apresentada neste trabalho. ? verificado a varia??o dos par?metros diel?tricos da fruta
em diversas frequ?ncias, durante o processo de matura??o, considerando as
transforma??es fisiol?gicas para a taxa de respira??o, forma??o de etileno, do amido,
dos a??cares e ?cidos sol?veis titul?veis. Os resultados mostram uma grande correla??o
entre os par?metros diel?tricos e o est?dio de matura??o da manga, quando se verifica a
varia??o di?ria da constante diel?trica e fator de perdas. A assinatura diel?trica obtida
permite identificar o pico de matura??o, que, para o manejo da amostra utilizada, ocorre
aproximadamente no sexto dia ap?s a colheita. Os resultados tamb?m sugerem que os
par?metros diel?tricos podem ser importantes indicadores na an?lise da manga Tommy
Atkins, principalmente pela possibilidade de efetuar medidas externas, n?o destrutivas,
o que pode se tornar uma ferramenta imprescind?vel para os produtores.

Previous issue date: 2017-10-26 / Coordena??o de Aperfei?oamento de Pessoal de N?vel Superior - CAPES / Adenylosuccinate lyase (ASL) belongs to aspartase/fumarase superfamily of enzymes which share a general acid-base catalytic mechanism with ?-elimination of fumarate as common product. ASL is involved in both de novo and salvage pathways of purine biosynthesis. Cloning, expression, and a method to obtain homogeneous recombinant ASL from Leishmania braziliensis (LbASL) are described. Mass spectrometry analysis of recombinant LbASL, oligomeric state determination and multiple sequence alignment are presented. Steady-state kinetics of LbASL showed a Michaelis-Menten pattern. Isothermal titration calorimetry binding assays suggested that LbASL follows a Uni-Bi ordered kinetic mechanism, in which release of fumarate is followed by AMP to yield free enzyme. Initial velocity data for the reverse reaction and the Haldane relationship allowed calculation of an unfavorable equilibrium constant for LbASL-catalyzed chemical reaction. The activation energy and thermodynamic activation parameters were estimated. Solvent kinetic isotope effects V/K and V suggest a modest contribution of solvent proton transference during the rate-limiting step of the reaction. Proton inventory data show that the modest normal effect on V arises from a single protonic site, and the transition state fractionation factor value of 0.74 suggests participation of solvent proton transfer in transition-state vibrations perpendicular to the reaction coordinate. pH-rate profiles for kcat and kcat/KM suggested amino acid residues involved in, respectively, catalysis and substrate binding. A model of LbASL was built to provide a structural basis for the experimental data. A better understanding of the mode of action of LbASL is useful for the rational design of antileishmaniasis agents. / A enzima Adenilosuccinato liase (ASL) pertence a superfam?lia de enzimas aspartase/fumarase, as quais compartilham o mecanismo catal?tico ?cido-b?sico com ?-elimina??o de fumarato como o produto comum. A ASL est? envolvida tanto na bioss?ntese de novo quanto na via de salvamento de purinas. Aqui s?o descritos os m?todos de clonagem, express?o e obten??o da prote?na recombinante ASL de Leishmania braziliensis (LbASL) na sua forma homog?nea. An?lises da prote?na recombinante por espectrometria de massa, determina??o do estado oligom?rico e alinhamento m?ltiplo de sequ?ncias tamb?m s?o apresentados. Ensaios de cin?tica em estado estacion?rio mostraram que a LbASL segue o perfil de Michaelis-Menten. Experimentos de titula??o isot?rmica por calorimetria sugerem que a LbASL segue um mecanismo cin?tico Uni-Bi ordenado, no qual o fumarato ? liberado primeiro do s?tio ativo seguido pelo AMP. Dados de velocidade iniciais para a rea??o reversa e a rela??o de Haldane permitiram calcular uma constante de equil?brio desfavor?vel para a rea??o qu?mica catalisada pela enzima. Os par?metros de energia de ativa??o e termodin?mica tamb?m foram estimados. Os efeitos isot?picos do solvente V/K e V sugerem uma modesta contribui??o da