Angiogênese é o processo pelo qual novos vasos sanguíneos são formados. Está associada ao desenvolvimento da neoplasia intra-epitelial de alto grau e do carcinoma invasor do colo uterino. Objetivos: O objetivo deste estudo foi avaliar a imuno-expressão da granzima B e do fator de crescimento endotelial vascular (VEGF) com as variantes da neoplasia intra-epitelial escamosa cervical. Métodos: A expressão imuno-histoquímica da granzima B foi estudada no epitélio, estroma e ainda no epitélio - estroma de 142 fragmentos de colo uterino sendo, 34 de neoplasia intra-epitelial cervical de grau I (NIC I), 36 de neoplasia intra-epitelial cervical de grau II (NIC II), 33 de neoplasia intra-epitelial cervical de grau III (NIC III) e 39 fragmentos de colo uterino sem atipias – grupo controle. Foi estudada a imuno-expressão para o fator de crescimento endotelial vascular (VEGF) em 160 fragmentos de colo uterino sendo, 43 de neoplasia intra-epitelial cervical de grau I (NIC I), 33 de neoplasia intra-epitelial cervical de grau II (NIC II), 31 de neoplasia intra-epitelial cervical de grau III (NIC III) e 53 fragmentos de colo uterino sem atipias – grupo controle. Resultados: No estroma, a imunoexpressão para a granzima B na neoplasia intra-epitelial cervical de grau I mostrou-se menor do que na neoplasia intra-epitelial cervical de grau III. Encontrou-se alta imuno-expressão para o fator de crescimento endotelial vascular na neoplasia intra-epitelial cervical de grau III e baixa imuno-expressão na neoplasia intra-epitelial de grau I e no grupo controle. Conclusão: Quanto maior a gravidade da lesão intra-epitelial cervical maior imuno-expressão para a granzima B. Encontrou-se aumento progressivo da imuno-expressão para o fator de crescimento endotelial vascular no grau intenso, de acordo com a severidade das neoplasias intra-epiteliais cervicais. / Introduction: The granzyme family designates a special group of serinoproteases which are stored fully processed and in the active form in cytoplasmic granules of the cytolytic T lymphocytes cells (CTL). Angiogenesis is the process in which new blood vessels are formed. It is associated with development of high degree intraepithelial neoplasia and invasive carcinoma of the uterine cervix. Objectives: This study objective was to evaluate the immuno-expression of granzyme B and vascular endothelial growing factor (VEGF) in the variants of cervical squamous intraepithelial neoplasia. Methods: Granzyme B immunohistochemical expression was studied in the epithelium, stroma and in both epithelium - stroma of 142 fragments of uterine cervix, being 34 grade I cervical intraepithelial neoplasia (CIN I), 36 grade II cervical intraepithelial neoplasia (CIN II), 33 grade III cervical intraepithelial neoplasia (CIN III) and 39 uterine cervix fragments without abnormalities - control group. Immunoexpression of vascular endothelial growing factor was studied in 160 uterine cervix fragments, being 43 grade I cervical intraepithelial neoplasia (CIN I), 33 grade II cervical intraepithelial neoplasia (CIN II), 31 grade III cervical intraepithelial neoplasia (CIN III) and 53 uterine cervix fragments without abnormalities - control group. Results: In the stroma, immuno-expression of granzyme B in grade I cervical intraepithelial neoplasia was smaller than in grade III cervical intraepithelial neoplasia. High VEGF immune-expression was found in grade III cervical intraepithelial neoplasia while it was low in grade I cervical intraepithelial neoplasia and in control group. Conclusion: The higher the severity of the cervical intraepithelial lesion, the higher the immuno-expression of granzyme B. A progressive increasement in VEGF immune-expression was found in the intense grade, according to the severity of cervical intraepithelial neoplasia. / TEDE / BV UNIFESP: Teses e dissertações
