Evaluation of a Miniaturized Rotating Disk Apparatus for In Vitro Dissolution Rate Measurements in Aqueous Media : Correlation of In Vitro Dissolution Rate with Apparent Solubility

The general aim of this thesis was to evaluate a newly designed and constructed miniaturized rotating disk apparatus for in vitro dissolution rate measurements of different drug substances from all of the classes in the Biopharmaceutical Classification System (BCS). The new equipment is based on a low volume flow-through cell of Plexiglas, a gold plated magnetic bar and a special designed press. The disk of drug substance (approx. 5 mg) is placed eccentrically in the bar. Rotation speeds were set with a graded magnetic stirrer. An external HPLC pump delivered a continuous flow of aqueous medium to the flow-through cell during dissolution testing. A reversed phase high-performance liquid chromatography system using diode array detection (RP-HPLC-DAD) was coupled online to the new equipment. The injections from the miniaturized rotating disk outlet into the quantifying HPLC system were controlled by a six-position switching valve. The injection volumes from the valve and the autosampler, used for the external standards, were statistically evaluated to match each other volumetrically. No analyses were longer than three minutes, using isocratic mode. A traditional USP rotating disk apparatus was used as a reference system and the two instruments were shown to be statistically dissimilar in the numerical dissolution rate values probably due to different hydrodynamics, but had approximately the same precision/repeatability. When correlating the logarithmic values of the in vitro dissolution rate (G) with the apparent solubility (S), using shake-flask methodology in the solubility studies, the two apparatuses gave the same correlation patterns. Further correlation studies were done where the media components were altered by the use of different buffer species or additives into the buffers, such as inorganic salts. Chemometric tools, e.g. orthogonal partial least squares (OPLS), were used to better evaluate the most influential factors for G and S in different media. The most significant factor for a model basic drug substance (terfenadine) was pH, followed by the ionic strength (I) and added sodium chloride in one of the media. However, the surfactants in the Fasted State Simulated Intestinal Fluid (FaSSIF-V2) were found to be insignificant for G and S in this study (using a 95% confidence interval). The new miniaturized apparatus is a promising prototype for in vitro dissolution rate measurements both for early screening purposes and in dissolution testing during drug development, but needs further instrumental improvements.

