Padronização da metodologia de congelamento de células da granulosa antrais humanas para suporte no co-cultivo com oócitos imaturos / Cryopreservation of human granulosa cells for future use in assisted reproductive procedures

As técnicas de cultivo de folículos e oócitos in vitro, com o objetivo de se obter oócitos maduros para procedimentos de Reprodução Assistida (RA), têm sido aplicadas em diferentes contextos. O sucesso destes procedimentos está diretamente relacionado ao sistema de cultivo utilizado. A utilização de células da granulosa (CG) humanas cultivadas in vitro como um suporte para o co-cultivo destes oócitos imaturos e folículos tem sido descrita por alguns autores. A criopreservação destas células, considerando-se o contexto de sua obtenção em procedimentos de RA, permitiria a viabilização da aplicação destas células na prática clínica diária. Sendo assim, o objetivo deste estudo foi padronizar o congelamento de células da granulosa (CG) humanas para aplicação em sistemas de co-cultivos de folículos e oócitos imaturos. Foram obtidas CG de 20 voluntárias em tratamento de reprodução assistida, células de 10 voluntárias foram cultivadas em meio ?-MEM suplementado para interrupção da luteinização e congeladas após 48 horas em container \"Cryostep\" (grupo 2C- 2 cultivos) (etapa 2) e células de 10 voluntárias foram congeladas em container \"Cryostep\" sem cultivo prévio (grupo CD- congelamento direto) (etapa 3). Após o descongelamento estas células foram (re)cultivadas por 144 horas, com troca de meio em 48, 96 e 144 horas para avaliações da produção de estradiol (E2) e progesterona (P4) (ng/mL). Verificamos redução na contagem celular e na viabilidade celular tanto no método de congelamento direto (CD) quanto no método com dois cultivos (2C) após o descongelamento (p<0,05), e isso se refletiu na produção de estradiol e progesterona que foi maior nas culturas de células frescas em relação às células criopreservadas (p<0,05). Porém, a relação de E2/célula foi mantida após o descongelamento, sugerindo que esta redução na produção se deve à redução no número de células, as que sobrevivem se mantém normofuncionantes (p=0,23).O CD foi mais eficiente pois permitiu uma maior recuperação celular e uma melhor viabilidade quando comparado ao grupo 2C. A relação estradiol/progesterona foi mantida em todos os tempos de cultivo, fresco, CD e 2C (p>0,05), indicando que a característica funcional destas células foi preservada após o descongelamento. Concluímos que a criopreservação de CG humanas obtidas durante a captação de oócitos compromete a contagem celular e a viabilidade geral da cultura, entretanto, a capacidade funcional e a característica destas células se mantêm preservadas (manutenção das relações E2/célula e E2/P4) / Follicle and oocyte in vitro culture techniques, aiming to obtain mature oocytes for Assisted Reproductive Treatments (ART), have been applied to different contexts. The success of these procedures depends on the culture system used. The use of human granulosa cells (GC) in co-culture systems for follicle and oocyte maturation have been described by some authors. The cryopreservation of these cells, considering the context in which they are obtained during ART, would enable the usage of these cells in such procedures in daily clinical practice. Thus, the objective of this study was to standardize the freezing protocol for human granulosa cells (GC) for future applications in co-culture systems for follicle and oocyte maturation. Twenty volunteers submitted to ART donated their granulosa cells after oocyte retrieval, 10 were cultivated previously in order to interrupt the luteinization process and then frozen \"Cryostep\" container (group 2C- two cultures) (step 2) and 10 were directly frozen with no previous culture in the \"Cryostep\" container (group DF- direct freeze) (step 3). After thawing these cells were (re)cultured for 144 hours, with medium exchange at 48, 96 and 144 hours to evaluate the estradiol (E2) and progesterone (P4) production (ng/mL). After thawing, there was a reduction in the cell number (p<0,05) and cell viability in both methods, the direct freezing (DF) and the two cultures (2C) (p<0,05); this had an impact in the production of estradiol and progesterone, which were higher in fresh cultures than in the frozen ones (p<0,05). However, the E2/cell ratio was maintained after thawing (p=0.23), suggesting that this impairment in steroid production was probably due to the reduction in the cell count. The cells that survive remain functionally normal. The DF was more efficient since it allowed greater cell recovery and better viability when compared to 2C. The estradiol/progesterone ratio was maintained in all culture times, in the fresh, DF or 2C groups (p>0.05), indicating that the functional characteristic of these cells was preserved post-thawing. We conclude that cryopreservation of human GC obtained during oocyte retrieval compromises the cell count and the overall viability of the culture; however, the functional capacity and the characteristic of these cells are preserved (maintenance of E2/cell and E2/P4 relations)