transfer?ncia de pr?tons do solvente durante o passo limitante da rea??o. Os dados obtidos no invent?rio de pr?tons mostram um modesto efeito em V resultante de um ?nico s?tio prot?nico, e o valor de transi??o do fator de estado de fracionamento de 0,74 sugere a participa??o da transfer?ncia de pr?tons do solvente em vibra??es de estado de transi??o perpendiculares ? coordenada da rea??o. Experimentos de perfil de pH para kcat e kcat/KM sugerem os res?duos de amino?cidos envolvidos, respectivamente, na cat?lise e liga??o do substrato. due to its range of applications, easily obtained in the laboratory and reusability after
calcination, so the knowledge regarding their properties is of utmost importance. In this
study were synthesized layered double hydroxides of two systems, Mg-Al and Zn-Al,
and such materials were analyzed with X-ray diffraction and, from these data, we
determined the volume density, planar atomic density, size crystallite, lattice
parameters, interplanar spacing and interlayer space available. Such materials were
also subjected to thermogravimetric analysis reasons for heating 5, 10, 20 and 25 ? C /
min to determine kinetic parameters for the formation of metaphases HTD and HTB
based on theoretical models Ozawa, Flynn-Wall Starink and Model Free Kinetics. In
addition, the layered double hydroxides synthesized in this working ratios were
calcined heating 2.5 ? C / min and 20 ? C / min, and tested for adsorption of nitrate
anion in aqueous solution batch system at time intervals 5 min, 15 min, 30 min, 1h, 2h
and 4h. Such calcined materials were also subjected to exposure to the atmosphere
and at intervals of 1 week, 2 weeks and 1 month were analyzed by infrared
spectroscopy to study the kinetics of regeneration determining structural called
"memory effect" / Os Hidr?xidos Duplos Lamelares tem se tornado materiais extremamente
promissores devido a sua gama de aplica??es, f?cil obten??o em laborat?rio e a

Previous issue date: 2007-03-28 / Petr?leo Brasileiro SA - PETROBRAS / The production of oil and gas is usually accompanied by the production of water, also known as produced water. Studies were conducted in platforms that discharge produced water in the Atlantic Ocean due to oil and gas production by Petrobras from 1996 to 2006 in the following basins: Santos (Brazilian south region), Campos (Brazilian southeast region) and Ceara (Brazilian northeast region). This study encompasses chemical composition, toxicological effects, discharge volumes, and produced water behavior after releasing in the ocean, including dispersion plumes modeling and monitoring data of the marine environment. The concentration medians for a sampling of 50 samples were: ammonia (70 mg L-1), boron (1.3 mg L1), iron (7.4 mg L-1), BTEX (4.6 mg L-1), PAH (0.53 mg L-1), TPH (28 mg L-1); phenols (1.3 mg L-1) and radioisotopes (0.15 Bq L-1 for 226Ra and 0.09 Bq L-1 for 228Ra). The concentrations of the organic and inorganic parameters observed for the Brazilian platforms were similar to the international reference data for the produced water in the North Sea and in other regions of the world. It was found significant differences in concentrations of the following parameters: BTEX (p<0.0001), phenols (p=0.0212), boron (p<0.0001), iron (p<0.0001) and toxicological response in sea urchin Lytechinus variegatus (p<0.0001) when considering two distinguished groups, platforms from southeast and northeast Region (PCR-1). Significant differences were not observed among the other parameters. In platforms with large gas production, the monoaromatic concentrations (BTEX from 15.8 to 21.6 mg L-1) and phenols (from 2 to 83 mg L-1) were higher than in oil plataforms (median concentrations of BTEX were 4.6 mg L-1 for n=53, and of phenols were 1.3 mg L-1 for n=46). It was also conducted a study about the influence of dispersion plumes of produced water in the vicinity of six platforms of oil and gas production (P-26, PPG-1, PCR-1, P-32, SS-06), and in a hypothetical critical scenario using the chemical