Physico-Chemical Characterization of Drugs: Acidity and Solubility

The aim of the present work was to contribute to establish robust and high throughput methodology of interest in the "Drug Discovery" step commonly done in pharmaceutical laboratories. This purpose involves the exploration of the possibilities of the potentiometric Sirius methodology to determine both acidity constants and solubilities of drugs and other bioactive compounds and also to do a study about how to improve bioavailability of a model drug, Amphotericine 8, by increasing its dissolution rate. In the first part of this project, the acidic dissociation enthalpies and constants of anilinium, protonated tris (hydroxymethyl)- aminomethane (HTris+), benzoic and acetic acids, have been determined at several temperatures in pure water and in methanol/water mixtures by potentiometry method. The pK(a) values determined by this technique are in accordance with those values determined by ITC method in our laboratory and also with those other values from literature. Also dissociation enthalpies can be obtained from potentiometric pK(a) values by means of the Van't Hoff approach and these obtained values are in agreement with those ones determined directly by calorimetry in our laboratory. In the second part, we focused on studying about solubility. The Chasing Equilibrium method offers an alternative to the classical procedures to measure the solubility of compounds with acid-base properties. The method is fast and yields accurate results. In this work, the solubility of several compounds including acids and bases was determined through the Chasing Equilibrium approach. A study of experimental conditions in terms of sample weight was performed to measure solubilities. The study shows that only a limited range of weights, depending on the nature and solubility of the compounds, is adequate to obtain reliable results. In the third part of this work, the solubility vs. pH profiles of five ionizable drugs of different nature (a monoprotic acid, a monoprotic base, a diprotic base and two amphoteric compounds showing a zwitterionic species each one) have been determined through two different methodologies: the classical Shake-Flask (S-F) and the potentiometric Cheqsol methods using in both instances the appropriate Henderson-Hasselbalch (H-H) or derived relationships. The results obtained independently from both approaches are consistent. A critical revision about the influence of the electrolyte used as buffering agent in the S-F method on the obtained solubility values is also performed. Thus, some deviations of the experimental points with respect the H-H profiles can be attributed to specific interactions between the buffering electrolyte and the drug due to the hydrotrophic character of citric and lactic acids. In other cases, the observed deviations are independent of the buffers used since they are caused by the formation of new species such as drug aggregates (cefadroxil) or the precipitation of a salt from a cationic species of the analysed compound (quetiapine). In the forth part, the objective was to compare the dissolution behavior of tablets prepared from solid dispersions prepared in DMSO dissolvent with and without drug-carrier and also with and without surfactants in aqueous and acidic solutions. Amphotericine B was used as a model drug. Two types of carriers were used; mannitol, inulin. Solid dispersions with two different drug loads were prepared by freeze drying method. It was found that the drug dissolution rate in aqueous and acidic solutions was significantly increased in the presence of drug-carrier and surfactants. X-ray powder diffraction revealed that all solid dispersions were fully amorphous. / El objetivo del presente trabajo ha sido contribuir a establecer metodología robusta y de high throughput de interés en la etapa conocida como "Drug Discovery" que tiene lugar en los laboratorios farmacéuticos al inicio del proceso de desarrollo de nuevos fármacos. Este objetivo ha implicado la exploración de las posibilidades de la metodología potenciométrica establecida y comercializada por Sirius Analytical Ltd. para la determinación de las constantes de acidez y de la solubilidad de compuestos bioactivos y también un estudio sobre la mejora de la biodisponibilidad de un fármaco muy insoluble tomado como modelo mediante el aumento de su velocidad de disolución. En la primera parte de esta Tesis se han determinado potenciométricamente las constantes de disociación ácida y la variación de entalpía asociada de dos bases y dos ácidos tomados como modelo en agua pura y en mezclas de metanol/agua (0-60% w/w) a varias temperaturas (25-55°C). Esto ha implicado la puesta a punto de la estandarización del sistema potenciométrico en las condiciones de trabajo. Los valores de pK(a) determinados son concordantes con los que ofrece la literatura. Se han calculado también las entalpias de disociación en los distintos solventes binarios estudiados mediante la ecuación de Van't Hoff a partir de los valores experimentales de pK(a). La consistencia de los resultados obtenidos con los de la literatura, obtenidos directamente por calorimetría, confirma la robustez de la metodología. En la segunda parte de este trabajo, el estudio se centró sobre la determinación potenciométrica de la solubilidad de ácidos y bases mediante el método conocido como Chasing Equilibrium, como alternativa a los procedimientos clásicos de equilibración. El método es rápido y produce resultados precisos. Se ha realizado un estudio sobre las condiciones experimentales óptimas en términos de peso de la muestra para medir eficazmente la solubilidad. El estudio muestra que, en función de la naturaleza y solubilidad de los compuestos, existe un intervalo limitado de peso de muestra adecuado para obtener resultados fiables. En la tercera parte de la presente memoria, se estudian los perfiles de solubilidad en función del pH de cinco fármacos ionizables de naturaleza diferente, un ácido y una base monopróticos, una base diprótica y dos compuestos anfóteros que muestran una especie zwitteriónica cada uno. Se han determinado los perfiles de solubilidad mediante el método clásico de equilibración (Shake-Flak, S-F) y el potenciómétrico y, en ambos casos, se han utilizado las relaciones apropiadas de Henderson-Hasselbalch (H-H) o derivadas. Los resultados obtenidos de forma independiente por ambos métodos son consistentes. Se ha hecho un estudio crítico acerca de la influencia del electrolito utilizado como agente tampón en el método S-F en los valores de solubilidad obtenidos y se han observado algunas desviaciones de los puntos experimentales con respecto a los perfiles esperados que pueden ser debidas a interacciones específicas entre el electrolito tampón y el fármaco. En otros casos, las desviaciones observadas son independientes de los tampones utilizados y se pueden atribuir a la formación de nuevas especies tales como agregados iónicos del fármaco en estudio o la precipitación de una sal a partir de una especie catiónica del compuesto analizado. En la cuarta parte de esta memoria el objetivo ha sido estudiar la velocidad de disolución de comprimidos preparados a partir de dispersiones sólidas de un fármaco modelo con y sin portador del fármaco y también en presencia y en ausencia de tensioactivo en soluciones acuosas neutras y ácidas. Como fármaco modelo se estudió la Anfotericina B y se utilizaron como portadores manitol e inulina y como tensioactivos se ensayaron el deoxicolato de sodio (SDC) y el laurilsulfato de sodio (SLS). La difracción de rayos X reveló que el fármaco en estudio se hallaba en estado amorfo en todas las dispersiones sólidas estudiadas. Se puede concluir que la velocidad de disolución del fármaco se incrementa significativamente en presencia de portador y tensioactivo.