Com o objetivo de analisar a influência do fenômeno de congelamento e o tempo de criopreservação sobre as propriedades biomecânicas de tendões, os autores estudaram 40 tendões de Aquiles obtidos de 20 cadáveres humanos. De cada cadáver foram retirados dois tendões, sendo que um foi testado a fresco e o contralateral congelado a - 85o C, durante um período de seis ou 12 semanas. Os corpos de prova foram submetidos a ensaios de tração, com análise de suas propriedades biomecânicas. Os resultados foram comparados estatisticamente pelo método de "t-student", com índice de significância de 0,05, não havendo diferença significativa entre os grupos "congelado" e "a fresco" / For the purpose of analyzing the influence of the freezing phenomenon and cryopreservation time over the biomechanical properties of tendons, the authors studied 40 Achilles tendons from 20 human cadavers. Each cadaver had two tendons removed, one of which was tested whilst fresh and its contralateral whilst frozen at - 85o C, for a period of six or 12 weeks. The trial items were submitted to tensile testing in order to analyze their biomechanical properties. The results were compared statistically using the T-Student method, with a significance ratio of 0.05, there being no significant difference between the 'frozen' group and the 'fresh' group

Concreto de alto desempenho aplicado a sistemas de processamento e armazenagem de alimentos em baixas temperaturas / High performance concrete applied to storage and processing system buildings at low temperatures