characteristics of each effluent. Through this study, using CORMIX and CHEMMAP models for dispersion plumes simulation of the produced water discharges, it was possible to obtain the dilution dimension in the ocean after those discharges. The dispersion plumes of the produced water modelling in field vicinity showed dilutions of 700 to 900 times for the first 30-40 meters from the platform PCR-1 discharge point; 100 times for the platform P-32, with 30 meters of distance; 150 times for the platform P-26, with 40 meters of distance; 100 times for the platform PPG-1, with 130 meters of distance; 280 to 350 times for the platform SS-06, with 130 meters of distance, 100 times for the hypothetical critical scenario, with the 130 meters of distance. The dilutions continue in the far field, and with the results of the simulations, it was possible to verify that all the parameters presented concentrations bellow the maximum values established by Brazilian legislation for seawater (CONAMA 357/05 - Class 1), before the 500 meters distance of the discharge point. These results were in agreement with the field measurements. Although, in general results for the Brazilian produced water presented toxicological effects for marine organisms, it was verified that dilutions of 100 times were sufficient for not causing toxicological responses. Field monitoring data of the seawater around the Pargo, Pampo and PCR-1 platforms did not demonstrate toxicity in the seawater close to these platforms. The results of environmental monitoring in seawater and sediments proved that alterations were not detected for environmental quality in areas under direct influence of the oil production activities in the Campos and Ceara Basin, as according to results obtained in the dispersion plume modelling for the produced water discharge / A produ??o de ?leo e g?s ? normalmente acompanhada da produ??o de ?gua, que ? denominada ?gua produzida. Nos demais par?metros n?o foram observadas diferen?as significativas. Nos casos de maior produ??o de g?s as concentra??es de monoarom?ticos (BTEX de 15, 8 a 21,6 mg L-1) e fen?is (2 a 83 mg L-1) foram mais elevadas do que nas plataformas de ?leo (concentra??es medianas de BTEX de 4,6 mg L-1 para n=53, e de fen?is de 1,3 mg L-1, para n=46).
Foi realizado um estudo sobre a influ?ncia das plumas de dispers?o da ?gua produzida no entorno de seis plataformas de produ??o de ?leo e g?s (P-26, PPG-1, PCR-1, P-32, SS-06, e um cen?rio cr?tico hipot?tico) usando as caracter?sticas qu?micas de cada um dos efluentes. Atrav?s deste estudo, utilizando os modelos CORMIX e CHEMMAP para simula??o de plumas de dispers?o da ?gua produzida descartada em mar foi poss?vel obter uma dimens?o das dilui??es ocorridas no oceano a partir destes descartes. As modelagens das plumas de dispers?o da ?gua produzida para o campo pr?ximo mostraram dilui??es de cerca de: 700 a 900 vezes logo nos primeiros 30 a 40 metros do ponto de descarte para a plataforma PCR-1; 100 vezes para a plataforma P-32, em dist?ncia de 30 metros; 150 vezes para a plataforma P-26, em dist?ncia de 40 metros; 100 vezes para a plataforma PPG-1, em dist?ncia de 130 metros; 280 a 350 vezes para a plataforma SS-06, em dist?ncia de 130 metros, 100 vezes para o cen?rio critico hipot?tico, em dist?ncia de 130 metros. As dilui??es continuam no campo afastado, e com os resultados das simula??es foi poss?vel verificar que todos os par?metros atingem concentra??es abaixo dos valores limites admitidos pela Resolu??o CONAMA 357/05 para ?guas salinas de classe 1, antes de 500m de dist?ncia do ponto de descarte. Estes resultados est?o coerentes com as medidas de campo. Os estudos realizados na ?gua produzida brasileira apresentaram, de maneira geral, toxicidade para os organismos marinhos, no entanto verifica-se que dilui??es de 100 vezes s?o suficientes para n?o causar o efeito t?xico. Dados de monitoramento da ?gua do mar em torno das plataformas de Pargo, Pampo e PCR-1 n?o evidenciaram toxicidade na ?gua do mar no entorno daquelas plataformas.