Improvement of dissolution rate of a new antiretroviral drug using an anti-solvent crystallization technology / Amélioration de la cinétique de dissolution nouvelle molécule antirétrovirale en utilisant la cristallisation par effet antisolvant

Cette étude concerne une nouvelle molécule antirétrovirale nommée CRS 74. Cette molécule présente une biodisponibilité limitée à cause de sa faible solubilité en phase aqueuse, sa mauvaise mouillabilité et sa faible vitesse de dissolution. Afin d'améliorer sa biodisponibilité, la molécule CRS 74 a été recristallisée par effet anti-solvant. Le solvant choisi est l'éthanol et l'anti-solvant l'eau. L'équilibre solide-liquide dans des mélanges binaires éthanol/eau a été mesuré à 30°C. Les solubilités obtenues ont été représentées en utilisant le modèle UNIQUAC pour le calcul des coefficients d'activité. Les solubilités expérimentales et calculées ont permis d'évaluer le ratio éthanol/eau optimum (25/75 % m/m) pour maximiser le rendement théorique en solide. Un mélange double jet avec pré-mélangeur type mélangeur en T a été choisi pour réaliser la cristallisation. Le solide cristallisé dans ces conditions semble plus aggloméré et son profil de dissolution comparé à celui du solide initial est inchangé. De plus, l'étude des cristaux obtenus en sortie de pré-mélangeur a montré que les vitesses de croissance et d'agglomération des cristaux sont élevées. Des additifs ont donc été utilisés en vue de modifier les propriétés de dissolution des cristaux, et d'optimiser les paramètres de formulation et de cristallisation. Les microcristaux produits en présence d'additifs présentent des profils de dissolution significativement plus rapides que les cristaux de la molécule initiale. Cette modification est attribuable à la modification de taille des cristaux et l'amélioration du mouillage en raison des interactions spécifiques entre la surface des cristaux et les additifs. / This study concerns a new antiretroviral drug named CRS 74. This molecule has a limited bioavailability because of its low aqueous solubility, poor water wettability and low dissolution rate. In an attempt to improve these properties, CRS 74 was recrystallized by using a Liquid Anti-Solvent (LAS) crystallization process. The chosen solvent is the ethanol and the anti-solvent the water. So solid-liquid equilibria in binary mixtures ethanol/water were measured at 30°C. The obtained solubility data were represented using UNIQUACbased model. The experimental and calculated solubilities permitted to estimate the optimal ethanol/water mass ratios (25/75 % w/w) in order to maximize the theoretical yield of solid. A double-jet with premixing (T-mixer) has been used to mix the two solutions. Particles of recrystallized CRS 74 seemed more agglomerated and the dissolution profile was not modified compared to the original drug. Furthermore, the study of crystals obtained at the exit of the mixer showed that the growth and agglomeration rates of crystals are high.In an attempt to improve its dissolution properties, CRS 74 has been recrystallized using different additives to optimize process and formulation parameters. Conclusively, produced microcrystals exhibited significantly faster dissolution rates than the original CRS 74 crystals. The improved dissolution is attributable to the modification of the particle size of drug crystals and enhancement of wetting properties due to specific interactions between the drug and the additives.

Enhancing Mineral Carbonation of Olivine with CO2 / Förbättring av mineral kolsyrning av olivin med CO2