O desenvolvimento de um concreto de alto desempenho aplicado a sistemas de processamento e armazenagem de alimentos em baixas temperaturas teve por objetivo atender às indústrias de alimentos no Brasil que se utilizam da tecnologia do frio em seu processo industrial. A hegemonia mundial do mercado de carnes é brasileira, sendo que nossas indústrias são responsáveis por 33% desse mercado. A ausência de estudos para aprimorar e tornar as plantas industriais mais duráveis e condizentes com a importância do setor foi determinante na delineação do tema desta tese. Desenvolveu-se um concreto de alto desempenho frente às possíveis situações em uma planta industrial: baixas temperaturas em ambientes secos e baixas temperaturas em ambientes sujeitos à umidade. A temperatura mínima a qual foram submetidos os concretos analisados foi de 35°C. O método de dosagem e as diretrizes calcadas no reforço da matriz da pasta de cimento e refinamento de poros mostraram-se como uma alternativa para vencer as agressividades causadas pelas baixas temperaturas ao concreto. Até então, estudos sugeriam um sistema de ar cujos vazios teriam diâmetros em torno de 250 µm espaçados com esta mesma distância entre eles e com conteúdo de 6±1% para a proteção de concretos expostos a baixas temperaturas. A produção de um concreto coeso, com teor de ar de 3,5%, poros com diâmetro médio de 0,02 µm e com área específica de 2,84m²/g, provaram ser duráveis quando expostos ao congelamento e a ciclos de gelo e degelo. O fator de durabilidade para esses concretos foi de 97%. Além da durabilidade, o projeto de dosagem contemplou o aspecto econômico, produzindo 1MPa com 7,8 kg de cimento por metro cúbico de concreto. Os resultados foram obtidos por meio de ensaios dinâmicos, microscopia eletrônica de varredura, porosimetria por intrusão de mercúrio, absorção de água por imersão, além dos ensaios mecânicos de resistência à compressão, à tração na flexão e módulo de elasticidade. / Brazil has rapidly emerged as one of the world\'s leading meat exporters, being responsible by 33% of this market. In face of that, a high performance concrete (HPC), that can be applied to storage and processing system buildings at low temperatures, has been developed with the aim of answering the needs of brazilian food manufacturers that use cooling technology during industrial process. The lack of studies to improve industrial plants and make them more durable was determinant to delineate this thesis subject. A high performance concrete was designed to resist to low temperatures at dry environments as well as at moist ones. Concrete was exposed to a target temperature of -35°C. The dosage method and the guidelines based on the strengthening of bulk cement and the porous refinement showed to be an alternative to overcome the aggressiveness caused by low temperatures to concrete. To this day, studies have suggested air voids systems to protect concrete from low temperature effects. According to them, the air voids should have about 250 µm of diameter, spaced from each other this same distance and with an air content of 6±1%. However, making a cohesive concrete, with 3,5% of air content and an average diameter porous of 0,02 µm with an specific surface of 2,84m²/g, resulted in a durable concrete, even when exposed to frost / thawing cycles. The durability factor of these concretes achieved 97%. Besides durability, the dosage design brought economical advantages, producing 1 MPa with 7,8 kg of cement per cubic meter of concrete. The results were obtained through dynamic tests, scanning electron microscopy, mercury intrusion porosimetry and water absorption by immersion. In addition, tests of compression, tension strength and elastic modulus were carried out.

