Aktivist, Javisst! : En kvalitativ fallstudie om Extinction Rebellion Svergies kommunikation på Facebook under Noveberupproret 2020

Social movements focusing on environmental issues experienced a renaissance in September 2018, and concurrently the environmentally focused social movement Extinction Rebellion was founded. Previous research reveals that social media has become an important tool for social movements for both mobilising and spreading knowledge and information. Therefore, the purpose of this study was to examine Extinction Rebellion Sweden's strategic communication and work processes during the campaign Novemberupproret (The November revolt), and to identify what characterized the posts on Extinction Rebellion Sweden's Facebook during the campaign. The theoretical frameworks used in this study was Grunig's two-way symmetrical and asymmetrical communication (Falkheimer & Heide 2014), Habermas theory regarding social actions (Falkheimer & Heide 2018) and ultimately semiotics by Barthes (1977). The findings are based on interviews with two members of Extinction Rebellion Sweden, along with a qualitative content analysis and a semiotic analysis. The findings of this study reveal that Extinction Rebellion Sweden mainly focuses, both in terms of strategies and work processes, on content regarding information and knowledge sharing, rather than mobilising content. The empirical material shows that the posts with two-way asymmetrical communication have generated more interaction in terms of likes, comments and sharing on Facebook, which opposes Grunig's theory regarding two-way communication. Further, it is revealed that Extinction Rebellions Sweden's most frequently used communication strategy during Novemberupproret were consciously hidden strategic actions. Lastly the findings show that the movement´s work process during Novemberupproret was built on democratic and collective principles both between Extinction Rebellion Sweden's internal project groups and within the national media group.

Exploring the dawn of the universe with the Sino-French SVOM mission / Explorer l'aube de l'univers avec la mission franco-chinoise SVOM

Cette thèse s’inscrit dans le cadre de la préparation de la mission spatiale franco­-chinoise SVOM (Space Variable Object Monitor) opérationnelle à l’horizon 2021 et qui aura pour but de détecter et caractériser les sursauts gamma. La première partie de la thèse consiste au développement d’un logiciel de simulation de COLIBRI (Catching OpticaL and Infrared BRight transIents), un télescope de suivi au sol de SVOM. Il comporte un simulateur de sursauts gamma, un exposure time calculator, un simulateur d'image, un algorithme estimant le redshift photométrique. Il a permis d'une part d’estimer les performances scientifiques du télescope afin de s'assurer qu'elles satisfont le cahier des charges, et d'autre part d'estimer une précision sur l'estimation de la distance du sursaut de l'ordre de 10% et 15% sur le redshift pour les sursauts à 3.5 < z < 8 et z > 8. La deuxième partie de la thèse porte sur l’étude de la propriété des poussières au sein des galaxies dans lesquelles se produisent ces sursauts gamma, mais aussi à l’étude de leur activité de formation stellaire. J'ai comparé les courbes d'extinction mesurées sur la ligne de visée de 7 GRBs à la courbe d'atténuation de leurs galaxies à l'aide d'un code de SED fitting, CIGALE. En comparant à un code de transfert radiatif on obtient des informations sur la distribution étoiles / poussière au sein de ces galaxies. Sur un échantillon élargi, on montre que les courbes d'atténuation pentues (plates) corrèlent avec des galaxies avec une faible (grande) atténuation due à la poussière, les moins (plus) activent en formation stellaire et les moins (plus) massives / This thesis lies within the framework of the sino-french SVOM (Space Variable Object Monitor) mission to be launched in 2021 whose objective is the detection and characterisation of Gamma-Ray Bursts (GRBs). The first part consists in the development of an end-to-end software for COLIBRI (Catching OpticaL and Infrared BRight transIents), a SVOM ground follow-up telescope. It consists in a GRB simulator, an Exposure Time Calculator, an image simulator and a photometric redshift algorithm. It allowed to estimate the telescope scientific performances to ensure that the optical design was fulfilling the scientific requirements. The relative accuracy on the photometric redshift delivered by COLIBRI is estimated to be about 10% and 15% for GRBs at 3.5 < z < 8 et z > 8. The second part of the thesis deals mainly with the study of dust properties in GRB host galaxies, but also with the star formation activity in these galaxies. We compare extinction curves measured along the GRB line of sight for 7 GRBs to the attenuation curve of their host galaxies measured with CIGALE, a SED fitting code. By comparing these curves to the results of a radiative transfer code, we obtain information about the geometrical distribution of dust and stars in these galaxies. On a larger sample of 23 galaxies, we show that the steepest (flattest) attenuation curves are associated to galaxies with a large (small) amount of dust attenuation, less (more) active in star formation and less (more) massive galaxies