Koldioxidutsläpp (CO2) från energiproduktionsindustrin och transportsektorn globalt påverkar miljön negativt. Länder har enats om att minska utsläppen för att nå målet om en genomsnittlig temperaturökning på 1,5 °C till 2030. Trots detta förväntas de globala utsläppen av CO2 från fossila bränslen och industriella processer vara cirka 40 Gton per år fram till 2100. För att dra nytta av CO2-utsläppen och skapa värdefulla produkter med negativa utsläpp är mineralkarbonatisering en önskvärd process. Denna process innebär att CO2 och mineraler löses upp i en alkalisk lösning och bildar stabila produkter. Faktorer som partikelstorlek hos mineralerna och CO2-lösningshastigheten påverkar mineralkarbonatiseringens hastighet. Experiment utfördes med en batchreaktor från Paebbl AB och en matematisk modell utvecklades i Matlab. Resultaten jämfördes för olika partikelstorlekar i tre motståndsfall. Större partikelstorlek hos olivin visade sig öka tiden för total konvertering, oavsett motståndstyp. De modellerade motstånden beskrev inte tillräckligt processen och indikerade att alla tre motstånd har en samtidig och enhetlig effekt på olivinmineralisering, utöver eventuella begränsningar som föroreningar och biprodukter. Mineraliseringsexperiment med 20 μm partiklar under en timme gav 34,4% omvandling, medan 10 μm partiklar under två timmar gav 46,7% omvandling. En inledande undersökning av massöverföringsbegränsningar visade att CO2-lösningshastigheten inte är den begränsande faktorn, utan lägre omrörningshastigheter och beteendet hos (CO2 + olivin)-systemet behöver ytterligare studeras. Framtida forskning bör fokusera på att lösa dessa begränsningar. / Carbon dioxide (CO2) emissions from the energy production industry and the transportation sector globally negatively affect the environment. A prominent example is the interconnection of carbon with the greenhouse effect. Countries have agreed to mitigate their emissions and try to fulfill the target of 1.5 oC average temperature increase by 2030, but in order to do so the global emissions of CO2 from fossil fuels and industrial processes will still lead up to the astonishing amount of 40 Gtons of CO2 each year until 2100.  It is apparent that processes that try to take advantage of the emitted CO2 creating valuable products with negative emissions are highly desired. One of these is mineral carbonation, where CO2 and minerals dissolve in an alkaline solution and form stable products. Many factors affect the rate at which mineral carbonation happens. The effect of the particle size of the mineral in the process will be investigated, along the CO2 dissolution rate through the overall gas-liquid mass transfer coefficient (kLa), in order to get a better understanding of the process. Experiments were conducted with a batch reactor provided by Paebbl AB and a mathematical model was developed in Matlab. The experimental and numerical results, in regards to the particle size, were then compared for the cases of three resistances. This model can be developed further for use in a continuous mineralization process. The results revealed that increasing the particle size of olivine leads to a significant increase in the time required for total conversion, irrespective of the resistance type. The modelled resistances were found to inadequately describe the process, suggesting a simultaneous and uniform effect of all three resistances on olivine mineralization, in addition to the effect of other possible limitations such as impurities and by-products. Mineralization experiments with 20μm particles and a duration of 1 hour led to 34.4% conversion, whereas experiments with 10μm particles and a duration of 2 hours resulted in 46.7% conversion. Finally, the initial investigation of the mass transfer limitations in a system of CO2 and water led to an average kLa coefficient of 191 h-1, suggesting that the CO2 dissolution rate is not the limiting factor. However, the impact of lower stirring rates remains unexplored due to the absence of appropriate instrumentation and the behaviour of the (CO2 + olivine) system should also be studied. Future research should aim to address these limitations.

L’importante augmentation de fluage des plaques de plâtre en milieu humide est un vieux problème dans l’industrie du bâtiment, dont l’origine n’est pas encore établie. Afin d’en comprendre le mécanisme une étude à trois échelles (macro : réponse mécanique, micro : cinétique de dissolution et nano : observation atomique) a été réalisée. Une corrélation forte existe entre le fluage du plâtre en immersion et la cinétique de dissolution du gypse. La concordance de cette corrélation avec une loi de déformation par dissolution sous contrainte, très utilisés en géologie, permet de proposer la dissolution sous contrainte comme un des mécanismes responsables du fluage du plâtre en immersion. L’évolution de la topographie de la surface du cristal de gypse immergé dans une solution aqueuse de gypse est observée par microscopie à force atomique (AFM). La cinétique de migration des marches atomiques est très dépendante de la sous-saturation de la solution, de la force d’appui de la pointe de l’AFM ainsi que des additifs utilisés. L’étude de l’influence de la force d’appui sur les vitesses des marches met en évidence la présence de deux mécanismes complètement différents. A fortes forces (> 15 nN) on observe un mécanisme d’usure de la surface, tandis qu’à faibles forces (< 10 nN) le mécanisme observé semble être la dissolution sous contrainte. L’évolution des vitesses des marches atomiques avec la force appliquée par la pointe est concordante avec une loi connue de dissolution sous contrainte. / The huge enhancement of the creep of plasterboard by humid environments is an old problem in the building industry, but its origin remains unknown. To understand this mechanism a three scales study (macro : mechanical behavior, micro : dissolution kinetics, nano : atomic observation) has been done. There is a strong correlation between wet plaster creep and gypsum dissolution kinetics. The concordance between this correlation and the law of deformation by pressure solution, well-known in geology, permits to propose pressure solution as one of the mechanisms responsible of wet plaster creep. The topological evolution of the cleaved surface of a gypsum single crystal during its dissolution in a flowing under-saturated aqueous solution has been observed with an atomic force microscope. The kinetics of step migration strongly depends on the saturation state of the solution, the force applied by the tip on the surface, as well as the used additives. The study of the influence of the force applied by the tip on the step velocity evidence two different dissolution enhancement regimes. At high forces (> 15 nN) a corrosive wear behavior is observed, while at low forces (< 10 nN) pressure solution is the observed mechanism. The step velocity evolution with the force obeys the known kinetic law of pressure solution.

