Interação entre polimorfismos de genes envolvidos na homeostase energética e na sensibilidade à insulina e a resposta a uma intervenção dietética para a redução do peso corporal em indivíduos obesos / Interaction between polymorphisms of genes involved in energy homeostasis andinsulin sensitivity and response to a dietary intervention for weight reduction in obese individuals

Neste estudo, foram incluídos 201 indivíduos com idade de 30 a 80 anos, sendo 100 obesos (IMC> 30 kg/m2) e 101 indivíduos do grupo controle. Os obesos participaram do programa de orientação nutricional para redução do peso corporal. As medidas antropométricas, avaliação da composição corporal e o perfil metabólico foram avaliados no grupo total. Os polimorfismos genéticos foram analisados pela técnica de PCR em tempo real e apenas o polimorfismo do gene IL 6 por PCR convencional. A avaliação de consumo alimentar foi realizada em apenas 73 indivíduos obesos que completaram o programa de orientação nutricional. Não encontramos associação entre os polimorfismos estudados e a obesidade em nosso estudo. No polimorfismo LEP -2548G>A, os indivíduos do grupo controle apresentaram concentração elevada de VLDL-c e triglicérides. Para o polimorfismo LEP 19A>G, os indivíduos obesos apresentaram concentração elevada de VLDL-c, insulina, HDL-c e valores aumentados de Homaβ e o grupo controle apresentou a concentração elevada de hemoglobina glicada. Para o polimorfismo LEPR Lys109Arg (c.326A>G), os obesos apresentaram valores aumentados de CA e RCQ e concentração elevada de ApoB e HDL-c, enquanto que os indivíduos do grupo controle apresentaram os valores aumentados de IMC, CA e teor de gordura e usPCR. Para o polimorfismo Gln223Arg (c.668A>G), os obesos apresentaram valores aumentados de teor de gordura e a concentração elevada de hemoglobina glicada e usPCR . Os indivíduos obesos, para o polimorfismo ADIPOQ 45T>G, apresentaram as concentrações elevadas de colesterol total e LDL-c ao passo que no grupo controle as concentrações elevadas de glicose e triglicérides. Para o polimorfismo 276G>T, os obesos apresentaram valores aumentados de IMC, CA, RCQ e teor de gordura e concentração elevada de IL-6. Os mesmos indivíduos apresentaram também o valor de IMC aumentado e a concentração de usPCR elevada para o polimorfismo PPARG Pro12Ala (34G>C). Já para o polimorfismo 161C>T os indivíduos obesos demosntraram a concentração elevada de HDL-c enquanto o grupo controle apresentou a concentração elevada de insulina e IL-1β. Os indivíduos obesos para o polimorfismo IL-6 -174 G>C, apresentaram os valores de CA aumentados e a concentração elevada de PAI-1 já para os indivíduos do grupo controle a concentração elevada de ApoA, IL1β e usPCR. No programa intervencional, foi possível observarmos uma adequação em relação ao consumo recomendado