Chemical contaminants in Chinese aquaculture imports, U.S. import security, and exposure assessment amongst vulnerable sub-populations

Doctor of Philosophy / Food Science / Justin Kastner / Many Chinese aquaculture farmers use unapproved chemicals to treat their fish, many of which are diseased as a result of the country’s poor waste management and environmental practices. During 2006-2007, the United States (U.S.), the European Union, and Japan rejected large amounts of Chinese seafood imports due to the presence of unapproved chemicals or the presence of approved chemicals at concentrations that exceeded permitted levels. This dissertation examines the sources of environmental health and food safety problems in China; it also examines how effective the U.S. and Chinese governments’ regulations are in protecting consumers from hazards in Chinese aquaculture products. The study looks at specific chemical contaminants found in Chinese aquaculture imports, explores their potential toxicity or carcinogenicity, and examines the reasons for their prohibition from human food. The study exploits the available U.S. seafood consumption patterns (courtesy of the National Health and Nutrition Examination Survey—NHANES—database) and then uses probabilistic modeling (courtesy of CREMe Software Limited) to determine the extent to which specific sensitive U.S. consumer subpopulations were exposed to aquaculture chemical contaminants in the food supply in a contrived scenario using real consumption data (from NHANES) and actual contamination data (from the FDA). The study compares exposure between children and adult consumers, and also looks at exposure among women aged 18 years and older and the elderly aged 60 years and older. This study suggests a strong likelihood that NHANES children, as well as female consumers aged 18 years and older and elderly consumers aged 60 years and older, were (in the contrived scenario) all exposed to violative intake levels of chemical contaminants from Chinese aquaculture imports. Children forming the 99.5th and 99.9th percentiles of NHANES seafood consumers were exposed to higher levels of nitrofuran, gentian violet, and malachite green contaminants per kilogram of body weight than were their adult counterparts. Conversely, children were exposed to lower levels of fluoroquinolone contaminants per kilogram of body weight than were their adult counterparts. The 50th, 95th, and 99.9th percentiles of female consumers aged 18 and older and elderly consumers aged 60 years and older were exposed to violative daily intake levels of contaminants in Chinese aquaculture. The study concludes by examining what the U.S. and Chinese governments should do to address aquaculture safety.

Le travail de thèse décrit dans ce manuscrit porte sur l’étude de la présence de biocides désinfectants dans les produits laitiers et sur l’impact des procédés de transformation du lait sur l’apparition de produits néoformés. Les biocides désinfectants sont des composés chimiques utilisés quotidiennement en industrie laitière lors des procédures de nettoyage-désinfection (ND) des surfaces en contact avec les aliments. Les ammoniums quaternaires de type chlorure de Benzalkonium (BACs) et chlorure de dialkyldimethylammonium (DDACs), et l’Aminopropyldodécylpropane diamine sont les désinfectants les plus largement utilisés en industrie laitière. Ces biocides peuvent entrainer des résidus sur les surfaces alimentaires, ce qui présente un risque pour la santé du consommateur. Dans le but de mesurer l’exposition des consommateurs, deux méthodes analytiques fiables ont été développées pour l'analyse de ces substances dans les produits laitiers impliquant la chromatographie liquide couplée à la spectrométrie de masse en tandem. Le lait cru de vache, la poudre de lait entier, les fromages à pâte pressée cuite et les fromages fondus ont été sélectionnés pour représenter la diversité des produits laitiers. L'évaluation des performances de chacune des méthodes a été réalisée par une approche globale basée sur le profil d'exactitude. Pour la plupart des composés et des matrices étudiés, les méthodes d'analyse ont été validées sur l’intervalle de dosage de 5 à 150 μg / kg. Pour répondre au questionnement de l'impact des procédés de transformation du lait sur les résidus de biocides désinfectants, l'évolution des teneurs en composés et leur devenir dans les différentes matrices issues du lait ont été étudiées. Pour cela, deux études de faisabilité mettant en oeuvre des comparaisons d'empreintes chimiques globales, acquises par spectrométrie de masse à haute résolution, de fromages fondus et de fromages à pâte dure (contaminés vs témoins) ont été réalisées. Ces études ont permis de détecter 4 ions discriminants liés à la présence de biocides dans le fromage fondu. Leur identification reste à réaliser. Tout ce travail a été réalisé à des fins de sécurité alimentaire. La première partie est à l'élaboration de méthodes analytiques ciblées pour les résidus de biocides dans le lait et les produits laitiers permettant ainsi de mesurer les résidus de biocides sur les denrées alimentaires. Ces mesures sont nécessaires pour effectuer une analyse du risque liée à ces résidus. La deuxième partie est en relation avec la question du comportement des résidus de biocides lors de la transformation du lait présentant la stratégie, les résultats que nous pourrions obtenir et la perspective de travaux futurs. / The thesis work focuses on the presence of disinfectants biocides in dairy products and on the impact of milk processing on the possible appearance of process-induced food contaminants related to the exposition of milk with biocides. Disinfectants biocides are chemicals daily used in the dairy industry in cleaning-disinfection (CD) processes of food contact surfaces. Quaternary ammoniums as benzalkonium chloride (BACs) and dialkyldimethylammonium chloride (DDACs), and the Aminopropyldodecylpropane diamine are the most widely used disinfectants in dairy industry. These biocides can lead residues on the surfaces of food contact materials therefore present a health risk for the consumer. With aim of measuring consumers exposure, two reliable analytical methods have been developed for the analysis of these substances in dairy products involving liquid chromatography hyphenated with tandem mass spectrometry. Raw cow's milk, whole milk powder, hard pressed and processed cheeses were selected as representing the diversity of dairy products. The evaluation of the performances of each of the methods was carried out by the global approach based on the accuracy profile. For most of compounds and matrices studied, analytical methods were validated within the range of 5 to 150 μg/kg. To answer to the questioning of the impact of milk processes on biocides disinfectants residues, the evolution of compounds contents and their fate in the various matrices resulting from the milk were studied. For this, two proof-of-concept studies implementing global chemical fingerprint comparisons, acquired by High Resolution Mass Spectrometry, of processed cheese and hard pressed cheese (contaminated vs control) samples were carried out. These studies allowed to detect 4 discriminant ions linked to the presence of biocides in processed cheese. Their identification remains to be done. Whole this work is related for food safety purposes. The first part was linked to elaborate targeted analytical methods for biocides residues in milk and milk products thus allowing the measurement of biocides residues on food. These measurements are necessary for the risk analysis linked to these residues. The second part is in relation with the question of the behavior of biocides residues within milk processing presenting the strategy, the results we could obtain and the perspective for future works.