Untersuchungen zur Qualität von peripheren Blutstammzellpräparaten

Das moderne Therapiekonzept der Hochdosischemotherapie von soliden Tumoren und hämatologischen Neoplasien erfordert die prätherapeutische Sammlung, die extrakorporale Reinigung und die nachfolgende Tieftemperaturlagerung von menschlichen Blutstammzellen zur späteren Retransplantation. Stammzellpräparate (Transplantate) von 22 Patienten wurden durch extrakorporale Trennung von peripheren Blut hergestellt. Von jedem Patienten wurde eine Probe mit Dimethylsulfoxid (DMSO) versetzt, bei - 196 oC gelagert und nach ca. 3 Wochen aufgetaut. Eine Vergleichsprobe jedes Patienten wurde ohne den Zusatz von DMSO und ohne einen Einfrierschritt untersucht. Die Exposition von DMSO, das Frieren und Auftauen der Zellen zeigte eine geringfügige des Zell- und Kerndurchmessers, eine konstante Anzahl an Mitochondrien und eine Reduktion der Vesikel. Ein markantes Merkmal der Schädigung nach Tieftemperaturlagerung war das Auftreten von Flüssigkeitseinlagerungen in die Kerndoppelmembran und die Ausbildung von zisternenartigen Erweiterungen des endoplasmatischen Retikulums. Regelmäßig konnten Mitochondrien von verringerter Größe und randständig kondensierter Cristae gefunden werden. Insgesamt konnten keine schwerwiegenden zellulären Schäden beim Vergleich der unbehandelten und der DMSO- versetzten , eingefrorenen und wieder aufgetauten Proben festgestellt werden. Die morphologischen Ergebnisse korrespondieren mit der vollständigen Restitution aller hämatopoetischer Zelllinien nach Transplantation. / The modern therapeutic concept of high-dose chemotherapy of solid tumors and hematologic neoplasias demands a pretherapeutic harvest, an extracorporal purification and an consecutive deep temperature storage of human blood stem cells which will be retransplanted later. Stem cell preparates (transplants) of 22 patients were produced by extracorporal separation of peripheral blood. From each patient a stem cell specimen was mixed with dimethylsulfoxide (DMSO), storaged at - 196 °C and thawed after about 21 days. A corresponding specimen of each patients material was investigated without DMSO addition and without freezing under native conditions. The DMSO exposed, frozed and again defrosted cells showed a mild increase of total cell and nucleus diameters, a constant number of mitochondrias and a reduction of vesicles. A markedly feature of deep temperature damage was the occurance of liquide storages in the nucleus double membrane and the forming of cisterne-like enlargement of the endoplasmatic reticulum. Persistantly we found mitochondrias with reduced size and marginal condensed cristae. Alltogether there were no severe cellular damages in the comparative investigated overlifed specimen cells of the same patient with and without DMSO and deep temperature storage. The morphological results correspond with clinical investigations of a sufficient restitution of all hematopoietic cell lineages in transplanted patients.