Os resultados de monitoramento ambiental de ?gua do mar e sedimentos comprovam que n?o foram detectadas altera??es da qualidade ambiental na ?rea sob influ?ncia direta das atividades de produ??o de petr?leo nas Bacias de Campos e do Cear?, corroborando todos os resultados de modelagem da pluma de dispers?o do descarte da ?gua produzida

As concentra??es dos par?metros org?nicos e inorg?nicos observadas para as plataformas brasileiras foram similares aos dados de refer?ncias mundiais para ?gua produzida no Mar do Norte e outras regi?es do mundo. Quando considerados os dois grupos distintos, plataformas da regi?o sudeste e nordeste (PCR-1) foram verificadas diferen?as significativas nas concentra??es dos par?metros: BTEX (p<0,0001), Fen?is (p=0,0212), Boro (p<0,0001), Ferro (p<0,0001), Toxicidade com organismo Lytechnus Variegatus (p<0,0001). Nos demais par?metros n?o foram observadas diferen?as significativas. Nos casos de maior produ??o de g?s as concentra??es de monoarom?ticos (BTEX de 15, 8 a 21,6 mg L-1) e fen?is (2 a 83 mg L-1) foram mais elevadas do que nas plataformas de ?leo (concentra??es medianas de BTEX de 4,6 mg L-1 para n=53, e de fen?is de 1,3 mg L-1, para n=46).
Foi realizado um estudo sobre a influ?ncia das plumas de dispers?o da ?gua produzida no entorno de seis plataformas de produ??o de ?leo e g?s (P-26, PPG-1, PCR-1, P-32, SS-06, e um cen?rio cr?tico hipot?tico) usando as caracter?sticas qu?micas de cada um dos efluentes. Atrav?s deste estudo, utilizando os modelos CORMIX e CHEMMAP para simula??o de plumas de dispers?o da ?gua produzida descartada em mar foi poss?vel obter uma dimens?o das dilui??es ocorridas no oceano a partir destes descartes. As modelagens das plumas de dispers?o da ?gua produzida para o campo pr?ximo mostraram dilui??es de cerca de: 700 a 900 vezes logo nos primeiros 30 a 40 metros do ponto de descarte para a plataforma PCR-1; 100 vezes para a plataforma P-32, em dist?ncia de 30 metros; 150 vezes para a plataforma P-26, em dist?ncia de 40 metros; 100 vezes para a plataforma PPG-1, em dist?ncia de 130 metros; 280 a 350 vezes para a plataforma SS-06, em dist?ncia de 130 metros, 100 vezes para o cen?rio critico hipot?tico, em dist?ncia de 130 metros. As dilui??es continuam no campo afastado, e com os resultados das simula??es foi poss?vel verificar que todos os par?metros atingem concentra??es abaixo dos valores limites admitidos pela Resolu??o CONAMA 357/05 para ?guas salinas de classe 1, antes de 500m de dist?ncia do ponto de descarte. Estes resultados est?o coerentes com as medidas de campo. Os estudos realizados na ?gua produzida brasileira apresentaram, de maneira geral, toxicidade para os organismos marinhos, no entanto verifica-se que dilui??es de 100 vezes s?o suficientes para n?o causar o efeito t?xico. Dados de monitoramento da ?gua do mar em torno das plataformas de Pargo, Pampo e PCR-1 n?o evidenciaram toxicidade na ?gua do mar no entorno daquelas plataformas.