de carboidrato e lipídios, excedendo apenas proteínas. O consumo de carboidratos foi maior nos grupos com graus II e III de obesidade. Após a intervenção nutricional, observamos mudanças no hábito alimentar dos envolvidos devido à diminuição da ingestão calórica e á redução do perfil lipídico, inflamatório, nos polimorfismos LEP, LEPR, ADIPOQ, PPARG e IL6. Os indivíduos obesos portadores dos haplótipos TG+GG/GG, para os polimorfismos do gene ADIPOQ, apresentaram valor aumentado de CA, RCQ e concentração elevada de glicose. Os portadores do haplótipo TG+GG/GT+TT demonstraram hemoglobina glicada e ApoB. Os indivíduos obesos portadores dos haplótipos AG+GG/GG, para os polimorfismos do gene LEPR, apresentaram valores aumentados de CA, RCQ e teor de gordura, já portadores do haplótipo AG+GG/AG+GG demonstraram concentração elevada de leptina, adiponectina, glicose, usPCR e HDL-c. / This study included 201 individuals aged 30 to 80 years, and 100 obese (BMI> 30 kg/m2) and 101 control subjects. Anthropometric measurements, assessment of body composition and metabolic profile were evaluated in the total group. Obese people, for the ADIPOQ polymorphism 45T> G, showed elevated concentrations of total cholesterol and LDL-C while in the control group the concentrations of glucose and triglycerides. For the polymorphism 276G> T, the obese had increased values of BMI, WC, WHR and fat content and high concentration of IL-6. The same individuals also had increased the value of BMI and the concentration of high usPCR for the PPARg polymorphism Pro12Ala (34G> C).As for the polymorphism 161 C> T obese individuals demonstrated the high concentration of HDL-C while the control group had a high concentration of insulin and IL-1β. Obese individuals for polymorphism IL-6 -174 G> C showed increased values of WC and the high concentration of PAI-1 as for individuals in the control group, the high concentration of ApoA, and IL1β usPCR. In the interventional program was able to observe a suitability in relation to recommended intake of carbohydrates and lipids, exceeding only protein. The carbohydrate intake was higher in groups II and III degree of obesity. After nutritional intervention, we observed changes in dietary habits of those involved due to decreased caloric intake and reducing the lipid profile, inflammatory, polymorphisms in the LEP, LEPR, ADIPOQ, PPARg and IL6. Obese individuals carring the haplotype TG+GG/GG, for the polymorphisms ADIPOQ gene showed increased values of WC, WHR end elevated glucose. Carries of haplotype TG+GG/GT showed glycate hemoglobin and ApoB. Obese individuals carrying the haplotype GG + AG / GG for the LEPR gene polymorphisms showed increased values of WC, WHR and fat, as carriers of the haplotype AG+GG/AG+GG showed high concentration of leptin, glucose, HDL-c and us PCR.