Desenvolvimento e avaliação comparativa de column switching e LC-MS em escalas convencional e miniaturizada para determinação de contaminantes emergentes em água / Development and comparative evaluation of conventional and miniaturized column switching and LC-MS for the determination of emerging contaminants in water

O interesse por técnicas analíticas que ofereçam a sensibilidade adequada, que sejam econômicas, ambientalmente corretas e capazes de processar rapidamente um grande número de amostras de diferentes complexidades é cada vez maior. Nesse aspecto, a utilização da LC no modo multidimensional é alternativa promissora, especialmente quando estratégias de extração e pré-concentração em fase sólida acopladas de forma on-line com a separação são exploradas, tanto em escala convencional quanto em escala miniaturizada. A ocorrência ambiental de contaminantes emergentes (EDCs) e seus impactos nas formas de vida aquática e terrestre têm sido motivo de preocupação. A quantificação desses analitos no ambiente exige métodos com limites de detecção (LDs) muito baixos, fazendo das estratégias column switching, com a inerente pré-concentração on-line, pertinentes para satisfazer as exigências na determinação desses compostos. Nesse cenário, o desenvolvimento e avaliação de estratégias column switching para determinação de contaminantes emergentes em amostras de interesse ambiental como aplicação modelo no estudo comparativo entre as escalas convencional e miniaturizada se faz impactante. Várias estratégias abrangendo column switching capilar e convencional foram investigadas. Colunas extratoras preenchidas com fases comerciais foram avaliadas, e tiveram seu desempenho testado no acoplamento com diversas colunas analíticas. Métodos em column switching capilar e convencional foram desenvolvidos e validados, tendo sido avaliadas as principais figuras de mérito. Os LQs atingidos em CS-UHPLC-ESI-MS/MS variaram de 0,001 ng mL-1 (BP e BzP) a 0,1 ng mL-1 (E3, E2 e EE2); para o método CS-cLC-ESI-MS/MS, os LQs foram maiores, variando entre 0,008 ng mL-1 (BP e BzP) a 0,8 ng mL-1 (E3, E2 e EE2). A linearidade dos métodos foi atestada avaliando-se os gráficos de resíduos, coeficiente de determinação e aplicando-se ponderações quando pertinente. A exatidão e a precisão foram investigadas, apresentando valores dentro dos critérios de aceitação. Por fim, os métodos foram aplicados em amostras ambientais aquosas coletadas em São Carlos - SP, mostrando que os métodos são adequados ao fim a que se destinam. / The interest in analytical techniques that offer adequate sensitivity, that are economical, environmentally friendly and capable of rapidly processing many samples of different complexities is increasing. In this regard, the use of LC in the multidimensional mode is a promising alternative, especially when solid phase extraction and preconcentration strategies coupled online with the separation are explored, both on a conventional scale and on a miniaturized scale. The environmental occurrence of emerging pollutants (EDCs) and their impacts on aquatic and terrestrial life forms have been of concern. Quantification of these analytes in the environment requires methods with very low detection limits (LDs), making column switching strategies with the inherent preconcentration online relevant to satisfy the requirements in determining these compounds. In this scenario, the development and evaluation of column switching strategies for the determination of emerging contaminants in samples of environmental interest as an application model in the comparative study between conventional and miniaturized scales is striking. Several strategies covering capillary and conventional column switching were investigated. Extraction columns filled with commercial phases were evaluated, and their performance was tested in the coupling with several analytical columns. Methods in capillary and conventional column switching modes were developed and validated, and the main figures of merit were evaluated. The LQs reached in CS-UHPLC-ESI-MS / MS ranged from 0.001 ng mL-1 (BP and BzP) to 0.1 ng mL-1 (E3, E2 and EE2); for the CS-cLC-ESI-MS / MS method, the LQs were higher, ranging from 0.008 ng mL-1 (BP and BzP) to 0.8 ng mL-1 (E3, E2 and EE2). The linearity of the methods was attested by evaluating the residual plots, coefficient of determination and applying weighted calibration when needed. Accuracy and precision were investigated, presenting values within the acceptance criteria. Finally, the methods were applied in samples collected in São Carlos - SP, showing that the methods are suitable for the purpose.

