a 4.000 (céls/mm3), havendo algumas diferenças significativas tanto em relação aos locais de coleta como em relação às estações do ano. Os valores mais elevados de THC ocorreram no verão e em relação aos três locais de coleta, os maiores valores foram geralmente encontrados nas ostras do manguezal. A porcentagem de HG variou de 34 a 82%, havendo diferenças significativas ao longo das estações e em relação aos três locais de coleta. Nas ostras coletadas no manguezal, os HG foram sempre predominantes (mais de 50%). A concentração de proteínas totais do plasma (CP), estimada pelo método de Bradford, variou de 0,11 a 3,0 mg/ml e mostrou um padrão sazonal similar nas ostras das três localidades. Os maiores valores foram atingidos no outono e os menores na primavera. O título aglutinante do plasma contra eritrócitos A humanos (2%) variou de 32 a 16.384, sendo os maiores valores sempre encontrados no cultivo. A análise histológica da glândula digestiva e das brânquias de C. rhizophorae revelou um padrão de organização semelhante ao de outras espécies de ostras. Além disso, não foram observadas alterações nas estruturas e arquitetura destes tecidos.

Este ecossistema desempenha um importante papel socioeconômico para a comunidade local, além de possuir uma grande diversidade ecológica. Os impactos produzidos pelos despejos de efluentes hospitalares e domésticos, aumento do trafego de embarcações motorizadas, carreamento de agrotóxicos de plantações das proximidades e uso destes para a pesca predatória, provocam um acentuado desequilíbrio no estuário, refletindo diretamente na população local, pela introdução de metais-traço nas áreas costeira e estuarina. Baseado nestas prerrogativas, o presente estudo visou a determinação das concentrações de metais-traço (zinco, selênio, arsênio, alumínio, ferro, manganês, chumbo e cromo) no sedimento superficial e em duas espécies de moluscos filtradores (Crassostrea rhizophorae e Mytella charruana), objetivando basicamente avaliar o grau de contaminação do estuário e fornecer indicadores para o estabelecimento de um futuro monitoramento ambiental da área. Amostras de sedimentos superficiais foram coletadas em 04 estações ao longo do estuário, durante os períodos seco e chuvoso do ano de 2009 nas margens direita e esquerda do estuário do rio Formoso. A profundidade do material coletado esteve compreendida entre 0 e 10 cm, onde as amostras foram colhidas durante as baixa-mares. Amostras do material biológico foram coletadas em bancos naturais do estuário localizados próximos à estação 2. Os parâmetros hidrológicos foram coletados nas mesmas estações do sedimento, na parte mais profunda da área. Os resultados obtidos indicaram elevadas concentrações de zinco, arsênio, ferro, manganês, chumbo e cromo nos sedimentos e arsênio, ferro e manganês nos moluscos bivalves. Para análise e quantificação dos metais nos sedimentos e moluscos, foram realizadas digestões ácidas próprias para cada matriz e utilizado o Espectrômetro de Emissão Ótica em Plasma Indutivamente Acoplado (ICP-OES). Os dados produzidos indicaram contaminação das matrizes utilizadas na pesquisa, onde os resultados obtidos foram comparados aos valores de referência das agências ambientais nacionais e internacionais

Ecological, Evolutionary, and Taphonomic Comparisons of Brachiopods and Bivalves at Multiple Spatial and Temporal Scales

The fossil record is the primary source of information on the history of life. As such, it is important to understand the limitations of this record. One critical area in which there is still much work to be done is in understanding how the fossil record, and our interpretation of it, may be biased. Herein, the fidelity between the life and death assemblage of an extant brachiopod with respect to morphological variability is studied using geometric morphometrics. The results from several analyses confirm a high degree of morphological variability with little change in mean shape between the living and sub-fossil assemblage. Additionally, there is no evidence of distinct morphogroups in either assemblage. These trends persist at all depths and size classes indicating that this species could be recognized as a single, rather than multiple, species if only fossil data were available. The second chapter involves the recognition and quantification of a worker bias in monographs of brachiopods and bivalves. Most specimens studied came from the 65th to 69th percentile of their species' bulk-collected size-frequency distribution. This indicates a significant bias toward monograph specimens that are larger than the mean size of the bulk sample. When compared at the species level, this bias was found to be highly consistent among the 86 species included in the study. Thus, size measurements of monographed specimens reliably and consistently record a similar size class for any given species, and this bias is easily corrected during meta-analyses. Chapter three focuses on bivalves and brachiopods from a modern tropical shelf and quantifies the magnitude of time averaging (temporal mixing) for these two different organisms. This is accomplished by dating a suite of shells from each site using amino acid racemization calibrated with several radiocarbon dates. By studying the age distributions for each species it is determined that, despite some site to site differences, both bivalve and brachiopod species exhibit a similar time averaging magnitude when collected from the same region or depositional system. This indicates that fossil assemblages of these species may have very similar resolution. / Ph. D.