O presente estudo tem como objetivo investigar se o treinamento periodizado e a dieta hiperlipídica influenciam os níveis de atividade física espontânea e voluntária e a alteração na ingestão alimentar de ratos. Para isso, o estudo foi dividido em duas etapas: na primeira o objetivo foi verificar a acurácia e confiabilidade de um sistema de videogrametria na análise do deslocamento de ratos Wistar, e na segunda etapa, verificar se o aumento da atividade física, por meio do treinamento periodizado, é acompanhado por uma redução compensatória na atividade física e alteração da ingestão alimentar. Foram utilizados Ratos (n=32) da linhagem Wistar (Rattus Norvegicus Albinus Wistar) com idade inicial de 120 dias mantidos em gaiolas coletivas durante 8 semanas, após a separação dos grupos: Grupo controle (GC), Grupo controle com dieta hiperlipídica (GDH), Grupo de treinamento periodizado (TP), e Grupo treinamento periodizado dieta hiperlipídica (TPH), os quais realizaram um protocolo de treinamento periodizado de natação, durante 6 semanas, após 2 semanas de adaptação. O limiar anaeróbio pelo teste de lactato mínimo (Lam) foi realizado para a separação dos animais em grupos homogêneos e para verificação dos efeitos do treinamento. Foram verificados o efeito do treinamento físico periodizado sobre o acúmulo de atividade física espontânea (AFE -deslocamento pela filmagem) e voluntária (AFV - roda de atividades), além da verificação da interferência da dieta hiperlipídica na atividade física espontânea e voluntária, no perfil lipídico (triglicerídeos, HDL), glicemia, comportamento alimentar, medidas e gordura corporal. A análise de variância foi utilizada para verificar as diferenças das variáveis entre os grupos, enquanto que a correlação de Pearson foi utilizada para verificar a associação entre a carga de treinamento com a ingestão alimentar, AFE e AFV, considerando alfa de 5%. Na primeira etapa a acurácia e a confiabilidade do sistema de videogrametria para a análise do deslocamento foi confirmada. Na segunda etapa verificou-se que os valores de glicemia, HDL e TG não apresentaram diferença significativa entre os grupos após 8 semanas de experimento. Os grupos treinados apresentaram maiores valores de Lam em relação ao grupo GC e GDH e não apresentaram ganho excessivo de gordura corporal. Em relação ao consumo alimentar, os grupos alimentados com dieta hiperlipídica apresentaram menor consumo calórico em relação aos grupos alimentados com dieta padrão. O acúmulo de AFV não apresentou diferença entre os grupos, porém a AFE foi maior no grupo controle em relação ao demais. A associação entre a carga de treino total semanal e o consumo alimentar e carga de treino total semanal e AFV foram fracas. Para associação entre a carga de treino total semanal e a AFE, houve forte associação com o grupo TP . O TP e a dieta hiperlipídica levaram a uma redução da AFE, assim como nas semanas onde os treinamentos foram intensos houve um nível de AFE menor. Nosso resultados indicam que existem fatores biológicos responsivos ao treinamento e a dieta hiperlipídica capazes de interferir nos níveis de AFE dos animais. / It is believed that there is a set point that limits the energy expenditure using as a strategy the control of the levels of physical activity in our organism aiming the maintenance of the energy homeostasis similar to other variables regulated biologically. The present study aims to investigate whether the periodized training and the hyperlipidic diet influence the levels of spontaneous and voluntary physical activity and the alteration in dietary intake of rats. For this, the study was divided in two phases: in the first the objective was to verify the accuracy and reliability of a videogrammetry system in the analysis of Wistar rats displacement and in the second stage, to verify if the increase of the physical activity through periodized training is accompanied by a compensatory reduction in physical activity and alteration in food intake. Rats (n = 32) Wistar (Rattus Norvegicus Albinus Wistar) with initial age of 120 days were kept in collective cages for 8 weeks, after separation of the groups: Control group (CG), Control group with hyperlipidic diet (GHD), Periodized training group (PT), and Periodic training group hyperlipidic diet (PTH), who performed a periodized swimming training protocol for 6 weeks after 2 weeks of adaptation. The anaerobic threshold by lactate minimum (Lam) test was performed to separate the animals into homogeneous groups and to verify the effects of the training. The effect of the periodic physical training on the accumulation of spontaneous physical activity (SPA - displacement by filming) and voluntary (VPA - activity wheel) was verified, as well as the verification of the interference of the hyperlipidic diet in spontaneous and voluntary physical activity, in the lipid profile (triglycerides, HDL), glycemia, eating behavior, measures and body fat. The analysis of variance was used to verify the differences of the variables between the groups, while the Pearson correlation was used to verify the association between training load and food intake, SPA and VPA, considering alpha of 5%. In the first phase the accuracy and reliability of the videogrammetry system for displacement analysis was confirmed. In the second phase, it was verified that the values of glycemia, HDL and TG did not present significant difference between the groups after 8 weeks of experiment. The trained groups had higher values of Lam in relation to the CG and GHD groups and did not present an excessive gain of body fat. In relation to the food consumption, the groups fed with a hyperlipid diet presented lower caloric intake than the groups fed a standard diet. The VPA accumulation did not present difference between the groups, but the SPA was higher in the control group in relation to the others. The association between total weekly training load and total weekly training and feeding load and VPA were weak. For association between the total weekly training load and the SPA, there was a strong association with the PT group. The PT and the hyperlipid diet led to the reduction of SPA, just as in the weeks where the training was intense there was a lower level of SPA. Our results indicate that there are training-responsive biological factors and the hyperlipidic diet capable of interfering with the SPA levels of the animals.