Catalytic activity of sewage sludge-derived char composite catalysts towards the oxidation of organic contaminants in water / Performances catalytiques de catalyseurs composites dérivés de charbons préparés à partir de boues de station d'épuration dans l'oxydation de polluants organiques dans l'eau

La gestion des boues de station d'épuration est un problème majeur. Dans ce travail, des charbons préparés à partir de boues de station d'épuration (SC) ont été utilisés comme support de catalyseurs. Les performances de ces catalyseurs ont été évaluées dans trois réactions d'oxydation pour le traitement de l'eau : le procédé Fenton, l'oxydation en voie humide catalytique et l'ozonation catalytique. Le catalyseur à base d'oxyde de fer supporté sur ce charbon (FeSC) est très actif dans le procédé de type Fenton pour la décoloration et la minéralisation de l'acide orange II (AOII). Les impuretés inorganiques présentes dans le charbon (cendres), telles que SiO2 et AI2O3, peuvent jouer le rôle de co-catalyseur. Le catalyseur FeSC est également très performant dans l'oxydation en voie humide catalytique du 2-chlorophénol à 120°C sous 0.9 MPa de pression partielle d'oxygène. Cependant, une lixiviation du fer est observée en cours de réaction du fait de la production de HC1 et de petits acides carboxyliques. La lixiviation du fer peut toutefois être évitée lorsque le pH du mélange réactionnel est maintenu en dessus de 4.5, sans que les performances catalytiques n'en soient affectées. Enfin, un catalyseur Mn-g-C3N4 supporté par un charbon obtenu à partir de boue de station d'épuration obtenu par modification de nitrure de carbone par des nanoparticules de manganèse a été synthétisé. L'activité catalytique de ce catalyseur composite est 1.6 fois supérieure à celle du catalyseur Mn-g-C3N4 non supporté. Ce catalyseur composite présente par ailleurs une bonne résistance à l'oxydation et une bonne stabilité, sans qu'aucune lixiviation du manganèse ne soit observée / The disposal of sewage sludge has become an issue of particular concern. In this thesis, sewage sludge derived carbon (SC) was employed as a catalyst support. The catalytic behavior of the prepared SC-based composite catalysts was investigated in three kinds of typical oxidation reactions, including heterogeneous Fenton-like oxidation, catalytic wet air oxidation (CWAO) and catalytic ozonation. Sewage sludge-derived carbon supported iron oxide catalyst (FeSC) showed high Fenton-like performances in the discoloration and mineralization of acid orange II (AOII). Inorganic components in the SC, such as SiO2 and AI2O3 may present have a co-catalytic effect upon Fenton-like reaction. FeSC catalyst also performed quite well in the CWAO of 2-CP at 120°C under 0.9 MPa oxygen partial pressure. However, iron leaching was observed due to the generation of HC1 and some small chain organic acids. Iron leaching could be efficiently prevented when the pH of the solution was maintained at values higher than 4.5, while the catalytic activity was only slightly reduced. Finally, Mn2O3 nanoparticles modified g-C3N4 (Mn-g-C3N4) was synthetized as a novel ozonation catalyst. To enlarge the adsorption capacity of the catalyst and improve its performances in the ozonation of sulfamethoxazole, the Mn-g-C3N4 catalyst was further supported over the sewage sludge-based activated carbon (SBAC). The catalytic activity of the composite catalyst was ca. 1.6 times higher compared to the unsupported Mn-g-C3N4 catalyst. The composite catalyst also exhibited very good resistance towards oxidation, limited Mn leaching and high stability

Estudo dos efeitos de contaminadores sobre o desempenho das células a combustível de membrana de eletrólito polimérico / Diagnosing the effects contaminants have over polymer electrolyte membrane fuel cells

Os componentes do conjunto membrana/eletrodos (MEA) das células a combustível de membrana de eletrólito polimérico/Polymer Electrolyte Membrane Fuel Cells (PEMFC) são sensíveis a impurezas, as quais podem vir do ar, do gás combustível e/ou da degradação dos componentes do módulo. Amônia, sulfeto de hidrogênio e monóxido de carbono são juntos os três principais subprodutos cotaminadores nos processos de geração de hidrogênio por reforma de combustíveis. Estes contaminadores afetam negativamente o desempenho das PEMFCs, assim é importante o entendimento destes efeitos para mitigá-los e introduzir a tecnologia das PEMFCs no mercado consumidor. Desta forma experimentos foram realizados visando diagnosticar os efeitos da amônia e do sulfeto de hidrogênio sobre os componentes do MEA das PEMFCs. Para a contaminação por sulfeto de hidrogênio foi provado, utilizando-se da técnica de cromatografia gasosa e de stripping, que a contaminação ocorre através da interação química e eletroquímica do contaminador com a superfície do catalisador de platina, e que estas interações ocorrem via um processo dissociativo e um processo oxidativos respectivamente. Estes processos de interação geram