Heterosis in the freezing tolerance of Arabidopsis thaliana (L.)Heynh

Die vorliegende Arbeit beschäftigt sich mit Heterosis in der Frosttoleranz von Arabidopsis-Kreuzungen. Die genetische Basis von Heterosis, der Verbesserung heterozygoter F1-Hybriden gegenüber den homozygoten Eltern, ist unbekannt. Vermutet werden Dominanz, Überdominanz oder Epistasie. Die zur Kreuzung verwendeten Parentalakzessionen entstammen unterschiedlichen Klimaten und differieren stark in ihrer Frosttoleranz (Lethaltemperatur, LT50). Von 24 reziproken Kreuzungen mit C24 und Col-0 wurden LT50, Prolin- und Zuckergehalt bestimmt. Die Untersuchungen an nicht akklimatisierten und kälteakklimatisierten Pflanzen zeigen häufiger Heterosis in C24- als in Col-Hybriden mit klarem Anstieg nach dem Akklimatisieren. Es besteht eine klare Abhängigkeit der Frosttoleranz zum Zucker-, Prolin- und Flavonoidgehalt, sowie zwischen der Stärke der Heterosis in Frosttoleranz und Metabolitgehalten. GCMS-Messungen bestätigen diese Ergebnisse. Es wurden 40 Metabolite detektiert, von denen viele signifikant mit der Frosttoleranz korrelieren und/oder deren Heterosis mit der Heterosis der LT50 korreliert. Sechs dieser Stoffe sind wichtige Komponenten des Citratzyklus, was auf eine Rolle von Teilen des Zyklus in der Heterosis der Frosttoleranz und eine Veränderung seiner Flussraten hindeutet. / We investigated heterosis in freezing tolerance of 24 reciprocal Arabidopsis-crosses with C24 and Col-0. The underlying genetic mechanisms of Heterosis, the enhancement of F1-hybrids in comparison to their homozygous parents, are unknown. Different mechanisms such as dominance, overdominance or epistasis are suggested. Parental freezing tolerance (LT50) was shown to correlate with the original habitat temperature. Besides the LT50, proline and sugar contents (glc, fru, suc, raf) have been measured on non-acclimated and cold-acclimated plants. Metabolite profiling and flavonoid measurements revealed significant stronger heterosis in C24- than in Col-crosses. Heterosis increases after cold acclimation. Freezing tolerance clearly correlate with the contents of sugars, proline and several flavonols, as well as the strength of heterosis in freezing tolerance with the metabolite content. GCMS-measurement confirmed these results. Fourty metabolites, of which many significantly correlate with LT50 and/or with heterosis in metabolite content and in LT50, were found. Six of these are important compounds in the TCA-cycle. Changes in flux rates of the TCA-cycle could be connected to Heterosis for the first time. Negative correlation between Heterosis in freezing tolerance and metabolite accumulation, points to a role of parts of the cycle in crosses and to a change of flux rates.