Valores de referência para metais potencialmente tóxicos em sedimentos e em moluscos filtradores no Rio Ribeira de Iguape / Reference values to potentially toxic metals in sediments and mussels in the River Ribeira de Iguape

O alto da Bacia Hidrográfica do Rio Ribeira de Iguape foi submetido a intensa atividade mineira, cujo objeto mineral principal era a obtenção de chumbo (Pb). Durante longo período as condições de explotação foram rudimentares, não havendo controle sobre o impacto ambiental gerado durante as fases de extração e beneficiamento do minério, cujos rejeitos (escória e concentrado) foram lançados por anos diretamente no rio Ribeira de Iguape. Com o propósito de se criar uma indicação clara do comprometimento ambiental ou não da bacia hidrográfica do rio Ribeira de Iguape quanto ao teor dos metais Cd, Pb, Cr, Zn e Cu, este trabalho exibe resultados de valores que podem servir de referência em teor para estes metais. Para tanto, foram coletadas amostras de duas espécies de moluscos bivalves, Anodontites tenebricosus e Corbicula fluminea e de sedimento aluvionar, em uma porção localizada a montante das fontes de contaminação pelas atividades de mineração e fundição. Os moluscos foram coletados por exame tátil do substrato, seus tecidos foram separados das conchas, liofilizados e analisados em espectrômetro de massa (ICP/MS) e em espectrômetro de absorção atômica (AES). Os valores médios foram em sua maioria inferiores aos valores obtidos por outros autores em pontos localizados ao longo do rio Ribeira de Iguape sob a influência das atividades de mineração e fundição. Em relação às amostras de sedimento aluvionar estas foram separadas em duas porções, uma contendo sedimentos com minerais não magnéticos e a outra sedimentos com minerais magnéticos. Estas amostras foram tratadas granulometricamente e analisadas por fluorescência de raio X (FRX), a fim de se obter a concentração dos metais contidos em cada fração granulométrica. Nas amostras de sedimento não magnético, o composto químico em maior concentração foi SiO2. Quanto aos metais potencialmente tóxicos aqui investigados, todos foram encontrados nestas amostras. Já em relação ao sedimento magnético, as amostras exibiram constituição química composta principalmente por Fe2O3. Os metais Pb, Cr, Zn e Cu, foram encontrados em maior concentração nas amostras referentes as frações granulométricas mais finas, sendo a concentração do Cr nitidamente mais elevada. A elevada concentração deste metal nas amostras de sedimento magnético, quando comparada com as obtidas nas amostras não magnéticas, pode indicar uma provável origem do Cr a partir do mineral cromita (FeCr2O4). Estes dados juntamente com informações contida na literatura específica, indicam que provavelmente a origem do Cr, Pb, Zn e Cu na bacia do rio Ribeira de Iguape possui origem geogênica. De maneira geral, as concentrações dos metais potencialmente tóxicos aqui verificadas, tanto nas amostras referentes às duas espécies de moluscos quanto nas amostras de sedimentos, são inferiores, em sua maioria, às obtidas em pontos ao longo do rio Ribeira de Iguape estudados por outros autores onde há influência das atividades de mineração, o que permite considerar esses dados como referência. / The Ribeira do Iguape River Basin (RIRB) was historically influenced by intense mining activities mainly related to lead (Pb) extraction. For many decades, such exploitation was conducted under rudimentary conditions without control of environmental impacts produced during the stages of ore extraction and beneficiation. Thus, mining wastes and metallurgical slags were directly discharged into the Ribeira de Iguape River, causing environmental contamination by metals. The present investigation has the purpose of developing reference values to some metals (Cd, Pb, Cr, Zn and Cu) to RIRB, in order to provide information that allow the environmental monitoring and the determination of environmental contamination to Ribeira do Iguape River Basin. To achieve that, samples of two bivalves species were collected: Corbicula fluminea and Anodontites tenebricosus. Besides, alluvial sediments from a site located upstream to the contamination sources for metals (mining and smelting activities) were also collected. For both species, soft tissues were firstly separated from their respective shells, freeze-dried and then analyzed in a mass spectrometer (ICP/MS) and atomic absorption spectrometer (AAS). Alluvial sediment samples were splitted into