Os resultados de monitoramento ambiental de ?gua do mar e sedimentos comprovam que n?o foram detectadas altera??es da qualidade ambiental na ?rea sob influ?ncia direta das atividades de produ??o de petr?leo nas Bacias de Campos e do Cear?, corroborando todos os resultados de modelagem da pluma de dispers?o do descarte da ?gua produzida

Some organisms similar to B. hominis have been described in a range of non-human hosts and are denominated Blastocystis sp., but their zoonotic potential still needs clarification. The objective of this work was to investigate the occurrence of natural infection by Blastocystis sp. in three species of domesticated birds sold at cooperative markets (with individual vendor stalls) in two districts of the city of Rio de Janeiro and to characterize the morphology of these organisms found in bird droppings, to enable a precise diagnosis. A total of 214 birds were purchased: 35 ducks (Anas platyrhynchos) from market A and 38 from market B, 35 quails (Coturnix japonica) from market A and 36 from market B and 35 chicks (Gallus gallus) from market A and 35 from market B. Of the birds studied from market A, 8 chicks (22.9%), 15 quails (42.9%) and 13 ducks (37.1%) presented biological forms of Blastocystys sp. in their droppings, while at the market B, the respective figures were 15 chicks (42.9%), 2 quails (5.56%) and 21 ducks (55.3%). There was no statistically significant difference in relation to the occurrence of infection in ducks and chicks between the two markets studied, but there was between the quails at the two markets. Considering the number of infected birds at the two markets irrespective of species, the difference was statistically significant. From the observations of the stained fecal smears of the animals that were positive for Blastocystis sp., it was possible to characterize six forms of organisms, namely vacuolar, granular, ameboid, cystic, avacuolar and multivacuolar. It was also possible to characterize the reproductive forms of Blastocystis sp. as binary division, endodyogeny, budding, plamotomy and schizogeny. Even though there are no studies confirming the zoonotic and pathogenic potential of Blastocystis sp., either to human or animal hosts, we suggest that environmental contamination can be occurring at these markets, due to the poor hygienic and sanitary conditions, thus posing a risk to both the animals and the people who circulate there. The knowledge of evaluative forms that were found and characterized in this work is important to gives a precise and faithful diagnosis of Blastocystis sp. in dropping. / Blastocystis hominis ? um dos parasitos de maior preval?ncia no trato gastrintestinal de humanos, por?m seu papel patog?nico ainda permanece incerto. Alguns organismos semelhantes ? B. hominis t?m sido descritos em uma variedade de hospedeiros n?o-humanos sendo denominado Blastocystis sp., mas seu potencial zoon?tico n?o est? esclarecido. O objetivo desse trabalho foi verificar a ocorr?ncia de infec??o natural de Blastocystis sp. em tr?s esp?cies de aves dom?sticas comercializadas em dois mercados municipais do Rio de Janeiro, caracterizando morfologicamente esses organismos encontrados em material fecal para a emiss?o de um diagn?stico preciso. Ao considerarmos o n?mero de aves infectadas nos dois mercados, independentemente da esp?cie, observou-se que n?o houve uma diferen?a estatisticamente significativa entre os dois mercados. Das observa??es de esfrega?o de fezes coradas com Giemsa daqueles animais que apresentaram positividade para Blastocystis sp., foi poss?vel caracterizar seis formas do organismo, sendo estas: vacuolar, granular, ameb?ide, c?sticas, avacuolar e multivacuolar. Tamb?m foi poss?vel caracterizar as formas reprodutivas de Blastocystis sp. como: divis?o bin?ria, endodiogenia, brotamento, plamatomia e esquizogonia. Mesmo n?o tendo estudos que confirmem o potencial zoon?tico e patog?nico de Blastocystis sp., tanto para hospedeiros humanos quanto para animais, sugere-se que nesses mercados de comercializa??o pode ocorrer a contamina??o ambiental, devido ?s condi??es higi?nicas e sanit?rias serem de p?ssima qualidade, sendo um risco para os animais, assim como para a popula??o humana que comumente freq?entam esses ambientes. an?logas ou similares na engenharia de po?os de petr?leo, como por exemplo, os
antiespumantes, dispersantes ou redutores de fric??o, controladores de filtrado e retardadores
de pega. Este estudo foi desenvolvido com o objetivo de avaliar e especificar produtos
qu?micos alternativos para compor sistemas de aditivos adequados ? formula??o de pastas de
cimento para aplica??o em po?os de petr?leo onshore caracterizados por temperaturas a partir
de 180?F (82?C), po?os depletados e po?os sujeito ? inje??o de vapor. Durante o per?odo de
execu??o do estudo, foram selecionados produtos qu?micos de acordo com sua fun??o. Estes
produtos foram especificados segundo normas internas da Petrobras e os testes foram
realizados de acordo com a norma API RP 10B. Alguns dos aditivos apresentaram grande
potencial para aplica??o em pastas de cimento para temperaturas elevadas, incluindo
antiespumantes, controladores de filtrado, dispersantes e retardadores de pega. / The civil construction industry employs a wide diversity of admixtures developed to
use in concrete and mortars, but that exhibits excellent performance for oilwell slurries. Some
properties meet analogous functions, for example, antifoam, dispersants or friction reducers,
fluid loss control agent and set retarders. This study was developed with the objective to
evaluate and specify alternative chemical products to be used as admixtures for the
formulation of cement slurries for application in onshore oil wells, characterized by
temperatures of 180?F (82?C), mature wells and wells subjected to steam injection. During
the study, chemical products were selected according its functions. These additives were
specified according to PETROBRAS internal standards and the tests were realized according
to API RP 10B standards. Some additives presented great potential for application in cement
slurries for high temperatures, including antifoams, fluid loss control agents, dispersants and
setting retarders.