enxofre adsorvido sobre a superfície da platina, a qual é bloqueada para posterior oxidação de hidrogênio, gerando sobrepotenciais que reduzem a diferença de potencial da célula. Utilizando-se da técnica de cromatografia gasosa e agora de voltametria cíclica foi mostrado na PEMFC, que durante o processo de remoção do enxofre adsorvido a platina dióxido de enxofre é gerado. Ainda na PEMFC, foi mostrado utilizando-se da técnica de \"air bleed\" que maiores tolerâncias ao sulfeto de hidrogênio podem ser alcançadas, apesar de ser insignificante. Para o caso da contaminação da PEMFC por amônia, indiretamente foi mostrado, utilizando-se técnicas eletroquímicas solução de ácido perclórico, que amônia pode afetar a reação de redução de oxigênio pela sua adsorção sobre a superfície do catalisador, ou pelo bloqueio da mesma para posterior adsorção/redução de oxigênio. Em estudos de absorção de água e condutividade de membranas de NafionTM, sob diferentes frações catiônicas (prótons/amônio), em contato com água na fase vapor sob diferentes atividades, foi mostrado que quanto maior a concentração de íons contaminadores no eletrólito menor a quantidade de água absorvida e menor a condutividade da membrana. Também foi mostrado que se tais membranas fossem usadas como eletrólito em PEMFCs, o desempenho da célula seria afetado drasticamente por perdas ôhmicas. Também foi mostrado que sob contaminação por amônia, PEMFCs sofrem aumentos em resistências ôhmicas devido a reduções na condutividade do eletrólito, contudo foi provado que esta representa menos de dez por cento do total de perdas observadas no desempenho da célula. Desde estudo foi concluído que amônia afeta o desempenho das PEMFCs principalmente pela redução na atividade dos prótons na camada catalítica catódica, que causa reduções no potencial misto de equilíbrio da reação de redução de oxigênio, e portanto na diferença de potencial da célula. Finalmente foi provado indiretamente que amônia deixa a célula através do equilíbrio de amônio com água, o qual deslocado gera amônia, a qual deixa a célula junto com o fluxo de gás cotódico. / The Membrane Electrode Assembly components of a PEMFC are sensitive to impurities, which can came with the air or hydrogen stream, or from the degradation of the stack components. Ammonia, hydrogen sulfide and carbon monoxide are together the main sub-products of fuel reforming processes for generating hydrogen. These contaminants negatively affect the PEMFC performance, so it is important to understand what those effects are in order to mitigate them and introduce PEMFC technology in the mass market. Therefore, experiments were carried out to diagnose the effects hydrogen sulfide and ammonia have on the MEA components of PEMFCs. For contamination by hydrogen sulfide it was proved utilizing EMS and stripping techniques that the poisoning process happens by chemical and electrochemical interactions of the contaminant with the Platinum catalyst surface, and that these interactions happen by a dissociative and oxidative process, respectively. Those processes generate sulfur adsorbed on the Platinum surface, which blocks it for further hydrogen oxidation, generating overpotentials, which reduce the cell potential. Utilizing the EMS and now the cyclic voltammetry technique it was shown that during the process of removing sulfur from the Platinum surface one generates sulfur dioxide. Using the Air Bleed technique it was shown that higher tolerances of the PEMFC against hydrogen sulfide can be reached, despite being insignificant. For contamination of the cell by ammonia it was indirectly proved utilizing electrochemical techniques in perchloric acid solutions that ammonia can affect the oxygen reduction reaction by adsorbing on the catalyst surface, or by blocking the surface for further oxygen adsorption/reduction. Studying water uptake and ionic conductivity of Nafion membranes under many different cation fractions (proton/ammonium) in contact with water vapor at different temperatures and water activities, it was proved that the more ammonium one has in the membrane the less will be the water uptake and ionic conductivity of it. It was also shown that if those membranes were used as electrolyte in PEMFC the cell performance would be severely affected by ohmic losses. It was also shown that under ammonia exposure PEMFCs suffer by ohmic resistance increases due to the lowering in the ionic conductivity of the electrolyte, however it was proved that it represent less than ten percent of the observed losses in the cell performance. From this study it was concluded that ammonia mainly affect the PEMFC performance by lowering the cathode catalyst layer proton activity, which lowers the oxygen reduction reaction equilibrium potential, and then the cell potential. Finally it was indirectly proved that ammonia leaves the cell by the equilibrium of ammonium and water, which dislocated generates ammonia that leaves the cell together with the cathode gas stream.