A microphysical model of scattering, absorption, and extinction in electromagnetic theory

Sorensen / This work presents a microphysical model of the classical interaction of electromagnetic waves with arbitrary single and multiple particles. The model is based on the volume integral equation solution to the macroscopic time-harmonic Maxwell equations. The integral is discretized over a particle's volume. The near and far-field scattered wave is then described by the secondary radiation from the discretized elements. The physical origin of the angular structure of the scattered wave is characterized by the superposition of these secondary waves. A graphical technique is developed to visualize how this superposition relates to the physical features of a particle, e.g., its size, shape, and refractive index. Numerical and analytical implementations of the model are presented for spherical and spheroidal particles and fractal-like spherical-particle aggregates. The connection between the reflection symmetry of a particle and the polarization state of its far-field scattered wave is illustrated. The model is used to explain the cause of the angular power-law patterns in a particle's scattered intensity. An analysis of the internal field distribution in fractal-like aggregates is performed and the results are compared to the Rayleigh-Debye-Gans theory. Extinction and the optical theorem are examined within the context of the model, resulting in a new understanding for the physical mechanism causing extinction and implications regarding its measurement. The culmination of this work is the unification of multiple scattering-concepts, often regarded as distinct, and the resulting insight afforded by the unified microphysical picture. This unified view is shown to reveal a new and simple explanation for the famous extinction paradox.

Extinction in the solar neighborhood : A comparative study of two methods used to measure reddening towards individual stars

Interstellar extinction and reddening are inescapable sources of uncertainty in the study of astronomical objects. Many creative ways to measure its effects have been developed, two of them being evaluated in this study. I apply two recently improved methods to estimate extinction toward individual stars, one based on interstellar absorption of sodium and the other based on opacity maps of the interstellar medium. The methods are applied to 14 stars in the local neighborhood, all within a distance of 250 pc from the Sun, and their results are compared. I find that both methods have severe limitations and large uncertainties, but can still be useful under the right conditions. I also provide suggestions on how the methods can be improved. / Interstellär extinktion och rödfärgning är oundvikliga osäkerhetskällor i studiet av astronomiska objekt. Många kreativa sätt att mäta dess effekter har utvecklats, av vilka två utvärderas i denna studie. Jag tillämpar två nyligen förbättrade metoder för att uppskatta utsläckning mot enskilda stjärnor, en baserad på interstellär absorption av natrium och den andra baserad på opacitetskartor över det interstellära mediet. Metoderna tillämpas på 14 stjärnor i solens närområde, alla inom ett avstånd av 250 pc, och deras resultat jämförs. Jag finner att båda metoderna har allvarliga begränsningar och stora osäkerheter, men ändå kan vara användbara under rätt förutsättningar. Jag ger också förslag på hur metoderna kan förbättras.