As part of this cultural heritage, highlights the importance of historical buildings. For the principles of restoration are met its important to know the state of conservation and the existing materials. Given the above, this work aims to study the pathological manifestations of the mortar coating from the Grupo Escolar Augusto Severo. For this, was conducted historical survey of the building, on-site visits with visual observation, collecting samples of mortar coating and characterization through visual and laboratory analysis, XRF, XRD, TG/DTG, OM and SEM-EDS. From the observations, pathological manifestations were found as: cracks, detachment of the coating, dirt, use of inappropriate material basis of cement, efflorescence, mold and mildew and also incompatible material base of cement. It was found that the pathological manifestations were intensified due to lack of coverage in some spots and especially the abandonment of the building, that many years does not have restoration or any other type of preventive and corrective maintenance. The results of laboratory tests indicated that the mortars studied are based on lime, with calcitic nature, and siliceous aggregate, with the presence of clay in two samples. From the samples collected, two are composed of lime, sand and clay and two by lime and sand. In this regard, it is important to conduct periodic inspections and maintenance, as well as carrying out restoration with use of material compatible with the original / A busca pela preserva??o do patrim?nio cultural deve ser uma constante, para que n?o haja a perda da identidade hist?rica e cultural de um pa?s. Como parte deste patrim?nio cultural, destaca-se a import?ncia das edifica??es hist?ricas. Para que os princ?pios de restauro sejam atendidos ? de suma import?ncia conhecer o estado de conserva??o desses bens e os materiais existentes. Diante do exposto, este trabalho tem como objetivo estudar as manifesta??es patol?gicas presentes nos revestimentos argamassados, bem como caracterizar as argamassas, de maior valor patrimonial, do Grupo Escolar Augusto Severo. Para isso, foi realizado um levantamento hist?rico da edifica??o, visitas in loco com observa??o visual e registro fotogr?fico, coleta de amostras de argamassas de revestimento e caracteriza??o por meio de an?lise visual e laboratorial, FRX, DRX, TG/DTG e MEV-EDS. A partir das observa??es realizadas, foram identificadas manifesta??es patol?gicas como: fissuras, desprendimento do revestimento, sujidade, efloresc?ncia, mofo e bolor, al?m da utiliza??o de material inadequado ? base de cimento. Constatou-se que as manifesta??es patol?gicas foram intensificadas devido ? falta de cobertura em alguns pontos e, sobretudo pelo abandono da edifica??o, que h? muitos anos n?o passa por restaura??o ou qualquer outro tipo de manuten??o preventiva e corretiva. O mesmo tem como objetivo caracterizar tecnologicamente as mat?rias-primas: res?duo termopl?stico de bot?o e res?duos de gesso para incorporar ao cimento, visando a produ??o de comp?sitos com propriedades de isola??o t?rmica: condutividade, difusividade, capacidade calor?fica e resistividade. thermal comfort, plus the advent of new laws regulating the minimum requirements of
comfort, disposal of solid industrial waste, construction waste, the requirements of consumers
by adopting construction methods "cleaner", encouraged the development of this work. Aims
technologically characterize the composite proposed in three types of samples (10%, 30% and
50% of thermoset plastic industrial waste) and raw materials: gypsum waste, cement and
plastic thermosetting industrial waste in order to produce the composite with properties of
thermal insulation: conductivity, thermal diffusivity, specific heat and resistivity. The
physical, structural and morphological properties of the raw materials were investigated by
thermogravimetry analysis (TG / DSC), X-ray diffraction (DRX), X-ray fluorescence (FXR)
and scanning electron microscopy (MEV). Obtaining mechanical properties through the
compression strength test. The analysis results indicate characteristics suitable for cement
matrix composite production with the addition of thermosetting plastic industrial waste and
gypsum waste, with potential application of these materials in composites with properties of
thermal insulation. Finally, assessing what proportion showed up with better performance.