two aliquots, one containing magnetic minerals and another with sediment containing non-magnetic minerals. These samples were separated for particle size and analyzed by X-ray fluorescence (XRF) in order to obtain the metals concentrations for each class of particle size. For presence of metals in bivalves, concentrations were mostly lower than those reported by other authors along RIRB areas influenced by mining and smelting residues. In the non-magnetic sediment fraction, the prominent substance was SiO2. Regarding to potentially toxic metals investigated in this study, all of them were found in such samples. On the other hand, in the magnetic sediment fraction, the prominent chemical was Fe2O3. Pb, Cr, Zn and Cu were found in highest concentrations in the finer grain fractions. The Cr concentration was noticeably high. The higher concentration of this metal in magnetic sediment fractions, when compared to the non-magnetic ones, may indicate a probable source of Cr, Pb, Zn e Cu from the mineral chromite (FeCr2O4). These data, together with that from specific literature, indicate that the origin of Cr at RIRB is probably natural .In general, the potentially toxic metals concentrations found in this study for both species of mussel and sediment are mostly lower than the concentrations obtained by others authors for other portions of Ribeira de Iguape River under influence of mining activities, thus, such results suggest that these concentrations may be considered as local reference values for RIRB.

Effets de l’exposition parentale au diuron sur le méthylome et transcriptome de l’huître du Pacifique Crassostrea gigas / Effects of parental exposure to diuron on methylome and transcriptome of the Pacific oyster Crassostrea gigas

L’huître du pacifique Crassostrea gigas est l'espèce marine la plus cultivée avec une production supérieure à 4 millions de tonnes pour l'année 2010. En France, C. gigas est cultivée depuis la fin des années 1970. Cependant, cette espèce souffre d’un syndrome de mortalité estivale depuis les années 1980, avec une amplification depuis 2008 qui touche jusqu'à 100 % des naissains. Ce syndrome de mortalité est un phénomène multifactoriel, basé sur l’interaction de nombreux facteurs: stress environnementaux, caractéristiques physiologiques et génétiques de l’huître, présence et virulence de pathogènes. L’huître du pacifique C. gigas est une espèce estuarienne qui est soumise aux pressions anthropiques comme la pollution du milieu côtier. Ces événements représentent des sources potentielles de stress en zones ostréicoles. Cependant, les connaissances sur les effets des polluants comme les pesticides sur C. gigas restent fragmentaires. Les périodes d’épandage d’herbicides coïncident parfois avec la période de reproduction des huîtres, raison pour laquelle nous considérons que ces produits chimiques pourraient affecter la génération suivante d'huîtres. Parmi les pesticides, le diuron est le plus fréquemment détecté sur les côtes françaises, avec une concentration maximale rapportée de 0,78 µgL-1. L'exposition directe aux herbicides affecte le transcriptome des huîtres qui est le premier niveau de réponse face à l'exposition du polluant. Il a été démontré que l'exposition parentale au diuron a des effets génotoxiques chez C. gigas au stade de naissain. Une variabilité phénotypique de trait d’ histoire de vie a été observée aussi pour ces naissains. Un autre effet possible des pesticides serait la modification de marques épigénétiques. Il est connu que les facteurs environnementaux telle que la pollution par des composés chimiques peuvent modifier l'épigénome et par conséquent le phénotype des individus et de leurs descendance en agissant au niveau trans-générationnel. Ces dernières observations nous permettent d’émettre l’hypothèse de l’implication de mécanismes épigénétiques suite à l’interaction avec des produits phytosanitaires. Ces mécanismes modifieraient le phénotype des huîtres au stade de naissains par l'exposition parental. Pour tester cette hypothèse nous avons étudié la méthylation globale de l'ADN (méthylome), qui est un de principal marques épigénétiques, et le transcriptome des naissains issus de géniteurs exposé au Diuron. Nous avons identifié des modifications du méthylome et du transcriptome qui ont un lien avec le phénotype de trait d'histoire de vie de ces naissains. Ces résultats démontreraient qu’une exposition indirecte ou parentale du diuron modifie la méthylation et l'expression de fonctions