Faktory ovlivňující mrazitelnot ejakulátu býků / Factors influencing the bull sperm freezability

An egg yolk is a common component of diluents used as protectants of sperm cells during the process of cryopreservation. Its substitution by low density lipoproteins (LDL) has shown that it is the LDL that provides the egg yolk with its cryoprotective characteristics: it protects sperms against cold shock and other changes, thus helping to preserve their fertilization ability even after the freezing-thawing process. However, the sperm quality is affected by many other internal and external factors. Therefore, the aim of this study was to evaluate if the effect of the addition of LDL to the diluent of the ejaculate is significantly influenced by the following selected factors - the type of used diluent, the bull´s breed and its individuality and the date of the sampling. Experimental insemination doses were repetitively (four times) obtained from a group of six bulls (three Holstein bulls and three Czech Fleckvieh bulls) at the Natural Hradišťko insemination service s.r.o. The samples of semen were diluted with two types of non-egg diluents containing soybean lecithin extracts (AndoMed and BioXCell). To each of these diluents, LDL at 4, 6, and 8% concentration was added; a non-LDL diluent served as a check. All the insemination doses were frozen by a standard procedure and then stored in liquid nitrogen. The CASA system (Computer Assisted Sperm Analysis) was selected for the evaluation of the sperm motility. Immediately after thawing and then after two hours of incubation in water bath (37 °C), the values of kinematic parameters were obtained from the samples - the total percentage of motile sperm and the percentage of progressively motile sperm. These data were then statistically processed; based on the outputs, VAP, VCL, ALH and the percentage of progressively motile sperm (PMOT) were chosen as representative kinematic parameters. The values of the parameters were higher in almost all evaluated samples diluted with BioXCell, compared to those diluted with AndroMed. Even though we demonstrated the existence of a high variability of results depending on the time of incubation, bull breed, the individuality of the bull and the date of the sampling, it could be said that the best concentration of added LDL is 6 % for BioXCell and 8 % for AndroMed. It would be useful to perform more experiments evaluating the effect of adding LDL to non-egg diluents on the quality of the thawed sperm. In these experiments, more sperm quality parameters should be examined and factors influencing the variability of results demonstrated in this work should be taken into consideration.

The goal of this paper is to clarify Martin Buber's concept of guilt. To make the notion of guilt clear we will use mainly Buber's papers 'I and Thou','The Problem of Man','Images of Good and Evil' and the short essay 'Guilt and Guilt Feelings'. During our analysis we will also focus on other important notions of Buber's philosophical system and their possible development. Among other things, it will be the notion of the world, relationship and essential determination.The whole paper is intended to clarify Buber's dispute with psychoterapists, mainly with S. Freud, over the nature of guilt in the essay 'Guilt and Guilt Feelings'.

Guinea pigs represent domesticated precocious rodents which became common pets. From the first day after birth they are fully vocal. This thesis was devoted to the vocalization of pups. The aim of the thesis was to 1) determine the age when the vocal individuality in whistle sound is demonstrable; 2) test possible ultrasonic signals emitted by young guinea pigs; 3) study the antipredator reactions and alarm calls to aerial (bird of prey) and terrestrial (dog) predators and human (control test). A total of 16 guinea pig pups were tested for vocal individuality, 28 pups for ultrasonic vocalization and 27 adolescents for predator recognition. When testing vocal ontogeny during the first 9 days after birth, I observed changes in temporal, frequency and parameter of intensity . When I test vocal individuality by cross-validated discriminant function analysis (DFA) based on ten acoustic parameters, calls were classified to correct animals with following success: day 1 = 71.9%, day 3 = 58.8%, day 5 = 53.10%, day 7 = 50.60% and day 9 = 63.10%. The highest frequency in whistle was 30.03 kHz. In predator discrimination reactions as freezing, fleeing and vigilance were observed. In the presence of dog, guinea pigs reacted for the longest time and most frequently by freezing. When confronted with a bird of prey, I observed for a longest time and most often fleeing and then freezing. When exposed to a human, guinea pigs showed mostly vigilance. Almost no vocalization was observed except for two events of alarm calls - drrr as in the presence of dog and chirrup as reaction to bird of prey. The following conclusions can be drawn from the presented results: 1) vocal individuality of guinea pigs is demonstrable immediately after birth and the rate of individually different vocal parameters changes with age; 2) guinea pigs are able to produce sound up to 30 kHz, i.e. within the ultrasound range, but signals of high frequencies are not crucial for their communication; 3) guinea pigs discriminate between terrestrial and aerial predators, but they emit alarm calls rather rarely.

