Utilizace trehalózy u orchidejí: evoluce genů trehalázy / Utilization of trehalose in orchids: evolution of trehalase genesŠoch, Jan January 2017 (has links)
All orchid species studied so far have been shown to participate in orchideoid mycorrhizal symbiosis. Morover, this symbiosis is absolutely vital component of their life cycle. Exchange of nutrients occurs between symbionts where the fungi provides the orchid with energy and carbon supply at least in its early developmental stages. This study focuses on the possible role of trehalose in this transfer. In vitro experiments have showed in five species from three different subfamilies of Orchidaceae family that they can utilize trehalose comparably with sucrose and glucose. Thus, the ability of trehalose utilization seems to be conserved among orchids. Trehalase enzyme activity was localized histochemically in orchid mycorrhizas. The activity strongly colocalized with colonized tissue supporting a hypothesis that trehalose transfer occurs in this site and is mediated by trehalase. Using bioinformatic methods, trehalase gene duplications were identified in many taxons of Embryophyta including three orchid species. Interestingly, highest number of trehalase gene copies was identified in genome of orchid Dactylorhiza majalis. Hematopoiesis is a precisely regulated process, dependent on the activity of hematopoietic cytokines and their receptors. Due to an extra round of whole genome duplication in teleost fish, two paralogs of many important genes, including some hematopoietic cytokines and their receptors, are present in the zebrafish (Danio rerio) genome. In this project, we have been investigating the role of zebrafish Kit ligands in hematopoiesis. Kit ligand is a pleiotropic cytokine, which is essential for vertebrate erythropoiesis; however, in zebrafish, no such role has been reported so far. To determine the function of zebrafish paralogs of Kit ligand (Kitlga and Kitlgb) in hematopoiesis, we performed in vivo and ex vivo gain- and loss-of-function experiments. Strikingly, we were the first to report the synergistic cooperation of zebrafish Kitlga with erythropoietin and dexamethasone, enabling the growth of kidney marrow-derived suspension cells and providing optimal conditions for the expansion of adult erythroid progenitors. Odorant-binding proteins (OBPs) are small soluble proteins expressed at high levels in the proximity of olfactory receptors. OBPs act as solubilizers and carriers of the lipophilic odorants in the aqueous mucus of mammals and other vertebrates. OBPs have now been studied nearly thirty years, but in comparison to the wealth of data available on their structural chemistry and molecular biology, our knowledge about gene expression and function of these proteins is still insufficient. This work provides new insights into the tissue specificity of OBP and presents several new sequences of genes governing these proteins in selected species of mice.

Polyploidization (whole-genome duplication) is widely considered one of the most important evolutionary forces driving the diversification of flowering plants. Polyploids tend to originate recurrently and many plant species retain individuals of two or more different ploidy levels in certain parts of their distributional range of even within their populations. The main aim of this thesis was to address the understudied aspects of polyploid speciation by employing new, convenient methods and/or studying plant model systems with unique features. Difference in monoploid genome size of Tripleurospermum inodorum (Asteraceae) cytotypes provided a unique opportunity for addressing the rate of spontaneous polyploidization in natural populations by enabling the easy distinction of neopolyploid mutants from long-established polyploids in routine flow-cytometric analyses. Repeated ploidy screening in mixed-ploidy populations of annual T. inodorum have been, to our knowledge, the very first attempt to document temporal changes in cytotype composition in situ. In spite of considerable between- year oscillations in cytotype frequencies, both diploids and tetraploids usually persisted locally for several consecutive years. Adaptive evolution of Toll-like receptors in birds Hana Velová, PhD thesis 6 Abstract Toll-like receptors (TLRs) are one of the key and presumably also evolutionary most original components of animal immune system. As Pattern recognition receptors they form the first line of innate immune defence against various pathogens. The proper receptor binding of pathogenic ligands is crucial for their correct recognition and for subsequent triggering of an appropriate immune response. Because there exists a direct interaction between the receptor surface and the pathogenic ligand, host-pathogen coevolution on molecular level can be predicted. Thus, through variability of their ligands, TLRs are exposed to extensive selective pressures that may be detected on both genetic and protein levels. Surprisingly, the variability we revealed in birds is even higher than previously expected based on the reports from other vertebrates, mainly mammals. In my doctoral thesis I summarise the results of my contribution to the avian TLR research. We were the first who experimentally verify the absence of functional TLR5 in several avian species and duplication of TLR7 in others. This thesis is focused on network behavior analysis (NBA) designed to detect network attacks. The goal of the thesis is to increase detection accuracy of obfuscated network attacks. Methods and techniques used to detect network attacks and network traffic classification were presented first. Intrusion detection systems (IDS) in terms of their functionality and possible attacks on them are described next. This work also describes principles of selected attacks against IDS. Further, obfuscation methods which can be used to overcome NBA are suggested. The tool for automatic exploitation, attack obfuscation and collection of this network communication was designed and implemented. This tool was used for execution of network attacks. A dataset for experiments was obtained from collected network communications. Finally, achieved results emphasized requirement of training NBA models by obfuscated malicious network traffic.