de gènes spécifiques, expliquant en partie la variabilité phénotypique observée. / The Pacific Oyster Crassostrea gigas is the most cultivated marine species in the world with a production superior to 4 millions of tons in 2010. In France, C. gigas is cultivated since the end of 1970s. However, this specie suffers from a syndrome of summer mortalities since the 1980s, with an amplification since 2008 affecting up to 100% of spats. This syndrome of mortality is a multifactorial phenomenon, based on the interaction of many factors: Environmental factors, genetic and physiologic features of the oysters, and the presence and virulence of pathogens. The Pacific Oyster C. gigas is an estuarine specie which is subjected to anthropogenic pressures such as pollution of the coastal environment. These events represent a potencial source of stress in oyster farm areas. However, the knowledge about the effects of pollutants such as pesticides on C. gigas remain fragmented. The herbicide application periods may coincide with the oyster breeding period, reason for which we consider that these chemicals could affect the next generation of oysters. Among pesticides, diuron is the most frequently detected on the French coast, with a maximum reported concentration of 0.78 µgL-1.The direct exposure to herbicides affects the transcriptome of oysters which is the first level of response to the exposure of pollutants. It was shown that parental exposure to diuron has genotoxic effects on C. gigas at the spat stage. A phenotypic variability of life history traits has also been observed for these spats. Another possible effect of pesticides would be the modification of epigenetic marks. It is known that environmental factors such as pollution by chemical compounds can alter the epigenome and consequently the phenotype of individuals and of their offspring acting at a transgenerational level. These last observations allow us to hypothesize the involvement of epigenetic mechanisms in response to interactions with herbicide products. These mechanisms could modify the phenotype of oysters spat state by parental exposure. To test this hypothesis we studied the genome-wide DNA methylation (methylome), which is a main epigenetic mark, and the transcriptome of the spat from diuron-exposed genitors. We identified methylome and transcriptome changes that are related to the phenotype of life history trait of these spats. These results show that an indirect or parental exposure to the diuron is able to modify the methylation and the expression of specific gene functions, partially explaining the phenotypic variability observed.

Estes animais atuam como bioindicadores de contaminação fecal e, ao serem ingeridos crus ou mal cozidos, podem transmitir os patógenos acumulados em seus tecidos. Diversos estudos em diferentes países reportaram a presença de protozoários patogênicos como Cryptosporidium e Giardia em moluscos bivalves destinados ao consumo humano, porém, no Brasil estes dados são inexistentes. Diante deste cenário, é interessante e relevante investigar a ocorrência destes protozoários neste tipo de alimento, consumidos em diversas regiões do país. Este estudo teve a finalidade de avaliar a contaminação natural por oocistos e cistos dos protozoários parasitos em moluscos bivalves localizados em um ambiente de estuário do litoral norte do Estado de São Paulo; detectar as formas parasitárias em água de rio e água do mar; verificar se existe correlação entre fatores climáticos, físico-químicos e microbiológicos e a ocorrência natural dos parasitos nestes ambientes aquáticos e verificar se existe correlação entre a qualidade das águas e a contaminação dos moluscos que vivem nesta região. Os exemplares de moluscos foram processados através da homogeneização de tecidos específicos e submetidos a centrífugo-concentração com éter-PBS e as amostras hídricas foram analisadas pela técnica de filtração em membranas. A visualização dos protozoários foi realizada através da reação de imunofluorescência direta e a confirmação da morfologia dos protozoários feita com a utilização de microscopia de contraste de fase e a incorporação de um corante vital (DAPI). Cistos de Giardia foram detectados em 8 das 12 amostras de água do rio e uma amostra continha oocistos de Cryptosporidium. Ambos os patógenos foram detectados na água do mar, onde Cryptosporidium esteve presente em 40,0 % e Giardia em 20,0 % do total amostrado. O pool de homogenados teciduais de brânquias foi