Molecular mechanisms of D-cycloserine in a fear extinction posttraumatic stress disorder (PTSD) animal model

Thesis (PhD)--Stellenbosch University, 2014. / ENGLISH ABSTRACT: Posttraumatic stress disorder (PTSD) is a severe, chronic and debilitating psychiatric disorder that can present after the experience of a life-threatening traumatic event. D-cycloserine (DCS), a partial N-methyl-D-aspartate (NMDA) receptor agonist, has been found to augment cognitive behavioural therapy by facilitating fear extinction; however, the precise mechanisms whereby DCS ameliorates fear triggered by a traumatic context remains to be fully elucidated. This study aimed to (i) identify the molecular mechanisms of intrahippocampally administered DCS in facilitating fear extinction in a rat model of PTSD by investigating gene expression profiles in the left dorsal hippocampus (LDH) of male Sprague Dawley rats and (ii) determine whether microRNA (miRNA) expression and DNA methylation mediated these gene expression changes. An adapted version of the PTSD animal model described by Siegmund and Wotjak (2007) was utilised. The total number of 120 rats were grouped into four experimental groups (of 30 rats per group) based on fear conditioning and the intrahippocampal administration of either DCS or saline: (1) fear conditioned + intrahippocampal saline administration (FS), (2) fear conditioned + intrahippocampal DCS administration (FD), (3) control + intrahippocampal saline administration (CS) and (4) control + intrahippocampal DCS administration (CD). Behavioural tests (the light/dark [L/D] avoidance test, forced swim test and open field test) were conducted to assess anxiety and PTSD-like behaviours. The L/D avoidance test was the most sensitive behavioural test of anxiety and was subsequently used to differentiate maladapted (animals that displayed anxiety-like behaviour) and well-adapted (animals that did not display anxiety-like behaviour) subgroups. In order to identify genes that were differentially expressed between FS maladapted (FSM) (n = 6) vs. FD well-adapted (FDW) (n = 6) groups, RNA sequencing was performed on the Illumina HiSeq 2000 which generated more than 60 million reads per sample. This was followed by subsequent bioinformatics analyses (using the software programs TopHat, Bowtie, Cuffdiff and Bio-Ontological Relationship Graph (BORG) database (that identifies genes that may be biologically relevant) to identify biologically relevant differentially expressed genes between the treatment groups. Epigenetic mechanisms mediating observed differences in gene expression were investigated by conducting DNA methylation and miRNAseq analyses in the FDW and FSM experimental groups. DNA methylation was investigated using real-time quantitative PCR (qPCR) amplification followed by high resolution melt analysis on the Rotor-GeneTM 6000. Differences in miRNA expression levels between the FDW and FSM groups were investigated by sequencing the miRNA fraction on the MiSeq platform. The bioinformatics pipeline used to analyse the RNAseq data identified 93 genes that were significantly downregulated in the FDW group compared to the FSM group. Forty-two of these genes were predicted to be biologically relevant (based on BORG analysis). Integrative network analyses revealed subsets of differentially expressed genes common across biological functions, pathways and disorders. The co-administration of DCS and behavioural fear extinction downregulated immune system genes and genes that transcribe proinflammatory and oxidative stress molecules. These molecules mediate neuroinflammation and subsequently cause neuronal damage. DCS also regulated genes involved in learning and memory processes. Additionally, a subset of the genes, which have been found to be associated with disorders that commonly co-occur with PTSD (such as cardiovascular disease, metabolic disease, Alzheimer‘s and Parkinson‘s disease), was downregulated by the co-administration of DCS and behavioural fear extinction. In order to determine whether real-time qPCR analysis would be sensitive enough to detect differential expression in those genes found to be differentially expressed in RNAseq analysis, the expression of nine genes was analysed using SYBR Green qPCR technology. In the LDH, six of the nine genes were found to be differentially expressed between FDW and FSM groups and one gene, matrix metallopeptidase 9 (MMP9), was observed to be differentially expressed between these two groups in the blood. Three of the nine genes for which differential expression levels were investigated using SYBR Green real-time qPCR, contained CpG islands and