Sustainable Agri-food Production and Consumption

Agri-food production is necessary to sustain the growing global population, but it adversely impacts the environment in various ways, including climate change, eutrophication, acidification, land and water uses, and loss of biodiversity, etc. These environmental impacts can also negatively affect human health, which could in theory outweigh the health benefits of nutritious food. While better agricultural practices need to be developed and applied to minimize the environmental burdens associated with the production chains, consumers are expected to implement more sustainable lifestyles and eat more environment-friendly foods. Life Cycle Assessment (LCA) is an analytical tool to evaluate the sustainability of a product by examining all the resources used and emissions generated during its life cycle. The first part of this work focused on the upstream production. An LCA of organic blueberry production was conducted to evaluate the trade-off between seasonal and local options and answer the question of whether imported fresh or domestic frozen blueberries are more sustainable. Fresh blueberries from Chile showed superior environmental performance within 2-week storage, due to lower electricity use associated with refrigeration and higher farming yield. Furthermore, length of storage and transportation distance were also found important; if farming yields are comparable, consumption of locally produced, fresh blueberries will be a better choice because of less energy use and shorter transportation distance. The second part of this work targeted at the downstream consumption and aimed to reduce the U.S. environmental footprint through changing adult eating habits. Supplemental functional units were applied in the LCA to incorporate the functions of food to provide nutrition and satiety. With controlled caloric intake, vegetarian diets were found overall more sustainable. However, large possible variations in the environmental impacts of the compared diets were observed due to wide range of nutritional quality of selected foods. Animal products, including meat and dairy especially, and discretionary foods were identified as hotspots in the American diet, that is, reducing the consumption of these foods or deliberately choosing more sustainable alternatives within the same food categories, like chicken and low-fat milk, can significantly improve the sustainability of current American dietary patterns. <br>

Mecanismos dopaminérgicos na aquisição e expressão do medo condicionado: envolvimento de receptores D1 e D2 / Dopaminergic mechanisms in the acquisition and expression of conditioned fear: involviment of D1 and D2 receptors

O aumento do reflexo de sobressalto na presença de um estímulo que tenha sido previamente pareado a choques nas patas é tomado como índice de medo e nomeado sobressalto potencializado pelo medo (SPM). O congelamento, interrupção de todos os movimentos observáveis, exceto aqueles associados com a respiração, também tem sido utilizado como índice de medo em ratos. Um crescente número de evidências sugere o envolvimento de mecanismos dopaminérgicos em diferentes aspectos da memória afetiva, como sua formação, evocação e expressão. No entanto, resultados sobre como e por meio de quais receptores os mecanismos dopaminérgicos influenciam o medo têm sido inconsistentes. O presente estudo examina o envolvimento dos receptores dopaminérgicos na aquisição e na expressão do medo condicionado à luz. Para isso, foram analisados os efeitos do antagonista D1, SCH 23390, do agonista D1, SKF 38393, do antagonista D2, sulpirida, e do agonista D2, quimpirole, no SPM e no congelamento. A atividade motora dos animais também foi avaliada no teste do campo aberto. SCH 23390, SKF 38393, sulpirida e quimpirole, administrados antes do condicionamento, não produziram efeitos no SPM, mas SCH 23390 diminuiu o congelamento. As administrações de SCH 23390, SKF 38393 e sulpirida antes do teste também não produziram efeitos no SPM e no congelamento. Quimpirole, em doses que agem em receptores pré-sinápticos, causou uma redução significativa no SPM e no congelamento, quando administrado antes do teste. A ação das drogas não foi devida a efeitos não-específicos uma vez que elas não produziram efeitos no teste do campo aberto. Os resultados sugerem que mecanismos dopaminérgicos devem estar envolvidos tanto na aquisição, quanto na expressão do medo condicionado à luz. Receptores D1 pós-sinápticos parecem participar da aquisição do congelamento condicionado à luz-CS, mas não do SPM. Por outro lado, receptores D2 pré-sinápticos parecem estar envolvidos na expressão do medo condicionado à luz-CS. / The increase in the startle reflex in the presence of a stimulus that has been previously paired to footshock is taken as an index of fear and named fear potentiated startle (FPS). Freezing behavior, a cessation of all observable movements, except those associated with respiration, has also been used as an index of fear in rats. A growing body of evidence has suggested that dopaminergic mechanisms are implicated in different aspects of affective memory, namely its formation, expression or retrieval. However, the results of studies that have examined how, and through which receptors, dopaminergic mechanisms influence fear have been inconsistent. This work is aimed at examining the involvement of dopaminergic receptors in the acquisition and expression of conditioned fear to ligth-CS. We evaluated the effects of systemic administration of the D1 antagonist, SCH 23390, the D1 agonist, SKF 38393, the D2 antagonist, sulpiride, and the D2 agonist, quinpirole before and after conditioning on FPS and freezing. The motor activity of the animals was also evaluated in an open field test. SCH 23390, SKF 38393, sulpiride and quinpirole, injected before conditioning sessions, did not produce any effect on FPS, but SCH 23390 decreased freezing. Injections of SCH 23390, SKF 38393 and sulpiride before testing session did not produce any effect on FPS or freezing. Quinpirole, injected at doses acting at presynaptic level, caused significant reduction in FPS and freezing, when injected before testing. Drugs’ action was not due to nonspecific effects since they had no effect in the open field test. Our findings indicate that DA mechanisms are involved in the acquisition and expression of conditioned fear using light-CS. Dopaminergic mechanisms mediated by postsynaptic D1 receptors seem to be involved in the acquisition of conditioned freezing to light-CS, but not in FPS. On the other hand, dopaminergic mechanisms mediated by presynaptic D2 receptors seem to be involved in the expression of conditioned fear to light-CS.