Chitin is
hydrolyzed by a chitinolytic system classified as: endo-chitinases, exo-chitinases and N-acetyl-b-D-glucosaminidases. In this study a Litochitinase1 extracted from the cephalotorax of the shrimp Litopenaeus Schmitt was purified 987.32 times using ionexchange chromatography DEAE-Biogel and molecular exclusion Sephacryl S-200.
These enzyme presented a molecular mass of about 28.5 kDa. The results, after kinetic assay with the Litochitinase1 using as substrate p-nitrophenyl-N-acetyl-b-Dglucosaminideo, showed apparent Km of 0.51 mM, optimal activity at pH ranging from
5.0 to 6.0, optimum temperature at 55?C and stability when pre-incubated at temperatures of 25, 37, 45, 50 and 55?C. The enzyme showed a range of stability at pH 4.0 to 5.5. HgCl2 inhibited Litochitinase1 while MgCl2 enhances its activity.
Antimicrobial tests showed that Litochitinase1 present activity against gram-negative bacterium Escherichia coli in the 800 μg/mL concentration. The larvicidal activity against Aedes aegypti was investigated using crude extracts, F-III (50-80%) and Litochitinase1 at 24 and 48 hours. The results showed larvicidal activity in all these samples with EC50 values of 6.59 mg/mL for crude extract, 5.36 mg/mL for F-III and 0.71 mg/mL for Litochitinase1 at 24 hours and 3.22 and 0.49 mg/mL for the F-III and Litochitinase1 at 48 hours, respectively. Other experiments confirmed the presence of chitin in the midgut of Aedes aegypti larvae, which may be suffering the action of
Litochitinase1 killing the larvae, but also the absence of contaminating proteins as serine proteinase inhibitors and lectins in the crude extract, F-III and Litochitinase1, indicating that the death of the larvae is by action of the Litochitinase1. We also
observed that the enzymes extracted from intestinal homogenate of the larvae no have activity on Litochitinase1. These results indicate that the enzyme can be used as an alternative to control of infections caused by Escherichia coli and reducing the infestation of the mosquito vector of dengue. / As quitinases s?o enzimas envolvidas na degrada??o da quitina e est?o presentes em uma gama de organismos, inclusive os que n?o cont?m quitina, tais como bact?rias, v?rus, plantas e animais desempenhando importantes papeis fisiol?gicos e ecol?gicos. contra Aedes aegypti foi investigada com os extratos brutos, F-III (50-80%) e na Litochitinase1 com tempo de 24 e 48 horas. Os resultados obtidos mostraram atividade larvicida em todas estas amostras com valores de EC50 de 6,59 mg/mL
para o extrato bruto, 5,36 mg/mL para F-III e 0,71 mg/mL para Litochitinase1 com 24 horas de ensaio e 3,22 e 0,49 mg/mL, para F-III e Litochitinase1 no tempo de 48 horas. Outros experimentos realizados confirmaram a presen?a de quitina no intestino m?dio das larvas de Aedes aegypti, as quais podem estar sofrendo a a??o da Litochitinase1 provocando suas mortes, como ainda a aus?ncia de prote?nas bioativas como inibidores de proteases ser?nicas e lectinas no extrato bruto, F-III e Litochitinase1, indicando que a morte das larvas ? mesmo por a??o da
Litochitinase1. Observou-se ainda que, as enzimas extra?das do homogenato intestinal das larvas n?o interferiram na atividade da Litochitinase1. Estes resultados indicam que esta enzima pode ser usada como alternativa para controlar infec??es causadas por Escherichia coli, como tamb?m na redu??o da infesta??o do mosquito vetor da dengue.