O objetivo da presente pesquisa foi desenvolver uma dieta artificial que possa suprir as necessidades nutricionais de Euschistus heros (Fabricius, 1798) (Hemiptera: Pentatomidae) para produzir insetos em quantidade, comparáveis aos da natureza e que produzam descendentes, através das gerações, possibilitando a criação dos parasitoides de ovos, Telenomus podisi Ashmead, 1893 e Trissolcus basalis (Wollaston, 1858) (Hymenoptera: Platygastridae), para a liberação em campo, em Programas de Controle Biológico Aplicado. Verificou-se o efeito das dietas artificiais selecionadas para a criação do percevejo, sobre a sua capacidade alimentar, utilizando-se o teste da fucsina ácida e a capacidade de parasitismo de ambos os parasitoides, como forma de avaliar a qualidade da dieta. Assim, realizou-se o estudo da biologia de E. heros em condições controladas de temperatura (25±2°C), umidade relativa (60±10%) e fotofase de 14 horas. Treze dietas foram testadas numa sequência de experimentos: no primeiro, compararam-se a dieta natural (vagem de feijão e amendoim) e a dieta artificial de Panizzi et al. (2000) modificada por Fortes et al. (2006); na segunda fase, incorporaram-se os componentes da dieta natural numa dieta artificial com vagem de feijão liofilizada (DAL), comparando-se proporções destes componentes e diferentes anticontaminantes e doses. Foram selecionadas, para a criação de E. heros, duas dietas artificiais secas compostas de vagem de feijão (Phaseolus vulgaris L., 1753) liofilizada e moída (35%), amendoim (Arachis hypogaea L., 1753) triturado em liquidificador (35%), sacarose (5%) e água (25%) mais anticontaminantes, pois foram mais eficientes do que as dietas natural e liofilizada sem anticontaminantes, ao longo das gerações. As dietas artificiais selecionadas têm a mesma composição, porém com anticontaminantes diferentes; a primeira delas contém nipagin (10000 ppm) + ácido sórbico (800 ppm) e a outra, nipagin (10000 ppm) + ácido sórbico (800 ppm) + tetraciclina (0,0076 ppm). Os insetos da quarta geração, criados nestas dietas artificiais com material liofilizado, tiveram características biológicas semelhantes àqueles obtidos na dieta natural, incluindo a razão sexual, a duração do desenvolvimento (ovoadulto), as deformações de asas e pernas, a longevidade, o período de préoviposição, a porcentagem de fêmeas que ovipositaram e o número de ovos por postura. A viabilidade para o período ovoadulto foi quase duas vezes superior à viabilidade obtida na dieta natural, sendo de 68% em relação aos 38% obtidos na dieta natural; embora o peso de adultos tenha sido 17% inferior aos criados em dieta natural, a fecundidade foi superior ao valor obtido na dieta natural (282 ovos/fêmea, em média), atingindo a média de 430 ovos/fêmea. A taxa líquida de reprodução (Ro) aumentou com o passar das gerações para as dietas com material liofilizado com anticontaminantes, ocorrendo o inverso com a dieta de material liofilizado sem anticontaminantes, mostrando a respectiva adaptação e degeneração ao longo das gerações. As dietas com material liofilizado mostraram-se adequadas, pois os percevejos causaram danos aos grãos de soja, introduzindo seus estiletes de forma análoga àqueles criados em dieta natural, e os ovos de E. heros, criados nestas dietas, foram igualmente parasitados por T. basalis e T. podisi, em relação àqueles colocados pela espécie criada em dieta natural. As 2 dietas liofilizadas selecionadas permitiram a produção de E. heros ao longo de, pelo menos 10 gerações, sem degeneração e com potencial de produção dos parasitoides de ovos T. podisi e T. basalis, para programas de Controle Biológico Aplicado na cultura da soja. / The objective of the present research was to develop an artificial diet to meet the nutritional needs for the mass production of individuals of Euschistus heros (Fabricius, 1798) (Hemiptera: Pentatomidae), comparable to those produced in Nature and which can produce successive generations thus making possible the rearing of the egg parasitoids, Telenomus podisi Ashmead, 1893 and Trissolcus basalis (Wollaston, 1858) (Hymenoptera: Platygastridae), for field releasing in Applied Biological Control Programs. The effect of selected artificial diets on stink bug feeding capacity, using the acid fuchsin test, and the parasitism capacity of both parasitoids, were verified as a way of evaluating diet quality. The biology of E. heros was studied under controlled conditions of temperature (25±2°C), relative humidity (60±20%) and a 14 hour photophase. Thirteen diets were tested in a series of experiments: in the first experiment, a natural diet (dry bean and peanut pods) and Panizzi et al.s (2000) artificial diet, modified by Fortes et al. (2006), were compared; in the second phase, the components of the natural diet were incorporated into an artificial diet with lyophilized drybean pods (DAL), comparing different ratios of these components and different anti-contaminants and dosages. Two dry artificial diets, composed of lyophilized and ground up dry bean pods (Phaseolus vulgaris L., 1753) (35%), peanuts (Arachis hypogaea L., 1753) crushed in a blender (35%), sucrose (5%) and water (25%), plus anticontaminants, were chosen for rearing E. heros since over the generations they were found to be more efficient than the natural diet and the lyophilized diet without anti-contaminants. The selected artificial diets have the same composition but with different anti-contaminants: one with nipagin (10000 ppm) + sorbic acid (800 ppm) and the other with nipagin (10000 ppm) + sorbic acid (800 ppm) + tetracyclin (0.0076 ppm). The 4th generation insects reared on these artificial diets with lyophilized material had similar biological characteristics as those obtained from the natural diet, including the sex ratio, development time (eggadult), wing and leg deformations, longevity, pre-oviposition period and the percentage of females which oviposited and number of eggs per oviposition. The viability for the egg-adult period (68%) was almost twice as high for the artificial diet as that obtained for the natural diet (38%). However, adult weight was 17% less than for individuals reared on natural diet, although fecundity was higher for insects reared on the artificial diet, reaching a mean of 430 eggs/female compared to 282 eggs/female on the natural diet. The net reproduction rate (Ro) increased with the number of generations for diets containing lyophilized material plus anti-contaminants, with the opposite occurring for the diet of lyophilized material without anti-contaminants, showing the respective adaptation and degeneration over the generations. The diets with lyophilized material were considered suitable because the stink bugs damaged the soybean grains, introducing their stylets in the same way as those insects reared on the natural diet. Similarly, E. heros eggs reared on these diets were equally parasitized by T. basalis and T. podisi, in the same way as those laid by the species reared on the natural diet. The two lyophilized diets selected allowed E. heros production during at least 10 generations, without any degeneration and with a potential to produce the egg parasitoids T. podisi and T. basalis, for Applied Biological Control Programs in soybeans.