The celiac disease has been object of frequent technological and clinical research on food alternatives to persons having the disease. The objective of this study was to develop the gluten free baked products, having a good sensorial acceptance, using cassava starch with the addition of soybeans derived products . From the rice cream, the cassava starch and the maize starch used in the formulation, the experimental design for Simplex-Centróide mixtures was carried through, and after that the software called Statistica 7.0 was used, inserting the maximum limit restriction of 50% of cassava starch, resulting in the experimental design for mixtures, giving then seven samples. Using these seven samples, sensorial and instrumental analysis composed of specific volume, elasticity and firmness analysis was carried through. For the sensorial analysis, the optimized formulation of mixture was 50% of rice cream and 50% of cassava starch, and for the instrumental analysis, the optimal simultaneous point for the three conducted analyses was 20% of rice cream, 50% of maize starch and 30% of cassava starch. From these two optimal points, a second sensorial analysis was realized, and the optimal formulation obtained from instrumental analyses was significantly preferred (hedonic scale average=6,96±1,71, using scale of 1 to 9) comparing to the first sensorial analysis (hedonic scale average=5,77±1,83, using scale of 1 to 9). From these samples, it was realized the comparative analysis of specific volume, elasticity, firmness and triangular test between baked bread and pre-baked and freeze bread. The baked sample and the pre-baked and freeze sample showed differences only in product elasticity, having not showed sensorial difference between the samples analyzed. For the muffin, a sensorial analysis was conducted, that resulted on hedonic scale average 7,76±1,07, using scale of 1 to 9. It was realized instrumental analysis of specific volume, elasticity and firmness, and comparative analysis between baked muffin and pre-baked and freeze muffin, adopting the same procedure used for bread analysis. The baked muffin and pre-baked and freeze muffin differed on specific volume and elasticity, however they didn´t have any sensorial difference when the triangular test was applied. For both, the bread and muffin, it was realized the physic-chemistry, nutritional and microbiological analysis, and both were according to the current law. It was realized cost analyses of the developed products, and the conclusion was that they are economically according with similar products. / A doença celíaca está relacionada à ingestão de alimentos que contêm glúten, proteína presente em algumas matérias-primas vegetais, como trigo, centeio, cevada e aveia, danificando a superfície da mucosa do intestino, dificultando a absorção de nutrientes pelo organismo. A doença celíaca tem sido objeto freqüente de estudos clínicos e tecnológicos em busca de alternativas alimentares para indivíduos que sofrem da enfermidade. O objetivo desse trabalho foi desenvolver produtos panificados isentos de glúten, de boa aceitação sensorial, utilizando amido de mandioca e adicionados de derivados de soja. A partir do amido de mandioca, creme de arroz e amido de milho utilizados na formulação, foi realizado o delineamento experimental para misturas Simplex-Centróide, e posteriormente os dados foram lançados no software Statistica 7.0, inserindo-se a restrição do limite máximo de 50 % de amido de mandioca, que resultou no delineamento para misturas originando sete amostras. A partir dessas sete amostras, foram realizadas análises sensorial e instrumentais de volume específico, elasticidade e firmeza. Para a análise sensorial, a formulação ótima de mistura encontrada foi 50% de creme de arroz e 50% de amido de mandioca, e para a análise instrumental, o ponto ótimo simultâneo das três análises realizadas foi 20% de creme de arroz, 30% de amido de mandioca e 50% de amido de milho. A partir dos dois pontos ótimos obtidos, foi realizada uma segunda análise sensorial, na qual a formulação ótima obtida a partir das análises instrumentais realizadas foi significativamente preferida (média escala hedônica=6,96±1,71, numa escala de 1 a 9) em relação à formulação ótima obtida na primeira análise sensorial (média escala hedônica 5,77±1,83, numa escala de 1 a 9). A partir dessa amostra, foram realizadas análises comparativas de volume específico, elasticidade e firmeza, e teste triangular entre o pão assado e o pão pré-assado e congelado. A amostra assada e a amostra pré-assada e congelada do pão diferiram apenas com relação à elasticidade do produto, não sendo percebida nenhuma diferença sensorial entre as amostras analisadas.Para o muffin foi realizada análise sensorial, a qual apresentou média de escala hedônica de 7,76±1,07, numa escala de 1 a 9. Foram realizadas análises sensorial e instrumentais de volume específico, elasticidade e firmeza comparativas entre a amostra assada e a amostra pré-assada e congelada, adotando o mesmo procedimento aplicado ao pão. O muffin assado e o muffin pré-assado e congelado diferiram com relação ao volume específico e elasticidade, porém não apresentaram diferença sensorial significativa aplicando-se o teste triangular. Para ambos os produtos foram realizadas análises físico-químicas, nutricionais e microbiológicas, as quais mostram estar de acordo com a legislação vigente. Foram realizadas levantamento de custo dos produtos desenvolvidos, os quais mostram estar economicamente de acordo com produtos similares encontrados no mercado.