positivo para oocistos e cistos em dois períodos, com concentrações de 12/oocistos/animal e 50 cistos/animal. Não houve associação significativa entre os fatores climáticos, físico-químicos e microbiológicos e o número de oocistos detectados na água do rio e na água do mar; para os cistos, foi encontrada uma correlação significativa com coliformes termotolerantes (p = 0,007) na água do mar. Ademais, não foi possível estabelecer correlação entre a qualidade da água e o número de cistos e oocistos detectados nos tecidos das ostras. Esta é a primeira vez que oocistos de Cryptosporidium e cistos de Giardia foram encontrados em ostras de uma região costeira do país e também na América Latina. Estes achados têm importância para a saúde pública, pois refletem a extensão da contaminação dos ambientes e dos alimentos de origem marinha, requerendo a padronização de um controle de qualidade. Palavras-chave: Cryptosporidium spp.; Giardia spp.; moluscos bivalves; Crassostrea rhizophorae; água do mar; água de rio; estuário; esgoto / Abstract: Bivalve molluscs are great filter feeders that may accumulate and concentrate substances, particles and even pathogens in their tissues, if the places that they inhabit or are cultivated receive contaminated effluents dispose. These animals, which serve as biological indicators of faecal contamination, may transmit the protozoa accumulated in their tissues, whenever they are ingested in a raw or lightly cooked form. Several studies in different countries reported the presence of parasite protozoa like Cryptosporidium and Giardia in bivalve molluscs destined for human consumption, however, in Brazil, these data are non-existent. For this reason, it is interesting and relevant the investigation of the occurrence of these protozoan in this type of food, consumed in different regions of the country. The aim of this study was to evaluate the natural contamination of oocysts and cysts of parasite protozoan in bivalve molluscs located in an estuarine environment in the Northern coast of São Paulo State; detect this protozoan in river and seawater; verify if there is a correlation between climatic, microbiological, physical and chemical factors with the natural occurrence of parasites in aquatic environments and verify if there is a correlation between water quality and the contamination of bivalve molluscs present at the region. Bivalve molluscs were processed through the homogenization of specific tissues and submitted to centrifugal concentration with ether-PBS, and water samples were analyzed by membrane filtration technique. Protozoa were visualized with direct immunofluorescence antibody test and to confirm the morphology of the parasites, contrast-phase microscopy and the incorporation of a vital dye (DAPI) were used. Giardia cysts were detected in 8 of 12 samples of river water and one sample contained Cryptosporidium oocysts. Both pathogens were detected in marine water where Cryptosporidium were present in 40,0% and Giardia in 20,0% of all samples analyzed. Gill pools homogenates of oysters were positive for oocysts and cysts in two periods with a mean concentration of 12 oocysts/oyster and 50 cysts/oyster. There was no significant association between the climatic, physical, chemical and microbiological factors with the number of oocysts detected in river water and in seawater; for cysts, it was found a high association only with thermotolerant coliforms (p = 0,007) in seawater. Moreover, it was not possible to establish correlation between water quality and the number of oocysts and cysts detected in oysters tissues. This is the first time that Cryptosporidium oocysts and Giardia cysts were found in oysters from the coastal region of Brazil, and it is also the first report in Latin America to date. These findings might be of public health importance, reflecting the extension of the contamination in marine environment and on seafood, requiring a need for quality control standards. A partir de 1984 as pesquisas brasileiras se concentraram na Baía do Almirantado, na Ilha Rei George, onde se localiza a base do país. A Baía do Almirantado foi eleita uma "Área Antártica Especialmente Gerenciada" pelas Partes Consultivas do Tratado Antártico e trata-se de um local representativo do ecossistema costeiro antártico, principalmente em termos de distribuição e composição de espécies. Lá os pesquisadores tem estudado a fauna bentônica da região, entre eles os Mollusca, que são abundantes e importantes no estudo dos