A qualidade da água de abastecimento é de vital importância à saúde da população dos núcleos urbanos do mundo todo. Por essa razão, muitas pesquisas enfocam esse tema. Além disso, os ataques terroristas recentes ocorridos nos Estados Unidos e Europa, vêm fomentando a antiga preocupação relativa a possíveis injeções de contaminantes em redes de distribuição de água para abastecimento, evidenciando a importância da efetiva vigilância de tais sistemas. O presente trabalho investiga a rede de monitoramento ótima para detecção de injeções intencionais de poluentes e/ou contaminantes em concentrações e/ou quantidades suficientes para que se propaguem nas direções do fluxo da água no interior das redes, do ponto de vista de objetivos múltiplos. A metodologia aqui apresentada representa uma extensão de propostas anteriores e é demonstrada ilustrativamente, através de redes já utilizadas na literatura. Com base no conceito da rede auxiliar proposto por Kessler et al. (1998), propõe-se o emprego de algoritmos genéticos multiobjetivo para considerar os níveis de serviço em termos do volume consumido, do tempo e da extensão da rede atingida antes da detecção. São criadas matrizes de poluição para os níveis de serviço considerados e, então, o algoritmo genético multiobjetivo SPEA é aplicado para identificar as soluções não-dominadas, em conformidade com o conceito de otimalidade de Pareto. Os resultados demonstram o potencial do método proposto em identificar tais soluções / The water supply quality is very important to the healthy of urban nucleus populations around the world. This is the reason why many researches focus on such theme. Besides this, recent terrorist attacks occurred in USA and Europe, have incited the old apprehension related to possible deliberate intrusions of contaminants into the water supply networks, making evident the importance of the effective vigilance of such systems. This work investigates the optimal monitoring network for detection of deliberate intrusions of pollutes and/or contaminants at concentrations and/or quantities enough for propagation inside the networks, on the point of view of multiple objectives. The method here proposed represents an extension to earlier proposals and is demonstrated with the support of networks from literature. Based on the concept of auxiliary network proposed by Kessler et al. (1998), a multiobjective genetic algorithm is used in order to consider the levels of service in terms of the consumed volume, time period and length of the network reached before detection. Pollution matrixes are built for the levels of service considered and the multiobjective genetic algorithm SPEA applied in the identification of the non-dominated solutions, according to the Paretto optimality concept. The results demonstrate the potential of the method in the identification of such solutions

Avaliação de resíduos de etilenotiouréia (ETU) em frutas comercializadas na cidade de São Paulo / Evaluation of ethylene thiourea (ETU) in fruits traded in the city of São Paulo.

Etilenotiouréia (ETU) é uma substância tóxica, formada pela degradação e/ou biotransformação dos fungicidas etilenobisditiocarbamatos (EBDC). Seus resíduos podem ser encontrados em plantas e no ambiente, após o uso de EBDC na agricultura, ou em animais e no ser humano, quando expostos a esses produtos. Comprovadamente, a ETU tem a capacidade de induzir tumor na tireóide de roedores e no fígado de camundongo, apresentando evidência suficiente para carcinogenicidade em animais e evidência inadequada para carcinogenicidade em seres humanos. Os objetivos deste estudo foram: validar método analítico para determinação de resíduos de ETU nas matrizes estudadas; verificar a presença de resíduos de ETU em amostras de frutas (mamão, maçã e morango), coletadas em diferentes pontos de comercialização da cidade de São Paulo; avaliar os resultados e a contribuição de risco à saúde da população consumidora. Foram analisadas 90 amostras, sendo 30 de cada fruta (maçã, mamão e morango), adquiridas em diferentes pontos de comercialização da cidade São Paulo, no período de dezembro de 2005 a dezembro de 2006, distribuídas nas diferentes regiões do referido município e durante as estações do ano. Os parâmetros de validação avaliados foram: seletividade, linearidade, limite de quantificação, limite de detecção, exatidão e precisão. Os estudos de recuperação foram realizados com fortificações em três níveis (1, 2 e 10 LQ) em amostras controle. A determinação de resíduos de ETU foi feita por cromatografia a líquido de alto desempenho, com detector de absorção no ultravioleta (HPLC-UV) e cromatografia a líquido acoplada à espectrometria de massas em tandem (LC/MS/MS). Como todos os resultados obtidos por HPLC-UV apresentaram níveis de ETU abaixo do LQ limite de quantificação (10,0?g/kg), as quantificações foram realizadas no LC/MS/MS, com limite de quantificação e de detecção respectivamente de 1,0?g/kg e 0,5?g/kg. O método por LC/MS/MS mostrouse adequado para análise de ETU nos níveis de até 2,0?g/kg para maçã e morango e de até 10,0 ?g/kg para mamão, com recuperações médias de 75 a 110% e coeficientes de variação de 6 a 17%. Foram encontrados resíduos de ETU em 10 (33%) das amostras de maçã, em níveis que variaram de 1,0 a 3,7 ?g/kg (ppb); em 20 (67%) das amostras de mamão, de 1,0 a 5, 3?g/kg; em 2 (7%) das amostras de morango, de 1,0 a 1,4 ?g/kg. Estes valores estão abaixo do limite de 50?g/kg, estabelecido na União Européia. A estimativa da ingestão de resíduos de ETU pelo consumo das frutas estudadas, considerando o maior nível encontrado nesta pesquisa e dados fornecidos pelo IBGE, representam respectivamente 0,05 e 0,20 % da IDA (Ingestão Diária Aceitável, estabelecida pelo Codex Alimentarius), para a população em geral e crianças. A estimativa da ingestão de resíduos de EBDC pelo consumo dos alimentos (maçã, tomate, mamão, alface, morango, banana, laranja, cenoura), monitorados pelo PARA no período de 2001 a 2004, considerando os maiores níveis encontrados e 100% dos resíduos de ditiocarbamatos (CS2) como sendo originários do uso de EBDC, foi de 7,2% para a população em geral e de 28,9 % para crianças. Dados de ETU e EBDC em outros alimentos, incluindo os industrializados, em água de consumo e em amostras ambientais, além de dados mais refinados de consumo alimentar para subgrupos mais sensíveis (como crianças e mulheres grávidas), são necessários para se avaliar o risco de maneira mais global e condizente com a realidade, em prol da Saúde Pública. / Ethylene thiourea (ETU) is a toxic substance generated by the degradation and/or biotransformation of ethylenebisdithiocarbamates (EBDC) fungicides. Their residues may be found in plants and in the environment after EBDC's use in agriculture or in animals and humans when exposed to such products. ETU is confirmedly able to induce tumors in rodents' thyroids and in mouse's liver, showing enough evidence of carcinogenicity in animals and inadequate evidence of carcinogenicity in humans. The aims of this study were validating an analytical method for determining ETU residues in the studied sources, verifying the presence of ETU residues in fruit samples (papaya, apple and strawberry) collected from different commercial centers in the city of São Paulo; evaluating the results and their contribution as a risk to the consumers' health. Ninety fruit samples (30 samples of each fruit - papaya, apple and strawberry) were analyzed. The samples were purchased at different commercial centers in different regions of the city of São Paulo and over all seasons during December 2005 through December 2006. The validation parameters used were selectivity, linearity, accuracy and precision. Recovery studies were carried out with fortifications in 3 levels (1, 2 and 10 times the Limit of Quantification, LQ) in control samples. The determination of ETU residues was carried out by high performance liquid chromatography, ultra-violet absorption detector (HPLC/UV) and liquid chromatography coupled on tandem mass spectrophotometry (LC/MS/MS). Since all results obtained through HPLC/UV showed levels of ETU below the LQ (10.0 ?g/kg), quantification were carried out through LC/MS/MS with quantification and detection limits of 1.0?g/kg and 0.5 ?g/kg, respectively. The LC/MS/MS method proved adequate for analyzing quantities of ETU as low as 2.0 ?g/kg in apple and strawberry and 10.0 ?g/kg in papaya, with average recovery of 75 to 110 % and variation coefficients of 6 to 17 %. ETU residues were found in 10 (33%) apple samples with levels ranging from 1.0 to 3.7 ?g/kg (ppb); in 20 (67%) of papaya samples with levels ranging from 1.0 to 5.3 ?g/kg (ppb); and in 2 (7%) strawberry samples with levels ranging from 1.0 to 1.4 ?g/kg (ppb). These values are below the limit established in European Community (50 ?g/kg). The assessment of ETU intake, considering the consumption informed by IBGE (Instituto Brasileiro de Geografia e Estatística, the Brazilian Institute for Geography and Statistics) and the highest level observed in total fruit samples in this research, represent 0.05 % and 0.20 % ADI (Acceptable Daily Intake, established by the Codex Alimentarius) for geral population and children, respectively. The assessment of EBDC intake, considering the consumption informed by IBGE (Instituto Brasileiro de Geografia e Estatística, the Brazilian Institute for Geography and Statistics) and the highest level observed in food samples (apple, tomato, papaya, lettuce, strawberry, banana, orange, carrot), monitoring from 2001 through 2004 year in the PARA (Programa de Análise de Resíduos de Agrotóxicos em Alimentos, Program Monitoring of Pesticide Residues in Food, Brazil), and that 100% of dithiocarbamate (CS2) residues from EBDC's use, represent 7.2% and 28.9% ADI for geral population and children, respectively. ETU and EBDC data in other foods (including industrialized foods), water and environment samples, together with more refined food consumption data considering more sensitive population subgroups (as children and pregnants) are necessary to evaluate the risk in a more global and realistic way in favor of Public Health.