A comparative study of the effect of spray drying and hot-melt extrusion on the properties of amorphous solid dispersions containing felodipine

No / OBJECTIVES: To compare the properties of solid dispersions of felodipine for oral bioavailability enhancement using two different polymers, polyvinylpyrrolidone (PVP) and hydroxypropyl methylcellulose acetate succinate (HPMCAS), by hot-melt extrusion (HME) and spray drying. METHODS: Felodipine solid dispersions were prepared by HME and spray drying techniques. PVP and HPMCAS were used as polymer matrices at different drug : polymer ratios (1 : 1, 1 : 2 and 1 : 3). Detailed characterization was performed using differential scanning calorimetry, powder X-ray diffractometry, scanning electron microscopy and in-vitro dissolution testing. Dissolution profiles were evaluated in the presence of sodium dodecyl sulphate. Stability of different solid dispersions was studied under accelerated conditions (40 degrees C/75% RH) over 8 weeks. KEY FINDINGS: Spray-dried formulations were found to release felodipine faster than melt extruded formulations for both polymer matrices. Solid dispersions containing HMPCAS exhibited higher drug release rates and better wettability than those produced with a PVP matrix. No significant differences in stability were observed except with HPMCAS at a 1 : 1 ratio, where crystallization was detected in spray-dried formulations. CONCLUSIONS: Solid dispersions of felodipine produced by spray drying exhibited more rapid drug release than corresponding melt extruded formulations, although in some cases improved stability was observed for melt extruded formulations.

Coupling source term, mineral reactivity and flow in radionuclide transport

The focus of this work is to investigate the dissolution of MW25, a non-radioactive simulant of UK high-level nuclear waste borosilicate glass, and to predict its performance in the near field of a geological repository. A single-pass flow-through (SPFT) experimental system was used to measure the forward dissolution rates of MW25. Experiments were conducted in two parts. Experiment Part 1 considers the dissolution of the waste glass in deionised water at 40 and 90 oC and circum-neutral pH. Experiment Part 2 considers the dissolution of the waste glass in simulant groundwaters, with similar compositions to groundwaters of Callovo-Oxfordian clay (lower-strength sedimentary rock (LSSR)) and Borrowdale Volcanic Group rocks (higher-strength rock (HSR)), at 40 oC and pH 7. The forward dissolution rate measured in deionised water was found to be approximately one order of magnitude higher at 90 oC than at 40 oC. A similar release was observed for Si, Mg and Al at 40 oC and 90 oC, whereas the B, Cs, Na, Li and Mo showed an order of magnitude increase when the temperature was increased from 40 to 90 oC for low q/S values. The activation energy (Ea) of the reactions shows that the dissolution process is a surface phenomenon. At 90 oC the net effect of the processes governing MW25 dissolution led to the preferential release of boron and alkali metals relative to the release of Si during the transient dissolution stage, accompanied by an increase in the concentration of silicic acid. This suggests that the solution activity of silicic acid at a higher temperature has a weak influence on the release of the mobile elements. The forward dissolution rate measured in LSSR simulant groundwater was found to be slightly higher than that measured in HSR simulant groundwater. The dissolution behaviour of MW25 in both groundwaters is consistent with its behaviour in deionised water at 40 oC, with the dissolution rates of elements increasing as flow rates were increased. However, forward dissolution rates measured in the simulant groundwaters were lower than the forward dissolution rates measured in deionised water under these experimental conditions. This is attributable to the interaction of the components of the simulant groundwaters with the glass, as revealed by post-reaction surface analyses, and a consequential lower alkalinity of the leachates collected in the experiments with simulant groundwater than in deionised water. Reactive chemical transport simulations of waste glass dissolution and radionuclide release in a hypothetical near field were conducted over a time span of a million years with GoldSim. The results showed that enclosing the waste glass in a steel canister covered by a copper canister and emplacing the waste package in a granite host rock is optimal for the long-term isolation of the radionuclides. The waste glass was found to play a significant role in the overall performance of the near field. This study features a new method for estimating the surface area of reacted glass powder more accurately than the geometric surface area estimate, which is the preferred standard method among researchers.