The thesis of this work is a treatise on liberalism in general firstly, what contains the etymology of the word liberalism. There are explained the beginnings of this orientation, dating back to ancient times and Middle Ages. The description of the beginnings of liberalism in England follows. The historical events of the time, which played an important role in the birth of liberalism, are accented. Liberalism is based on certain principles as well as any other social or political theory and in this work this principles are enriched especially by ideas of John Locke. Liberalism reached its height in England in the nineteenth century when it defended its leading position. Liberalism is not only social and political orientation but it plays an important role in economy also. It supports free trade without state's interventions and it promotes private enterprise. Although liberalism was often criticized by church representatives and particularly by Conservatives, liberal ideas are still current. Liberal Democrats are the third-largest party in the House of Commons in Great Britain. They have made a coalition government together with the Conservatives since 2010. This thesis proceeds from the Toni Morrison's novel Sula and it mainly concentrates on a mutual relationship between an individual and society as reflected in the book. It consists of two main parts. The first one describes some aspects of the African-American history; the second one focuses on the individual characters in Sula and analyzes the Bottom society, charted against principles of US society in general. Individuality as the basic point of this thesis is shown as the moving element in the development of the society that also gives the direction of that development. Relationship between an individual and society is considered dialectic - on one hand, particular individuals participate in the development of the society, destroy stereotypes and violate dogmas; on the other hand, these people are often subdued and limited for the sake of the proper functioning of the society. Primary basis of this work is an assumption of the necessity of individual driving elements in the society for its (social) ceaseless development and subjectivism as the basic point of view on human existence. This work is strictly theoretical reasoning and comparation of the two important trains of thought, humanistic socialism and liberal competition, which conflict with each other in everyday events of the present world. I am trying in this work to consider arguments, which F.A. Hayek is using to defend his idea about functioning of a liberal order which according to him absolutely spontaneusly creates the best possible environment for an assertion and development of a free individual and these arguments i am trying to follow subsequently with humanistic thinking of Erich Fromm placing the emphasis on his different conception of the individual but also spontaneus freedom and to analyse if it is possible for such individual who is humanly oriented to find full exercise of his inner needs and at the same time to remain useful member just of such society which Hayek submit us. Aim of this work is consideration of every discovered contrast of these two conceptions and thinking about humanistic ideas if they are for the liberal order only past drowned in selfish competitive acting or if they are still actual or if they even have opportunity to fully develop in such order. Benefit of this work I see first of all in the analysis of though not quite sought after but extremely actual problem if free individual is able to spontaneusly create such social order over the time in which his individuality may fully develop or if he dies out in unconconsciously created all embracing conformability.

This dissertation thesis describes the phenomenon of audiovisual performance in different connotations. A substantial part is devoted to psychological survey into my own subconscious and subsequently to metaphorical processing of this introspection. An important part of the work is a resume of the general knowledge of the physical properties and its metaphorical application on mental and creative processes. This analysis and explanation of internal cognitive models are implemented in terms of the study of relationships, interactions based on cooperation or confrontation, but also the inability to grasp the objective reality from the subjective position due to the merged distinctions of real and fictional. The importance of audiovisual performance is also studied in linear perspective of history, in order to point out the general principle of repetition patterns, which reveals that audiovisual performances is an activity which is a necessary expression of life itself and not just the concept in the field of arts.

It's aim is to understand the question: "what does it mean to be individual being?" from the point of view of traditional thinker who is considered to stand on the top of traditional philosophical thinking. In the work I interpret the most important passages of Phenomenology. I point out that to the individuality we can look only as on a single example of subjectivity. Every specific quality and determination of individuality come from outside, individuality is determinated by the common reason and the society where she lives. Individuality in Hegel doesn't mean some sort of special beeing, concrete life (which search modern philosophy of 20th century) but it means only occurence of something "eternal", the consciousness.

Lidská důstojnost a sestra / Human dignity and nurse

The aim of the thesis is to find out the conception of human dignity at medical staff. At nurses, in addition, with a hidden observation to verify to what extent they meet their statements in practice, whether for nurses a patient does not become only an object of relevant therapeutic and nursing acts. The empirical part of this thesis was processed in form of a qualitative research. The data collection technique was a non-standardised interview with nurses at standard in-patient departments, students of the nursery Master study programme, students of a higher nursing school and patients at a standard department. At nurses it was verified, using the technique of hidden observation, to what extent their actual behaviour in practice corresponds to the information mentioned in interviews. Therefore in their interviews we differentiated the mentioned and non-mentioned elements. The interviews were anonymous. They were recorded in writing because only in one case the respondent agreed for us to use a dictaphone. Collection of empirical data proceeded from June to August 2008. Unlike our expectations the research showed that respondents have no conception of human honour and have no complex conception of its keeping in practice. We categorised the rather poor information into five basic categories that covered 92% (intimacy, communication, individuality, empathy and equality of approach). Statements that we did not manage to categorise here create other 5 categories (worthy dying, consider seriously, pain calming, lege artis process and faith). Most often the respondents mentioned the following categories: intimacy (80.4 %), communication (76.8 %), individuality (60.7 %), empathy (39.3 %) and equality of approach (33.9 %). The hidden observation showed that most nurses (13 out of 16, i.e. 81 %) adhered to their fundamentals of keeping human dignity when working with ill people in a satisfactory way. The other 3 nurses made mistakes in all categories except keeping intimacy; here only one of them did wrong. Results of the thesis should contribute to improvement of quality of care and help to better respect for human dignity.