were used for CpG island DNA methylation analysis. Results indicated that CpG island DNA methylation did not mediate differential gene expression of TRH, NPY or MT2A. Bioinformatics analysis of miRNAseq data identified 23 miRNAs that were differentially expressed between the FDW and FSM groups. Several of these miRNAs have previously been found to be involved in brain development and behavioural measures of anxiety. Furthermore, functional luciferase analysis indicated that the upregulation of rno-mi31a-5p could have facilitated the downregulation of interleukin 1 receptor antagonist gene (IL1RN) as detected in RNAseq. RNAseq and miRNAseq analyses in this PTSD animal model identified differentially expressed genes and miRNAs that serve to broaden our understanding of the mechanism whereby DCS facilitates fear extinction. To this end, immune system genes and genes transcribing proinflammatory and oxidative stress molecules were among the genes that were found to be differentially expressed between the FDW and FSM groups. Based on the results obtained, it can be hypothesised that DCS attenuates neuroinflammation and subsequent neuronal damage, and also regulates genes involved in learning and memory processes. Concomitantly, these gene expression alterations mediate optimal neuronal functioning, plasticity, learning and memory (such as fear extinction memory) which contribute to the fear extinction process. Furthermore, biologically relevant differentially expressed genes that were associated with DCS facilitation of fear extinction and with other chronic medical conditions, such as cardiovascular disease and metabolic diseases, might help to explain the co-occurrence of these disorders with PTSD. In conclusion, Identifying the molecular underpinnings of DCS-mediated fear extinction brings us closer to understanding the process of fear extinction and could, in future work be used to explore novel therapeutic targets to effectively treat PTSD and related disorders. / AFRIKAANSE OPSOMMING: Posttraumatiese stressindroom is 'n ernstige, kroniese aftakelende psigiatriese toestand wat kan ontwikkel na 'n lewensgevaarlike traumatiese gebeurtenis. Nuwe generasie RNA volgordebepaling (RNAseq) van die LDH RNA en daaropvolgende bioinformatiese analise was uitgevoer om gene te identifiseer wat differensieel uitgedruk is tussen die twee behandelingsgroepe van belang in die betrokke studie, naamlik FS wanaangepaste (FSM) teenoor FD goedaangepaste (FDW) groepe. Epigenetiese analises was uitgevoer om te bepaal of differensieel uitgedrukte miRNAs of CpG-eiland DNA metilasie die differensiële geenuitdrukking bemiddel het. Bioinformatiese analises van die RNAseq data het 93 gene geïdentifiseer waarvan die geen uitdrukking beduidend onderdruk was in die FDW groep in vergelyking met die FSM groep; 42 van hierdie gene was voorspel om biologies relevant te wees. Geïntegreerde netwerk analise het onthul dat sekere van die differensieel uitgedrukte gene gemeenskaplik was tussen verskeie biologiese funksies, padweë en versteurings. Funksionele luciferase analises het verder aangedui dat die verhoogde uitdrukking van rno-mi31a-5p moontlik die verlaagde geen uitdrukking van IL1RN, soos waargeneem in die RNAseq data, kon bewerkstellig het. RNAseq en miRNAseq analises in hierdie posttraumatiese stressindroom dieremodel het differensieël uitgedrukte gene en miRNAs geïdentifiseer wat dien om die verstaanswyse te verbreed van hoe DCS die vrees uitwissings proses fasiliteer. Die meganismes waardeur DCS vrees uitwissings bewerkstellig het sluit die verlaging van immuun-sisteem geen-uitdrukking in, sowel as verlaagde uitdrukking van gene wat pro-inflammatoriese en oksidatiewe stress gene transkribeer. DCS het daardeur neuro-inflammasie en gevolglike neurale skade voorkom. DCS het daarmee saam ook gene gereguleer wat betrokke is by leer en geheue prosesse. Hierdie gesamentlike veranderings in geen uitdrukking het gelei tot die uiteindelike bewerkstelling van optimale neurale funksionering, plastisiteit, leer en geheue prosesse wat uiteindelik bygedra het tot vrees uitwissing. Biologies relevante differensieël uitgedrukte gene wat ook geassosieer was met ander kondisies, soos middel verwante versteurings en metaboliese versteurings, kan help om die komorbiditeit met posttraumatiese stressindroom te verklaar. Identifisering van die molekulêre grondslae van DCS bemiddelde vrees uitwissing verbreed ons begrip en verstaan van vrees uitwissing en kan moontlik, in toekomstige navorsing gebruik word om nuwe innoverende terapeutiese teikens te verken om sodoende posttraumatiese stressindroom meer effektief te kan behandel.