processos que determinam a estrutura e o funcionamento das comunidades bentônicas marinhas. Este estudo se focou na classe Bivalvia, provendo um novo material de referência para identificação destes animais, e mais especificamente daqueles da Baía do Almirantado, apresentando-se como um guia ilustrado, com descrições detalhadas das conchas dessas espécies. Além disso, dados de distribuição geográfica e batimétrica foram reunidos para uma análise dessa fauna, na tentativa de qualificar a importância da região das Ilhas Shetland do Sul como um local estratégico para estudos em biogeografia. Foram identificadas 39 espécies de 18 famílias distintas, e seus dados de biologia, distribuição batimétrica e geográfica reunidos. As famílias mais representativas foram Philobryidae, Nuculanidae e Lasaeidae, contabilizando 15 espécies. As regiões com mais espécies em comum com a área estudada foram o Estreito de Magalhães, Ilha Georgia do Sul, Mar de Weddell, Península Antártica e sul do Chile; regiões notáveis pela riqueza de Bivalvia. As famílias mais largamente distribuídas, encontradas neste estudo, foram LImidae e Carditidae; e as mais restritas foram Siliculidae e Nuculanidae. A espécie com maior distribuição foi Cyclocardia astartoides, e as mais restritas Ennucula georgiana e Mysella narchii. As espécies encontradas se concentram nas profundidades até 1000 m, com seu número caindo para menos da metade além disso. Apenas seis espécies ocorrendo além dos 2000 m: Yoldiella profundorum, Limopsis marionensis, Limopsis lilliei, Adacnarca nitens, Limatula pygmaea e Cyclocardia astartoides. Este estudo demonstram a região da Baía do Almirantado como abrigo de uma fauna de Bivalvia composta por espécies estritamente antárticas, e outras que ocorrem ao norte da Convergência Antártica; sendo assim uma região importante para o estudo da distribuição destes animais / Abstract: Brazil abided to the "Antarctic Treaty" in 1975, and since 1982 maintains the "Brazilian Antarctic Program". As from 1984 the Brazilian researches focused on Admiralty Bay, in King George Island, where the country?s Antarctic base is located. Admiralty Bay was elected a "Antarctic Especially Managed Area" by the Antarctic Treaty Consultative Parties and is a representative place for the Antarctic coastal ecosystem, especially in views of species distribution and composition. In there researchers have been studying the regional benthic fauna, amidst it the Mollusca, abundant and important animals for the study of determinant process in the structure and operation of the marine benthic community. The present study focused on the class Bivalvia, providing a new reference source for identification of the group, and specifically for those of Admiralty Bay, presented as an illustrated guide with detailed shell descriptions for all the species found. Furthermore, data on geographic and bathymetric distribution were gathered for an analysis of this fauna, in attempt to qualify the importance of the South Shetland Islands as a strategic place for studies on biogeography. Thirty-nine species, of 18 distinct families, were identified, and their biological, geographical and bathymetrical data gathered. The most representative families were Philobryidae, Nuculanidae and Lasaeidae, counting 15 species in sum. The geographic regions with more species in common with the studied area were Strait of Magellan, South Georgia Island, Weddell Sea, Antarctic Peninsula and Southern Chile; notable regions for Bivalvia richness. The families found with the largest geographic distribution were Limidae and Carditidae, and the most restricted ones were Siliculidae and Nuculanidae. The species with largest geographical distribution was Cyclocardia astartoides, while the most restricted were Ennucula georgiana and Mysella narchii. The species found in this study are concentrated in depths until 1000 m, beyond that their number shortens to less than half. Only six occur deeper than 2000 m: Yoldiella profundorum, Limopsis marionensis, Limopsis lilliei, Adacnarca nitens, Limatula pygmaea and Cyclocardia astaroides. The present study shows Admiralty Bay as shelter to a Bivalvia fauna with strictly Antarctic species and northern ones, common north to the Antarctic Convergence, thus being an important place for studying the distribution of these animals / Mestrado / Ecologia / Mestre em Ecologia

Les phénoloxydases chez l’huître creuse Crassostrea gigas : biomarqueurs potentiels de stress environnemental / Phenoloxidases in the Pacific oyster Crassostrea gigas : potential biomarkers of environmental stress

