There is described a electromigration separation method for the analysis of low concentrations of copper with the possibility to detect a Wilson disease in my thesis. There were developed a methodology and propitious electrolyte systems, which can pre-concentrate copper prior to its on-line analysis. The developed electrolyte system for analysis had the following composition: LE1 (1.10-2 M CH3COOH, 2.10-2 M CH3COONH4, 2.10-4 M SPADNS), LE2 (1.10-2 M CH3COONH4, 1.10-2 M NH4OH, 1.10-4 M BKP, 5.10-5 M PAR, 5.10-3 M C6H17N3O7) and DE (7.10-5 M Cu(CH3COO)2, 1.10-3 M CH3COONH4 a 1.10-2 M CH3COOH). With a suggested electrolyte was achieved almost seventyfold accumulation of copper during 42 minutes, which lowered the concentration detection limit on the level convenient for the detection of Wilson disease. Complete analysis took about 98 minutes. There was accomplished a conductometric detection of copper in my thesis, however the developed electrolyte system can also analyze a copper by photometric detection, which is more sensitive.

The thesis deals with the determination of selected ion herbicides in surface waters and in the first part describes the herbicides. Given the broad spectrum herbicide further work deals mainly with glyphosate. The next section describes the electromigration methods and the methods by which the herbicides were determined. Finally, it describes how the development of an electrolyte system along with the use of graphical possibilities of computer technology. In conclusion are presented and commented on the results of measurements of glyphosate in surface waters, compared with foreign literature and assessing the suitability of methods for the analysis of contaminants in the environment.

The presence of inorganic ions is one of the criteria for thermal water assessing. The most important anions that affect the water quality and allow their use for therapeutic purposes, include sulfates, chlorides, nitrates and bicarbonates. Ions (as sodium, potassium, calcium and magnesium) are the significant cations. The ion chromatography is used for separation of these substances and also some methods based on electrophoretic migration of ions in an electric field. The electromigration method of isotachophoresis was chosen for determination of selected ions in water samples collected from thermal boreholes in Pasohlávky.

The diploma thesis deals with the determination of selected heavy metals in wastewater, while the first part describes electromigration methods and individual heavy metals. In the next part of the thesis is describe, the problems of sewage sludge and methods commonly used for determination of heavy metals in wastewater. The practical part of the thesis describes the development of the electrolyte system together with the use of computer technology. Following is a discussion of the possibility of removing heavy metals from wastewater. In conclusion, the results of heavy metal measurements, comparison with foreign literature and assessment of suitability of the method for analysis of hazardous substances at low concentrations in the environment are presented and commented.

Evaluating data in capillary zone electrophoresis usually involves many steps that require using several different programmes. Apart from evaluating the electrophoreogram itself, it is usual to process the obtained data in some other way. For example, a suitable model is fit to the data in order to obtain physical and chemical parameters of the separation (e.g. stability constant in case of complexation). It is also important to know the accuracy of the evaluation (the calculation error). In this work, new parts of the Eval programme, originally developed for electrophoreogram evaluation, were implemented. The programme now automatically estimates the Haarhoff-van der Linde function (solution of continuity equation in capillary) parameters for analyte peak. Complexing agents are often used to improve the separation in the capillary zone electrophoresis. Complexation in the capillary can be described by its physical and chemical parameters. A new part was added to the Eval programme that allows the user to fit a rectangular hyperbole function to the obtained data. Thus, the regression parameters of this dependence can be gained. Nelineární jevy v elektrokinetické chromatografii / Nonlinear phenomena in electrokinetic chromatography

Capillary electrophoresis often uses complexing agents since the interaction between the analyte and the complexing agent can result in achieving or improving the separation. Examples of such methods can be electrokinetic chromatography or affinity capillary electrophoresis (ACE). ACE is used to determine the complexing parameters. In case of chiral separation, this issue gets complicated, since the parameters of the two analytes (enantiomers) are not completely independent to one another. Therefore, a procedure has been proposed in this thesis, that should always be used to evaluate the complexing parameters of two enantiomers. Statistical evaluation of these parameters was assessed as well. This work also proposes a method that allows to determine the relative migration order of two enantiomers in two different complexing separation systems. The mathematical description of electrophoresis is based on continuity equations, that are inherently nonlinear. However, these equations can be linearized to obtain an approximate analytical solution. There was recently presented a generalized model, that enables inclusion of complete complexing equilibria in the theoretical description of electromigration.