Improvements to the modelling of radiowave propagation at millimetre wavelengths : in-depth studies are reported on resonance phenomena in the scattering of spherical ice particles, extinction and backscattering properties of clouds and on the absorption and dispersion spectra of atmospheric gases

Various physical mechanisms that affect radiowave propagation at millimetre wavelengths are considered. Current modelling weaknesses are highlighted and new improved models or more appropriate modelling approaches are suggested. Interference and resonance phenomena in the scattering of spherical ice and water particles are reviewed. The long standing problem of the numerous resonances observed in the scattering diagrams of dielectric spheres is answered. The spatial structure and the physical characteristics of non-precipitable ice and water clouds are reviewed. Extinction and back scattering calculations for a wide variety of cloud models over the entire millimetre frequency spectrum are given. Multiple scattering and the effects of super-large drops in clouds are also dealt with. The potential of a spaceborne instrument in deducing information about the vertical structure of various cloud types is examined. Attenuation and reflectivity profiles resulting from various cloud types are calculated for a nadir pointing fixed beam millimetre wave radar operating at 94 GHz. The physics and application of the equation of radiative transfer to millimetre wave propagation in the earth's atmosphere are given and also is the solution of this equation for a typical millimetre wave remote sensing application. The theory of gaseous absorption at millimetre wavelengths is presented and an improved modelling approach is proposed for the calculation of the absorption and dispersion spectra of atmospheric gases. The effects of trace gases on communication systems operating at high altitudes are for the first time reported. Finally the use of the 60 GHz oxygen absorption band for top-side air traffic control/navigation and broadband transmission purposes is studied.

The effects of habitat loss and fragmentation caused by woody plant encroachment on native plant diversity and on an invasive grass

Habitat loss, habitat fragmentation and species invasions have been recognized as three of the leading threats to biodiversity. I examined the effects of habitat loss and fragmentation on native and invasive plants in central Texas. During the last century, the density and abundance of woody plants has been increasing in the savannas of eastern Edwards Plateau. This process, known as woody plant encroachment, not only reduces the amount of open herbaceous habitat but also fragments that habitat creating smaller and more isolated patches. In three studies, I investigated the consequences of this habitat loss and fragmentation for plants which do not occur under the cover of woody plants including native grasses and forbs and the invasive Eurasian bunchgrass, Bothriochloa ischaemum (King Ranch Bluestem). In the first study, I show that woody plant encroachment reduces native herbaceous species richness (the number of species in a given area). Using a collection of historical aerial photographs, I demonstrate that current native herbaceous species richness was most strongly related to recent habitat amount, but to the degree of habitat fragmentation at least 50 years ago. In a second study, I show that the presence of B. ischaemum was negatively related to the degree of fragmentation in the surrounding landscape. Finally, I found that B. ischaemum had higher rates of germination and growth in experimental plots where the species commonly lost with woody plant encroachment were removed than in unmanipulated control plots. Together, this work suggests that woody plant encroachment is directly slowing the spread of an invasive species while indirectly facilitating its establishment. / text