Determinação de nutrientes e contaminantes Inorgânicos em café solúvel por espectrometria de emissão atômica com plasma de argônio induzido (ICP-AES) / Determination of nutrients and inorganic contaminants in soluble coffee by atomic emission spectrometry with inductively-coupled plasma (ICP-AES)

A produção industrial do café solúvel brasileiro destina-se, na sua maior parte, ao mercado externo, constituindo-se em um importante ítem de exportação do país. O objetivo do presente trabalho foi o desenvolvimento e aprimoramento de métodos de análise química adequados à investigação de componentes inorgânicos em café solúvel. Inicialmente foi definido o método de mineralização das amostras através da avaliação de 6 procedimentos diferentes, classificados em 3 classes: (I) microondas focalizado; (II) via úmida convencional e (III) sistema sob pressão em frascos de decomposição com tampas (bombas de teflon). As determinações dos nutrientes e contaminantes inorgânicos: Na, K, Mg, Al, P, S, Ca, Mn, Fe, Ni, Cu, Zn, Cd, Sb, Pb, Cr, Sn, As, Se e Hg, após a utilização do procedimento de mineralização que emprega o sistema de microondas focalizado com os reagentes HNO3 e H2O2, foram realizadas por espectrometria de emissão atômica com plasma induzido, ICP-AES (da nomenclatura inglesa Inductively Coupled Plasma-Atomic Emission Spectrometry). Interferências de matriz e/ou espectrais foram detectadas nas determinações de Pb, Cr, Sn, As, Se e Hg. No caso do Sn, optou-se pela correção da radiação de fundo através do software do equipamento, pois outras linhas de emissão deste analito eram menos sensíveis e igualmente sujeitas a interferências espectrais. Para o Pb e Cr a escolha de comprimentos de ondas alternativos eliminou os problemas de interferências espectrais devido à matriz das amostras. A aplicação das técnicas de geração de hidretos para o As e Se e a geração de vapor frio para o Hg, possibilitaram as determinações destes analitos nas amostras de café solúvel, aumentando a sensibilidade das análises. Os resultados obtidos para os teores dos nutrientes e contaminantes inorgânicos em 21 amostras de café solúvel disponíveis no mercado brasileiro, foram aceitáveis para o consumo humano, tanto no aspecto nutricional como toxicológico, com exceção de uma amostra com teor de Cr igual a 0,52 ± 0,02 mgKg-1, acima do permitido pela legislação brasileira que fixa o valor máximo de 0,10 mgKg-1. A aplicação de técnicas estatísticas, análise de clusters hierárquicos e componentes principais aos resultados obtidos, possibilitaram investigar a variabilidade das composições minerais e classificar grupos de amostras similares. O emprego da técnica instrumental ICP-AES através do sistema de nebulização convencional e a utilização do sistema de geração de hidretos e vapor frio, mostraram-se adequados para as determinações dos nutrientes e contaminantes inorgânicos em café solúvel, uma vez que níveis de precisão e exatidão aceitáveis foram obtidos. / The production of Brazilian soluble coffee is almost entirely destined for exporting. Therefore, the objective of this work was the development and the establishment of appropriate methods of chemical analysis of inorganic components in soluble coffee. Firstly, the appropriate digestion procedure for soluble coffee samples was defined through the evaluation of six different digestion techniques that were classified into three groups: (I) focused microwave system, (II) conventional wet digestion and (III) pressure digestion system in teflon bombs. The determinations of nutrients and toxic elements: Na, K, Mg, Al, P, S, Ca, Mn, Fe, Ni, Cu, Zn, Cd, Sb, Pb, Cr, Sn, As, Se, and Hg after the utilization of the focused microwave system with HNO3 and H2O2 reagents, were made by Inductively Coupled Plasma Atomic Emission Spectrometry (ICP-AES) technique. Matrix and spectral interferences were detected in the determination of Pb, Cr, Sn, As, Se and Hg. Correction of the background enhancement by the equipment software was made in the determination of Sn, as other analytical lines were unsuitable for this application. For Pb and Cr, the choice of other analytical lines eliminated the problem of matrix and spectral interferences. The application of the hydride generation technique for As and Se and cold atomic vapour with ICP-AES for Hg, were suitable methods to determine these analytes in soluble coffee samples with increased sensitivity of the analysis. The results obtained of nutrients and toxic elements in twenty-one samples of soluble coffee collected from Brazilian market were acceptable to human consumption at nutritional and toxic level with exception of a sample with 0,52 ± 0,02 mgKg-1 of Cr, above the value specified in Brazilian Food Legislation, that fixes the maximum Cr contend in 0,10 mgKg-1. The aplication of statistical techniques analysis of principal components and the hierarchical cluster on the results of mineral compositions, enabled to investigate the variability in the composition and to classify groups of similar samples. The application of the ICP-AES technique using the conventional pneumatic nebulization system, the hydride generation technique and the cold vapour for Hg were suitable to determine the nutrients and toxic elements in soluble coffee, as they provided acceptable results indicating accuracy and precision.