L’huître creuse Crassostrea gigas est le produit aquacole le plus commercialisé dans le monde. Cependant, en France et dans d’autres régions du monde, des mortalités estivales massives affectent de façon alarmante les populations naturelles et cultivées de cette espèce, surtout aux stades juvéniles. Ces évènements sembleraient être liés à un déséquilibre entre les acteurs de la triade hôte – agent pathogène – environnement, favorisant l’affaiblissement des mécanismes de défense de l’hôte, et par conséquent, l’apparition et/ou l’augmentation de maladies. Parmi les facteurs environnementaux pouvant contribuer à ce déséquilibre, les contaminants chimiques sont connus pour moduler les capacités de défense de divers organismes aquatiques. Les phénoloxydases (PO) sont les enzymes-clés d’une cascade biochimique responsable de la production de mélanine et sont impliquées dans les défenses immunitaires et dans la reconnaissance du non-soi chez les invertébrés. Récemment, une activité de type PO a été détectée chez C. gigas, et a été proposée comme biomarqueur potentiel de la contamination chimique. Cependant, de nombreuses inconnues existent quant aux différents types d’activité PO présents chez C. gigas, et à leur implication dans les mécanismes de défense chez cette espèce. Dans ce contexte général,les objectifs de cette thèse ont été d’ 1) identifier les différents types de PO présents dans différents tissus de l’huître 2)évaluer le potentiel des PO en tant que biomarqueurs dans des expériences d’exposition in vivo à des contaminants organiques, 3) évaluer le potentiel des PO en tant que biomarqueurs dans des études de biosurveillance in situ. Nous avons pu montrer, pour la première fois chez cette espèce, l’existence de deux types d’activité PO : une activité catécholase et une activité laccase. De plus, nous avons pu mettre en évidence in vitro un effet bactéricide lié à l’activité des PO contre deux souches pathogènes de l’huître, Vibrio splendidus et V. aesturianus, dont leur présence a été fréquemment associée aux phénomènes de mortalités estivales. Enfin, grâce à une analyse multi-biomarqueurs sur différents tissus de C. gigas,l’ensemble des résultats ont permis de proposer que les PO pourraient être utilisées comme biomarqueurs de stress environnemental, plutôt que de contamination, dans la surveillance de la qualité des eaux côtières et estuariennes. / The Pacific oyster Crassostrea gigas is the leading aquaculture product at the worldwide level. However, massive summer mortalities affect dramatically cultivated and natural oyster populations, especially at young life stages. These events could be linked to an unbalance between the different actors of the triad host – pathogen agent – environment, which could favour a weakening of defence mechanisms in the host, and consequently, the emergence and/or increase of diseases. Among environmental factors that could contribute to this unbalance, chemical contaminants are known to modulate defence capacities in different aquatic organisms. Phenoloxidases (POs) are the key enzymes of a biochemical cascade responsible for the production of melanin and are implicated in immune defences and in self/non-self recognition in invertebrates.Recently, a PO-like activity was detected in C. gigas, and was suggested as a potential biomarker of chemical contamination. However, little is known on the different PO activities present in C. gigas, and on their implication indefence mechanisms in this species. In this general context, the aims of this work were to 1) identify the different types of PO present in different tissues of C. gigas 2) evaluate their potential as biomarkers in experiments of in vivo exposure to organic contaminants, 3) evaluate their potential as biomarkers in in situ biomonitoring studies. We showed, for the first time in this species, the existence of two PO-type activities: a catecholase activity and a laccase activity. Moreover, we observed an in vitro bactericidal effect associated to PO activities against two oyster pathogens, Vibrio splendidus and V.aesturianus, which have often been associated to massive summer mortalities in oysters. Finally, based on a multiple biomarkeranalysis in different tissues of C. gigas, results suggest that POs could be used as biomarkers of environmental stress, rather than of contamination, for biomonitoring studies in coasts and estuaries.