La cognition se réfère aux mécanismes par lequel l'animal perçoit, apprend, mémorise et agit selon les informations auquel il est confronte dans son environnement. Les animaux on chacun leur propre monde sensoriel et il est primordial qu'ils s'y adaptent en développant des compétences spécialisées en fonction des informations sensorielles qui lui sont le plus utile. Il en est de même des informations qu'il est utile de stocker afin de pouvoir les utiliser ultérieurement. Les mécanismes sous-jacents à ces processus d'adaptation comportementale sont lies à la plasticité du système. Comment cette plasticité permet la mise en place de modules adaptatif reste actuellement une question sans complète explication. Le thème de cette thèse porte sur la plasticité et la modularité des capacités d'apprentissage et de mémoire olfactive chez Drosophila melanogaster. Dans la nature, la drosophile est confrontée à des environnements sensoriels complexes comprenant plusieurs stimuli sensoriels qu'elle doit associées à des renforcements négatifs ou positifs selon les conditions. En laboratoire il est possible de reproduire ce genre d'événement et j'ai ainsi pu tester le niveau d'adaptation des drosophiles à différent niveaux de traitement de l'information. Ces processus ont révèle l'existence de modules cognitifs plus ou moins spécialisés qui permettent a l'animal de s'adapter spécifiquement a son milieu. De plus, la réalisation d'une sélection artificielle sur les compétences à stocker les informations révèle l'implication de l'évolution dans la mise en place de ces modules.

Mid-Pleistocene Extinction of Deep-Sea Ostracoda?

A global extinction event has been documented in protozoan foraminifera in the late Pliocene to Pleistocene. The timing of the extinction event varied depending on location, however for Ocean Drilling Project Site 1125, disappearances occurred between 2.5 and 0.57 Ma, with the major decline approximately 1.1 Ma. In order to determine if this event affected benthic organisms other than protozoans, this study was undertaken to determine how podocopid ostracods (Crustacea) recovered Ocean Drilling Program Site 1125 responded. The present study was hindered by the small number of valves recovered; the fact that a large proportion of taxa found were undescribed and new to science; and the current state of taxonomic scheme that is under significant revision. These factors meant that a comprehensive comparison could not be achieved. Despite this, counts of ostracod valves and assessments of diversity from this study reveal a significant increase in both parameters from approximately 900-600 ka. Three possible causes were investigated to account for this increase, sediment type and sample size; affects of taphonomy, mainly dissolution; or an actual biotic 'event'. Statistical analyses showed that although sample size did have some effect, it was not the sole reason for the increase in ostracod numbers. Dissolution had an expected affect on the percentage of juveniles but no correlations were found with other sample characteristics. Sedimentation rate was investigated but this also proved unrelated. Therefore, it is suggested that the increase in total ostracod valves and diversity which occurs between 900 and 600 ka was in fact a natural, biotic 'event'. This preliminary evidence suggests that an oceanographic event that has negatively impacted on the foraminifers has had the reverse affect on the ostracod assemblage, in the sense that both population size and diversity increase during that time.

Effects of LTD-blocking Tat-GluR2 Peptide on Contextual Fear Memory Impairments Induced by Cannabinoids

The mechanisms underlying cannabinoid impairment of fear memory is not clear. This study investigated the effects of the synthetic cannabinoid HU210 and the endocannabinoid hydrolysis inhibitor JZL 195 on fear memory following contextual fear conditioning (CFC; an animal model of fear). The long-term depression (LTD)-blocking peptide Tat-GluR2 was utilized to investigate whether the expression of cannabinoid-induced LTD (CB-LTD) is required for the cannabinoid impairment of acquisition and consolidation of contextual fear memory. HU210 reduced freezing throughout the test phase of the acquisition protocol, which was not affected by pre-administration of Tat-GluR2. High and moderate doses of HU210 reduced freezing during the first and last half, respectively, of the test phase of the consolidation protocol, which was prevented by pre-treatment with Tat-GluR2. HU210 did not affect freezing during the test phase of the retrieval protocol. Thus, these results suggest that HU210 impairs acquisition and consolidation, but not retrieval of contextual fear memory, and that in vivo CB-LTD expression is required for HU210 impairment of the consolidation, but not acquisition, of contextual fear memory. We also observed that HU210 and JZL 195 do not facilitate the acquisition of contextual fear memory extinction.