Mecanismos dopaminérgicos na aquisição e expressão do medo condicionado: envolvimento de receptores D1 e D2 / Dopaminergic mechanisms in the acquisition and expression of conditioned fear: involviment of D1 and D2 receptors

O aumento do reflexo de sobressalto na presença de um estímulo que tenha sido previamente pareado a choques nas patas é tomado como índice de medo e nomeado sobressalto potencializado pelo medo (SPM). O congelamento, interrupção de todos os movimentos observáveis, exceto aqueles associados com a respiração, também tem sido utilizado como índice de medo em ratos. Um crescente número de evidências sugere o envolvimento de mecanismos dopaminérgicos em diferentes aspectos da memória afetiva, como sua formação, evocação e expressão. No entanto, resultados sobre como e por meio de quais receptores os mecanismos dopaminérgicos influenciam o medo têm sido inconsistentes. O presente estudo examina o envolvimento dos receptores dopaminérgicos na aquisição e na expressão do medo condicionado à luz. Para isso, foram analisados os efeitos do antagonista D1, SCH 23390, do agonista D1, SKF 38393, do antagonista D2, sulpirida, e do agonista D2, quimpirole, no SPM e no congelamento. A atividade motora dos animais também foi avaliada no teste do campo aberto. SCH 23390, SKF 38393, sulpirida e quimpirole, administrados antes do condicionamento, não produziram efeitos no SPM, mas SCH 23390 diminuiu o congelamento. As administrações de SCH 23390, SKF 38393 e sulpirida antes do teste também não produziram efeitos no SPM e no congelamento. Quimpirole, em doses que agem em receptores pré-sinápticos, causou uma redução significativa no SPM e no congelamento, quando administrado antes do teste. A ação das drogas não foi devida a efeitos não-específicos uma vez que elas não produziram efeitos no teste do campo aberto. Os resultados sugerem que mecanismos dopaminérgicos devem estar envolvidos tanto na aquisição, quanto na expressão do medo condicionado à luz. Receptores D1 pós-sinápticos parecem participar da aquisição do congelamento condicionado à luz-CS, mas não do SPM. Por outro lado, receptores D2 pré-sinápticos parecem estar envolvidos na expressão do medo condicionado à luz-CS. / The increase in the startle reflex in the presence of a stimulus that has been previously paired to footshock is taken as an index of fear and named fear potentiated startle (FPS). Freezing behavior, a cessation of all observable movements, except those associated with respiration, has also been used as an index of fear in rats. A growing body of evidence has suggested that dopaminergic mechanisms are implicated in different aspects of affective memory, namely its formation, expression or retrieval. However, the results of studies that have examined how, and through which receptors, dopaminergic mechanisms influence fear have been inconsistent. This work is aimed at examining the involvement of dopaminergic receptors in the acquisition and expression of conditioned fear to ligth-CS. We evaluated the effects of systemic administration of the D1 antagonist, SCH 23390, the D1 agonist, SKF 38393, the D2 antagonist, sulpiride, and the D2 agonist, quinpirole before and after conditioning on FPS and freezing. The motor activity of the animals was also evaluated in an open field test. SCH 23390, SKF 38393, sulpiride and quinpirole, injected before conditioning sessions, did not produce any effect on FPS, but SCH 23390 decreased freezing. Injections of SCH 23390, SKF 38393 and sulpiride before testing session did not produce any effect on FPS or freezing. Quinpirole, injected at doses acting at presynaptic level, caused significant reduction in FPS and freezing, when injected before testing. Drugs’ action was not due to nonspecific effects since they had no effect in the open field test. Our findings indicate that DA mechanisms are involved in the acquisition and expression of conditioned fear using light-CS. Dopaminergic mechanisms mediated by postsynaptic D1 receptors seem to be involved in the acquisition of conditioned freezing to light-CS, but not in FPS. On the other hand, dopaminergic mechanisms mediated by presynaptic D2 receptors seem to be involved in the expression of conditioned fear to light-CS.

La lutte contre la délinquance de lucre et de trafic repose aujourd’hui en France sur une stratégie fondée principalement sur l’appréhension des avoirs illicites ainsi générés par cette activité. En pratique, la loi française a conféré aux magistrats, et sous certaines conditions aux enquêteurs, des prérogatives très offensives permettant de placer sous-main de justice des biens patrimoniaux très en amont de la procédure, au plus tôt, et ce indépendamment du droit de propriété et de la présomption d’innocence. Cependant, si la loi a grandement réformé le droit des saisies en créant les saisies pénales spéciales, elle n’a pas donné le cadre à un droit de l’exécution des confiscations. En effet, la confiscation reste le pivot de l’enquête pénale patrimoniale, même si les saisies pénales spéciales en sont aujourd’hui le moteur. Pourtant, l’absence d’une procédure post-sentence est de nature à ouvrir un nouvel espace d’opportunité pour permettre aux condamnés de dissiper une partie de leurs biens.