Efekt kalorického příjmu a aromatických látek na kvalitu lidské tělesné vůně / The effect of caloric intake and aromatic substances on the human body odour quality

Previous work has shown that mate choice is in part affected by cues associated with quality and condition of potential mate. These cues could be based on visual characteristics or as shown recently on chemical cues. There is growing evidence which suggest that body odour plays relatively significant role in mate choice. Moreover, body odour is to some extent affected by ingested diet. Therefore, we focused on the effect of caloric restriction and garlic consumption on axilary odour. Our results suggest that restoration of food intake improve body odour in terms of pleasantness and attractiveness (probably as a consequence of recommended specific diet), but during caloric restriction it is rated lower. The garlic consumption improves hedonic assessment of body odour. This effect could be ascribed to his well-known health effects. Our findings thus confirm assumption that ingested diet affects body odour and these changes in odour attractiveness could serve as a cue to individual condition and quality.

An investigation of body fat accrual in an ethnically diverse cohort of British Columbian children and youth: patterns, obesity classification, and determinants

Obesity during childhood and adolescence is a serious public health concern in Canada and globally. Obesity is a complex disease with genetic, environmental, social, and behavioural determinants. However, our understanding of obesity and its development is limited by a reliance on proxy measurements of adiposity such as body mass index (BMI) and cross-sectional study designs that limit our ability to assess temporality. In this dissertation, I present the first set of body fat percent (BF%) accrual and velocity percentile curves for Canadian children and youth, investigate the relationship between BMI- and BF%-based definitions of obesity, and examine the longitudinal influence of sedentary time, moderate-to-vigorous physical activity (MVPA) and caloric intake on the development of BF%. My analyses are based on the UBC Healthy Bones III Study (HBSIII), a mixed longitudinal study of boys and girls aged 8-12 years at baseline, measured between 1999 and 2012. In HBSIII, adiposity was measured directly as BF% from total body dual energy x-ray absorptiometry (DXA) scans and MVPA and sedentary time were measured objectively using accelerometers. For the first study in my dissertation, I used generalized additive models for location scale and shape (GAMLSS) to develop sex- and ethnic-specific BF% accrual and velocity percentile curves. I present separate curves for Asian and Caucasian boys and girls aged 9-19 years at the 3rd, 10th, 25th, 50th, 75th, 95th, and 97th centiles. In this descriptive study, I found materially different shaped BF% percentile curves for Asian and Caucasian girls but not for boys. Second, I examined the relationship between BMI- and BF%-based definitions of obesity for Asian and Caucasian boys and girls aged 9-19 years. I used multivariable regression models, sensitivity and specificity analysis, receiver operating characteristic (ROC) curves, and Youden’s Index to explore this relationship. I found that BMI identified <50% of those classified with obesity based on BF%, and that classification performance of BMI differed significantly by age and sex subgroups for Asian and Caucasians. In my third analysis, I explored the longitudinal relationship between BF% and sedentary time, MVPA, and caloric intake as boys and girls mature. I fit polynomial multilevel models using MO (years from age at peak height velocity, APHV) as the time variable. Rate of change in BF% across maturity differed between boys and girls and differences in MVPA, sedentary time, and caloric intake between individuals influenced BF% at APHV (MO=0) and rate of change in BF% across maturity. Together, these studies advance our understanding of how body fat accrues as children and youth mature, and highlight the heterogeneity in predictors of adiposity and adiposity measurement accuracy across age, sex, and ethnic groups. / Graduate

Sleep and Eating Behavior Among Adolescent Females with Overweight or Obesity: The Role of Appetite-Related Cognitive Processes

Insufficient sleep duration and poor sleep quality can potentiate weight gain and obesity in adolescents. Furthermore, overweight and obese females are at unique risk for insufficient sleep and associated health complications. We examined self-reported sleep duration and self-reported adequacy of sleep duration as potential moderators of the relationship between eating behavior and several cognitive processes including hedonic hunger, executive dysfunction, and self-control. We used a multisystemic conceptual framework to highlight the pathways that may explain the relationship between sleep behaviors and the Healthy Eating Index (HEI). The study employed a cross-sectional design. Participants completed baseline measures of height and weight, self-control, executive functioning, hedonic hunger, and sleep functioning. Self-report of poor sleep adequacy directly influenced executive dysfunction which consequently explained a decrease in self-control functioning. Moreover, we evaluated whether sleep deprivation and extension influences caloric intake. We offer novel yet promising evidence that 9 hrs of sleep fostered greater self-control functioning which promoted intake of 484.69 fewer calories per day compared to sleep deprivation. Our study is well-positioned to improve understanding of individual cognitive subsystems and the mechanism that underlies the influence of sleep behavior on weight-related behaviors among overweight and obese females. Findings from this study have the potential to inform health interventions that promote healthy eating and sleep behaviors.