Approche intégrative de la réponse d'un organisme marin face au changement climatique : la coquille Saint-Jacques Pecten maximus et les stress thermique et hypoxique / Integrative approach of the response of marine organisms to climate change : heat- and hypoxia- stresses in the great scallop Pecten maximus

Les écosystèmes côtiers sont parmi les plus vulnérables aux changements globaux actuels, qui entraînent notamment une augmentation de la température de l'eau, ainsi que de la fréquence des épisodes hypoxiques. La coquille Saint-Jacques, Pecten maximus, est une espèce subtidale évoluant à des profondeurs de 2 à 210 m. Malgré son intérêt commercial et un intérêt écologique majeur, cette espèce n'a fait l'objet que de peu d'études au niveau moléculaire. L'objectif de cette thèse était de caractériser les mécanismes moléculaires régissant l'acclimatation de cette espèce aux contraintes thermique et hypoxique. Nous avons dans un premier temps caractérisé les modifications d'expression des gènes/protéines, par des approches transcriptomiques (RNAseq) et protéomiques (2-DE), dans un tissu, le manteau, d'animaux exposés à une contrainte thermique prolongée (56 jours). Nous avons ainsi pu identifier les voies majeures de régulation (eg., AP-1), les grandes fonctions (eg., cytosquelette) et processus (eg., apoptose) impliqués dans la réponse, mais également d'observer les grandes orientations du métabolisme (eg., dégradation des lipides de réserve). La réponse des organismes à l'hypoxie dépend de leur manière de gérer les faibles teneurs en oxygène. Nous avons d'abord, par une approche comparative avec une espèce intertidale, la moule (Mytilus spp.), caractérisée la réponse physiologique de la coquille Saint-Jacques à l'hypoxie. Nous avons pu ainsi déterminer ses paramètres d'oxyregulation, plus particulièrement son Point critique en 02 (Pc02). Le développement d'une approche protéomique, couplant l'effet de la température et de l'hypoxie, nous a ensuite permis d'identifier plusieurs protéines (CK2, GLN, etc.) potentiellement impliquées dans la réponse au niveau moléculaire. Enfin, dans l'optique de mieux comprendre la physiologie particulière de ces mollusques dans leur environnement naturel, nous avons comparé les signatures protéomiques de deux populations de P. maximus évoluant dans des écosystèmes contrastés, i.e. en limite nord- (Norvège) et au centre- (Brest) de l'aire de répartition de l'espèce. Les résultats suggèrent des différences majeures entre les deux populations au niveau du cytosquelette. En conclusion, ce travail ouvre des perspectives nouvelles pour la compréhension des mécanismes moléculaires régissant l'adaptation des mollusques aux contraintes thermiques et hypoxiques, deux stress particulièrement importants pour les organismes marins dans le contexte du changement global. / Coasts are among the most vulnerable ecosystems to the ongoing global changes, which result in increased water temperatures and frequencies of hypoxic episodes. The great scallop, Pecten maximus, is a subtidal species living at depths of 2-210 m. In spite of its commercial and major ecological values, only few studies at the molecular level were performed on this species. This thesis aimed at characterizing the molecular mechanisms implied in acclimation of this species to thermal and hypoxia stresses. We first characterized the changes of expression of the genes / proteins in response to a long-term thermal stress (56 days), by using both a transcriptomic- (RNAseq) and a proteomic- (2-DE based) approaches, in the mantle tissue of scallops. This allowed us to identify key regulatory pathways (eg., AP-1), the major functions (eg., cytoskeleton) and processes (eg., apoptosis) involved in the response, but also to observe the main orientations of metabolism (eg., degradation of lipid reserves). The response of organisms to hypoxia depends on how they cope with low oxygen availability. Therefore, we first carried out a comparative approach with an intertidal species, the mussel (Mytilus spp.) to characterize the physiological response of P. maximus to hypoxia. Of note, we could determine its oxyregulatory parameters, particularly its critical point in 02 (Pc02). Then, coupling the effects of temperature and of hypoxia, we developed a proteomic approach that allowed us to identify several proteins (CK2, GLN, etc.) potentially involved in the response at the molecular level. Finally, in an effort to better understand the particular physiology of these mollusks in their natural environment, we compared the proteomic signatures of two populations of P. maximus living in highly contrasted ecosystems, ie in the northern limit- (Norway) and the center- (Brest) of the biogeographical distribution of this species. The results suggest major differences between the two populations, especially at the cytoskeleton level. In all, this work opens new avenues for understanding the molecular mechanisms governing the adaptation of mollusks to heat and hypoxia, two stresses that will most probably greatly influence the lifestyle of marine organisms and populations in future years.
